Slightly squashed and transversely sectioned sprouts (seedlingroot + hypocotyl to cotyledons) of Pisum arvense, Ornithopussativus and Helianthus annuus were examined. The presence ofstomata is described on seedling roots, including the root hairzone, in Pisum and Ornithopus and in the root hair zone of thehypocotyl of Helianthus. The stomata found in the root hairzones are almost always open, usually without chloroplasts andare not sensitive to the action of abscisic acid (ABA). TheABA sensitivity of the stomata appears first above the roothair zone and increases gradually towards the cotyledons. Thepossible role of stomata in the root hair zone is discussed. Hydraulic properties of entire root systems and isolated rootsof three contrasting sugarcane clones were evaluated using transpiration-induceddifferences in hydrostatic pressure across intact root systems,root pressure-generated xylem sap exudation, and pressure-fluxrelationships. Regardless of the measurement technique employed,the clones were ranked in the same order on the basis of theirleaf area–specific total root system hydraulic conductance(Croot). All methods employed detected large developmental changesin Grootroot with maximum values occurring in plants with approximately02 m2 total leaf area. Genotypic ranking according to Groot,was reflected as a similar ranking according to root length-specifichydraulic conductance (L) of individual excised roots. Genotypicdifferences in Groot and L were consistent with anatomical characteristicsobserved in individual roots. Patterns of Groot, during soildrying and following re-irrigation suggested that the declinein Groot, observed during soil drying occurred within the rootsrather than at the soil–root interface and may have beencaused in part by xylem cavitation in the roots. Uptake of the dinitroaniline herbicide oryzalin (3,5-dinitro-N4,N4-dipropylsulfamlamide) and its effect on root growth werestudied using 5 mm corn (Zea mays L.) and pea (Pisum sativum)root apices. Pea root growth was much less susceptible to oryzalinthan corn root growth. Uptake studies showed that pea root apicesalso accumulated much less [14C]oryzalin and had a lower bindingaffinity for this herbicide. [14C]oryzalin was not metabolizedin root apices from either species. Thus, the differential susceptibilityto oryzalin in the case of corn versus pea can be explained,at least in part, by differences in oryzalin uptake and accumulationby roots. Erica x darleyensis grows rhythmically when plants are propagatedin vitro on a medium with nitrate nitrogen. The aim of thisstudy was to determine if transforming the root system withAgrobacterium rhizogenes (strain A4) alters the rhythmic developmentof the aerial shoots of Erica x darleyensis. Isolated root cultures which could be maintained over severalmonths by serial subculture were established from Brassica oleraceavar. italica cv. Green Comet F1. A modified White's medium wasfound to be the best of several salt compositions tested. Theeffects on isolated root growth of the following were also examined;nutritional components, culture vessel type and closure, pHof the medium and auxin type and concentration. Using a mediumdevised for Green Comet, root cultures were established fromsix other B. oleracea, B. napus and B. campestris cultivars. It was possible to regenerate shoots from segments of culturedroots by incubation on agar-solidified media containing cytokininand auxin. The effects on regeneration of various auxins andcytokinins were investigated; the combination of Picloram withKN gave the highest frequency of shoot formation. It was demonstratedthat isolated roots retained their regenerative ability overa period of 5 months in culture. We recently described an in vitro approach that uses root culturesto study haustorial formation in Striga asiatica. Previous studieshave used haustoria formed on intact radicles of Striga seedlings.In vitro cultured roots formed haustoria that appeared morphologicallysimilar to those formed by Striga radicles, but were 5–10-foldlarger. In this study, we provide biochemical and histologicalevidence to support further the similarity of root culture haustoriato haustoria formed on radicles of seedlings. We examined invivo protein synthesis during haustorial development on rootcultures and radicles by 2-D PAGE. Four proteins increased inabundance in both root cultures and radicles after 6 h of haustorialinduction. All four proteins appeared transiently in root culturesand radicles, being more abundant at 6 h, and less abundantafter 24 h of haustorial induction. Only three of the four haustorial-specificproteins were more abundant in root cultures after 2 h of haustorialinduction; all four had decreased in abundance after 12 h ofhaustorial induction. Using light microscopic analysis we comparedthe ontogeny of root culture haustoria to that of haustoriaon radicles. These studies revealed that root culture haustoriaundergo developmental changes similar to those reported forradicle haustoria such as early expansion of cortical cells,the emergence of haustorial hairs from epidermal cells, andthe development of densely staining cells at the haustorialapex. In addition, these changes occurred within a similar time-frameand sequence in root culture and radicle haustoria. Griselinia littoralis roots quickly and vigorously from cuttingsof seedlings and mature plants and also forms roots on detachedleaves. Cuttings root in the dark but leaves must be present.In contrast G. lucida roots vigorously only from cuttings takenfrom seedlings. Light is essential for root formation on oldermaterial. Seed swelling, germination, root extension, lateral root initiationand shoot growth were studied in soils of different water contents,using non-destructive, serial neutron radiography. Seeds fromthree varieties of soya beans (Glycine max L.) and one varietyeach of maize (Zea mays L.) and vetch (Vicia sativa L.) wereused. The seeds germinated when they had increased in size bya certain amount, if germination is taken as the time when theradicle first appears. The rate at which roots and shoots extendalso depend on soil water content. Species of the genus Veronica differ in habitat preferences,growth form and in adventitious root production. The annualspecies rarely or never produce adventitious roots in intactplants in the field but some, for example V. persica and V.arvensis will root vigorously from single node stem segmentsin culture. Others, such as V. agrestis require the presenceof IAA for substantial levels of root formation to occur incultured stem segments. Veronica hederifolia cuttings rarelyproduce roots. Stem cuttings of the perennial species, in general,rooted more vigorously than those of annual plants. Both V.fihiformis and V. serpyllifolia root very strongly. The position of root production from the stem cuttings differedfrom species to species. Roots arose either from the node, theregion of the base or at some intermediate point. Veronica arvensis,V. chamaedrys and V. persica rooted mainly from the basal regionwhereas V. filiformis rooted mainly from the node. Veronicaserpyllifolia cuttings rooted at both of these locations. Veronica filiformis, a perennial species that is infertile inBritain, produces root primordia in intact plants at nodes whichare close to the shoot apex. Thus, even very young stem segmentshave ‘preformed’ root primordia. For this reason,detached stem segments of V. filiformis root very rapidly andthis probably has been of great significance in its successfulinvasion and spread in lawns and short turf areas. A recessive maize mutant with disrupted seedling developmentwas isolated following transpositional mutagenesis with Mutator.This mutant, initially identified during germination on thebasis of abnormal growth of the scutellar node, was designatedlsn1 (l arge s cutellar n ode). The mutant seedling exhibitsan enlarged primary root with a longitudinal groove and multipleseparate root tips. The mutant root is shorter than normal,because of defective cell elongation, and lacks lateral roots.The mutant plant shows defective leaves and reduced internodeelongation. Histological analyses on primary root, shoot, scutellarnode and juvenile leaves revealed a series of defects, all relatedto an irregular differentiation of vascular elements. In addition,in situ hybridization of mutant leaves demonstrates an abnormalpattern of expression of Knotted-1, a marker of meristem function.The presence of multiple roots fused together can be interpretedas suppression of the negative control responsible for the differentiationof only one root primordium. Stem cuttings of Chrysanthemum morifolium Ram. were treatedwith aqueous solutions of (2-chloroethyl)phosphonic acid (Ethrel)and indolebutyric acid (IBA). Ethrel (1 mg per 1) as a dip oras two foliar sprays, promoted root length and branching inMrs. Roy and Clipper which are difficult-to-root cultivars,but had no effect on Improved Mefo, an easy-to-root cultivar.IBA increased root number in both Clipper and Improved Mefo.The results suggest that IBA and Ethrel act at different stagesof the rooting process, with IBA promoting initiation and Ethrelstimulating elongation and branching. Increases in root numberwith IBA treatment, and in root length with Ethrel treatment,were accompanied by decreases in the soluble carbohydrate concentration,particularly in stem bases.  相似文献

Patterns of root cortex cell development and ultrastructurewere analysed in Sagittaria lancifolia L., Thalia geniculataL. and Pontederia cordata L. using scanning and transmissionelectron microscopy (SEM, TEM). In all three species, cortexcells were arranged in radial columns extending from the endodermisto the hypodermis/epidermis. During gas space formation, thecortex cells elongated parallel to the root radius and shrankin the plane perpendicular to the radius leaving long and thinrows of cortex cells extending from the endodermis to the epidermis.Although the cortex cells appeared collapsed in tissue withwell-developed gas spaces, TEM revealed that the cortical cellsas well as the epidermal cells maintained intact membranes andmany normal organelles. Formation of root cortex tissue withwell-developed gas spaces does not require cell death in thesespecies. Living cortex cells in root tissue with mature gasspaces could provide a symplastic pathway for transport betweenthe root stele and the living epidermal cells. The constraints on water uptake imposed by individual root tissueswere examined forOpuntia ficus-indicaunder wet, drying, andrewetted soil conditions. Root hydraulic conductivity (LP) andaxial conductance (Kh) were measured for intact root segmentsfrom the distal region with an endodermis and from midroot witha periderm;LPwas then measured for each segment with successivetissues removed by dissection. Radial conductivity (LR) wascalculated fromLPandKhfor the intact segment and for the individualtissues by considering the tissue conductivities in series.Under wet conditions,LRfor intact distal root segments was lowestfor the cortex; at midroot, where cortical cells are dead,LRforthe cortex was higher and no single tissue was the predominantlimiter ofLR.LRfor the endodermis and the periderm were similarunder wet conditions. During 30d of soil drying,LRfor the distalcortex increased almost three-fold due to the death of corticalcells, whereasLRfor the midroot cortex was unaffected;LRforthe endodermis and the periderm decreased by 40 and 90%, respectively,during drying. For both root regions under wet conditions, thevascular cylinder had the highestLR, which decreased by 50–70%during 30d of soil drying. After 3d of rewetting, new lateralroots emerged, increasingLRfor the tissues outside the vascularcylinder as well as increasing uptake of an apoplastic tracerinto the xylem of both the roots and the shoot. The averageLRforintact root segments was similar under wet and rewetted conditions,but the conductivity of the tissues outside the vascular cylinderincreased after rewetting, as did the contribution of the apoplasticpathway to water uptake. Root caps of primary, secondary, and seminal roots of Z. mayscv. Kys secrete large amounts of mucilage and are in close contactwith the root all along the root apex. These roots are stronglygraviresponsive. Secondary and seminal roots of Z. mays cv.Ageotropic are also strongly graviresponsive. Similarly, theircaps secrete mucilage and closely appress the root all alongthe root apex. However, primary roots of Z. mays cv. Ageotropicare non-responsive to gravity. Their caps secrete negligibleamounts of mucilage and contact the root only at the extremeapex of the root along the calyptrogen. These roots become graviresponsivewhen their tips are coated with mucilage or mucilage-like materials.Peripheral cells of root caps of roots of Z. mays cv. Kys containmany dictyosomes associated with vesicles that migrate to andfuse with the plasmalemma. Root-cap cells of secondary and seminal(i.e. graviresponsive) roots of Z. mays cv. Ageotropic are similarto those of primary roots of Z. mays cv. Kys. However, root-capcells of primary (i.e. non-graviresponsive) roots of Z. mayscv. Ageotropic have distended dictyosomal cisternae filled withan electron-dense, granular material. Large vesicles full ofthis material populate the cells and apparently do not fusewith the plasmalemma. Taken together, these results suggestthat non-graviresponsiveness of primary roots of Z. mays cv.Ageotropic results from the lack of apoplastic continuity betweenthe root and the periphery of the root cap. This is a resultof negligible secretion of mucilage by cells along the edgeof the root cap which, in turn, appears to be due to the malfunctioningof dictyosomes in these cells. Plants of Plantago lanceolata L. and Zea mays L., cv. ‘Campo’were grown at two levels of light intensity. Especially in theroots, the rate of dry matter accumulation decreased at lowlight intensity. The carbohydrate content of both roots andshoots of P. lanceolata was not affected by light intensity.The relative contribution of SHAM1-sensitive respiration, thealternative chain, to total root respiration of both P. lanceolataand Z. mays, was not affected by light intensity during thedaytime. The alternative pathway was somewhat decreased at theend of the dark period, but not in the root tips (0–5mm) where it still contributed 56% in respiration. It was, therefore,concluded that photosynthesis is not a major factor in regulationof root growth in the species investigated. To see whether the effect of light intensity on root growthrate was via transpiration, plants of Z. mays were grown atdifferent air humidities. Both high humidity and low light intensityaffected the root morphology in such a way that the distancebetween the apex and the first laterals on the primary rootaxis increased. It is suggested that this effect on root morphologyis due to transpiration and the subsequent removal of root-producedinhibitors of lateral root growth; although light intensityalso affected the rate of dry matter accumulation of roots andthe rate was not affected by the humidity of the air. It is,therefore, concluded that the effect of light intensity on therate of dry matter accumulation of roots of Z. mays is not viaan effect on transpiration.  相似文献

When stressed by low nutrient availability, young sunflowerplants (Helianthus annuus) showed responses seen in many otherspecies: increases in root uptake capacity (Vmax, l/Km), root:shoot ratio, and putative nutrient-use efficiency, nUE=l/(tissuenutrient content). A straightforward mechanistic model is derivedfor relative growth rate (RGR) in solution culture in termsof these factors. A linear regression based on the model indicatesa negative role for nUE, which violates a premise of the model.A revised model proposes that primary adaptations are only inuptake rate and growth or nutrient allocations, and these actthrough the photosynthetic utility of nutrient. The tissue nutrientcontent and associated nUE become dependent quantities. Thepredictions for RGR, as tested by linear regression, are improved.The model predicts that nUE can increase as external solutionconcentration decreases, but decreases with increased uptakeadaptations in one given environment. The decrease in nUE compromisespotential gains in RGR from uptake adaptations, and makes increasesin root: shoot ratio a nearly insignificant contributor to earlyRGR. The model and associated regression analyses are generalizedfor additional adaptations such as increased root fineness andfor different quantitative ways that a nutrient may limit photosynthesis.The model and analyses are also generalized to plant growthin soil and growth without functional balance between root andshoot. Anatomical investigations were carried out on the structureand development of the mature secondary haustorium in Alectravogelii growing on Arachis hypogaea or Vigna unguiculata. Followingthe formation of the young secondary haustorium, both the cambiumand pericycle of the host root directly opposite the young secondaryhaustorium are stimulated to divide and form new tissues andorgans including haustorial roots. Further proliferations ofthe host root pericycle and the haustorial cortex give riseto a large, tuberous and complex mature secondary haustoriumwithin which the tissues of the host and parasite remain inintimate contact forming a perfect graft union with a wide zoneof contact. Apart from the haustorial axial xylcm strand whichnormally connects the xylem of the parasite secondary root withthat of the host, direct xylary connections are also establishedbetween the axial xylem of the haustorium and the xylem of thehaustorial roots. The entire surface of the mature secondaryhaustorium of Alectrais covered with these haustorial rootsas was previously observed in its mature primary haustorium. Anatomical studies were carried out on initiation of the secondaryhaustorium in Alectra vogelii, a root parasite of leguminouscrops in Nigeria. In both the normal and self-haustorium, theformation of the haustorial initial on the parasite root soonafter initial contact between the host and parasite roots isfollowed by the penetration of the host root by the haustorium.Specialized penetrating cells (intrusive cells) at the haustorialfront prise apart and loosen the host root cortical cells, whichlater become digested. Through the same processes, a few ofthese columnar intrusive cells at the haustorial front piercethe endodermis to make contact with the xylem of the host root.Thereafter, a true conductive bridge consisting of short, isodiametric,reticulate vessel elements is established between the parasiteand host roots through the secondary haustorium. No pholem tissuewas observed in the connection. There is a close similaritybetween the mode of initiation of the secondary haustorium ofAlectra vogelii and that previously described for its primaryhaustorium.
