Bacillus thuringiensis serovar japonensis strain Buibui experimental formulations containing Cry8Cal delta-endotoxin were evaluated in the field for the control of oriental beetle, Anomala orientalis Waterhouse; Japanese beetle, Popillia japonica Newman; Asiatic garden beetle, Maladera castanea (Arrow); and European chafer, Rhizotrogus majalis (Razoumowsky) larvae infesting turfgrasses. In 17 fall applications, control of A. orientalis and P. japonica was achieved at rates as low as 100 g of toxin per ha; however, rates of 301 or 593 g/ha were consistently more effective. A. orientalis larvae were the most susceptible followed by P. japonica, R. majalis, and M. castanea. Two spring applications of 120 g of toxin per ha were not effective against oriental or Japanese beetles.  相似文献

Bacillus thuringiensis subspecies kurstaki and aizawai are important control agents for lepidopteran pests. Bioassays were designed to test B. t. kurstaki and aizawai against second- and-fourth instar black cutworm larvae with and without Bacillus sp. NFD2 and Pseudomonas sp. FNFD1 bacteria. B. thuringiensis subsp. aizawai (XenTari) was more toxic to both second- and fourth-instar black cutworm, Agrotis ipsilon (Hufnagel) (Lepidoptera: Noctuidae), larvae than B. t. kurstaki (DiPel) at 7 d after treatment (DAT). When DiPel was combined with NFD2 or FNFD1 versus second instars, the LC50s were 5.0X and 4.7X lower, respectively, than with DiPel alone. DiPel combined with both NFD2 and FNFD1 versus second instars resulted in an LC50 value 7.7X lower than with DiPel alone. When XenTari was combined with NFD2 or FNFD1 versus second instars, the LC50s were 5.2X and 3.8X lower, respectively, than with XenTari alone. XenTari combined with both NFD2 and FNFD1 versus second instars resulted in an LC50 9.7X lower than with XenTari alone. When DiPel was combined with NFD2 or FNFD1 versus fourth instars, the LC50s were 4.4X and 3.4X lower, respectively, than with DiPel alone. DiPel combined with both NFD2 and FNFD1 versus fourth instars resulted in an LC50 5.0X lower than with DiPel alone. When XenTari was combined with NFD2 or FNFD1 versus fourth instars, the LC50s were 5.7X and 3.3X lower, respectively, than with XenTari alone. XenTari combined with both NFD2 and FNFD1 versus fourth instars resulted in an LC50 6.7X lower than with XenTari alone.  相似文献

Isolation of Bacillus thuringiensis (Bt) strain or its cry gene encoding insecticidal crystal protein (ICP) with specific toxicity is of great importance to biological control of insect pests. In this study, by screening 66 strains of Bt isolated from soil samples collected in Shandong Province, China, a new cry8-type gene from Bt strain B-JJX was identified via PCR-RFLP method. This novel gene, cry8Ab1, was cloned from the Bt strain B-JJX and expressed in an acrystalliferous mutant strain HD-73?. The open reading frame of the cry8Ab1 gene consists of 3543 bp with a G + C content of 37.99% and encodes a protein of 1180 amino acids with a putative MW of 133.3 kDa which was confirmed by SDS-PAGE analysis. The Cry8Ab1 protein was expressed and released as spherical parasporal crystals from Bt acrystalliferous mutant strain HD-73? along with the presence of spores. In bioassays, this protein was toxic to 3-day-old larvae of the scarabaeid pests, Holotrichia oblita and H. parallela, with an LC50 of 5.72 and 2.00 μg toxin g?1 soil, respectively. The results are in accordance with the insecticidal activities of the original Bt strain B-JJX, which had an LC50 of 1.72 and 0.96 μg toxin g?1 soil against H. oblita and H. parallela, respectively.  相似文献

After greenhouse and outdoor microplot experiments, a critical density of two third instars per microplot for third instars of European chafer, Rhizotrogus (Amphimallon) majalis (Razoumowsky), in corn, Zea mays L., was derived. On average, the number of missing or damaged plants increased approximately 8% from zero to two larvae per 900 cm2. Furthermore, 23 fields in 2 yr were sampled for larvae along transects by using a golf cup cutter as the sampling tool and the critical density of 0.2 larva per sampling unit as the critical density. The sampling unit was one golf cup cutter with a diameter of 10.8 cm or 91.4 cm2 (10 sampling units approximately 900 cm2 approximately 1 foot2). Fieldwide means and variation were modeled to Taylor's power law, a = 1.42 and b = 1.47, and 20 of 23 fields fit the negative binomial probability distribution. Wald's formula for a sequential sampling plan was most accurate and least time-consuming, according to the operating characteristic and the average sample number function, relative to Iwao's and converging lines formulae. Percentage of sand, topography, soil bulk density, and proximity to trees were measured as potential predictors of areas with high larval density. Percentage of sand and soil bulk density were significant predictors, and topography and proximity to trees were not significant predictors. Field areas where the percentage of sand is high and the soil bulk density is low to moderate or where the percentage of sand is moderate and the soil bulk density is low should be chosen as sampling locations.  相似文献

【目的】明确昆虫病原线虫 Heterorhabditis beicherriana LF品系(LF)与苏云金芽孢杆菌 Bacillus thuringiensis HBF-18菌株(Bt HBF-18)混用后对华北大黑鳃金龟 Holotrichia oblita 幼虫的致病力的协同增效作用,为该害虫的防治提供新的技术措施。【方法】在室内测定了LF在不同使用剂量、不同环境温度及不同土壤湿度条件下对华北大黑鳃金龟7-10日龄幼虫的致病力;通过室内生测测定了Bt HBF-18对LF存活的影响,以及Bt HBF-18与LF两者混用后对7-10日龄华北大黑鳃金龟幼虫的防治效果;同时通过室外盆栽试验测定了两者混用对华北大黑鳃金龟幼虫的防治效果。【结果】华北大黑鳃金龟幼虫死亡率随LF施用剂量和处理时间的增加而升高,其中,侵染期线虫(infective juveniles, IJs)800 IJs/100 μL及以上剂量处理7 d后幼虫死亡率达到了100%;25℃为该线虫侵染的最适宜环境温度;适宜土壤湿度范围为14%~20%,湿度过低或过高都会显著影响其侵染效率。室内生测结果表明, Bt HBF-18处理9 d对华北大黑鳃金龟幼虫的致死中浓度(LC 50 )为 1.44× 10^8 CFU/g土,此浓度对LF的存活基本没有影响。另外,室内生测和室外盆栽试验结果均表明,将LF与Bt HBF-18混用能显著提高对华北大黑鳃金龟幼虫的防治效果,混用后具有不同程度的加成或协同增效作用。室内生测试验中LC 50 Bt+200 IJs/100 μL LF混用处理3 d后,较单独LF和Bt HBF-18处理幼虫死亡率分别提高了约43.07%和36.05%,具有显著的协同增效作用;室外盆栽试验中1/2 LC 50 Bt+1 000 IJs/mL LF, LC 50 Bt+1 000 IJs/mL LF和1/2 LC 50 Bt+1 500 IJs/mL LF均具有协同增效作用,其中1/2 LC 50 Bt+1 500 IJs/mL LF增效作用最佳,较单独LF和Bt HBF-18处理幼虫死亡率分别提高了约38.89%和80.55%。【结论】将昆虫病原线虫LF与Bt HBF-18混用对华北大黑鳃金龟幼虫的防治具有加成或协同增效作用。  相似文献

1 Although management of the oriental beetle Anomala orientalis (Waterhouse) by mating disruption shows promise across a range of agricultural systems, relatively little is known about aspects of the reproductive biology of this species relevant to its management. We studied the effects of delayed mating on several aspects of the oviposition behaviour and biology of the oriental beetle using females mated in the laboratory at 4–13 days posteclosion. 2 Females exhibited a gradual decline in fecundity with increased age at mating that was largely a function of a decline in duration of the oviposition period, as well as lower female fertility: females mated at 11 days were less likely to lay any fertile eggs. However, egg fertility did not vary with female age at mating. 3 Because mating delay did not affect longevity, females mated at older ages experienced decreased oviposition periods; however, females laid more eggs per day with increased age at mating, which partially offset shorter oviposition periods. 4 A mating delay of ≥ 6 days relative to females mated within the first day of reaching sexual maturity resulted in an approximately 35–50% lower mean fecundity. 5 These results suggest that, for mating disruption to be a successful management tool for the oriental beetle, mating must be prevented rather than delayed. We compare the findings of the present study with the published research on delayed mating in moths and discuss the importance of these results in relation to management of the oriental beetle using female sex pheromones.  相似文献

This study compared the efficacy of SPLAT-OrB, a new pheromone formulation for oriental beetle mating disruption that can be mechanically applied, with hand-applied plastic dispensers in commercial blueberry fields. Both formulations were tested at 2.5 and 5 g of the major sex pheromone component (Z)-7-tetradecen-2-one per hectare, and evaluated by measuring trap shut-down, mating success of caged females, and the number of grubs in sentinel blueberry pots baited with tethered females. All pheromone-treated plots had fewer male captures in traps and lower mating success of caged females compared with untreated plots. SPLAT-OrB, and plastic dispensers at the higher rate, also reduced the number of grubs in sentinel pots. To understand the mechanism of mating disruption in oriental beetle, males were observed approaching the pheromone sources in disrupted plots. In addition, male oriental beetle captures were quantified in plots treated with varying SPLAT-OrB dollop densities per ha. Consistent with predictions for competitive attraction, field observations revealed males approaching the pheromone source and male captures decreasing concavely with increasing dollop density. In a mark-release-recapture study, male oriental beetles responded to SPLAT-OrB dollops and plastic dispensers at least 60 m from the source. Additionally, SPLAT-OrB emitted pheromone that was attractive to male oriental beetles for >5 wk; however, emission rates and attraction dropped rapidly during the first 3-4 wk. This study demonstrates the feasibility of using SPLAT-OrB for oriental beetle mating disruption as an alternative to hand-applied plastic dispensers, and conclusively reveals that a principal mechanism is the competitive attraction of males.  相似文献

Field and laboratory studies were conducted to identify potential resistance among crape myrtles, Lagerstroemia spp., to Japanese beetle, Popillia japonica Newman and to flea beetles, Altica spp. Damage ratings revealed variation among cultivars in susceptibility to beetle feeding. Cultivars with Lagerstroemia fauriei Koehne in their parentage exhibited the least amount of damage in choice and no-choice experiments, with few exceptions. The data indicate that both beetle species cause more feeding damage on certain cultivars of Lagerstroemia indica L., such as 'Country Red', 'Twilight', and 'Carolina Beauty' than interspecific cultivars with L. fauriei in their parentage, such as 'Natchez', 'Tonto', and 'Muskogee'. When comparing the effect of parentage on all of the major pests of crape myrtle, L. faurei confers resistance to all pests except crape myrtle aphid. No correlation was found between leaf toughness, leaf color, and leaf nutrients in estimating flea beetle cultivar preference. With this information, growers can more effectively target scouting measures to the most susceptible cultivars. and breeders can select plants that will require the fewest chemical inputs.  相似文献

Pristionchus pacificus has been developed as a nematode satellite organism in evolutionary developmental biology. Detailed studies of vulva development revealed multiple differences in genetic and molecular control in P. pacificus compared to the model organism Caenorhabditis elegans. To place evolutionary developmental biology in a comprehensive evolutionary context, such studies have to be complemented with ecology. In recent field studies in western Europe and eastern North America we found 11 Pristionchus species that are closely associated with scarab beetles and the Colorado potato beetle. However, P. pacificus was not commonly found in association with scarab beetles in these studies. Here, we describe the results of a similar survey of scarab beetles in Japan. Pristionchus pacificus was the most common Pristionchus species on scarab beetles in Japan, with 40 out of 43 (93%) isolates. The other Pristionchus isolates represent three novel species, which we refer to as Pristionchus sp. 11, Pristionchus sp. 14, and Pristionchus sp. 15. Thirty-seven of the established P. pacificus strains were found on the oriental beetle Exomala orientalis. Laboratory studies with the sex pheromone (Z)-7-tetradecen-2-one of the oriental beetle revealed that P. pacificus shows strong olfactory attraction to the beetle's sex pheromone, which provides a potential mechanism for the recognition and interaction of P. pacificus and E. orientalis. Together, this study identifies P. pacificus as the most common Pristionchus nematode in field studies in Japan, identifies E. orientalis as an important host species, and provides the basis for the ecology of P. pacificus.  相似文献

The oriental beetle Exomala (Anomala) orientalis (Waterhouse) is an important pest of turfgrass in Korean golf courses, and although a few chemical insecticides are registered for insect pest control, they are not very effective against scarab larvae. There is also a growing concern in Korea about the run-off of insecticides into sensitive habitats and the potential for groundwater contamination. A safe and environmentally sound alternative is needed to conventional insecticides. We therefore evaluated six Korean entomopathogenic nematode isolates: S. carpocapsae Pocheon, S. glaseri Dongrae, S. glaseri Mungyeong, S. longicaudum Gongju, S. longicaudum Nonsan, and Heterorhabditis sp. Gyeongsan for their potential as bioinsecticides for control of E. orientalis. In addition, we evaluated a reduced chemical insecticide approach that combined chlorpyrifos-methyl with nematodes. In laboratory tests Heterorhabditis sp. Gyeongsan was the most efficacious, causing 100% mortality of the second and 38% of the third instars. All other nematode isolates caused 50-80% mortality of the second and 15-30% of the third instars. E. orientalis pupae were highly susceptible to all the Korean entomopathogenic nematode isolates except S. carpocapsae. In artificially infested field plots, all Korean nematode isolates cause 50-70% mortality of the third instar. A combination of a one-half rate of Heterorhabditis sp. and a one-half rate chlorpyrifos-methyl was synergistic, causing 91% mortality compared with 69% for the full rate of Heterorhabditis sp. or 22% for the full rate of chlorpyrifos-methyl. In a second field trial, a natural infestation of preoverwintering third instar was treated. In this trial a one-half rate of S. longicaudum Nonsan plus a one-half rate of chlorpyrifos-methyl caused 96.8% mortality, much more than a full rate of S. longicaudum Nonsan (45.9% mortality) or a full rate of chlorpyrifos-methyl (28.7% mortality). The interactions of Heterorhabditis sp. and S. longicaudum Nonsan with chlorpyrifos-methyl in our field trials appear to be synergistic.  相似文献

Abstract The dung beetle genus Phanaeus as currently recognized by Edmonds (1994) consists of 51 species placed in 13 species groups and two subgenera. Here, I examine the phylogeny and biogeography of this genus by analysing the mitochondrial cytochrome oxidase subunit I (530 bp), nuclear large subunit ribosomal RNA (28S, D2 region), and 67 morphological characters for 28 species of Phanaeus. Both maximum parsimony and Bayesian analyses from the combined data yielded well‐resolved trees, although low bootstrap and posterior probability support were found for basal nodes. The phylogenetic hypotheses presented here suggest that the subgenera Phanaeus s.str. and Notiophanaeus should each be elevated to the status of full genus. With the exception of the eucraniine outgroups, the paleano species group of the genus Phanaeus is recovered as sister to all other taxa, including the outgroups Oxysternon, Sulcophanaeus and Coprophanaeus. High bootstrap values and posterior probabilities supported the species groups endymion, tridens and vindex. Biogeographical analyses suggest an ancestral distribution for Phanaeus in the Andes in South America, although numerous dispersal events evidently have produced a complicated biogeographical history.  相似文献

Three cultivars of tall fescue, Festuca arundinacea Schreb., were compared with three cultivars each of fine fescue (Festuca spp.), Kentucky bluegrass (Poa pratensis L.), and perennial ryegrass (Lolium perenne L.) to evaluate tolerance to root-feeding by European chafer grubs, Rhizotrogus majalis (Razoumowsky). Potted turfgrasses were infested with initial densities equivalent to 33 or 66 grubs per 0.1 m2 on 19 August 2000. More grubs were added in late September and October, bringing the total to 66 or 143 grubs per 0.1 m2. Plant growth, root loss, weight gain, and survival of grubs were measured. The experiment was repeated in fall of 2001 with an initial density of 66 grubs per 0.1 m2. The proportion of root mass lost as a result of grub feeding was a function of turf species, root growth, grub survival, and grub growth during the test. Grubs gained the most weight and consumed the most roots when feeding on fine fescue. Fine fescue suffered the greatest percentage of root loss in 2000, despite having the most rapid root growth and largest mass in control pots. Cultivars of tall fescue appeared to be the most tolerant of grub feeding, having the smallest reduction in root mass in both years. Data from fine fescue, Kentucky bluegrass, and perennial ryegrass cultivars were not as consistent as tall fescue, because for some cultivars root growth and grub survival were different between years. We also found that grubs increased in mass by 20% when the mass of available roots was doubled.  相似文献

Sublethal exposure of Colorado potato beetle, Leptinotarsa decemlineata (Say), larvae to the delta-endotoxin of Bacillus thuringiensis variety tenebrionis (Berliner) caused a dose-dependent reduction in feeding and weight gain when tested in a leaf disk bioassay. The highest doses of chronic (continuous-lower concentration) exposure resulted in peak foliage consumption on day 1 as compared with peak consumption on days 3 and 4 when exposure was acute (24-h higher concentration). Dose and exposure regimen interacted significantly in their effects on the extension of development. When development time was analyzed separately for each exposure regimen, only acute exposure caused significant delays in development that extended through to adult eclosion. The efficiency of conversion of ingested material to biomass (ECI) declined significantly with both exposure regimens. The lethal and most sublethal effects of exposure to delta-endotoxin were not cumulative, in that similar total doses, whether delivered acutely or chronically, produced different effects. Female adults that survived acute and chronic exposure to delta-endotoxin as larvae had significantly reduced weight and longevity, and tended to produce fewer eggs (45 and 44% reductions in acute and chronic exposures, respectively) when compared with control adults. The intrinsic rate of increase (r) and net reproductive rate (R0) also appeared to be reduced.  相似文献

Laboratory trials examined the effect of the bioinsecticide Novodor® (active ingredient 3% Bacillus thuringiensis var. tenebrionis (w/w) (Btt)), on the Australian plague soldier beetle, Chauliognathus lugubris, a predator of the Tasmanian eucalyptus leaf beetle Chrysophtharta bimaculata . Spraying of C. lugubris and the C. bimaculata eggs supplied as food showed that neither Novodor® FC with active nor inactive Btt caused any decrease in predatory performance, longevity or fecundity of C. lugubris during the period of their population peak (December). Towards the end of their peak population levels, in late January, there was a steady increase in mortality which was not related to either of the Novodor® FC treatments. On average, male:female pairs of C. lugubris laid one egg batch and ate 30 C. bimaculata eggs a week whether untreated or sprayed with either Novodor® FC. This Btt-based insecticide appears to be a suitable component of an integrated pest management (IPM) regime that aims to conserve natural predators.  相似文献

Japanese beetle, Popillia japonica Newman is a major pest of turf and ornamentals. Laboratory bioassays were conducted to evaluate the potential interactions between a biological control agent, Heterorhabditis marelatus (Nematoda: Heterorhabditidae) IN strain and the insecticide halofenozide against both overwintered and nonoverwintered 3rd instars of Japanese beetle. Treatments consisted of all combinations of 2 rates of halofenozid with H. marelatus nematodes Imidacloprid was used as a standard. Percentage larval mortality was evaluated at 7, 14, and 21 d after treatment. No deleterious effects were observed. The nematode treatments generally produced significantly greater larval mortality relative to both chemical treatments. Twenty-one days after treatment, both rates of nematodes resulted in 100% mortality, whereas insecticide treatments did not surpass 60% mortality. No synergism was detected in any of the combination treatments. There were no significant differences in nematode reproduction in larvae exposed to halofenozide and nematodes versus larvae exposed to only nematodes.  相似文献

The use of Bacillus thuringiensis as a biopesticide is a viable alternative for insect control since the insecticidal Cry proteins produced by these bacteria are highly specific; harmless to humans, vertebrates, and plants; and completely biodegradable. In addition to Cry proteins, B. thuringiensis produces a number of extracellular compounds, including S-layer proteins (SLP), that contribute to virulence. The S layer is an ordered structure representing a proteinaceous paracrystalline array which completely covers the surfaces of many pathogenic bacteria. In this work, we report the identification of an S-layer protein by the screening of B. thuringiensis strains for activity against the coleopteran pest Epilachna varivestis (Mexican bean beetle; Coleoptera: Coccinellidae). We screened two B. thuringiensis strain collections containing unidentified Cry proteins and also strains isolated from dead insects. Some of the B. thuringiensis strains assayed against E. varivestis showed moderate toxicity. However, a B. thuringiensis strain (GP1) that was isolated from a dead insect showed a remarkably high insecticidal activity. The parasporal crystal produced by the GP1 strain was purified and shown to have insecticidal activity against E. varivestis but not against the lepidopteran Manduca sexta or Spodoptera frugiperda or against the dipteran Aedes aegypti. The gene encoding this protein was cloned and sequenced. It corresponded to an S-layer protein highly similar to previously described SLP in Bacillus anthracis (EA1) and Bacillus licheniformis (OlpA). The phylogenetic relationships among SLP from different bacteria showed that these proteins from Bacillus cereus, Bacillus sphaericus, B. anthracis, B. licheniformis, and B. thuringiensis are arranged in the same main group, suggesting similar origins. This is the first report that demonstrates that an S-layer protein is directly involved in toxicity to a coleopteran pest.  相似文献

Susceptibility to Cry3Bb1 toxin from Bacillus thuringiensis (Bt) was determined for western corn rootworm, Diabrotica virgifera virgifera LeConte, neonates from both laboratory and field populations collected from across the Corn Belt. Rootworm larvae were exposed to artificial diet treated with increasing Cry3Bb1 concentrations, and mortality and growth inhibition were evaluated after 4-7 d. The range of variation in Bt susceptibility indicated by growth inhibition was similar to that indicated by mortality. Although interpopulation variation in susceptibility was observed, the magnitude of the differences was comparable with the variability observed between generations of the same population. In general, the toxin was not highly toxic to larvae and estimated LC50 and EC50 values were several times higher than those reported for lepidopteran-specific Cry toxins by using similar bioassay techniques. These results suggest that the observed susceptibility differences reflect natural variation in Bt susceptibility among rootworm populations and provide a baseline for estimating potential shifts in susceptibility that might result from selection and exposure to Cry3Bb1-expressing corn hybrids.  相似文献
