Interactive effects of K+ and N (principally NH4+) on plant growth and ion uptake were investigated using hydroponically grown rice (Oryza sativa L. cv. M202) seedlings by varying the availability of NH4+ or NO3? and K+ during an 18d growth period, a 3d pretreatment period and during flux measurements. Plants grew best in media containing 100 mmol m?3 NH4+ and 200mmolm?3 K+ (N100/K200), followed by N2/K200 < N100/K2 < N2/K2. 86Rb+(K+) fluxes were increased by exposure to N during the 18 d growth period and the 3 d of pretreatment, but decreased by the presence of NH4+ during flux measurements. This inhibition was a function of prior N/K provision and the [NH4+]0 present during flux determinations. NH4+ was least inhibitory to 86Rb+(K+) influx in high-N/low-K plants. Pretreatments with K+ failed to stimulate NH4+ uptake, and the presence of K+ in the uptake solutions reduced NH4+ fluxes only in high-N/low-K plants.  相似文献

Steady state kinetics were used to examine the influence of Cd2+ both on K+ stimulation of a membrane-bound ATPase from sugar beet roots (Beta vulgaris L. cv. Monohill) and on K+(86Rb+) uptake in intact or excised beet roots. The in vitro effect of Cd2+ was studied both on a 12000–25000 g root fraction of the (Na++K++Mg2+)ATPase and on the ATPase when further purified by an aqueous polymer two-phase system. The observed data can be summarized as follows: 1) Cd2+ at high concentrations (>100 μM) inhibits the MgATPase activity in a competitive way, probably by forming a complex with ATP. 2) Cd2+ at concentrations <100 μM inhibits the specific K+ activation at both high and low affinity sites for K+. The inhibition pattern appears to be the same in the two ATPase preparations of different purity. In the presence of the substrate MgATP, and at K+ <5 mM, the inhibition by Cd2+ with respect to K+ is uncompetitive. In the presence of MgATP and K+ >10 μM, the inhibition by Cd2+ is competitive. 3) At the low concentrations of K+, Cd2+ also inhibits the 2,4-dinitrophenol(DNP)-sensitive (metabolic) K+(86Rb+) uptake uncompetitively both in excised roots and in roots of intact plants. 4) The DNP-insensitive (non metabolic) K+(86Rb+) uptake is little influenced by Cd2+. As Cd2+ inhibits the metabolic uptake of K+(86Rb+) and the K+ activation of the ATPase in the same way at low concentrations of K+, the same binding site is probably involved. Therefore, under field conditions, when the concentration of K+ is low, the presence of Cd2+ could be disadvantageous.  相似文献

Effects of interrupted K+ supply on different parameters of growth and mineral cation nutrition were evaluated for spring wheat (Triticum aestivum L. cv. Svenno). K+ (2.0 mM) was supplied to the plants during different periods in an otherwise complete nutrient solution. Shoot growth was reduced before root growth after interruption in K+ supply. Root structure was greatly affected by the length of the period in K+ -free nutrient solution. Root length was minimal, and root branching was maximal within a narrow range of K+ status of the roots. This range corresponded to cultivation for the last 1 to 3 days, of 11 in total, in K+ -free nutrient solution, or to continuous cultivation in solution containing 0.5 to 2 mM K+. In comparison, both higher and lower internal/external K+ concentrations had inhibitory effects on root branching. However, the differing root morphology probably had no significant influence on the magnitude of Ca2+, Mg2+ and Na+ uptake. Uptake of Ca2+ and especially Mg2+ significantly increased after K+ interruption, while Na+ uptake was constant in the roots and slowly increased in the shoots. The two divalent cations could replace K+ in the cells and maintain electroneutrality down to a certain minimal range of K+ concentrations. This range was significantly higher in the shoot [110 to 140 μmol (g fresh weight)?1] than in the root [20 to 30 μmol (g fresh weight)?1]. It is suggested that the critical K+ values are a measure of the minimal amount of K+ that must be present for physiological activity in the cells. At the critical levels, K+ (86Rb) influx and Ca2+ and Mg2+ concentrations were maximal. Below the critical K+ values, growth was reduced, and Ca2+ and Mg2+ could no longer substitute for K+ for electrostatic balance. In a short-term experiment, the ability of Ca2+ to compete with K+ in maintaining electroneutrality in the cells was studied in wheat seedlings with different K+ status. The results indicate that K+, which was taken up actively and fastest at the external K+ concentration used (2.0 mM), partly determines the size of Ca2+ influx.  相似文献

Net rates of NO3? and K+ uptake were compared for oilseed rape (Brassica napus L. cv. Jet neuf), perennial ryegrass (Lolium perenne L. cv. S23), Italian ryegrass (Lolium multiflorum Lam. cv. Augusta) and wheat (Triticum aestivum L. cv. Fen-man) in flowing solution culture during a 4-day sequence of low-low-high-high natural irradiance. Concentrations of NO3? (10 μM) and K+ (2.5 μM) in solutions were maintained automatically and hourly variation in net uptake of these ions was measured. During the 2 days of low irradiance (<1 MJ m?2 day?1) the uptake rates of both ions by all species were low at <1 mmol NO3?, m?2 h?1 and <0.4 mmol K+ m?2 h?1. Uptake increased in each species during the first day of high irradiance (7.90 MJ m?2 day?1) to >4 mmol NO3? m?2 h?1 and >1.4 mmol K+ m?1 h?1. These higher rates were maintained throughout the following night. The lag-time between maximum irradiance and the onset of the highest acceleration in uptake was greater for NO3? (5–8 h) than for K+ (≤1 h) in rape, wheat and Italian ryegrass. Uptake of NO3?, by perennial ryegrass showed an almost constant acceleration for 18 h following maximum irradiance. In all species the measured maximum inflows (uptake rate per unit root length) of both ions were greater than theoretical maximum potential inflows to a non-competing infinite-sink root in soil, by factors of 7 and 36, respectively, for NO3? and K+, averaged over all species.  相似文献

A microsomal (Na++ K++ Mg2+)ATPase preparation from sugar beet roots was used. The activation by simultaneous addition of Na+ and K+ at different levels was examined in terms of steady state kinetics. The observed data can be summarized in the following way: 1. The apparent affinity between the enzyme and the substrate MgATP depends on the ratio between Na+ and K+. At low Na+ concentration (below 5 mM), the apparent Km decreases with increasing concentrations of K+ (1–20 mM). At 5 mM Na+, the K+ level does not change the apparent Km, while at Na+ levels above 10 mM, the apparent Km between enzyme and substrate increases with increasing concentration of K+. 2. When the MgATP concentration is kept constant, homotropic cooperativity (concerning one type of ligand) and heterotropic cooperativity (concerning different types of ligands) exist in the activation by Na+ and K+. The Na+ binding is cooperative with different Km values and Hill coefficients (n) in the presence of low and high concentration of K+. At low Na+ level (< 5 mM). a negative cooperativity exists for Na+ (nNa < 1) which is more pronounced in the presence of high [K+]. When the concentration of Na+ is raised the negative cooperativity disappears and turns into a positive one (nNa > 1). Only K+ binding in the presence of low [Na+] shows cooperativity with a Hill coefficient that reflects changes from negative to positive homotropic cooperativity with increasing concentrations of K+ (nK < 1 → nK > 1). In the presence of [Na+] > 10 mM, the changes in nk are insignificant. 3. A model is proposed in which one or two different K sites and one or two Na sites control the catalytic activity, with multiple interactions between Na+, K+ and MgATP. 4. In the presence of Na+ (< 10 mM), K+ is probably bound to two K sites, one of which translocates K+ through the membrane by an antiport Na+/K+ mechanism. This could be connected with an elevated K+ uptake in the presence of Na+ and could therefore explain some field properties of sugar beets.  相似文献

Tomato growth was examined in solution culture under constant pH and low levels of NH4+ or NO3?. There were five nitrogen treatments: 20 mmoles m?3 NH4+, 50 mmoles m?3 NO3?, 100 mmoles m?3 NH4+ 200 mmoles m?3 NO3?, and 20 mmoles m?3 NH4++ 50 mmoles m?3 NO3?. The lower concentrations (20 mmoles m?3 NH4+ and 50 mmoles m?3 NO3?) were near the apparent Km for net NH4+ and NO3? uptake; the higher concentrations (100 mmoles m?3 NH4+ and 200 mmoles m?3 NO3?) were near levels at which the net uptake of NH4+ or NO3? saturate. Although organic nitrogen contents for the higher NO3? and the NH4++ NO3? treatments were 22.2–30.3% greater than those for the lower NO3? treatment, relative growth rates were initially only 10–15% faster. After 24 d, relative growth rates were similar among those treatments. These results indicate that growth may be only slightly nitrogen limited when NH4+ or NO3? concentrations are held constant over the root surface at near the apparent Km concentration. Relative growth rates for the two NH4+ treatments were much higher than have been previously reported for tomatoes growing with NH4+ as the sole nitrogen source. Initial growth rates under NH4+ nutrition did not differ significantly (P≥ 0.05) from those under NO3? or under combined NH4++ NO3?. Growth rates slowed after 10–15 d for the NH4+ treatments, whereas they remained more constant for the NO3? and mixed NH4++ NO3? treatments over the entire observation period of 24–33 d. The decline in growth rate under NH4+ nutrition may have resulted from a reduction in Ca2+, K+, and/or Mg2+ absorption.  相似文献

Ammonium (NH4+) is a central intermediate in the N metabolism of plants, but the quantitative importance of NH4+ in transporting N from root to shoot and the capability of plants to store NH4+ in leaves are still matters of substantial controversy. This paper shows that some of these controversies have to be related to the use of inadequate analytical procedures used for extraction and quantification of NH4+ in plants. The most frequently used methods for determination of NH4+, viz. colorimetric methods based on the classical Berthelot reaction, suffered severely from interference caused by amino acids, amines, amides and proteins. For some of these metabolites the interference was positive, while for others it was negative, making correction impossible. Consequently, colorimetric analysis is inapplicable for determination of NH4+ in plants. Results obtained by ion chromatography may overestimate the NH4+ concentration due to co‐elution of NH4+ with amines like methylamine, ethylamine, ethanolamine and the non‐protein amino acid Γ‐aminobutyric acid. Derivatization of NH4+ with o‐phthalaldehyde at alkaline pH and subsequent quantification of NH4+ by fluorescence spectroscopy was also associated with interference. However, when pH was lowered to 6.8 during derivatization and 2‐mercaptoethanol was used as reductant, NH4+ could be determined with a high selectivity and sensitivity down to a detection limit of 3.3 μM in a 10‐μl sample volume. Derivatization was performed on‐line using a column‐less HPLC system, enabling rapid quantification of NH4+ in a few minutes. Flow injection analysis with on‐line gas dialysis was, likewise, free from interference, except when applied on highly senescent plant material containing volatile amines. Labile N metabolites in leaf tissue extract, xylem sap and apoplastic fluid were degraded to NH4+ during extraction and subsequent instrumental analysis if the samples were not stabilised. A simple and efficient stabilisation could be obtained by addition of 10 mM ice‐cold HCOOH to the plant extraction medium or to the samples of apoplastic fluid or xylem sap. We conclude that significant concentrations of NH4+, exceeding 1 mM, may occur in xylem sap, leaf apoplastic fluid and leaf tissue water of nitrate‐grown tomato and oilseed rape plants. The measured NH4+ concentrations were not a result of excessive N supplies, as even plants grown under mildly N‐deficient conditions contained NH4+.  相似文献

Long-term effects of 1-naphtaleneacetic acid (NAA), benzyladenine (BA), gibberellic acid (GA3), abscisic acid (ABA) and ethylene on K+ levels, K+ uptake and translocation to the shoot were studied in young wheat plants (Triticum aesticum L. cv. Martonvásári-8) grown at different K+ supplies. Na+ levels and K+/Na+ selectivity were also investigated. Both in shoots and roots, NAA, BA and ABA decreased K+ and Na+ levels more effectively in high-K+ plants than in low-K+ plants. GA, and ethylene did not influence K+ and Na+ levels. K+/Na+ selectivity in roots of low-K+ plants was increased in favour of K+ by BA, NAA and to a lesser extent by ABA. In high-K+ plants only BA increased the K+/Na+ ratio, whereas the effects of the other hormones were the opposite (NAA) or less pronounced (ABA). K+(86Rb) uptake was inhibited by NAA and BA in low-K+ plants but not in high-K+ plants. K+(86Rb) uptake was inhibited throughout by 10 μM ABA. K+(86Rb) translocation to the shoot was influenced by the hormones similarly to the uptake patterns, with the exception of ABA, which inhibited translocation in low-K+ plants but not in high-K+ plants. The results show that hormonal effects may quantitatively and qualitatively be modified by K+ levels in the plant and that internal K+ concentration may play a role in the mechanisms regulating the effects of NAA, BA and ABA but probably not in those of GA3 or ethylene.  相似文献

Abstract Fusicoccin (FC)-stimulated K+ (86Rb) uptake and proton extrusion of maize (Zea mays) root apical segments were inhibited by pretreatment of 4-day-old seedlings with the herbicide Chlorsulfuron. In the range of Chlorsulfuron concentrations 0.01-10 mmol m?3, the percentage of inhibition was 15% at 0.01 mmol m?3 and progressively increased with Chlorsulfuron concentration up to 60% at 10 mmol m?3. At the maximum concentration tested (10 mmol m?3), the inhibition was evident after 1.5 h of pre-treatment. The binding of FC to microsomal fractions of root segments from Chlorsulfuron-pretreated seedlings was inhibited by 30%. It is suggested that Chlorsulfuron causes an alteration at the plasmalemma level involving the FC binding sites. The ineffectiveness of Chlorsulfuron in inhibiting FC-stimulaled K+ uptake when administered to excised segments, while inhibiting the enzyme acetolactate synthase, pointed out by Ray (1984) as the site of action of Chlorsulfuron in pea plants, suggests that the observed inhibition of K+ uptake and H+ extrusion is not induced by Chlorsulfuron inhibition of this enzyme. An alternative site of action of Chlorsulfuron is hypothesized in maize plants.  相似文献

The influx of Rb+ into the roots of two barley varieties (Hordeum vulgare L. cv. Salve and cv. Ingrid) from a K+-free 86Rb-labelled nutrient solution with 2.0 mM Rb+, was checked at intervals from day 6 to day 18. The control plants were continuously grown in complete nutrient solution containing 5.0 mM K+, while two other groups of plants were grown in K+-free nutrient solution starting on day 6 and between day 6 and day 9, respectively. The pattern of Rb+ influx was similar for both varieties, although their efficiencies in absorbing Rb+ were different. The relationship between Rb+ influx and K+ concentration of the root could be interpreted in terms of negative feedback through allosteric control of uptake across the plasmalemma of the root cells. Hill plots were bimodal, but in the opposite direction. The Hill coefficients, reflecting the minimum number of interacting allosteric binding sites for K+ (Rb+), were low (≤–3.0). It is discussed whether the threshold value, that is the breaking point in the Hill plot, is indicative of a changed efficiency of transporting units for K+ (Rb+) transport to the xylem. Moreover, feedback regulation might be involved in transport of K+ between root and shoot. The variation in K+ concentrations in the roots and shoots of control plants were cyclic but in phase opposition despite an exponential growth. In isolated Elodea densa leaves, the relationships between H+ extrusion (-ΔH+), K+ fluxes and membrane potential (Em) were investigated for two different conditions of activation of the ATP-dependent H+ pump. The ‘basal condition’ (darkness, no pump activator present) was characterized by low values of-ΔH+ and K+ uptake (ΔK+), wide variability of the ?ΔH+/ΔK+ ratio, relatively low membrane polarization and Em values more positive than EK for external K+ concentrations (|K+]o of up to 2mol m?3. A net K+ uptake was seen already at [K+]o below 1 mol m?3, suggesting that K+ influx in this condition was a thermodynamically uphill process involving an active mechanism. When the H+ pump was stimulated by fusicoccin (FC), by cytosol acidification, or by light (the ‘high polarization condition’), K+ influx largely dominated K+ and C? efflux, and the ?ΔH+/ΔK+ ratio approached unity. In the range 50 mmol m?3?5 mol m?3 [K+]0, Em was consistently more negative than EK. The curve of K+ influx at [K+]0 ranging from 50 to 5000mmol m?3 fitted a monophasic, hyperbolic curve, with an apparent half saturation value = 0–2 mol m?3. Increasing |K+]0 progressively depolarized Em, counteracting the strong hyperpolarizing effect of FC. The effects of K+ in depolarizing Em were well correlated with the effects on both K+ influx and ?ΔH+, suggesting a cause-effect chain: K+0 influx → depolarization → activation of H+ extrusion. Cs+ competitively inhibited K+ influx much more strongly in the ‘high polarization’ than in the ‘basal’ condition (50% inhibition at [Cs+]/[K+]0 ratios of 1:14 and 1:2, respectively) thus confirming the involvement of different K+ uptake systems in the two conditions. These results suggest that in E. densa leaves two distinct modes of interactions rule the relationships between H+ pump, membrane polarization and K+ transport. The nitrogen requirement of plants is predominantly supplied by NH4+ and/or NO3? from the soil solution, but the energetic cost of uptake and assimilation is generally higher for NO3? than for NH4+. We found that CO2 enrichment of the atmosphere enhanced the root uptake capacity for NO3?, but not for NH4+, in field-grown loblolly pine saplings. Increased preference for NO3? at the elevated CO2 concentration was accompanied by increased carbohydrate levels in roots. The results have important implications for the potential consequences of global climate change on plant-and ecosystem-level processes in many temperate forest ecosystems.  相似文献

Klotz, M. G. and Erdei, L. 1988. Effect of tentoxin on K+ transport in winter wheat seedlings of different K+-status. The influence of the phytoeffective mycotoxin, tentoxin, [cyclo-(L-leucyl-N-methyltrans-dehydronhenyl-alanyl-glycyl-N-methyl-L-alanyl)] (in K+ uptake and on translocation of K+ from roots to shoot was studied in 14-day-old winter wheat plants (Triticum aestivum L. cv. Martonvásári-8) grown at different levels of K+ supply. For comparison, the effects of 2,4-dinilrophcnol and valinomycin were also investigated. In I-h experiments I pM tentoxin reduced K+ influx in the routs over the external K+ concentration range 0.1 to 1 mM (low-K+ plants), whereas stimulation was observed al lower and higher K+ concentrations. On the other hand, in plants grown at 0.3 mM K+, tentoxin stimulated the translocation of K+ from roots to shoots in 5-h experiments. Valinomycin affected K+ transport only al high K+-status (slight stimulation). In low-K+ plants 2,4-dinitrophenol (DNP) caused drastic inhibition of K+ uptake, but in high-K+ plants uptake was only slightly inhibited and translocation slightly stimulated, It is concluded that the opposite effects of tentoxin on K+ uptake and translocation agree1 with the directions of the H+-ATPases pumping H+ towards the apoplast and located at the cortex plasmalemma and the xylem parenchyma plasma-membrane, respectively. These effects should probably be attributed to the interaction between tentoxin and the K+-carrier protein rather than to a direct influence of tentoxin on H+-ATPase.  相似文献

Kinetic parameters for NH4+ and NO3? uptake were measured in intact roots of Lolium perenne and actively N2-fixing Trifolium repens. Simultaneously, net H+ fluxes between the roots and the root medium were recorded, as were the net photosynthetic rate and transpiration of the leaves. A Michaelis–Menten-type high-affinity system operated in the concentration range up to about 500 mmol m?3 NO3? or NH4+. In L. perenne, the Vmax of this system was 9–11 and 13–14 μmol g?1 root FW h?1 for NO3? and NH4+, respectively. The corresponding values in T. repens were 5–7 and 2 μmol g?1 root FW h?1. The Km for NH4+ uptake was much lower in L. perenne than in T. repens (c. 40 compared with 170 mmol m?3), while Km values for NO3? absorption were roughly similar (around 130 mmol m?3) in the two species. There were no indications of a significant efflux component in the net uptake of the two ions. The translocation rate to the shoots of nitrogen derived from absorbed NO3?-N was higher in T. repens than in L. perenne, while the opposite was the case for nitrogen absorbed as NH4+. Trifolium repens had higher rates of transpiration and net photosynthesis than L. perenne. Measurements of net H+ fluxes between roots and nutrient solution showed that L. perenne absorbing NO3? had a net uptake of H+, while L. perenne with access to NH4+ and T. repens, with access to NO3? or NH4+, in all cases acidified the nutrient solution. Within the individual combinations of plant species and inorganic N form, the net H+ fluxes varied only a little with external N concentration and, hence, with the absorption rate of inorganic N. Based on assessment of the net H+ fluxes in T. repens, nitrogen absorption rate via N2 fixation was similar to that of inorganic N and was not down-regulated by exposure to inorganic N for 2 h. It is concluded that L. perenne will have a competitive advantage over T. repens with respect to inorganic N acquisition.  相似文献

Abstract: With a partially purified, membrane-bound (Ca + Mg)-activated ATPase preparation from rat brain, the K0.5 for activation by Ca2+ was 0.8 p μm in the presence of 3 mm -ATP, 6 mm -MgCl2, 100 mM-KCI, and a calcium EGTA buffer system. Optimal ATPase activity under these circumstances was with 6-100 μm -Ca2+, but marked inhibition occurred at higher concentrations. Free Mg2+ increased ATPase activity, with an estimated K0.5, in the presence of 100 μm -CaCl2, of 2.5 mm ; raising the MgCl2 concentration diminished the inhibition due to millimolar concentrations of CaCl2, but antagonized activation by submicromolar concentrations of Ca2+. Dimethylsulfoxide (10%, v/v) had no effect on the K0.5 for activation by Ca2+, but decreased activation by free Mg2+ and increased the inhibition by millimolar CaCl2. The monovalent cations K+, Na+, and TI+ stimulated ATPase activity; for K+ the K0.5 was 8 mm , which was increased to 15 mm in the presence of dimethylsulfoxide. KCI did not affect the apparent affinity for Ca2+ as either activator or inhibitor. The preparation can be phosphorylated at 0°C by [γ-32P]-ATP; on subsequent addition of a large excess of unlabeled ATP the calcium dependent level of phosphorylation declined, with a first-order rate constant of 0.12 s?1. Adding 10 mm -KCI with the unlabeled ATP increased the rate constant to 0.20 s?1, whereas adding 10 mm -NaCl did not affect it measurably. On the other hand, adding dimethyl-sulfoxide slowed the rate of loss, the constant decreasing to 0.06 s?1. Orthovanadate was a potent inhibitor of this enzyme, and inhibition with 1 μm -vanadate was increased by both KCI and dimethylsulfoxide. Properties of the enzyme are thus reminiscent of the plasma membrane (Na + K)-ATPase and the sarcoplasmic reticulum (Ca + Mg)-ATPase, most notably in the K+ stimulation of both dephosphorylation and inhibition by vanadate.  相似文献

In order to identify physiological components that contribute to salinity tolerance, we compared the effects of Na+, Mg2+ and K+ salts (NaCl, Na2SO4, MgCl2, MgSO4, KCl and K2SO4), Ca2+ (CaSO4), mannitol and melibiose on the wild type and the single-gene NaCl-tolerant mutants stl1 and stl2 of Ceratopteris richardii. Compared with gametophytic growth of the wild type, stl2 showed a low level of tolerance that was restricted to Na+ salts and osmotic stress. stl2 exhibited high tolerance to both Na+ and Mg2+ salts, as well as to osmotic stress. In response to short-term exposure (3 d) to NaCl, accumulation of K+ and Na+ was similar in the wild type and stl1. In contrast, stl2 accumulated higher levels of K+ and lower levels of Na+. Ca2+ supplementation (1.0 mol m?3) ameliorated growth inhibition by Na+ and Mg2+ stress in wild type and stll, but not in stl2. In addition, under Na+ stress (175 mol m?3) wild-type, stll and stl2 gametopbytes maintained higher tissue levels of K+ and lower levels of Na+ when supplemented with Ca2+ (1.0 mol m?3). stl2 gametophytes were extremely sensitive to K+ supplementation. Growth of stl2 was greater than or equal to that of the wild type at trace concentrations of K+ but decreased substantially with increasing K+ concentration. Supplementation with K+ from 0 to 1.85 mol m?3 alleviated some of the inhibition by 75 mol m?3 NaCl in the wild type and in stl1. In stl2, growth at 75 mol m?3 NaCl was similar at 0 and 1.85 mol m?3 K+ supplementation. The effects of an auxin herbicide, 2,4-D, at a concentration of 0.01 mM, on the K+ uptake and efflux of excised roots of wheat (Triticum aestivum L. cv. Rannaya) were investigated at different pH values. The K+ movement was monitored with a K+ (86Rb) tracer. In parallel experiments the ATPase activities of microsomal fractions were determined by the inorganic phosphate liberation method. 2,4-D inhibited the K+ uptake especially at low pH, irrespective of whether Ca2+ was present or not. No marked changes were observed in the K+ efflux properties at pH values above 4. The inhibitory effect on K+ uptake exhibited a correlation with the hydrocarbon solubility of the herbicide, but not with the 2,4-D-induced decrease of the ATPase activity. It is suggested that 2,4-D exerts a non-specific effect on the lipid-protein interactions, giving rise to a generalized alteration of the transport barrier properties of the plasma membrane even at as low a concentration as 0.01 mM.  相似文献

Abstract: Rat brain microsomes accumulate Ca2+ at the expense of ATP hydrolysis. The rate of transport is not modulated by the monovalent cations K+, Na+, or Li+. Both the Ca2+ uptake and the Ca2+-dependent ATPase activity of microsomes are inhibited by the sulfated polysaccharides heparin, fucosylated chondroitin sulfate, and dextran sulfate. Half-maximal inhibition is observed with sulfated polysaccharide concentrations ranging from 0.5 to 8.0 µg/ml. The inhibition is antagonized by KCl and NaCl but not by LiCl. As a result, Ca2+ transport by the native vesicles, which in the absence of polysaccharides is not modulated by monovalent cations, becomes highly sensitive to these ions. Trifluoperazine has a dual effect on the Ca2+ pump of brain microsomes. At low concentrations (20–80 µM) it stimulates the rate of Ca2+ influx, and at concentrations >100 µM it inhibits both the Ca2+ uptake and the ATPase activity. The activation observed at low trifluoperazine concentrations is specific for the brain Ca2+-ATPase; for the Ca2+-ATPases found in blood platelets and in the sarcoplasmic reticulum of skeletal muscle, trifluoperazine causes only a concentration-dependent inhibition of Ca2+ uptake. Passive Ca2+ efflux from brain microsomes preloaded with Ca2+ is increased by trifluoperazine (50–150 µM), and this effect is potentiated by heparin (10 µg/ml), even in the presence of KCl. It is proposed that the Ca2+-ATPase isoform from brain microsomes is modulated differently by polysaccharides and trifluoperazine when compared with skeletal muscle and platelet isoforms.  相似文献

In the present study, we investigated whether growth and main nutrient ion concentrations of cabbage (Brassica campestris L.) could be increased when plants were subjected to different NH4^+/NO3- ratios. Cabbage seedlings were grown in a greenhouse in nutrient solutions with five NH4^+/NO3- ratios (1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75; and 0:1). The results showed that cabbage growth was reduced by 87% when the proportion of NH4^+-N in the nutrient solution was more than 75% compared with a ratio NH4^+/NO3- of 0.5:0.5 35 d after transplanting, suggesting a possible toxicity due to the accumulation of a large amount of free ammonia in the leaves. When the NH4+/NO3- ratio was 0.5:0.5, fresh seedling weight, root length, and H2PO4- (P), K^+, Ca^2+, and Mg^2+ concentrations were all higher than those in plants grown under other NH4^+/NO3- ratios. The nitrate concentration in the leaves was the lowest in plants grown at 0.5: 0.5 NH4^+/NO3-. The present results indicate that an appropriate NH4^+/NO3- ratio improves the absorption of other nutrients and maintains a suitable proportion of N assimilation and storage that should benefit plant growth and the quality of cabbage as a vegetable.  相似文献

Abstract Changes in the net uptake rate of K+ and in the average tissue concentration of K+ were measured over 14 d in response to changes in root temperature with oilseed rape (Brassica napus L. cv. Bien venu) and barley (Hordeum vulgare L. cv. Atem). Plants were grown in flowing nutrient solutions containing 2.5 mmol m?3 K+ and were acclimatized over 49 d (rape) or 28 d (barley) to low root temperature (5°C) prior to steady–state treatments at root temperatures between 3 °C and 25 °C, with common air temperature. Uptake of K+ was monitored continuously over 14 d and nitrogen was supplied as NH4++ NO?3 or NH+4 or NO?3. Unit absorption rates of K+ increased with time and with root temperature up to Day 4 or 5 following the change in root temperature. Thereafter they usually approached steady-state, with Q10? 2.0 between 7 °C and 17°C, although rates became similar between 7 °C and 13°C. Uptake of K+ by rape plants was invariably greater under NO?3 nutrition compared with NH+4. The percentage K+ in the plant dry matter increased with temperature from 2% at 3 °C to 4% at 25 °C in rape, but there was less effect of temperature on the average concentrations of K+ in the plant fresh weight or plant water content. Concentrations of K+ in the leaf water fraction of rape plants decreased with increasing root temperature, but in barley they increased with increasing root temperature. Concentrations of K+ in the root water fraction were relatively stable with respect to root temperature. The results are discussed in terms of compensatory changes in K+ uptake following a change in root temperature and the relationships between growth, shoot: root ratio and K+ composition of the plant.  相似文献
