Spawning of coal grunter Hephaestus carbo was successfully induced using doses of human chorionic gonadotrophin (hCG) between 500 and 3000 IU kg (body weight)−1. Water hardened eggs are telolecithal, amber in colour, spherical, transparent, demersal and slightly adhesive with a single large oil droplet and perivitelline space 47% of total egg volume. Cleavage begins 10–15 min after fertilization. Epiboly begins 6h after fertilization and continues for 4 h. Invagination of the neural tube is apparent 11·5 h after fertilization, followed by progressive organogenesis up to hatching 60–80 h after fertilization. An invagination in the yolk, consistent in shape, position and time of appearance among embryos spawned from numerous brood stock pairs, was visible in all fertilized eggs between neurulation (11-5 h) and early organogenesis (20 h). The functional significance of this yolk invagination is unknown. Newly-hatched larvae (4·2 mm L T) are elongate and possess well developed eyes, a functional mouth, and a large yolk sac. Yolk is fully resorbed and first feeding occurs at 6 days posthatching. The sequence of fin formation is caudal, second dorsal and anal, first dorsal, pectoral and pelvic. The prefiexion larval stage lasts for c. 8 days and flexion of the notochord is complete within a further 8–9 days. Squamation commences at 30 days posthatching and transition to the juvenile life stage is complete by 35–40 days posthatching.  相似文献

Regions of the gut of near-term embryos and new-born larvae of the viviparous rockfish Sebastes taczanowskii were examined histologically and immunohistochemically in order to ascertain whether they possessed the potential to absorb intact macromolecules. Compared with the hindgut of larvae 24 h after parturition, that of near-term embryos (stage 31) was found to be markedly hypertrophied and to react positively when tested for the presence of eosinophilic materials and with antibodies raised against extracts of the rockfish yolk (ab.a-E). In addition, near-term embryos incubated in Ringer solution containing 1% bovine serum albumin appeared to accumulate the protein within the epithelium of the hindgut. Furthermore, as confirmed by immunodiffusion analysis, the ovarian fluid contained materials which reacted positively to ab.a-E. These results suggest that ingestion of intact macromolecules occurs during the late embryonic stages and that the yolk-like substances detected within the hindgut epithelium of such embryos may be derived from the bathing ovarian fluid.  相似文献

对乌骨鸡胚胎发育过程中卵内各组成成分(蛋清、蛋黄、胚胎)在不同胚龄(即入孵的第0、3、6、9、12、15、18d和出壳)时蛋白质、脂肪含量变化进行测定与分析.结果表明:在不同胚龄时,蛋清中蛋白质含量显著高于蛋黄,蛋黄中脂肪含量始终高于蛋清;而胚胎中的蛋白质、脂肪逐渐增加。另外,胚胎发育过程中胚胎蛋白质的主要来源是蛋黄中的蛋白质,说明蛋黄是禽类胚胎发育过程中营养需要的主要来源。  相似文献

Egg size for Fundulus heteroclitus (L.) populations is concordant with the distribution of the two F . heteroclitus subspecies, i.e. F. h. heteroclitus eggs are considerably larger than F. h. macrolepidotus eggs. The influence of egg size on survival of embryos during incubation and survival and growth of newly-hatched larvae was estimated for four populations representing both subspecies along the Atlantic coast of the United States and in Delaware Bay. Survival of embryos was determined for incubation periods of 14, 21 and 28 days. Greatest differences in survival were detected following the longest incubation period where less than 50 per cent of the smaller F. h. macrolepidotus eggs survived while little or no mortality was detected among the larger F. h. heteroclitus eggs . Influence of egg size on larval survival was also greatest among those larvae hatched after 28 days where F. h. macrolepidotus larvae survived without food, for an average of 6 days, while F. h. heteroclitus larvae survived 11–12 days. F. h. heteroclitus larvae were significantly larger at hatching than F. h. macrolepidotus larvae. Larval growth rates were the same (0.4 mm day−1) in both subspecies. As a result, size differences at hatching were still maintained after 42 days of growth. The differences in egg size along with other morphological and reproductive characteristics of F. heteroclitus populations probably represent genetically based adaptations to environmental conditions, of which the length of the spawning season is one of the major components stimulating the coevolution of these traits.  相似文献

Embryonic development and larval morphology of Chromis crusma was described from five nests sampled between 21 and 25 m depth in central Chile (33°S). From each nest, a set of c. 100 randomly selected eggs were hand-collected and transported in seawater to the laboratory. Subsets of c. 30 eggs per nest were maintained in 50 ml glass containers at a constant ambient temperature of c. 12°C (range 11.5–12.9°C). Egg length (L) and width (W) and larval notochordal length (LN) were measured from photographs. Geometric morphometric analyses were performed in newly hatched and 1 week old larvae to quantify shape changes. Ellipsoid eggs had an average (mean ± SE) size of 1.12 ± 0.05 mm L and 0.67 ± 0.02 mm W, with volume being similar throughout 15 developmental stages (i.e., ellipsoid-shaped; 0.27 mm3). Planktonic larvae hatched between 5 and 11 days at 12°C and had a mean LN of 3.13 ± 0.25 mm, a yolk sack volume of 0.03 mm3 and an oil droplet volume of 0.005 mm3. Morphological traits at hatching included: (a) lack of paired fins and jaws; (b) single medial fin fold; (c) lack of eye pigmentation; (d) yolk sac present near anterior tip; (e) melanophores distributed along ventral surface with one pair over the forehead. In order to generate an up-to-date summary of developmental traits within Pomacentridae, we reviewed literature on egg development (e.g., shape and number of oil droplets), hatching and larval traits (e.g., morphology, pigmentation patterns). Thirty-two publications accounting for 35 species were selected, where eggs, embryonic development, hatching and larval traits were found for 26, 21, 24 and 34 species, respectively. In order to evaluate potential phylogenetic and environmental relationships within the early stages of Pomacentridae, cluster analyses (Bray Curtis similarity, group average) were also performed on egg and larval traits of 22 species divided by subfamily (Stegastinae, Chrominae, Abudefdufinae, Pomacentrinae) and thermal ranges (i.e., low: 16.5°C (range: 12–21°C), medium: 24.5°C (range:21–28°C) and high: 27°C (range: 26–28°C)), suggesting that early developmental patterns can be segregated by both temperature and phylogenetic relationships.  相似文献

A series of studies examined the influence and temporal interaction of energy substrate, media complexity, and tissue co-culture on the development of in vitro fertilized cat embryos and the persistence of the morula-to-blastocyst developmental block. In Study I, oocytes were fertilized and cultured for 144 hr in a simple culture medium (modified Krebs Ringer bicarbonate; mKrb), containing either glucose or glutamine, or cultured in mKrb w/ glutamine for the initial 72 hr with transfer to mKrb w/ glucose for the final 72 hr. Fertilization rate, percent development to morulae, and cell number per embryo were similar (P > 0.05) between treatments and blastocyst formation was universally low (<10%). In Study II, oocytes were fertilized and cultured in either mKrb (w/ glucose or glutamine) or in a complex medium, Ham's F10 (w/ 10% fetal bovine serum [FBS]). After 72 hr of initial culture, embryos in mKrb were transferred into Ham's F10. Fertilization rate was lower (P < 0.01) in Ham's F10 but embryo development to the morulae stage and cell number per embryo were comparable (P > 0.05) for all treatments. A higher percentage of blastocysts and morulae becoming blastocysts were observed after initial culture in mKrb w/ glutamine than after initial culture in mKrb w/ glucose. In Study III, oocytes were fertilized and cultured initially in mKrb (w/ glutamine), and then switched to either Ham's F10 or cat oviductal cell monolayers (in Ham's F10). Additional embryos were cultured exclusively in Ham's F10 or on cat oviductal cell monolayers. Fertilization rates were lower (P < 0.05) on oviductal cells but cell number per embryo was similar (P > 0.05) in all treatments. Blastocyst formation was lower (P < 0.05) on oviductal cells than in mKrb-Ham's F10 treatment and was <20% in all treatments. In summary, while in vitro fertilization-derived cat embryos develop to morulae under a variety of culture conditions, the morula-to-blastocyst developmental block was minimally responsive to alterations in energy substrate and medium complexity or fluctuations in their temporal availability. In addition, oviductal cell culture, alone or in combination with other culture variations, was ineffective in overcoming the developmental block. © 1996 Wiley-Liss, Inc.  相似文献

Comparison of the results of two methods for species recognition, an egg identification key and rearing of larvae, showed only small qualitative differences in the identification of fish species present in a stretch of the River Elbe, Germany. This study investigated the effect of strong magnetic fields as insecticidal activity on Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae) larvae and eggs at different stages of development and their preference by the egg parasitoid, Trichogramma embryophagum Hartig (Hymenoptera: Trichogrammatidae). Eggs ranging in age from 24-h to 48-h and 72-h-old and larvae (1 to 2 days) were exposed to 1.4 Tesla (T) magnetic fields from a DC power supply at 50 Hz for different time periods (3, 6, 12, 24, 48 and 72 h). Twelve hours of exposure at 1.4 T was toxic to 24-h-old eggs of E. kuehniella. The 72-h-old host eggs treated with 1.4 T for 6–72 h were not significantly preferred by T. embryophagum. The magnetic field was toxic to 24-h-old eggs of E. kuehniella exposed to 1.4 T for 12. The treatment of magnetic fields on the 72-h-old host egg with 1.4 T at 6–72 h was not significantly preferred by T. embryophagum. Magnetization of 24-h-old eggs of E. kuehniella for 3 h could be effectively used with T. embryophagum as sterilised host eggs. These eggs were markedly preferred by T. embryophagum. The LT50 and LT99 values of magnetic fields at different egg stages of E. kuehniella, and larvae were measured. A level of 1.4 T at 72 h completely prevented the development of the larvae. There was no significant effect on larval survival at 1.4 T at 48 and 72 h. Increasing magnetic fields exposure times for eggs that were 24-h, 48-h and 72-h-old prevented larval emergence and increased their mortality rate. Consequently, magnetic fields could be used in controlling stored-product pest eggs and larvae of E. kuehniella.  相似文献

Ultrastructural and histological changes in the embryonic and larval surface during ontogenesis of the endangered golden mahseer Tor putitora is studied here for the first time. Embryonic development was completed 91–92 h after fertilization at an ambient temperature of 23° ± 1° C (mean ± s.d. ). The gastrula stage was characterized by presence of the Kupffer's vesicle, notochord, ectoderm and endoderm cells. Primordial germ cells were clearly identifiable from c. 55 h post‐fertilization at the organogenesis stage. Mean total length of newly hatched larvae was 7·0 ± 0·5 mm. Scanning electron microscopy of newly hatched larvae demonstrated vitelline arteries, microridged epithelial cells and mucous gland openings over much of the body surface. Eye, oral cavity, pharyngeal arches, heart, intestinal loop, prosencephalon, cephalic vesicle and nasal epithelium were clearly distinguished in 3 day old hatched individuals. In 6 day old individuals, caudal‐fin rays and internal organs were evident. The dorsal fin became prominent at this stage and larvae began swimming at the surface. The reserved yolk material was totally absorbed 8–11 days after hatching and larvae began feeding exogenously. Tor putitora exhibited a longer early developmental period than other cyprinids reared at similar temperatures.  相似文献

The production of heat in the fertilized eggs, during the early development of the Japanese pond frog, Rana brevipoda porosa, was measured using a pyroelectric detector constructed with a polyvinylidene fluoride film. One and 2 μW of heat production was detected during the cleavage periods in the embryogenesis of the 2-cell embryo and the 4-cell embryo, respectively. The heat production increased stepwise with the cleavage in the embryogenesis of each stage. In comparison with the heat production during the inter-cleavage periods, a larger amount was noted at the cleavage periods in the two embryonic stages.  相似文献

Temperature development relationships were determined for batches of Irish Sea cod Gadus morhua eggs incubated in flow-through incubators. Hatching began 16·4 days after fertilization (DAF) at 6° C, 10·3 DAF at 8° C, 9·4 DAF at 10° C and 7·4 DAF at 12° C. Egg mortality increased at the higher temperatures, but survival was >80%. Results were compared with published data at four comparable stage end points: the end of blastula, the end of gastrula, the point of growth of the embryo completely surrounding the yolk and the point when 50% of the eggs were hatched. All the studies showed a curvilinear relationship between age at stage and temperature. There was a 12 day inter-study difference in time to 50% hatch at 2° C and 4 day difference at 10° C. There were no consistent trends that differentiated eastern v. western, or northern v. southern populations. A single model for cod egg incubation time from fertilization to 50% hatch was derived based on data from six cod populations, but it is recommended that individual stock relationships should be used where possible.  相似文献

The duration of the egg stage at constant temperature showed a decrease with increasing constant temperature up to 30°; above this, there was a slight increase in the duration and a decrease in survival. Regularly changing temperature had a slight but significant retarding effect on egg duration in one experiment but in the second, there was no significant effect. In two experiments in which there were two sudden changes of temperature in the same direction, there was no significant effect. The duration of the larval stages from hatching up to the third stage showed a decline with increasing constant temperature up to 25°. Survival was poor at 30°. The larvae were fed on E. coli cultures. In the regularly changing experiments on the larvae one showed a slight retardation and the second an acceleration in development. Both were significant (P < .05). In the experiments in which temperature changed in two sudden jumps in the same direction, one showed a slight acceleration and the other showed no effect. It is concluded that the effects of changing temperature while significant are probably not large enought to be of significance in nature. Newly hatched and third stage larvae grown at constant temperatures showed a tendency to be longer at higher temperatures.  相似文献

Niveoscincus metallicus is a small viviparous skink which provides a substantial amount of yolk to each of its developing embryos although some organic nutrients are also transferred across the placentae. The total amount of lipid present in the yolk of N. metallicus (37% of dry weight) is very much higher than that in the newborns (19% of dry weight), confirming that the yolk is utilised as an energy source during gestation. Triacylglycerols (TAG), which are storage compounds, are the major lipid resource available to the embryos and are present in relatively large amounts in the yolk of N. metallicus. Polar lipids (PL), which form the structural components of membranes, and sterols (ST), which are involved in the synthesis of hormones and vitamins, are also present in the yolk. The proportions of each of these lipid classes differs markedly between yolks and newborns. This may reflect variations in the role played by each lipid class in the provision of nutrients to, and development of, embryos.  相似文献

In cattle, oocytes obtained from follicles smaller than 3 mm in diameter can undergo maturation in vitro, progressing to MII and undergoing fertilization, but are developmentally incompetent. Cytoplasts were prepared from in vitro matured oocytes aspirated from small (1-3 mm) or large (6-12 mm) follicles and fused to serum starved mural granulosa cells. Following activation, reconstructed embryos were cultured for 7 days and classified G1 to G4, before being processed for nuclei counting or transferred to synchronized recipients. Oocytes from small follicles had lower rates of polar body extrusion (59.6 vs. 69%; 731/1230 vs. 608/857) and fusion (71.4 vs. 78.8%; 360/497 vs. 364/465; P < 0.06). There were no differences in total rate of blastocysts development (60 vs. 59.8%; small vs. large), or any grade classification. A significant interaction was detected between follicle size and embryo grade with G3 embryos from small follicles having a greater cell number. Developmental competence of G1 and G2 embryos did not differ at day 27 (48 vs. 46%; 16/33 vs. 17/37; small vs. large). Although there were no differences in fetal size between the two groups, differences in allantois length (53 vs. 86 mm; small vs. large; P < 0.002) and allantois width (9.5 vs. 13 mm; small vs. large; P < 0.06) were seen. No differences in survival to term (2/13 in each group) were observed. These results indicate that cytoplasts from follicles of 1-3 and 6-12 mm in diameter are equally developmentally competent when used in a nuclear transfer procedure.  相似文献

Regulation of river flow and the amount of winter rainfall are the major factors affecting the water temperature of the spawning grounds, for green sturgeon in the Klamath River. During the primary spawning period of green sturgeon, mid-April to June, the water temperature may vary from 8 to 21°C. To estimate the potential implications of this modified thermal regime, we examined the survival and development in three progeny groups of green sturgeon embryos from zygote to hatch, at constant incubation temperatures (11–26°C). Temperatures 23–26°C affected cleavage and gastrulation and all died before hatch. Temperatures 17.5–22°C were suboptimal as an increasing number of embryos developed abnormally and hatching success decreased at 20.5–22°C, although the tolerance to these temperatures varied between progenies. The lower temperature limit was not evident from this study, although hatching rate decreased at 11°C and hatched embryos were shorter, compared to 14°C. The mean total length of hatched embryos decreased with increasing temperature, although their wet and dry weight remained relatively constant. We concluded that temperatures 17–18°C may be the upper limit of the thermal optima for green sturgeon embryos, and that the river thermal regime during dry years may affect green sturgeon reproduction.  相似文献

An egg of the critically endangered flapper skate Dipturus intermedius was successfully incubated to hatching in captivity in what is believed to be a first for the species. Water conditions (temperature, salinity, flow rate) were recorded, with mean water temperatures ranging from a monthly mean of 8.3 ± 1.2 to 13.2 ± 0.3°C and salinity from a monthly mean of 30.5 ± 1.2 to 36.6 ± 2.3 ppt. Hatching occurred after 534 days, suggesting that flapper skate eggs take c. 5700 growing degree-days to incubate to hatching. The egg's prolonged embryonic development raises concerns about flapper skate eggs' vulnerability to anthropogenic disturbance.  相似文献

Variation in developmental conditions is known to affect fitness in later life, but the mechanisms underlying this relationship remain elusive. We previously found in jackdaws Corvus monedula that larger eggs resulted in larger nestlings up to fledging. Through a cross-foster experiment of complete clutches we tested whether this association can be attributed to egg size per se, or to more proficient parents producing larger eggs and larger nestlings, with the latter effect being more or less independent of egg size. Due to other manipulations post-hatching, we primarily investigated effects on nestling mass on day 5, which we show to predict survival until fledging. We introduce a new statistical approach to compare the competing hypotheses and discuss the multiple advantages of this approach over current practice of which we report the results for comparison. We conclude that 92% of the slope of the association between egg size and nestling mass can be attributed to a direct effect of egg size. The remaining 8% of the slope can be attributed to aspects of parental chick rearing ability as reflected in egg size, but this component did not deviate significantly from zero. Intriguingly, the effect of egg size on day 5 nestling mass was steeper (1.7 g cm−3) than the effect of egg size on day 1 hatchling mass (0.7 g cm−3). Early growth is exponential, and the difference in effect size may therefore be explained by hatchlings from large eggs being further in their development at hatching. The direct effect of egg size on nestling mass raises the question what causes egg size variation in jackdaws.  相似文献
