Oxygen consumption rates of nauplii of the brine shrimp Artemia franciscana Kellogg 1906 were determined over a range of salinities from 10 to 110 ppm, in temperatures from 0 to 30°C, using a multi-factorial design. The oxygen micro-sensors employed have a fast response time and are capable of accurately measuring oxygen concentrations at temperatures well below 0°C. Oxygen uptake rate ranged from 0.03 to 0.66 μmol O2 mg−1 h−1 and was sensitive to changes in both salinity and temperature. Temperature was the dominant factor affecting oxygen consumption rates, which showed a significant increase with increasing temperature. A slight decrease was measured in oxygen consumption with increasing salinity related to differential solubility of oxygen in waters of different salinities. Thermal sensitivity of oxygen consumption determined from calculations of Q 10, indicated physiological adaptation of Artemia nauplii to the ranges of temperatures tested. Chromosome set manipulation was used to produce rainbow trout, Oncorhynchus mykiss, with identical nuclear backgrounds, but different maternal backgrounds to determine mitochondrial effects on development rate and oxygen consumption. Significant differences in development rate and oxygen consumption were observed between groups from different females. Development rates ranged from a mean of 317.97 degree days (°d) to 335.25 °d in progeny from different females. Mean oxygen consumption rates ranged from 3.31 μmol O2 g− 1 wet mass h− 1 to 9.66 μmol O2 g− 1 wet mass h− 1. Oxygen consumption and development rate analysis revealed the two slowest developing groups had the highest oxygen consumption rates. Development rate differences between second generation clonal females indicate that mitochondrial genomes play a significant role on early development and are comparable to development rate differences between clonal lines of rainbow trout. These results indicate that selection for mitochondrial genomes could increase growth rates and possibly food conversion ratios in aquaculture species.  相似文献

Summary Under chemolithoautotrophic growth conditions with the organism Alcaligenes eutrophus H16 the exponential growth phase is characterized by two different growth rates, each associated with different specific rates of ammonium consumption. On the basis of the analytical determination of Poly--hydroxybutyric acid (PHB), it can be conclusively shown that PHB is synthesized even during the exponential growth phase at a specific rate proportional to the specific growth rates of total biomass. After complete consumption of ammonium, the increase of biomass is exclusively due to PHB synthesis, whereas protein and rest biomass (cell dry weight minus PHB) remain constant. After an extended period of fermentation, the PHB content reaches a saturation value. The transient phase between the growth and the storage phase is very short in comparison to the duration of the whole fermentation. 1. Temperature, organic carbon and oxygen consumption were measured over a year at 13 sites in four lowlands streams within the same region in North Zealand, Denmark with the objectives of determining: (i) spatial and seasonal differences between open streams, forest streams and streams with or without lakes, (ii) factors influencing the temperature dependence of oxygen consumption rate, (iii) consequences of higher temperature and organic content in lake outlets on oxygen consumption rate, and (iv) possible consequences of forecasted global warming on degradation of organic matter. 2. High concentrations of easily degradable dissolved (DOC) and particulate organic carbon (POC) were found in open streams downstream of plankton‐rich lakes, while high concentrations of recalcitrant DOC were found in a forest brook draining a forest swamp. Concentrations of predominantly recalcitrant POC and DOC were low in a groundwater‐fed forest spring. Overall, DOC concentration was two to 18 times higher than POC concentrations. 3. Oxygen consumption rate at 20 °C was higher during summer than winter, higher in open than shaded streams and higher in lake outlets than inlets. Rate was closely related to concentrations of chlorophyll and POC but not to DOC. The ratio of oxygen consumption rate to total organic concentrations (DOC + POC), serving as a measure of organic degradability, was highest downstream of lakes, intermediate in open streams and lowest in forest streams. 4. Temperature coefficients describing the exponential increase of oxygen consumption rate between 4 and 20 °C averaged 0.121 °C?1 (Q10 of 3.35) in 70 measurements and showed no significant variations between seasons and stream sites or correlations with ambient temperature and organic content. 5. Oxygen consumption rate was enhanced downstream of lakes during summer because of higher temperature and, more significantly, greater concentrations of degradable organic carbon. Oxygen consumption rates were up to seven times higher in the stream with three impoundments than in a neighbouring unshaded stream and 21 times higher than in the groundwater‐fed forest spring. 6. A regional climate model has calculated a dramatic 4–5 °C rise in air temperature over Denmark by 2070–2100. If this is realised, unshaded streams are estimated to become 2–3 °C warmer in summer and winter and 5–7 °C warmer in spring and, thereby, increase oxygen consumption rates at ambient temperature by 30–40% and 80–130%, respectively. Faster consumption of organic matter and dissolved oxygen downstream of point sources should increase the likelihood of oxygen stress of the stream biota and lead to the export of less organic matter but more mineralised nutrients to the coastal waters.  相似文献

Oxygen consumption and dissolved nitrogen fluxes at the water-sediment interface of an oyster-bed were measured in situ using transparent enclosures inserted on undisturbed sediment. Experiments were performed in summer, under dark and light conditions, with various densities of the oyster Crassostrea gigas (0–150 animals m – 2). The influence of oyster biomass on oxygen and ammonia exchange rates was similar in both lighting conditions. Oxygen consumption increased with increasing biomass, though not at the level of prediction for the highest biomasses, suggesting a depressed respiration rate. Ammonia release never matched the rates predicted by adding sediment efflux to oyster excretion, when biomass exceeded 100 g DW m – 2. The coupling between oxygen consumption and ammonia release (O : N ratio) was thus influenced by oyster biomass. Stabilization of nitrogen release was related to enhanced nitrification in the presence of oyster and/or sediment uptake of ammonia against the molecular gradient. Urea release was erratic but appeared uninfluenced by oyster biomass. Fluxes of oxygen and of each nitrogen compound displayed thus a specific response to modifications of the oyster biomass. Both the organisms and the sediment are involved in regulation processes of metabolic exchange rates at the water-sediment interface.  相似文献

Low dissolved oxygen (DO) levels often occur during summer in tidal creeks along the southeastern coast of the USA. We analyzed rates of oxygen loss as water-column biochemical oxygen demand (BOD5) and sediment oxygen flux (SOF) at selected tidal creek sites monthly over a 1-year period. Ancillary physical, chemical and biological data were collected to identify factors related to oxygen loss. BOD5 rates ranged from 0.0 mg l?1 to 7.6 mg l?1 and were correlated positively with organic suspended solids, total suspended solids, chlorophyll a concentrations, temperature, and dissolved oxygen, and negatively with pH and nitrate + nitrite. SOF rates ranged from 0.0 to 9.3 g O2 m?2 d?1, and were positively correlated with temperature, chlorophyll a, and total suspended solids, but negatively with dissolved oxygen. Both forms of oxygen uptake were seasonally dependent, with BOD5 elevated in spring and summer and SOF elevated in summer and fall. Average oxygen loss to sediments was greater and more variable than oxygen loss in the water column. Oxygen deficits at three of five locations were significantly related to BOD5 and SOF, but not at two sites where ground water discharges were observed. Correlation and principal component analyses suggested that BOD5 and SOF responded to somewhat different suites of environmental variables. BOD5 was driven by a set of parameters linked to warm season storm water inputs that stimulated organic seston loads, especially chlorophyll a, while SOF behaved less strongly so. Runoff processes that increase loads of organic material and nutrients and ground water discharges low in dissolved oxygen contribute to occurrences of low dissolved oxygen in tidal creeks.  相似文献

Growth and lactose metabolism of a Leuconostoc mesenteroides strain were studied in batch cultures at pH 6.5 and 30° C in 101 modified MRS medium sparged with different gases: nitrogen, air and pure oxygen. In all cases, growth occurred, but in aerobiosis there was oxygen consumption, leading to an improvement of growth yield Y x/s and specific growth rate compared to anaerobiosis. Whatever the extent of aerobic growth, oxygen uptake and biomass production increased with the oxygen transfer rate so that the oxygen growth yield, Y x/o2, remained at a constant value of 11 g dry weight of biomass/mol oxygen consumed. Pure oxygen had a positive effect on Leuconostoc growth. Oxygen transfer was limiting under air, but pure oxygen provided bacteria with sufficient dissolved oxygen and leuconostocs were able to consume large amounts of oxygen. Acetate production increased progressively with oxygen consumption so that the total molar concentration of acetate plus ethanol remained constant. Maximal Y x/s was obtained with a 120 l/h flow rate of pure oxygen: the switch from ethanol to acetate was almost complete. In this case, a 46.8 g/mol Y x/s and a 0.69 h–1 maximal growth rate could be reached.  相似文献

Spatial and temporal variations in the concentrations of dissolved gases (CH4, CO2, and O2) in peat cores were studied using membrane inlet mass spectrometry (MIMS). Variations in vertical gas profiles were observed between random peat cores taken from hollows on the same peat bog. Methane concentrations in profiles (0–30 cm) generally increased with depth and reached maximum values in the range of 200–450 m CH4 below about 13-cm depth. In some profiles, a peak of dissolved methane was observed at 7-cm depth. Oxygen penetrated to approximately 2-cm depth in the hollows. The sampling probe was used to continuously monitor CH4, CO2, and O2 concentrations at fixed depths in peat cores over periods of several days. The concentration of dissolved CO2 and O2 at 1-cm depth oscillated over a 24-h period with the maximum of CO2 concentration corresponding with the minimum of 02. Diurnal variations in CO2 but not CH4 were measured at 15-cm depth; dissolved CO2 levels decreased during daylight hours to a constant minimum concentration of 4.85 mm. This report also describes the application of MIMS for the measurement of gaseous diffusion rates in peat using an inert gas (argon); the value of D, the diffusion coefficient, was 2.07 × 10–8 m2 s–1. Oxygen consumption of luminous bacteria determined by the Thunberg micro respirometer and by the time which elapses before the luminescence of an emulsion of luminous bacteria in sea water begins to dim, when over 99 per cent of the dissolved oxygen has been consumed, agree exactly. Average values for oxygen consumption at an average temperature of 21.5°C. are 4.26 x 10–11 mg. O2 per bacterium; 2.5 x 104 mg. per kilo and 5.6 mg. O2 per sq. m. of bacterial surface. The only correct comparison of the oxygen consumption of different organisms or tissues is in terms of oxygen used per unit weight with a sufficient oxygen tension so that oxygen consumption is independent of oxygen tension. Measurement of the oxygen concentration which just allows full luminescence, compared with a calculation of the oxygen concentration at the surface of a bacterial cell just necessary to allow the observed respiration throughout all parts of the cell, indicates that oxygen must diffuse into the bacterium much more slowly than through gelatin or connective tissue but not as slowly as through chitin.  相似文献

Respiration of four freshwater species of the amphipod crustacean Gammarus: G. fossarum, G. lacustris (river and lake), G. pulex and G. roeseli were measured in a closed, stirred respirometric chamber with a micro-electrode. Oxygen consumption, expressed as weight-specific oxygen uptake (Rs) in relation to decreasing oxygen concentration, varied at air saturation from 0.86 (G. lacustris, lake) to 2.06 μl O2 mg−1 AFDW h−1 (G. pulex). Rs also differed intra-specifically among the two populations of G. lacustris. G. lacustris (river), G. pulex and G. roeseli expressed moderate ability to regulate their oxygen consumption at decreasing oxygen concentrations, whereas the regulation ability was higher in G. lacustris (lake) and in G. fossarum, which maintain high oxygen uptake at oxygen levels >2 mg O2 l−1. All four Gammarus species are partial regulators in response to variations in oxygen concentration. The differences between species are considered too small to account for their natural distributions. It appears that the tolerances of Gammarus species to organic pollution depend only in part on oxygen conditions.  相似文献

1. Denitrification, net oxygen consumption and net nitrous oxide flux to the atmosphere were measured in three small rivers (discharge approximately 2–27 m3 s?1) at the whole reach scale during Spring and Summer, 2002. Two of these rivers (Iroquois River and Sugar Creek in north‐west Indiana – north‐east Illinois, U.S.A.) drained agricultural catchments and the other (Millstone River in central New Jersey, U.S.A.) drained a mixed suburban–agricultural catchment. 2. Denitrification, oxygen consumption and N2O flux were measured based on net changes in dissolved gas concentrations (N2, O2, and N2O) during riverine transport, correcting for atmospheric exchange. On each date, measurements were made during both light and dark periods. 3. Denitrification rates in these rivers ranged from 0.31 to 15.91 mmol N m?2 h?1, and rates within each river reach were consistently higher during the day than during the night. This diurnal pattern could be related to cyclic patterns of nitrification driven by diurnal variations in water column pH and temperature. 4. Oxygen consumption ranged from 2.56 to 241 mmol O2 m?2 h?1. In contrast to denitrification, net oxygen consumption was generally higher during the night than during the day. 5. River water was consistently supersaturated with N2O, ranging from 102 to 209% saturated. Net flux of N2O to the atmosphere ranged from 0.4 to 60 μmol N m?2 h?1. Net flux of N2O was generally higher at night than during the day. The high flux of N2O from these rivers strengthens the argument that rivers are an important contributor to anthropogenic emissions of this greenhouse gas.  相似文献

In this work the concept of bubble-free oxygenation that was able to ensure oxygen supply and carbon dioxide extraction for a chemostat culture of Escherichia coli was experimentally demonstrated. It was operated at the dilution rate of 0.275 h–1 under atmospheric pressure and at 35.5 °C. Foralkyl, a commercial perfluorocarbon, added in the influent medium under emulsified form and at 50% volumic fraction, was able to provide 0.17 g O2/l/h and extract 0.23 g CO2/l/h for a culture at 0.74 g/l of biomass. This oxygen supply was close to the maximum oxygenation Foralkyl was theoretically able to provide at this pressure when imposing a minimum oxygen concentration of 1 mg/l in the water phase. The quantification of transfer was not done from a direct measurement of oxygen transfer rates because conventional oxygen concentration measurement by membrane polarographic probe in an emulsion was not judged reliable. Stomatal movement is an energetic oxygen-requiring process. In the present study, the effect of oxygen concentration on mitochondrial respiratory activity and red-light-dependent photosynthetic oxygen evolution by Vicia faba and Brassica napus guard cell protoplasts was examined. Comparative measurements were made with mesophyll cell protoplasts isolated from the same species. At air saturated levels of dissolved oxygen in the protoplast suspension media, respiration rates by mesophyll protoplasts ranged from 6 to 10μmoles O2 mg?1 chl h?1, while guard cell protoplasts respired at rates of 200–300 μmoles O2 mg chl?1 h?1, depending on the species. Lowering the oxygen concentration below 50–60 mmol m?3 resulted in a decrease in guard cell respiration rates, while rates by mesophyll cell protoplasts were reduced only at much lower concentrations of dissolved oxygen. Rates of photosynthesis in mesophyll cell protoplasts isolated from both species showed only a minor reduction in activity at low oxygen concentrations. In contrast, photosynthesis by guard cell protoplasts isolated from V. faba and B. napus decreased concomitantly with respiration. Oligomycin, an inhibitor of oxidative phos-phorylation, reduced photosynthesis in mesophyll cell protoplasts by 27–46% and in guard cell protoplasts by 51–58%. The reduction in both guard cell photosynthesis and respiration following exposure to low oxygen concentrations suggest close metabolic coupling between the two activities, possibly mediated by the availability of substrate for respiration associated with photosynthetic electron transport activity and subsequent export of redox equivalents.  相似文献

Synopsis The effects of constant and diurnally fluctuating levels of dissolved oxygen on the growth of young-of-the-year winter flounder,Pseudopleuronectes americanus, were examined under controlled laboratory conditions. Fish were exposed for either 10 or 11 weeks to constant levels of 6.7 (high) and 2.2 (low) mg l–1, and a diurnal fluctuation, ranging from 2.5 to 6.4 mg 02l–1. Growth rates, calculated for both standard length and weight, for fish exposed to low and diurnally fluctuating levels were significantly reduced (p < 0.001) as compared to those for fish exposed to the high level. Growth rates of fish exposed to the high level were over twice those of fish held under low oxygen conditions. Under fluctuating conditions, fish grew at intermediate rates. Following these exposures, all fish were subsequently held at 7.2 mg Oz l–1 for five weeks. Growth rates increased over two and a half times for fish previously exposed to the low oxygen level and were significantly (p < 0.001) higher than for the other two groups.  相似文献

We investigated the effect of dissolved organic carbon (DOC) on hypolimnetic metabolism (accumulation of dissolved inorganic carbon (DIC) and methane (CH4)) in 21 lakes across a gradient of DOC concentrations (308 to 1540 mol C L–1). The highly colored nature of the DOC in these lakes suggests it is mostly of terrestrial origin. Hypolimnetic methane accumulation was positively correlated with epilimnetic DOC concentration (Spearman rank correlation = 0.67; p < 0.01), an indicator of allochthonous DOC inputs, but not with photic zone chlorophyll a concentration (Spearman rank correlation = 0.30; p = 0.22). Hypolimnetic DOC concentrations declined in 19 of 21 lakes during the stratified period at rates that ranged from 0.06 to 53.9 mmol m–2 d–1. The hypolimnetic accumulation of DIC + CH4 was positively correlated with, and, in most cases of comparable magnitude to, this DOC decline suggesting that DOC was an important substrate for hypolimnetic metabolism. The percentage of surface irradiance reaching the thermocline was lower in high DOC lakes (0.3%) than in low DOC lakes (6%), reducing hypolimnetic photosynthesis (as measured by the depth and magnitude of the deep dissolved oxygen maxima) in the high DOC lakes. In June, the hypolimnia of lakes with < 400 mol L–1 DOC had high concentrations of dissolved oxygen and no CH4, while the hypolimnia of lakes with DOC > 800 mol L–1 were completely anoxic and often had high CH4 concentrations. Thus, DOC affects hypolimnetic metabolism via multiple pathways: DOC was significant in supporting hypolimnetic metabolism; and at high concentrations depressed photosynthesis (and therefore oxygen production and DIC consumption) in the hypolimnion.  相似文献

The Fenton-type reaction between ferrous diethylenetriamine pentaacetic acid (Fe2+-DTPA, 50–200 μM) and H2O2 (20–1000 μM) in phosphate buffer at pH 7.0 results in consumption of dissolved oxygen. This observation differs from many prior reports that oxygen is liberated when more concentrated solutions of H2O2 are decomposed by iron salts. The rate and total quantity of oxygen consumed were dependent upon the concentrations of ferrous chelate, H2O2, and excess DTPA. Evidence is provided that both the ferrous-DTPA chelate and free DTPA can participate in the oxygen-consuming reactions. Oxygen was also consumed during the Fenton reaction between ferrous ions and H2O2 when DTPA and phosphate buffer were omitted. Under these conditions, oxygen evolution was observed at higher H2O2 concentrations (e.g., 400 μM). The consumption of oxygen during the Fenton-type reaction of an iron chelate at neutral pH may be relavant to events that take place in biologic systems.  相似文献

Summary Instantaneous oxygen consumption, muscle potential frequency, thoracic and ambient temperature were simultaneously measured during heating in individual workers and drones of honey bees. Relationships between these parameters and effects of thoracic temperature on power input and temperature elevation were studied. Oxygen consumption increased above basal levels only when flight muscles became active. Increasing muscle potential frequencies correlated with elevated oxygen consumption and raised thoracic temperature. The difference between thoracic and ambient temperature and oxygen consumption were linearly related. Oxygen consumption per muscle potential (l O2 · g –1 thorax · MP–1) was two-fold higher in drones than in workers. However, oxygen consumption for heating the thorax (l O2 · g –1 thorax · (Tth-Ta) · °C–1) was nearly the same in workers and drones. Thoracic temperature affected the amount of oxygen consumed per muscle potential (R10=1.5). Achieved temperature elevation per 100 MP was more temperature sensitive in drones (R10=6–10) than in workers (R10=3.6). Q10 values for oxygen consumption were 3 in workers and 4.5–6 in drones. Muscle potential frequency decreased with a Q10=1.8 in workers and 2.7 in drones. Heating behaviour of workers and drones was different. Drones generated heat less continuously than workers, and showed greater interindividual variability in predilection to heat. The oxygen and nitrate reduction kinetics of a nonflocculating strain of Zoogloea ramigera were determined. Axenic, nitrate-reducing bacterial suspensions were acclimated to various oxygen levels in a chemostat while measuring nitrate reduction in the presence of high ammonium nitrogen concentrations. Significant nitrate reduction was observed at oxygen concentrations up to 8 mg L–1. Oxygen consumption was inhibited by oxygen concentrations in excess of 2 mg L–1.  相似文献

The effect was studied of oxygen supply on the changes in total and specific rate of oxygen consumption by the cells, oxygen transfer rate, saturation concentrations of dissolved oxygen and the yields of batch and continuous cultivations. Experiments were done on the microorganismKlebsiella aerogenes CCM 2318 growing on synthetic glucose medium. Continuous cultivations were carried out at dilution rates of 0.96 and 0.178 h−1. The rate of oxygen transfer was determined by the sulphite method and the coefficient KLa was assessed using the dynamic method with a correction for changes in the saturations of dissolved oxygen. A lowered oxygen supply in batch cultivations caused deformations in the course of cell respiration. Comparison of results of batch and continuous cultivations showed that the highest yields Yx/s and Yx/o are attained at low dilution rates without oxygen limitation. Batch cultivations, on the other hand, exhibit the lowest yields and the highest cell respiration levels. In both types of cultivations, a respiration peak was ascertained under the conditions of growth limitation by oxygen.  相似文献
