A culture of the thermophilic, unicellular, blue-green alga Synechococcus lividus was freed from two thermophilic bacteria by use of equilibrium (isopycnic) centrifugation in Ficoll density gradients. After removal of the bacteria, the alga would grow only on agar plates in a high carbon dioxide atmosphere. Intermittent illumination, equilibrium centrifugation, and differential centrifugation were tested as techniques for obtaining synchronized cultures of the alga. Daughter cells selected by differential centrifugation in Ficoll density gradients yielded the best synchrony, and a cross-wall index of 85% was observed during the period of cell division.  相似文献

We describe afast, low-cost, and reliable way of creating arrays from sample molecules of interest present within microformatted sample vessels (such as 1536-well microplates). The principle involves simple centrifugal transfer of molecules of interest onto a solid planar or membrane surfaces placed over the initial sample vessel. Tools and procedures are presented that validate the robustness and precision of this facile solution to an otherwise difficult problem in modern molecular genetics. The availability of transferred DNA molecules for hybridization is also demonstrated. In conclusion, this "centrifugal-array" concept should help research studies to be applied on ever-greater scales with very simple machinery.  相似文献

Measles virus has been centrifuged on different density gradients. It sediments at densities of 1,20 g/cm3 in K-tartrate, of 1,18–1,21 g/cm3 in sucrose, 1,19–1,23 g/cm3 in CsCl and 1,19 g/cm3 in metrizamide gradients. Metrizamide reduced measles virus infectivity. In sucrose gradients sometimes more than one infectious peak was observed. Control Vero cells produced particles of the same densities as measles virus peaks. These peaks did contain actin as the major protein. The relevance of this finding in relation to the presence of actin in measles virus is discussed.  相似文献

Sample centrifugation onto membranes for sequencing   总被引:2,自引:0,他引:2  
This paper presents a new method for adsorption of proteins in solution onto a polyvinylidene diflouride (PVDF) membrane using centrifugation. The technique uses a low molecular weight cut-off membrane (LMW) placed underneath a PVDF membrane. The paired membranes are placed in a receptacle which in turn fits into a microcentrifuge tube. During sample centrifugation, the LMW acts to increase the amount of protein that is concentrated and adsorbed onto the hydrophobic surface of the PVDF membrane. By alternating between two receptacle sizes, this method can accommodate large (greater than 10 micrograms) and small (less than 10 micrograms) amounts of sample. This paper demonstrates sample recovery for a variety of proteins as quantitated by radioactivity and amino acid analysis after centrifugation onto PVDF. Amino acid and sequence analysis results demonstrate the efficiency with which interfering buffers and sodium dodecyl sulfate are removed as a result of sample centrifugation and washing. Finally, we demonstrate the utility of this technique with samples in the low picomole range to obtain useful sequence information following electrophoretic isolation of cyanogen bromide fragments purified by high performance electrophoresis chromatography.  相似文献
