The changes in germinability and phospholipid content of naturallyand artificially aged tomato seeds, variety Kingley Cross, werecompared. Subsequent low temperature pre-sowing treatment (LTPST)was unable to prevent decreases in the germination capacityof aged samples, but significantly improved germination rates.However, in contrast to previous studies on artificial ageingusing the variety Moneymaker, losses of total phospholipid werenot always well correlated with decreased germinability. Totalphospholipid levels in naturally aged seeds were largely unaffectedby pre-sowing treatment, but the proportion of phosphatidylcholinewas greatly increased. These results strongly suggest that lossesof phospholipid are coincidental rather than causative eventsin the loss of seed viability during ageing, and that the patternof changes depends on the type of ageing experienced by theseeds. The influence of prehydration in water or priming in –1.5 MPa polyethylene glycol 8000 solution for various periods,followed by redrying, on germination rate and longevity of lettuce(Lactuca sativa L.) seeds (achenes) was determined during controlleddeterioration at 10% moisture content (fresh weight basis) and40°C. Short prehydration treatments (up to 1 h) had littleeffect on either germination rate or longevity, but significantlyimproved root growth rates. Increasing durations of prehydrationor priming reduced the mean time to germination by up to 61%relative to untreated seeds, but also reduced mean seed longevityby as much as 84% Prehydration and priming altered the relationshipsbetween germination rate and viability and between normal andabnormal seedlings during ageing. Prehydration in abscisic acidor at a temperature inhibitory to germination did not preventthe reduction in longevity under controlled deterioration conditions.While prehydration or priming treatments effectively acceleratesubsequent germination rates of lettuce seeds, the redried seedsare nonetheless highly susceptible to deterioration in storage. The influence of seed priming and ageing treatments on viabilityand rate of germination of tomato (Lycopersicon esculentum Mill.)seeds was examined under both long-term and controlled-deteriorationstorage conditions. Seeds of a single lot of tomato were eitherprimed or aged to increase or decrease the rate of germination(Argerich and Bradford, 1989). They were then stored at 6% moisturecontent (dry weight basis) at either 4 ?C or 30 ?C for 1 year.Both viability and germination rate were unaffected by eitherstorage temperature in control seeds, or by 4 ?C storage inprimed or aged seeds. At 30 ?C, however, viability and germinationrate of primed and aged seeds was markedly reduced after 6 monthsof storage. The temperature dependence of the germination rateand the spread of germination times within the population wasalso adversely affected by high temperature storage, particularlyfor primed seeds. Under controlled deterioration conditions(13.5% moisture content and 50 ?C), the rate of loss of viabilitywas greater for primed seed than for control or aged seeds.The relationship between seed viability and the mean germinationrate, however, was not influenced by the seed treatments. Thesedata are analysed in relation to current models of seed deteriorationduring storage and seed repair during priming. The results indicatethat enhancement of seed germination rates by priming treatmentssimultaneously lowers the resistance of seeds to deterioration.Primed tomato seeds must, therefore, be considered to be vigorousseeds with a reduced storage life. Damage and degradation of cellular proteins is observed duringage-induced seed deterioration. L-Isoaspartyl protein methyltransferase(EC 2.1.1.77 [EC] ) is an enzyme hypothesized to play a role in limitingand repairing age-induced damage to proteins. Tomato (Lycopersiconesculentum Mill. ‘New Yorker’) seeds were assayedfor changes in L-isoaspartyl methyl-transferase activity duringaccelerated ageing and after osmotic priming. Accelerated ageingof seeds for 1–4 d at 45C and 100% relative humidityreduced germination from 94% to 71%, increased the mean timeof germination (MTG) from 2.4 to 5.8 d, and was accompaniedby a correlative decrease in L-isoaspartyl methyltransferaseactivity (r2=0.90). Aged and untreated seeds were primed for7 d at 20C in darkness using aerated solutions of 3% KNO3 orpolyethylene glycol 8000 (PEG) with equivalent osmotic potential(–1.25 MPa). Priming with KNO3 decreased the MTG, butdid not improve germination percentage for untreated seeds.Priming did not affect L-isoaspartyl methyltransferase activityin untreated seeds, but restored activity in aged seeds primedin KNO3 to levels near that of untreated seeds. Priming withPEG did not effectively improve the MTG or increase L-isoaspartylmethyltransferase activity. During germination, L-isoaspartylmethyltransferase activity remained constant for 48 h post-imbibitionand then declined, suggesting that the enzyme was developmentallyregulated and inactivated or degraded as radicle emergence occurred. The possibility of using ATP content as an indicator of seedquality was studied in onion seeds {Allium cepa cv. Wdenswil).The percentage germination and ATP content of imbibed seedswere compared during 145 weeks of storage at three temperatures(3, 15 and 30C). ATP content, which was undetectable in airdriedseeds (moisture content: 9%, w/w), increased rapidly as a functionof imbibition time, as did the fresh weight and respirationrate, reaching a steady-state level after about 17 h. After36 weeks of storage, the rate of ATP formation was greater forthe seeds stored at 3C than for those kept at 15 and 30C.Furthermore, the onset of ATP synthesis was delayed. These phenomena,which are likely to be an expression of seed ageing, are usefulindicators, allowing the prediction of the loss of seed viabilitybefore the decrease in percentage germination which occurredbeyond 36 weeks of storage. In addition, the correlation betweenATP content and germination capacity of seeds during 145 weeksof storage was excellent (r=0.95 at 15C and 0.97 at 30C),provided that a 17 h-imbibition time, specific for onion seeds,was chosen. These results are discussed in terms of the controversyconcerning the correlation between the ATP content and germinationpercentage of seeds. The Effects of Priming and Ageing on Seed Vigour in Tomato   总被引:1,自引:0,他引:1  
A comparison was made of the effects of seed priming or ageingtreatments on the performance of tomato (Lycopersicon esculentumMill. cv. UC204C) seeds according to a number of indices ofseed vigour. A single lot of tomato seeds was primed in 120mol m–3 K2HPO4 + 150 mol m–3 KNO3 for 5 d at 20?C, or aged at 13% moisture content (dry weight basis) and 50?C for 6 d. Germination percentage (>98%) was unaffectedby priming and reduced to 85% by ageing. X-ray photographs andlongitudinal sections revealed the formation of free space surroundingthe embryo in dry primed seeds, which was not evident in controlor aged seeds. Priming increased the rate of germination atall temperatures above the base temperature (Tb), while ageingdecreased it. Tb was unaffected by priming and only slightlyincreased by ageing. The variation in individual times to germinationwas approximately doubled in both primed and aged seed comparedto the control, based upon the slopes of probit germinationpercentage versus log thermal time curves. Root growth aftergermination tests and seedling growth in both greenhouse andfield tests were not influenced by either priming or ageing.The conductivity test was found to be unreliable as a vigourtest for tomato seeds. The results identify several indiceswhich can be used to quantify seed vigour in tomato. They alsoillustrate that seed priming can enhance seed performance accordingto some criteria, while having no effect or decreasing qualityaccording to other criteria. Seed vigour can apparently be separatedinto various components which can be independently influencedby seed enhancement treatments. Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. The germination of two species of salt-marsh plants Limoniumvulgare Mill., and L. humile Mill., is investigated in relationto their ecology. Germination is inhibited by sea water, butgermination even in fresh water is low. Inhibition of germinationby sea water is negatively correlated with water uptake. However,a treatment in sea water stimulates subsequent germination infresh water. The effect of the sea-water treatment appears tobe one of osmotic shock which weakens the seed coat. Variationin response to the treatment is partly genetical and partlyenvironmental. Seeds in different parts of the salt-marsh willbe subject to differing conditions, both during seed developmentand during the interval between seed maturation and seed dispersal.In connection with this the effect of leaching by sea waterand fresh water is considered. The response to a sea-water treatmentvaries with time. Considered in the long term it shows somecyclical variation which might correspond with the tidal cycle.It is shown that seeds of Limonium can survive long periodsin sea water. There is a slow rate of germination during immersion.Most of those seeds which do not germinate in sea water surviveto germinate rapidly in fresh water. The response to oxygenconcentration, light, and temperature is also investigated.Experiments on seedling growth and establishment show the needfor fresh water and a suitable substrate. Mud gives a lowergermination but a higher survival, while sand gives a highergermination but a lower survival. The dispersal of Limoniumseeds in nature is investigated. Seeds of Limonium are mostabundant in the drift line, but even these only represent asmall fraction of the total produced. It is considered thatreproduction by seed is only important when a new area is beinginvaded. Subsequent reproduction is largely by vegetative spread.The ecological implications of the germination experiments arediscussed.  相似文献

Carboxypeptidase, aminopeptidase and proteinase activities weremeasured in endosperms from dry and germinating x Haynaldoticumsardoum naturally aged seeds. Carboxypeptidase activity, presentin dry seeds, decreased slightly during germination and remainednearly unchanged during the storage period. Aminopeptidase activityincreased during germination in younger seeds, but decreasedin non-viable seeds. Proteinase activity was absent in dry seeds,increased during germination in younger seeds and disappearedin the older ones. Proteinase activity was not recovered in old endosperms followingtransplantation of young embryos, and was recovered only toa very small extent in young endosperms following transplantationof old embryos. Young endosperms onto which young embryos hadbeen transplanted gave maximum recovery of enzyme activity,although this was lower than in young intact seeds. 为了确定沙拐枣植物种子的萌发特性及最优播前预处理方法,在实验室条件下,对10种沙拐枣植物的种子进行了磨砺、硫酸和热水浸泡、冷藏、种子浸出液处理,然后进行发芽实验研究。萌发实验的结果表明,10种沙拐枣植物对于不同的种子预处理,均表现出相似的萌发反应。磨砺、硫酸浸泡和冷藏处理对种子萌发有明显地促进作用。与对照相比,种子浸出液处理对种子的发芽率、发芽速度均具有明显地抑制作用,并能增强种子的休眠。冷藏处理具有打破有活力的种子休眠、促进种子萌发的作用,但它与热水浸泡处理一样,对有活力种子表现出一定的致死作用。沙拐枣植物的萌发模式在不同种子预处理问均表现出明显的差异性。机械磨擦和硫酸处理能够促进种子的萌发率及发芽势。泡果沙拐枣(Callingonum junceum)在本项实验中表现出很强的萌发能力。  相似文献

Seeds with low vigour as a result of ageing showed a large improvementin seed quality following a 2 h soak in water, reflected inan increased rate of germination, higher emergence in soil,and the retention of high germination after 24 h rapid ageingin a controlled deterioration vigour test. Similar, but smallerimprovements were seen after a priming treatment using polyethyleneglycol. Seeds dried to their initial moisture content aftertreatment retained these beneficial effects. The treatmentshad little effect on high vigour seed. These findings suggestthat seed invigoration by hydration treatments results fromthe repair of previously sustained deterioration. Sweet pepper seeds were osmotically conditioned in 0.4 M mannitolsolution for 4 d (at 25 °C, in darkness) before or afterstorage at 35 °C for up to six months, and their germinationand viability was compared with that of untreated seeds storedunder the same conditions. Seeds that had been osmoconditionedprior to storage retained a high rate of germination and germinatedto a high final percentage (from 80 to 50 per cent) at both15 and 25 °C throughout the storage period. By contrast,both the rate and total level of germination of untreated pepperseeds declined rapidly at both germination temperatures, andby three months of storage the total level of seed viabilitywas already less than 10 per cent. Seeds that were first storedat 35 °C, and then osmoconditioned just prior to germination,showed a decline in germinability which when tested at 25 °Cwas the same as for untreated seeds, while tested at 15 °Coccurred at a slightly slower rate than for untreated seeds. It is evident that osmoconditioning prior to storage, in additionto the acceleration of germination, resulted in a dramatic delayof the ageing rate, thus increasing considerably the longevityof seeds. On the other hand, osmoconditioning after storagedid not seem to have any significant effect on seed viability,though it enhanced the germination rate. Moist chilling (cold stratification) is well-known as a simpleand effective means of overcoming physiological seed dormancyand enhancing germination of many temperate tree and shrub species.However, the apparent activation of intracellular repair mechanismsin non-dormant black spruce (Picea mariana) seeds followingmoist chilling, has not previously been reported. The presentcontribution records the beneficial effects of moist chillingafter accelerated ageing (40°C/98% relative humidity) for3–10 d of black spruce seeds. This treatment was maximallyeffective after 7 d of accelerated ageing, increasing germinationfrom 43 to 61%. Ultrastructural examination of embryo cellsindicated that intracellular activation occurred during moistchilling even when seeds had not been subjected to acceleratedageing, and that the increasing levels of damage accumulatingover the 3–7 d period of ageing were substantially reversedduring subsequent moist chilling. Using the method of room temperature phosphorescence (RTP), we divided air-dry pea (Pisum sativum L.) seeds subjected to accelerated ageing (40°C, 85% relative humidity) into three fractions: (I) high-quality seeds, (II) weakened seeds, and (III) dead seeds. In the process of ageing, seed germinability firstly decreased and then increased due to so-called “improved” seeds of fraction II, which returned to fraction I as judged from the RTP level; the germinability of these seeds became equal to that of fraction I seeds. Seeds capable of germination (fractions I and II) differed in the rates of imbibition, which depended on plasma membrane permeability (opened or closed water channels) but not on the presence of the seed coat. A low activation energy of seed imbibition in fraction II (less than 5 kcal/mol) indicates that water channels are open. A mercury-containing compound (5 μM p-chloromercuribenzoate (PCMB) reduced the rate of water uptake by these seeds, and dithiothreitol restored it. A high activation energy of fraction I seed imbibition (more than 12 kcal/mol) corresponded to the water uptake mainly across the lipid bilayer when water channels are closed. PCMB did not affect the rate of fraction I seed imbibition. We supposed that mature air-dry pea seeds had open water channels. During the first stages of fraction I seed imbibition, these channels were closed, limiting water uptake. NaF (100 μM), an inhibitor of phosphatase, prevented channel closing and accelerated the imbibition of fraction I seeds. It did not affect the imbibition rate of fraction II seeds, indicating their water channels to be opened. However, NaF did not affect the water uptake of “improved” fraction II seeds as well. It seems likely that their channels were closed during accelerated ageing but otherwise than via dephosphorylation. The results obtained indicate the possibility of water inflow regulation in the weakened seeds via the state of aquaporins, which form water channels in the membranes.  相似文献

The bases of differences in germination rates (GRg = inverseof time to germination [tg] of percentage g) among three cold/salt-toleranttomato (Lycopersicon esculentum Mill.) accessions (PI 341988,PI 120256, and PI 174263) and one cold/salt-sensitive tomatocultivar (T5) were investigated. The effects of seed priming(6 d imbibition in aerated –1.2 MPa polyethylene glycolsolution at 20 ?C followed by redrying) and of removing theendosperm/testa cap covering the radicle on the temperaturesensitivity of GRg, and the interaction of these treatmentswith genotypes, were also examined. GRg decreased linearly withdecreasing temperature for all genotypes and seed treatments.The minimum or base temperatures for germination (Tb) variedby 1 ?C among the tomato lines, so genotypic differences inGRg were due to differing thermal time requirements for germination.The mean thermal time requirement for germination of T5 seeds was 22% and 19% greater than that of PI 341988 andPI 120256 seeds, respectively, but only 9% greater than thatof PI 174263 seeds. Seed priming did not lower Tb of any genotype,but significantly reduced by 24, 49, 41, and 49% in T5, PI 341988, PI 120256, and PI 174263, respectively,indicating that priming increased the rate at which the seedsprogressed towards germination when T>Tb, but did not lowerthe minimum temperature at which germination could occur. Primingincreased the GRg of T5 seeds to equal or exceed those of control(non-primed) seeds of the cold/salt-tolerant genotypes at anyT>Tb, but the PI lines exhibited an even greater responseto priming. Times to germination within each seed lot were normallydistributed on a logarithmic scale. Priming increased the variancein tg within a seed lot when compared to control seeds. However,the variation in thermal time for germination between the 10thand 90th percentiles of the seed population (T(10–90))was relatively unaffected by priming due to the reduction in in primed seeds. Removing the endosperm cap and testa opposite the radicle tip decreased almost 6-fold and and reduced Tb by 5 ?C in T5 and PI 341988,implicating processes in the endosperm/testa as the limitingfactors in germination at suboptimal temperatures. Seeds of Hancornia speciosa germinated best at a temperatureof 20–30 °C. The viability of the seeds during storagewas short and the best storage conditions for viability entailedkeeping the seeds in polyethylene bags. Seed viability was maintainedonly when the seeds were stored at a moisture content above30%; storage conditions which allowed dehydration resulted ina rapid loss of viability (the seeds showed recalcitrant behaviour). Low temperature during storage did not improve longevity. Arelationship between germination and moisture content was established,but when the moisture content fell below 25% there was a drasticreduction of germination. After 9 weeks of storage, even athigh moisture content, seeds lost viability. Loss of seed viability during seed dehydration was associatedwith increased leakage of electrolytes and organic solutes,and reduced tetrazolium staining during subsequent imbibition. The seeds ofNasturtium palustreDC. do not germinate, eitherin the light or darkness, at various constant temperatures,but require for their full germination a certain period of alow temperature (5°) applied immediately after light irradiation.These results indicate the existance of at least two processes,a light-dependent process and a low temperature-requiring process,in the initiation of germination ofNasturtiumseeds. Experimentalevidence indicated further that the light exposure causes twodifferent processes in the seed germination. 2. When a dark period at 23° was inserted between the lightirradiation and the low temperature treatment the germinationwas suppressed. The inhibitory effect of the inserted dark periodat 23° was eliminated by a short irradiation during thedarkness (light-break). 3. Prolonged exposure ofNasturtium seeds to any concentrationof gibberellin brought about no germination when exposure wasgiven in complete darkness. The germination was promoted onlywhen light irradiation was applied to the seeds. A short applicationof gibberellin at a fairly high concentration was, however,remarkably effective for the germination even in the darkness,and the germination was inhibited as the gibberellin applicationwas lengthened. Three-month ‘old’ and ‘fresh’ seedsof Talinum triangulare were subjected to various treatmentsto induce early and rapid germination. Scarification and activated carbon were the most effective treatmentsin improving total germination in fresh seeds, while the 3 and5 per cent thiourea treatments were most effective in improvingtotal germination in old seeds. Activated carbon, scarificationand 5 per cent thiourea treatments enhanced early germinationin both old and fresh seeds. Cumulative percentage germinationwas very high in fresh seeds after scarification or after treatmentwith activated carbon and 5 per cent thiourea, and lowest inseeds treated with 3 per cent thiourea and hot water. In oldseeds, highest cumulative percentage germination was obtainedwith 3 and 5 per cent thiourea treatments and scarification.Generally, higher germination was obtained with fresh seedsthan with old seeds. Partial seed-coat removal and treatment with 5 per cent thiouriaresulted in a higher rate of and cumulative percentage germinationcompared with seeds with the coat partially removed but nottreated with thiourea. Constantly high temperature (34 °C) increased both rateand total germination compared with seeds planted at room temperature(20–23 °C). Treatments that did not induce germinationwere 1 per cent thiourea, H2SO4, cold water soaking at roomtemperature, 6 per cent hydrogen peroxide and soil planting.These treatments effected less than 3 per cent germination. The photorequirement for the germination of Eragrostis seedsdecreases with the progress of their after-ripening, and thegermination occurs whether in continuous light or in darknessat the final stage of after-ripening. The dehydration of seedsor the puncturing of seed coats also results in a decrease ofphotorequirement for germination. The rate of water absorptionof seeds increases with the germination capacity under continuousdark condition. However, there is no correlation between therespiration rate and the germination capacity; respiration isstimulated in the punctured seeds, but not in the after-ripenedseeds. The after-ripened or punctured seeds which no longer have aphotorequirement become light-sensitive again, when they areallowed to germinate in the air of low oxygen concentrations.The assumption is presented that the permeability to oxygenof the seed coat may be a factor controlling the seed germinationof this species.
