Summary The purpose of this work was to determine if hypotonicity, in addition to the stimulation of active Na+ transport (Venosa, R.A., 1978,Biochim. Biophys. Acta 510:378–383), promoted changes in (i) active K+ influx, (ii) passive Na and K+ fluxes, and (iii) the number of3H-ouabain binding sites.The results indicate that a reduction of external osmotic pressure () to one-half of its normal value (=0.5) produced the following effects: (i) an increase in active K+ influx on the order of 160%, (ii) a 20% reduction in Na+ influx and K+ permeability (P K), and (iii) a 40% increase in the apparent density of ouabain binding sites. These data suggest that the hypotonic stimulation of the Na+ pump is not caused by an increased leak of either Na+ (inward) or K+ (outward). It is unlikely that the stimulation of active Na+ extrusion and the rise in the apparent number of pump sites produced by hypotonicity were due to a reduction of the intracellular ionic strength. It appears that, at least in part, the stimulation of active Na+ transport takes place whenever muscles are transferred from one medium to another of lower tonicity even if neither one was hypotonic (for instance =2 to =1 transfer). Comparison of the present results with those previously reported indicate that in addition to the number of pump sites, the cycling rate of the pump is increased by hypotonicity. Active Na+ and K+ fluxes were not significantly altered by hypertonicity (=2).  相似文献

Summary It has been suggested that during the oxytocin-induced hydrosmotic response, water crosses the luminal membrane of urinary bladder epithelium cells through membranespanning proteins. Although specific inhibitors of osmotic water transport have not been found, certain sulfhydryl reagents such as mercurial compounds may help to identify the proteins involved in this permeation process. We tested the effects ofp-chloromercuribenzene sulfonate (PCMBS) and of fluoresceinmercuric acetate (FMA) on the net water flux, the microtubule and microfilament structures of the frog urinary bladder, and the distribution of intramembrane particle aggregates in the luminal membrane.We observed that: (i) 5mm PCMBS at pH 5 and 0.5mm FMA at pH 8 added to the mucosal bath at the maximum of the response to oxytocin partially inhibited the net water flux. Inhibition then increased progressively when the preparation was repeatedly or continuously stimulated, until it reached a maximal inhibition at 120 min. This inhibition was not reversed even when cystein was added in the mucosal bath. PCMBS and FMA effects were also observed when cyclic AMP (3,5 cyclic adenosine monophosphate) was used to increase water permeability. (ii) PCMBS mucosal pretreatment did not modify the basal water flux but potentiated the inhibitory effect of PCMBS or FMA on the hydrosmotic response to oxytocin. (iii) Microtubule and microfilament network, visualized in target cells by immunofluorescence, was not affected by PCMBS. (iv) The maximal PCMBS or FMA inhibition was not associated with a reduction of aggregate surface area in the apical membrane.The persistence of the intramembrane particle aggregates associated with the oxytocin-induced hydrosmotic response during the net water flux inhibition by PCMBS, suggests that the PCMBS effect occurs possibly at the level of sulfhydryl groups of the water channel itself.  相似文献

Summary Addition of glucose or the nonmetabolizable analogue -methyl-d-glucoside to rabbit proximal tubules suspended in a glucoseand alanine-free buffer caused a sustained increase in intracellular Na+ content (+43±7 nmol · (mg protein)–1) and a concomitant but larger decrease in K+ content (–72±11 nmol· (mg protein)–1). A component of the net K+ efflux was Ba2+ insensitive, and was inhibited by high (1mm) but not low (10 m) concentrations of the diuretics, furosemide and bumetanide. The increase in intracellular Na+ content is consistent with the view that the increased rates of Na+ and water transport seen in the proximal tubule in the presence of glucose can be attributed (at least in part) to a stimulation of basolateral pump activity by an increased [Na+] i .  相似文献

To characterize mechanisms of esophageal desalination, osmotic water permeability and ion fluxes were measured in the isolated esophagus of the seawater eel. The osmotic permeability coefficient in the seawater eel esophagus was 2·10-4 cm·s-1. This value was much lower than those in tight epithelial, although the eel esophagus is a leaky epithelium with a tissue resistance of 77 ohm·cm-2. When the esophagus was bathed in normal Ringer solutions on both sides no net ion and water fluxes were observed. However, when mucosal NaCl concentration was increased by a factor of 3, Na+ und Cl- ions were transferred from mucosa to serosa (desalination). If only Na+ or Cl- concentration in the mucosal fluid was increased by a factor of 3, net Na+ and Cl- fluxes were reduced to 30–40%, indicating that 60–70% of the net Na+ and Cl- fluxes are coupled mutually. The coupled NaCl transport seems to be effective in desalting the luminal high NaCl. The remaining 30–40% of the total Na+ and Cl- fluxes seems to be due to a simple diffusion, because these components are independent of each other and follow their electrochemical gradients, and also because these fluxes remain even after treatment with NaCN or ouabain. A half of the coupled NaCl transport could be explained by a Na+/H+–Cl-/HCO 3 - double exchanger on the apical membrane of the esophageal epithelium, because mucosal amiloride and 4.4-diisothiocyanatostilbene-2,2-disulphonic acid inhibited the net Na+ and Cl- fluxes by approximately 30%. Summary Transepithelial Li+ influx was studied in the isolated epithelium from abdominal skin ofRana catesbeiana. With Na+-Ringer's as inside medium and Li+-Ringer's as outside medium, the Li+ influx across the epithelium was 15.6 A/cm2. This influx was considerably reduced by removal of either Na+ or K+ from the inside bath or by the addition of ouabain or amiloride. Epithelial K+ or Na+ concentration was respectively lower in epithelia bathed in K+-free Ringer's or Na+-free Ringer's. In conditions of negligible Na+ transport, a 20mm Li+ gradient (outin) produced across the short-circuited epithelium a Li+ influx of 11.8 A/cm2 and a mean short-circuit current of 10.2 A/cm2. The same Li+ gradient in the opposite direction produced a Li+ outflux of only 1.9 A/cm2. With equal Li+ concentration (10.3 and 20.6mm) on both sides of the epithelium, plus Na+ in the inside solution only, a stable Li+-dependent short-circuit current was observed. Net Li+ movement (outin) was also indirectly determined in the presence of an opposing Li+ gradient. Although Li+ does not substitute for Na+ as an activator of the (Na++K+)-ATPase from frog skin epithelium, Li+ influx appears to be related to Na+–K+ pump activity. It is proposed that the permeability of the outer barrier to Na+ and Li+ is regulated by the electrical gradient produced by electrogenic Na+–K+ pumps located in the membrane of the deeper epithelial cells.  相似文献

Summary The present study describes a new perfusion technique—based on the use of a routine spectrofluorometer—which enables fluorometric evaluation of polarity, regulation and kinetics of Na+/H+ exchange at the level of an intact monolayer. Na+/ H+ exchange was evaluated in bicarbonate-free solutions in OK (opossum kidney) cells, a renal epithelial cell line. Na+/H+ exchange activity was measured by monitoring changes in intracellular pH (pH i ) after an acid load, using the pH-sensitive dye 27-bis (carboxyethyl) 5–6-carboxy-fluorescein (BCECF). Initial experiments indicated that OK cells grown on a permeable support had access to apical and basolateral perfusion media. They also demonstrate that OK cells express an apical pH i , recovery mechanism, which is Na+ dependent, ethylisopropylamiloride (EIPA) sensitive and regulated by PTH. Compared to resting conditions (pH i =7.68; pH o =7.4) where Na+/H+ exchange is not detectable, transport rate increased as pH i decreased. A positive cooperativity characterized the interaction of internal H+ with the exchanger, and suggests multiple H+ binding sites. In contrast, extracellular [Na+] increased transport with simple Michaelis-Menten kinetics. The apparent affinity of the exchanger for Na+ was 19mM at an intracellular pH of 7.1 and 60mM at an intracellular pH of 6.6. Inhibition of Na+/H+ exchange activity by EIPA was competitive with respect to extracellular [Na+] and theK i was 3.4 M. In conclusion, the technique used in the present study is well suited for determination of mechanisms involved in control of epithelial cell pH i and processes associated with their polarized expression and regulation.  相似文献

Summary Exposing the apical membrane of toad urinary bladder to the ionophore nystatin lowers its resistance to less than 100 cm2. The basolateral membrane can then be studied by means of transepithelial measurements. If the mucosal solution contains more than 5mm Na+, and serosal Na+ is substituted by K+, Cs+, or N-methyl-d-glucamine, the basolateral membrane expresses what appears to be a large Na+ conductance, passing strong currents out of the cell. This pathway is insensitive to ouabain or vanadate and does not require serosal or mucosal Ca2+. In Cl-free SO 4 2– Ringer's solution it is the major conductive pathway in the basolateral membrane even though the serosal side has 60mm K+. This pathway can be blocked by serosal amiloride (K i=13.1 m) or serosal Na+ ions (K i 10 to 20mm). It also conducts Li+ and shows a voltage-dependent relaxation with characteristic rates of 10 to 20 rad sec–1 at 0 mV.  相似文献

Summary The Na+ requirement for active, electrogenic Cl absorption byAmphiuma small intestine was studied by tracer techniques and double-barreled Cl-sensitive microelectrodes. Addition of Cl to a Cl-free medium bathingin vitro intestinal segments produced a saturable (K m =5.4mm) increase in shortcircuit current (I sc) which was inhibitable by 1mm SITS. The selectivity sequence for the anion-evoked current was Cl=Br>SCN>NO 3 >F=I. Current evoked by Cl reached a maximum with increasing medium Na concentration (K m =12.4mm). Addition of Na+, as Na gluconate (10mm), to mucosal and serosal Na+-free media stimulated the Cl current and simultaneously increased the absorptive Cl flux (J ms Cl ) and net flux (J net Cl ) without changing the secretory Cl flux (J sm Cl ). Addition of Na+ only to the serosal fluid stimulatedJ ms Cl much more than Na+ addition only to the mucosal fluid in paired tissues. Serosal DIDS (1mm) blocked the stimulation. Serosal 10mm Tris gluconate or choline gluconate failed to stimulateJ ms Cl . Intracellular Cl activity (a Cl i ) in villus epithelial cells was above electrochemical equilibrium indicating active Cl uptake. Ouabain (1mm) eliminated Cl accumulation and reduced the mucosal membrane potential m over 2 to 3 hr. In contrast, SITS had no effect on Cl accumulation and hyperpolarized the mucosal membrane. Replacement of serosal Na+ with choline eliminated Cl accumulation while replacement of mucosal Na+ had no effect. In conclusion by two independent methods active electrogenic Cl absorption depends on serosal rather than mucosal Na+. It is concluded that Cl enters the cell via a primary (rheogenic) transport mechanism. At the serosal membrane the Na+ gradient most likely energizes H+ export and regulates mucosal Cl accumulation perhaps by influencing cell pH or HCO 3 concentration.  相似文献

Summary Na+ transport was characterized in normal human fibroblasts and neoplastic H.Ep. 2 cells in order to investigate the role of the endogenous peptidic factor inhibitin that is secreted by a variety of neoplastic cells (including H.Ep. 2) and inhibits Na+/Na+ exchange in human erythrocytes. Although active (Na+, K+-ATPase mediated) Na+ fluxes were similar in the two cell types, H.Ep. 2 cells maintained higher intracellular Na[su+] concentration (26mm) compared to fibroblasts (12mm). An analysis of passive Na+ fluxes showed a difference in the handling of Na+ via ouabain and bumetanide-insensitive transport between the two cell types: H.Ep. 2 cells achieved net Na+ influx via an amiloride-sensitive pathway that was only demonstrated in fibroblasts when 10% fetal calf serum (FCS) was present. Kinetic studies were undertaken to investigate the interaction between Na+ flux via Na+/H+ and Na+/Na+ exchanges. for this purpose, an outwardly directed Na+ gradient was created by loading the cells with Na+ (Na i >100mm) to activate the reverse functioning of Na+/H+ exchange (i.e., Na out + H in + ). The rates of ouabain-and bumetanide-insensitive Na+ efflux were measured over a range of extracellular Na+ concentrations (Na o + 14–140mm). In the presence of 10% FCS, the two cell types showed different responses: in fibroblasts the Na+ efflux rate showed an inverse correlation with extracellular Na+ concentration, while H.Ep. 2 cells significantly increased their rate of Na+ efflux as extracellular Na+ concentration increased. So although the thermodynamic force would direct net Na+ efflux when Na i + >Na o + , H.Ep.2 cells were under kinetic control to perform Na+/Na+ exchange.When exogenous inhibitin was tested on fibroblasts, the steady-state intracellular Na+ concentration increased from 14 to 19mm (p<0.01). In Na+-loaded fibroblasts, serum-stimulated Na+ efflux was partially inhibitin sensitive and the maximal inhibitory effect was seen when extracellular Na+ concentration was 14mm and presumably the Na+/H+ exchanger operating in the reverse mode. This study demonstrated that, in contrast to fibroblasts, H.Ep.2 cells have a modified Na+/H+ exchange system whereby it acts in the Na in + H out + mode without exogenous growth factor activation and resists functioning in the reversed mode. It is proposed that inhibitin, is the endogenous modifier of this transport system in H.Ep.2 cells with the result that H.Ep.2 cells maintain a higher concentration of intracellular Na+ compared to fibroblasts.  相似文献

It is proposed that the activity of an epidermal cotransport system for Na+ and dicarboxylic amino acids accounts for the small amounts of L-glutamate and L-aspartate in the otherwise amino-acid-rich blood plasma of insects. This Na+-dependent transport system is responsible for more than 95% of the uptake of these amino acids into the larval epidermis of the beetle Tenebrio molitor. Kinetic analysis of uptake showed that the Na+-dependent co-transporter has medium affinity for L-glutamate and L-aspartate. The K m for L-glutamate uptake was 146 mol·l-1, and the maximum velocity of uptake (V max) was 12.1 pmol·mm-2 of epidermal sheet per minute. The corresponding values for L-aspartate were 191 mol·l-1 and 8.4 pmol·mm-2·min-1. The Na+/L-glutamate co-transporter has a stoichiometry of at least two Na+ ions for each L-glutamate-ion transported (n=217). The co-transporter has an affinity for Na+ equivalent to a K m of 21 mmol · l-1 Na+. Na+ is the only external ion apparently required to drive L-glutamate uptake. Li+ substitutes weakly for Na+. Removal of external K+ or addition of ouabain decreases uptake slowly over 1 h, suggesting that these treatments dissipate the Na+/K+ gradient by inhibiting epidermal Na+/K+ ATPase. Several structural analogues of L-glutamate inhibit the medium-affinity uptake of L-glutamate. The order of potency with which these competitive inhibitors block glutamate uptake is L-cysteatethreo-3-hydroxy-Dl-aspartate > D-aspartateL-aspartate> L-cysteine sulphinate > L-homocysteateD-glutamate. L-trans-Pyrrolidine-2,4-dicarboxylate, a potent inhibitor of L-glutamate uptake in mammalian synaptosomes, is a relatively weak blocker of epidermal uptake. The epidermis takes up substantially more L-glutamate by this Na+-dependent system than tissues such as skeletal muscle and ventral nerve cord. The epidermis may be a main site regulating blood L-glutamate levels in insects with high blood [Na+]. Summary The effect of extracellular and intracellular Na+ (Na o + , Na i + ) on ouabain-resistant, furosemide-sensitive (FS) Rb+ transport was studied in human erythrocytes under varying experimental conditions. The results obtained are consistent with the view that a (1 Na++1 K++2 Cl) cotransport system operates in two different modes: modei) promoting bidirectional 11 (Na+–K+) cotransport, and modeii) a Na o + -independent 11 K o + /K i + exchange requiring Na i + which, however, is not extruded. The activities of the two modes of operation vary strictly in parallel to each other among erythrocytes of different donors and in cell fractions of individual donors separated according to density. Rb+ uptake through Rb o + /K i + exchange contributes about 25% to total Rb+ uptake in 145mm NaCl media containing 5mm RbCl at normal Na i + (pH 7.4). Na+–K+ cotransport into the cells occurs largely additive to K+/K+ exchange. Inward Na+–Rb+ cotransport exhibits a substrate inhibition at high Rb o + . With increasing pH, the maximum rate of cotransport is accelerated at the expense of K+/K+ exchange (apparent pK close to pH 7.4). The apparentK m Rb o + of Na+–K+ cotransport is low (2mm) and almost independent of pH, and high for K+/K+ exchange (10 to 15mm), the affinity increasing with pH. The two modes are discussed in terms of a partial reaction scheme of (1 Na++1 K++2 Cl) cotransport with ordered binding and debinding, exhibiting a glide symmetry (first on outside = first off inside) as proposed by McManus for duck erythrocytes (McManus, T.J., 1987,Fed. Proc., in press). N-ethylmaleimide (NEM) chemically induces a Cl-dependent K+ transport pathway that is independent of both Na o + and Na i + . This pathway differs in many properties from the basal, Na o + -independent K+/K+ exchange active in untreated human erythrocytes at normal cell volume.     Summary In the NaK-ATPase proteoliposomes (PLs), the NaK-pump activity, Na+ uptake, and ATP hydrolysis were apparently enhanced by carbonyl cyanidem-chlorophenylhydrazone (CCCP) and other ionophores without ion gradients. These ionophore effects were not cation specific. Without ionophores, the PL's ATPase activity fell to its steady-state value within 3 sec at 15°C. This decrease in activity disappeared in the presence of CCCP. Since CCCP is believed to enhance proton mobility across the lipid bilayer and dissipate membrane potential (V m ), we postulated that aV m build-up partially inhibits the PLs by changing the conformation of the NaK-pump, and that CCCP eliminated this partial inhibition. Since this activation required extracellular K+ and high ATP concentration in the PLs, CCCP must affect the conversion between the phosphorylated forms of NaK-ATPase (EP); this step has been suggested by Goldschlegger et al. (1987) to be the voltage-sensitive step (J. Physiol. (London) 387:331–355). Although cytoplasmic K+ accelerated the change of ADP-and K+-sensitive EP (E*P) to K+-sensitive ADP-insensitive EP (E2P), CCCP did not compete with cytoplasmic K+ when cytoplasmic Na+ was saturated. When the PLs were phosphorylated with 20 m ATP and 20 m palmitoyl CoA instead of with high concentration of ATP, CCCP increased the E*P content and decreased the ADP-sensitive K+-insensitive EP (E1P). The results described above suggest that CCCP affects the E1P to E*P change in the E1PE*PE2P conversion and that this reaction step is inhibited byV m .  相似文献

    Summary This report details preliminary findings for ion channels in the plasma membrane of protoplasts derived from the cotyledons ofAmaranthus seedlings. The conductance properties of the membrane can be described almost entirely by the behavior of two types of ion channel observed as single channels in attached and detached patches. The first is a cation-selective outward rectifier, and the second a multistate anion-selective channel which, under physiological conditions, acts as an inward rectifier.The cation channel has unit conductance of approx. 30 pS (symmetrical 100 K+) and relative permeability sequence K+>Na+>Cl (10.160.03); whole-cell currents activate in a time-dependent manner, and both activation and deactivation kinetics are voltage dependent. The anion channel opens for hyperpolarized membrane potentials, has a full-level conductance of approx. 200 pS and multiple subconductance states. The number of sub-conductances does not appear to be fixed. When activated the channel is open for long periods, though shuts if the membrane potential (V m ) is depolarized; at millimolar levels of [Ca2+]cyt this voltage dependency disappears. Inward current attributable to the anion channel is not observed in whole-cell recordings when MgATP (2mm) is present in the intracellular solution. By contrast the channel is active in most detached patches, whether MgATP is present or not on the cytoplasmic face of the membrane. The anion channel has a significant permeability to cations, the sequence being NO 3 >Cl>K+>Aspartate (2.0410.18 to 0.090.04). The relative permeability for K+ decreased at progressively lower conductance states. In the absence of permeant anions this channel could be mistaken for a cation inward rectifier. The anion and cation channels could serve to clampV m at a preferred value in the face of events which would otherwise perturbV m .  相似文献

    Summary We have investigated the kinetic properties of the human red blood cell Na+/H+ exchanger to provide a tool to study the role of genetic, hormonal and environmental factors in its expression as well as its functional properties in several clinical conditions. The present study reports its stoichiometry and the kinetic effects of internal H+ (H i ) and external Na+ (Na o ) in red blood cells of normal subjects.Red blood cells with different cell Na+ (Na i ) and pH (pH i ) were prepared by nystatin and DIDS treatment of acid-loaded cells. Unidirectional and net Na+ influx were measured by varying pH i (from 5.7 to 7.4), external pH (pH o ), Na i and Na o and by incubating the cells in media containing ouabain, bumetanide and methazolamide. Net Na+ influx (Na i <2.0 mmol/liter cell, Na o = 150mm) increased sigmoidally (Hill coefficient 2.5) when pH i fell below 7.0 and the external pH o was 8.0, but increased linearly at pH o 6.0. The net Na+ influx driven by an outward H+ gradient was estimated from the difference of Na+ influx at the two pH o levels (pH o 8 and pH o 6). The H+-driven Na+ influx reached saturation between pH i 5.9 and 6.1. TheV max had a wide interindividual variation (6 to 63 mmol/liter cell · hr, 31.0±3, mean±sem,n=20). TheK m for H i to activate H+-driven Na+ influx was 347±30nm (n=7). Amiloride (1mm) or DMA (20 m) partially (59±10%) inhibited red cell Na+/H+ exchange. The stoichiometric ratio between H+-driven Na+ influx and Na+-driven H+ efflux was 11. The dependence of Na+ influx from Na o was studied at pH i 6.0, and Na i lower than 2 mmol/liter cell at pH o 6.0 and 8.0. The meanK m for Na o of the H+-gradient-driven Na+ influx was 55±7mm.An increase in Na i from 2 to 20 mmol/liter cell did not change significantly H+-driven net Na+ influx as estimated from the difference between unidirectional22Na influx and efflux. Na+/Na+ exchange was negligible in acid-loaded, DIDS-treated cells. Na+ and H+ efflux from acid-loaded cells were inhibited by amiloride analogs in the absence of external Na+ indicating that they may represent nonspecific effects of these compounds and/or uncoupled transport modes of the Na+/H+ exchanger.It is concluded that human red cell Na+/H+ exchange performs 11 exchange of external Na+ for internal protons, which is partially amiloride sensitive. Its kinetic dependence from internal H+ and external Na+ is similar to other cells, but it displays a larger variability in theV max between individuals.  相似文献

    Summary The studies reported here were carried out to characterize further previously described changes in membrane localized amino acid transport associated with simian virus 40 transformation of the mammalian cell line, Balb/c3T3. Membrane vesicles were prepared from confluent cultures of both simian virus 40 transformed Balb/c3T3 (SV3T3) and the untransformed parent line, Balb/c3T3 (3T3). An initial, externally imposed out>in, 100mm Na+ gradient produces acceleration of early ingress of -aminoisobutyric acid (AIB) in vesicles from both cell lines, but transient, concentrative uptake (overshooting) only in SV3T3 vesicles. Early ingress ofl-leucine is also accelerated in SV3T3 vesicles by a Na+ gradient, and overshooting is also demonstrable.Na+-gradient independent AIB permeability of SV3T3 and 3T3 membranes was estimated using uptake data, a first order rate equation and measurements of vesicle size derived from quasi-elastic light-scattering studies. AIB permeability of SV3T3 membranes is greater than that of 3T3 membranes (113 Å/min and 43 Å/min, respectively), suggesting that overshooting in 3T3 vesicles is not attenuated by a Na+-independent AIB leak.     Summary The role of transmembrane pH gradients on the ouabain, bumetanide and phloretin-resistant Na+ transport was studied in human red cells. Proton equilibration through the Jacobs-Stewart cycle was inhibited by the use of DIDS (125 m) and methazolamide (400 m). Red cells with different internal pH (pH i =6.4, 7.0 and 7.8) were prepared and Na+ influx was measured at different external pH (pH o =6.0, 7.0, 8.0). Na+ influx into acid-loaded cells (pH i =6.4) markedly increased when pH o was raised from 6.0 to 8.0. Amiloride, a well-known inhibitor of Na+/H+ exchange systems blocked about 60% of the H+-induced Na+ entry, while showing small inhibitory effects in the absence of pH gradients. When pH0 was kept at 8.0, the amiloride-sensitive Na+ entry was abolished as pH i was increased from 6.4 to 7.8. Moreover, measurements of H+ efflux into lightly buffered media indicated that the imposition of an inward Na+ gradient stimulated a net H+ efflux which was sensitive to the amiloride analog 5-N-methyl-N-butyl-amiloride. Furthermore, in the absence of a chemical gradient for Na+ (Na i + =Na 0 + =15mm,Em=+6.7 mV), an outward H+ gradient (pH i =6.4, pH0=8.0) promoted a net amiloride-sensitive Na+ uptake which was abolished at an external pH of 6.0.     Summary The relation of osmotic to diffusional water permeability of human red blood cells was compared after treating the cells with different concentrations of PCMBS (p-chloromercuribenzene sulfonate). After subtracting the PCMBS-insensitive permeability (presumably the water permeability of the lipid bilayer) from each, the ratio of osmotic to diffusional permeability remains invariant (11) as more and more water channels are inhibited by increasing concentrations of PCMBS. This result implies that the channels close in an all-or-none way and suggests a two-state model. Analysis of the dependence of osmotic water permeability on PCMBS concentration in terms of the model reveals a 11 stoichiometry and a dissociation constant for the PCMBS/membrane receptor complex of about 0.019mm at 37°C. Temperature dependence studies show that the reaction is entropically driven (H o25 kcal/mol, S o100 cal/moldeg) and suggest the involvement of hydrophobic interactions.  相似文献

    Summary Unitary K+ currents in single cells isolated from ventricular muscle of newborn rat hearts were measured in response to different potentials and [K] o . TheI/V curves were linear for potentials more negative than the zero-current voltage: especially in high [K] o (150nm KCl), no clear outward currents could be detected indicating a drastic rectification in the inward direction. The channel is mainly selective to K+ but Na+ ions are also carried (P Na/P K=0.056). The channel conductance is proportional to the square root of [K] o but Na+ ions seem to have a facilitatory effect on K, the single-channel conductance. The channel activity, measured asP o, i.e. the probability to find the channel in open state, decreased as the membrane was hyperpolarized. This behavior was tentatively explained by an inactivation process as the membrane becomes more negative. The rate constants of the transitions between the different states were calculated according to a C–O–C model. A control of the gating process by permeant ion K+ was postulated, based on the increase of one of the rate constants from the closed to the open state with [K] o . Finally, the macroscopicI/V curves calculated fromP o and i, the unit current, were found to be characteristic of a ion-blocked inward rectifier.  相似文献

    Summary The volume regulatory response of the Ehrlich ascites tumor was studied in KCl-depleted, Na+-enriched cells. Subsequent incubation in K+-containing NaCl medium results in the reaccumulation of K+, Cl, water and the extrusion of Na+. The establishment of the physiological steady state is due primarily to the activity of 2 transport systems. One is the Na/K pump (K M for K 0 + =3.5mm;J max=30.1 mEq/kg dry min), which in these experiments was coupled 1K+/1 Na+. The second is the Cl-dependent (Na++K+) cotransport system (K M for K 0 + =6.8mm;J max=20.8 mEq/kg dry min) which mediates, in addition to net ion uptake in the ratio of 1K+1Na+2Cl, the exchange of K i + for K 0 + . The net passive driving force on the cotransport system is initially inwardly directed but does not decrease to zero at the steady state. This raises the possibility of the involvement of an additional source of energy. Although cell volume increases concomitant with net ion uptake, this change does not appear to be a major factor regulating the activity of the cotransport system.  相似文献

    The free-living cyanobacterium Anabaena variabilis showed a biphasic pattern of 14CH3NH 3 + uptake. Initial accumulation (up to 60 s) was independent of CH3NH 3 + metabolism, but long-term uptake was dependent on its metabolism via glutamine synthetase (GS). The CH3NH 3 + was converted into methylglutamine which was not further metabolised. The addition of l-methionine-dl-sulphoximine (MSX), to inhibit GS, inhibited CH3NH 3 + metabolism, but did not affect the CH3NH 3 + transport system.NH 4 + , when added after the addition of 14CH3NH 3 + , caused the efflux of free CH3NH 3 + ; when added before 14CH3NH 3 + , NH 4 + inhibited its uptake indicating that both NH 4 + and CH3NH 3 + share a common transport system. Carbonylcyanide m-chlorophenylhydrazone and triphenyl-methylphosphonium both inhibited CH3NH 3 + accumulation indicating that the transport system was -dependent. At pH 7 and at an external CH3NH 3 + concentration of 30 mol dm-3, A. variabilis showed a 40-fold intracellular accumulation of CH3NH 3 + (internal concentration 1.4 mmol dm-3).
