采用苗期缺磷和全营养对照处理,以70个中国春—野生亲缘种属二体添加系及中国春为材料,根据苗期表观遗传性状、磷吸收率和利用率相对生物量对其进行耐低磷胁迫能力筛选鉴定和基因染色体定位。结果表明:大麦4H和长穗偃麦草7E染色体上携带有耐低磷胁迫的优异基因;长穗偃麦草6E、黑麦1R和6R、卵穗山羊草4Ug和6Mg、易变山羊草4Sv染色体携带促进小麦根系生长发育的基因;拟斯比尔托山羊草5S和簇毛麦4V染色体分别携带高磷吸收率和磷利用率的基因。通过染色体工程技术,可以将携带耐低磷胁迫基因的外源染色体片段导入普通小麦,为小麦耐低磷胁迫育种和了解植物耐低磷胁迫的分子机理奠定基础。  相似文献

为了提高黄淮海麦区小麦育种材料的赤霉病抗性,采用分子标记辅助选择的方法,将来自望水白的4个抗赤霉病主效QTL 3B-QTL、4B-QTL、5A-QTL和6B-QTL导入不同的感病背景中,在后代BC1F3和BC1F4株系中评价它们的抗病效应和农艺性状回复情况。结果表明:(1)导入4个抗病QTL株系的平均病小穗率和病粒率分别为12.2%和6.3%,而受体亲本则分别达到59.1%和44.2%,抗病性显著提高;(2)病小穗数和病粒率与穗长及株高极显著负相关,但与可育小穗数、百粒重、旗叶长和旗叶宽等农艺性状指标没有显著相关性。因此,通过导入抗病主效QTL可以显著改善感病材料的抗性,为进一步选育高产抗病品种提供基础材料。不良农艺性状的紧密连锁阻碍着抗赤霉病主效QTL的高效利用,需要通过继续回交或与其他品种杂交来打破这种遗传连锁关系。  相似文献

The problem of chemically synthesized nanoproducts motivated scientific community to explore ecofriendly methods of nanosynthesis. Diatoms belong to a group of aquatic, unicellular, photosynthetic microalgae have been scarcely investigated as a source of reducing and capping agents for nanosynthesis of pesticides and antibiotics. The present study reports a novel ecofriendly method for the fabrication of bioactive gold nanoparticles using locally isolated Nitzschia diatoms. The diatom-fabricated gold nanoparticles show characteristic ruby red colored with sharp absorbance peak at 529 nm. Electron microscopy confirmed irregular shape of gold nanoparticles, with average size of 43 nm and zeta potential of −16.8 mV. The effects of gold nanoparticles on diatom viability were investigated using light and electron microscopy. The mechanistic approach to shed light on how diatoms reacted after exposure to gold metal salt revealed that exposure to gold chloride triggers elevated levels of catalase and peroxidase (12.76 and 14.43 unit/mg protein, respectively) to relieve reactive oxygen species (ROS) stress induced by gold salt exposure. Investigation studies on mechanisms behind Nitzschia-mediated gold nanoparticles fabrication outlined the role of diatom proteins, polysaccharides in reduction, and stabilization of nanoparticles as confirmed by FT-IR analysis. Bioactivity of gold nanoparticles was accessed by coupling them with antibiotics (penicillin and streptomycin), which increased their antibacterial activity compared to individual nanoparticles and antibiotics (Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus). Overall, the present novel phyco-nanotechnological approach is a promising tool to be used as sustainable strategy in green nanotechnology as well as to reduce use of antibiotics in microbial control.  相似文献

甜荞是异花授粉作物,自然界中的甜荞种质资源多存在生物学混杂现象。每份甜荞种质资源可以分化出若干份新的遗传资源类型,其遗传背景复杂。简化甜荞种质资源的遗传背景,准确描述新品种的亲子代关系和遗传背景一直是世界荞麦工作者面对的技术难题。甜荞种质资源的繁育需要采取隔离措施,实施过程中需要考虑甜荞花粉粒大小、甜荞授粉机制、访问昆虫种类以及种子量多少等。为解决上述问题,本研究建立了以隔离杂交圃、隔离单繁圃、隔离扩繁圃、隔离比较圃和田间试验圃的荞麦繁育新方法——"五圃法",以期开发出不同类型的新的荞麦资源,在育种中可针对性的配制强优势组合,为新品种选育提供种质材料。  相似文献

Paleo-sea-surface temperatures in the northeastern- and southeastern-parts of the Japan Sea were reconstructed for the last 160 kyr using alkenone temperatures (UK37-temperatures). UK37-temperatures at two sites show distinct glacial–interglacial changes during the last 160 kyr except for the interval corresponding to middle MIS 3 to MIS 2. On orbital-timescales, UK37-temperature tends to be high during MIS 5e, MIS 5c, and MIS 5a, which coincides with the intervals of stronger East Asian summer monsoon activity. The amplitude of temperature fluctuations in the Japan Sea is significantly higher than those in the neighboring seas. We suggest that the SST variation was amplified by the increasing source water (Kuroshio water) temperature and the changes in the volume transport of the Tsushima Warm Current (TWC) and/or the north–south oscillation of the sub-polar front position within the Japan Sea. Millennial-scale temperature fluctuations in the Japan Sea show that the temperature at the northern site was higher than that at the southern site during warmer periods of MIS 5, which is called “temperature reversal.” By analogy with modern oceanography, the temperature reversal could reflect the enhanced volume transport of the TWC and the spatial relationship between the studied site and the branches of the TWC, which is an essential factor in north–south temperature reversal around the eastern Japan Sea. Temperature drops were found at 114 ka, 111 ka, 93 ka, 87 ka, and 77 ka in MIS 5. Those events were associated with an increase in organic carbon and alkenone contents and can be correlated with the abundance peaks of ice-rafted debris (IRD) at Site GH05-1208 in the northern Japan Sea, suggesting that the surface water was cooled by enhanced mixing and consequent upwelling in a stronger winter monsoon regime.  相似文献

A permeabilized rat adipocyte model was developed which permitted an examination of: 1) insulin receptor autophosphorylation, 2) phosphorylation of a putative insulin receptor substrate of 160 kDa, pp160, and 3) the dephosphorylation reactions associated with each of these phosphoproteins. Rat adipocytes, preincubated with [32P]orthophosphate for 2 h, were exposed to insulin (10(-7) M) at the time of digitonin permeabilization. Phosphorylation of pp160 and autophosphorylation of the insulin receptor increased as a function of Mn2+ concentration in the media with near maximum responses at 10 mM. Maximum response was at least as large as the intact cell response to 10(-7) M insulin. In contrast, magnesium did not increase phosphorylation of pp160 although an increase in receptor autophosphorylation was observed. Autophosphorylation was preserved at digitonin concentrations of 20-100 micrograms/ml, but pp160 phosphorylation was negligible beyond 40 micrograms/ml. Our previous work demonstrated that the insulin receptor was associated with a phosphotyrosine phosphatase activity in permeabilized adipocytes (Mooney, R., and Anderson, D. (1989) J. Biol. Chem. 264, 6850-6857). The current permeabilized adipocyte model made possible an examination of the effects of phosphotyrosine phosphatase inhibitors, including several divalent metal cations (Zn2+, Co2+, and Ni2+), vanadate, and molybdate on both net phosphorylation of pp160 and autophosphorylation of the insulin receptor. Zn2+ at 100 microM, Ni2+ at 1 mM, and Co2+ at 1 or 5 mM increased insulin-dependent phosphorylation of pp160 at least 5-fold and autophosphorylation 2-fold. At higher concentrations of Zn2+ (1 mM) and Ni2+ (5 mM), however, no increase in phosphorylation of pp160 was observed and autophosphorylation was inhibited. Vanadate (1 mM) and molybdate (100 microM) increased insulin-dependent phosphorylation of pp160 by 3-fold when tested separately and 7-fold in combination. Insulin receptor autophosphorylation was increased 50% by each and 3-fold when the agents were combined. Dephosphorylation of pp160 and the insulin receptor was analyzed directly by permeabilizing prelabeled insulin-treated adipocytes in the presence of EDTA (10 mM). Dephosphorylation of pp160 was especially rapid with a t1/2 of approximately 10 s. The t1/2 for the insulin receptor was 37 s. Zn2+ at 1 mM (a concentration that inhibited the insulin receptor kinase) was a strong inhibitor of dephosphorylation, prolonging the rate of pp160 dephosphorylation more than 12-fold and insulin receptor dephosphorylation 3-fold.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

In a recent article in this journal, Ethology, 2014 present arguments as to why fighting in piglets during suckling may not be an epiphenomenon as we had previously suggested Ethology, 119, 2013, 353 and provide several alternative explanations. Although the new information Skok and ?korjanc bring to this debate is carefully considered and important, I argue here that it is not sufficient to counter our original claim that fighting among piglets (and kittens) during suckling may be an epiphenomenon, a largely inconsequential by‐product, of developmental processes anticipating the use of these behaviors and associated anatomical structures in later functional contexts. However, the points Skok and ?korjanc raise contribute importantly to identifying areas where further information and experimental work are needed to resolve this and related behavioral issues. I therefore conclude that this debate, independent of any particular final outcome, constructively points to the general difficulty and need for caution in interpreting behavior in seemingly ‘obvious’ functional terms, and particularly during development.  相似文献

Wild‐type p53 functions as a tumour suppressor while mutant p53 possesses oncogenic potential. Until now it remains unclear how a single mutation can transform p53 into a functionally distinct gene harbouring a new set of original cellular roles. Here we show that the most common p53 cancer mutants express a larger number and higher levels of shorter p53 protein isoforms that are translated from the mutated full‐length p53 mRNA. Cells expressing mutant p53 exhibit “gain‐of‐function” cancer phenotypes, such as enhanced cell survival, proliferation, invasion and adhesion, altered mammary tissue architecture and invasive cell structures. Interestingly, Δ160p53‐overexpressing cells behave in a similar manner. In contrast, an exogenous or endogenous mutant p53 that fails to express Δ160p53 due to specific mutations or antisense knock‐down loses pro‐oncogenic potential. Our data support a model in which “gain‐of‐function” phenotypes induced by p53 mutations depend on the shorter p53 isoforms. As a conserved wild‐type isoform, Δ160p53 has evolved during millions of years. We thus provide a rational explanation for the origin of the tumour‐promoting functions of p53 mutations.  相似文献
