Cultures of the obligate psychrophilic diatom Fragilariopsis cylindrus (Grunow) were grown for 4 months under steady-state conditions at −1 °C and +7 °C (50 μmol photons m−2 s−1) prior to measurements in order to investigate long-term acclimation of photosynthesis to both temperatures. No differences in maximum intrinsic quantum yield of PS II (FV/FM) and relative electron transport rates could be detected at either temperature after 4 months of acclimation. Measurements of photosynthesis (relative electron transport rates) vs. irradiance (P vs. E curves) revealed similar values for relative light utilization efficiency (α = 0.57 at −1 °C, α = 0.60 at +7 °C) but higher values for irradiance levels at which photosynthesis saturates (EK) at −1 °C and, therefore, higher maximum photosynthesis (PMAX = 54 (relative units) at −1 °C, PMAX = 49 at +7 °C). Nonphotochemical quenching (NPQ) measurements at 385 μmol photons m−2 s−1 indicated higher (37%) NPQ for diatoms grown at −1 °C compared to +7 °C, which was possibly related to a 2-fold increase in the concentration of the pigment diatoxanthin and a 9-fold up-regulation of a gene encoding a fucoxanthin chlorophyll a,c-binding protein. Expression of the D1 protein encoding gene psbA was ca. 1.5-fold up-regulated at −1 °C, whereas expression levels of other genes from Photosystem II (psbC, psbU, psbO), as well as rbcL, the gene encoding the Rubisco large subunit were similar at both temperatures. However, a 2-fold up-regulation of a plastid glyceraldehyde-P dehydrogenase at −1 °C indicated enhanced Calvin cycle activity. This study revealed for the first time that a polar diatom could efficiently acclimate photosynthesis over a wide range of polar temperatures given enough time. Acclimation of photosynthesis at −1 °C was probably regulated similarly to high light acclimation.  相似文献

The effects of temperature and larval density on survival of larvae, growth rate, age at pupation, and adult size (measured as wing length and dry weight) of laboratory-reared Anopheles gambiae (Diptera: Culicidae) were studied. Larvae were reared at three temperatures (24, 27 and 30°C) and three densities (0.5, 1 and 2 larvae/cm2). The effects of density and temperature strongly interacted to determine the mosquitoes' life-history parameters. Survival was highest at the intermediate temperature of 27°C. The differences between the temperatures increased with increasing density. At 30°C survival decreased as density increased, but at 27°C increasing density led to higher survival. Age at pupation increased as temperature decreased from 30°C to 24°C and as density decreased from 2 to 0.5 larvae/cm2. Adult size also increased as temperature decreased, but showed a negative correlation with density only at 27°C. In contrast, at 24°C and 30°C a decrease in density led to a decrease in adult size. Growth rate showed a similar pattern. At 27°C growth rate decreased as density increased, but at other temperatures the opposite trend was observed.  相似文献

Cell suspension cultures of Catharanthus roseus G. Don, Glycine max (L.) Merr. and Nicotiana tabacum L. were incubated with [14C]acetate, [14C]oleic acid and [14C]linoleic acid at five different temperatures ranging from 15 to 35° C. When the incubation temperature was increased, [14C]acetate was incorporated preferentially into [14C]palmitate, with a concomitant drop in [14C]oleate formation. Between 15 and 20° C, [14C]oleic acid accumulated in C. roseus cells. In all cultures, optimum desaturation of [14C]oleic acid to [14C]linoleic acid occurred between 20 and 25° C, and in G. max this was also the optimal range for desaturation of [14C]linoleic acid to [14C]linolenic acid. Elongation of [14C]palmitic acid was inhibited when cultures grown at 15° C for 25 h were subsequently incubated with [14C]acetate at 25° C. [14C]oleic acid accumulated in G. max and C. roseus cultures grown at 35° C for 25 h and subsequently incubated at 25° C. Desaturation of [14C]oleic acid increased up to 25° C, but then decreased or leveled off depending on the cell line and on the temperature prior to incubation.  相似文献

Winter rye (Secale cereale L. cv Musketeer) grown at 5 °C/250 µmol photons m–2 s–1 exhibited a relative reduction state of PS II comparable to that of rye grown at 20 °C but high light (800 µmol photons m–2 s–1) (1-qP = 0.32) whereas winter rye grown at 20 °C/250 µmol photons m–2 s–1 exhibited values of 1-qP ( 0.15) comparable to plants grown at 5 °C but low light (50 µmol photons m–2 s–1). The apparent size of the electron donor pool to PS I, estimated either in vivo or in vitro in the presence of methylviologen by A820 was positively correlated with the relative reduction state of PS II under the steady-state growth conditions. Immunoblotting of rye thylakoid polypeptides indicated that the relative contents of Lhcb1, Lhcb2, D1, Cyt f, PC, PsaA/PsaB heterodimer and the -subunit of ATPase complex exhibited minimal changes on a Chl basis. In contrast, a 2-fold increase in plastoquinone A content was associated with increasing growth irradiance at growth temperatures of either 5 or 20 °C. We suggest that the increases in the apparent size of the electron donor pool to PS I associated with rye grown at either 5 °C/250 µmol photons m–2 s–1or 20 °C/800 µmol photons m–2 s–1 may be explained by an increased thylakoid plastoquinone A content, coupled with possible enhanced PS I cyclic electron transport and/or increased capacity for electron donation from the stroma to the intersystem electron transport chain. The results are discussed with respect to photosynthetic adjustment to changes in PS II excitation pressure in winter rye.  相似文献

Branchipus visnyai is presently known from only one location, in Italy, on Monte Catabio (central Appennine, Latium, 1590 m a.s.l). It lives in a large astatic pond and in a small temporary pool close to the pond. Typically two generations appear in the pond (in spring after snowmelt and in autumn after dilution by rainfall), unless an exceptionally dry season occurs. Yet, due to shallowness and small size, filling and duration of the pool is erratic and depends on the annual amount of snow and rainfall. Consequently, B. visnyai does not always appear in the pool.Field studies suggest that either a rise in temperature or a drop in conductivity may trigger cyst hatching in the two biotopes. Also, the species exhibits a high tolerance to change in environmental factors like water temperature. Hatching occurs within a range of 10–22 °C, adults survive at temperatures from 1 to 27 °C. Carbon incorporation rates of Simocephalus vetulus were measured to study the effects of the physical state of the animals, size of the animal, varying temperature and light conditions. Physical state of the animal showed little effect on incorporation rates after the first hour. Incorporation rates increased in proportion to the third power of animal size. Experimental animals collected at temperatures of 12, 20 or 25°C fed maximally at 10, 15 and 25°C respectively, when subjected to a feeding temperature range of 5 to 30°C. We have interpreted this as an indication that S. vetulus is able to acclimate and incorporate maximally at various temperatures after prolonged exposure to that temperature. When fed over an irradiation range of 0 to 4.68 × 10–3 cal cm–2 s–1 incorporation rates were indirectly proportional to irradiance. This suggests a response to decreased irradiance in the weedy, littoral habitat of these animals.  相似文献

Synopsis The most important factor affecting the potential range of 14 non-native fishes in Florida appears to be their lack of tolerance to low temperatures. In this study, temperatures associated with reduction in feeding, cessation of feeding, loss of equilibrium and death were identified by decreasing water temperature 1°C day–1. Fishes tested and their mean lower lethal temperatures were: Astronotus ocellatus (12.9°C), Belonesox belizanus (9.7°C), Cichlasoma bimaculatum (8.9°C), C. cyanoguttatum (5.0°C), C. meeki (10.3°C), C. octofasciatum (8.0°C), C. trimaculatum (10.9°C), Clarias batrachus (9.8°C), Hemichromis bimaculatus (9.5°C), Hypostomus sp. (11.2°C), Tilapia aurea (6.2°C), T. mariae (11.2°C), T. melanotheron (10.3°C) and T. mossambica (9.5°C). Invertebrate offspring are usually larger in colder environments. To test for possible effects of covariates (e.g. maternal phenotype and feeding conditions) on this pattern, we performed a laboratory experiment to look at the effect of temperature on newborn weight in the planktonic crustacean Daphnia magna. Three tempèratures (12°C, 16°C and 22°C) and two food levels (10,000 cells ml–1 and 150,000 cells ml–1) were used, and offspring were examined from the first five clutches of mothers that had been maintained under the constant experimental conditions for three generations. Preliminary analysis suggested that newborn weight was significantly affected by temperature although patterns in the data were not clear cut. In addition, the covariates mother weight and clutch size were positively and negatively correlated with newborn weight, respectively; and later clutches tended to contain heavier offspring. Therefore, in an effort to control for the effects of the covariates, repeated-measures analysis of covariance was performed using ratio values of newborn weight/mother weight (relative newborn weight) as the dependent variable, clutch size as the covariate and clutch number as the repeated measures term. Now, temperature as a main effect in an ANCOVA model did not significantly influence relative newborn weight. The repeatedmeasure term clutch number also became nonsignificant, indicating that when differences in mother weight due to age were accounted for there were no overall differences in relative newborn weight between clutches from a particular mother. Temperature effects on relative newborn weight were only significant as part of interaction terms with food concentration and with clutch number. Thus there were different weight responses to temperature within food levels, and between clutch numbers within food levels. Under the low-food conditions newborn were heaviest at 16°C, lightest at 12°C and intermediate at 22°C. Conversely, under the high-food condition newborn were lightest at 16°C, heaviest at 12°C and again intermediate at 22°C. However, newborn tended to be heavier under the low food condition, and food concentration was highly significant as a main effect. Mother growth rate showed no significant relationship with newborn weight. It is concluded that direct temperature effects on relative newborn weight are marginal and nonsignificant. Temperature effects through interactions with food concentration and clutch number are important determinants of newborn weight, but relatively speaking account for only a small proportion of observed variance in newborn weight (25%), compared with the direct effect of food concentration (67%).  相似文献

Summary The fine structure of the indirect flight muscles was studied by electron microscopy in the following Notch locus mutants of Drosophila melanogaster reared at 18° C or 29° C for 6 days after eclosion: Ax 16172/Ax16172, Ax28/ Ax28, l(1)Nts1/l(1)Nts1,l(1)Nts1/Y and in wild-type controls. The flies were raised up to eclosion at 25° C or 18° C. It was observed that the l(1)Nts1 flies gradually became flightless within a few days if reared at 29° C as adults, and gross changes in the fine structure of the flight muscles were also observed in flies of this genotype. Peripheral myofilaments of myofibrils were disarranged and the mitochondria diminutive. At 18° C the flight muscles remained normal. In all of the Abruptex (Ax) combinations the flight muscles remained similar to the wild-type controls at both 18° C and 29° C, i.e. they were normal. The results suggest that the Notch gene is active in adult flies in addition to its activity during embryonic, larval and pupal stages, and is directly or indirectly involved in the adult development of the muscle tissue.  相似文献

We evaluated the combined effects of food (0.5 × 106, 1.0 × 106 and 2.0 × 106 cells ml−1 of Chlorella vulgaris) and temperature (15, 20 and 25 °C) on life history variables of B. havanaensis. Regardless of Chlorella density there was a steep fall in the survivorship of B. havanaensis at 25 °C. Both food level and temperature affected the fecundity of B. havanaensis. At any given food level, rotifers cultured at 15 °C showed extended but low offspring production. At 25 °C, offspring production was elevated, the duration of egg laying reduced and the fecundity was higher during the latter part of the reproductive period. The effect of food level was generally additive, at any given temperature, and higher densities of Chlorella resulted in higher offspring production. Average lifespan, life expectancy at birth and generation time were 2–3 times longer at 15 °C than at 25 °C. At 20 °C, these remained at intermediate levels. The shortest generation time (about 4 days) was observed at 25 °C. Gross and net reproductive rates and the rate of population increase (r) increased with increasing temperature and generally, at any given temperature, higher algal food levels contributed to higher values in these variables. The r varied from 0.11 to 0.66. The survival patterns and lower rates of reproduction at 15 °C suggest that the winter temperatures (10–15 °C) prevailing in many waterbodies in Mexico City allow this species to sustain throughout the year under natural conditions.  相似文献

Preimaginal development, mortality, aphid consumption rate, and size and weight upon reaching the adult stage of the aphidophagous coccinellids Hippodamia convergens Guérin-Méneville and Coccinella septempunctata L. collected from Karditsa, central Greece, were examined at four constant temperatures (14, 17, 20 and 23 °C) and L16:D8. The coccinellids fed on the tobacco aphid, Myzus persicae nicotianae Blackman. Egg, larval and pupal mortality was highest at 14 °C reaching 85.0, 73.8 and 29.4% in H. convergens and 49.3, 75.4 and 58.8% in C. septempunctata, respectively. Total preimaginal development ranged from 57.2 to 70.4 days at 14 °C, and to 16.9 and 22.1 days at 23 °C in H. convergens and C. septempunctata, respectively. Heavier and larger adults of H. convergens were obtained at 17 and 20 ° C. In C. septempunctata temperature did not affect adult weight while the lowest size was observed at 14 and 17 °C. Day-degrees requirements for preimaginal development in H. corvengens were 212.9 above a developmental threshold of 11.0 °C. The corresponding values for C. septempunctata were 281.5 and 10.7 °C. In H. convergens total and daily aphid consumption ranged from 46.8 aphids at 14 °C to 85.0 aphids at 23 °C and from 1.5 aphids at 14 °C to 9.2 aphids at 23 °C, respectively. The corresponding values for C. septempunctata were 112.0 and 2.7 at 14 °C and 157.7 and 12.4 at 23 °C. The results show the high potential of both predators as biological control agents against the tobacco aphid. The knowledge obtained could be essential for their appropriate use and for the improvement of mass rearing systems.  相似文献

To investigate the influence of temperature andholding time on the pyrolyzate yields of Chlorella protothecoides, the microalgal cells weresubjected to pyrolysis at 200, 300, 400, 500 and 600 °Cfor 5, 20, 60 and 120 min, separately. High oil yields above 40% dry weight cells wereobtained both at relatively low temperature (300 °C)with relatively long holding times (20–120min) and relatively high temperatures (400–500 °C)with relatively short holding times (5–20min). The maximum oil yield of 52.0% was achieved at500 °C for 5 min. The gas yield was generallyincreased with the increasing temperature and holdingtime. It could reach 63.3–76.0% at 600 °C.High pyrolytic rates of 72–87% were obtained at allexperiments except at 200 °C for 5–20 min or300 °C for 5 min. Thermogravimetric analysisindicated that the main thermal degradation of thismicroalga occurred at 200–520 °C. The resultsimply that C. protothecoides is a good candidatefor the production of renewable fuels by pyrolysis.  相似文献

The thermoregulatory characteristics of three species of Cryptomys from Zambia and Angola are examined and, together with published data on four other species of Cryptomys from southern Africa, used to determine whether scaling occurs in this genus of subterranean rodents. The thermoregulatory properties of acclimated giant Zambian mole-rats, Cryptomys mechowi ( =267 g), Angolan mole-rats, Cryptomys bocagei ( =94 g) and Zambian common mole-rats Cryptomys hottentotus amatus ( =77 g) are as follows. Mean resting metabolic rates (RMRs) within the respective thermoneutral zones were 0.60±0.08 cm3 O2 g-1 h-1 (n=12) for C. mechowi; 0.74±0.06 cm3 O2 g-1 h-1 (n=8) for C. bocagei and 0.63±0.06 cm3O2 g-1 h-1 (n=21) for C. h. amatus. The thermoneutral zones (TNZs) of all three species are narrow: 29–30°C for C. mechowi; 31.5–32.5°C for C. bocagei and 28–32° C for C. h. amatus. The increase in mean RMR at the lowest temperatures tested (15° C for C. mechowi, 18° C for C. bocagei and C. h. amatus) was 2.35, 2.2 and 3.82 times their RMR in the TNZ respectively. Body temperatures are low, 34±0.53° C (n=24) for C. mechowi, 33.7±0.32° C (n=20) for C. bocagei and 33.8±0.43° C (n=40) for C. h amatus. At the lower limit of thermoneutrality, conductances are 0.09±0.01 cm3 O2 g-1 h-1 °C-1 (n=30) in C. mechowi; 0.12±0.01 cm3 O2 g-1 h-1 °C-1 (n=20) in C. bocagei and 0.12±0.03 cm3 O2 g-1 h-1 °C-1 (n=32) in C. h. amatus. The range in mean body mass among the seven species of Cryptomys examined for scaling was 60 g (C. darlingi) to 267 g (C. mechowi). There is no clear relationship between RMR within the TNZ and body mass. The resultant relationship is represented by the power curve RMR=2.45 mass-0.259.  相似文献

Summary The pattern of aldehyde oxidase (AO) activity was determined in wing discs of Drosophila melanogaster larvae homozygous for the mutants apt 73n, Beaded, and vestigial (vg) in order to determine if reduction in field size in the pouch could be related to alterations of the wild-type AO pattern, as suggested by the Kauffman (1978) hypothesis. The pattern in wild-type discs was resolved into six areas for comparison with mutant discs. vg discs developed at 25° C showed restriction of the pattern into a small area on the anterior side of the disc, and comparison of vg and wild-type prepupal wings allowed positive identification of the AO pattern elements which remained. AO patterns in vg wing discs grown at 27°, 29°, and 31° C were progressively more complete and similar to wild-type, reflecting the reduction in cell death in discs grown at higher temperatures. These results show that cell loss during the third instar in vg development at 25° C is responsible for the alteration of the AO pattern, rather than field size reduction, and that determination of the pattern must take place much earlier than the time of its first appearance during the third larval instar, and before cell death in vg discs begins. Thus mutants acting at earlier stages will be necessary for further tests of the Kauffman hypothesis.  相似文献

Sampling and experiments were performed in brackish fishponds, located near the Ebrie Lagoon, Ivory Coast. Moina developed here at salinities up to about 4 g l-1. Mean embryonic and juvenile development times in experimental conditions were 0.82 and 0.80 day, respectively, with 2 juvenile stages at 30 °C (salinity of 3 g l-1; 29 µg chlorophyll-a l-1) and 1.22 and 1.37, with 2 stages for most individuals and 3 or 4 stages for some of them at 26 °C (2 g l-1 and 36 µg l-1). The corresponding Q10 for embryonic development was relatively high (2.7). Growth rates in weight at 30 ° C were 2.11 and 1.37 µg µg-1 d-1, respectively, for the embryonic and juvenile stages, 0.21 for mean somatic growth of the five first female adult stages, and 1.00 for primipara, summed for somatic growth, production of eggs, and embryonic growth. Consequently, differences in growth rates between young and adults were small. M. micrura also was a high fecundity species according to its size, fecundity in nature varying from 2.2 to 7.7 eggs per adult female. Rates of population increase evaluated for some sequences of parthenogenetic growing periods, reached up to 0.9. Most of the daily P/B evaluated by the cumulative growth method reached values above 1.0 (28 to 31 °C), minima being about 0.7 for lower temperatures (approximatively 26 °C). Populations were also characterized by low juvenile to adult ratios (down to 0.9) and high daily birth rates (up to 1.2). Overall, M. micrura is a highly productive, opportunistic species, well adapted to the low salinities that occur during part of the year in the ponds.Biological and population characteristics of this species, and literature data on regulation mechanisms, possible use for aquaculture, and on size in tropical species are discussed.  相似文献

Photosynthetic activity, in leaf slices and isolated thylakoids, was examined at 25° C after preincubation of the slices at either 25° C or 4° C at a moderate photon flux density (PFD) of 450 mol·m–2·s–1, or at 4° C in the dark. The plants used wereSpinacia oleracea L.,Cucumis sativus L. andNerium oleander L. which was acclimated to growth at 20° C or 45° C. The plants were grown at a PFD of 550 mol·m–2·s–1. Photosynthesis, measured as CO2-dependent O2 evolution, was not inhibited in leaf slices from any plant after preincubation at 25° C at a moderate PFD or at 4° C in the dark. However, exposure to 4° C at a moderate PFD induced an inhibition of CO2-dependent O2 evolution within 1 h inC. sativus, a chilling-sensitive plant, and in 45° C-grownN. oleander. The inhibition in these plants after 5 h reached 80% and 40%, respectively, and was independent of the CO2 concentration but was reduced at O2 concentrations of less than 3%. Methyl-viologen-dependent O2 exchange in leaf slices from these plants was not inhibited. There was no photoxidation of chlorophyll, in isolated thylakoids, or any inhibition of electron transport at photosystem (PS)II, PSI or through both photosystems which would account for the inhibition of photosynthesis. The conditions which inhibit photosynthesis in chilling-sensitive plants do not cause inhibition inS. oleracea, a chilling-insensitive plant, or in 20° C-grownN. oleander. The CO2-dependent photosynthesis, measured at 5° C, was reduced to about 3% of that recorded at 25° C in chilling-sensitive plants but only to about 30% in the chilling-insensitive plants. Methyl-viologen-dependent O2 exchange, measured at 5° C, was greater than 25% of the activity at 25° C in all the plants. The results indicate that the mechanism of the chilling-induced inhibition of photosynthesis does not involve damage to PSII. The effect of relative meal size (0.5–24% body mass) on specific dynamic action (SDA) was assessed in Chinese catfish (Silurus asotus Linnaeus) (30.90±1.30 g) at 25.0°C; the cutlets of freshly killed loach without viscera, head and tail were used as a test meal. There was no significant difference in either SDA duration or peak oxygen consumption (VO2) among low meal size ranges. But both increased linearly as meal size increased from 2 to 24% without reaching a plateau. Factorial metabolic scope was 5.92 in fish fed with 24% body mass, the highest documented feeding metabolic scope value in fish till now. The Peak VO2 of satiated meal size groups (175.85±10.55 mg O2 h−1) was above 80% of maximum metabolic rate during locomotion recovery process (215.48±7.07 mg O2 h−1). The relationship between energy expended on SDA (E) and energy ingested (I) was described as: E=0.0000432I 2+0.140I+2.12. The lowest value of SDA coefficient appeared at 2% body mass group.  相似文献

Rhodospirillum rubrum grown either chemotrophically or phototrophically at 14°C and 30°C, was employed to study the effect of temperature on fatty acid composition as well as on several membrane bound functions involved in energy metabolism. Upon growth at both temperatures the fatty acid composition of membranes showed differences, which could be attributed to an incomplete formation of photosynthetically active membranes rather than specifically to the growth temperature. Activities of NADH dependent respiration and light induced proton extrusion by cells did not show discontinuities in Arrhenius plots down to temperatures of 15°C and 5°C, respectively. In contrast, coupling factor Mg2+- and Ca2+-ATPase as well as succinate cytochrome c oxidoreductase showed significant breaks at 20°C and 18°C, respectively. Similarly, in Rhodopseudomonas sphaeroides. NADH dependent respiration and light induced proton extrusion by cells was continuous over the entire range of temperatures applied. ATPase as well as succinate cytochrome c oxidoreductase, on the other hand, featured discontinuities in Arrhenius plots at 20°C and 19°C. Summary We have isolated a mutant of the yeast Schizosaccharomyces pombe which exhibits sensitivity to UV light when grown at either 30° or 37°C, as compared to the parental wild-type strain. This increased sensitivity is more pronounced when cells are grown at 37°C. The mutant is also sensitive to 18 MeV electrons at the high temperature. Tetrad analysis of spores generated by crossing the mutant and a Rad+ strain revealed that sensitivity to both types of radiation cosegregate 2:2, relative to wild-type resistance, indicating that a single altered chromosomal locus is responsible for the radiation sensitivities observed. In addition, analysis of spores resulting from crosses between the mutant and all other known S. pombe rad mutants indicates that the temperature-dependent sensitivity described in this report is mediated by a mutation in a previously unidentified rad locus.  相似文献

Plastic enclosures (limnocorrals) were used to assess the impact of adding NO3 - to an oligotrophic pond. The pond, Mare's Egg Spring, is spring fed at a constant temperature of 4.5 °C (the main portion of the pool never exceeded 6 °C) and a molar ratio of N:P of 0.5. The pool is dominated by the cyanobacterium (blue green alga), Nostoc pruniforme, which is capable of nitrogen fixation. This, in combination with the low N:P ratio, suggested that non-nitrogen fixing primary producers were limited by nitrogen. Over a period of one month, growth and nitrogen fixation of Nostoc, suspended chlorophyll, sediment denitrification, and benthic diatom diversity were not dependent upon nitrate concentration. However, sedimentary chlorophyll levels increased slightly when NO inf3 sup- levels were increased from 0.71 µM (ambient) to 214 µM. Limnocorrals with slit sides gave water replacement rates between those for complete enclosure and those in open water. In the open channel, Nostoc growth was highest, and suspended chlorophyll the lowest. In the closed corrals, Nostoc growth was the lowest and suspended chlorophyll the highest, with intermediate values in the slit corrals. Therefore, short term increases of NO inf3 sup- levels in nitrogen poor aquatic systems do not necessarily affect the biological community, but slowing water replacement in small ponds may have significant effects.  相似文献
