Polymorphisms for melanic form of insects may provide various selective advantages. However, melanic alleles may have significant/subtle pleiotrophic “costs.” Several potential pleiotrophic effects of the W (=Y)‐linked melanism gene in Papilio glaucus L. (Lepidoptera) showed no costs for melanic versus yellow in adult size, oviposition preferences, fecundity, egg viability, larval survival/growth rates, cold stress tolerance, or postdiapause emergence times. Sexual selection (males choosing yellow rather than mimetic dark females) had been suggested to provide a balanced polymorphism in P. glaucus, but spermatophore counts in wild females and direct field tethering studies of size‐matched pairs of virgin females (dark and yellow), show that male preferences are random or frequency‐dependent from Florida to Michigan, providing no yellow counter‐advantages. Recent frequency declines of dark (melanic/mimetic) females in P. glaucus populations are shown in several major populations from Florida (27.3°N latitude) to Ohio (38.5° N). Summer temperatures have increased significantly at all these locations during this time (1999–2018), but whether dark morphs may be more vulnerable (in any stage) to such climate warming remains to be determined. Additional potential reasons for the frequency declines in mimetic females are discussed: (i) genetic introgression of Z‐linked melanism suppressor genes from P. canadensis (R & J) and the hybrid species, P. appalachiensis (Pavulaan & Wright), (ii) differential developmental incompatibilities, or Haldane effects, known to occur in hybrids, (iii) selection against intermediately melanic (“dusty”) females (with the W‐linked melanic gene, b+) which higher temperatures can cause.  相似文献

Diazotrophy of Rhodopseudomonas acidophila and Rhodopseudomonas capsulata was not obligatorily linked to photosynthesis. In the dark R. acidophila grew with dinitrogen as sole nitrogen source at a dissolved oxygen tension of 15 Torr (= 2.0 kPa); the doubling time was 8 h. Acetylene reduction by whole cells was more sensitive to oxygen in the light than in the dark. 16.5 mg N2 were fixed per g lactic acid consumed. R. capsulata synthesized nitrogenase and fixed dinitrogen in the dark at a dissolved oxygen tension of less than one Torr (= 0.13 kPa). Molecular methods are a necessary tool for sexing monomorphic birds. These molecular approaches are usually reliable, but sexing protocols should be evaluated carefully because biochemical interactions may lead to errors. We optimized laboratory protocols for genetic sexing of a monomorphic shorebird, the upland sandpiper (Bartramia longicauda), using two independent sets of primers, P2/P8 and 2550F/2718R, to amplify regions of the sex‐linked CHD‐Z and CHD‐W genes. We discovered polymorphisms in the region of the CHD‐Z intron amplified by the primers P2/P8 which caused four males to be misidentified as females (n = 90 mated pairs). We cloned and sequenced one CHD‐W allele (370 bp) and three CHD‐Z alleles in our population: Z° (335 bp), Z (331 bp) and Z″ (330 bp). Normal (Z°Z°) males showed one band in agarose gel analysis and were easily differentiated from females (Z°W), which showed two bands. However, males heterozygous for CHD‐Z alleles (Z′Z″) unexpectedly showed two bands in a pattern similar to females. While the Z′ and Z″ fragments contained only short deletions, they annealed together during the polymerase chain reaction (PCR) process and formed heteroduplex molecules that were similar in size to the W fragment. Errors previously reported for molecular sex‐assignment have usually been due to allelic dropout, causing females to be misidentified as males. Here, we report evidence that events in PCRs can lead to the opposite error, with males misidentified as females. We recommend use of multiple primer sets and large samples of known‐sex birds for validation when designing protocols for molecular sex analysis.  相似文献

Uncultivated microbial clades (‘microbial dark matter’) are inferred to play important but uncharacterized roles in nutrient cycling. Using Antarctic lake (Ace Lake, Vestfold Hills) metagenomes, 12 metagenome-assembled genomes (MAGs; 88%–100% complete) were generated for four ‘dark matter’ phyla: six MAGs from Candidatus Auribacterota (=Aureabacteria, SURF-CP-2), inferred to be hydrogen- and sulfide-producing fermentative heterotrophs, with individual MAGs encoding bacterial microcompartments (BMCs), gas vesicles, and type IV pili; one MAG (100% complete) from Candidatus Hinthialibacterota (=OLB16), inferred to be a facultative anaerobe capable of dissimilatory nitrate reduction to ammonia, specialized for mineralization of complex organic matter (e.g. sulfated polysaccharides), and encoding BMCs, flagella, and Tad pili; three MAGs from Candidatus Electryoneota (=AABM5-125-24), previously reported to include facultative anaerobes capable of dissimilatory sulfate reduction, and here inferred to perform sulfite oxidation, reverse tricarboxylic acid cycle for autotrophy, and possess numerous proteolytic enzymes; two MAGs from Candidatus Lernaellota (=FEN-1099), inferred to be capable of formate oxidation, amino acid fermentation, and possess numerous enzymes for protein and polysaccharide degradation. The presence of 16S rRNA gene sequences in public metagenome datasets (88%–100% identity) suggests these ‘dark matter’ phyla contribute to sulfur cycling, degradation of complex organic matter, ammonification and/or chemolithoautotrophic CO2 fixation in diverse global environments.  相似文献

In two races of European corn-borer moths (ECB), the E-race females emit and males respond to 99:1 sex pheromone blend of (E)/(Z)-11-tetradecenyl acetates, whereas the Z-race females and males produce and respond to the opposite 3:97 pheromone blend of (E)/(Z)-11-tetradecenyl acetates, respectively. We previously have shown that female production of the final blend ratio is under control of a major autosomal locus but that the sequence of male upwind flight responses to the blend is controlled by a sex-linked (Z-linked) locus. This sex-linked control of behavioral responses in crosses of E and Z ECB now is confirmed by use of sex-linked TPI (triose phosphate isomerase) allozyme phenotypes to determine the origin of the sex chromosomes in F2 populations. F1 males from reciprocal E × Z crosses generate similar behavioral-response profiles in wind-tunnel studies, with moderate numbers responding to the Z pheromone and intermediate blends (35%–65% Z), but very few responding to the E pheromone. The F2 behavioral-response profiles indicate that they are composed of 1:1 mixtures of hybrids and paternal profiles. Analysis of TPI allozyme differences allowed us to separate male F2 populations into individuals whose Z chromosomes both originated from their grandfathers, and individuals who had one Z chromosome originating from each grandparent. With these partitioned F2s, the TPI homozygotes exhibited behavioral-response profiles very much like their grandfathers, whereas the TPI hybrids produced response profiles similar to their heterozygous F1 fathers. These results demonstrate incontrovertibly that the response to sex pheromone in male ECB is controlled by a sex-linked gene that is tightly linked to the TPI locus and therefore is independent of the locus controlling pheromone blend production in females.  相似文献

Monema flavescens Walker (Lepidoptera: Limacodidae) is a multivoltine, generalist moth whose larvae cause serious damage to many types of trees. Pheromone lures prepared according to a study of a Japanese population were found to be ineffective at attracting M. flavescens nettle caterpillars in China, and some studies have shown intraspecific geographical differences in the composition of sex pheromones. We therefore reexamined the sex pheromone composition of M. flavescens in a Chinese population. In this study, the electroantennographically (EAG) active compounds in an extract from Chinese virgin females of M. flavescens were identified as (E)‐8‐decen‐1‐ol (E8‐10:OH), (Z)‐7,9‐decadien‐1‐ol (Z7,9‐10:OH), (Z)‐9,11‐dodecadien‐1‐ol (Z9,11‐12:OH), and (Z)‐9,11‐dodecadienal (Z9,11‐12:Ald) via coupled gas chromatographic‐electroantennographic detection (GC‐EAD) and coupled GC‐mass spectrometry (MS). Pheromone dimorphism might occur in this species, as this mixture of compounds in Chinese females was different from that of E8‐10:OH and E7,9‐10:OH extracted from Japanese females in previous research. In wind tunnel and field tests, the males were significantly attracted to a blend of the pheromone components E8‐10:OH, Z7,9‐10:OH, and Z9,11‐12:OH in a 100:5:4 ratio. The addition of Z9,11‐12:Ald did not change the male response. The optimized three‐component lure blend may provide a useful tool for monitoring and controlling Chinese populations of M. flavescens.  相似文献

In 2010, abrupt outbreaks of the African armyworm, Spodoptera exempta (Walker), occured on the Tarama, Iriomote and Kikai Islands in southwestern Japan. Analysis by gas chromatography-electroantennographic detection (GC-EAD) revealed two EAG-active compounds on male antenna in crude extract of virgin females. These compounds were identified as (9Z)-9-tetradecenyl acetate (Z9-14:Ac) and (9Z,12E)-9,12-tetradecadienyl acetate (Z9E12-14:Ac) in ca. 90:10 ratio by subsequent GC-MS analyses. (11Z)-11-Hexadecenyl acetate (Z11-16:Ac), which had previously been identified as a third component in the Kenyan population, was not detected. Binary blends of Z9-14:Ac and Z9E12-14:Ac at ratios between 99:1 and 90:10 showed a potent attractiveness in the field, superior to that of virgin females and comparable to that of the three-component formulation determined in Kenya. For the population survey, a 98:2 blend was used. In Tarama, only a few moths of S. exempta were captured with a light trap during the night when more than 600 males were captured with synthetic sex pheromone; more S. exempta captures with a light trap had been reported than with sex-pheromone traps in Kenya. This indicates that the Okinawan population has different properties from the Kenyan population in pheromone composition and behavioral response to light.  相似文献

Endogenous cytokinins were quantified in synchronized Chlorella minutissima Fott et Novákova (MACC 361) and Chlorella sp. (MACC 458) grown in a 14:10 light:dark (L:D) photoperiod. In 24 h experiments, cell division occurred during the dark period, and cells increased in size during the light period. Cytokinin profiles were similar in both strains, consisting of five cis‐zeatin (cZ) and three N6‐(2‐isopentenyl)adenine (iP) derivatives. Cytokinin concentrations were low during the dark period and increased during the light period. In 48 h experiments using synchronized C. minutissima (MACC 361), half the cultures were maintained in continuous dark conditions for the second photoperiod. Cell division occurred during both dark periods, and cells increased in size during the light periods. Cultures kept in continuous dark did not increase in size following cell division. DNA analysis confirmed these results, with cultures grown in light having increased DNA concentrations prior to cell division, while cultures maintained in continuous dark had less DNA. Cytokinins (cZ and iP derivatives) were detected in all samples with concentrations increasing over the first 24 h. This increase was followed by a large increase, especially during the second light period where cytokinin concentrations increased 4‐fold. Cytokinin concentrations did not increase in cultures maintained in continuous dark conditions. In vivo deuterium‐labeling technology was used to measure cytokinin biosynthetic rates during the dark and light periods in C. minutissima with highest biosynthetic rates measured during the light period. These results show that there is a relationship between light, cell division, and cytokinins.  相似文献

To date, information is wanting with regard to the use of new exotic parasitoids against olive fruit fly, Bactrocera (=Dacus) oleae (Gmelin) (Diptera: Tephritidae), a serious pest of olives Olea europaea L., in the Mediterranean basin. We investigated the oviposition response and developmental biology on B. oleae of Fopius (=Biosteres) arisanus (Sonan) (=Opius oophilus Fullaway) (Hymenoptera: Braconidae), an egg-pupal parasitoid of tephritid fruit flies, never tested before as a potential parasitoid of this host. Our results showed that olive fruits infested with B. oleae eggs exerted a relevant attraction to gravid F. arisanus and represented a stimulus for oviposition. Nevertheless they were not as attractive to female parasitoids as the Mediterranean fruit fly, Ceratitis capitata Wiedemann (Diptera: Tephritidae), eggs infested papaya fruits (Carica papaya L.). In our experimental conditions, F. arisanus completed development in B. oleae within 33 ± 1.7 days (males) and 35 ± 1.6 (females). Increases in host egg to female parasitoid ratios of 1:1, 5:1, 10:1 and 20:1 corresponded with decreases in the percentage of B. oleae parasitisation and host killing but corresponded also with increases in absolute parasitisation. Our findings are discussed in light of possibilities of utilising F. arisanus for biological control of olive fruit fly.  相似文献

Abstract

Free amino acids in EUGLENA. — The free amino acids present in Euglena gracilis strain Z, cultivated in the light and the dark, as well as in the strain bleached by streptomycin, have been identified by the conventional paper chromatographic method.

Variations have been observed when the photosynthetic apparatus is lost. Generally in the photosynthetic Euglena cells the pool of aminoacids reminds that of green algae, whereas bleached cells are similar to protozoa. It is possible that these variations are linked to the metabolic patways of green and bleached forms.  相似文献

We describe three coccoid green algal strains belonging to a new genus and species, Marinichlorella kaistiae Z. Aslam, W. Shin, M. K. Kim, W.‐T. Im et S.‐T. Lee, in seawater samples from the South Sea of Korea. These strains were maintained at 25°C–30°C under a 12:12 light:dark (L:D) photoregime in an ASN‐III medium at a pH of 7.5. These strains were tolerant of high salinity (7.5% NaCl) (w/v) and temperature (40°C). Molecular phylogenetic analyses using 18S rRNA gene sequence data resolved these organisms to a clade separate from green coccoid algae with similar morphology. The DNA–DNA hybridization results demonstrated very low relatedness of these organisms to phylogenetically related species of the genera Chlorella and Parachlorella. The molar guanine + cytosine content (G + C mol%) of the genomic DNA of these organisms ranged from 64.7 to 69.1 mol%. Based on molecular phylogeny, DNA–DNA hybridization, and other morphological studies, we propose a new taxon, Marinichlorella kaistiae, to describe these strains and classify them in the family Chlorellaceae. The Japanese giant looper, Ascotis selenaria cretacea, is a serious defoliator of tea gardens in Japan. The females produce racemic (Z,Z)-6,9-cis-3,4-epoxynonadecadiene (epo3,Z6,Z9-19:H, main component) and (Z,Z,Z)-3,6,9-nonadecatriene (Z3,Z6,Z9-19:H, minor component). The orientation of the males to the synthetic pheromone placed in a trap was strongly disrupted by Z3,Z6,Z9-19:H or a mixture of its monoepoxy derivatives (epoxydiene mixture, EDM) impregnated in septa and placed around the trap. Based on this result, polyethylene tubes containing Z3,Z6,Z9-19:H or EDM were prepared and effect of these dispensers was examined in a field. Disruption of male orientation to synthetic pheromone traps was achieved in orchards permeated with Z3,Z6,Z9-19:H at dispenser density of 3000 and 5000 tubes ha–1 (release rate: 0.55–0.61 mg day–1 tube–1) and with EDM at every tested dose, 250–5000 tubes ha–1 (release rate: 0.25–0.39 mg day–1 tube–1). Furthermore, disruption of mating in tethered females was observed in these orchards; particularly, the mating was perfectly inhibited in the areas treated with EDM at 3000 and 5000 tubes ha–1. This is the first formulation for the mating disruption of a geometrid pest.  相似文献

Analysis by gas chromatography with electroantennographic detection of extracts of pheromone glands derived from calling females of the sugarcane‐borer Diatraea flavipennella revealed two antennally active compounds. These components were identified as (Z)‐9‐hexadecenal (Z9–16:Ald) and (Z)‐11‐hexadecenal (Z11–16:Ald) by comparison of the retention times of the natural compounds and the synthetic compounds supported by two‐dimensional gas chromatography – time‐of‐flight mass spectrometric analysis and the positions of the double bounds in the chains were confirmed from the mass spectral fragmentation patterns of their dimethyldisulphide adducts. The analysis indicated that Z9–16:Ald and Z11–16:Ald were present in the sex pheromone in the proportions 25 : 75. Trace amounts of tetradecanal, hexadecanal, (Z)‐7‐hexadecenal (Z7–16:Ald), (Z)‐9‐hexadecen‐1‐ol and (Z)‐11‐hexadecen‐1‐ol were also found in the extract, but of these only Z9–16:Ald and Z11–16:Ald appeared to be antennally active. Behavioural bioassays demonstrated that a binary blend composed of Z9–16:Ald and Z11–16:Ald in the ratio of 25 : 75 induced a response in D. flavipennella virgin males similar to that elicited by live virgin females or by an hexane extract of the pheromone glands of calling females. Z9–16:Ald and Z11–16:Ald are, therefore, considered to be the major constituents of the female sex pheromone of D. flavipennella.  相似文献

Abstract The potato psyllid, Bactericera (= Paratrioza) cockerelli (?ulc) (Hemiptera: Triozidae), is a major pest of potato. Studies were conducted to determine the age at which both males and females reach reproductive maturity and the effect of age and time of day on sex attraction. Adult B. cockerelli reach reproductive maturity within 48 h post‐eclosion, with females being mature on the day of eclosion and males at 1 day post‐eclosion. Oviposition generally began 2 days after mating but was delayed when females mated within 2 days post‐eclosion. In laboratory olfactometer assays, the age of females used as odor sources and the age of males assayed to these odors did not affect sex attraction, with both younger (1–4‐day‐old) and older (8–10‐day‐old) males being attracted to females, regardless of female age (1–4‐day‐old or 8–10‐day‐old). Males assayed to live females at different times during the photophase (between 8:00 and 20:00 hours) were attracted to females between 11:00 and 17:00 hours, showing a temporal periodicity in sex attraction with B. cockerelli at least during the photophase.  相似文献

Fopius (= Biosteres) arisanus (Sonan) (= Opius oophilus Fullaway) (Hymenoptera: Braconidae) is an egg-pupal parasitoid of tephritid fruit flies. Small scale releases of F. arisanus were undertaken in coffee, Coffea arabica L., on the island of Kauai, to evaluate its potential for suppression of fruit fly populations. In conjunction with field releases, assays were conducted to determine the quality of pre-released parasitoids. In addition, the suitability of various sugars as potential food supplements in the field was evaluated. Spermathecal dissection showed that >70% of a total 1.7 million females released in 1998 and 1999 were successfully mated. Moreover, a sperm rating of 2.5–2.6 indicated that the spermathecae of inseminated females were 50% full. There were 85-137 matured eggs (mean =101.7±11.5) in the ovaries of mated or virgin females. When honey was replaced with ripe coffee and water, ripe coffee, or water alone, 10 days after emergence, females remained alive for another 5.5, 5.4, and 4.9 days, respectively. There was corresponding deterioration in the ovaries of gravid females. Mean eggs of honey-deprived females declined from >100 to <70 shortly before their death. Suitability test of various sugars showed that females lived longer when fed with honey (26 days), maple syrup (25 days) or molasses (20 days). Moreover, females provided with these sugars produced mean cumulative progeny per female of 77.2±19, 67±7, and 81±14, respectively, during a 2-week egging period. Our findings provided a basis for development of field release strategy for F. arisanus.  相似文献

In the fall armyworm, Spodoptera frugiperda (Lepidoptera, Noctuidae), two sympatric strains have been recognized that have been termed corn strain (C) and rice strain (R), referring to their most common host plants. Both strains are reproductively isolated via a distinct prezygotic barrier as well as via an intriguing postzygotic phenomenon: when R females have mated with C males, the resulting RC hybrid females exhibit dramatically reduced fertility independent of their mating partner. Here, we demonstrate that the reduced fertility is caused by the fact that these females refrain from mating, that is, females are behaviorally sterile. We identified a Z‐chromosomally linked sterility locus that is most likely incompatible with yet to be identified autosomal (or cytoplasmic) factors, leading to the observed sexual abstinence. Within‐chromosome mapping revealed the sterility locus to be located in an area of strongly reduced interstrain recombination.  相似文献

Life span differs between the sexes in many species. Three hypotheses to explain this interesting pattern have been proposed, involving different drivers: sexual selection, asymmetrical inheritance of cytoplasmic genomes, and hemizygosity of the X(Z) chromosome (the unguarded X hypothesis). Of these, the unguarded X has received the least experimental attention. This hypothesis suggests that the heterogametic sex suffers a shortened life span because recessive deleterious alleles on its single X(Z) chromosome are expressed unconditionally. In Drosophila melanogaster, the X chromosome is unusually large (~20% of the genome), providing a powerful model for evaluating theories involving the X. Here, we test the unguarded X hypothesis by forcing D. melanogaster females from a laboratory population to express recessive X‐linked alleles to the same degree as males, using females exclusively made homozygous for the X chromosome. We find no evidence for reduced life span or egg‐to‐adult viability due to X homozygozity. In contrast, males and females homozygous for an autosome both suffer similar, significant reductions in those traits. The logic of the unguarded X hypothesis is indisputable, but our results suggest that the degree to which recessive deleterious X‐linked alleles depress performance in the heterogametic sex appears too small to explain general sex differences in life span.  相似文献

Capillary gas chromatography indicated the presence of (Z)-7-dodecenyl alcohol and (Z)-7-dodecenyl acetate in the abdominal tip extracts of female Cornutiplusia circumflexa (L.) (Lepidoptera: Noctuidae: Plusiinae) moths. Gas chromatography combined with mass spectrometry and dimethyl disulfide derivatization confirmed the structure of these two principal pheromone components. In a flight-tunnel bioassay, a 5:1 blend of alcohol and acetate elicited the complete courtship sequence by males. The response was comparable to that evoked by a live virgin female, but males spent significantly longer time at calling females than at the synthetic source. The 5:1 blend was also attractive to male C. circumflexa in the field, as indicated by catches in traps baited with this mixture. (Z)-7-dodecenyl alcohol alone was slightly attractive whereas (Z)-7-dodecenyl acetate alone was completely inactive. C. circumflexa is the first reported Plusiinae species which utilizes (Z)-7-dodecenyl alcohol as the major pheromone component.  相似文献

Speciation is a continuous and dynamic process, and studying organisms during the early stages of this process can aid in identifying speciation mechanisms. The mallard (Anas platyrhynchos) and Mexican duck (A. [p.] diazi) are two recently diverged taxa with a history of hybridization and controversial taxonomy. To understand their evolutionary history, we conducted genomic scans to characterize patterns of genetic diversity and divergence across the mitochondrial DNA (mtDNA) control region, 3523 autosomal loci and 172 Z‐linked sex chromosome loci. Between the two taxa, Z‐linked loci (ΦST = 0.088) were 5.2 times more differentiated than autosomal DNA (ΦST = 0.017) but comparable to mtDNA (ΦST = 0.092). This elevated Z differentiation deviated from neutral expectations inferred from simulated data that incorporated demographic history and differences in effective population sizes between marker types. Furthermore, 3% of Z‐linked loci, compared to <0.1% of autosomal loci, were detected as outlier loci under divergent selection with elevated relative (ΦST) and absolute (dXY) estimates of divergence. In contrast, the ratio of Z‐linked and autosomal differentiation among the seven Mexican duck sampling locations was close to 1:1 (ΦST = 0.018 for both markers). We conclude that between mallards and Mexican ducks, divergence at autosomal markers is largely neutral, whereas greater divergence on the Z chromosome (or some portions thereof) is likely the product of selection that has been important in speciation. Our results contribute to a growing body of literature indicating elevated divergence on the Z chromosome and its likely importance in avian speciation.  相似文献

The cosmopolitan genus Bembidion is represented in New Zealand by 20 species, of which 19 are endemic; B. brullei appears to be a recent introduction. On phenetic characters the species fall into 7 subgenera, as follows: Zeplataphus n.subg.—maorinum Bates, dehiscens Broun, charile Bates, granuliferum n.sp., townsendi n.sp., tairuense Bates; Zeactedium Netolitzky—orbiferum Bates, musae Broun; Zeperyphodes n.subg.—callipeplum Bates; Zeperyphus n.subg.—actuarium Broun; Zemetal‐lina n.subg.—chalceipes Bates, solitarium n.sp., anchonoderum Bates, tekapoense Broun, wanakense n.sp., urewerense n.sp., hokitikense Bates, parviceps Bates; Ananotaphus Netolitzky—rotundicolle Bates; Notaphus Stephens—brullei Gemminger & Harold. The North Island population of maorinum is distinct from the typical South Island form in having reduced microscrulpture on the elytra, and is here separated as levatum n.ssp. An apparent geographic isolate of anchonoderum, represented by 2 females from Stewart Island, is provisionally recognised as stewartense n.ssp. The polymorphic complex within subg. Ananotaphus is here regarded as a single species, of which the North Island population is sufficiently distinct to warrant subspecific status as eustictum Bates; however, intergrades occur in the north‐west of the South Island. The following names fall into synonymy: latiusculum Broun (= maorinum); diaphanum Broun (= musae); nesophilum Broun (= callipeplum)’, tinctellum Broun (= chalceipes);antipodum Broun (= anchonoderum)’, tantillum Broun and probably attenuatum Broun (=hokitikense)’, clevedonense Broun and waikatoense Broun (= rotundicolle, ssp. eustictum)’, gameani Jeannel (= brullei).
