Submerged cultures of Pleurotus ostreatus, when supplemented with β-myrcene, produced perillene, a rare furanoid monoterpene with a unique flowery citrus-like flavour. The major volatile products of the conversion were identified from which a substantiated conversion pathway has been derived. The isomeric epoxides 1,2- and 3,10-epoxymyrcene were found among the first enzymatic oxidation products. An unusual opening of both oxirane rings to the E/Z isomers of α-acaridiol was thought to be key to the fungal formation of perillene. Further oxidation of α-(Z)-acaridiol via the corresponding hydroxyaldehydes and lactols resulted in perillene formation. A new natural compound, 3-(4-methyl-3-pentenyl)-2,5-dihydrofuran-2-ol (α,α-acarilactol), was among the transformation products.  相似文献

A β-tubulin gene (TUB1) from the basidiomycete Pleurotus sajor-caju was sequenced. TUB1 encodes a 446-amino-acid protein. The coding region is interrupted by 9 introns, all of which had a 5'-GTRNGT…YAG-3' sequence at the boundaries. Locations of the introns in TUB1 were common between the β-tubulin genes of other basidiomycetes, but not with animals, ascomycetes, or plants. This suggests that the introns were inserted independently into the β-tubulin gene after these divisions had diverged.  相似文献

Summary Two isolates of Pleurotus ostreatus from North America and one from Germany are interbreedable. Under identical conditions at low temperatures, their fruiting bodies are hard to distinguish. However, shape and colour and several other characters vary with culture conditions. The American stocks fruit well at temperatures from 4 to 24 °C, the German ones only below 15 °C. Four types of hybrids between German and American Pleupotus were obtained: i) The whole fruiting process is temperature sensitive as in German Pleurotus. ii) It proceeds at 4–24 °C as in American stocks, iii) Fruiting initiation is insensitive but sporophore development is sensitive to elevated temperatures, iv) Primordia formation and initial sporophore development depend on temperatures below 15 °C, but pileus expansion and spore discharge continue above 20 °. The involvement of separate genes for the single developmental steps and the use of temperature sensitivity for commercial varieties are discussed. One sporeless strain, F42x11, with considerable fruiting bodies has been obtained. In intrastock di-mon-matings this character was dominant.  相似文献

白黄侧耳Pleurotus cornucopiae微卫星间区(ISSR)分析   总被引:3,自引:0,他引:3  
本试验对我国1982年至2004年22年间栽培的白黄侧耳Pleurotus cornucopiae 30个菌株进行了锚定ISSR分析,试验表明,引物P4和P5都能对白黄侧耳P.cornucopiae进行多态性扩增,P4将供试菌株扩增出45个条带,大小在200~20000bp,P5将供试菌株扩增出39个条带,大小在500~15000bp,扩增出的条带100%具多态性。聚类分析在遗传相似性61%的水平下将30个供试菌株划分为15个类群,即15个具一定遗传差异的菌株;具有相同ISSR图谱、遗传相似性程度100%的可能为同一菌株,属于同物异名。试验表明我国的食用蕈菌野生环境面临人工栽培种质的污染,采自河北、山东、云南自然环境下的白黄侧耳P.cornucopiae与此前大量栽培的一些商业品种具完全相同的ISSR指纹图谱,聚类分析相似性系数100%。  相似文献

Supplementation of the key metabolite, alpha-(Z)-acaridiol, to stirred-tank cultured Pleurotus ostreatus was used to demonstrate that integrated in situ product recovery resulted in high conversion rates and quantitative separation of the target product perillene from the nutrient medium. The conversion of beta-myrcene by P. ostreatus was scaled-up from shake-flasks into a controlled, stirred tank bioreactor equipped with gas stripping and adsorption on a polystyrene fixed bed. The formation of the attractive flavour compound perillene was measured daily using standard controlled capillary gas chromatography. The formation of alpha-(Z)-acaridiol was the metabolic bottleneck of the conversion of beta-myrcene to perillene. Efficient in situ recovery of the volatile product enabled quantitative separation of the pure flavour compound. Appropriated bioprocessing, i.e. in situ separation of product, steadily shifted the metabolic equilibria and thus accomplished high conversion rate and pure product.  相似文献

Dimethyl sulfoxide (DMSO) is commonly used as a co-solvent to dissolve poorly water-soluble biologically active agents to assess their biological activities such as for enzyme induction. The question addressed was whether DMSO can be assumed to be an inert co-solvent. The influence of DMSO on the production of extracellular enzymes by Pleurotus ostreatus was investigated. DMSO functioned as either an inducer or a repressor, depending on the enzyme studied. The production of laccase and endo-1,4-β-xylanase increased by 29 and 250%, respectively, in presence of DMSO. However, DMSO repressed the activities of manganese peroxidase, β-glucosidase, β-xylanase, and endo-1,4-β-glucanase by 30, 33, 99 and 16%, respectively. These results raise concerns about the interpretation of bioactivity measurements when DMSO is assumed to function as an inert co-solvent to solubilize water-insoluble molecules. 糙皮侧耳(Pleurotus ostreatus)的AFLP指纹图谱分析   总被引:7,自引:0,他引:7

在对糙皮侧耳(Pleurotus ostreatus)的AFLP分析条件进行优化的基础上,利用该技术建立了14株产自不同地区的糙皮侧耳:DNA指纹图谱,并进行了数据分析。结果表明,在合成的14条引物的不同组合中,引物对E-3/M-3可以产生较多的DNA多态片段,E-AGC/M-CAT引物对的扩增效果最好,共获得184条DNA扩增带,其中多态性条带i101条,占54.89%。利用UPGMA法对所获数据进行聚类分析,计算得到糙皮侧耳菌株之间的遗传距离,发现不同品种间遗传距离差异较大,从0.192到0.754,说明糙皮侧耳的遗传多样性比较丰富。绘制的指纹分析树状图表明,14个糙皮侧耳菌株被分为6个组群,其中P17和杂3的相似性系数最高,达到了81.2%,而侧5与其他菌株的亲缘关系相对最远。  相似文献

以糙皮侧耳Pleurotus ostreatus菌株新831和豫6为材料,从这两个菌株的菌丝体中提取了糙皮侧耳病毒基因组,共有3个dsRNA片段,大小分别为8.2kb、2.6kb和1.1kb。采用菌丝尖端分离脱毒、原基组织分离脱毒和原生质体再生脱毒技术对糙皮侧耳菌丝体进行脱毒处理,利用dsRNA技术对脱毒效果进行检测。结果显示,原基组织分离脱毒后3个条带依然存在;菌丝尖端分离脱毒后,8.2kb和1.1kb 2个条带完全脱除,2.6kb的条带亮度有所减弱;原生质体再生脱毒后3个条带完全脱除;对3种脱毒技术得到的菌株进行生理生化指标测定,结果显示,原生质体再生脱毒菌株的菌丝生长速度、生物量、呼吸强度、纤维素酶活等均明显优于出发菌株、原基组织分离脱毒和菌丝尖端分离脱毒菌株;栽培结果表明,原生质体再生脱毒菌株前两茬菇的生物学效率达到96.4%-99.1%,比出发菌株提高19.9%-25.4%,并且菌盖宽度和长度有所增加,表明原生质体再生技术可以有效脱除糙皮侧耳菌株病毒,提高糙皮侧耳栽培产量。  相似文献

为研究微生物法预处理对红麻秸秆中木质素的降解及后续的红麻纤维素酶促糖化和发酵效率的影响,将白腐真菌Pleurotus sajor-caju接种在红麻秸秆培养基上固态培养,对红麻秸秆进行预处理。经P. sajor-caju培养25~35 d后,有效转化红麻秸秆中的木质素,转化率最高可达50.20%,并提高红麻纤维素的酶促水解效率,糖化率达69.33%~78.64%,与对照组相比提高了3.5~4.1倍。以微生物法预处理后的红麻秸秆样品为底物的同步糖化发酵实验表明,发酵72 h,发酵液中乙醇浓度达到18.35~  相似文献

Alternative substrates for cryopreservation at −20 °C have been little explored for basidiomycetes and could bring new possibilities of lower cost cryopreservation. Nevertheless, freezing temperatures between −15 and −60 °C are very challenging because they frequently result in cryoinjuries. The objective of this study was to evaluate substrates associated to cryoprotective agents for Pleurotus ostreatus cryopreservation at −20 or −70 °C in order to develop alternative techniques for basidiomycete cryopreservation. P. ostreatus was grown on potato dextrose agar or whole grains of oat, wheat, rice or millet and transferred to cryovials with cryoprotective solution with 1 % dimethyl sulfoxide, 5 % glycerol, 10 % saccharose, 4 % glucose, 6 % polyethylene glycol-6000 or 5 % malt extract. The mycelium in the cryovials were cryopreserved at −20 or −70 °C and recovered for evaluation of the mycelial growth viability after 1 and 3 years. Both substrates and cryoprotectants affect the viability of the mycelial growth cryopreserved at −20 or −70 °C; wheat grains combined with cryoprotectants such as saccharose or glucose are effective for keeping mycelium viable after cryopreservation at −20 °C for 1 or 3 years; for cryopreservation at −70 °C after 1 or 3 years, any substrate combined with any cryoprotectant is effective for preserving the mycelium viable, except for millet grains with polyethylene glycol after 3 years; semi-permeable cryoprotective agents such as saccharose and glucose are the most effective for cryopreservation at −20 or −70 °C for at least 3 years.  相似文献

Both enantiomers of aliphatic and aromatic cyanohydrins were hydrolyzed with the aid of Rhodococcus butanica ATCC 21197 to afford optically active α-hydroxy acids. Two water-soluble polysaccharide-protein complexes, extracted from Pleurotus tuber-regium sclerotia, were modified chemically to obtain their sulfated and carboxymethylated derivatives. While C6 position of glucan was fully substituted, C2, C3, and C4 were only partially substituted by sulfate groups. C3, C4, and C6 position of glucan were partially substituted during the carboxymethylation. Chain conformation and antitumor activity of the native samples and their derivatives were studied. The native samples and derivatives existed in sphere conformation, and showed potent in vitro antitumor activities. Water-solubility and introduction of ionic groups played important roles in enhancing the antitumor activities of the polysaccharide–protein complexes.  相似文献

本文选用刺芹侧耳Pleurotus eryngii 24号(广杏)为自交材料,对其自交S1代群体的质量性状(颜色、畸形、生长特性)和数量性状(形状、产量、大小、数目)进行了综合分析.分析结果表明:自交后代的出菇率为70%,自交导致后代群体菌丝平均生长速度和平均产量都低于亲本,16%的后代菌株单产高于亲本,最高单产达204.0g,高出亲本45.7%.木屑培养基上菌丝日均生长速度与产量无显著相关性(R=-0.028).均菇数目与单产成显著正相关性(R=0.543).大部分菌株(52%)菌盖颜色为亲本颜色(灰色),自交后代出现20%非亲本形状(棒状)菌株,畸形菇占19%.后代群体盖径、柄径、柄长的变异程度相当,差异性不显著.可根据育种目标对自交子代的较优菌株进行定向选择利用.  相似文献

The cauliflower mosaic virus (CaMV) 35S promoter has been most commonly used in plant transformation studies, but its activity in mushrooms has not been reported. p301-b is a binary vector containing a bialaphos resistance gene driven by the promoter of Lentinus edodes glyceraldehyde-3-phosphate dehydrogenase (GPD) gene. CaMV 35S-GUS was inserted into p301-b, and the resulting construct p301-bG was transformed to protoplasts of Ganoderma lucidum and basidiospores of Pleurotus citrinopileatus. GUS activity was observed in the transformants, indicating that CaMV 35S promoter can direct expression of exogenous gene in the mushrooms. This is the first report on the application of CaMV 35S promoter in genetic modification of mushrooms.  相似文献

β-D-Glucan, a polysaccharide isolated from an edible mushroom Pleurotus pulmonarius (Fr.) Quel., presented antinociceptive activity in mice. This study evaluated the involvement of transient receptor potential (TRP) channels and protein kinase C (PKC) on antinociceptive effect of a (1→3),(1→6)-linked β-D-glucan (GL) in mice. Intraperitoneal administration of GL potently inhibited nociceptive responses induced by intraplantar injections of capsaicin, cinnamaldehyde, menthol, acidified saline and phorbol myristate acetate (PMA). Moreover, Western blot analysis revealed that GL treatment also prevented PMA-induced PKC? activation. Collectively, present results demonstrate that GL could constitute an attractive molecule of interest for the development of new analgesic drugs.  相似文献

Hot aqueous extraction of the basidiocarps of the mushroom Pleurotus sajor-caju provided a cold water-soluble, gel-like glucan, which was characterized chemically, and its effects on RAW 264.7 cell line (mouse leukaemic monocyte macrophage) activation were determined. NMR spectroscopy, HPSEC, methylation analysis, and a controlled Smith degradation showed it to have a branched structure with a (1→3)-linked β-Glcp main-chain, substituted at O-6 by single-unit β-Glcp side-chains, on the average of two to every third residues of the backbone, with a molar mass of 9.75×10(5)gmol(-1). In macrophage cell culture, the β-glucan induced production of NO and the cytokines TNF-α, IL-1β, these effects being very similar as those of Escherichia coli serotype 0111:B4 Sigma-Aldrich lipopolysaccharide (LPS), although not modifying the response of LPS-activated macrophages. The results suggest that the (1→3), (1→6)-linked β-glucan from P. sajor-caju may have potential for immunological activities, although additional experiments are necessary for a better understanding of the mechanisms involved.  相似文献

变色圈试验证明平菇可以选择性优先降解稻草中的木质素,培养15d后,平菇对稻草中的木质素降解率为17.86%,对综纤维素降解率为2.44%,选择性指数为9.79。生料栽培平菇后,稻草中的木质素被降解50.24%。用气—质色谱(GC/MS)和红外光谱(IR)对木质素降解产物分析结果表明,平菇对稻草中木质素的降解效果十分明显,降解产物中检测出了大量含有苯环的小分子,证明木质素聚合体的降解首先发生在单体的侧链及单体间的连键上,发生Cα-Cβ、β-O-4等断裂,形成了单体。在进一步的降解过程中,平菇表现了其自身特有的降解机制,取代苯环单体上的甲氧基为甲基,而后发生苯环的开裂,这与报道的白腐菌降解过程有所不同。红外光谱分析中,平菇对木质素的降解明显,降解产物中含有很多木质素单体所特有的基团,如紫丁香基、愈创木基等,说明木质素的降解首先发生的是侧链的氧化反应。  相似文献

用从校内采集的P 96 6和从德阳市采集的P DY97两个侧耳属野生种与栽培种P 17之间的单核体进行配对具有不亲和性 ,证明了这 3个物种间遗传的独立性。同时通过P 17,P 96 6 ,P DY97各自的单核体自交实验 ,表明这 3个物种的交配系统是双因子控制系统。  相似文献
