Sclerotia of Botrytis cinerea were found to be a source of primary inoculum for bunch rot of grapes in the Hunter Valley of New South Wales. Conidia-bearing sclerotia were found on canes on grapevines. Resporulation of sclerotia was observed and sporogenic germination led to infection of flowers and berries. Sclerotia survived for eight months in untreated soil in the laboratory. Application of dicarboximides (procymidone, iprodione and vinclozolin) reduced infection of flowers and bunch rotcaused by the sporogenic germination of sclerotia.  相似文献

The flowering process in a female tree ofSalix tetrasperma was analysed by culturing its reproductive buds at different developmental stages during the dormant period on a chemically defined medium and examining the nature of sprouts produced by them. Buds at the upper eight nodes of the actively growing shoots developing in an acropetal sequence were cultured in separate lots. While all the buds collected from the 1st and 2nd nodes of the branches from the top downwards were vegetative and produced shoots, a considerable number of those collected from the 3rd and 4th nodes were reproductively determined and produced catkins. All the buds obtained from the 5th node and below were reproductive. Reproductive buds were cultured at regular time intervals during the dormant period. Freshly formed buds cultured in March during the spring growth flush produced catkins and were therefore reproductively determined. However, such a determination was not tantamount to flowering, as the floral meristems present in the axils of catkin bracts remained quiescent. Floral meristems of the buds cultured during April to August developed into small vegetative shoots. This was followed by the crucial period during September to December when the hitherto vegetative sprouts of the floral meristems showed a gradual transition into ovaries (female flowers) resulting in fertile catkins. Catkins produced from buds cultured in January and February produced well-developed ovaries.  相似文献

Summary The production of actinorhodin by Streptomyces coelicolor in a defined medium was examined using spore and vegetative inocula. The spore inoculum yielded higher concentrations of biomass and actinorhodin as well as a higher maximum specific growth rate compared with the vegetative inoculum. Nevertheless, the productivity of the batch culture for actinorhodin formation with vegetative inoculum was higher than that with spore inoculum.  相似文献

Waterlogged archaeological timbers of theMary Rose were shown to support a range of fungi, including marine soft rot fungi. These isolates, and other wood degrading fungi, were inactivated by gamma irradiation at doses of 3.1–15.0 kGy. No clear pattern of variation in radioresistance appeared between the Ascomycota, Basidiomycota and Deuteromycota. Terrestrial fungi were generally more resistant than marine fungi. Little variation in radioresistance was observed between vegetative hyphae and sporulating cultures/fruiting bodies. Sublethal doses resulted in reduced viability in some species. Gamma irradiation is suggested as a possible means of controlling fungal colonisation of waterlogged archaeological wood.  相似文献

Different methods to inoculate seedlings of Pinus pinaster and P. sylvestris with edible Lactarius species under standard greenhouse conditions were evaluated. Fungal inoculations were performed both under pure culture synthesis in vitro, followed by transplantation of acclimatized seedlings, and directly in the greenhouse using different techniques for inocula production (mycelial slurries, vegetative inoculum grown in peat-vermiculite and alginate-entrapped mycelium). In vitro inoculations with L. deliciosus produced thoroughly colonized seedlings. However, a sharp decrease in mycorrhizal colonization was detected on transplanted seedlings after 4 month's growth in the greenhouse. On the other hand, all the inocula applied directly in the greenhouse, except the alginate-entrapped mycelium, produced a variable number of mycorrhizal seedlings and colonization rates after the first growing season, depending on the plant-fungal combination and the inoculation method. Inoculations with vegetative inocula of the strain 178 of L. deliciosus were the most effective in producing mycorrhizal seedlings. All the seedlings inoculated with this strain were colonized although the colonization rates were relatively low. The commercial feasibility of the different inoculation methods for the production of seedlings colonized with edible Lactarius species is discussed.  相似文献

Several species of Botr yosphaeria are known to occur on grapevines, causing a wide range of disorders including bud mortality, dieback, brown wood streaking and bunch rot. In this study the 11 Botryosphaeria spp. associated with grapevines growing in various parts of the world, but primarily in South Africa, are distinguished based on morphology, DNA sequences (ITS-1, 5.8S, ITS-2 and EF1-α) and pathological data. Botryosphaeria australis, B. lutea, B. obtusa, B. parva, B. rhodina and a Diplodia sp. are confirmed from grapevines in South Africa, while Diplodia porosum, Fusicoccum viticlavatum and F. vitifusiforme are described as new. Although isolates of B. dothidea and B. stevensii are confirmed from grapevines in Portugal, neither of these species occurred in South Africa, nor were any isolates of B. ribis confirmed from grapevines. All grapevine isolates from Portugal, formerly presumed to be B. ribis, are identified as B. parva based on their EF1-α equence data. From artificial inoculations on grapevine shoots, we conclude that B. australis, B. parva, B. ribis and B. stevensii are more virulent than the other species studied. The Diplodia sp. collected from grapevine canes is morphologically similar but phylogenetically distinct from D. sarmentorum. Diplodia sarmentorum is confirmed as anamorph of Otthia spiraeae, the type species of the genus Otthia (Botryosphaeriaceae). A culture identified as O. spiraeae clustered within Botryosphaeria and thus is regarded as probable synonym. These findings confirm earlier suggestions that the generic concept of Botryosphaeria should be expanded to include genera with septate ascospores and Diplodia anamorphs.  相似文献

Basidiospores were isolated from the fruiting bodies of Ganoderma infecting oil palms from an estate in Johor and from ornamental palms (including oil palms) from Singapore. The spores were then germinated to obtain homokaryotic mycelia. Based on clamp connection formation in paired hyphal fusions, tester strains were identified from the homokaryons isolated. Compatibility tests were then carried out using these testers to determine the relatedness of the homokaryotic Ganoderma isolates, both from Johor and from Singapore. Results from the compatibility tests showed that Ganoderma from both locations belong to the same species, while the Ganoderma isolates from Singapore share some common alleles. The pathogenicity tests carried out on Chrysalidocarpus lutescens seedlings using inoculum growing on rubber wood blocks showed that dikaryotic mycelia can cause basal stem rot infection.  相似文献

Esca, a Grapevine Trunk Disease (GTD), is of major concern for viticulture worldwide. Our study compares the fungal communities that inhabit the wood tissues of vines that expressed or not foliar esca-symptoms. The trunk and rootstock tissues were apparently healthy, whether the 10 year-old plants were symptomatic or not. The only difference was in the cordon, which contained white rot, a typical form of esca, in 79% of symptomatic plants. Observations over a period of one year using a fingerprint method, Single Strand Conformation Polymorphism (SSCP), and the ITS-DNA sequencing of cultivable fungi, showed that shifts occurred in the fungal communities colonizing the healthy wood tissues. However, whatever the sampling time, spring, summer, autumn or winter, the fungi colonizing the healthy tissues of asymptomatic or symptomatic plants were not significantly different. Forty-eight genera were isolated, with species of Hypocreaceae and Botryosphaeriaceae being the most abundant species. Diverse fungal assemblages, made up of potentially plant-pathogenic and -protective fungi, colonized these non-necrotic tissues. Some fungi, possibly involved in GTD, inhabited the non-necrotic wood of young plants, but no increase in necrosis areas was observed over the one-year period.  相似文献

Of seven isolates tested for pathogenecity, all could infect ten species of citrus fruits and edible parts of five species of noncitrus crops. Recovery of quiescent filamentous fungi from non‐symptomatic grape berries and dormant buds demonstrated dominance of Alternaria, Aureobasidium, Cladosporium, Ulocladium and other dematiaceous hyphomycetes. Up to 78% of berries contained fungi prior to harvest. Botrytis cinerea was recovered from 0.2 to 0.5% of surface‐disinfested berries just subsequent to fruit set, and 1.6–4.8% of surface‐disinfested, over‐wintered dormant buds. In laboratory inoculations of mature grape berries with strains of Alternaria, Aureobasidium, Cladosporium, Ulocladium and Botrytis, only the latter was aggressive in rotting berry fruits. Inoculations with B. cinerea alone and in combination with strains of Alternaria, Aureobasidium, Cladosporium and Ulocladium recovered from grape demonstrated that prior occupation of wound sites by the latter fungi resulted in reduced lesion size compared to inoculation with B. cinerea alone.  相似文献

Mulberries are cultivated for different purposes: for feeding larvae of silkworm, Bombyx mori, as fresh and dry food resources, in wood instrument industry, in pharmaceutical industry and as outdoor ornamental trees in Iran. In recent years, twig and branch canker disease symptoms have been noticed on mulberry trees in northwestern parts of Iran. Diplodia isolates were repeatedly recovered from symptomatic tissues. Based on cultural and morphological features, the isolates were identified as Diplodia seriata. The identity of the isolates was further confirmed using sequence data from internal transcribed spacer (ITS)-rDNA and ef-1α gene. A phylogenetic analysis using ITS sequence data clustered the isolates obtained in this study together with known Diplodia seriata isolates of other woody hosts from GenBank. Inoculation studies carried out on white mulberry twigs using an excised shoot method revealed that the isolates are pathogenic on this host. D. seriata have been reported from other woody host plant species such as Juglans nigra and Vitis vinifera in Iran, however, to the best of our knowledge, the occurrence of D. seriata on mulberry trees is new for Iran. The distribution and reaction of different Morus spp. to D. seriata remain to be studied.  相似文献

Surface-sterilized sheared-root inocula of two vesicular-arbuscular mycorrhizal (VAM) fungi (Glomus intraradices and G. versiforme) from pot cultures associated with excised tomato roots showed significant sporulation and the production of an extensive hyphal biomass. As many as 102–103 axenic mature spores were recovered in Petri dishes during 3 months incubation in the dark. Propagules of both species were able to complete their vegetative life cycle in vitro and efficiently colonize Acacia albida roots after 1 month under greenhouse conditions. The effectiveness of 0.5 cm pieces of VAM roots as starter inocula indicates the high inoculum potential of intravesicle propagules.  相似文献

AIMS: The incidence of toxigenicity among Australian isolates of Aspergillus niger and Aspergillus carbonarius was assessed. Aspergillus rot and concomitant production of ochratoxin A (OA) in bunches inoculated with A. carbonarius were also investigated. METHODS AND RESULTS: Aspergillus niger and A. carbonarius were isolated from vineyard soils. Aspergillus niger was more widespread than A. carbonarius, and two restriction fragment length polymorphism types of A. niger, N and T, were present. Three of 113 A. niger isolates and all 33 A. carbonarius isolates produced OA. Aspergillus carbonarius was inoculated onto Semillon bunches with and without damage in the month before harvest. Damaged berries at greater than 12.3 (o) Bx were particularly susceptible to Aspergillus rot and production of OA, which was concentrated in severely mouldy berries. CONCLUSIONS: OA in Australian grapes results mainly from infection of berries by A. carbonarius. It is concentrated in discoloured, shrivelled berries. The potential for Aspergillus rot and OA production appears to commence after veraison and increase with berry damage and ripeness. SIGNIFICANCE AND IMPACT OF THE STUDY: Minimizing damage to grapes between veraison and harvest significantly reduces Aspergillus rot and OA formation. Monitoring the extent of Aspergillus rot in bunches infected with toxigenic Aspergillus spp. may give some indication of OA contamination.  相似文献

Botryosphaeriaceae are common dieback and canker pathogens of woody host plants, including stone fruit trees. In the present study the diversity of members of the Botryosphaeriaceae isolated from symptomatic wood of Prunus species (plum, peach, nectarine and apricot) was determined in stone fruit-growing areas in South Africa. Morphological and cultural characteristics as well as DNA sequence data (5.8S rDNA, ITS-1, ITS-2 and EF-1a) were used to identify known members and describe novel members of Botryosphaeriaceae. From the total number of wood samples collected (258) 67 isolates of Botryosphaeriaceae were obtained, from which eight species were identified. All species were associated with wood necrosis. Diplodia seriata (= "Botryosphaeria" obtusa) was dominant, and present on all four Prunus species sampled, followed by Neofusicoccum vitifusiforme and N. australe. First reports from Prunus spp. include N. vitifusiforme, Dothiorella viticola and Diplodia pinea. This is also the first report of D. mutila from South Africa. Two species are newly described, namely Lasiodiplodia plurivora sp. nov. from P. salicina and Diplodia africana sp. nov. from P. persica. All species, except Dothiorella viticola, caused lesions on green nectarine and/or plum shoots in a detached shoot pathogenicity assay.  相似文献

One of the greatest challenges in contemporary society is to reduce and treat household solid waste. The choice of inoculum to be used for start-up in reactors that degrade organic waste is critical to the success of organic waste treatment. In this study, the functional diversity, phylogenetic identification, and biogas production of bacterial communities from six inoculum sources were investigated. We used BIOLOG EcoPlates to evaluate the metabolic abilities of the bacterial communities, followed 16S rRNA gene sequence analysis to determine the phylogenetic affiliation of the bacteria responsible for carbon consumption. We observed great diversity in the physiological profiles. Of the six inocula tested, the sludge from an upflow anaerobic sludge blanket reactor (SRU) contained the most diverse, metabolically versatile microbiota and was characterized by the highest level of biogas production. By contrast, the sludge of the anaerobic lagoon (SAL) showed the worst performance in BIOLOG EcoPlates assays, but it exhibited the most diversity and generated the second largest amount of biogas. The bacterial isolates retrieved from BIOLOG EcoPlates were characterized as aerobic and/or facultative anaerobic, and were mainly Gram-negative. Phylogenetic analysis revealed that the isolates belonged to three major phyla: Proteobacteria, Firmicutes and Actinobacteria, represented by 33 genera. Proteobacteria exhibited the most diversity. The distribution of the bacterial genera differed considerably among the six inocula. Pseudomonas and Bacillus, which are able to degrade a wide range of proteins and carbohydrates, predominated in five of the six inocula. Analysis of the bacterial communities in this study indicates that both SRU and SAL microbiota are candidates for start-up inocula in anaerobic reactors. These start-up inocula must be studied further in order to identify their practical applications in degrading organic waste.  相似文献

The architectural development ofViburnum dilatatum andV. wrightii was investigated quantitatively. In both species, the major axis is developed from terminal buds of vegetative shoots and from axillary buds on the most distal nodes of reproductive shoots. The architecture of the two species is formed mainly by four branching patterns: a monopodial pattern (M), a sympodial pattern producing a pair of opposite daughter shoots (SP), a sympodial pattern producing a single daughter shoot (SS), and a pattern terminated with a dormant or dead bud (D). In the process of the architecture formation, four successive stages are recognized: 1) height growth, where the M pattern is dominant; 2) crown formation, in addition to the M pattern, the SP pattern occurs frequently; 3) crown expansion, the M and SP patterns are also frequent; 4) over mature, the M pattern becomes dominant again. These four stages are common to the two species, butViburnum wrightii proceeds with the crown formation stage more rapidly and stays in the crown expansion stage for a longer time thanV. dilatatum. The crown ofViburnum wrightii is thus more branched than that ofV. dilatatum.  相似文献

Massive flowering of tropical Phalaenopsis orchids is coordinated by the cold-induced release of reproductive bud dormancy. Light and temperature are the two key factors integrated by the dormancy mechanism to both stop and reactivate the meristem development of many other angiosperm species, including fruit trees and ornamental plants. It is well established that leaves and roots play a major role in inducing flower development; however, currently, knowledge of molecular events associated with reproductive bud dormancy release in organs other than the bud is limited. Using differential gene expression, we have shown that the leaves of a hybrid of Phalaenopsis species, Doritaenopsis ‘Tinny Tender’, undergo major metabolic modifications. These changes result in the production of sucrose and amino acids, both of which can sustain bud outgrowth, and auxin and ethylene, which may play important roles in awaking the dormant buds. Intake of abscisic acid and synthesis of the hormone jasmonate may also explain the inhibition of vegetative growth that coincides with bud growth. Interestingly, many genes that were upregulated by cold treatment are homologous for genes involved in flower induction and vernalization in Arabidopsis, indicating that processes regulating flowering induction and those regulating reproductive bud dormancy release may use similar pathways and effector molecules.  相似文献

Reestablishing native perennial plants and reducing invasive species are pivotal for many ecological restoration projects. The interactions among plant species, arbuscular mycorrhizal fungi, and soil P availability may be critical determinants of the success of native and non‐native plants in restoration and species invasions. Here we assessed mycorrhizal responsiveness for three late‐successional and three early‐successional plant species native to Rocky Mountain National Park and for the non‐native Downy brome, cheatgrass (Bromus tectorum L.) using field soil and commercial inoculum. Factorial greenhouse experiments were conducted to compare biomass of plant species with and without field soil and commercial inoculum treatments along a phosphorus (P) gradient, which ranged from ambient field levels to 12% of field levels, using dilutions of native soils. The two field soil inoculum treatments resulted in significant biomass differences for all species studied. Late‐successional species responded positively to field inoculum, whereas early‐successional species responded negatively. The two commercial inocula had low colonization rates (14 of 166 inoculated plants). The commercial inocula substrates had significant treatment effects on five of seven species included in the study in the apparent absence of mycorrhizal symbiosis. Soil P levels influenced mycorrhizal responsiveness in only one species, Smooth blue aster (Aster laevis L.). Our results show that, at least for the species studied here, locally collected field inoculum is the best choice for reestablishment of late‐successional native plant species.  相似文献

Safflower seedlings were used to discriminate two morphologically similar species of Phytophthora namely P. melonis and P. drechsleri . All isolates of P. melonis from different sources could not infect safflower seedlings under high inoculum potential whereas all isolates of P. drechsleri from various hosts attacked safflower within a short period. Of 31 authentic Phytophthora species inoculated to safflower seedlings only seven species including Phytophthora asparagi , Phytophthora cactorum , Phytophthora cryptogea , Phytophthora drechsleri , Phytophthora erythroseptica , Phytophthora palmivora and Phytophthora quercina caused hypocotyls infection. All cucurbit isolates of Phytophthora from different parts of Iran and one from China could not infect safflower seedlings and were identified as P. melonis which had been confirmed previously by molecular analysis.  相似文献

In this work, we studied the ecological interactions between grape berry microorganisms and Drosophila sp. flies involved in sour rot disease during grape ripening. After veráison the total microbial counts of grape berries affected by sour rot increased from about 2 log CFU/g of berries to more than 7 log CFU/g. Berry damage provoked a clear shift in yeast diversity from basidiomycetes to ascomycetous fermentative species. The latter were mostly Pichia terricola, Hanseniaspora uvarum, Candida zemplinina, and Zygoascus hellenicus. However, these species were not able to produce the metabolites characteristic of sour rot (gluconic and acetic acids) in inoculated berries. On the contrary, the acetic acid bacteria Gluconacetobacter saccharivorans produced high levels of these acids, mainly when berries were incubated in the presence of the insect Drosophila sp. Sour rot was not observed when grape bunches were physically separated from insects, even when berries were artificially injured. The wounds made in berry skin healed in the absence of insects, thus preventing the development of sour rot. Therefore, in the vineyard, the induction of sour rot depends on the contamination of wounded berries by a microbial consortium--yeasts and acetic acid bacteria--transported by drosophilid insects which disseminate sour rot among damaged berries. In the absence of these insects, plant defense mechanisms are effective and lead to skin healing, preventing disease spread. Thus, we showed that Drosophila sp. act as a vector for microorganisms associated with grape sour rot disease.  相似文献
