Summary A series of experiments was done to determine the best conditions for the extraction of protein from mould spores (Penicillium roqueforti). These experiments included studies on the influence of the extraction technique, the nature of the extracting fluid, spores glass: beads: extracting fluid ratio, and the time of extraction.Some work was done in fractionating the extractable nitrogenous substances and proteins of the spores. It was found that the extracted nitrogen could be fractionated into 12% dialysable low molecular weight nitrogen compounds and 88% crude protein, the latter was made up of about 40% albumens and 60% globulins.Visiting Research Professor, on scientific leave from National Research Center, Cairo, U.A.R.; Appointment supported by the International Cooperation Administration under the Visiting Research Scientists Program administered by the National Academy of Sciences of the United States of America. To investigate the nutritional value of the diatom Cyclotella cryptica Reimann, Lewin, and Guillard as an alternative feed for the use in the aquaculture industry, the heterotrophic growth characteristics, total fatty acids, and the resultant fatty acid profile of the microalga were studied when cultivated with sodium nitrate, ammonium chloride, or urea. All three nitrogen sources supported growth under heterotrophic conditions, and their uptake affected the pH of the cultivation medium, even when buffered. The use of sodium nitrate or urea resulted in a significant increase in the pH of the cultivation media, whereas the use of ammonium chloride caused a minor decrease in the pH of the cultivation media. The maximum specific growth rate was highest when urea and ammonium chloride were supplied at a low concentration; however, the total fatty acid content was not significantly affected (P = 0.101) by the nitrogen source when supplied at 10.7 mM nitrogen. The total fatty acid content and fatty acid profile of C. cryptica was more affected by the growth phase (predominately influenced by the initial nitrogen concentration) than by the source of nitrogen.  相似文献

Studies on the nutritive ecology of red colobus monkeys (Colobus badius preussi) in Korup National Park, Cameroon, investigated food plants and the parts eaten as well as nutritive values of the various food items. Food plants were identified through direct observations and collected for laboratory determination of nutrient content. Food plants were analysed for crude protein (CP), crude fibre (CF), ether extract (EE), nitrogen‐free‐extractive (NFE) and total ash (A). A total of seventeen species belonging to nine different families were identified as food plants for red colobus. Food and feeding habits showed that the species were highly folivorous, tending to prefer the young leaves of food plants. Lecomtedoxa klaineana (27.0%) and Xylopia aethiopica (22.2%) of families Sapotaceae and Annonaceae, respectively, constituted about 50% of total food items consumed. Results of nutrient composition indicated individual nutrients varied significantly (P < 0.01) with food species.  相似文献

Summary The influence of some physiological and environmental conditions on the growth and nitrogen fixation by a local strain of Nostoc commune revealed that the variation of hydrogen ion concentration of the nutritive medium, light intensity and temperature greatly affected the growth rate, cellular nitrogen and extracellular nitrogen of the experimental organism. It was possible however, to formulate the optimum conditions for growth and nitrogen fixation by this organism.  相似文献

A population growth model is considered for a one locus two allele problem with selection based entirely on fertility differences. A local stability analysis is carried out for the critical points — which include possible polymorphic states — of the resulting nonlinear differential equations. The methods of dynamical systems theory are applied to obtain limiting genotypic proportions for every initial state. Summary NIH3T3 cells grow in a serum-free basal nutrient medium supplemented with fibronectin, transferrin, insulin, epidermal growth factor (EGF) and high density lipoprotein (HDL). The individual omission from the serum-free medium of insulin, EGF, or HDL results in greatly reduced cell growth. These growth-restrictive conditions can be used to select for cells transformed with SV40, the polyomavirus middle T antigen gene, the activated humanras gene, and the mouse c-myc gene. Wild edible plants form an important constituent of traditional diets in the Himalaya. In the Sikkim Himalaya a total of 190 species have been screened as edible species out of which nearly 47 species come to the market. The present paper deals with nutritive values of 27 most commonly consumed wild edible plants in the Sikkim Himalaya. Of 27 plant species that were analyzed for their nutritive values, 22 were edible for their fruits and five for leaves/shoots. Among different plant parts, generally higher nutrient concentration was recorded for leaves, followed by new shoots and fruits. For different species the crude fiber content ranged between 2.15–39.90%, and the total soluble salts between 4.66–21.00%, and the vitamin C content from 6–286 mg/100 g. The fat content was determined high in the fruits ofCastanopsis hystrix, Machilus edulis, andCinnamomum species, while the protein content was highest inHippophae rhamnoides, Cucumis melo, andEleagnus latifolia. The total carbohydrate content ranged from 32–88% in the fruits of various wild edibles, the reducing sugar from 1.25–12.42%, total sugar from 2.10–25.09%, the lignin content varied from 9.05–39.51%, the hemicellulose between 25.63–55.71% and cellulose content varied from 9.57–33.19% in different species. Among the various macronutrients estimated in the plant samples of different wild edible species, nitrogen was present in highest quantity, followed by potassium, calcium, magnesium, phosphorus, and sodium. Micronutrients, such as iron, zinc, magnesium, and copper contents were analyzed in different plant parts of various wild edible species. The iron content was higher in leaves and new shoots. The nutritive values of certain wild edible species determined in this study are comparable with various commercial fruits. It is suggested that a few wild edible species need to be grown for commercial cultivation and adopted in the traditional agroforestry systems, which will lead to reduced pressure on them in natural forest stands as well as producing economic benefits for poor farmers.  相似文献

We consider the problem of quantitatively modeling movements of marked flea beetles in cultivated arrays of the cole crop, collards (Brassica oleraceae). 为生产合适的硒源提供一种思路,以菌丝体生物量、含硒量、还原糖、氨态氮和蛋白质为指标,采用四因素三水平正交设计法优化虎掌菌的富硒发酵条件,探讨不同浓度的硒对虎掌菌固体培养菌丝生长和液体培养产生的生物组分的影响。结果表明,高浓度的硒抑制虎掌菌菌丝体生长;正交试验选择不同的衡量指标,由极差分析得出各因素的影响程度大小,结合证实试验得到以还原糖为指标的最优组合为葡萄糖6%(质量分数)、酵母浸膏3%(质量分数)、KH_2PO_4 0.1%(质量分数)、Na_2SeO_3 0.6 mmol/L,其菌丝体生物量和氨态氮含量较高。与对照相比,加硒后虎掌菌发酵液中氨态氮、还原糖和总糖含量显著增加(P0.05);当硒浓度为0.5 mmol/L时,氨态氮、还原糖和总糖含量均达到最高值。菌丝体生物量和可溶性蛋白质分别在硒浓度为0.2 mmol/L和0.4 mmol/L时达到最大值。虎掌菌富硒培养后,发酵液的营养成分含量会发生变化。  相似文献

Summary Mycorrhizal and nonmycorrhizal plants of Panicum coloratum L. were grown in a factorial treatment design under two nitrogen levels and two clipping heights with an unclipped control. The nitrogen concentration in different plant components was determined following 9 weeks of growth under experimental conditions. Mycorrhizal infection increased green leaf and sheath nitrogen concentration by a relatively small, but significant percentage and had no effect on nitrogen allocation to the various plant components. Clipping increased leaf nitrogen concentration but inhibited growth to the extent that, when compared with the unclipped controls, less nitrogen remained in residual plant biomass with up to half of the total nitrogen allocated to offtake (the material removed by clipping). Plants receiving the higher nitrogen fertilization had higher tissue concentration of N and more N allocated to above-ground living tissues. Mycorrhizal infection interacted with clipping height and also with N availability significantly. Infection was unable to ameliorate the negative effects of the most severe clipping regime and of the low nitrogen availability on leaf and sheath N content. This is possibly due to mycorrhizal demand for carbohydrates competing with the carbohydrate requirement of roots for nitrogen uptake.  相似文献

Hepatotoxin production in cyanobacteria has been shown to correlate to external stimuli such as light and nutrient concentrations and ratios, although conflicting results have been reported. Specific growth rates and protein and microcystin content of M. aeruginosa PCC7806 and M. aeruginosa UV027 were determined under nonlimiting batch culture conditions for a range of medium nitrogen and phosphorous atomic ratios. Both strains exhibited a similar optimal medium N:P ratio for increased cellular microcystin levels. Additionally, total cellular protein content and intracellular microcystin content were significantly correlated to each other (r2 = 0.81, p < 0.001). Microcystin and protein content increased considerably as the maximum specific growth rate for the experimental conditions was reached. The significant correlation of cellular protein and microcystin content and their relative increase with increasing specific growth rate, within defined ranges of medium N:P ratios, suggest a close association between microcystin production and N:P ratio–dependent assimilation of nitrogen, and resulting total cellular protein levels, which may be further modulated by specific growth rate.  相似文献

土壤中氮素的吸收、转化及含量的变化是影响植被生长的关键因素。为探讨湿地植被不同退化状态对土壤氮组分含量和相关酶活性的影响,以及土壤氮组分含量与相关酶活性之间的关系,以甘南尕海湿地不同植被退化状态样地(未退化CK、轻度退化SD、中度退化MD和重度退化HD)为研究对象,采用野外采样与室内实验相结合的方法,分析了植被不同退化状态下不同形态氮组分(全氮、铵态氮、硝态氮和微生物量氮)含量的变化特征,以及土壤氮转化酶(蛋白酶、脲酶、硝酸还原酶和亚硝酸还原酶)活性之间的相关关系。结果表明:(1)在植被退化状态下,土壤含水量逐渐减小,土壤温度呈先减小后增大的趋势;(2)随着植被退化程度的加剧,硝态氮含量呈增加趋势,而全氮、铵态氮和微生物量氮含量均随退化程度加剧呈减小趋势;土壤蛋白酶活性随退化程度的加剧而减小,脲酶活性呈先减小后增大的趋势,重度退化活性最高,轻度退化最低;硝酸还原酶活性随退化程度的加剧而增加,亚硝酸还原酶活性表现为"升-降-升"的变化趋势,即轻度退化活性最高,未退化和中度退化较低;(3)土壤蛋白酶活性与全氮、铵态氮和微生物量氮呈极显著正相关关系(P<0.01),与硝态氮含量呈显著负...  相似文献

The effects of macronutrient balance on nutrient intake and utilization were examined in Manduca sexta larvae parasitized by Cotesia congregata. Insects fed an artificial diet having constant total macronutrient, but with varied ratios of protein and carbohydrate, with altered diet consumption in response to excesses and deficiencies of the individual macronutrients. Bivariate plots of protein and carbohydrate consumption for non-parasitized larvae demonstrated a curvilinear relationship between points of nutrient intake for the various diets, and the larvae grew best on carbohydrate-biased diets. The relationship was linear for parasitized larvae with the growth uniform across diets. On protein-biased diets, the larvae regulated the nitrogen content, containing similar amounts of nitrogen regardless of consumption. Efficiency of nitrogen conversion in non-parasitized larvae was greatest on carbohydrate-biased diets, while nitrogen conversion by parasitized larvae was greatest with intermediate nutrient ratios. Accounting for carbohydrate consumption, the lipid content decreased as dietary carbohydrate increased, but parasitized larvae contained significantly less lipid. The total biomass of parasites developing in individual host larvae was positively correlated with host protein consumption, but the individual parasites were similar in size. Parasitism influences host nutrient consumption in a manner that achieves uniform host growth under diverse nutritional regimes, thereby constraining blood nutrient concentrations within limits suitable for parasite growth and development.  相似文献

Summary The synthesis of protein and nucleic acids was studied by isotope incorporation and dilution in the plasmodia ofPhysarum polycephalum during periods of growth and differentiation (spherule formation). The total protein content decreased during starvation, but protein synthesis still occurred, probably at the expense of proteins previously synthesized during growth. Studies on leucine incorporation showed that protein synthesized during growth had a greater turnover than did protein formed by starving cultures, when both types of cultures were transferred to starvation conditions. Protein synthesis after prolonged starvation was rapidly and markedly decreased following the inhibition of RNA synthesis, whereas no such direct dependence on RNA synthesis was observed in growing cultures or during early starvation.The kinetics of RNA synthesis and the types of RNA formed were also shown to differ in growth and starvation. RNA turnover was low in growing cultures but substantial in starving cultures that were returned to growth medium. Qualitative differences in pulse-labeled RNA extracted from growing or starving cultures were revealed by methylated-albumin-kieselguhr column chromatography and sucrose gradient centrifugation. In starving cultures proportionately more labeled RNA was found in the lighter, non-ribosomal region of the gradient, and RNA from this region hybridized with denatured DNA to a greater extent than did other RNA fractions.This work was supported in part by Grant CA-07175 from the National Cancer Institute and by a grant from the Alexander and Margaret Stewart Trust Fund. Summary Forty-six cell cultures established from amniocentesis fluids were preserved in liquid nitrogen and later recovered from the frozen state with little loss of viability as compared to prefreeze viability. Five to 10% glycerol was found to be optimal for preservation in liquid nitrogen, and as few as 5×105 viable cells per frozen ampule could initiate cell growth. Storage in liquid nitrogen did not affect the genetic stability of glucose-6-phosphate dehydrogenase, lactate dehydrogenase, malic dehydrogenase, leucine aminopeptidase, acid phosphatase, or 6-phosphogluconic acid dehydrogenase isozymes of the amnion cultures. The total nitrogen and amino acid composition of seven Azolla strains were compared at four different growth phases. Total nitrogen content of the individual strains ranged from 2.6% to 5.7% of dry matter and was not significantly influenced by growth phase or population density. The concentration of the sixteen amino acids determined was maximal during the linear growth stage and specific differences occurred among Azolla strains. AnAzolla microphylla strain was the best source of amino acids and anA. filiculoides strain the poorest under the cultural conditions used. The chemical index score demonstrated the potential of some species, such as theA. microphylla strain, as contributor of protein for animals. Strains of other species, such asA. filiculoides, had several limiting amino acids and appear more suited for use solely as a green manure. All Azolla strains contained a similar proportion of essential (55%) and non-essential (45%) amino acids. Leucine, lysine, arginine and phenylanine+tyrosine were the predominant essential amino acids whereas the sulfur containing amino acids (methionine and cystine) were present in smaller amounts.  相似文献

Entomophthora virulenta was tested for growth and sporulation under various nutritional conditions in shake cultures containing combinations of dextrose and protein hydrolysates. Darkness, a temperature of 25°C, and a pH near 6.5 induced the formation of the greatest number of zygospores when the carbon and nitrogen sources were held constant. The influence of the total concentration of nutrients and of the ratio of the carbon and nitrogen sources depended on the nitrogen source used: The highest amount of spores under these conditions was produced with yeast extract as nitrogen source, a carbon/nitrogen source ratio of 2, and a 15% total nutrient concentration. The influence of the preinoculum and inoculum on sporulation is discussed.  相似文献

The possibility that c-phycocyanin serves as a nitrogen source in Spirulina platensis during nitrogen starvation was studied. The following evidence was obtained in support of this idea. 1. Under favourable conditions for growth, c-phycocyanin existed in large excess in the algal cells. 2. When the supply of nitrogen was low, about 30–50% of the c-phycocyanin disappeared without any effect on the maximal growth rate. 3. A culture which was deprived of nitrogen continued to grow unaffectedly for a period, the duration of which depended on the c-phycocyanin content in the cell before nitrogen starvation was initiated. 4. c-phycocyanin was the only nitrogenous compound that was depleted during the course of nitrogen starvation when growth was yet unaffected. 5. When protein synthesis was inhibited either by nitrogen starvation or by methionine sulfoximine (MSO), phycocyanin content began to decline immediately and growth continued at normal rates as long as c-phycocyanin did not decline below 50%. 6. The decrease in c-phycocyanin content during nitrogen starvation was accompanied by an increase in proteolytic activity.  相似文献

Two Caribbean strains (1651 and 1655) of the ciguatera-causing dinoflagellate Gambierdiscus toxicus were grown in xenic, batch culture under defined, measured nutrient conditions with nitrate, ammonium, urea, a mix of free amino acids (FAA), or putrescine as the nitrogen source. Cultures were maintained at 27 °C, salinity 35, 110 μmol m−2 s−1 (12 h:12 h light:dark cycle) on L2 medium at an initial nitrogen concentration of 50 μM N. Toxicity was determined using a ouabain/veratridine-dependent cytotoxicity assay (N2A assay) standardized to a ciguatoxin standard. Nitrate, ammonium, FAA, and putrescine supported growth, but urea did not. The appearance of ammonium in the organic nitrogen cultures indicated that G. toxicus and/or associated bacteria remineralized the available organic nitrogen. Both strains were exposed to nitrogen-limiting conditions as evidenced by chlorophyll a content per cell, nitrogen content, and nitrogen (N) to phosphorus (P) (N:P) ratio significantly declining once nitrogen was no longer available in the medium and cells entered stationary phase. Strain 1651 grew significantly faster than strain 1655 when nitrate, FAA, and putrescine was the nitrogen source, but not ammonium. Nitrogen source had no effect on growth rate (0.14 d−1) in strain 1651. The growth rate of strain 1655 (0.10–0.13 d−1) was significantly faster on ammonium than the other nitrogen sources. Strain 1655 was significantly more toxic (10-fold) than strain 1651 except when growing on ammonium at exponential phase. Toxicity ranged from 1.3 to 8.7 fg C-CTX1-Eq cell−1 in strain 1651 and from 30.7 to 54.3 fg C-CTX1-Eq cell−1 in strain 1655. Nitrogen source had no significant affect on toxicity. Toxicity was greater in stationary versus exponential phase cells for strain 1651 when grown on nitrate and strain 1655 regardless of nitrogen source. The difference in toxicity between growth phases may result from an increase in ciguatoxin and/or maitotoxin. Our results suggest that some strains of G. toxicus when associated with bacteria are able to take advantage of organic as well as inorganic nitrogen sources on short time scales to support future growth. The uncoupling of total nitrogen and phosphorus pools from conditions in the water column suggest that instantaneous growth rates can be supported by nutrients acquired hours to days earlier.  相似文献

Emerging evidence suggests that protein phosphatases play an important role in the growth and development of higher plants. We report here on the effects of okadaic acid and calyculin-A, two specific and potent inhibitors of the type-1 and type-2A families of serine/ threonine protein phosphatases, on the growth and development ofArabidopsis thaliana L. roots. Application of these drugs in nanomolar ranges arrested root hair growth, severely affected the shape of cells within the zone of elongation and inhibited root growth rates. Root hair elongation was inhibited by concentrations of okadaic acid and calyculin-A as low as 3 nM. The pleiotropic effects of okadaic acid and calyculin-A point to multiple functions for type-1 and -2A protein phosphatases in controlling root growth and development.We thank Kelly Aldrich for excellent technical assistance.
