Summary Extracellular amylase, lipase and protease produced by haploids, diploids and heterocaryons of Aspergillus nidulans were analysed. Three morphologically normal strains and 8 morphologic mutants as well as various genetic combinations of the 11 strains were examined in solid culture medium containing specific substrates. The enzyme production of each strain was determined by measuring the halo around the colony. It was observed that the colonies showing less growth also showed more alterations in enzyme production. The compact strains (BVIII and B6) and the slow-growing heterocaryons (pp+M32 and pp+M35) showed the highest enzymatic index for the three enzymes simultaneously. If colony growth is not considered, then for amylase and protease the highest values were reached by some diploid and heterocaryons and for lipase by one morphological strain. The results showed that morphological mutants and some combinations could be used for higher production of amylase, lipase and protease.  相似文献

Summary A high efficiency of Hordeum bulbosum-mediated haploid production in barley has been achieved using a floret culture technique in which florets pollinated with Hordeum bulbosum are cultured on modified N6 media containing 0.5 mg/l kinetin and 1.2 mg/l2,4-D. Cultures were maintained at 25 °C with a 16 h photoperiod for 9 days before embryo rescue. In a comparison of haploid production efficiency using five F1 hybrids from winter x winter and winter x spring barley crosses, 41.6 haploid plants/100 florets pollinated were produced using floret culture. Using detached tiller culture, 13.5 haploid plants/100 florets pollinated were produced. Higher efficiencies achieved with floret culture are attributed to the formation of larger, differentiated embryos. Such embryos lead to higher frequencies of plant regeneration. The F1 from a winter x winter cross was inferior in haploid production compared to F1s from winter x spring crosses. Summary Different methods of line development using doubled haploids in recurrent selection are presented. They are divided into two types: recurrent selection with progeny testing and recurrent selection on the phenotype of lines. It is shown that one of the best methods of line development is based on single doubled haploid descent recurrent selection. Only one line is studied per plant of the population, and the best lines are intercrossed. Using this method it is very easy to extract lines for variety development.  相似文献

Kleine Liebling is a haploid of unusual interest; not only is it a monoploid, but at the same time a successful horticultural variety. Analysis made of pollen mother cell meiosis revealed a very low frequency of bivalents, and evidence of neocentric activity among the univalents. Colchicine-induced diploid plants were synthesised, but contrary to expectation, microsporogenesis was very erratic, and pairing never complete. As in the haploid from which they were derived, a high proportion of dyads formed the final product of meiosis. It was concluded that both these irregularities were the result of mutations effecting the genetic control of meiosis.  相似文献

The efficiency of visual assessment for grain yield and its components in spring barley rows was examined using a number of assessors. In 20 out of 28 combinations of assessor and character, only 20 or less lines were needed to be retained from the set of 99 lines to save at least 50% of the best 10 lines. Assessments of tillers/row and to a lesser extent 1000-grain weight were generally more effective than assessments of yield/row and grain/ear. The low effectiveness of assessment of grains/ear was attributed to inadequate sampling of ears. Four out of five assessors showed bias towards assessment of tillers/row, the most easily assessed character, in their assessments of yield/row. Experienced barley workers were more successful in their assessments than others less familiar with the crop. Specially developed keys, aimed at making visual assessment more objective, generally had only small positive effects on the efficiency of assessment. Repeated assessments of characters by some assessors were consistent. It was concluded that visual assessment rather than direct measurement should be recognised as a basic tool of breeding in the early generations, using a large number of lines as a ‘safety net’ allowing the loss of some of the best lines.  相似文献

Different pretreatments were given to anthers of barley before culturing, and their effects assessed on the frequency of embryos and green doubled haploid plants produced. Mannitol pretreatment was better than cold pretreatment for some low responding cultivars. Optimal concentration of mannitol for pretreatment depended on cultivar. Low responding genotypes needed a higher concentration of mannitol than responsive ones. The addition of Ficoll to liquid medium increased the number of embryos and green plants. The influence of the growth regulators 2,4-D and TIBA was assayed using ten cultivars of barley grown in Spain. The anti-auxin TIBA gave good embryo production with some of the low responding cultivars. Two row-type cultivars always produced higher number of embryos and green plantlets than six row-type. The application of these modifications to 10 F1 hybrids with potential agronomic value, allowed the production of almost 1000 doubled haploid plants from only 3500 anthers. Up to two doubled haploid plants per flower were produced from the cross Monlon × Sonja. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Normal synaptonemal complexes have been found in haploid barley meiotic prophase at stages equivalent to pachytene in diploids. Reconstructions of serially sectioned nuclei have shown that up to 60% of the haploid chromosomes may pair in either intra- or interchromosomal associations. The extent and nature of the synaptonemal complex formation suggest that the chromosome pairing is non-homologous. From the virtual absence of chiasmata in metaphase I stages of the haploids it is inferred that crossing over requires a more precise DNA alignment than is provided by synaptonemal complex formation alone.  相似文献

Local populations of Cladophoropsis membranacea exist as mats of coalesced thalli composed of free-living haploid and diploid plants including clonally reproduced plants of either phase. None of the phases are morphologically distinguishable. We used eight microsatellite loci to explore clonality and fine-scale patch structure in C. membranacea at six sites on the Canary Islands. Mats were always composites of many individuals; not single, large clones. Haploids outnumbered diploids at all sites (from 2:1 to 10:1). In both haploid and diploid plants, genetic diversity was high and there was no significant difference in allele frequencies. Significant heterozygote deficiencies were found in the diploid plants at five out of six sites and linkage disequilibrium was associated with the haploid phase at all sites. Short dispersal distances of gametes/spores and small effective population sizes associated with clonality probably contribute to inbreeding. Spatial autocorrelation analysis revealed that most clones were found within a radius of approximately 60 cm and rarely further than 5 m. Dominance of the haploid phase may reflect seasonal shifts in the relative frequencies of haploids and diploids, but may alternatively reflect superiority of locally adapted and competitively dominant, haploid clones; a strategy that is theoretically favoured in disturbed environments. Although sexual reproduction may be infrequent in C. membranacea, it is sufficient to maintain both life history phases and supports theoretical modelling studies that show that haploid-diploid life histories are an evolutionarily stable strategy.  相似文献

Eight commercial varieties of spring barley (Getman, Tabora, Adagio, Galaktik, Europrestige, Korona, Nevada, and Stalker) were used for the in vitro culture establishment and the development of an efficient protocol for the induction of the callus formation and plant regeneration. Mature embroys of the above-mentioned genotypes were used as explants. The induction of callus formation, culture passage, and plant regeneration were carried out using a medium containing MS salts supplemented with casein hydrolysate (1 g/l), L-proline (690 mg/l), thiamine-HCI (1 mg/l), maltose (30 g/l), 2,4-D (2 mg/l), CuSO4 (12.5 mg/l), myoinositol (250 mg/1), and gelrite (3.5 g/l) at pH 5,6–5,8. The ability of callus formation was observed in all studied varieties, and the callus formation’s frequency varied from 65 ± 3.4% to 100%. The highest regeneration potential was revealed for the Korona (88 ± 2,8%), Europrestige (89 ± 6.5%), Tabora (93 ± 3.4%), Getman (99 ± 0.8%), and Nevada (100%) varieties. For all cultivars the generation of plants from a calli occurred via the organogenesis and somatic embryogenesis. The highest total regeneration potential was registered for the Getman variety (50 ± 5%), which was selected for the further development of effective genetic transformation protocols.  相似文献

Summary The relationship between in vitro performance of haploid embryos and the agronomic performance of the derived doubled haploid (DH) lines during the stages of field evaluation was investigated. The results showed a positive correlation between coleoptile height of haploid plantlets in culture and final plant height of their DH progeny lines in the field. These results illustrate that in vitro selection for plant height and other linked quantitative or pleiotropic characters can be carried out at the initial stages of a DH breeding programme.  相似文献

The effect of glucose concentration as a carbon source in the range of 5-20 g/L on the fermentative productions of intra-and extra-cellular ethanol, acetate, formate, oxalate, lactate, and pyruvate, as well as pyruvate decarboxylase in A. orientalis were investigated, depending on the incubation period. Intra-and extra-cellular pyruvate levels increased with rising glucose concentrations up to 15 and 20 g/L of glucose, respectively. In addition, intra-cellular pyruvate levels reached their maximum on the 48th hour in the range of 12.5-20 g/L of glucose, except for 5 and 10 g/L while extra-cellular pyruvate were at the 48th and 60th hours. As a fermentative end product, intra-and extra-cellular ethanol levels increased with increasing glucose concentrations of the growth medium and with incubation period. Activity of pyruvate decarboxylase, one of the key enzymes of the alcoholic fermentation, increased significantly with increasing glucose concentrations up to the 48th hour. Intra-and extra-cellular acetate levels increased significantly with increasing glucose concentrations of the growth medium and reached their maximums on the 48th hour, as was the case also for pyruvate. Intra-cellular formate levels increased up to 15 g/L, while extra-cellular levels increased with increasing glucose concentration. The maximum intra-and extra-cellular lactate levels were determined at 12.5 g/L and 20 g/L of glucose on the 48th hour, respectively. The results suggest that elevated ethanol production suppressed lactate and formate production, supported via possibly formed CO(2). In addition, pyruvate, as well as acetate, were used as carbon sources due to the depletion of glucose contents in the growth medium.  相似文献

Double haploids (DH), obtained during androgenesis in vitro or by genome diploidisation in regenerated haploids, are one type of basic materials used in triticale breeding programmes. The aim of this study was to improve DH production by a combination of colchicine treatment methods on a sample of five winter and five spring triticale hybrids. Colchicine was applied in vitro either in the C17 medium to induce embryo-like structures (ELS) or in the 190-2 medium for green plant (GP) development. Regenerants which remained haploid were immersed in a colchicine solution either when placed on the medium prior to transferring to soil or when growing in pots, followed by the application or absence of cooling. Colchicine treatment during anther culture affected neither ELS nor GP development, but significantly increased the number of DH plants in comparison to spontaneous chromosome doubling. The highest efficiency was recorded when colchicine was applied in the induction medium (55%) versus the regeneration medium (44.5%) or no colchicine treatment (30%). The effectiveness of chromosome duplication in haploid plants ranged from 32 to 64.5% and it was the highest for the treatment on the medium followed by cooling. Individual hybrids differed regarding their capability of regeneration and chromosome doubling, which were consistent only to a low or moderate extent. However, taken together, winter and spring hybrids did not differ significantly. Combined colchicine application resulted in a high yield of DH production, 82.6% for all triticale hybrids, and can provide a considerable number of fertile DH lines for triticale breeding programmes.  相似文献

A map of the barley genome consisting of 107 loci was constructed using double haploid lines. 33 RAPD loci marks all barley chromosomes. The total length of the obtained map is 1047 centiMorgans (cM) with a 9.8 cM average distance between markers. 7 linkage groups was identified as certain chromosomes. The distribution of the markers was analyzed, comparison with other published maps was made. Some statistical considerations are presented. Alternative strategies of gene mapping are considered.  相似文献

Observations on the Ml, M2 and M3 generations from three barley crosses confirmed that pollen irradiation can cause deviations from expected segregation ratios for certain characters. The reduced fertility observed in the Ml and M2 generations of these crosses could be problematical in breeding programmes. It was of particular interest that at the highest dose rate used for the cross TS117 × Scots Bere there was no expression of the 6-row character, which is controlled by a recessive paternal factor.  相似文献

Abstract

Doubled haploid (DH) plants are completely homozygous individuals that can be generated in just a few months via androgenesis in vitro. DHs are useful tools in genetic research and in plant breeding. Isolated microspore culture (IMC) is the most efficient way to produce DHs, but a strong genotype dependency imposes limitations to its wide application. Six-row, spring barley genotypes are considered as particularly recalcitrant due to a low frequency of embryogenesis and a high rate of albinism. Seeking to develop an efficient IMC protocol for this type of barley, we explored four important factors: (1) the harvest stage of immature spikes, (2) the type of pretreatment applied, (3) the osmotic potential in the induction medium, and (4) the plating density of microspores. This work was first performed using four barley genotypes: two typical six-row spring cultivars (ACCA and Léger), a two-row spring (Gobernadora) and a two-row winter (Igri) cultivar. First, by optimizing the harvest stage for each genotype we obtained a twofold to fourfold increase in the yield of embryogenic microspores. Second, two pretreatments (0.3 M mannitol for 2 days, or a combination of cold and heat over 15 days) both performed significantly better than the commonly used cold pretreatment (28 days at 4 °C). Third, an induction medium-containing mannitol (32 g/l) doubled green plant regeneration. Fourth, a plating density of 106 microspores/ml yielded the highest number of green regenerated plants. Our most important findings were then confirmed using sets of F1s from a six-row, spring-type breeding program.  相似文献

The susceptibility of barley doubled haploids (DH) to Fusarium head blight (FHB) was investigated. Heads of 24 DH lines (11 two-rowed and 13 six-rowed) derived from F1 Maresi (two-rowed) × Pomo (six-rowed) hybrids were inoculated with a conidial suspension of the single isolate IPO348-01 of Fusarium culmorum. The experiment was carried out in three consecutive years (1996–98) in one location. The number of kernels per ear, 1000-kernel weight and kernel weight per ear were recorded in inoculated and control plots. In the infected kernels nivalenol (NIV) content and deoxynivalenol (DON) content were determined. The effects of genotype, year and genotype-year interaction on reduction of yield traits were significant. For mycotoxin content only genotype and year effects were found to be important. The average NIV concentration in kernels of inoculated lines ranged from 0.15 mg/kg in the two-rowed line MP7 to 6.36 mg/kg in the six-rowed line MP113. A low accumulation of DON was observed in the studied population (from 0.01 to 0.20 mg/kg). Generally, no significant differences in mycotoxin content were found between two-rowed and six-rowed genotypes. The line MP7 was found to be superior — with the lowest mycotoxin accumulation, and reduction in yield traits. Environmental conditions (years) affected DON and NIV level in kernels; however, the tendency to a lower or higher accumulation of mycotoxin in individual lines was stable over the years.  相似文献
