Psychophysical measurements were made of the sensory effects of l-menthol applied topically to the forearm under controlled thermal conditions. In the first experiment, subjects judged the intensity and quality of sensations produced by warming or cooling the skin in the presence of menthol or the vehicle. During cooling, menthol intensified cutaneous sensations and increased reports of burning. During warming, menthol intensified sensations transiently at low temperatures and weakened them lastingly at higher temperatures; the frequency of reports of burning varied with intensity. A second experiment tested the hypothesis that menthol would lower the threshold for warmth and raise the threshold for heat pain. No change in either threshold was observed. The primary sensory effects of l-menthol on hairy skin are therefore to heighten the perception of cooling and to attenuate the perception of moderate warming. In contrast with other common chemical irritants, menthol's pungent qualities appear to be enhanced by cooling and suppressed by warming; this suggests that its sensory irritancy may be attributable to the stimulation of a population of high-threshold cold fibers or cold-sensitive nociceptors.  相似文献

This study utilized the methodology of Functional Measurement theory to investigate the additivity of painful and non-painful thermally induced experiences at one body site with those produced by brief noxious and innocuous electrical stimuli at another. Forty healthy young subjects were tested, using a Peltier thermode to induce tonic pain and an electrocutaneous stimulator for presenting phasic pain, under conditions of either full attention or visual/cognitive distraction (counting numerous light signals) in order to evaluate whether the summed effects are attributable to refocused attention. Six levels of intensity were combined in a factorial design for both tonic and phasic pain. Subjects indicated the overall strength of their dual perception on a visual analog scale. Stimuli showed complex patterns of interaction. Two stimuli were generally rated as greater than one, but the summation was far from additive and greatly influenced by the intensity of the stronger stimulus, suggesting inhibitory action. In general, tonic heat pain strongly affected the perception of phasic electrocutaneous pain whereas the reverse was only partly true. Distraction had a very small effect, suggesting that the “pain inhibits pain” phenomenon attributable to diffuse noxious inhibitory controls (DNIC) is not due to attentional processes. Our data also relate to issues regarding spatial summation across dermatomes and to adaptation level effects in pain, in which a strong painful experience serves as an anchor or comparison point by which others are judged. The psychophysical findings provide a perceptual foundation for clinical phenomena in which patients face with comorbid pain disorders.  相似文献

This study of the bed bug, Cimex lectularius, examines tolerance of adult females to extremes in temperature and loss of body water. Although the supercooling point (SCP) of the bed bugs was approximately −20°C, all were killed by a direct 1 h exposure to −16°C. Thus, this species cannot tolerate freezing and is killed at temperatures well above its SCP. Neither cold acclimation at 4°C for 2 weeks nor dehydration (15% loss of water content) enhanced cold tolerance. However, bed bugs have the capacity for rapid cold hardening, i.e. a 1‐h exposure to 0°C improved their subsequent tolerance of −14 and −16°C. In response to heat stress, fewer than 20% of the bugs survived a 1‐h exposure to 46°C, and nearly all were killed at 48°C. Dehydration, heat acclimation at 30°C for 2 weeks and rapid heat hardening at 37°C for 1 h all failed to improve heat tolerance. Expression of the mRNAs encoding two heat shock proteins (Hsps), Hsp70 and Hsp90, was elevated in response to heat stress, cold stress and during dehydration and rehydration. The response of Hsp90 was more pronounced than that of Hsp70 during dehydration and rehydration. Our results define the tolerance limits for bed bugs to these commonly encountered stresses of temperature and low humidity and indicate a role for Hsps in responding to these stresses.  相似文献

The dynamics and the level of accumulation of small heat shock proteins (sHSP group 21–27) after a heat exposure were studied in three Drosophila species differing in thermotolerance. The southern species Drosophila virilis, having the highest thermotolerance, surpassed thermosensitive D. lummei and D. melanogaster in the level of sHSPs throughout the temperature range tested. The results suggest an important role of sHSPs in the molecular mechanisms of adaptation to adverse environmental conditions, particularly to hyperthermia.  相似文献

用PCR的方法获得AtJ2和AtJ3基因的3'非编码区的核甘酸片段作为探针,Northern杂交结果表明:AtJ2和AtJ3基因在植物的根、茎、叶、花蕾、花和长角果中都有表达,并在植物整个生长周期中都有表达,但随着植株的衰老表达量有所下降.不同环境胁迫的实验结果表明:热激使AtJ2和AtJ3基因的表达迅速升高;冷胁迫也能诱导这两个基因表达的明显增加,但需要的时间比热激要长得多,达9 h;水分胁迫能引起AtJ2和AtJ3基因表达量的微弱增加;可盐胁迫对AtJ2和AtJ3基因的表达没有影响.说明AtJ2和AtJ3基因可能参与对除盐胁迫以外多种环境刺激的响应.  相似文献

疼痛长期困扰人类健康,其发病机制纷繁复杂,究竟谁在其中扮演了重要的作用是目前亟待解决的重大问题。随着对疼痛研究的不断深入,超极化激活的环核苷酸门控通道逐渐引起广泛关注。在炎性痛和神经病理性痛过程中,它都扮演了至关重要的作用,其数目改变和开放频率增加都参与介导了疼痛部位的异常放电,成为诱发疼痛的开关。在给与阻断剂或敲除通道2亚型后,能明显缓解炎性痛和神经病理性痛的不良反应,成为可以缓解疼痛发生的新靶点。超极化激活的环核苷酸门控通道在机体分布广泛,参与多种重要生理功能的调节,但目前还没有针对该门控通道某种亚型的特异性阻断剂。在今后,也许超极化激活的环核苷酸门控通道会成为临床治疗疼痛的新靶点,该通道的特异性药物也将为广大患者带来新的福音。  相似文献

The authors sought to determine whether developmental differences in the magnitude of embryonic mortality caused by heat stress in vivo are caused by changes in resistance of embryos to elevated temperature. In this regard, responses of oocytes, two-cell embryos, four- to eight-cell embryos, and compacted morulae to heat shock were compared. An additional goal was to define further the role of cumulus cells and glutathione in thermoprotection of oocytes. In experiment 1, heat shock (41°C for 12 hr) decreased the number of embryos developing to the blastocyst stage for two-cell (26% vs. 0%) and four- to eight-cell (25% vs. 10%) embryos but did not affect morulae (37% vs. 42%). In experiment 2, exposure of two-cell embryos to 41°C for 12 hr reduced the number of four- to eight-cell embryos present 24 hr after the end of heat shock (88% vs. 62%). In experiment 3, heat shock reduced the number of two-cell embryos developing to blastocyst (49% vs. 8%) but did not affect subsequent development of oocytes when heat shock occurred during the first 12 hr of maturation (46% vs. 41% development to blastocyst); membrane integrity was not altered. In experiment 4, oocytes were cultured with an inhibitor of glutathione synthesis, DL-buthionine-[S,R]-sulfoximine (BSO), for 24 hr and exposed to 41°C for the first 12 hr of maturation. Percentages of blastocysts were 35% (39°C), 18% (41°C), 17% (39°C+BSO), and 11% (41°C+BSO). For experiment 5, oocytes were either denuded or left with cumulus intact and were then radiolabeled with [35S]methionine and [35S]cysteine at 39°C or 41°C for 12 hr. Exposure of oocytes to 41°C for 12 hr reduced overall synthesis of 35S-labeled TCA-precipitable intracellular proteins (18,160 vs. 14,594 dpm/oocyte), whereas presence of cumulus increased synthesis (9,509 vs. 23,246). Analysis by two-dimensional SDS PAGE and fluorography revealed that heat shock protein 68 (HSP68) and two other putative heat shock proteins, P71 and P70, were synthesized by all oocytes regardless of treatment. Heat shock did not alter the synthesis of HSP68 or P71 but decreased amounts of newly synthesized P70. Cumulus cells increased synthesis of P71 and P70. Results indicate there is a biphasic change in resistance to elevations in temperature as oocytes mature, become fertilized, and develop. Resistance declines from the oocyte to the two-cell stage and then increases. Evidence suggests a role for cumulus cells in increasing HSP70 molecules and protein synthesis. 目的:通过对慢性非特异性颈部不适飞行员与无症状飞行员之间头颈夹肌表面肌电特征的比较,为慢性非特异性颈部不适飞行员的早期诊断提供参考依据。方法:用表面肌电仪测量52名慢性非特异性颈部不适飞行员与11名无症状飞行员双侧头颈夹肌的松弛状态、等长收缩以及异长收缩时的表面肌电特征,分析不同状态下双侧头颈夹肌疲劳性肌电指标MFs、MPFs、ZCRs以及平均肌电AEMG值,用统计学软件SPSS 18.0分析实验结果。结果:非特异性颈部不适飞行员与无症状飞行员双侧头颈夹肌在松弛状态时疲劳性肌电指标MFs、MPFs、ZCRs比较均无统计学差异(P0.05),其双侧头颈夹肌在等长收缩和异长收缩时间有统计学差异(P0.05)。头颈夹肌处于松弛状态时,两组间平均肌电AEMG比较无统计学差异(P0.05),而在肌肉等长收缩和异长收缩时,两组间比较有统计学差异(P0.05)。结论:慢性非特异性颈部不适飞行员与无症状飞行员之间头颈夹肌表面肌电相关指标存在统计学差异,慢性颈部不适的飞行员相比无症状飞行员更易出现头颈夹肌的疲劳以及头颈夹肌的功能下降。头颈夹肌表面肌电特征有助于早期诊断慢性非特异性颈部不适飞行员的肌肉功能状态的改变。  相似文献

Relationships between the appearance of low-molecular-weight heat-shock proteins (LMW HSPs) in maize, winter wheat, and winter rye mitochondria and the tolerance of the mitochondria to hyperthermia (42°C, 3 h) were studied using one-dimensional SDS-PAGE, immunochemical methods, and polarography. Heat shock inhibited respiration to a greater extent in the wheat and rye than in the maize mitochondria. A single 20-kD LMW HSP was found both inside and on the surface of mitochondria isolated from heat-treated wheat and rye seedlings. After heating maize seedlings, two LMW HSPs (28 and 23 kD) appeared inside the mitochondria, and three proteins (22, 20, and 19 kD) appeared on their surface. We suppose that the latter three proteins play an essential role in the protection of mitochondria from hyperthermic damage. It seems likely that the diversity of the hyperthermia-induced LMW HSPs in plant mitochondria affects their thermal stability.  相似文献

Physical or chemical damage to peripheral nerves can result in neuropathic pain which is not easily alleviated by conventional analgesic drugs. Substantial evidence has demonstrated that voltage-gated Na+ channels in the membrane of damaged nerves play a key role in the establishment and maintenance of pathological neuronal excitability not only of these peripheral nerves but also in the second- and third-order neurons in the pain pathway to the cerebral cortex. Na+ channel blocking drugs have been used in treating neuropathic pain with limited success mainly because of a preponderance of side-effects. We have developed an analogue of mexiletine which is approximately 80 times more potent than mexiletine in competing with the radioligand, 3H-batrachotoxinin for binding to Na+ channels in rat brain membranes and also it is much more lipophilic than mexiletine which should enhance its uptake into the brain to block the increased expression of Na+ channels on second- and third-order neurons of the pain pathway. This analogue, HFI-1, has been tested in three different rat models of neuropathic pain (formalin paw model, ligated spinal nerve model and contusive spinal cord injury model) and found to be more effective in reducing pain behaviours than mexiletine.  相似文献

目的:观察电针不同穴位对慢性炎症痛模型大鼠行为学改变的影响,进一步探讨电针对慢性痛所致心理行为改变的调节规律:方法:将40只雄性Wistar大鼠随机分为正常组、模型组、电针百会组、电针关元组、电针足三里组,每组各8只。除正常组外,其余各组均采用弗氏完全佐剂(CFA)制备佐剂性关节炎(AA,即慢性炎症痛)大鼠模型,于造模第2天各治疗组捆绑固定后给予电针20min,正常组及模型组捆绑束缚20min,隔日一次,共10次。于造模前、造模后、治疗5次、10次分别采用高架十字迷宫检测各组大鼠开放臂进入次数比例(OE%)及开放臂停留时间比例(OT%)的变化。结果:与造模前比较,电针百会穴组大鼠造模后0E%和0T%明显减少(P〈0.05,P〈0.05),且治疗5次后OT%、治疗10次后0E%与0T%均降低明显(P〈0.01,P〈0.01,P〈0.05),而电针关元组大鼠治疗10次后OE%和OT%则明显升高(P〈0.05,P〈0.05),电针足三里组大鼠造模后0T%、治疗5次后大鼠OE%与OT%、治疗10次后OT%均明显降低(P〈O.01,P〈0.05,P〈0.01,P〈0.05);与造模刚结束比较,电针百会组大鼠在治疗5次、10次后0E%由升高逐步降低,而OT%由低逐渐升高(P〉0.05),电针关元组模型大鼠治疗10次后模型大鼠0E%和0T%均明显升高(P〈0.05,P〈0.05),电针足三里组模型大鼠治疗10次后OT%升高明显(P〈0.05);与治疗5次后比较,电针百会组大鼠在治疗10次后OE%降低、OT%升高(P〉O.05),电针关元组与电针足三里组大鼠治疗10次后OE%和OT%均明显升高(P〈0.05,P〈0.05;P〈0.01,P〈0.01)。与电针百会组比较,电针关元组与电针足三里组大鼠在治疗10次时OE%均明显升高(P〈0.01,P〈0.01)结论:电针不同穴位具有一定抗抑郁和抗焦虑作用,可改善慢性炎症痛所致大鼠焦虑、情绪行为的变化;不同穴位效应不同,关元穴与足三里穴具有综合调节、抗焦虑效应,且具有相对特异性。  相似文献

Evidence supporting an association between phenotypic plasticity and invasiveness across a range of plant taxa is based primarily on comparisons between invasive species and native species whose potential invasiveness is typically unknown. Comparison of invasive and non-invasive exotic species would provide a better test of whether plasticity promotes invasion. Such comparisons should distinguish between adaptive and non-adaptive plasticity because they have different consequences for invasiveness. Adaptive plasticity is expected to promote the invasion of multiple habitats, but non-adaptive plasticity may reflect specialization for invading more favorable habitats only. We grew four invasive and four non-invasive species of the Commelinaceae with and without competitors and compared their putatively adaptive plasticity of three traits related to competitive ability and non-adaptive plasticity in performance. The invasive species grew significantly more than the non-invasive species only in the non-competitive environment. The invasive species had greater plasticity of performance (total biomass) in response to competition than non-invasives, but there was no consistent difference in the plasticities of the traits related to competitive ability. These results are consistent with specialization of these invasive taxa for invading the more productive non-competitive environment rather than a superior ability to invade both competitive and non-competitive environments. A comprehensive understanding of the relationship between plasticity and invasiveness will require many more comparisons of the plasticity of invasive and non-invasive taxa in a range of traits in response to a variety of environments.  相似文献

选用朱砂叶螨Tetranychus cinnabarinus阿维菌素抗性品系和敏感品系,测定了热预刺激后其在极限高温下的存活率,并应用SDS-PAGE技术研究了热激蛋白(HSPs)的种类及其含量。结果表明:非致死的热预刺激能显著提高朱砂叶螨耐极限温度的能力。两个品系在不同温度热激处理后,其蛋白质种类和含量发生了变化。正常情况下,朱砂叶螨敏感品系与阿维菌素抗性品系相比缺失8条条带;敏感品系热激后,增加了分子量分别为97.2,74.3,62.4,53.0和30.3 kDa的5条条带; 抗性品系热激后没有特异蛋白带的产生,但进一步高温胁迫后有些蛋白表达增强。此结果有助于解释朱砂叶螨抗性品系存在高温适合度优势现象。  相似文献

The relationship between induction of central sensitization and facilitation of temporal summation to repetitive stimulation is still unclear. The aim of this study was to investigate temporal summation before and after the induction of secondary hyperalgesia by two different experimental methods: capsaicin injection and controlled heat injury. The effect of each injury model was assessed on a separate day with an interval of at least 5 days. Twelve healthy volunteers participated. Each experiment was performed using electrical, radiant heat, mechanical impact, and punctuate stimuli consecutively. The pain threshold (PT) to a single stimulus and the summation threshold to five repetitive stimuli for electrical (2?Hz) and radiant heat (0.83?Hz) were assessed within the secondary hyperalgesic area. The degree of temporal summation for stimulus intensities of 0.8, 1.0, and 1.2 times the baseline pain thresholds were evaluated by the increase in visual analogue scale (VAS) scores from the first to the fifth stimulus of the train. Further, the degrees of temporal summation were assessed for mechanical impact and punctuate stimuli within the primary and secondary hyperalgesic areas. The contra-lateral forearm served as control (no injury). The pain threshold and the summation threshold to electrical and heat stimuli decreased significantly within the secondary hyperalgesic area after the injury induced by both heat injury or capsaicin injection. However, there was no temporal summation for heat and electrical stimuli in either model. In contrast, for the mechanical impact and punctuate mechanical stimuli the degree of temporal summation was significantly facilitated in the secondary hyperalgesic areas compared with the baseline and the control arm in both models. In the primary hyperalgesic area, the degree of temporal summation was facilitated to mechanical impact and punctuate stimuli but only following the capsaicin injection. In conclusion, the temporal summation mechanism for mechanical stimuli was facilitated in the secondary hyperalgesic area.  相似文献

In a previous article in the Journal of Visualized Experiments we have demonstrated skin microdialysis techniques for the collection of tissue-specific nociceptive and inflammatory biochemicals in humans. In this article we will show pain-testing paradigms that are often used in tandem with microdialysis procedures. Combining pain tests with microdialysis provides the critical link between behavioral and biochemical data that allows identifying key biochemicals responsible for generating and propagating pain.Two models of evoking pain in inflamed skin of human study participants are shown. The first model evokes pain with aid of heat stimuli. Heat evoked pain as described here is predominantly mediated by small, non-myelinated peripheral nociceptive nerve fibers (C-fibers). The second model evokes pain via punctuated pressure stimuli. Punctuated pressure evoked pain is predominantly mediated by small, myelinated peripheral nociceptive nerve fibers (A-delta fibers). The two models are mechanistically distinct and independently examine nociceptive processing by the two major peripheral nerve fiber populations involved in pain signaling.Heat pain is evoked with aid of the TSA II, a commercially available thermo-sensory analyzer (Medoc Advanced Medical Systems, Durham, NC). Stimulus configuration and delivery is handled with aid of specific software. Thermodes vary in size and shape but in principle consist of a metal plate that can be heated or cooled at various rates and for different periods of time. Algorithms assessing heat-evoked pain are manifold. In the experiments shown here, study participants are asked to indicate at what point they start experiencing pain while the thermode in contact with skin is heated at a predetermined rate starting at a temperature that does not evoke pain. The thermode temperature at which a subject starts experiencing pain constitutes the heat pain threshold.Mechanical pain is evoked with punctuated probes. Such probes are commercially available from several manufacturers (von Frey hairs). However, the accuracy of von Frey hairs has been criticized and many investigators use custom made punctuated pressure probes. In the experiments shown here eight custom-made punctuated probes of different weights are applied in consecutive order, a procedure called up-down algorithm, to identify perceptional deflection points, i.e., a change from feeling no pain to feeling pain or vice versa. The average weight causing a perceptional deflection constitutes the mechanical pain threshold.  相似文献
