The frequency responses of 11 rapidly adapting (RA) fibers in cat were studied by representing the average firing rate as a function of sinusoidal stimulus amplitude and stimulus frequency. Specifically, rate-intensity functions at different stimulation frequencies were fitted by four-parameter (a0, a1, a2, a3), piece-wise linear functions using nonlinear regression (n = 59; R2 > 0.877). Rate-intensity functions at intermediate frequencies were found by linear interpolation. The result of this analysis is rate–amplitude–frequency functions plotted as two-dimensional surfaces. The surfaces consist of five regions separated and sufficiently defined by four space curves. At 14 different frequencies, the statistical distribution of each rate-intensity-function parameter could be approximated by a particular lognormal distribution (n = 56; R2 > 0.796). The Kolmogorov–Smirnov test fails to reject this hypothesis for each combination of frequency and parameter (56 tests; p > 0.39). Therefore, at a given frequency, the variation of the parameters can be represented by lognormal distributions with specific means and standard deviations. Responses of six RA fibers, which are different from the data-set used for modeling, were compared with the stochastic model at different frequencies. The parameters of those fibers were tested against the null hypotheses that they were sampled from the particular parameter distributions dictated by the model. The Kolmogorov–Smirnov test fails to reject all the hypotheses at the α = 0.05 level (44 tests). At the α = 0.10 level, only a few test parameters were found to be departing from the model (a0 and a1 at 5?Hz; a2 at 20?Hz; a2 and a3 at 50?Hz). The remaining test parameters could be accurately described by the model. Having confirmed the validity of the model, the logarithmic means and the logarithmic standard deviations of the lognormally distributed rate-intensity-function parameters were estimated in the frequency range of 4–200?Hz. The rate–amplitude–frequency surfaces sampled from the established stochastic model completely characterize the rate responses of RA fibers to sinusoidal stimuli and are superior to tuning curves which require selecting criterion responses. The current rate-response model is promising for future computational work, especially on population modeling.  相似文献

The population response of rapidly-adapting (RA) fibers is one component of the physiological substrate of the sense of touch. Herein, we describe a computational scheme based on the population-response model by K.O. Johnson (J. Neurophysiol. 37: 48–72, 1974) which we extended by permitting the capability to include the spatial distributions of receptors in the glabrous skin linked to RA fibers. The hypothetical cases simulated were rectangular, uniformly random and proximo-distally Gaussian distributions. Each spatial organization produced qualitatively distinct population-response profiles that also varied due to stimulus parameters. The effects of stimulus amplitude, average innervation density and contactor-probe location were studied by considering various response measures: number of active fibers, summated firing rate and the average firing rate of a subset of the modeled population. The outcome of the measures were statistically compared among simulated anatomical distributions. The response is the same for rectangular and uniformly random distributions, both of which have a homogeneous innervation density. However, the Gaussian distribution produced statistically different responses when the measure was not averaged over the subset population which represented the receptive field of a higher-order neuron. These results indicate that, as well as stimulus parameters, the anatomical organization is a significant determinant of the population response. Therefore, reconstructing population activity for testing psychophysical hypotheses must presently be done with care until the organization of the receptors within the skin has been clarified.  相似文献

The goal of this study was to investigate the relationship between the psychophysical vibrotactile thresholds of the Pacinian (P) channel and the mechanical properties of the skin at the fingertip. Seven healthy adult subjects (age: 23–30) participated in the study. The mechanical stimuli were 250-Hz sinusoidal bursts and applied with cylindrical contactor probes of radii 1, 2, and 3.5?mm on three locations at the fingertip. The duration of each burst was 0.5?s (rise and fall time: 50?ms). The subjects performed a two-interval forced-choice task while the stimulus levels changed for tracking the threshold at 75% probability of detection. There were significant main effects of contactor radius and location (two-way ANOVA, values of p?<?0.001). The thresholds decreased as the contactor radius increased (i.e., spatial summation effect) at all locations. The thresholds were lowest near the whorl at the fingertip. Additionally, we measured the mechanical impedance (specifically, the storage and loss moduli) at the contact locations. The storage moduli did not change with the contactor location, but the loss moduli were lowest near the whorl. While the loss moduli decreased, the storage moduli increased (e.g., more springiness) as the contactor radius increased. There was moderate and barely significant correlation between the absolute thresholds and the storage moduli (r?=?0.650, p?=?0.058). However, the correlation between the absolute thresholds and the loss moduli was high and very significant (r?=?0.951, p?<?0.001). The results suggest that skin mechanics may be important for locally shaping psychophysical detection thresholds, which would otherwise be expected to be constant due to uniform Pacinian innervention density at the fingertip.  相似文献

The presence of some cytoskeletal proteins related to the intermediate filaments glial fibrillary acidic protein -GFAP and vimentin) and S-100 protein has been investigated in sensory formations of the glabrous skin of the rat. A positive reaction both for S-100 protein and vimentin was found in the inner core and related cells of glomerular and simple sensory corpuscles; in contrast, no positive reaction was shown for GFAP. The authors discuss these results on the basis of the glial origin of the inner core and related cells in sensory formations.  相似文献

The usual pigmentation pattern in mammalian skin consists of fixed melanocytes in the basal layer of the epidermis, supplying keratinocytes with melanosomes. We observed that the glabrous skin (rhinaria and footpads) of dogs deviates from this pattern. In dogs, melanocytes are found in both the dermis and epidermis. The epidermal melanocytes are situated in the intercellular spaces of the basal and spinous layers. They are characterized by a quantity of cytoplasm containing a centriole, also developing melanosomes, and in some cases annulate lamellae. There is a high frequency of closely apposed melanocytes in the epidermis. Melanosomes in different stages of formation are also abundant. The morphology of the glabrous skin of dogs suggests transport of melanocytes from the dermis into the epidermis and formation of melanosomes in the epidermis. A distributed and intense pigment formation may be necessary to achieve the black noses of many dog breeds and wild canids, as well as dark footpads despite heavy abrasion and rapid skin renewal.  相似文献

Fast axoplasmic transport through the sensory fibers of the sciatic nerve has been compared in rats and mice. The use of in vitro incubation permits high levels of specific activity to be attained when labeling with [35S]l-methionine. The specific activity of the transported proteins was about 10-fold greater in mice than in rats. Proteins labeled with radioactive methionine were examined after separation on polyacrylamide gels. There are no differences between mice and rats when the proteins carried by rapid transport are compared. Similarly, the proteins synthesized by the Schwann cells of these two species are not distinguishable. The dorsal root ganglia of mice, however, yield a band of radioactivity that is not seen in ganglia from rats. This band migrates with an apparent molecular weight of 31,000 daltons.  相似文献

We have recently demonstrated that in a rat model of chemotherapy-induced peripheral neuropathy (CIPN), there is a significant decrease in the duration of the depolarization-evoked Ca2+ transient in small diameter, IB4+, and capsaicin-responsive neurons innervating the glabrous skin of the hindpaw. This change was specific to the transient duration and significantly smaller if not undetectable in neurons innervating the dorsal skin of the hindpaw or the skin of the inner thigh. Given the importance of mitochondria in intracellular Ca2+ regulation and the findings of chemotherapy-associated increase in mitotoxicity along the sensory neuron axons, we hypothesized that CIPN is due to both increases and decreases in mitochondria function, with changes manifest in distinct subpopulations of afferents. To begin to test this hypothesis, we used confocal microscopy and Ca2+ imaging in combination with pharmacological manipulations to study paclitaxel-induced changes in retrograde tracer-labeled neurons from naïve, vehicle-treated, and paclitaxel-treated rats. Paclitaxel treatment was not associated with decreased mitochondrial membrane potential or increased superoxide levels in the somata of putative nociceptive glabrous skin neurons. However, it was associated with significant increases in the relative contribution of mitochondria to the control of the evoked Ca2+ transient duration in putative nociceptive glabrous skin neurons, as well as increases in mitotracker and Tom20 staining which reflected an increase in mitochondrial volume. Furthermore, the relative contribution of the sarco-endoplasmic reticulum Ca2+ ATPase to the regulation of the duration of the depolarization evoked Ca2+ transient was also increased in this subpopulation of neurons from paclitaxel treated rats. Our results indicate that the paclitaxel-induced decrease in the duration of the evoked Ca2+ transient is due to both direct and indirect influences of mitochondria. It remains to be determined if and how these changes contribute to the manifestation of CIPN.  相似文献

Caveolin-3, the muscle-specific isoform of the caveolae-associated protein caveolin, is often thought to be localized exclusively in the surface membrane in mature fibers and associated with transverse (t)-tubular system only transiently during development. Skeletal muscle fibers present a model where the surface membrane (sarcolemma) can be completely separated from the cell by mechanical dissection. Western blotting of matching portions of individual fibers from adult rat muscle in which the sarcolemma was either removed (skinned segment), or left in place (intact segment), revealed that ≥ 70% of caveolin-3 is actually located deeper in the fiber rather than in the sarcolemma itself. Triton solubility of caveolin-3 was no different between sarcolemmal and t-tubule compartments. Confocal immunofluorescence microscopy showed caveolin-3 present throughout the t-system in adult fibers, with ‘hot-spots’ at the necks of the tubules in the sub-sarcolemmal space. A similar representation was seen for the muscle specific voltage-dependent sodium channel Nav1.4 and it was found that at least some Nav1.4 co-immunoprecipitated with caveolin-3 in skinned muscle fibers. The caveolin-3 hot-spots just inside the opening of t-tubules may form regions that localize ion channels and kinases at the key place needed for efficient electrical transmission into the t-tubules as well as for other signaling processes.  相似文献

Summary Explants of fetal rat liver maintained in organ culture lost about 40% of their mass in 42 hr of incubation as a result of decrease in blood cells and hepatocytes. Proteins from the cytosol and particulate elements of the tissue were found in the culture medium. About 60% of this protein was degraded to peptides during culture. The transfer of malate and lactate dehydrogenases from tissue to medium paralleled that of proteins. Glutamate dehydrogenase was lost from the mitochondria and in part leaked through the cell membrane into the medium. Net loss of activity of the three enzymes occurred, probably as a consequence of proteolytic degradation. Of 12 enzymes in liver tissue, the specific activities of eight—soluble malate dehydrogenase, glutamate dehydrogenase, succinate dehydrogenase, phosphopyruvate carboxylase, hexosediphosphatase, glucose-6-phosphatase, tyrosine, aminotransferase, and alanine aminotransferase—were unchanged or increased. Glycogen synthetase, aspartate aminotransferase, pyruvate kinase, and lactate dehydrogenase decreased. Although changes in membrane permeability may have had some influence on the results reported, the predominant effect was due to loss of protein from tissue as a result of discharge of total contents of some of the cells into the medium. The residual explanted tissue retained its structural integrity. It is concluded that fetal rat liver in organ culture provides a suitable model system for controlled studies with this organ in vitro. The effect of a high bromide intake on the kinetics of iodide uptake and elimination in the thyroid and skin of adult male rats was studied. In rats fed a diet with sufficient iodine supply (>25 μg I/d), the iodide accumulation in the skin predominated during the first hours after 131I -iodide application. From this organ, radioiodide was gradually transferred into the thyroid. A high bromide intake (>150 mg Br/d) in these animals led to a marked decrease in iodide accumulation, especially by the thyroid, because of an increase in iodide elimination both from the thyroid and from the skin. In rats kept under the conditions of iodine deficiency (<1 μ I/d), the iodide accumulation in the thyroid, but not in the skin, was markedly increased as a result of a thyrotropic stimulation. The effect of a high bromide intake (>100 mg Br/d) in these animals was particularly pronounced because the rates of iodide elimination were most accelerated both from their thyroid and from their skin. The route of administration of the NSAID, flurbiprofen (sq vs. po) resulted in positive and negative results respectively with regard to enhanced cancellous and cortical bone accumulation in the immature rat. This pharmacokinetic study was an effort to understand the pharmacodynamic difference between the two routes of administration observed when the same dose range of drug, given as single daily doses, had been employed in both studies. Conventional chiral pharmacokinetics were evaluated in young rats. A significant difference was observed in the Tmax of the active (S)-enantiomer by both administration routes (sq 4 h and po 1 h). The bioavailability, as evaluated by AUCs favored the sq route as expected. The plasma concentrations over 18 h, at steady state, for one po dose group (0.5 mg/kg/day) fell well within the therapeutic window described by the 0.1 and 0.5 mg/kg sq doses which had demonstrated anabolic bone activity. Oral dosing had exhibited no significant bone activity. We conclude that the pharmacodynamic difference between routes of administration cannot be simply explained on a pahrmacokinetic basis. Consequently, experiments detailing the pharmacodynamics and pharmacokinetics of single and multiple dose administration of aryl-propionic acids in normal and osteopenic states need further pharmacologic study. © 1994 Wiley-Liss, Inc.  相似文献

We intended to use the rat model to study the effect of autoantibodies on implantation and fetal viability. However, we have since found an effect of normal rat serum on fetal resorption rate and fetal viability. The objective of this study was to determine the consistency of this effect. Wistar strain albino rats were used for injection of 150 μl normal rat serum into the lumen of uterine horn on days L2–L6. The other uterine horn, used as a control, was injected with 150 μl normal saline. Percent implantation, fetal resorption rate and fetal viability were determined following the intrauterine injection of normal rat serum as compared with normal saline. A significant increase in fetal resorption rate was observed following the injection of rat serum on days L4 and L5 (P = 0.003 and P = 0.001, respectively). A significant decrease in fetal viability was demonstrated following the injection of rat serum on day L5 (P = 0.01). The rat can provide a suitable animal model for further studies.  相似文献

Evoked neural responses to tactile stimulation were recorded electro-physiologically from the mechanoreceptive afferent fibers innervating the buccal and submandibular regions of Wistar rats anesthetized with sodium thiopental. Miniature probes 200 μm in diameter were used, and data analysis was performed on the mechanosensitivity of responses to tactile stimulation in the areas innervated by the mental, mylohyoid, auriculotemporal, and cervical nerves. Mechanosensitivity of each area showed a characteristic distribution of slowly adapting (SA), rapidly adapting (RA), C-fiber (CF), and hair follicle (HF) units in individual receptive fields. The density of the SA units was high in the areas innervated by the mylohyoid and auriculotemporal nerves. The CF units were concentrated in the small dome in the area of the mylohyoid nerve and the auriculotemporal nerve, as shown by a significant response to the dynamic features of stimulation. Estimation of the current needed for tactile acuity suggests an important role of the SA fibers in the areas innervated by the auriculotemporal, mylohyoid, and cervical nerves.  相似文献

Human parathyroid hormone (hPTH 1-34) stimulates an anabolic response in human and animal skeletons; however, it is unclear if the effect is strain dependent. To determine if the anabolic response to hPTH (1-34) was dependent upon strain in rats we used 2 outbred strains (Sprague Dawley, Wistar), 2 inbred strains (Fischer 344, Wistar spontaneously hypertensive:SHR), and 2 mutant strains (Zucker obese, Zucker lean) of rats. Male rats, 5 weeks of age, from each strain were treated subcutaneously with 80 microg/kg body weight hPTH (1-34) or vehicle for 12 days. The response to PTH was similar in all strains whereby PTH exerted an anabolic effect on femoral bone mass and cancellous bone histology that was independent of strain differences. Histomorphometric indices of bone volume, mineralized surface and bone formation in lumbar vertebrae increased in all PTH-treated rats. Additionally, femur bone mineral content and bone mineral density measured by dual energy X-ray absorptiometry (DEXA), and ash weight increased in all PTH-treated rats. These increases occurred regardless of strain. In summary, PTH exerted comparable anabolic effects on bone mass, bone mineral density and bone formation in all rat models tested demonstrating that the skeletal responsiveness to PTH was not dependent upon strain.  相似文献

Pulmonary hypertension (PH) is a life-threatening lung disease. PH with concomitant lung diseases, e.g., idiopathic pulmonary fibrosis, is associated with poor prognosis. Development of novel therapeutic vasodilators for treatment of these patients is a key imperative. We evaluated the efficacy of dual activation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) using an active, small-molecule phosphodiesterase (PDE4)/PDE5 dual inhibitor (Compound A). Compound A increased both cAMP and cGMP levels in WI-38 lung fibroblasts and suppressed the expressions of type-1 collagen α1 chain and fibronectin. Additionally, compound A reduced right ventricular weight/left ventricular weight+septal weight ratio, brain natriuretic peptide expression levels in right ventricle, C─C motif chemokine ligand 2 expression levels in lung, and plasma surfactant protein D. Our data indicate that dual activation of cAMP/cGMP pathways may be a novel treatment strategy for PH.  相似文献

One of the main difficulties with primary rat brain endothelial cell (RBEC) cultures is obtaining pure cultures. The variation in purity limits the achievement of in vitro models of the rat blood-brain barrier. As P-glycoprotein expression is known to be much higher in RBECs than in any contaminating cells, we have tested the effect of five P-glycoprotein substrates (vincristine, vinblastine, colchicine, puromycin and doxorubicin) on RBEC cultures, assuming that RBECs would resist the treatment with these toxic compounds whereas contaminating cells would not. Treatment with either 4 microg/mL puromycin for the first 2 days of culture or 3 microg/mL puromycin for the first 3 days showed the best results without causing toxicity to the cells. Transendothelial electrical resistance was significantly increased in cell monolayers treated with puromycin compared with untreated cell monolayers. When cocultured with astrocytes in the presence of cAMP, the puromycin-treated RBEC monolayer showed a highly reduced permeability to sodium fluorescein (down to 0.75 x 10(-6) cm/s) and a high electrical resistance (up to 500 Omega x cm(2)). In conclusion, this method of RBEC purification will allow the production of in vitro models of the rat blood-brain barrier for cellular and molecular biology studies as well as pharmacological investigations.  相似文献

The manifestation of hybrid vigor with regard to the inheritance of body weight was established as a postweaning phenomenon in rats. Liver weight gain generally corresponded to body weight gain and also reflected hybrid vigor. The protein/DNA ratios of the chromatin from livers of inbred and heterotic hybrid male rats were found to increase with increasing age in growing rats. Hybrid chromatin had higher protein/DNA ratios throughout the age range studies. The difference between hybrids and inbreds were greatest in the early postweaning stages of growth and maturation. The RNA/DNA ratios of chromatin from livers of inbred and hybrid male rats were also found to exhibit trends within certain developmental periods.This investigation was supported by matching Federal and West Virginia State funds from the Hatch 190 grant and is published with the approval of the Director of the Agricultural Station as Scientific Paper No. 1293.  相似文献
