In order to synthesize a sugar ester at high concentration, 1,2-O-isopropylidene-α-d-glucofuranose (IpGlc), which is one of the sugar acetals and is more hydrophobic than unmodified glucose, was esterified with palmitic acid at 40°C using immobilized lipase from Candida antarctica in some organic solvents or their mixtures. Acetone + t-butyl alcohol (3:1 v/v) improved both the initial reaction rate and yield after 80 h: the product reached its maximum value (240 mmol/kg solvent; ca. 110 g/kg solvent) when 400 mmol IpGlc/kg solvent and 1,200 mmol palmitic acid/kg solvent were used in this solvent mixture.  相似文献

The cAMP-dependent protein kinase catalytic (C) subunit is inhibited by two classes of functionally nonredundant regulatory (R) subunits, RI and RII. Unlike RI subunits, RII subunits are both substrates and inhibitors. Because RIIβ knockout mice have important disease phenotypes, the RIIβ holoenzyme is a target for developing isoform-specific agonists and/or antagonists. We also know little about the linker region that connects the inhibitor site to the N-terminal dimerization domain, although this linker determines the unique globular architecture of the RIIβ holoenzyme. To understand how RIIβ functions as both an inhibitor and a substrate and to elucidate the structural role of the linker, we engineered different RIIβ constructs. In the absence of nucleotide, RIIβ(108-268), which contains a single cyclic nucleotide binding domain, bound C subunit poorly, whereas with AMP-PNP, a non-hydrolyzable ATP analog, the affinity was 11 nM. The RIIβ(108-268) holoenzyme structure (1.62 Å) with AMP-PNP/Mn2+ showed that we trapped the RIIβ subunit in an enzyme:substrate complex with the C subunit in a closed conformation. The enhanced affinity afforded by AMP-PNP/Mn2+ may be a useful strategy for increasing affinity and trapping other protein substrates with their cognate protein kinase. Because mutagenesis predicted that the region N-terminal to the inhibitor site might dock differently to RI and RII, we also engineered RIIβ(102-265), which contained six additional linker residues. The additional linker residues in RIIβ(102-265) increased the affinity to 1.6 nM, suggesting that docking to this surface may also enhance catalytic efficiency. In the corresponding holoenzyme structure, this linker docks as an extended strand onto the surface of the large lobe. This hydrophobic pocket, formed by the αF-αG loop and conserved in many protein kinases, also provides a docking site for the amphipathic helix of PKI. This novel orientation of the linker peptide provides the first clues as to how this region contributes to the unique organization of the RIIβ holoenzyme.  相似文献

 Cyclic AMP (cAMP) is an important regulator of liver growth and differentiation. The main intracellular cAMP receptor, cAMP-dependent protein kinase (PKA), consists of two regulatory (R) and two catalytic (C) subunits. There are two classes, RI and RII, of the regulatory subunit, giving rise to type I (RI2C2) and type II (RII2C2) PKA. The RI/RII ratio generally decreases during organ development, and increases during carcinogenesis. Alterations in this ratio have been implicated as an important factor in experimental and clinical carcinogenesis. We have studied the expression of RIα, RIIα, Cα, and an important substrate of PKA, the cAMP-response element binding protein, during rat liver carcinogenesis. Two-color immunofluorescence and confocal laser scan microscopy were used to characterize localization of the cAMP-dependent signal transducers in hepatocytes, bile ducts, oval cells, and preneoplastic lesions. We found that bile ducts and oval cells (putative liver stem cells) contained a higher RI/RII ratio than hepatocytes and preneoplastic lesions. Thus, an altered RI/RII ratio was not detected during early rat liver carcinogenesis, but may contribute to differentiation of putative liver stem cells to hepatocytes. The inheritance of resistance to sunflower downy mildew (SDM) derived from HA-R5 conferring resistance to nine races of the pathogen has been determined and the new source has been designated as Pl 13 . The F2 individuals and F3 families of the cross HA-R5 (resistant) × HA 821 (susceptible) were screened against the four predominant SDM races 300, 700, 730, and 770 in separate tests which indicated dominant control by a single locus or a cluster of tightly linked genes. Bulked segregant analysis (BSA) was carried out on 116 F2 individuals with 500 SSR primer pairs that resulted in the identification of 10 SSR markers of linkage groups 1 (9 markers) and 10 (1 marker) of the genetic map (Tang et al. in Theor Appl Genet 105:1124–1136, 2002) that distinguished the bulks. Of these, the SSR marker ORS 1008 of linkage group 10 was tightly linked (0.9 cM) to the Pl 13 gene. Genotyping the F2 population and linkage analysis with 20 polymorphic primer pairs located on linkage group 10 failed to show linkage of the markers with downy mildew resistance and the ORS 1008 marker. Nevertheless, validation of polymorphic SSR markers of linkage group 1 along with six RFLP-based STS markers of linkage group 12 of the RFLP map of Jan et al. (Theor Appl Genet 96:15–22, 1998) corresponding to linkage group 1 of the SSR map, mapped seven SSR markers (ORS 965-1, ORS 965-2, ORS 959, ORS 371, ORS 716, and ORS 605) including ORS 1008 and one STS marker (STS10D6) to linkage group 1 covering a genetic distance of 65.0 cM. The Pl 13 gene, as a different source with its location on linkage group 1, was flanked by ORS 1008 on one side at a distance of 0.9 cM and ORS 965-1 on another side at a distance of 5.8 cM. These closely linked markers to the Pl 13 gene provide a valuable basis for marker-assisted selection in sunflower breeding programs.  相似文献

The types I, II, and III receptors (RI, RII, RIII) for transforming growth factor-β (TGF-β) become down-regulated in response to ligand, presumably via their internalization from the cell surface. This report examines the down-regulation of full-length RI, RII, and RIII in cells endogenously or transiently expressing these receptors. Down-regulation occurred rapidly (within 2 h after TGF-β1 treatment at 37°C) and showed a dose response, between 10 and 200 pM TGF-β1, in cells expressing RI, RII, and RIII (Mv1lu and A549 cells). A comparison between Mv1Lu and mutant cell derivatives R-1B (lacking RI) or DR-26 (lacking RII) indicated that all three receptors were necessary for efficient down-regulation. Down-regulation experiments, utilizing TGF-β-treated 293 cells transiently expressing different combinations of these receptors indicated that neither RII or RIII were down-regulated when expressed alone and that RI was required for maximal down-regulation of RII. RII and RIII were partially down-regulated when these receptors were coexpressed in the absence of RI (in R-1B and 293 cells). Surprisingly, TGF-β receptors were partially down-regulated in Mv1Lu, A549, and 293 cells treated with TGF-β1 at 4°C. Microscopic examination of 293 cells coexpressing RI fused to green fluorescent protein (RI–GFP) and RII indicated that, after treatment with TGF-β1 at 4°C, RI–GFP formed aggregates at the cell surface at this temperature. RI–GFP was not detected at the surface of these cells after TGF-β1 treatment at 37°C. Our results suggest a two phase mechanism for TGF-β1 receptor down-regulation involving receptor modulation (aggregation) at the cell surface and internalization.  相似文献

The effects of the C/N ratio, CaCO3 and PO4 addition, and temperature profile on reactor-based composting of weathered hydrocarbon-contaminated soil were evaluated in a series of 30-day tests in temperature-controlled mini-composters. Soil containing 17,000 mg (kg dry soil)–1 mineral oil and grease (MOG) was composted with maple leaves and alfalfa. Although the leaves and alfalfa also contained MOG, degradation of contaminated soil derived MOG (total MOG degradation minus MOG degradation in a control with no soil) increased from 0 to 45% as the quantity of co-substrate increased from 0 to 63%. Simulation of biopile conditions (i.e., aeration and addition of mineral salts but no co-substrate) resulted in only 6% MOG degradation. Addition of CaCO3 before composting increased total MOG degradation from 23% to 43%. Total MOG degradation increased with decreasing C/N ratio. At a molar C/N ratio of 17, 43% of the total MOG was degraded in 30 days, while at a C/N ratio of 40 there was no total MOG degradation. When temperatures ranging from 23 to 60 °C were investigated, 50 °C maintained for 29 days resulted in the maximum degradation which was 68% of total initial MOG.  相似文献

The acute toxicity of standardized extract of Orthosiphon stamineus was studied in Sprague Dawley rats. The rats were administered a single dose of 5000 mg/kg body weight (BW) orally on Day 0 and observed for 14 days. There were no deaths recorded and the animals did not show signs of toxicity during the experimental period. The effect of the extract on general behavior, BW, food and water intake, relative organ weight per 100 g BW, hematology and clinical biochemistry were measured. All the parameters measured were unaffected as compared to the control. The acute toxicity LD50 was estimated to be >5000 mg/kg BW.  相似文献

Marine sewage outfalls are the cheapest method of disposal for municipalities with restricted funds, despite their negative effects on marine ecosystems. This study was conducted on the marine sewage outfalls of the Turkish cities Rize (Black Sea) and Yalova (Sea of Marmara). Heavy metals, polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), and organochlorine pesticides (OCPs) were investigated in the sediments for determine pollution degree. Two of Yalova stations were classified as having high contamination levels (1034.436 and 1476.483 ng/g, respectively) and all the other stations suffered moderate pollution in terms of ∑16PAHs. ∑DTTs in sediments above probable effect levels (PELs), were observed both stations Yalova and Rize. The highest metal concentrations in the sediments from the eight stations ranged from 4.97 to 29.65 mg/kg for Ni; 7.57–44.14 mg/kg for Cr; 9.84–42.76 mg/kg for Cu; and 48.08–103.77 mg/kg for Zn. In sum, examination of pollutant sources provides clear evidence of an anthropogenic effect on the discharge environments.  相似文献

The objective was to examine the effect of maternal nutrient restriction followed by realimentation during midgestation on uterine blood flow (BF). On Day 30 of pregnancy, lactating, multiparous Simmental beef cows were assigned randomly to treatments: control (CON; 100% National Research Council; n = 6) and nutrient restriction (RES; 60% of CON; n = 4) from Day 30 to 140 (period 1), and thereafter, realimented to CON until Day 198 of gestation (period 2). Uterine BF, pulsatility index (PI), and resistance index (RI) were obtained from both the ipsilateral and contralateral uterine arteries via Doppler ultrasonography. Generalized least square analysis was performed. Ipsilateral uterine BF in both groups increased quadratically (P < 0.01) during period 1 and linearly (P < 0.01) during period 2. There was a treatment (P = 0.05) effect during period 2; where RES cows had greater ipsilateral BF versus CON. Ipsilateral uterine PI and RI decreased linearly (P ≤ 0.01) during period 1 across treatments. Contralateral uterine BF in CON cows tended (P < 0.09) to be greater versus RES in both periods. Contralateral PI in both groups increased linearly (P ≤ 0.01) during period 1. Contralateral uterine RI was increased (P ≤ 0.05) in RES cows versus CON in both periods. There was no interaction or treatment effect (P ≥ 0.24) for total BF during either period. Nutrient restriction does not alter total uterine BF, but it may increase vascular resistance. However, up on realimentation, local conceptus-derived vasoactive factors appear to influence ipsilateral uterine BF.  相似文献

Concentration, source, and ecological risk of polycyclic aromatic hydrocarbons (PAHs) were investigated in 22 stations from surface sediments in the areas of anthropogenic pollution in the Klang Strait (Malaysia). The total PAH level in the Klang Strait sediment was 994.02±918.1 µg/kg dw. The highest concentration was observed in stations near the coastline and mouth of the Klang River. These locations were dominated by high molecular weight PAHs. The results showed both pyrogenic and petrogenic sources are main sources of PAHs. Further analyses indicated that PAHs primarily originated from pyrogenic sources (coal combustion and vehicular emissions), with significant contribution from petroleum inputs. Regarding ecological risk estimation, only station 13 was moderately polluted, the rest of the stations suffered rare or slight adverse biological effects with PAH exposure in surface sediment, suggesting that PAHs are not considered as contaminants of concern in the Klang Strait.  相似文献

Immunocytochemical localization of Na+ channel subtypes RI and RII showed that RI immunoreactivity is relatively low and homogeneous along the rostral-caudal extent of sagittal brain sections, whereas RII staining is heterogeneous and relatively dense in the forebrain, substantia nigra, hippocampus, and cerebellum. The somata of the dentate granule cells, hippocampal pyramidal cells, cerebellar Purkinje cells, and spinal motor neurons are immunoreactive for RI but not RII. In contrast, areas rich in unmyelinated nerve fibers, such as the mossy fibers of the dentate granule cells, the stratum radiatum and stratum oriens of the hippocampus, and the molecular layer of the cerebellum, are strongly immunoreactive for RII but not RI. Differential regulation of expression of RI and RII genes may allow differential modulation of Na+ channel density in somata and axons. The sites of RI localization correlate closely with sites where sustained Na+ currents have been recorded.  相似文献

The early corpus luteum (CL) (before Day 6) does not regress after a single PGF treatment. We hypothesized that increasing PGF dose or number of treatments would allow regression of the early CL (Day 5). Nonlactating Holstein cows (N = 22) were synchronized using the Ovsynch protocol. On Day 5 (Day 0 = second GnRH treatment), cows were assigned to: (1) control (N = 5): no further treatment; (2) 1PGF (N = 6): one dose of 25 mg PGF; (3) 2PGF (N = 5): two doses of 25 mg PGF (50 mg) given 8 hours apart (second PGF on Day 5 at the same time as the other PGF treatments); (4) DPGF (N = 6): double dose of 25 mg PGF (50 mg) given on Day 5. Blood samples were collected to monitor progesterone (P4) profiles in two periods. In the first period (0 to 24 hours), there were effects of treatment (P = 0.01), time (P < 0.01), and an interaction of treatment and time (P = 0.02). Group 1PGF versus control was different only at 12 hours (P = 0.02). Cows treated with DPGF were different than control at 4 hours (P = 0.04), 12 hours (P < 0.01), and 24 hours (P < 0.01). Only cows treated with 2PGF had lower P4 than control during the entire period and low P4 (0.37 ± 0.17 ng/mL) at 24 hours, usually indicative of luteolysis. In the second period (Day 5 to 15 of the cycle), there were effects of treatment (P < 0.01), time (P < 0.01), and interaction of treatment and time (P = 0.002). Group 1PGF was not different than control from Day 5 to 13 and P4 was greater than control on Day 14 (P = 0.01) and 15 (P < 0.01). Circulating P4 in DPGF cows was lower than control from Day 7 (P = 0.05) through 12 (P < 0.01). Likewise, there were differences between control and 2PGF from Day 7 to 13, but not on Day 14 and 15. On Day 15, all PGF-treated groups had circulating P4 consistent with an active CL. Ultrasound evaluation confirmed that no CL from any group completely regressed during the experiment and no new ovulations occurred to account for functional CL later in cycle. In summary, a double dose of PGF (twice on Day 5 or 8 hours apart) can dramatically decrease P4, consistent with classical definitions of luteolysis; however, these CL recover and become fully functional. Thus, the Day 5 CL of mature Holstein cows do not regress even to two doses of PGF.  相似文献

 After having established the specificity of the antibodies for the rat testis by western blot analysis, the potential target cells for transforming growth factors (TGFβs) were identified by immunohistochemical detection of both type I (TβRI) and type II (TβRII) transducing receptors for TGFβs in the adult rat testis in situ. Leydig cells showed a strong TβRII immunoreactivity whereas the TβRI staining was weak. Only TβRII was detectable in Sertoli cells. In germ cells, staining for TβRI was stronger than for TβRII and the expression of both receptors depended on the seminiferous cycle stage. TβRI first appeared in pachytene spermatocytes and was absent in elongated spermatids from stage XIV onwards. Labelling for TβRII was observed as early as the spermatogonia stage; it increased in pachytene spermatocytes at the onset of TβRI and disappeared in elongating spermatids from stage XI onwards. These results show that TGFβs can affect somatic cells functions and suggest that these factors are involved in the control of meiosis and early spermiogenesis, exerting a direct effect on germ cells. This study examines the therapeutic and year-round prophylactic efficacy of different formulations used in dogs in three Spanish areas where canine thelaziosis is endemic. The study was conducted as a Good Clinical Practice, multicentre, randomised field study in privately owned outdoor dogs naturally infected with Thelazia callipaeda. The active pharmaceutical ingredients tested were: an oral formulation of milbemycin oxime 12.5 mg combined with praziquantel 125 mg (A), a subcutaneous sustained-release formulation of moxidectin 10 g (B), a moxidectin 2.5% weight/volume (w/v) spot-on formulation combined with imidacloprid 10% w/v (C), and an eye drop formulation (6 µg) of ivermectin 10 mg/ml diluted 10% in propylene glycol (D). Infected dogs were randomly allocated to treatment Groups A, B, C and D. Dogs testing negative for T. callipaeda inspection in two visits (Day 7/Day 14 and D30) were enrolled in the prophylaxis trial and reallocated to the corresponding study group (A, B, C or D). Treatment efficacy ranged from 70.4% recorded in Group A 1 week after treatment, to 100% recorded in Group C on Day 30 and in Group B on Day 60. Treatment was more efficacious in Group D (85.7% 1 week after treatment) than A, but was never 100% efficacious as in Groups B and C. Year-round prophylactic efficacy was 83.3% in Group A, 100% in Group B, 93.5% in Group C and 87.5% in Group D. In conclusion, products containing moxidectin were highly efficacious both in treating and preventing canine thelaziosis. Milbemycin also emerged as a good option. However, the off-label use of topical or subcutaneous ivermectin should be avoided due to possible adverse reactions such as pruritus, irritation or redness. In endemic areas, monthly prophylaxis to limit the spread of T. callipaeda to new areas across Europe and reduce zoonotic risks is essential.  相似文献

This study was designed to evaluate the reproductive performance of lactating dairy cows (Holstein Friesian) after the injection of PGF analogue on Day 15 postpartum, and GnRH analogue on Day 23 after artificial insemination (AI) with Presynch (two injections of PGF, administered 14 days apart starting at 30–35 days postpartum) + Ovsynch-based (GnRH–7 days–PGF–2 days–GnRH–16–20 hours–timed artificial insemination) treatments, during the warm and cold periods of the year. All the cows (n = 313) were assigned to one of the four groups including: M1 (n = 72) in which the cows were treated with PGF on Day 15 postpartum + Presynch-Ovsynch + GnRH on Day 23 post-AI; M2 (n = 41) in which the cows received PGF on Day 15 postpartum + Presynch-Ovsynch; M3 (n = 100) including the cows that got Presynch-Ovsynch; and control group (n = 100) including the cows that were not treated and were inseminated at natural estrus. Pregnancy diagnosis was performed 28 to 35 days post-insemination by means of ultrasound. The results showed that treatment with PGF on Day 15 postpartum significantly decreased the days to conception and the number of services per conception (P < 0.01) and it also improved the first service conception rate (P < 0.1) only in cows that were treated with M2 protocol. Whereas, the days to first service was not influenced by the treatment of PGF on Day 15 postpartum (P > 0.05). In contrast, administration of GnRH on Day 23 post-AI increased the days to conception and the number of service per conception (P < 0.01) and tended to decrease the first service conception rate (P < 0.1) in cows that were treated with M1 compared with M2 protocol. Therefore, it was concluded that Presynch-Ovsynch protocol could be more reproductive and beneficial when a single treatment with PGF was administered at 15 days postpartum (15 days after the PGF, Presynch-Ovsynch protocol was initiated). Interestingly, the administration of a GnRH agonist on Day 23 post-AI not only did not improve the reproductive performance of the cows receiving first postpartum timed artificial insemination after Presynch-Ovsynch protocol but also reduced that.  相似文献

This work was to investigate distribution characteristics, human health risk assessment, and possible sources of 16 priority polycyclic aromatic hydrocarbons (PAHs) in corn and surface soils of farmlands along main roadside in Changchun City, Jilin Province, China. Total concentrations of 16 PAHs ranged from 1572.4 to 4390.2 µg/kg with a mean value of 2954.9 µg/kg in soils and from 219.9 to 627 µg/kg with a mean value of 362µg/kg in corn. Light-molecular-weight PAHs (2–3 rings) concentration was dominant in soils, accounting for 51%, whereas high-molecular-weight PAHs (5–6 rings) concentration was highest in corn, accounting for 48%. The results of plant concentration factor indicated that high-molecular-weight PAHs have greater mobility. To evaluate potential risk to human health, hazard index (HI) and risk index (RI) were employed. The values of HI for corn and soils were both smaller than 1, indicating that exposure of PAHs posed no or little potential risk to local residents. The fact that values of RI for corn and soils were smaller than 1 × 10–4 suggested that exposure of PAHs posed no or little cancer risk to local residents. The possible sources of PAHs in corn and soils were both identified as mixture patterns of pyrogenic and petrogenic sources.  相似文献

The objective of this study was to determine the effects of progesterone and cloprostenol (a PGF analogue) on ovarian follicular development and ovulation in prepubertal heifers. In Experiment 1, crossbred Hereford heifers (Bos taurus; 10 to 12 mo old, 255 to 320 kg) were assigned randomly to three groups and given (1) an intravaginal progesterone-releasing insert (CIDR; P group, n = 13); (2) a CIDR plus 500 μg cloprostenol im (PGF analogue) at CIDR removal (PPG group, n = 11); or (3) no treatment (control group, n = 14). The CIDR inserts were removed 5 d after follicular wave emergence. Progesterone-treated heifers (P and PPG groups) had a larger dominant follicle than that of the control group (P = 0.01). The percentage ovulating was highest in the PPG group (8 of 11, 73%), intermediate in the P group (4 of 13, 31%), and lowest in the control group (1 of 14, 7%; P < 0.02). In Experiment 2, 16 heifers (14 to 16 mo old, 300 to 330 kg) were designated to have follicular wave emergence synchronized with either a CIDR and 1 mg estradiol benzoate im (EP group, n = 8) on Day 0 (beginning of experiment) or by transvaginal ultrasound-guided ablation of all follicles ≥5 mm on Day 3 (FA group, n = 8). On Day 7, CIDRs were removed in the EP group, and all heifers received 500 μg cloprostenol im. Ovulation was detected in 6 of 8 heifers (75%) in both groups. In summary, the use of PGF with or without exogenous progesterone treatment increased the percentage ovulating in heifers close to spontaneous puberty.  相似文献
