An intranuclear microsporidium is described from hemoblastic cells of the chinook salmon, Oncorhynchus tshawytscha. The infection is associated with an acute anemia in the fish. Up to 47% of the hemoblast nuclei were infected in anemic fish. The organisms, found only in spleen and kidney tissues, were 1-2 microns in diameter and consisted of vegetative and early sporulation forms. This microsporidium differs from known species which parasitize fish in its tissue location; however, the absence of mature spores and other life cycle stages precludes determination of its precise taxonomic identity.  相似文献

An unusual xenoma-forming microsporidium was discovered in the central nervous system of moribund zebrafish from a laboratory colony in Eugene, Oregon. Infected fish were often emaciated and lethargic, and histological examination commonly revealed severe myelitis and myositis associated with the infection. Based on its structure, development, and small subunit ribosomal DNA sequence it is unique among fish microsporidia. Spores are uninucleate, ovoid to pyriform, with a prominent posterior vacuole. Spores average 5.4 x 2.7 microm with 13-16 coils of the polar filament. The microsporidium produces xenomas within the spinal cord and hindbrain of fish, and xenomas contained sporophorous vesicles with up to 16 spores. Sporoblasts and presporoblast stages (probably sporonts) are found occasionally in small aggregates dispersed randomly throughout xenomas. It clustered in the "Ichthyosporidium group" along with other fish microsporidian genera based on rDNA sequence analysis. The rDNA sequence of the zebrafish microsporidium was most similar to that of Ichthyosporidium, but showed only 12.1% similarity and therefore this microsporidium can be considered a distinct genus and species, which we have named Pseudoloma neurophilia n. g., n. sp.  相似文献

ABSTRACT. Spinal and cranial ganglia of American angler fish, Lophius americanus, are often infected with microsporidia. This protozoon elicits the formation of large, spore-filled, hypertrophied host cells, cysts. Previous reports of microsporidia in European lophiids identify the parasite as Spraguea lophii, a genus which has recently been shown to be dimorphic. The spores from L. americanus are monomorphic (2.8 × 1.5 μm) and uninucleate. Each spore contains a polar tube that forms six to nine coils. Spraguea lophii differs from the microsporidium described in L. americanus in several ways. Spraguea lophii has two spore types: a large spore (4.0 × 1.25 μm) containing a diplokaryon and three to four polar tube coils and a smaller uninucleate spore (3.5 × 1.5 μm) with five to six polar tube coils. Because of these major differences, the microsporidium from L. americanus is removed from the genus Spraguea and returned to its original genus, Glugea, as a new species, G. americanus n. sp. Other ultrastructural characteristics of G. americanus are included: the posterior vacuole encloses two distinct membranous structures; one is tubular and resembles a “glomerular tuft” and the second is lamellar and composed of concentric membrane whorls, additionally, the straight or manubroid portion of the polar tube proceeds beyond the posterior vacuole before it turns anteriorly and begins to coil.  相似文献

Spore polypeptide profiles of Nosema locustae and an unidentified microsporidium infecting Aulocara elliotti and Psoloessa delicatula are compared with sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis (PAGE). Unique spore polypeptide profiles are not detected for the unknown microsporidium from A. elliotti and P. delicatula whereas these profiles are distinctly different from N. locustae spore polypeptide profiles. These results indicate that the microsporidium is not a polymorphic form of N. locustae but a separate species.  相似文献

The natural ecology of a heterosporous microsporidium, Amblyospora connecticus was investigated at three different salt marsh habitats during 1986–1989. The parasite has a well-defined seasonal transmission cycle that occurs regularly each year and intimately involves the primary mosquito host, Aedes cantator, and the intermediate copepod host, Acanthocyclops vernalis. In the spring, the microsporidium is horizontally transmitted from the copepod, where it appears to overwinter, to the mosquito via the ingestion of haploid spores produced in the copepod. Mosquitoes develop a benign infection, and females transmit the microsporidium transovarially to their progeny via infected eggs. Oviposition occurs during the summer and infected eggs hatch synchronously in the fall causing widespread epizootics. Infected larvae die, and the cycle is completed when meiospores are released into the pool and subsequently are eaten by A. vernalis, which reappears in the fall and early winter. Amblyospora connecticus thereby persists by surviving in one of two living hosts throughout most of its life cycle rather than in the extra-corporeal environment. This represents an important survival strategy for A. connecticus as results show the salt marsh habitat to be a relatively unstable environment that is subject to periodic flooding and drying. The adaptive significance of utilizing an intermediate host in the life cycle is discussed as it directly facilitates transmission and enhances survival of the microsporidium.  相似文献

Characteristic configurations of the nuclei and synaptonemal complexes, indicative of the onset of meiosis, were observed in the meronts of the monomorphic diplokaryotic microsporidium, Paranosema grylli. This finding indicates that a process similar to the meiosis previously reported in polymorphic and some monomorphic monokaryotic microsporidia probably occurs in the development of P. grylli. It is the first evidence for the possible presence of a sexual phase in the life cycle of this microsporidium, which for a long time has been considered asexual.  相似文献

A new genus, Endoreticulatus n. g., is described for the microsporidium Pleistophora fidelis (Hostounský & Weiser, 1975) based on light and electron microscopic studies of a microsporidium in the Colorado potato beetle, Leptinotarsa decemlineata (Say). This latter microsporidium is considered to be conspecific with P. fidelis because both isolates have been shown to be infectious for L. decemlineata where infection is limited to the epithelial cells of the midgut; both are haplokaryotic and develop as groups of sporoblasts and spores in subpersistent vacuoles in the host cell. In addition, meronts, sporonts, and spores of each isolate often occur simultaneously in a common cell, and by light microscopy they both appear similar. Ultrastructural studies of the isolate from L. decemlineata revealed that all developmental stages occur in parasitophorous vacuoles derived from cisternae of rough endoplasmic reticulum of the host cell cytoplasm. Based on the unique nature of the parasitophorous vacuole, a new genus, Endoreticulatus, is proposed for P. fidelis. The genus is compared with the other genera whose species undergo multisporous sporogony in sporophorous vesicles. In addition, the nature of the parasitophorous vacuole of Endoreticulatus fidelis (Hostounský & Weiser, 1975) n. comb. is compared with the parasitophorous vacuole known to encase various developmental stages of several other microsporidian species.  相似文献

Convergent lady beetles, Hippodamia convergens Guérin-Méneville, are a popular choice for aphid control in North America. An unidentified microsporidium was found in H. convergens adults that were purchased from a commercial insectary in 2004. This study examined egg cannibalism and egg predation as a means of horizontal transmission of the unidentified microsporidium among H. convergens larvae and three coccinellid species found in Nova Scotia: Coccinella septempunctata (seven-spotted lady beetle), C. trifasciata perplexa (three-banded lady beetle), and Harmonia axyridis (multicolored Asian lady beetle). The microsporidium was transmitted with 100% efficiency when first instars fed on microsporidia-infected eggs. Mean spore count data from smear preparations of infected beetles suggest that the infection was as heavy in C. trifasciata perplexa (a native coccinellid) (11.2 ± 0.96 spores/100 μm2) as it was in H. convergens (the natural host) (12.8 ± 1.16) but lighter in the introduced species C. septempunctata (7.5 ± 0.65) and H. axyridis (0.8 ± 0.11). For all of the beetle species examined, larval development was significantly longer for microsporidia-infected individuals than for their uninfected cohorts. The microsporidium had no effect on larval mortality. Based on the results of this study, field-collected H. convergens should be examined for microsporidia and uninfected individuals should be used to rear individuals for release in biological control programs. However, this is unlikely to happen because H. convergens are relatively easy and inexpensive to collect from their overwintering sites for redistribution.  相似文献

The microsporidium Nudispora biformis n. g., n. sp., a parasite of a larva of the damsel fly Coenagrion hastulatum in Sweden, is described based on light microscopic and ultrastructural characteristics. Merogonial stages and sporonts are diplokaryotic. Sporogony comprises meiotic and mitotic divisions, and finally eight monokaryotic sporoblasts are released from a lobed plasmodium. Sporophorous vesicles are not formed. The monokaryotic spores are oval, measuring 1.4–1.8 × 2.8–3.4 μm in living condition. The thick spore wall has a layered exospore, with a median double-layer. The polaroplast has two lamellar parts, with the closest packed lamellae anteriorly. The isofilar polar filament is arranged in 6 (to 7) coils in the posterior half of the spore. Laminar and tubular extracellular material of exospore construction is present in the proximity of sporogonial stages. In addition to normal spores teratological spores are produced. The microsporidium is compared to the microsporidia of the Odonata; its possible relations to the genus Pseudothelohania and to the Thelohania-like microsporidia are discussed. The new genus is provisionally included in the family Thelohaniidae.  相似文献

The microsporidian Trachipleistophora hominis was isolated in vitro from the skeletal muscle of an AIDS patient. Since its discovery several more cases of myositis due to Trachipleistophora have been diagnosed but the source of infection is unknown. Morphologically, T. hominis most closely resembles Pleistophora and Vavraia, which undergo polysporous sporogony in sporophorous vesicles, but differs from these genera in the mode of formation of sporoblasts and in the morphology of the sporophorous vesicles. Alignment and analyses of the small subunit ribosomal DNA sequences of T. hominis and several other polysporoblastic genera indicated that its closest phylogenetic relationships were with species of the genera Pleistophora and Vavraia, in line with morphological predictions. The type species of the latter two genera are Pleistophora typicalis and Vavraia culicis; these are parasites of fish and mosquitoes, respectively. These results suggest two possible routes and sources of infection to AIDS patients, these being perorally by ingestion of inadequately cooked fish or crustaceans or percutaneously during a bloodmeal taken by a haematophagous insect. Support for an insect source has been provided by recent detection of a microsporidium from mosquitoes in human corneal tissue.  相似文献

Parasite infections are common during the critical developmental period in children. The occurrences of intestinal parasites are also common in orphanage, nurseries and schools in Turkey. The study was carried out to determine the percentage of microsporidium and intestinal parasites in children from Malatya, Turkey. This study was carried out at the Department of Parasitology of Inonu University, Turgut Ozal Medical Center, during January–December 2006. Totally, 1,181 stool samples were examined using the native-Lugol, sedimentation-techniques, modified trichrome (MTS), acid-fast-trichrome stain and calcofluor staining methods. In addition, perianal region material was taken from the children to examine with cellophane tape method. Power analyses were performed for statistical analyses used. Microsporidia were found in 92 (7.8%) of the samples, and also intestinal parasites were detected in 329 (27.8%). The numbers of infections according to the species were as follows: 69 (5.8%) Entamoeba coli, 7 (0.6%) Blastocystis hominis, 114 (9.7%) Giardia intestinalis, 15 (1.3%) Iodomoeba butchlii, 8 (0.7%) Dientamoeba fragilis, 7 (0.6%) Taenia spp. 70 (5.9%) Enterobius vermicularis, 11 (0.9%) Hymenolepis nana, 25 (2.1%) Trichomonas intestinalis, 1 (0.1%) Ascaris lumbricoides and 2 (0.2%) Chilomastix mesnilii. Also, greater than 90% power values were achieved for statistical analyses. Whereas the detection rates of microsporidium and intestinal parasites were found to be low, it was concluded that in addition to intestinal parasites, microsporidium should be also searched for in children with complaints of intestinal system.  相似文献

We describe a unique microsporidian species that infects the green stink bug, Chinavia hilaris; the brown marmorated stink bug, Halyomorpha halys; the brown stink bug, Euschistus servus; and the dusky stink bug, Euschistus tristigmus. All life stages are unikaryotic, but analysis of the consensus small subunit region of the ribosomal gene places this microsporidium in the genus Nosema, which historically has been characterized by diplokaryotic life stages. It is also characterized by having the reversed arrangement of the ribosomal gene (LSU –ITS‐ SSU) found in species within the “true Nosema” clade. This microsporidium is apparently Holarctic in distribution. It is present in H. halys both where it is native in Asia and where it is invasive in North America, as well as in samples of North American native C. hilaris collected prior to the introduction of H. halys from Asia. Prevalence in H. halys from mid‐Atlantic, North America in 2015–2016 ranged from 0.0% to 28.3%, while prevalence in C. hilaris collected in Illinois in 1970–1972 ranged from 14.3% to 58.8%. Oral infectivity and pathogenicity were confirmed in H. halys and C. hilaris. Morphological, ultrastructural, and ecological features of the microsporidium, together with a molecular phylogeny, establish a new species named Nosema maddoxi sp. nov.  相似文献

Microsporidia are opportunistic pathogens that infect a wide range of invertebrates and vertebrates. To assess the potential role of dogs in the transmission of these zoonotic pathogens, a total of 282 fecal samples from dogs in the Central Anatolia Region of Turkey were analyzed by utilizing species specific polymerase chain reaction for the four most frequent human microsporidia. Two microsporidia species were recognized in 41 samples (14.5%). Encephalitozoon intestinalis was detected in 35 samples (12.4%) and it was the most common microsporidium. The second microsporidium, E. cuniculi, was identified in six (2.1%) of the samples. Sequence analysis of the intergenic spacer of the ribosomal ribonucleic acid (RNA) internal transcribed spacer (ITS) gene revealed the presence of three E. intestinalis haplotypes closely associated with each other. No polymorphic region was found among the ITS sequences of E. cuniculi isolates and they were characterized as genotype III. This study provides the first data on the zoonotic microsporidia species from dogs in Turkey.  相似文献

An undescribed Nosema was found infecting adults of the mecopteran Hylobittacus apicalis. This microsporidium is described herein as the first record of a microsporidium from the order Mecoptera. The slightly pyriform spores measured 4.5 × 2.4 μm. Mature spores had 9.5–10 polar filament coils irregularly grouped in the posterior end. The life cycle and ultrastructure of the developmental stages were described, and were typical of other Nosema spp. This microsporidium was regularly recorded from adult Hylobittacus apicalis populations over a 10-year period and tbe incidence of infection increased during the summer.  相似文献

Microsporidia are highly successful parasites that infect virtually all known animal lineages, including the model Danio rerio (zebrafish). The widespread use of this aquatic model for biomedical research has resulted in an unexpected increase in infections from the microsporidium Pseudoloma neurophilia, which can lead to significant physical, behavioral, and immunological modifications, resulting in nonprotocol variation during experimental procedures. Here, we seek to obtain insights into the biology of P. neurophilia by investigating its genome content, which was obtained from only 29 nanograms of DNA using the MiSeq technology and paired‐end Illumina sequencing. We found that the genome of P. neurophilia is phylogenetically and genetically related to other fish‐microsporidians, but features unique to this intracellular parasite are also found. The small 5.25‐Mb genome assembly includes 1,139 unique open‐reading frames and an unusually high number of transposable elements for such a small genome. Investigations of intragenomic diversity also provided strong indications that the mononucleate nucleus of this species is diploid. Overall, our study provides insights into the dynamics of microsporidian genomes and a solid sequence reference to be used in future studies of host–parasite interactions using the zebrafish D. rerio and P. neurophilia as a model.  相似文献

Few microsporidia have been reported from whitefish species (subfamily Coregoninae). For the most part, these microsporidia have been incompletely described. In a survey of parasites of mountain whitefish Prosopium williamsoni collected from Kootenay Lake, British Columbia, we encountered an unusual microsporidium infecting the endomysium of the skeletal musculature. Spores were uninucleate, ovoid to pyriform, and were 5.6 (5-7) microm x 3.2 (3-4) microm with 13-16 coils in the polar filament. We describe here this organism as a new species based on its site of development and its relationship among fish microsporidia based on small subunit ribosomal DNA sequence data, i.e., our analysis showed that it is not closely related to other microsporidia for which ribosomal DNA sequence is available thus far.  相似文献

ABSTRACT. The SSUrDNA and the ITS of different microsporidia from eight fishes, four insects and a shrimp were amplified and digested with restriction enzymes. The generated riboprints suggest a close evolutionary relationship between Glugea americanus and Spraguea lophii suggesting that Glugea americanus should be renamed Spraguea americanus and that the tissue infected and host origin should be considered of greater taxonomic importance for defining a genus than previously considered. Phylogenetic analysis of the riboprints demonstrates an unidentified microsporidium from a bumper fish ( Chloroscombrus chrysurus ) is related although not identical to Microgemma oviodea , a parasite from red band fish. We were also able to distinguish between Glugea anomala and Glugea atherinae and Glugea stephani but were not able to differenciate among the latter two. Insects isolates, Nosema costelytrae, N. bombycis, N. trichoplusiae, Nosema sp. and a shrimp isolate, Agmasoma penaei , are not related to the fish isolates.  相似文献
