In studies on the effect of pH and temperature on cellulolytic and pectolytic activity of C. destructans, it was found that the isolates used produced only endoglucanases. The temperature and pH affected the synthesis of these enzymes. Fungi cultured at 26°C produced more of these enzymes than those grown at the two other temperatures. At 10°C, only one isolate produced minute amounts of endoglucanases. None of fungi studied exhibited cellulolytic activity in cultures grown at 20°C. Cellulolytic activity was found only in acidic media (pH 5.0). The fungi studied exhibited higher pectolytic than cellulolytic activity. In the post culture liquids of these organisms, both types of pectolytic enzymes (exo- and endo-PMG) were detected. Different temperature and pH values affected the production of these enzymes differently in various isolates.  相似文献

The production of cytokinin-like substances by different strains of Cylindrocarpon destructans either pathogenic or non pathogenic to fir (Abies alba Mill.) was studied. The cytokinins were extracted from the fungal culture media and separated by means of column chromatography. The biological activities in different fractions were assayed by callus growth of soybean (Glycine max Merill var. Acme). The amount of these substances was calculated on the basis of fresh weight of callus obtained from cultures containing authentic cytokinin. Cytokinin-like substances were detected in 5 of 6 pathogenic isolates tested and in 2 of 5 non-pathogenic isolates. On the basis of their occurrence in particular column fractions, most of the substances seem to be identical or similar to zeatin and zeatin riboside. In addition the fungi studied produced other, not identified cytokinin-like substances. The synthesis of Ri P can not be excluded. However the method applied did not allow to separate Ri P from zeatin and zeatin riboside.  相似文献

Culture fluids obtained from submerged cultures of white, brown and gray wood rot fungi were assayed for the presence of cellulolytic activity complexes against the model substrated carboxymethylcellulose-Na and Standard Whatman cellulose and natural substrates, i.e. celluloses isolated from pine bark and sawdust. The cellulolytic activity of the examined fungal species was highly differentiated. The use of model and natural substrates allowed determination of the high substrate specificity of the cellulase complexes produced by the fungi. Not all the fungi were found to produce EC 3.2.1.4. endo-1, 4-beta-glucanase under the culture conditions employed. All the fungi were, however, able to produce a complex of EC 3.2.1.4. exo-1, 4-beta-glucanases. All the examined fungi were also able to degrade, although to a varied extent, such higher forms of cellulose as Standard Whatman cellulose or natural celluloses isolated from pine bark and sawdust. Determination of the cellulolytic activity of fungi against the above-mentioned specific natural substrates affords the possibility of their practical use.  相似文献

Marchalina hellenica (Gennadius) (Hemiptera: Margarodidae) is a scale insect, endemic in pine (Pinus halepensis) forests of Greece and other Mediterranean countries, which plays a major role in the production of honeydew honey. We investigated the morphological and the biological characteristics of M. hellenica in the pine forests of Mt. Parnis in comparison with those in the high-altitude fir (Abies cephalonica) forest of Mt. Helmos where it has been recently established, after anthropogenic intervention. Morphologically, the final body size of the 1st instar and the adult stages of M. hellenica in the fir forest were equal to those in the pine forest. Biologically, in the fir forest, the insect exhibited a long 1st instar’s period, which was the stage of its overwintering. In the pine forest, the 1st instar period was short and the insect overwinters in the stage of 2nd instar. The number of eggs per female in the fir forest was quite low (25–145) compared with the number of eggs in the pine forest (200–300). Concerning the qualitative parameters of the resulting honeydew-honey, statistically significant differences were found in Diastase and HMF (p < 0.0001). No difference was found in sucrose, fructose + glucose and water content, neither to electrical conductivity and total acidity. All values were within the EU limits (EU Directive 2001).  相似文献

The goal of this study is to identify and characterize the cellulose degrading microorganisms in the larval gut of the white grub beetle, Lepidiota mansueta. Thirty bacterial strains were isolated and tested for cellulolytic activity using soluble carboxymethyl cellulose (CMC) degrading assays. Of these strains, five (FGB1, FB2, MB1, MB2, and HB1) degrade cellulose. Cellulolytic activity was determined based on formation of clear zone and cellulolytic index on CMC plate media. The highest cellulolytic index (2.14) was found in FGB1. Partial 16S rDNA sequencing, morphological, and biochemical tests were used to identify and characterize the five isolates, all Citrobacter sp. (Enterobacteriaceae). This study identifies new cellulose degrading microorganisms from the larval gut of L. mansueta. The significance of identifying these strains lies in possible application in cellulose degradation.  相似文献

Abstract

The study reports the characterization of 10 isolates of mulberry black root rot causing fungus, Botryodiplodia theobromae obtained from the infected gardens of Karnataka, Andhra Pradesh and Tamil Nadu. The analysis based on cultural, morphological, pathogenicity and molecular markers (RAPD and SSRs) revealed significant variations among the isolates. Based on the disease reaction on susceptible V-1 variety, isolates were grouped as pathogenic (60%), moderate pathogenic (20%) and non-pathogenic (20%). Among all isolates, RAPDs revealed higher marker polymorphism however, based on Shannon’s Information Index (I) SSRs were more informative (0.781) compared to the former (0.444). Stepwise Multiple Regression Analysis (MRA) indicated a total of 5, 5 and 3 molecular markers were found to correlate with disease symptoms. Screening of germplasm using multiple strains of virulent isolates will enhance possibilities of locating diverse resistant genes. Pyramiding of these genes will aid in development of mulberry variety with durable resistance and sustainable sericulture.  相似文献

Pink snow mould is a serious disease on grasses and winter cereals in cold and temperate zones during winter. To better understand the basis for the variation in pathogenicity between different isolates of Microdochium nivale and M. majus and to simplify selection of highly pathogenic isolates to use when screening for resistance to pink snow mould in perennial ryegrass, we sought traits correlated with pathogenicity. Isolates of M. nivale were more pathogenic on perennial ryegrass than isolates of M. majus, as measured by survival and regrowth of perennial ryegrass after infection and incubation under simulated snow cover. Pathogenicity as measured by relative regrowth was highly correlated with fungal growth rate on potato dextrose agar (PDA) at 2°C. Measuring fungal growth on PDA therefore seems to be a relatively simple method of screening for potentially highly pathogenic isolates. In a study of a limited number of isolates, highly pathogenic isolates showed an earlier increase and a higher total specific activity of β‐glucosidase, a cell wall‐degrading enzyme, compared with less pathogenic isolates. None of the M. majus isolates was highly pathogenic on perennial ryegrass. Our results indicate biological differences between M. nivale and M. majus and thus strengthen the recently published sequence‐based evidence for the elevation of these former varieties to species status.  相似文献

为更好地弄清草鱼(Ctenopharyngodon idella)肠道纤维素降解细菌的种类,采用羧甲基纤维素(CMC)作为唯一碳源的选择性培养基,分别从草鱼肠道内容物和肠道黏膜中分离到了40株产纤维素酶细菌。16S rRNA基因序列的分析结果显示,大多数产纤维素酶细菌为气单胞菌属(Aeromonas)的种类,其次为肠杆菌属(Enterobacter)的细菌以及未经分离纯培养的细菌(Uncultured bacterium)。进一步研究细菌产纤维素酶能力发现,纤维素酶活性显著性高于其他菌株的分别是A. veronii MC2、A. veronii BC6、肠杆菌科(Enterobacteriaceae)中一种未经分离纯培养的细菌BM3(Uncultured bacterium BM3)和A. jandaei HC9。草鱼肠道中简答气单胞菌(A. jandaei)、类志贺邻单胞菌(Plesiomonas shigelloides)、阴沟肠杆菌(E. cloacae)以及产气肠杆菌(E. aerogenes)是被作为产纤维素酶细菌的首次报道。    相似文献

Nine thermophilic cellulolytic clostridial isolates and four other noncellulolytic bacterial isolates were isolated from self-heated biocompost via preliminary enrichment culture on microcrystalline cellulose. All cellulolytic isolates grew vigorously on cellulose, with the formation of either ethanol and acetate or acetate and formate as principal fermentation products as well as lactate and glycerol as minor products. In addition, two out of nine cellulolytic strains were able to utilize xylan and pretreated wood with roughly the same efficiency as for cellulose. The major products of xylan fermentation were acetate and formate, with minor contributions of lactate and ethanol. Phylogenetic analyses of 16S rRNA and glycosyl hydrolase family 48 (GH48) gene sequences revealed that two xylan-utilizing isolates were related to a Clostridium clariflavum strain and represent a distinct novel branch within the GH48 family. Both isolates possessed high cellulase and xylanase activity induced independently by either cellulose or xylan. Enzymatic activity decayed after growth cessation, with more-rapid disappearance of cellulase activity than of xylanase activity. A mixture of xylan and cellulose was utilized simultaneously, with a significant synergistic effect observed as a reduction of lag phase in cellulose degradation.  相似文献

Pathogenic and non-pathogenic isolates of Streptococcus suis type 2 were screened to determine whether differences in superoxide dismutase (SOD) synthesis could explain the observed differences in their pathogenicity and intracellular fate in macrophages. A single band of SOD activity of similar Rf value was visualised in PAGE gels in all isolates and inhibition studies suggested that the cofactor present was manganese. There was no correlation between specific SOD activity and virulence. It is unlikely, therefore, that SOD produced by S. suis type 2 mediates intracellular survival of pathogenic isolates in macrophages.  相似文献

Cellulase production by Aureobasidium pullulans from the temperate regions has remained speculative, with most studies reporting no activity at all. In the current study, tropical isolates from diverse sources were screened for cellulase production. Isolates were grown on a synthetic medium containing cell walls of Msasa tree (Brachystegia sp.) as the sole carbon source, and their cellulolytic activities were measured using carboxymethyl cellulose and alpha-cellulose as substrates. All isolates studied produced carboxymethyl cellulase (endoglucanase) and alpha-cellulase (exoglucanase) activity. Endoglucanase-specific activities of ten selected isolates ranged from 2.375 to 12.884 micromol glucose.(mg protein)-1.h-1, while activities on alpha-cellulose (exoglucanase activity) ranged from 0.293 to 22.442 micromol glucose.(mg protein)-1.day-1. Carboxymethyl cellulose induced the highest cellulase activity in the selected isolates, while the isolates showed variable responses to nitrogen sources. The current study indicates that some isolates of A. pullulans of tropical origin produce significant extracellular cellulolytic activity and that crude cell walls may be good inducers of cellulolytic activity in A. pullulans.  相似文献

The main aim was to evaluate the occurrence of cellulolytic bacteria from the Stain house Lake, located at Admiralty Bay, Antarctica. Thick cotton string served as a cellulose bait for the isolation of bacteria. A total of 52 bacterial isolates were recovered and tested for their cellulase activity, and two of them, isolates CMAA 1184 and CMAA 1185, showed significant cellulolytic activity on carboxymethylcellulose agar plates. Phylogenetic analysis placed the isolates into the Bacillus 16S ribosomal RNA gene subclade. Both isolates produced a cold-active cellulase which may play a crucial role in this extreme environment.  相似文献

Summary The extent and type of chloroplast DNA restriction fragment length polymorphism was determined among individual tree samples of coast redwood, Douglas fir, incense-cedar, and loblolly pine. A total of 107 trees was surveyed for three restriction enzymes (BamHI, EcoRI, HindIII) and six chloroplast DNA probes from petunia (P3, P4, P6, P8, P10, S8). The probes comprise 64% of the petunia chloroplast genome. Polymorphisms were detected in all species but loblolly pine. Coast redwood and incense-cedar had a small number of rare variants, whereas Douglas fir had one highly polymorphic region of insertions/deletions in sequences revealed by the P6 probe from petunia. The mutation hotspot is currently being studied by DNA sequence analysis.  相似文献

Summary We report the complementation of a genetic lesion in the genome of Erwinia carotovora subsp. carotovora (Ecc), a pathogenic bacterium that incites soft rot of plants. A Sau3AI genomic library of Ecc was constructed using the conjugal cosmid pLAFR-3 as a vector. Sixteen cosmid clones encoding various plant tissue-degrading enzymes were identified, including a proteolytic clone, five cellulolytic clones, and ten pectolytic clones. We detected a mutant of Ecc with no proteolytic activity following transposon mutagenesis with an unstable Tn5-carrying plasmid. Conjugal transfer of the protease-encoding cosmid to this mutant restored near-wildtype extracellular protease production. Further manipulation and study of genes encoding pathogenic determinants in Ecc will be possible using this system.  相似文献

The community of culturable cellulolytic bacteria was analyzed in two long-term experimental field sites on Albic Luvisol (silty sand) and Haplic Phaeozem (loam), with and without farmyard manure treatment. Against the backdrop of significant differences in soil properties, the bacterial community structure differed clearly between sites and was affected by manure application as analyzed by T-RFLP of 16S rDNA. The population densities of cellulolytic bacteria were significantly increased by manure application in Phaeozem. Cellulose decomposing potentials of 537 isolates were tested on soluble, colloidal, and crystalline cellulose. The results showed some evidence of a greater proportion of isolates with high decomposition activity in Luvisol, but no impact from manure application could be observed in both soils. Restriction analysis and sequencing of 16S rDNA of isolates revealed a rather simple community composition that was dominated by Streptomyces (67%). The composition of the RFLP groups was affected by manure application, which was most evident in Luvisol, whereas an effect of the soil type could not be found. Although abundant RFLP groups were assigned to phylogenetically different bacterial classes (Actinobacteria, Betaproteobacteria, and Gammaproteobacteria), cellulolytic activity could not consistently be differentiated. All in all, cellulolytic capabilities of the isolates were highly variable and did not map to phylogenetic affiliation.  相似文献

A clostridial bacterium is accepted to be one of the important and efficient microorganisms for the application in fuel fermentation process. However, the lack of cellulolytic activity of cellulosome in this organism appears to be one of the main important problems for efficient production of the fuel. It is therefore interesting to search for the genetic resource of natural clostridial bacteria for the application in bioengineering. Presently, Clostridium species selection and identification are based on various physiological properties tests. This article developed the way for a 4-step screening process via mainly three criteria and 16S rDNA identification. In this study, solvent-producing clostridial bacteria were successfully isolated from decomposed sources, cow feaces, and dry grass in Thailand. Anaerobes were screened by cellulolytic activity and butanol tolerance in selective media that composed of basal media supplemented with 2% cellulose and 5% butanol. Thirty isolates of cellulolytic and butanol-tolerant anaerobic bacteria were obtained from screening in this medium. Fifteen isolates were rapidly classified as in the class Clostridia by three selected criteria (endospore formation, sulfite-reducing ability, and metabolic products). Secondary metabolites of the bacteria such as acetone, butanol and ethanol were varied depending on the process. Clostridial differential medium was used as a genus identification tool. Finally, PCR-amplified gene fragments coding for 16S rDNA were analyzed as a key to identify bacteria species. This process can be used to screen and identify Clostridium species in short period. Cellulosome and non-cellulosome cellulases productivity were analyzed. The results revealed that the selected cellulolytic strains (such as Fea-PA) exhibited EngD non-cellulosome cellulase activity especially endoglucanase activity on carboxymethyl cellulose. The selective system in this research was appropriate for the screening of Clostridiaceae in a similarity range between 83% and 100%.  相似文献

Soft rot is the most important disease on calla lily in Poland. The isolation of the presumptive pathogen from symptomatic tubers on nutrient agar yielded bacteria with different colony morphology. Of 41 isolates collected, 10 showed pectolytic activity on crystal violet pectate medium and caused soft rot on potato slices. All pectolytic bacteria appeared to be Gram‐negative rods producing typical soft rot on inoculated leaf petioles of calla lily. Bacteria with colonies which morphologically resembled those used for inoculation were re‐isolated from diseased petioles. Their identification was based on phenotypic characters and sequence of the gene fragment coding 16S rRNA. It was found that, in addition to Pectobacterium carotovorum subsp. carotovorum, soft rot of calla lily can be caused by Pectobacterium carotovorum subsp. atrosepticum, Pseudomonas marginalis, Pseudomonas veronii and Chryseobacterium indologenes. The latter two are described for the first time as plant pathogens. The pectolytic activity of all identified bacteria, except that of P. carotovorum subsp. atrosepticum, was lower than that of P. carotovorum subsp. carotovorum, but strains of P. veronii showed a higher activity than P. marginalisand C. indologenes species.  相似文献

This is the first report of isolation of fungi present in fatty and defatted castor bean meal as well as the first of crop’s selection to test the cellulolytic potential, in order to verify the diversity and potential of cellulolytic fungi in castor bean waste (Ricinus communis L.). For the screening on solid medium, it was used carboxymethylcellulose (CMC) as the sole carbon source. The microcrystalline cellulose (Avicel) was used as a substrate for submerged fermentation for production of cellobiohydrolase (FPase) and the CMC to produce endoglucanases (CMCase) and β-glycosidases (BG). 189 cultures of fungi were isolated, including 40 species of filamentous fungi and three yeasts. The Aspergillus was the most frequent found genus. Regarding the distribution of isolated species from defatted castor bean meal, the A. niger was the most frequent one; and within the fatty castor bean meal, the Emericela variecolor prevailed among other species. Among the 67 fungal cultures tested in the initial screening on solid media to assess the cellulolytic potential, 54 disclosed Cellulolytic Index (CI) ranging from 1.04 to 6.00 mm. The isolates were selected for enzyme production in liquid medium with values above 2.0 CI. They were obtained with A. japonicus URM5620 FPase activity (4.99 U/ml) and BG (0.05 U/ml), and Rhodotorula glutinis URM5724 activity of CMCase 3.58 U/ml. These cases occurred after 168 h of submersion for both species of fungi. In our study, we could conclude that the castor bean is a promising source of fungi capable of producing cellulolytic enzymes.  相似文献

The virulence and vegetative compatibility of eight Dutch and four Italian isolates of Fusarium oxysporum obtained from lily were compared. The virulence was tested by determination of the specific interaction between the Fusarium isolates and eight lily cultivars. A specific interaction was not found, so the existence of races was not demonstrated. Six of the twelve isolates turned out to be non-pathogenic for lily. The pathogenic isolates fell in four vegetative compatibility groups. No vegetative compatibility was found between isolates of F. oxysporum f. sp. lilii and those of f. sp. gladioli.  相似文献

Pectolytic enzyme formation by whole cells of Erwinia aroideae was markedly stimulated when nalidixic acid was added to a culture medium. The activity of pectolytic enzyme was markedly stimulated by nalidixic acid when the activity was measured by the decrease of viscosity of pectin, while activities of both polygalacturonic acid trans-eliminase and polygalacturonase which were measured respectively by the increase of optical density at 230 nm and the liberation of aldehyde groups, were not stimulated. The analysis of pectolytic enzyme by carboxymethyl cellulose column chromatography indicated that there was a significant difference in the elution profiles between the pectolytic enzyme induced by nalidixic acid and that synthesized under normal conditions. Therefore, we conclude that two enzymes are distinct protein species.  相似文献
