Accumulation of misfolded proteins as insoluble aggregates occurs in several neurodegenerative diseases. In Parkinson's disease (PD) and dementia with Lewy bodies (DLB), alpha-synuclein (alpha S) accumulates in insoluble inclusions. To identify soluble alpha S oligomers that precede insoluble aggregates, we probed the cytosols of mesencephalic neuronal (MES) cells, normal and alpha S-transgenic mouse brains, and normal, PD, and DLB human brains. All contained highly soluble oligomers of alpha S whose detection was enhanced by delipidation. Exposure of living MES neurons to polyunsaturated fatty acids (PUFAs) increased alpha S oligomer levels, whereas saturated FAs decreased them. PUFAs directly promoted oligomerization of recombinant alphaS. Transgenic mice accumulated soluble oligomers with age. PD and DLB brains had elevated amounts of the soluble, lipid-dependent oligomers. We conclude that alpha S interacts with PUFAs in vivo to promote the formation of highly soluble oligomers that precede the insoluble alpha S aggregates associated with neurodegeneration.  相似文献

Numerical changes in dictyosomes during the formation of autospores in a green alga, Chlorococcum infusionum, were investigated by electron microscopy. Two dictyosomes were seen near the nucleus in young vegetative cells. Four dictyosomes were seen in large mononucleate cells which appeared to enter mitosis soon. Binucleate cells contained 4 or 8 dictyosomes, the latter number being found in the large binucleate cells. Large tetranucleate cells contained 16-25 dictyosomes in each cell. Dictyosomes consisted of about 4 cisternae with a diameter of 0.4-0.6 micron in mono- to tetranucleate cells. Cytokinesis began with the formation of the septa during the third nuclear division, and 16 cells were finally formed. Dictyosomes did not increase in number in 8- and 16-nucleate cells. In septum-forming cells, dictyosomes were 0.6-1.0 microns in diameter, with 6-9 cisternae. A single dictyosome was included in each of the 16 resultant cells. These observations suggest that the dictyosomes multiply in association with the multiplication of the nuclei without correlation with formation of the cell wall or cytokinesis.  相似文献

We find a differential distribution of dictyosomes in the avascular tip cells of the oat (Avena sativa) coleoptile upon geostimulation. A differential activation (increased vesicle production) of the dictyosomes with respect to gravity also occurs, but only in the tip cells of the lower tissues. Similar differences in distribution and activation of dictyosomes occur also in cells subjacent to the avascular tip (1st and 5th millimeter from the apex) of both the upper and lower half-tissues. When only the outer epidermal cells below the apex are considered, the differential distribution and activation of dictyosomes occur only in the lower outer epidermis. The changes in distribution of dictyosomes begin at 6 minutes, or sooner, from the start of geostimulation, before the onset of geotropism. The kinetics of amyloplast sedimentation and Golgi movement do not appear to differ in the cells of the avascular tip. We suggest that the Golgi participates in, and possibly initiates, the differential elongation of cells of geotropically stimulated coleoptiles.  相似文献

Visual hallucinations (VH) represent one of the core features in discriminating dementia with Lewy bodies (DLB) from Alzheimer’s Disease (AD). Previous studies reported that in DLB patients functional alterations of the parieto-occipital regions were correlated with the presence of VH. The aim of our study was to assess whether morphological changes in specific cortical regions of DLB could be related to the presence and severity of VH. We performed a cortical thickness analysis on magnetic resonance imaging data in a cohort including 18 DLB patients, 15 AD patients and 14 healthy control subjects. Relatively to DLB group, correlation analysis between the cortical thickness and the Neuropsychiatric Inventory (NPI) hallucination item scores was also performed. Cortical thickness was reduced bilaterally in DLB compared to controls in the pericalcarine and lingual gyri, cuneus, precuneus, superior parietal gyrus. Cortical thinning was found bilaterally in AD compared to controls in temporal cortex including the superior and middle temporal gyrus, part of inferior temporal cortex, temporal pole and insula. Inferior parietal and supramarginal gyri were also affected bilaterally in AD as compared to controls. The comparison between DLB and AD evidenced cortical thinning in DLB group in the right posterior regions including superior parietal gyrus, precuneus, cuneus, pericalcarine and lingual gyri. Furthermore, the correlation analysis between cortical thickness and NPI hallucination item scores showed that the structural alteration in the dorsal visual regions including superior parietal gyrus and precuneus closely correlated with the occurrence and severity of VH. We suggest that structural changes in key regions of the dorsal visual network may play a crucial role in the physiopathology of VH in DLB patients.  相似文献

Summary Transformation of the Golgi apparatus inMicrasterias americana at various stages after full growth and at the earliest stage of cell growth was investigated using an electron microscope. Silver-hexamine staining and the acid phosphatase (ACPase) test were also carried out. In cells cultured for two days after full growth, dictyosomes began to produce hypertrophied vesicles (HVs) along their five peripheral reagions. The HVs contained fibrous material, which was stained by silver-hexamine, and small granules which reacted with ACPase. The HVs were pinched off the dictyosomes and fused with each other and with the vacuoles. In the earliest stage of cell growth, the cisternae of the dictyosomes were stretched in one direction, which modified the shape from circular to elliptical and the dictyosomes curved along the long axis of the ellipse. These curved dictyosomes which produced middle sized vesicles (MVs) from the distal networks, divided into two identical parts along the short axis of the ellipse.  相似文献

The polymorphism of the dictyosomes in the root meristeme ofFagopyrum is connected with their various functions in secretory processes and cell differentiation. The dictyosomes containing vesicular dilatations of the cisternae, which in this object occur more frequently than in others, probably participate in a similar way as the Golgi apparatus of the animal cell in the formation of lysozomes, in the formation of elements belonging to the group of dense bodies analogical lysozomes. These bodies are present in large numbers in the cytoplasm of cells, containing dictyosomes with vesicular dilatations. The other forms of the dictyosomes reveal indications of their participation in the production of the carbohydrate material of the cell walls, like most dictyosomes of other plant objects. However, no fusion of the Golgi vesicles with the plasmalemma was observed. According to their morphological appearance the typical forms of dictyosomes were classified on the basis of their relationship to secretory processes. Simultaneously the morphology and function of the Golgi apparatus was compared in the animal and plant cell. Several morphological varieties of the dictyosomes of plant cells, observed after the action of pathogenic factors and the effect of the fixation procedures, were also noticed in small quantities in the cells of the investigated objects.  相似文献

Summary The fungal metabolite brefeldin A (BFA) causes inhibition of cell growth inMicrasterias denticulata after 2 h incubation, combined with slight malformation of the cell shape. The BFA effects on cell development are accompanied by a gradual decrease in the number of Golgi cisternae and severe structural and morphological changes of the dictyosomes which are already visible after only 10 min exposure. When the treatment is prolonged the number of dictyosomes is markedly reduced, leading to almost complete loss of Golgi bodies, particularly in the young semicell. Groups of primary wall material-containing vesicles accumulated in areas of former dictyosomes, and previously unknown vesicular bodies are found. Restitution of almost normal dictyosomes occurs within 5 h when the cells are allowed to recover from BFA treatment.Micrasterias cells incubated in BFA at concentrations below 15 M maintain their ability to divide over several generations. Our results indicate that, of the various inhibitors of the secretory pathway tested against growingMicrasterias cells, BFA is the only drug which induces complete and reversible dissociation of dictyosomes in the growing semicell. Summary Ultrastructural observations on the principal endomembranes (endoplasmic reticulum, Golgi apparatus) of synchronously growing of wild type and mutant (CW 2, CW 15) strains ofChlamydomonas reinhardii have been carried out. The dictyosomes of the Golgi apparatus in all three cases are highly polar in morphology but lack intercisternal filaments. A clear spatial relationship between dictyosomes and endoplasmic reticulum is seen and a transfer of vesicles from the latter to the former is easily visualized. Coated vesicles invariably appear to be restricted to the trans-pole of the dictyosomes. The endoplasmic reticulum adjacent to the cis pole of dictyosomes is considerably hypertrophied in the case of the wild type, only partially so in the mutant CW 2 but not at all in the mutant CW 15. In the wild type this swelling is most extreme during the period of wall deposition and for several hours afterwards. The results are discussed in relation to the biosynthesis and intracellular transport of, particularly O-glycosidically linked, glycoproteins.  相似文献

Summary High pressure freezing and freeze substitution methods significantly improve the antigenic preservation of S-locus specific glycoproteins (SLSG). The SLSG, which are implicated in the incompatibility response, are localized over the cell wall and cytoplasm. Labeling in the cytoplasm is mainly associated with dictyosomes and rough endoplasmic reticulum. Quantitative analysis show that in cryofixed papillae the labeling was enhanced by approximately 45% over the cell wall and approximately 90% over the dictyosomes compared to chemically fixed papillae.  相似文献

The number and distribution pattern of dictyosomes in cellsof a green alga,Closterium ehrenbergii, were examined by fluorescencemicroscopy. Dictyosomes absorbed the fluorescent dye, DiOC6(3) intensely, and strongly radiated fluorescent light. Dictyosomeswere distributed in the cytoplasm along the longitudinal chloroplast-ridges.They began to divide synchronously at a premitotic stage whenthe chloroplast started to divide, and duplicated in numberbefore the cell divided by a transverse septum. Summary Numerical and structural changes in dictyosomes during the germination of zygospores inClosterium ehrenbergii were examined by electron microscopy. In the dormant mature zygospores, two parallel cisternac were seen which were derived from the disorganization of dictyosomes during the maturation of zygospores. After the induction of germination, the two parallel cisternae developed into dictyosomes with ten or eleven cisternae. The dictyosomes doubled in number by division every day for four days and reached, at the time of germination, a density of distribution similar to that found in the youngest zygospore. On the 4th day after the induction of germination, dictyosomes produced two kinds of vesicles which appear to be involved in the formation of new cell wall layers. The germination of the zygospore was effected by the escape of the cell covered with the new cell wall layers through the broken old cell wall layers.  相似文献

Summary Transformation of the Golgi apparatus in the stages of the cell cycle ofMicrasterias americana was investigated with an electron microscope. In the resting cell, dictyosomes consisted of eleven cisternae, the distal-most cisterna of which was partially network-shaped. During the developmental stages of the new half cell followed by nuclear division, dictyosomes produced dark vesicles and large vesicles at the peripheries of the distal cisternae, thus diminishing the diameter of the distal cisternae and their numbers. Finally, dictyosomes with six or seven cisternae of small diameter were found when the new half cell reached to full size as a mother half cell. At this stage, the small dictyosomes produced flat vesicles. Thereafter, dictyosomes recovered the size and number of their constituent cisternae, being supplied a membrane and substrate from the primary vesicles. Lysosomes divided the dictyosomes in two, and these divided dictyosomes seemed to regenerate in one case and to disorganize in another. The occurrence of large vesicles was also confirmed using the negative staining method in growing cells.  相似文献

Ultrastructural changes of Golgi apparatus of frog urinary granular cells at antidiuretic hormone (ADH) stimulation of water transport were studied. During a short-time ADH action (5 min) the fragmentation of the complex on single dictyosomes and dilution of certain cisternae is discovered. A conclusion is made that the granular cell giant vacuoles may originate from the Golgi cisternae. It is suggested that the microtubules may be involved in the translocation of dictyosomes and migration of formed vacuoles. The quantity of microtubules increases during ADH action very significantly. Moreover, the involvement of the Golgi apparatus is shown in the maintenance of the cell membrane balance due to budding of tubular structures from transcisternae and shuttling between luminal and vacuolar membranes.  相似文献

The dictyosome (Golgi body) in the secondary spermatocyte of the cricket appears in electron micrographs as a duplex structure composed of (a) a group of parallel double-membraned lamellae and (b) a group of associated vacuoles arranged along the compact lamellae in a chain-like fashion. This arrangement of ultramicroscopic structure for the dictyosomes is strikingly comparable to that described for the Golgi apparatus of vertebrates. Accordingly, the two are considered homologous structures. Associated with the duplex structure of the dictyosomes is a differentiated region composed of small vacuoles. This is thought to represent the pro-acrosome region described in light microscope preparations. In the spermatid the dictyosomes fuse, giving rise to the acroblast. Like the dictyosomes, the acroblasts are made up of double-membraned lamellae and associated vacuoles. In addition, a differentiated acrosome region is present which, in some preparations, may display the acrosome vacuole and granule. Both the dictyosomes and acroblasts are distinct from mitochondria.  相似文献

Apolipoprotein E (APOE) is the only confirmed susceptibility gene for late-onset Alzheimer disease (AD). In a recent genomic screen of 54 families with late-onset AD, we detected significant evidence for a second late-onset AD locus located on chromosome 12 between D12S373 and D12S390. Linkage to this region was strongest in 27 large families with at least one affected individual without an APOE-4 allele, suggesting that APOE and the chromosome 12 locus might have independent effects. We have since genotyped several additional markers across the region, to refine the linkage results. In analyzing these additional data, we have addressed the issue of heterogeneity in the data set by weighting results by clinical and neuropathologic features, sibship size, and APOE genotype. When considering all possible affected sib pairs (ASPs) per nuclear family, we obtained a peak maximum LOD score between D12S1057 and D12S1042. The magnitude and location of the maximum LOD score changed when different weighting schemes were used to control for the number of ASPs contributed by each nuclear family. Using the affected-relative-pair method implemented in GENEHUNTER-PLUS, we obtained a maximum LOD score between D12S398 and D12S1632, 25 cM from the original maximum LOD score. These results indicate that family size influences the location estimate for the chromosome 12 AD gene. The results of conditional linkage analysis by use of GENEHUNTER-PLUS indicated that evidence for linkage to chromosome 12 was stronger in families with affected individuals lacking an APOE-4 allele; much of this evidence came from families with affected individuals with neuropathologic diagnosis of dementia with Lewy bodies (DLB). Taken together, these results indicate that the chromosome 12 locus acts independently of APOE to increase the risk of late-onset familial AD and that it may be associated with the DLB variant of AD.  相似文献

Colloidal gold-labelled antibody was used to localize indole-3-acetic acid in caps of primary roots of Z. mays cv. Kys. Gold particles accumulated on the nucleus, vacuoles, mitochondria, and some dictyosomes and dictyosome-derived vesicles. This is the first localization of indole-3-acetic acid in dictyosomes and dictyosome-derived vesicles and indicates that dictyosomes and vesicles constitute a pathway for indole-3-acetic acid movement in and secretion from root cap cells. Our findings provide cytochemical evidence to support the hypothesis that indole-3-acetic acid plays an important role in root gravitropism.  相似文献

The fine structure of epidermal cells, particularly in relationto dictyosomes, has been examined in different regions of dark-growncucumber hypocotyls and in response to auxin treatment, usingboth dot overlay and image analysis techniques. The most noticeablechange in cell structure along the hypocotyls is the increasein vacuolar volume. The volume fraction occupied by dictyosomesand secretory vesicles also increased, whereas that for mitochondriaremained relatively constant. During auxin treatment, the volumefraction for dictyosomes showed an increase after 30 min followedby a fall, whereas that occupied by secretory vesicles fellsteadily over 90 min. The number of cisternae per dictyosomeshowed some increase after 2 h of auxin treatment, althoughthe increase in dictyosomal material with cell expansion waslargely accounted for by an increase in the number of dictyosomes. Auxin-stimulated elongation growth of the hypocotyls was inhibitedby a range of calcium antagonists, chelators and ionophores.The most marked inhibitions were observed with calcium chloride,the chelator chlortetracycline and the ionophores verapamil,nigericin and monensin. Linear transducer experiments showedthat these compounds generally caused an immediate reductionin the rate of growth. Fine structural observations carriedout on epidermal cells showed the most obvious effects withmonensin and nigericin which caused dictyosomes and secretoryvesicles to swell. EGTA and LaCl3 caused secretory vesiclesto accumulate around dictyosomes, while the ionophore A23187had little effect. The results suggest that the concentration of Ca2+ in the cytoplasmmay be critical for cell elongation. Summary The morphology of the Golgi apparatus of Amoeba proteus can be influenced by substances inducing pinocytotic activity as well as by extreme temperatures. During the ingestion of a solution of 0.5% egg white the number of Golgi bodies decreases from 100% measured in control cells to 82% measured in cells showing induced pinocytosis. Simultaneously the ratio of the surface area of the cisternae at the proximal face to that of the vesicles at the distal face of single dictyosomes remains constant (1.74–1.72).The decrease and increase of the temperature of the culture medium to 4° C and 30° C respectively, causes the disappearance of most of the dictyosomes. After keeping the cells for 3–10 h at these temperatures the number of Golgi bodies was only 5–10% of the controls. A continued treatment with cold or warm culture medium leads to a partial reorganization of dictyosomes. After 15 h the number of Golgi bodies counted per cell returned to 57% at 4° C and 38% at 30° C. The ratio of the surface area of the Golgi cisternae to the surface area of the Golgi vesicles also alters under the influence of extreme temperatures. The values measured after treating the cells for 3 h, 4 h 10 h and 15 h at 4° C and 30° C amounted to 0.75, 0.85, 1.14 1.53 and 0.93, 0.38, 0.88, 1.60, respectively, compared to 1.72 of control amoebae.The different values of the ratio of the surface area of cisternae to that of vesicles indicate that there are strong morphological changes of single dictyosomes.  相似文献

Investigation of a possibility of long-term storage of frozen (-196 degrees C) viable neurons and nervous tissue is one of the central present day problems. In this study ultrastructural changes in neurons of frozen-thawed snail brain were examined as a function of time. We studied the influence of cryopreservation, cryoprotectant (Me2SO), cooling to 4-6 degrees C, and a prolonged incubation in physiological solution at 4-6 degrees C on dictyosomes of Golgi apparatus, endoplasmic reticulum (ER) cisternae and mitochondria. It has been found that responses of these intracellular structures of cryopreserved neurons to the above influences are similar: dissociation of Golgi dictyosomes, swelling of endoplasmic reticulum cisternae and mitochondrial cristae. Both freezing-thawing and cryoprotectant were seen to cause an increase in the number of lysosomes, liposomes, myelin-like structures, and to form large vacuoles. The structural changes in molluscan neurons caused by cryopreservation with Me2SO (2 M) were reversible.  相似文献

Golgi apparatus were isolated from untreated rat liver and separated into three fractions. One consisted mainly of vesicles, a second of tubular particles (dictyosomes) and the third was a mixed fraction. Large differences between these fractions could be seen in the electron microscope and by enzyme analysis. The total lipid content of the vesicles was 3.5-times greater than that of the dictyosomes and the neutral lipid value was 7-times greater. The ratio of phospholipids to protein was approximately the same in the three fractions. However, the phospholipid patterns differed between the vesicle and dictyosome fractions.  相似文献
