Proteases from pyloric caeca extract of three fish species including brownstripe red snapper (Lutjanus vitta), bigeye snapper (Priacanthus tayenus) and threadfin bream (Nemipterus marginatus) were comparatively studied. The extracts from bigeye snapper and threadfin bream exhibited the highest hydrolytic activities toward casein, α-N-benzoyl-dl-arginine-p-nitroanilide and α-N-ρ-tosyl-l-arginine methyl ester at pH 8.0 and 60 °C and pH 8.5 and 55 °C, respectively. The extract of brownstripe red snapper showed the optimal pH and temperature of 8.0 and 60 °C with all substrates used except the optimal temperature was 65 °C when casein was used. All proteases were strongly inhibited by soybean trypsin inhibitor (SBTI) and N-ρ-tosyl-l-lysine chloromethylketone (TLCK) and partially inhibited by N-tosyl-l-phenylalanine chloromethylketone for all substrates tested, suggesting that trypsin-like proteases were the major enzymes. Substrate-gel activity staining of 40–60% ammonium sulfate (AS) fraction revealed that major activity bands were observed with molecular mass of 24, 22 and 20 kDa for brownstripe red snapper, bigeye snapper and threadfin bream, respectively. Those activity bands were partially inhibited by SBTI and TLCK. AS fraction was further used to produce gelatin hydrolysate from the skin of brownstripe red snapper with different degrees of hydrolysis (DH). Hydrolysate with DH of 15% exhibited the highest DPPH and ABTS radical scavenging activities and ferric reducing antioxidant power (p < 0.05). Therefore, the extract from pyloric caeca could be used to produce the gelatin hydrolysates possessing antioxidative activities.  相似文献

Common carp (Cyprinus carpio, [Linnaeus, 1758]) have long been established in the United States and in some cases their populations can be very dense, leading biologists to expend significant effort towards reducing numbers of common carp in some populations. Estimating abundance of common carp prior to removal efforts would be useful in evaluating success of these removal efforts, but marking large numbers of fish can be expensive. Therefore, a low-cost tagging option is needed. In this study, we used tank and field experiments to determine the retention and longevity of hole-punch marks in the opercula of common carp. For the tank experiment, fish were double marked with a size-3 self-piercing tag and an operculum hole-punch mark (using a paper hole-punch tool with a hole diameter of 6.4 mm) on opposite sides of the fish. Over the entirety of the 180–day tank experiment, retention of the self-piercing tags and hole-punch marks was 100% and no marking mortality was observed. For the field experiment, 883 common carp were tagged at random in two groups, a double-marked group (n = 416, both self-piercing tags and hole-punch) and a single-mark group (n = 467, self-piercing tag). Fish were sampled monthly for 398 days. Because the length distribution sampled was bimodal, we evaluated tag retention of fish <330 mm TL (small fish, n = 273) and > 331 mm TL (large fish, n = 143), separately. Hole-punch mark retention was high for both size classes throughout most of the field experiment. For large fish, retention of hole-punch marks was 100% for the entire 398-day experiment. For small fish, retention of hole-punch marks was 100% through 184-day and remained above 93% through 328-days, but declined to 0% by day 398. Our results suggest that the operculum hole-punch mark is a valuable low-cost, long-term technique for tagging common carp.  相似文献

Turkey’s natural and ecological situations are very suitable for aquaculture. Turkey also has a wide variety of freshwater and marine species comprising trout, carp, sea bass, sea bream, turbot, mussel, crayfish, etc. The total production of fish and shellfish was 646,310 tons in 2008. The contribution of freshwater catch to total fishery production is relatively small. Capture fisheries production amounted to 494,124 tons whilst aquaculture production was 152,186 tons in the same year. In Turkey, Engraulis encrasicholus (anchovy) is the main caught sea fish species. Fisheries in the Black Sea are the most important fishery by far and show the greatest variations in total catch. Alburnus tarichii (a local species belonging to Cyprinidae) and Cyprinus carpio (the common carp) are the most important species caught from freshwaters. Aquaculture is going to play an increasingly important role in the Turkish economy, as fishery products are the only products of animal origin that can be exported to the EU. There has been a fast increase in the aquaculture production in Turkey with the implementation of scientific and technological modernization. For example, total aquaculture production for 1986 and 2008 was 3,075 and 152,186 tons, respectively. The percentage of aquaculture in total fish production has been rising every year. The ratio of cultured fish production to total fish production was 1.5% in 1990 s, 13.57% in 2000 and more than 20% in 2005. It was 23.55% in 2008. Trouts are the main cultured freshwater fish species. Raceways and floating cages are employed in culture of trout. Carps are also important cultured freshwater fish species. Sea bass and gilthead sea bream are grown marine fish species. Floating cages, off-shore and earthen ponds are used for marine fish species culture. There has been an increase in fishery exports and imports in recent years. It was more than US500 million in 2008, but that of 2004 was just over US 500 million in 2008, but that of 2004 was just over US 233 million. However, aquaculture production is still far away from the production targets and fisheries sector is not an important part of the economy at present.  相似文献

Lake Ziway harbours indigenous and exotic fish species including Nile tilapia (Oreochromis niloticus) and common carp (Cyprinus carpio). Nile tilapia was the dominant and preferred fish. However, its contribution to total catch has dramatically declined from 89.3% in 1994 to 27% in 2014 while the introduced common carp has increased from 0% before 2012 to 25% in 2014. Common carp potentially compete with Nile tilapia for available resources and could be a cause for the decline. Thus, the study explored the dietary overlap of the two species from April to August 2017. Schoener's overlap index (α) revealed significant dietary overlap between the two species (α = 0.84, between juveniles and α = 0.63, between adults). Juveniles fed mainly on animal origin (zooplankton and insects), while adult Nile tilapia have consumed plant origin (macrophyte and phytoplankton), and adult common carp fed on macrophytes and detritus. The presence of significant dietary overlap between the two species, particularly due to intense competition among juveniles, might cause the reduction of Nile tilapia stock because the native species has shown a competitive disadvantage for food in the presence of common carp. This study provides baseline information to researchers and decision makers working towards the sustainable resource utilisation of the system.  相似文献

The purpose of the present study was to investigate the effects of cryoprotectants (4 % sucrose?+?4 % sorbitol) on functional and rheological changes, including thermal stability, dynamic rheological and emulsifying properties, gel texture profile, gel whiteness, gel water-binding capacity, and gel microstructure, in the myofibrillar proteins (MP) of common carp (Cyprinus carpio) surimi subjected to multiple freeze-thaw cycles (FT, 0, 1, 3 and 5 times). Increased numbers of FT cycles were accompanied by reduced thermal transition temperature (T max), reduced enthalpy of denaturation (ΔH) (P?<?0.05), and enhanced susceptibility to thermal aggregation. The observation of the gel microstructure indicated that the FT processes produced empty spaces and changed the aggregate gel structure into a coarser-stranded network. The addition of cryoprotectants significantly prevented the reduction of MP thermal stability (P?<?0.05). The protective effect on protein functionality was demonstrated by the cryoprotectant’s efficacy in maintaining the three-dimensional gel network by small-strain oscillatory rheological testing and increasing the emulsifying capacity and emulsion stability of the MP. The FT process caused a decrease (P?<?0.05) in whiteness, water-binding capacity, and texture (hardness, springiness, and gumminess) of the MP gels, which were reduced by the addition of cryoprotectants. Overall, the FT-induced loss of the abovementioned functional and rheological properties was significantly reduced by the presence of cryoprotectants.  相似文献

Arrocampo Reservoir is used as a cooling basin for a nuclear power plant located in southern Europe. Its annual mean temperature is about 30 °C with maxims that achieved 41.5 °C near the hot water effluent. Common carp (Cyprinus carpio, Linnaeus 1758) is the dominant pelagic fish species in this reservoir. Hydroacoustic surveys were conducted bimonthly over 9 years to characterize common carp abundance and distribution in the reservoir on an annual and seasonal temporal scale. Mean fish density during the period was 0.029 fish m−3, varying from a maximum of 0.038 in 2003 to a minimum of 0.012 in 1998. There were no significant differences in the mean fish density among years. Each year, fish distribution showed significant seasonal variation. Carp were homogeneously distributed during autumn and spring; in winter carp occupied warmer outfall areas, whereas in summer, fish were distributed in the coldest area of the reservoir (dam area) but avoided the warm outfall area. These seasonal tendencies of carp distribution was repeated every years. Behavioural reaction appears to be particularly important in explain carp distribution in this reservoir.  相似文献

In order to investigate ontogenetic changes in diet and diet overlap between rohu (Labeo rohita) and common carp (Cyprinus carpio) in polyculture ponds, food preferences of different size classes of these fishes were quantified. Rohu diet consisted of both phytoplankton and zooplankton, and there was a distinct ontogenetic shift in the relative importance of these food items. Zooplankton was the dominant food for rohu up to 20.6 cm total length (TL) and then gradually decreased in importance as fish grew. Phytoplankton was initially a minor component of rohu diet but gradually increased in importance and became the dominant food for rohu at 24.2 cm TL. Phytoplankton biovolume in rohu guts was positively correlated with fish size (TL). Chesson’s α indicated that rohu of all sizes preferentially selected Cladocera and avoided Cyanophyceae and Euglenophyceae. Young rohu initially preferred Rotifera and Copepoda but gradually switched to Bacillariophyceae and Chlorophyceae. Common carp diet consisted of phytoplankton, zooplankton, and benthic macroinvertebrates, but was dominated by benthic macroinvertebrates (63–92% of total diet). As common carp grew, the proportion of zooplankton ingested decreased and the proportion of benthic macroinvertebrates increased. Benthic macroinvertebrate biovolume in common carp guts was positively correlated with fish size. Common carp of up to 15.4 cm TL preferentially selected zooplankton, but common carp larger than 18.9 cm TL avoided this food item. Common carp of all sizes avoided phytoplankton. A low dietary overlap was found between rohu and common carp (Schoener overlap index: 0.08–0.35), probably due to ingestion of smaller quantities of zooplankton by the latter. Dietary overlap also decreased with increasing rohu and common carp size because of divergent ontogenetic shifts in dietary preferences of the two species.  相似文献

Impact of cyprinids on zooplankton and algae in ten drainable ponds   总被引:4,自引:4,他引:0  
To study the impact of cyprinids on algae, zooplankton and physical and chemical water quality, ten drainable ponds of 0.1 ha (depth 1.3 m) were each divided into two equal parts. One half of each pond was stocked with 0 + cyprinids (bream, carp and roach of 10–15 mm), the other was free of fish. The average biomass of the 0 + fish at draining of the ponds was 466 kg ha–1, to which carp contributed about 80%.The fish and non-fish compartments showed significant differences. In the non-fish compartments the density of Daphnia hyalina was 10–30 ind. l–1 and that of Daphnia magna 2–4 ind. l-–1, whereas in the fish compartments densities were c. 1 ind. l–1. Cyclopoid copepods and Bosmina longirostris, however, showed higher densities in the fish compartments. The composition of algae in the two compartments differed only slightly, but the densities were lower in the non-fish compartments. The significant difference in turbidity was probably caused by resuspension of sediment by carp. No significant difference in nutrient concentration between the compartments was found.  相似文献

Juvenile walleye pollock, Theragra chalcogramma, is the dominant forage fish on the continental shelf of the Gulf of Alaska, yet little is known about the feeding habits of this important interval of pollock life history. The taxonomic composition and size of prey found in the stomachs of age-0 juveniles collected at three nearshore locations in the Gulf of Alaska in September 1990 were compared to the composition and size of zooplankton collected in concurrent plankton tows. The maximum length of prey consumed increased dramatically over the length range of pollock examined (58–110 mm) from approximately 7 mm to 30 mm, due mainly to the consumption of large euphausiids and chaetognaths by the bigger individuals. The maximum width of prey changed little over this size range although there was a general increase in prey width with increasing predator size. The minimum prey length and width did not change with increasing fish size. Juvenile pollock generally selected the larger prey sizes relative to what was available. Juvenile pollock showed a marked preference for adult euphausiids and decapod larvae and an avoidance of copepods and chaetognaths relative to the numbers collected in net tows. These results are discussed relative to the feeding ecology of these juvenile fishes. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

To elucidate the functional role of piscine incomplete ELR motif, the recombinant CXC and its mutants (mELR and mLoop) were produced in Escherichia coli M15 based on the predicted mature peptide coding sequence of the black sea bream CXC (BS CXC) chemokine. Assays showed that the BS rCXC proteins displayed strong ability to induce fish blood neutrophils and head kidney (HK) macrophage migration in a dose-independent manner (10 to 200 ng), both in black sea bream and common carp. Although the ELR motif and the N-terminal loop of ELR+CXC chemokines are essential for chemotactic activity and receptor binding in mammals, the mELR and mLoop mutants showed no significant difference in their induction of chemotaxis of fish blood neutrophils compared with the full-length rCXC at the same dose. Human recombinant IL-8 (hrIL-8) can clearly induce piscine blood neutrophil migration and has no effect on macrophages, whereas the BS rCXC cannot induce chemotaxis in higher vertebrates, such as rat blood neutrophils or macrophages, even if the incomplete ELR motif in rCXC was mutated to ELR. The BS CXC and its mutants can promote the phagocytosis ability of piscine blood neutrophils and HK macrophages both in black sea bream and common carp, but have no effect on rat neutrophils or macrophages. Results showed that the piscine ELR+CXC-like chemokine represents an ancient version of a CXC chemokine; the ELR motif still does not show the higher specific polarization of function as found in mammalian.  相似文献

Adenylosuccinate lyase (ADSL) is a bifunctional enzyme acting in de novo purine synthesis and purine nucleotide recycling. In the present study, we have constructed a grass carp (Ctenopharyngodon idella) intestinal cDNA library that has over 2.3 × 105 primary clones. An expressed sequence tag (EST) of grass carp adenylosuccinate lyase (gcADSL) gene was screened from this library. Both 5′-RACE and 3′-RACE were carried out in order to obtain the complete cDNA sequence, which contains a 1,446 bp open reading frame encoding 482 amino acids about 54.552 kDa. The deduced amino acid sequence shares high homology with its vertebrate counterparts, which shares 94% similarity with zebrafish, 81% with African clawed frog as well as chicken, 77% with human and 76% with mouse. This gcADSL genomic sequence, consisted of 13 exons and 12 introns, is 8,557 bp in size. Real-time quantitative PCR analysis revealed that the highest expression level of gcADSL was detected in muscle and the lowest in gill. In western blotting analysis, His6-tagged gcADSL protein expressed in Escherichia coli could be recognized not only by an anti-His6-tag monoclonal antibody but also by an anti-human ADSL polyclonal antibody, indicating immunological crossreactivity occurs between grass carp and human ADSL protein. 1,082 bp 5′-flanking region sequence was cloned and analyzed.  相似文献

In ten 0.1 ha drainable ponds O+ cyprinids were stocked to assess their impact on the aquatic community. Bream and roach were stocked in early June, 1986, and carp in early July, 1986. The ponds were drained mid November, 1986. The relationship between the fish community and its food resource was estimated in terms of production and consumption with use of the data on growth and mortality of the fish. The average gross production of the fish species in the ten ponds (48.4 kg during the experiment) was dominated by the carp (58.5%). A minimum estimate of the food consumption of the O+ cyprinids is 760 kJ.m–2. The average production of bream and roach was much lower than that of carp due to a higher mortality and a more specialized feeding pattern. The gross productions of carp, bream and roach in the ten ponds were not significantly related, although stomach analysis showed diet overlap between the carp and the bream.  相似文献

The aim of this study was to determine if saddleback syndrome (SBS) in a wild population of yellowfin bream (Acanthopagrus australis) was the result of a developmental defect or caused by physical injury. Information was collected in 2012 on the incidence of SBS and other abnormalities in this species in Moreton Bay, Australia. Abnormalities in adult fish (>250 mm Total Length, TL) with SBS (n = 47) were compared with those without SBS (n = 30). A sample of juvenile fish (n = 404) was checked for the presence of SBS. The results show that scale loss, scale pattern misalignment, lateral line fracture and pectoral fin abnormality were closely associated with SBS. SBS was uncommon (<2%) in juveniles <70 mm TL, but common (>12%) in the larger juveniles (70–215 mm TL). These results, together with the findings that scale loss associated with SBS in adult fish occurred in the range 80–245 mm back‐calculated TL, indicate that SBS and the related abnormalities in yellowfin bream are a result of physical injuries to larger juveniles (>70 mm TL). The reduction in the incidence of SBS from approximately 12% in the larger juveniles to 5% in adults is evidence of mortality associated with SBS.  相似文献

Sequence analysis of the cytochrome c oxidase I gene was used to confirm the identity of species in 50 commercial surimi products labeled as Alaskan pollock (Theragra chalcogramma). All sequences (655 base pairs long) produced from the samples were aligned and compared with reference sequences from the GenBank database. A Neighbor Joining tree clustered samples into eight groups. Results showed that only 16% (8/50) of the surimi samples were made from Alaskan pollock as declared. The remaining species detected in samples belonged to the families Sciaenidae (12%), Synodontidae (16%), Merlucciidae (38%), Nemipteridae (8%), Priacanthidae (2%), Mullidae (2%), and two species of Gadidae (4%).  相似文献

The effects of freezing on the heat-induced gelation, Ca2+-ATPase activity and myofibril structure of Antarctic krill muscle were investigated. Muscle from freshly caught krill was immediately stored at-20°C in the presence (to prevent freezing) (glycerol krill) and absence (frozen krill) of glycerol. Several protease inhibitors, monosodium glutamate and Ca2+ were individually added to glycerol krill to inhibit endogenous proteolysis. The examinations described above were carried out after about 3-month storage at-20°C. In glycerol krill (unfrozen state), viscoelastic parameters of the heat-induced gels and Ca2+-ATPase activities of all the krill samples were similar to those of “surimi” (raw fish meat paste) of Alaska pollack which gave gel of good quality, although the micro structure (Z-lines) of myofibrils was different among the glycerol krill samples. In frozen krill, however, the parameters of the gel were different from those of “surimi”, the ATPase activity was completely lost and disruption of the myofibril structure occurred. Refreezing (-20°C) of glycerol krill after removal of glycerol resulted in a marked decrease in the gelation ability. These results suggest that freezing of krill muscle causes deterioration of the gelation ability.  相似文献

The effect of common carp and bream on hydrochemical parameters, abundance, and structure of phyto-, zooplankton, and macrozoobenthos has been studied in semienclosed mesocosms installed in the littoral zone of a mesotrophic lake. Significant differences in the biomass of different algal groups in mesocosms with fish in respect to the control were established only for diatoms and were not found for other phytoplankton groups. Common carp had a greater effect on the abundance of large zooplankton species (Diaphanosoma brachyurum) compared to bream. The abundance of the small Bosmina longirostris increased in mesocosms both with bream and common carp. The macrozoobenthos biomass reduced at higher rates in mesocosms with common carp than in those with bream, with the strongest effect of common carp on mayfly larvae. The differences between the consumption of chironomid and oligochaete larvae were not established in mesocosms with common carp. Bream mainly affected the larvae of mayflies and oligochaetes and, to a lesser degree, the chironomid larvae.  相似文献

In 1987, the Bleiswijkse Zoom, a small, shallow lake in The Netherlands, was divided into two compartments to investigate the possible use of biomanipulation as a tool for restoring the water quality of hypertrophic lakes. The density of the fish stock before restoration was about 650 kg.ha–1, composed mainly of bream, white bream and carp. Pikeperch was the main fish predator in the lake. In April, 1987, in one compartment (Galgje) all planktivorous bream and white bream and about 85% of the benthivorous bream and carp were removed. Advanced pikeperch fry were introduced as predator during the transient period. The other compartment (Zeeltje) was used as a reference. Removal of the fish in Galgje resulted in low concentrations of chlorophyll-a, total phosphorus, nitrogen and suspended solids. The absence of bottom-stirring activity by benthivorous fish and the low chlorophyll-a concentrations led to an increase in the Secchi disk transparency from 20 to 110 cm. Within two months after removal of the fish, macrophytes, mainly Characeae, became abundant. Until July the high density of large zooplankton species caused low algal biomass. From June onwards, the zooplankton densities decreased, but the algal concentrations remained low. This is probably because of nutrient limitation or depression of algal growth by macrophytes or both. Compared with the non-treated compartment the number of fish species in the treated compartment was higher. Perch, rudd and roach, i.e. the species associated with aquatic vegetation, were found in the samples. The survival of the O+ pikeperch was poor. The pikeperch could not prevent the growth of young cyprinids. Within two months after the removal of the fish a habitat for northern pike was created.  相似文献

Effects of protein isolate from bambara groundnut (BGPI) at different levels (0–6 %, w/w) in combination with microbial transglutaminase (MTGase) at different concentrations (0, 0.3 and 0.6 U g?1surimi) on gels properties of sardine (Sardinella albella) surimi were investigated. In the absence of MTGase, the increases in breaking force and deformation of gels were obtained when BGPI at levels of 1.5–3 % was incorporated (P?<?0.05). The further increases in BGPI levels (4.5–6 %) resulted in the decrease in breaking force and deformation (P?<?0.05). When MTGase (0.3 and 0.6 U g?1surimi) was added, the increase in breaking force and deformation were noticed, regardless of BGPI levels, and the strengthening effect was in dose-dependent manner. The increases in hardness, gumminess and chewiness were also observed when surimi gel was added with BGPI and MTGase (P?<?0.05). Water-holding capacity of gels was improved with increasing level of BGPI, and MTGase incorporated (P?<?0.05). Whiteness of gels slightly decreased with increasing BGPI levels, however the addition of MTGase had no impact on whiteness (P?>?0.05). Based on electrophoretic study, myosin heavy chain decreased with addition of MTGase, indicating the formation of cross-links. More compact structure was observed in gel added with MTGase (0.6 U g?1surimi) and 6 % BGPI, and was accompanied by an increased gel strength.  相似文献

Thirty-six enclosures, surface area 4 m2, were placed in Little Mere, a shallow fertile lake in Cheshire, U.K. The effects of different fish species (common carp, common bream, tench and roach) of zooplanktivorous size, and their biomass (0, 200 and 700 kg ha–1) on water chemistry, zooplankton and phytoplankton communities were investigated. Fish biomass had a strong effect on mean zooplankton size and abundance. When fish biomass rose, larger zooplankters were replaced by more numerous smaller zooplankters. Consequently phytoplankton abundance rose in the presence of higher densities of zooplanktivorous fish, as zooplankton grazing was reduced. Fish species were also significant in determining zooplankton community size structure. In enclosures with bream there were significantly greater densities of small zooplankters than in enclosures stocked with either carp, tench and, in part, roach. When carp or roach were present, the phytoplankton had a greater abundance of Cyanophyta than when bream or tench were present. Whilst top-down effects of fish predation controlled the size partitioning of the zooplankton community, this, in turn apparently controlled the bottom-up regeneration of nutrients for the phytoplankton community. At the zooplankton–phytoplankton interface, both top-down and bottom-up processes were entwined in a reciprocal feedback mechanism with the extent and direction of that relationship altered by changes in fish species. This has consequences for the way that top-down and bottom-up processes are generalised.  相似文献
