太子陵旧石器地点于2005年4月发现,2005年5月又对其进行了复查。该地点位于天津蓟县东北部孙各庄乡清太子陵东侧冲沟西面的黄土台地中,在其浅黄色粉砂质黏土层和地表发现石制品58件,包括石核、石片、断块和石器。古人类选择地点附近的阶地底部河卵石为原料进行剥片和加工石器;硬锤锤击法为剥片的主要技术;石制品总体以小型和微型居多;石器主要以石片为毛坯,刮削器是主要类型;石器多由古人类在石片的一侧采用锤击法正向加工而成。地貌与地层对比则显示遗址的时代大致属于晚更新世晚期之末或全新世早期。  相似文献

The combination of gene therapy and virotherapy for cancer treatment has received close attention and has become a trend in the field of cancer biotherapy. A strategy called 'Cancer Targeting Gene-Viro-Therapy' (CTGVT) or 'Gene Armed Oncolytic Viral Therapy' (GAOVT) has been proposed, in which an antitumor gene is inserted into an oncolytic viral vector. In our previous study, a dual-regulated oncolytic adenovirus with enhanced safety for normal cells and strict liver cancer-targeting ability, designated Ad?enAFP?E1A?E1B (Δ55) (briefly Ad?enAFP?D55), was successfully constructed. In the current work, interleukin-24 (IL-24) and suppressor of cytokine signaling 3 (SOCS3) genes were packaged into Ad?enAFP?D55. The new constructs, Ad?enAFP?D55-(IL-24) and Ad?enAFP?D55-(SOCS3), showed improved tumoricidal activity in hepatoma cell lines compared with the oncolytic viral vector Ad?enAFP?D55. The co-administration of Ad?enAFP?D55-(IL-24) and Ad?enAFP?D55-(SOCS3) showed much better antitumor effect than Ad?enAFP?D55-(IL-24) or Ad?enAFP?D55-(SOCS3) alone both in vitro and in a nude mouse xenograft model. Moreover, our results also showed that blockade of the Jak/Stat3 pathway by Ad?enAFP?D55-(SOCS3) infection in HuH-7 cells could down-regulate some anti-apoptosis proteins, such as XIAP, Bcl-xL, and survivin, which might sensitize the cells to Ad?enAFP?D55-(IL-24)-induced apoptosis. These results indicate that co-administration of Ad?enAFP?D55-(IL-24) and Ad?enAFP?D55-(SOCS3) may serve as a candidate therapeutic approach for the treatment of liver cancer.  相似文献

As heart-rate variability (HRV) is under evaluation in clinical applications, the authors sought to better define the interdependent impact of age, maximal exercise, and diurnal variation under physiologic conditions. The authors evaluated the diurnal changes in HRV 24-h pre- and post-maximal aerobic exercise testing to exhaustion in young (19-25 yrs, n?=?12) and middle-aged (40-55 yrs, n?=?12) adults. Subjects wore a portable 5-lead electrocardiogram holter for 48?h (24?h prior to and following a maximal aerobic capacity test). Time-, frequency-, time-frequency-, and scale-invariant-domain measures of HRV were computed from RR-interval data analyzed using a 5-min window size and a 2.5-min step size, resulting in a different set of outputs every 2.5?min. Results were averaged (mean?±?SE) over four prespecified time periods during the morning, afternoon, evening, and night on Day 1 and Day 2. Diurnal changes in HRV in young and middle-aged adults were compared using a two-way, repeated-measures analysis of variance (ANOVA). Young adults demonstrated higher HRV compared to middle-aged adults during periods of wakefulness and sleep prior to maximal exercise stress testing (i.e., high-frequency power during Day 1: young adults: morning 1862?±?496?ms(2), afternoon 1797?±?384?ms(2), evening 1908?±?431?ms(2), and night 3202?±?728?ms(2); middle-aged adults: morning 341?±?53?ms(2), afternoon 405?±?68?ms(2), evening 469?±?80?ms(2), and night 836?±?136?ms(2)) (p < .05). Exercise resulted in reductions in HRV such that multiple measures of HRV were not significantly different between age groups during the afternoon and evening periods. All measures of HRV demonstrated between-group differences overnight on Day 2 (p < .05). Young adults are associated with higher baseline HRV during the daytime. Sleep increases variability equally and proportionally to daytime variability. Given the higher baseline awake HRV and equal rise in HRV during sleep, the change in HRV from sleep to morning with exercise is greater in younger subjects. These physiologic results have clinical significance in understanding the pathophysiology of altered variability in ill patients.  相似文献

Pyruvate was produced from glucose by Escherichia coli BW25113 that contained formate dehydrogenase (FDH) from Mycobacterium vaccae. In aerobic shake-flask culture (K (L) a?=?4.9?min(-1)), the recombinant strain produced 6.7?g pyruvate?l(-1) after 24?h with 4?g sodium formate?l(-1) and a yield of 0.34?g pyruvate?g?glucose(-1). These values were higher than those of the original strain (0.2?g?l(-1) pyruvate and 0.02?g pyruvate?g?glucose(-1)). Based on the reaction mechanism of FDH, the introduction of FDH into E. coli enhances the accumulation of pyruvate by the regeneration of NADH from NAD(+) since NAD(+) is a shared cosubstrate with the pyruvate dehydrogenase complex, which decarboxylates pyruvate to acetyl-CoA and CO(2). The oxygenation level was enough high to inactivate lactate dehydrogenase, which was of benefit to pyruvate accumulation without lactate as a by-product.  相似文献

Bioconversion of biodiesel-derived crude glycerol into carotenoids and lipids was investigated by a microbial conversion of an oleaginous red yeast Sporidiobolus pararoseus KM281507. The methanol content in crude glycerol (0.5%, w/v) did not show a significant effect on biomass production by strain KM281507. However, demethanolized crude glycerol significantly supported the production of biomass (8.64?±?0.13?g/L), lipids (2.92?±?0.03?g/L), β-carotene (15.76?±?0.85?mg/L), and total carotenoids (33.67?±?1.28?mg/L). The optimal conditions suggested by central composite design were crude glycerol concentration (55.04?g/L), initial pH of medium (pH 5.63) and cultivation temperature (24.01°C). Under these conditions, the production of biomass, lipids, β-carotene, and total carotenoids were elevated up to 8.83?±?0.05, 4.00?±?0.06?g/L, 27.41?±?0.20, and 53.70?±?0.48?mg/L, respectively. Moreover, an addition of olive oil (0.5???2.0%) dramatically increased the production of biomass (14.47?±?0.15?g/L), lipids (6.40?±?0.09?g/L), β-carotene (54.43?±?0.95?mg/L), and total carotenoids (70.92?±?0.51?mg/L). The oleic acid content in lipids was also increased to 75.1% (w/w) of total fatty acids, indicating a good potential to be an alternative biodiesel feedstock. Meanwhile, the β-carotene content in total carotenoids was increased to 76.7% (w/w). Undernutrition during pregnancy and/or lactation plays an important role on the overall health of offspring later in life. Using a rodent model, the present study was conducted to examine the effect of fetal and early postnatal iron deficiency on iron metabolism in adult animals. Rats were treated with three stages of low or normal iron diets from gestation until the end of the study. During the first stage (4?weeks prior to 3?weeks after pregnancy, total 7?weeks), two groups of adult females (dams) were fed with either a low-iron (7.4?mg iron/kg, group LD) or control-iron (274?mg/kg, group CD) diet. During the second stage (from 3 to 13?weeks of age, total 10?weeks), all pups from stage 1 (both the LD and CD groups) were placed on a control-iron diet for 10?weeks (groups LD?CCD and CD?CCD, respectively). During the third stage (from 13 to 29?weeks of age, total 16?weeks), both LD?CCD and CD?CCD groups from stage 2 were fed with a low-iron (named LD?CCD?CLD and CD?CCD?CLD groups, respectively). We found that the live birth rate of the offspring of the LD dams (84.7?%) was significantly lower than that of the CD dams (95.4?%). During stage 2, the mean body weight of the LD?CCD male or LD?CCD female rats exceeded the CD?CCD male rats (p?<?0.05). Compared with the CD?CCD?CLD rats, the LD?CCD?CLD rats had significantly increased total iron binding capacity, and higher levels of transferrin, serum erythropoietin (EPO), renal EPO mRNA, duodenal divalent metal transporter-1, and renal transferrin receptors. A recombinant enzyme from Lysinibacillus fusiformis was expressed, purified, and identified as an oleate hydratase because the hydration activity of the enzyme was the highest for oleic acid (with a k (cat) of 850?min(-1) and a K (m) of 540?μM), followed by palmitoleic acid, γ-linolenic acid, linoleic acid, myristoleic acid, and α-linolenic acid. The optimal reaction conditions for the enzymatic production of 10-hydroxystearic acid were pH 6.5, 35?°C, 4% (v/v) ethanol, 2,500?U ml(-1) (8.3?mg?ml(-1)) of enzyme, and 40?g l(-1) oleic acid. Previous studies have shown that the deletion of brnQ from the Corynebacterium glutamicum chromosome results in a significant reduction in L-isoleucine uptake rates, while overexpression of brnFE leads to enhanced L-isoleucine export rates. Given that net excretion rates would be an important factor for high titers of L-isoleucine accumulation, we have tested the notion that decreased L-isoleucine uptake combined with increased L-isoleucine excretion will further improve high-yield strains that are currently used for the industrial-scale production of L-isoleucine. To examine the effect of the two carriers on L-isoleucine accumulation in L-isoleucine producer C. glutamicum YILW, we constructed a brnQ deletion mutant (C. glutamicum YILW?brnQ) and two brnFE overexpressors (C. glutamicum YILWpXMJ19brnFE and C. glutamicum YILW?brnQpXMJ19brnFE). Compared to the original strain, the efflux rate of the brnQ mutant increased from 19.0 to 23.6?nmol?min(-1) mg (dry wt)(-1) and its L-isoleucine titer increased from 154.3?mM (20.2?g?l(-1)) to 170.3?mM (22.3?g?l(-1)). The efflux rates of C. glutamicum YILWpXMJ19brnFE and C. glutamicum YILW?brnQpXMJ19brnFE were 33.5 and 39.1?nmol?min(-1) mg (dry wt)(-1), and their L-isoleucine production titers were 197.2?mM (25.9?g?l(-1)) and 221.0?mM (29.0?g?l(-1)), respectively. This study evaluated the effects of anti-lipid peroxidases when supplemented to the thawing and incubation media of frozen-thawed boar spermatozoa. Semen pellets were thawed and incubated in media with 1.0 mM α-tocopherol or diethylenetriamine. Ecosystem nitrous oxide (N(2) O) emissions respond to changes in climate and CO(2) concentration as well as anthropogenic nitrogen (N) enhancements. Here, we aimed to quantify the responses of natural ecosystem N(2) O emissions to multiple environmental drivers using a process-based global vegetation model (DyN-LPJ). We checked that modelled annual N(2) O emissions from nonagricultural ecosystems could reproduce field measurements worldwide, and experimentally observed responses to step changes in environmental factors. We then simulated global N(2) O emissions throughout the 20th century and analysed the effects of environmental changes. The model reproduced well the global pattern of N(2) O emissions and the observed responses of N cycle components to changes in environmental factors. Simulated 20th century global decadal-average soil emissions were c. 8.2-9.5?Tg?N?yr(-1) (or 8.3-10.3?Tg?N?yr(-1) with N deposition). Warming and N deposition contributed 0.85?±?0.41 and 0.80?±?0.14?Tg?N?yr(-1) , respectively, to an overall upward trend. Rising CO(2) also contributed, in part, through a positive interaction with warming. The diversity and density of methanogenic archaea and methane production were investigated ex situ at different growth stages of rice plant cultivated in compost-treated tropical rice fields. The qPCR analysis revealed variation in methanogens population from 3.40?×?10(6) to 1.11?×?10(7) copies?g(-1) dws, in the year 2009 and 4.37?×?10(6) to 1.36?×?10(7) copies?g(-1) dws in the year 2010. Apart from methanogens, a large number of bacterial (9.60?×?10(9) -1.44?×?10(10) copies?g(-1) dws) and archaeal (7.13?×?10(7) -3.02?×?10(8) copies?g(-1) dws) communities were also associated with methanogenesis. Methanogen population size varied in the order: flowering > ripening > tillering > postharvest > preplantation stage. The RFLP-based 16S rRNA gene-targeted phylogenetic analysis showed that clones were closely related to diverse group of methanogens comprising members of Methanomicrobiaceae, Methanosarcinaceae, Methanosaetaceae and RC I. Laboratory incubation studies revealed higher amount of cumulative CH(4) at the flowering stage. The integration of methanogenic community structure and CH(4) production potential of soil resulted in a better understanding of the dynamics of CH(4) production in organically treated rice-field soil. The hypothesis that the stages of plant development influence the methanogenic community structure leading to temporal variation in the CH(4) production has been successfully tested.  相似文献

The first aim of the study was to assess clock-time patterning of work-related injuries (WRIs) of firemen (FM) of Sa?ne et Loire-71 (France) during the 4-yr span of 1 January 2004 to 31 December 2007. FM of this service are legally required to log every WRI and seek its evaluation by the medical service, whether the WRI was the result of worksite duties or exercise/sport activities at the station. WRI was defined specifically as a (nonexercise, nonsport, and nonemotional/stress) work-associated trauma, verified both by log book and medical records. For the corresponding years, the 24-h pattern of emergency calls (Calls) plus road traffic (Traffic) on the main roads of the service area was also assessed. Relative risk (R) of WRI was calculated as the quantity of WRIs/h divided by the quantity of Call responses/h?×?1000, which takes into account the number of at-risk FM/unit time, since each dispatched emergency vehicle is staffed with 4 FM. Comparably trained regular (RFM) and volunteer (VFM) FM experienced a total of 187 WRIs. The 24-h WRI curve patterns of RFM and VFM were correlated (r?=?0.4, p??.05). In the present study, chronic behavioural stress resulting from low social status affected the physiological responses of rainbow trout (Oncorhynchus mykiss) to a subsequent acute stressor, exposure to hypoxia. Rainbow trout were confined in fork-length matched pairs for 48-72?h, and social rank was assigned based on behaviour. Dominant and subordinate fish were then exposed individually to graded hypoxia (final water PO(2), PwO(2)?=?40?Torr). Catecholamine mobilization profiles differed between dominant and subordinate fish. Whereas dominant fish exhibited generally low circulating catecholamine levels until a distinct threshold for release was reached (PwO(2)?=?51.5?Torr corresponding to arterial PO(2), PaO(2)?=?24.1?Torr), plasma catecholamine concentrations in subordinate fish were more variable and identification of a distinct threshold for release was problematic. Gill remodeling in goldfish (Carassius auratus) is accomplished by the appearance or retraction of a mass of cells (termed the interlamellar cell mass or ILCM) between adjacent lamellae. Given the presumed effects of gill remodeling on diffusing capacity, the goals of the current study were (1) to determine the consequences of increased aerobic O(2) demand (swimming) on gill remodelling and (2) to assess the consequences of the presence or absence of the ILCM on aerobic swimming capacity. Fish acclimated to 7?°C exhibited a marked increase in the ILCM which occupied, on average, 70.0?±?4.1?% of the total interlamellar channel area in comparison to an average ILCM area of only 28.3?±?0.9?% in fish acclimated to 25?°C. Incrementally increasing swimming velocity in fish at 7?°C to achieve a maximum aerobic swimming speed (U (CRIT)) within approximately 3?h resulted in a marked loss of the ILCM area to 44.8?±?3.5?%. Fish acclimated to 7?°C were subjected to 35?min swimming trials at 30, 60 or 80?% U (CRIT) revealing that significant loss of the ILCM occurred at swimming speeds exceeding 60?% U (CRIT). Prior exposure of cold water-acclimated fish to hypoxia to induce shedding of the ILCM did not affect swimming performance when assessed under normoxic conditions (control fish U (CRIT)?=?2.34?±?0.30 body lengths s(-1); previously hypoxic fish U (CRIT)?=?2.99?±?0.14 body lengths s(-1)) or the capacity to raise rates of O(2) consumption with increasing swimming speeds. Because shedding of ILCM during U (CRIT) trials complicated the interpretation of experiments designed to evaluate the impact of the ILCM on swimming performance, additional experiments using a more rapid 'ramp' protocol were performed to generate swimming scores. Neither prior hypoxia exposure nor a previous swim to U (CRIT) (both protocols are known to cause loss of the ILCM) affected swimming scores (the total distance swum during ramp U (CRIT) trials). However, partitioning all data based on the extent of ILCM coverage upon cessation of the swimming trial revealed that fish with less than 40?% ILCM coverage exhibited a significantly greater swimming score (539?±?86?m) than fish with greater than 50?% ILCM coverage (285?±?70?m). Thus, while loss of the ILCM at swimming speeds exceeding 60?% U (CRIT) confounds the interpretation of experiments designed to assess the impact of the ILCM on swimming performance, we suggest that the shedding of the ILCM, in itself, coupled with improved swimming scores in fish exhibiting low ILCM coverage (<40?%), provide evidence that the ILCM in goldfish acclimated to cold water (7?°C) is indeed an impediment to aerobic swimming capacity.  相似文献

Helicobacter pylori infection could impair iron absorption from fortified products. The objective of the study was to determine the effect of H. pylori infection on iron absorption from asymptomatic adults consuming wheat flour fortified with iron and zinc. The (13)C urea breath test was used to assess H. pylori infection. Twenty-four H. pylori-positive and 26 H. pylori-negative volunteers completed the study. On day?1, the subjects were randomized to receive for breakfast bread fortified with either ferrous sulfate and zinc sulfate or ferrous fumarate and zinc oxide. Bread fortified with ferrous sulfate was labeled with (59)Fe as sulfate, and bread fortified with ferrous fumarate was labeled with (55)Fe as fumarate. On day?3, they received the other type of bread, with the respective tracers. On days?18-23, a proton pump inhibitor was administered to all subjects. The cofortification of milk with iron (Fe) and zinc (Zn) is a strategy used to prevent these deficiencies during childhood. Given that Zn can negatively interact with iron in aqueous solutions, the objective of the present study was to determine the effect of Zn on Fe absorption of milk fortified with Fe and Zn. Twenty-eight women between 33 and 47?years of age, with contraception and a negative pregnancy test, participated in one of two absorption studies. They received on four different days, after an overnight fast, 200?mL of milk (26?% fat) fortified with 10?mg Fe/L, as (A) ferrous sulfate, or the same milk but with graded doses of added Zn, as Zn sulfate of (B) 5, (C) 10, and (D) 20?mg/L (study 1, n?=?15). In study 2 (n?=?13), subjects received the same milk formulations, but these were also fortified with ascorbic acid (70?mg/L). Milk was labeled with radioisotopes 59Fe or 55Fe, and the absorption of iron was measured by erythrocyte incorporation of radioactive Fe. Screening of bacteria from Sambhar lake, an extreme hypersaline environment of India, led to the isolation of 93 haloalkaliphilic bacteria growing optimally in media with 2?C25?% salt and 6?C12 pH. Based on 16S rRNA gene sequences, 93 isolates were further categorized into 32 groups, with each group representing a different taxa belonging to 3 phyla (Firmicutes, Proteobacteria and Actinobacteria). Majority of the isolates (53.12?%) showed similarity with phylum Firmicutes which was followed by Proteobacteria (40.63?%) and Actinobacteria (6.25?%). The isolates belonging to 32 representative groups were further evaluated for the production of extracellular enzymes viz. amylase, cellulase, protease and xylanase, plant growth promoting attributes and BIOLOG? substrate usage. Among all the isolates, xylanase producing isolates were in maximum (68?%) as compared to protease (56?%), cellulase (40?%), and amylase (37?%) producing strains. Similarly, among plant growth promoting activities, ammonia producing isolates were highest (56?%) when compared to those producing ACC deaminase (53?%), IAA (50?%), hydrogen cyanide (28?%), siderophore (21?%) and solubilizing P (34?%). The gills contain essential cells for respiration and osmoregulation, whereas the hepatopancreas is the site of digestion, absorption, and nutrients storage. The aim of this work was to separate and characterize gill and hepatopancreatic cells of the mangrove crab, Ucides cordatus. For gills, the methodology consisted of an enzymatic cellular dissociation using Trypsin at 0.5%, observation of cellular viability with Tripan Blue, and separation of cells using discontinuous sucrose gradient at concentrations of 10%, 20%, 30%, and 40%. The hepatopancreatic cells were dissociated by magnetic stirring, with posterior separation by sucrose gradient at the same concentrations above. For gills, a high cellular viability was observed (92.5±2.1%), with hemocyte cells in 10% sucrose layer (57.99?±?0.17%, *P?相似文献
