为了探究外源物一氧化氮(NO)供体硝普钠(sodium nitroprusside,SNP)对Cd2+胁迫下小麦根系生长发育和活性氧代谢的影响,以小麦(Triticum aestivum L.)为材料,研究10 mmol/L CdCl2胁迫下,30 μmol/L硝普钠(含一氧化氮NO)对小麦根系生长发育和活性氧代谢的影响。结果显示,外施SNP后,Cd2+胁迫下的小麦根长度、鲜重与干重较单独镉胁迫处理分别上升了48.0%、107.7%和87.3%,根系超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)的活性分别上升了28.5%、7.4%、19.2%和9.8%,根中超氧自由基(O2.-)和过氧化氢(H2O2)的含量分别降低了80.5%和47.0%;同时外施SNP,使镉胁迫下小麦根中的可溶性糖含量和脯氨酸含量分别上升了24.7%和22.1%;使根中丙二醛(MDA)含量降低了30.2%;使根系活力上升了15.3%。因此,外源NO在一定程度上可以显著提高小麦根的抗氧化能力,增强小麦的抗逆性,缓解镉对小麦根系的毒害,进而促进小麦幼苗根系的生长发育。  相似文献

Nitric oxide (NO) is a bioactive gaseous, multifunctional molecule playing a central role and mediating a variety of physiological processes and responses to biotic and abiotic stresses including heavy metals. The present study investigated whether NO applied exogenously as sodium nitroprusside (SNP) has any protective role against arsenic (As) toxicity in Oryza sativa (rice). Treatment with 50 μM SNP (a NO donor) significantly ameliorated the As-induced (25 or 50 μM) decrease in root and coleoptile length of rice. Further, As-induced oxidative stress measured in terms of malondialdehyde (MDA), superoxide ion (), root oxidizability and H2O2 content was lesser upon supplementation of NO. It indicated a reactive oxygen species (ROS) scavenging activity of NO. NO addition reversed (only partially) the As-induced increase in activities of antioxidant enzymes – superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, and catalase. The study concludes that exogenous NO provides resistance to rice against As-toxicity and has an ameliorating effect against As-induced stress.  相似文献

【目的】研究干旱胁迫对加拿大飞蓬种子萌发及幼根生长的影响,为科学防控加拿大飞蓬入侵提供依据。【方法】以当年生加拿大飞蓬成熟种子为材料,分别用蒸馏水(CK)和不同浓度5%、10%、15%、20%、25%的聚乙二醇溶液(PEG)(分别表示轻度、中度、重度、极度和极重度干旱胁迫强度)对加拿大飞蓬种子进行处理。测定日相对发芽率、发芽率、发芽势等指标,分析加拿大飞蓬种子萌发情况;测定幼苗根系宽、根系高、根平均直径、根面积、总根尖数及根分叉数,分析其幼苗根生长情况;并测定种子恢复萌发情况。【结果】无干旱胁迫的加拿大飞蓬种子萌发指标最高,随着干旱胁迫强度增加,种子萌发指标减小,极重度干旱胁迫下加拿大飞蓬种子不萌发。与CK相比,轻度干旱胁迫和重度干旱胁迫对加拿大飞蓬根平均直径影响不显著,但会促进侧根与不定根生长;重度干旱胁迫下,加拿大飞蓬的总根尖数和分叉数显著增加。加拿大飞蓬种子在PEG浓度为20.13%的干旱胁迫下不发芽。种子在极度和极重度干旱胁迫下不萌发,但当水分条件充足时,种子依然具有萌发的能力。【结论】建议在加拿大飞蓬种子尚未成熟前进行人工拔除,或在加拿大飞蓬入侵地种植耐旱强、生长快、根系发达、竞争性强的植物,以控制加拿大飞蓬的生长发育。  相似文献

入侵种与本地种杂交可能会改变其某些性状,影响其生理生化过程,增强其对一些重金属的耐受性。镉是目前造成土壤污染的主要重金属元素之一,为探讨杂交是否能够增强植物对重金属镉的耐受性,采用培养皿滤纸法比较了不同浓度镉(0,10,40,80和120 mg/L)处理下瘤突苍耳(Xanthium strumarium)、苍耳(Xanthium sibiricum)及二者杂合体(X.strumarium♀×X.sibiricum♂,X.sibiricum♀×X.strumarium♂)种子萌发与幼苗生长情况。结果表明:1)入侵种瘤突苍耳与本地种苍耳之间存在杂交现象;2)镉促进了杂交瘤突苍耳(X.strumarium♀×X.sibiricum♂)种子的萌发,当镉浓度为80 mg/L时,其萌发率、发芽势、发芽指数最高;3)杂交瘤突苍耳在80、120 mg/L镉处理下根长和叶绿素含量低于瘤突苍耳,但子叶受到的氧化胁迫并未高于瘤突苍耳。研究结果显示,杂交有可能提高了瘤突苍耳在高浓度镉胁迫时对重金属镉的转运能力,幼苗根系吸收大量的镉并向上运输给子叶,导致根系生长受到抑制、子叶叶绿素含量下降,抗氧化酶系统可能在降...  相似文献

In the present study, we evaluate the protective effect of nitric oxide (NO) against senescence of rice leaves promoted by ABA. Senescence of rice leaves was determined by the decrease of protein content. ABA treatment resulted in (1) induction of leaf senescence, (2) increase in H2O2 and malondialdehyde (MDA) contents, (3) decrease in reduced form glutathione (GSH) and ascorbic acid (AsA) contents, and (4) increase in antioxidative enzyme activities (superoxide dismutase, ascorbate peroxidase, glutathione reductase, and catalase). All these ABA effects were reduced by free radical scavengers such as sodium benzoate and GSH. NO donors [N-tert-butyl-alpha-phenylnitrone (PBN), sodium nitroprusside, 3-morpholinosydonimine, and AsA + NaNO2] were effective in reducing ABA-induced leaf senescence. PBN prevented ABA-induced increase in the contents of H2O2 and MDA, decrease in the contents of GSH and AsA, and increase in the activities of antioxidative enzymes. The protective effect of PBN on ABA-promoted senescence, ABA-increased H2O2 content and lipid peroxidation, ABA-decreased GSH and AsA, and ABA-increased antioxidative enzyme activities was reversed by 2-(4-carboxy-2-phenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide, a NO-specific scavenger, suggesting that the protective effect of PBN is attributable to NO released. Reduction of ABA-induced senescence by NO in rice leaves is most likely mediated through its ability to scavenge active oxygen species including H2O2.  相似文献

【目的】山东省青岛市崂山区的黑松上发现小蠹危害情况,本研究对采集到的小蠹进行鉴定,以明确小蠹种类,为更好地调查生物多样性以及开展林间蛀干害虫监测和防控工作提供依据。【方法】对采自黑松树体上的小蠹进行形态学观察、拍照,并且提取其基因组DNA,利用小蠹亚科通用引物扩增线粒体细胞色素氧化酶亚基Ⅰ(COⅠ)序列进行测序,在NCBI数据库中进行序列比对。【结果】基于形态学和分子生物学2方面的鉴定,采集到的小蠹为松曲毛小蠹。其成虫体黄色,前胸背板前端有类似横向刻痕,鞘翅面有凹凸不平的沟,整个虫体侧缘有稀疏的绒毛,雌虫头部绒毛多于雄虫。基于COⅠ基因序列构建系统发育树( ML树) 显示,山东青岛小蠹样本与意大利样本序列聚成一支,置信度为 100%。【结论】黑松上发现的松曲毛小蠹为山东新记录种。此研究结果为小蠹快速DNA鉴定以及溯源分析提供基础数据,有关部门应加强黑松蛀干害虫的监测和防控。  相似文献

Flavonoids and coumaric acid derivatives were identified from dandelion flower (Taraxacum officinale). Characteristics of chain-breaking antioxidants, such as extended lag phase and reduced propagation rate, were observed in oxidation of linoleic acid emulsion with the addition of dandelion flower extract (DFE). DFE suppressed both superoxide and hydroxyl radical, while the latter was further distinguished by both site-specific and non-specific hydroxyl radical inhibition. DPPH-radical-scavenging activity and a synergistic effect with alpha-tocopherol were attributed to the reducing activity derived from phenolic content of DFE. A significant (p < 0.05) and concentration-dependent, reduced nitric oxide production from acterial-lipopolysaccharide-stimulated mouse macrophage RAW264.7 cells was observed with the addition of DFE. Moreover, peroxyl-radical-induced intracellular oxidation of RAW264.7 cells was inhibited significantly (p < 0.05) by the addition of DFE over a range of concentrations. These results showed that the DFE possessed marked antioxidant activity in both biological and chemical models. Furthermore, the efficacy of DFE in inhibiting both reactive oxygen species and nitric oxide were attributed to its phenolic content.  相似文献

Nitric oxide (NO) is a gaseous free radical that reacts with O2 in air and aqueous solution. NO donors have been widely used to circumvent the difficulties inherent in working with a reactive gas, but NO donors do not deliver NO at a constant rate for prolonged periods of time. Furthermore, some of the most commonly used NO donors produce additional, bioactive decomposition products. We designed and built an apparatus that allowed for the precise mixing of gaseous NO with air and the delivery of gas through sample vials at fixed rates. This experimental setup has the added advantage that continuous flow of gas over the sample reduces the buildup of volatile breakdown products. To show that this experimental setup was suitable for studies on the dormancy and germination of Arabidopsis thaliana seeds, we introduced vapors from water or sodium nitroprusside (SNP) into the gas stream. Seeds remained dormant when treated with water vapor, but gases generated by SNP increased germination to 90%. When pure NO was mixed with air and passed over dormant seeds, ∼ ∼30% of the seeds germinated. Because nitrite accumulates in aqueous solutions exposed to NO gas, we measured the accumulation of nitrite under our experimental conditions and found that it did not exceed 100 µM. Nitrite or nitrate at concentrations of up to 500 µM did not increase germination of C24 ecotype Arabidopsis seeds to more than 10%. These data support the hypothesis that NO participates in the loss of Arabidopsis seed dormancy, and they show that for some dormant seeds, exposure to exogenous NO is sufficient to trigger germination.  相似文献

随着毛竹杉木混交林面积的不断扩大,它们之间既会产生促进作用,又会产生抑制作用。本研究对比分析了不同浓度毛竹各器官(鲜叶、干叶、枝条、竹杆、竹鞭和鞭根)的浸提液及枯落物、根际土浸提液对杉木种子发芽率的影响,探讨了毛竹各器官浸提液及枯落物、根际土浸提液对杉木种子萌发的化感作用。结果表明:(1)处理前期,毛竹各器官浸提液对杉木种子萌发具有抑制作用,且当浸提液浓度为1∶25和1∶50的高浓度时,相对于低浓度浸提液对杉木种子萌发的抑制作用更为持久;(2)处理后期,低浓度的毛竹各器官浸提液对杉木种子萌发促进作用较快,而高浓度的浸提液仍呈现抑制作用,促进作用出现较为缓慢;(3)枯落叶浸提液总体上对杉木种子萌发具有抑制作用,在整个实验过程中低于对照组的发芽率,仅在1∶50浓度时略高对照组实验;而根际土浸提液对杉木种子萌发自始至终都具有促进作用,其中低浓度的促进作用十分显著。故在实际的毛竹杉木混交林生产营林过程中可选用毛竹根际土制备浸提液,促进杉木种子萌发,为毛竹杉木混交林的生产经营提供参考。  相似文献

化感作用是影响人工混交林种间关系的重要因素之一。已有研究表明:喜树南方红豆杉混交对喜树生长有明显促进作用,并从混交改善混交林地微气候角度解释了这种促进作用,但未从种间化感作用角度探讨这种作用。为了探究南方红豆杉是否对喜树具有潜在的化感促进作用,从而更全面了解喜树南方红豆杉混交林种间关系,考察了不同浓度(25、50 g/L和100 g/L)的南方红豆杉(Taxus chinensis var.mairei)鲜叶、凋落叶、枝和根水浸提液对喜树(Camptotheca acuminata)种子发芽和幼苗生长的影响。结果表明:(1)南方红豆杉凋落叶浸提液对喜树种子发芽和幼苗生长无显著影响(P>0.05),而鲜叶、枝和根浸提液对喜树发芽和幼苗生长均具有不同程度的促进作用(P<0.05),且作用强度大体与浓度呈正相关;(2)100 g/L的南方红豆杉鲜叶、枝和根浸提液浸泡的喜树种子,其发芽率比对照组分别提高8.1%、14.9%和25.6%;(3)南方红豆杉鲜叶浸提液只有在高浓度(100 g/L)时,对喜树幼苗基径生长具有促进作用(P<0.05),而对其株高、全株干重和净光合速率无显著影响;南方红豆杉根和枝浸提液对喜树幼苗株高、基茎、干重和净光合速率均具有促进作用(P<0.05),与对照组相比,100 g/L的根和枝浸提液浇灌喜树幼苗,可以使喜树幼苗株高分别提高14.2%和8.4%,基径分别提高19.0%和15.3%,干重分别提高23.1%和15.9%,净光合速率分别提高11.6%和6.1%。研究结果表明,在喜树南方红豆杉混交林中,南方红豆杉对喜树的正向化感作用可能是促进喜树生长的重要因素之一。  相似文献

Root exudates influence significantly physical, chemical and biological characteristics of rhizosphere soil. Their qualitative and quantitative composition is affected by environmental factors such as pH, soil type, oxygen status, light intensity, soil temperature, plant growth, nutrient availability and microorganisms. The aim of the present study was to assess the influence of growth substrate and plant age on the release of carboxylates from Lupinus albus L. and Brassica napus L.Both plant species were studied in continuously percolated microcosms filled with either sand, soil or sand + soil (1:1) mixture. Soil solution was collected every week at 7, 14, 21, 28 and 35 days after planting (DAP). Carboxylate concentrations were determined by reversed-phase liquid chromatography - electrospray ionization - time of flight mass spectrometry (LC-ESI-TOFMS).Oxalate, citrate, succinate, malate and maleate were detected in soil solutions of both plant species. Their concentrations were correlated with the physiological status of the plant and the growth substrate. Oxalate was the predominant carboxylate detected within the soil solution of B. napus plants while oxalate and citrate were the predominant ones found in the soil solutions of L. albus plants.The sampling determination of carboxylates released by plant roots with continuous percolation systems seems to be promising as it is a non-destructive method and allows sampling and determination of soluble low molecular weight organic compounds derived from root exudation as well as the concentration of soluble nutrients, which both might reflect the nutritional status of plants.  相似文献

为探究Zn、Cu与盐复合胁迫对翅碱蓬(Suaeda salsa)萌发生长的影响机理及其调控措施,以翅碱蓬为研究对象,采用水培试验方法,测定Zn、Cu和盐复合胁迫条件下翅碱蓬种子发芽率、萌发速率和幼苗渗透调节物质含量等指标,分析1.5 mg/L吲哚乙酸(IAA)、100 mg/L赤霉素(GA)和0.3%硝酸钾(KNO3)处理对Zn、Cu与盐复合胁迫条件下翅碱蓬萌发与生长的影响。结果表明:(1) Zn、Cu与高盐复合胁迫极显著降低翅碱蓬种子的发芽率,中高浓度的Zn、Cu与盐复合胁迫极显著降低翅碱蓬种子的萌发速率,Zn、Cu和盐复合胁迫对翅碱蓬种子的萌发生长影响表现为低促高抑效应,影响因子间存在明显的协同效应;(2)Zn、Cu和盐复合胁迫条件下,随着盐浓度的升高,翅碱蓬幼苗体内过氧化氢酶(CAT)、过氧化物酶(POD)和超氧化物歧化酶(SOD)3种抗氧化酶活性呈先升高后降低的变化趋势,Zn、Cu和高盐复合胁迫使翅碱蓬幼苗体内丙二醛(MDA)含量增加近2.5倍;(3)1.5 mg/L IAA溶液浸种12 h可显著提高Zn、Cu与高盐复合胁迫条件下翅碱蓬种子的发芽率和萌发速...  相似文献

为揭示植物适应锰胁迫的生理机制,通过在不同Mn2+浓度(0、1、5、10、15、20 mmol/L)下开展盐肤木(Rhus chinensis)种子萌发以及幼苗生长实验,检测锰胁迫处理7、15、30 d后幼苗生理生化特性的变化。结果表明:(1)随着Mn2+浓度的升高,盐肤木种子发芽率变化不显著,在80.0%-81.6%之间,发芽势、发芽指数和活力指数则呈先升后降的趋势;其幼苗生物量也呈现先升后降的趋势;(2)随着Mn2+浓度的升高与胁迫时间的延长,盐肤木幼苗叶绿素a、叶绿素b含量均呈现先增加后降低的趋势,类胡萝卜素含量呈现下降的趋势;(3)胁迫7 d时,随着Mn2+浓度的升高,盐肤木幼苗超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性均显著上升;胁迫15、30 d时,高Mn2+浓度(15-20 mmol/L)下POD、CAT活性则均降低;(4)胁迫7 d时,随着Mn2+浓度的升高,可溶性糖、可溶性蛋白、游离脯氨酸含量升高;胁迫15、30 d时,在Mn2+浓度为20 mmol/L时可溶性蛋白与游离脯氨酸含量显著降低;(5)随着Mn2+浓度的升高与胁迫时间的延长,丙二醛(MDA)含量均升高。研究说明盐肤木具有较强的耐受锰胁迫能力,它可通过增强抗氧化酶活性、积累渗透调节物质含量来应对锰胁迫。  相似文献

为探讨人工巨尾桉林叶片挥发物对周边农作物的化感作用,采用不同质量新鲜巨尾桉叶片及由新鲜叶片提取的桉叶油对玉米、辣椒、西红柿等三种植物种子进行处理,观测其种子萌发和幼苗生长状况。结果表明:(1)在三种被测植物中,桉树叶片挥发物对玉米种子萌发影响最小,对辣椒影响最大;(2)当叶片用量小于200g时,桉树叶片挥发物对三种植物种子萌发影响不明显,当叶片用量达400g时,能完全抑制辣椒、西红柿种子萌发,并能极显著降低玉米种子萌发(P0.01);(3)玉米幼苗芽生长随叶片用量的增加呈现先促进后抑制现象,对芽高、鲜重、干重的促进和抑制均达到显著(P0.05)或极显著水平(P0.01);(4)当叶片用量小于或等于100g时,桉树叶片挥发物对辣椒、西红柿幼苗生长影响不明显,当用量达到200g时则能极显著抑制辣椒、西红柿幼苗生长;(5)桉叶油对三种测试植物的抑制效果与叶片自然挥发物相似,且效果更显著。  相似文献

恶劣环境下,人工海防林因面临养分胁迫而经营困难。为探讨盐、磷胁迫对主要海防林树种木麻黄和台湾相思种子萌发及生长的影响,该研究分别用不同浓度的NaCl(盐)和KH2PO4(磷)溶液处理种子和浇灌幼苗,测定种子萌发和幼苗生长指标。结果表明:(1)高盐胁迫显著抑制种子萌发,对幼苗生长有一定影响,但两种植物影响程度不同;台湾相思种子萌发耐盐性高于木麻黄,前者相对盐害率最大值为23.03%,后者为89.15%;随着盐浓度增加,木麻黄和台湾相思种子的发芽率、发芽势、发芽指数和活力指数均降低,对应最大值分别为38.70%、34.67%、18.70、0.055和76.67%、62.22%、48.46、6.11。(2)两种植物的株高和根长随盐浓度增加而降低,木麻黄和台湾相思株高分别为12.29~6.01 mm和48.27~17.33 mm,根长分别为8.57~1.45 mm和33.41~5.88 mm;台湾相思根、茎、叶生物量及根冠比均随盐浓度的增加逐渐减小,木麻黄各处理差异较小。(3)台湾相思的种子和幼苗较木麻黄更耐低磷环境,二者最适磷浓度存在差异;木麻黄种...  相似文献

Following our previous findings of high extracellular redox activity in lichens, the results of the work presented here identify the enzymes involved as laccases. Despite numerous data on laccases in fungi and flowering plants, this is the first report of the occurrence of laccases in lichenized ascomycetes. Extracellular laccase activity was measured in 40 species of lichens from different taxonomic groupings and contrasting habitats. Out of 20 species tested from suborder Peltigerineae, 18 displayed laccase activity, while activity was absent in species tested from other lichen groups. Identification of the enzymes as laccases was confirmed by the ability of lichen leachates to readily metabolize substrates such as 2,2′-azino(bis-3-ethylbenzthiazoline-6-sulfonate) (ABTS), syringaldazine and o-tolidine in the absence of hydrogen peroxide, sensitivity of the enzymes to cyanide and azide, the enzymes having typical laccase pH and temperature optima, and an absorption spectrum with a peak at 614 nm. Desiccation and wounding stimulated laccase activity. Laccase activity was not increased after treatment with normal inducers of laccase synthesis, suggesting that they are constitutively expressed. Electrophoresis showed that the active form of laccase from Peltigera malacea was a tetramer with an unusually high molecular mass of 340 kDa and an isoelectric point (pI) of 4.7. The finding of abundant extracellular redox enzymes known to actively produce reactive oxygen species suggest that their roles may include increasing nutrient supply to lichens by delignification, and deterring pathogens by contributing to the oxidative burst. Furthermore, once released into the environment, they may participate in the carbon cycle by facilitating the breakdown or formation of humic substances.  相似文献

The phylogenetic distribution and structural diversity of the nitric oxide synthases (NOS) remain important and issues that are little understood. We present sequence information, as well as phylogenetic analysis, for three NOS cDNAs identified in two non-mammalian species: the vertebrate marine teleost fish Stenotomus chrysops (scup) and the invertebrate echinoderm Arbacia punctulata (sea urchin). Partial gene sequences containing the well-conserved calmodulin (CaM)-binding domain were amplified by RT-PCR. Identical 375-bp cDNAs were amplified from scup brain, heart, liver and spleen; this sequence shares 82% nucleic acid and 91% predicted amino acid identity with the corresponding region of human neuronal NOS. A 387-bp cDNA was amplified from sea urchin ovary and testes; this sequence shares 72% nucleic acid identity and 65% deduced amino acid identity with human neuronal NOS. A second cDNA of 381 bp was amplified from sea urchin ovary and it shares 66% nucleic acid and 57% deduced amino acid identity with the first sea urchin sequence. Together with earlier reports of neuronal and inducible NOS sequences in fish, these data indicate that multiple NOS isoforms exist in non-mammalian species. Phylogenetic analysis of these sequences confirms the conserved nature of NOS, particularly of the calmodulin-binding domains.  相似文献

为了探讨凋落物物理阻隔对格氏栲天然林自然更新状况的影响,通过模拟野外凋落物覆盖,设置格氏栲种子上层覆盖厚0 cm(CK)、2 cm(D2)、4 cm(D4)、6 cm(D6)、8 cm(D8)及种子下层铺垫厚2 cm(U2)、4 cm(U4)凋落物等7个处理,分析凋落物覆盖方式及厚度对格氏栲种子萌发及胚根生长的影响。结果表明:(1)凋落物覆盖方式及厚度对种子萌发进程存在显著影响。CK萌发持续时间最长,上层覆盖处理(D)次之,下层铺垫处理(U)的种子起始萌发时间显著滞后。(2)CK种子萌发率最高,其次D6处理发芽速度较快,发芽整齐;U处理较D处理的发芽率及发芽势均显著降低,且萌发抑制率显著增加。(3)D处理的种子胚根生长速度快,胚根长度大于CK;U处理的种子胚根生长速度呈先慢后快趋势。可见,凋落物是影响格氏栲种子萌发及胚根生长的重要因素,主要通过阻碍种子与土壤接触而抑制萌发,影响格氏栲林更新。  相似文献
