In ectomycorrhizae auxins are proposed to attenuate elicitor-induced defence reactions in the host plant. To examine this hypothesis we compared the elicitor-induced accumulation of peroxidase isoforms between suspension-cultured spruce (Picea abies[L.] Karst.) cells incubated in media with and without auxins. In spruce cells changes in ionically and covalently wall-bound as well as symplasmic peroxidase (EC 1.11.1.7) activities were observed when elicitors from the following fungal species were applied: (1) Hebeloma crustuliniforme, an ectomycorrhizal partner of spruce; (2) Suillus variegatus, an ectomycorrhizal fungus incompatible with spruce; (3) Heterobasidion annosum, a spruce pathogen. Activity staining after SDS-PAGE and western blotting showed an accumulation of an ionically wall-bound 38-kDa peroxidase isoform. In addition, two covalently wall-bound isoforms (34 and 53 kDa) that could be released from spruce cell walls by cellulase and pectinase treatment were also induced by elicitors from these fungi. Moreover, in cells cultured without auxins all the elicitors triggered a rapid and transient accumulation of ionically wall-bound peroxidases, which reached a maximum activity 48 h after elicitor application. This early and transient peroxidase accumulation was diminished and delayed in cells cultured in the presence of auxins. In contrast, activity of peroxidases released into the culture medium of spruce cells or into the medium of protoplasts was suppressed by the elicitors of Hebeloma crustuliniforme. However, this suppression was attenuated by the action of auxins. It is suggested that under natural conditions, in infected spruce roots, the elicitors of the compatible fungus cause both suppression of the peroxidase (which is secreted to the free space of the roots), and induction of wall-bound and symplasmic peroxidases. On the other hand, auxins synthesized by the fungus could weaken these different elicitor-mediated effects.  相似文献

Root cultures ofAngelica gigas Nakai were found to be sensitive to elicitation by polysaccharide elicitors, such as methyl-β-cyclodextrin, glucan, carboxymethyl-β-chitin, chitosan, yeast extract and pectin. For the production of decursinol angelate, carboxymethyl-β-chitin and glucan were found to be the most efficient elicitors. The enhanced accumulation of decursinol angelate was proportional to the increase of the phenylalanine ammonia-lyase (PAL) activity after the treatment with most of the elicitors. However, carboxymethyl-β-chitin treatment did not stimulate the PAL activity, despite the 1.6-fold increase in the decursinol angelate production.  相似文献

In suspension-cultured cells of tomato (Lycopersicon esculentum Mill.), the activity of 1-aminocyclopropane-1-carboxylate synthase (ACC-S) rapidly increases in response to fungal elicitors. The effect of inhibitors of protein kinases and protein phosphatases on the regulation of ACC-S was studied. K-252a, an inhibitor of protein kinases, prevented induction of the enzyme by elicitors and promoted its apparent turnover in elicitor-stimulated cells, causing a 50% loss of activity within 4 to 8 min in both the presence and absence of cycloheximide. Calyculin A, an inhibitor of protein phosphatases, caused a rapid increase of ACC-S in the absence of elicitors and an immediate acceleration of the rate of ACC-S increase in elicitor-stimulated cells. In the presence of cycloheximide there was no such increase, indicating that the effect depended on protein synthesis. Cordycepin, an inhibitor of mRNA synthesis, did not prevent the elicitor-induced increase in ACC-S activity but strongly reduced the K-252a-induced decay and the calyculin A-induced increase of its activity. In vitro, ACC-S activity was not affected by K-252a and calyculin A or by treatments with protein phosphatases. These results suggest that protein phosphorylation/dephosphorylation is involved in the regulation of ACC-S, not by regulating the catalytic activity itself but by controlling the rate of turnover of the enzyme.  相似文献

Much work has elucidated the pathways and mechanisms involved in the production of insect immune effector systems. However, the temporal nature of these responses with respect to different immune insults is less well understood. This study investigated the magnitude and temporal variation in phenoloxidase and antimicrobial activity in the mealworm beetle Tenebrio molitor in response to a number of different synthetic and real immune elicitors. We found that antimicrobial activity in haemolymph increased rapidly during the first 48h after a challenge and was maintained at high levels for at least 14 days. There was no difference in the magnitude of responses to live or dead Escherichia coli or Bacillus subtilis. While peptidoglylcan also elicited a long-lasting antimicrobial response, the response to LPS was short lived. There was no long-lasting upregulation of phenoloxidase activity, suggesting that this immune effector system is not involved in the management of microbial infections over a long time scale.  相似文献

Elicitor preparations containing the avr5 gene products from races 4 and 2.3 of Cladosporium fulvum, and tomato (Lycopersicon esculentum L.) cells containing the resistance gene Cf5 were used to investigate the involvement of redox processes in the production of active oxygen species associated with the plant response to the fungal elicitors. Here we demonstrate that certain race-specific elicitors of C. fulvum induced an increase in ferricyanide reduction in enriched plasma membrane fractions of tomato cells. The addition of elicitors to plasma membranes also induced increases in NADH oxidase and NADH-dependent cytochrome c reductase activities, whereas ascorbate peroxidase activity was decreased. These results suggest that changes in the host plasma membrane redox processes, transferring electrons from reducing agents to oxygen, could be involved in the increased production of active oxygen species by the race-specific elicitors. Our results also show that the dephosphorylation of enzymes involved in redox reactions is responsible for the race-specific induced redox activity. The effects of guanidine nucleotide analogs and mastoparan on the activation of plasma membrane redox reactions support the role of GTP-binding proteins in the transduction of signals leading to the activation of the defense response mechanisms of tomato against fungal pathogens.  相似文献

Depolarization of tomato leaf cells by oligogalacturonide elicitors   总被引:5,自引:0,他引:5  
The electrical potential difference (Em) across the plasma membrane of tomato leaf mesophyll cells consists of a cyanide-sensitive component, presumably produced by an H+-ATPase, and a cyanide-insensitive component. Variation of Em between different batches of tissue is mainly caused by variation in the cyanide-sensitive component. Oligogalacturonide elicitors that induce the synthesis of proteinase inhibitors in tomato seedlings depolarize the Em of tomato leaf mesophyll cells. This depolarization closely resembles that caused by cyanide: they are of similar magnitude and vary in a similar manner with variation in the initial Em of different batches of tissue. Treatments with cyanide and with the elicitors have similar effects on the small depolarization caused by KCl at 10 mol m?3. The results suggest that the elicitors depolarize Em by inhibiting the plasma membrane H+-ATPase, but that the detailed mechanism of inhibition by the elicitors is different from that caused by cyanide.  相似文献

Cerebrosides A and C, compounds categorized as glycosphingolipids, were isolated in our previous study from the rice blast fungus (Magnaporthe grisea) as novel elicitors which induce the synthesis of rice phytoalexins. In this paper, these cerebroside elicitors showed phytoalexin-inducing activity when applied to plants by spray treatment and also induced the expression of pathogenesis-related (PR) proteins in rice leaves. This elicitor activity of the cerebrosides showed the structural specificity as that for the induction of phytoalexins. Ceramides prepared from the cerebrosides by removal of glucose also showed the elicitor activity even in lower level compared to the cerebrosides. In field experiments, the cerebroside elicitors effectively protected rice plants against the rice blast fungus, an economically devastating agent of disease of rice in Japan. The cerebrosides elicitors protected rice plants from other disease as well and were found to occur in a wide range of different phytopathogens, indicating that cerebrosides function as general elicitors in a wide variety of rice-pathogen interactions.  相似文献

We previously reported elevated cytosolic calcium levels in tobacco cells in response to elicitors [D. Lecourieux, C. Mazars, N. Pauly, R. Ranjeva, A. Pugin, Analysis and effects of cytosolic free calcium elevations in response to elicitors in Nicotiana plumbaginifolia cells, Plant Cell 14 (2002) 2627-2641]. These data suggested that in response to elicitors, Ca2+, as a second messenger, was involved in both systemic acquired resistance (RSA) and/or hypersensitive response (HR) depending on calcium signature. Here, we used transformed tobacco cells with apoaequorin expressed in the nucleus to monitor changes in free nuclear calcium concentrations ([Ca2+](nuc)) in response to elicitors. Two types of elicitors are compared: proteins leading to necrosis including four elicitins and harpin, and non-necrotic elicitors including flagellin (flg22) and two oligosaccharidic elicitors, namely the oligogalacturonides (OGs) and the beta-1,3-glucan laminarin. Our data indicate that the proteinaceous elicitors induced a pronounced and sustainable [Ca2+](nuc) elevation, relative to the small effects of oligosaccharidic elicitors. This [Ca2+](nuc) elevation, which seems insufficient to induce cell death, is unlikely to result directly from the diffusion of calcium from the cytosol. The [Ca2+](nuc) rise depends on free cytosolic calcium, IP3, and active oxygen species (AOS) but is independent of nitric oxide.  相似文献

Suspension cultures of Habanero pepper (Capsicum chinense Jacq.) were exposed to salicylic acid or methyl jasmonate to change secondary metabolism. Both treatments led to the accumulation of capsaicinoids and their late biosynthetic intermediate, vanillin. Both elicitors had a positive effect on the activities of phenylalanine ammonia lyase and coumarate O-methyltransferase, but none of them represented the main limiting step for capsaicinoid accumulation since vanillin contents were two orders of magnitude higher than those of capsaicinoids.  相似文献

Rapid reactions comprising efflux of K+ and Cl, phosphorylation of a 63-kDa protein (pp63), extracellular alkalinization and synthesis of H2O2 are equally induced in cells of Picea abies (L.) Karst. by chitotetraose, colloidal chitin and cell wall elicitors from the ectomycorrhizal fungus Hebeloma crustuliniforme (Bull. ex Fries.) Quél. an ectomycorrhizal partner of spruce. Cleavage of fungal cell wall elicitors and of artificial chitin elicitors to monomeric and dimeric fragments by apoplasmic spruce chitinases (36-kDa class I chitinase, pI 8.0, and 28-kDa chitinase, pI 8.7; EC 3.2.1.14) equally prevented induction of these rapid reactions. Also, N-acetylglucosamine oligomers and elicitors from the fungal cell walls showed a similar dependence of their activity on the degree of polymerisation. From these results it is suggested that, during ectomycorrhiza formation, only some of the chitin-derived elicitors reach their receptors at the plant plasma membrane, initiating reactions of the hypersensitive response in the host cells. The remaining fungal elicitors will be degraded to varying extents by wall-localized chitinases of the host root, reducing the defence reactions of the plant and allowing symbiotic interactions of both organisms. Plant cells respond to elicitors by inducing a variety of defense responses. Some of these reactions are dependent on the activity of protein kinases. Recently, mitogen-activated protein kinases (MAPKs) have been identified to be activated by fungal and bacterial elicitors as well as by pathogen infection. In gel kinase assays of alfalfa cells treated with yeast cell wall-derived elicitor (YE) revealed that 44- and 46-kDa MAPKs are rapidly and transiently activated. Immunokinase assays with specific MAPK antibodies revealed that YE mainly activated the 46-kDa SIMK and the 44-kDa MMK3 and to a lesser extent the 44-kDa MMK2 and SAMK. When cells were treated with chemically defined elicitors potentially contained in the YE (chitin and N-acetylglucosamine oligomers, beta-glucan, and ergosterol), the four MAPKs were found to be activated to different levels and with different kinetics. Whereas SIMK and SAMK have been found to be activated by a number of diverse stimuli, MMK3 is activated during mitosis and was therefore assumed to participate in cell division (). No physiological process could be associated with MMK2 activity so far. This is the first report that MMK2 and MMK3 can be activated by external stimuli. Overall, our findings indicate that plant cells can sense different cues of a given microorganism through the activation of multiple MAPKs.  相似文献

Plant response to pathogens involves an intricate network of signal transduction pathways. Here, potato cell cultures were used to study signal transduction in response to elicitors from Phytophthora infestans. Pretreatment of cells with Ser/Thr protein kinase inhibitors, EGTA, calmodulin antagonists or a channel blocker abolished the induction of two enzymes involved in defence responses, phenylalanine ammonia‐lyase (PAL) and peroxidase. Phosphatase inhibitors caused an increase of these activities in the absence of elicitors. Hyphal cell wall components (HWC) from an incompatible race (HWC 0) produced a rapid and transient increment of histone phosphorylation, whereas induction by HWC from a compatible race (HWC C) was less pronounced and more sustained. As activities were calcium‐dependent, a fraction enriched in calcium‐dependent protein kinases (CDPKs) was obtained by DEAE chromatography. Fractions from HWC 0‐ and HWC C‐treated cells presented higher kinase activity than that from untreated cells. Moreover, total activity was higher in the incompatible than in the compatible interaction. Activity was calcium‐dependent, partially inhibited by calmodulin antagonists and able to phosphorylate syntide‐2, a specific substrate of CDPKs. An in‐gel kinase assay showed the presence of a band of approximately 50 kDa whose activity was higher in HWC 0‐ than in HWC C‐treated cells and was not detected in control extracts. This report presents evidences of the differential activation of CDPKs in response to elicitors from different races of P. infestans, revealing that these protein kinases participate in the defence response to oomycete.  相似文献

报道了不同真菌诱导子对悬浮培养的西洋参(Panaxquinquefolium)细胞生长、皂甙和多糖合成,以及细胞内和培养液中过氧化物酶活性的生理效应。悬浮培养的西洋参细胞经刺盘孢菌(Colletotrichumnicoltianae)丝体诱导子处理后,总皂甙产率可由对照的296mg/L增加到679mg/L(约占细胞干重的(16.3%),比对照提高约1.3倍,而且总皂甙的85%排放在培养液中;经黑曲霉(Aspergillusnigran)诱导子处理后,细胞多糖含量可达到11.79%(细胞干重),比对照增加1倍多。初步纯化的刺盘孢菌丝体诱导子和尖孢镰刀菌(Fusuriumoxysporum)滤液诱导子在诱导处理前期能明显促进西洋参细胞生长,同时细胞内及培养液中过氧化物酶活性显著增加;随时间延长,细胞生长和酶活性逐步受到抑制。  相似文献

Abstract. Two size ranges of oligosaccharide elicitors of pectic origin have been investigated for their effects on tomato plants. Both size ranges, with degrees of polymerization of 1–7 and 10–20 respectively, induced the accumulation of proteinase inhibitor (PI) activity in excised plants, and also induced changes in membrane potential of leaf mesophyll cells. The depolarizations were substantial, rapid, and reversible on removal of the elicitors. The effects are discussed in the context of early events in the signal transduction pathway linking oligosaccharides to changes in PI gene expression.  相似文献

The effects of yeast extract and selected polysaccharide elicitors on secondary metabolite production, particularly of anthocyanin and phenolic acid, in cell suspension cultures of Vitis vinifera were investigated. All elicitors either maintained or promoted cell growth in culture. Overall, secondary metabolite production in V. vinifera cell suspension cultures responded differently to different elicitors. Chitosan, pectin, and alginate enhanced production of anthocyanin within 13 days of culture with levels of 2.5-, 2.5-, and 2.6-fold increase, respectively, over that of control. Chitosan, alginate, and gum arabic significantly promoted accumulation of phenolic acids, particularly 3-O-glucosyl-resveratrol, in V. vinifera cultures, as well as in the culture medium. Intracellular phenolic acid production was significantly enhanced by alginate and chitosan, with 1.7- and 1.5-fold levels, respectively, of that of control. Extracellular phenolic acid production was also significantly increased in the presence of chitosan and gum arabic, with levels of 3.3- and 1.7-fold higher, respectively, than those of control. In addition, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity was enhanced in the presence of elicitors, and this was positively correlated with increased accumulation of anthocyanin in V. vinifera cell suspension cultures.  相似文献

The dynamics of induced accumulation of callose in tomato cells is studied. Localization of callose in the cells is studied by methods of fluorescence microscopy and the time needed for quantitative determination of callosa is optimized. The quantity of callose in tomato cells treated with different biotic elicitors is established. A nonlinear dependence of the deposition of callose on the concentration of chitin oligomers containing 3–5 fragments of N-acetylglucosamine is established and a proportional increase in the content of callose in the cells with increasing concentration of chitin dimer and chitosan in the culture medium is demonstrated.  相似文献

Biogenic elicitors (chitosan and its complex with salicylic acid) and an immunosuppressor (laminarin) were shown to increase the activity of L-phenylalanine ammonia lyase (EC 4.3.1.5) and protein synthesis in potato tubers. Laminarin did not decrease L-phenylalanine ammonia lyase activity.
