This study analyzed five varieties of P. tomentosa and their putative parents, P. alba, P. davidiana, P. adenopoda and P. tremula by RAPD and artificial cross. Of 78 bands revealed by 13 arbitrary primers selected, 58 (74.36%) bands were polymorphic among P. tomentosa and four putative parents. The RAPD statistical results showed the relationship between P. tomentosa and P. alba or P. adenopoda was closer than that between P. tomentosa and P. davidiana or P. tremula . There were higher similarity and lower genetic distance between P. tomentosa and P. alba or P. adenopoda than that between P. tomentosa and the other two putative parents. The results from UPGMA, Fitch margoliash phylogenetic dendrogram were similar. In addition, artificial hybrids including reciprocal cross between P. alba and P. adenopoda were used to compare with five varieties of P. tomentosa . And those hybrids between P. alba as female and P. adenopoda as male were closer to P. tomentosa than those from reciprocal cross. The results demonstrated that P. tomentosa was a natural hybrid between P. alba as female parent and P. adenopoda as male parent.  相似文献

把生长在全盐含量为 0 .361%的土壤中的毛白杨 ( Populus tomentosa Carr.)愈伤组织悬浮细胞培养在含不同浓度 Na Cl的液体培养基中 ,定期测量各组的细胞密度 ,根据生长曲线研究其耐盐性。结果显示 ,与不加 Na Cl的液体培养基相比 ,生长于含较低浓度 Na Cl( 2 .7‰和 5.5‰ )液体培养基中的细胞生长较好 ,而在含较高浓度 Na Cl( 8.2‰和 10 .9‰ )的液体培养基中 ,细胞生长受到较强抑制。  相似文献

Changes of abscisie acid (ABA) and gibberellin (GA3) contents during the release of dormancy in winter bud of Populus tomentosa Carr. were determinedwith GC. After leaf fall in autumn, content of ABA in the bud was 888.0 μg/kg. fr.wt. Obvious decrease in ABA content was observed during the bud released from dormancy. The bud kept in room temperature opened about two months earlier thanthat under natural condition; and the rate of decrease of ABA content in these budwas also more rapid. The ABA contents of buds with similar outer appearance werecompared, either the bud from outside under natural condition or under room temperature, they were similar, although time of their occurrence was quite different, withalmost a difference of about two months. From this fact it has been assumed that thereis a close relationship between the release of dormancy and the decrease in ABA content. Another fact was noticed before Dec. 6, no GA3 could be detected in the dormantbud. From Jan. 9 and thereafter, GA3 content increased gradually and reached itsmaximum (20 μg/kg. fr.wt) by Mar. 19, the bud was inflated. GA3 decreased again as the bud was opened, it seems to be that the process of releasing dormancy in bud mightbe promoted by GA3.  相似文献

以毛白杨基因型TC152无菌苗为材料,研究毛白杨悬浮细胞系建立与植株再生,结果表明,通过悬浮培养和固体培养两种方法诱导毛白杨悬浮细胞分化不定芽,最终获得无菌生根苗。愈伤组织在MS+1.5mg·L-12,4-D+30g·L-1蔗糖的液体培养基中振荡培养,12d可建立悬浮细胞系;悬浮细胞系继代培养基为MS+0.8mg·L-12,4-D+30g·L-1蔗糖,继代周期为7d,悬浮细胞在MS+1.0mg·L-16-BA+0.1mg·L-1NAA+0.5~1.0mg·L-1ZT+30g·L-1蔗糖培养基中悬浮培养,可分化大量不定芽,每个培养瓶中可得到40~50个芽,个别不定芽玻璃化;不定芽在1/2MS+0.6mg·L-1IBA+20g·L-1蔗糖+5.5g·L-1琼脂培养基上可分化不定根。悬浮细胞通过固体平板培养增殖为愈伤组织块后,在MS+1.0mg·L-16-BA+0.1mg·L-1NAA+1.0mg·L-1ZT+30g·L-1蔗糖+5g·L-1琼脂的固体培养基上,不定芽分化率可达到70.00%。  相似文献

外源突变基因AP1M2转化毛白杨的研究   总被引:1,自引:0,他引:1  
为探究毛白杨开花调控分子机制并获得不飞絮毛白杨株系,利用农杆菌介导法将显性负突变结构基因爿Pm挖转入毛白杨中,经PCR检测,共获得阳性转化植株7株。通过RT_qPCR检测转基因植株中外源基因AP1M2表达量,发现各株系间的表达水平差异显著,最高为内参的5.41倍,最低为1.89倍。对转基因毛白杨中内源刚尸,和其他开花关键基因的表达量进行检测,发现内源AP1的表达受到抑制,表达量明显下调;其他开花关键基LFY、AP3、PI、SEP3和FTI等的表达量也有不同程度的降低,而FT2表达量并没有明显变化。这些研究结果表明转4尸m扭毛白杨中API、LFY、AP3、PI、SEP3和FT2的表达受到明显抑制.对毛白杨花发育将有一定影响。  相似文献

植物脂肪酸合成的主要部位是叶绿体,叶绿体向外运输脂肪酸的种类和数量受到乙酰-乙酰载体蛋白硫脂酶(FATB)控制。FATB基因在植物生长过程起着非常关键的作用。本研究以毛白杨为材料,将生物信息学知识和分子生物学手段相结合,首先利用现有的杨树基因组EST序列库资源,通过同源序列搜索,经过多次拼接合并获得了理论的杨树脂肪酸去饱和酶基因PtFATB序列全长,利用RT-PCR手段成功克隆得到了毛白杨FATB基因全长编码序列cDNA,该cDNA全长1,450bp,包括起始密码子ATG和144bp的5′末端非编码区,终止密码子TGA和40bp的3′末端非编码区,开放阅读框编码421个氨基酸。通过RT-PCR半定量研究了PtFATB在叶片组织中的表达量最高,茎、根中的表达量依次降低。在低温、干旱、NaCl、ABA四种条件下诱导生长24h,只有在低温的条件下发现PtFATB表达量略微降低,其他几种情况未有变化,该结果表明PtFATB呈组成型表达。上述结果为植物脂肪酸的基因工程提供了基础。  相似文献

Cell wall structure of wheat coleoptiles grown under continuous hypergravity (300 g) conditions was investigated. Length of coleoptiles exposed to hypergravity for 2-4 days from germination stage was 60-70% of that of 1 g control. The amounts of cell wall polysaccharides substantially increased during the incubation period both in 1 g control and hypergravity-treated coleoptiles. As a results, the levels of cell wall polysaccharides per unit length of coleoptile, which mean the thickness of cell walls, largely increased under hypergravity conditions. The major sugar components of the hemicellulose fraction, a polymer fraction extracted from cell walls with strong alkali, were arabinose (Ara), xylose (Xyl) and glucose (Glc). The molar ratios of Ara and Xyl to Glc in hypergravity-treated coleoptiles were higher than those in control coleoptiles. Furthermore, the fractionation of hemicellulosic polymers into the neutral and acidic polymers by the anion-exchange column showed that the levels of acidic polymers in cell walls of hypergravity-treated coleoptiles were higher than those of control coleoptiles. These results suggest that hypergravity stimuli bias the synthesis of hemicellulosic polysaccharides and increase the proportion of acidic polymers, such as arabinoxylans, in cell walls of wheat coleoptiles. These structural changes in cell walls may contribute to plant resistance to hypergravity stimuli.  相似文献

Background and Aims In temperate regions, trees undergo annual cycles of cambial growth, with periods of cambial activity and dormancy. Environmental factors might regulate the cambial growth, as well as the development of cambial derivatives. We investigated the effects of low temperature by localized cooling on cambial activity and latewood formation in two conifers, Chamaecyparis obtusa and Cryptomeria japonica.Methods A plastic rubber tube that contained cooled water was wrapped around a 30-cm-wide portion of the main stem of Chamaecyparis obtusa and Cryptomeria japonica trees during seasons of active cambium. Small blocks were collected from both cooled and non-cooled control portions of the stems for sequential observations of cambial activity and for anatomical measurements of cell morphology by light microscopy and image analysis.Key Results The effect of localized cooling was first observed on differentiating tracheids. Tracheids narrow in diameter and with significantly decreased cambial activity were evident 5 weeks after the start of cooling in these stems. Eight weeks after the start of cooling, tracheids with clearly diminished diameters and thickened cell walls were observed in these stems. Thus, localized low temperature induced narrow diameters and obvious thickening of secondary cell walls of tracheids, which were identified as latewood tracheids. Two months after the cessation of cooling, a false annual ring was observed and cambium became active again and produced new tracheids. In Cryptomeria japonica, cambial activity ceased earlier in locally cooled portions of stems than in non-cooled stems, indicating that the cambium had entered dormancy sooner in the cooled stems.Conclusions Artificial cooling of stems induced latewood formation and cessation of cambial activity, indicating that cambium and its derivatives can respond directly to changes in temperature. A decrease in the temperature of the stem is a critical factor in the control of cambial activity and xylem differentiation in trees.  相似文献

Background and Aims

Cambium reactivation after dormancy and budbreak in deciduous trees requires a supply of mobilized reserve materials. The pathway and mode of transfer of these materials are poorly understood.

Methods

Transport of reserve materials during cambium reactivation in Populus nigra was investigated by conventional and immunocytochemical TEM analyses, SDS–PAGE, western blotting and intracellular microinjection of fluorescent dyes.

Key Results

Proteinaceous compounds stored in vacuoles and protein bodies of vascular cells and ray cells disappeared within 3 weeks after cambial reactivation and budbreak. Some of these proteins (32 kDa, 30 kDa and 15 kDa) were labelled by lectin antibodies in SDS–PAGE. The same antibodies were localized to plasmodesmata (PDs) between phloem parenchyma, ray cells and fusiform cambial cells. In addition, proteinaceous particles were localized inside the cytoplasmic sleeves of these PDs during budbreak. During this period, the functional diameter of PDs was about 2·2 nm which corresponds approximately to the Stokes'' radius of the detected 15-kDa protein.

Conclusions

Lectin-like reserve proteins or their degradation products seem to be transferred through PDs of phloem parenchyma and rays during cambial reactivation and budbreak. PD transfer of storage proteins is a novelty which supports the concept of symplasmic nutrient supply to the cambial region.  相似文献

The effect of gibberellic acid (GA) and naphthylacetic acid (NAA) on hypocotyl elongation and cell wall polysaccharides was studied using Phaseolus vulgaris seedlings grown in light condition. The hypocotyl was demarcated into two segments — one near the root was called lower and the one near the cotyledon was called upper. The upper segment showed a typical sigmoidal growth curve while lower segment did not show any growth at all. GA promoted the growth of upper segment while NAA showed clear inhibition in both the segments. Xyloglucan content showed a clear inverse correlation with growth. Pectic polysaccharides did not show a clear trend, though showed an initial inverse correlation with growth. It is concluded that degradation of low and high molecular weight xyloglucans are involved in cell wall loosening which in turn may be responsible for the elongation growth of Phaseolus hypocotyls in light.  相似文献

Background and Aims

Alterations of plasmodesma (PD) connectivity are likely to be very important for plant development. Here, the repetitive division pattern of cambial initials in Populus nigra ‘italica’ was studied to follow the development of the PD network during maturation. Furthermore, seasonal changes were investigated in order to trace indications for developmental and functional adaptations.

Methods

Cambium samples of P. nigra twigs, collected in summer, autumn and spring, were chemically fixed for transmission electron microscopy. The parameters, PD density (number of PDs per square micrometre cell-wall area) and PD frequency (total number of PDs per average cell-wall area), were determined for radial and tangential cell interfaces deposited in chronological order.

Key Results

Data sets, presented in plasmodesmograms, show a strong variability in the PD network throughout the year. In summer, high PD numbers occur at the division wall which, after PD doubling by longitudinal fission, decline with further development both at the xylem and the phloem side. In autumn, the number of PDs at the division wall is low as they are in subsequent tangential interfaces. In spring, the first cell division coincides with a massive increase in PD numbers, in particular at the division wall. Only the radial walls between initials maintain their PD equipment throughout the year. This feature can be exploited for identification of the initial layer.

Conclusions

PD networks in the cambium go through a strict developmental programme depending on the season, which is associated with changing functional requirements. For instance, PD numbers correlate with proliferative activity and potential pathways for intercellular signalling. Increases in PD numbers are ascribed to longitudinal fission as a major mechanism, whereas the decline in older derivatives is ascribed to PD degradation.  相似文献

This study aims to establish an efficient pretreatment process using cellulose-dissolution solvents to enhance the enzymatic saccharification. LiOH/urea, LiCl/DMAc, concentrated phosphoric acid, ionic liquid (1-butyl-3-methylimidazolium chloride; [BMIM]Cl) and N-methyl-morpholine-N-oxide (NMMO) were selected as the cellulose dissolution agents. Except the cellulosic sample regenerated from LiCl/DMAc system, all the other treated samples exhibited lower cellulose crystallinity and degree of polymerization (DP), and consequently, exhibited a significant enhancement on enzymatic hydrolysis kinetic. Ionic liquid pretreatment offered unique advantages in the hydrolysis rate in the first 10 h, probably due to the extensively structural transformation of cellulose from the crystalline to the amorphous region. Meanwhile, the regenerated cellulose from concentrated phosphoric acid almost completely consisted of cellulose II, and achieved the highest saccharification yield.  相似文献

Gene-derived simple sequence repeats (genic SSRs), also known as functional markers, are often preferred over random genomic markers because they represent variation in gene coding and/or regulatory regions. We characterized 544 genic SSR loci derived from 138 candidate genes involved in wood formation, distributed throughout the genome of Populus tomentosa, a key ecological and cultivated wood production species. Of these SSRs, three-quarters were located in the promoter or intron regions, and dinucleotide (59.7%) and trinucleotide repeat motifs (26.5%) predominated. By screening 15 wild P. tomentosa ecotypes, we identified 188 polymorphic genic SSRs with 861 alleles, 2–7 alleles for each marker. Transferability analysis of 30 random genic SSRs, testing whether these SSRs work in 26 genotypes of five genus Populus sections (outgroup, Salix matsudana), showed that 72% of the SSRs could be amplified in Turanga and 100% could be amplified in Leuce. Based on genotyping of these 26 genotypes, a neighbour-joining analysis showed the expected six phylogenetic groupings. In silico analysis of SSR variation in 220 sequences that are homologous between P. tomentosa and Populus trichocarpa suggested that genic SSR variations between relatives were predominantly affected by repeat motif variations or flanking sequence mutations. Inheritance tests and single-marker associations demonstrated the power of genic SSRs in family-based linkage mapping and candidate gene-based association studies, as well as marker-assisted selection and comparative genomic studies of P. tomentosa and related species.  相似文献

The spores of Anemia tomentosa var. anthriscifolia and A. tomentosa var. tomentosa were studied focusing the attention on their abnormalities. The study was based on fresh and herbarium material and the spores were examined with light, scanning and transmission electron microscopy. Normal, abnormal and abortive spores were observed in both taxa. The normal spores were trilete, triangular in polar view, and the ornamentation consisted of parallel ridges separated by narrow and smooth grooves. The spores were observed in monads, dyads, triads and tetrads. The abnormal spores were monolete, trilete, tetralete or alete with great variations in size. In fact, some spores were almost double the size of the normal ones. Some differences were also found in the ornamentation of the spores. Aborted and not completely developed spores were also observed in the specimens. The wall ultrastructure of the taxa was studied for the first time. The exospore was two-layered with numerous cavities inside its structure, and the perispore was also two-layered. The results revealed that the sporoderm ultrastructure of both normal and abnormal spores of the taxa analyzed was very similar.  相似文献

基于1982—2006年内蒙古地区17个小白杨物候观测站的气温以及小白杨春季和秋季物候资料,对该区小白杨春季和秋季物候的变化趋势进行了研究,分析了物候期变化与气候变暖的关系,并利用逐步回归方法建立了小白杨始花期和落叶期变化的预估模型.结果表明:1982—2006年间,研究区气温呈增加趋势,该区小白杨春季物候呈提前趋势,秋季物候呈延后趋势;小白杨始花期与冬、春季气温呈负相关,主要影响因子为3—4月的平均气温;落叶期与秋季气温呈正相关,主要影响因子为8—10月的平均气温;根据气候变化国家评估报告,未来内蒙古地区春、秋季平均气温分别升高1.5 ℃~5.4 ℃和1.2 ℃~4.4 ℃,小白杨始花期将提前3.9~17.8 d,落叶期将延后3.0~12.4 d.  相似文献
