The results of a systematic attempt to relate the growth-stimulatingfunction of compounds analogous to ethylene-bis-nitrourethaneto their chemical structure and properties showed that, withthe exception of a series of nitraminocarboxylic acids, NO2NH(CH2)COOH,activity was confined to compounds of the form R—N(NO2)—X—N(NO2)—R,where X is an alkylene or substituted alkylene chain. It issuggested that active compounds are converted to dinitramines(R = H) in the plant, and that the acidic nature of the —NHNO2groups is of importance. The oil/water partition coefficientsand the geometrical structure of the molecules are also shownto influence their growth-stimulating activity.  相似文献

In soyabean [Glycine max (L.) Merrill] the period between sowingand flowering is comprised of three successive developmentalphases—pre-inductive, inductive and post-inductive—inwhich the rate of development is affected, respectively, bytemperature only, by photoperiod and temperature, and then againby temperature only. A reciprocal-transfer experiment (carriedout at a mean temperature of 25°C) in which cohorts of plantswere transferred successively between short and long photoperiodsand vice-versa showed that eight combinations of three pairsof maturity alleles (E1/e1, E2 /e2, E3 /e3) had their greatesteffect on the duration of the inductive phase in long days.This phase was increased with the increasing photoperiod sensitivityinduced by the different gene combinations, and ranged fromabout 27 to 54 d according to genotype. In a short day regime(11·5 h d-1), less than the critical photoperiod, theduration of the inductive phase was brief—requiring about11 photoperiodic cycles in the less photoperiod-sensitive genotypesand only about seven cycles in the more sensitive ones. Thematurity genes also affected the duration of the two photoperiod-insensitivephases; these durations were positively correlated with thephotoperiod-sensitivity potential of the gene combinations.The largest effect was on the pre-inductive phase which variedfrom 3 to 11 d, while the post-inductive phase varied from about13 to 18 d. As a consequence of these non-photoperiodic effectsof the maturity genes, even in the most inductive regimes (daylengthsless than the critical photoperiod) the time taken to flowerby the less photoperiod-sensitive combinations of maturity geneswas somewhat less than in the more sensitive combinations—rangingfrom about 28 to 34 d. NG, a strain of cultured tobacco cells of Nicotiana glutinosahad high growth rates and carboxylate contents (C—A) of100 to 130 meq/100 g of dry cells on media containing 42 meqNO3/liter as the sole N source. (C—A) is the amount ofinorganic cations minus inorganic anions in meq per 100 g ofdry cells. NG, cultured on media containing NH4 10+NO3 42 in meq/liter,had lower growth rates and lower (C—A) values as comparedwith NG on media containing NO3 as the sole N source. NG, cultured on media containing NH4 30+NO3 42 in meq/liter,had high growth rates and (A—C) values of 22 to 53 meq/100gof dry cells. In this case, the (A—C) content may correspondto organic cations, basic organic N compounds such as free asprotein-bound basic amino acids. The easily absorbed Cl mayhave been required maintain good growth conditions such as ionicbalance and a favorable pH in the cells. Thus cultured cells of Nicotiana glutinosa may have physiologicaladaptability against variations in a relatively wide range of|C—A| contents [|C—A| being the absolute valuesof (C—A)]. A method is described for the assay of the uptake of amino-acidsfrom aqueous solution by fungus mycelia. The rate of uptakein Botrytis fabae was found to vary with the age of the fungus,the pH, and the concentration of the amino-acid. Both L- andD-isomers were accumulated, but the system had a greater affinityfor the L-isomer. Unsubstituted —NH2, and —COOHgroups were necessary for the process, which was constitutiveand inhibited by uncouplers of oxidative phosphorylation. Competitionexperiments with several amino-acids suggest that one uptakeprocess is common to all. Although both peptides and amineswere accumulated they did not compete effectively with amino-acidsfor uptake.  相似文献

Gibberellins A1 to A9 have been compared, each at several doselevels, in bioassays based on extension of stems of dwarf gardenpea (Pisum sativum), dwarf bean (Phaseolus vulgaris) and Lunariaannua, of hypocotyls of cucumber (Cucumis sativus) and lettuce(Lactuca sativa), and of leaf sheaths of three dwarf mutants(d–1, d–3, d–5) of maize (Zea mays). GibberellinsA3 (gibberellic acid) and A7 are of high potency in most bioassays.A8 is of negligible potency in all and is probably not a functionalhormone. The other gibberellins show a more or less marked tendencyto specificity. The plants used as bioassay material also differin the specificity of their response. Some, for example, maizedwarfs d–3 and d–5 and lettuce, respond well tomost gibberellins; others, for example, cucumber, respond onlyto a few; extreme specificity is shown by Lunaria annua which,in the unvernalized condition, responds by stem elongation onlyto gibberellin A7. Dose/response curves of the various gibberellinsare usually parallel, but certain exceptions to this have beenfound. Possible explanations of specificity are discussed inrelation to the results obtained, and it is concluded that insufficientevidence is available to make it possible to draw any validconclusions. Current definitions of gibberellins, whether basedon chemical structure or biological activity, are unsatisfactory.  相似文献

The extent to which photorespiration occurs in CrassulaceanAcid Metabolism (CAM) plants has received limited attention.No comparative studies of C3 and CAM development have been made.To address this problem, activities of several photorespiratoryenzymes were measured in a facultative CAM plant—Mesembryanthemumcrystallinum L.—during induction of CAM by water stress(NaCl-treatment). Salt-treatment over a 22 d period produceda progressive change in metabolism from C3 to CAM. This wasconfirmed by (I) changes in gas exchange from C3 fixation tothe characteristic CAM pattern of nocturnal CO2 uptake; (2)increases in did acid fluctuation and (3) a 30-fold increasein phosphoenol pyruvate (PEP) carboxylase activity. In contrast,no significant changes were observed in the activities of glycollateoxidase (GO), NAD-dependent-hydroxypyruvate reductase (HPR),glutamine synthetase (GS) or glutamate dehydrogenase (GDH) whenplants were induced into CAM. Ion exchange chromatography onDEAE Sephacel detected only one GS isoenzyme (GS2, chloroplastic)at all stages of CAM induction, Western blot analysis, however,detected an additional, although minor, band of GS1 (cytosolic),in C3 plants, which disappeared following CAM induction. Ourresults show that, after development of CAM, these plants stillretain the capacity to photorespire. This may be an essentialrequirement of CAM plants growing in habitats with variablerainfall. When water availability is high, stomata may openin the light allowing ribulose bisphosphate carboxylase oxygenase(Rubisco) activity and photorespiration to occur. Careri, G., Giansanti, A. and Fensom, D. 1986. Dielectnc-gravimetricmeasurements on Fucus vesiculosus and Ascophyllum nodosum duringdehydration.—J. exp. Bot. 37: 375–378. Samples of the marine algae Fucus vesiculosus and Ascophyllumnodosum were dehydrated between plates of a condenser so thatsimultaneous measurements of dielectric properties and rateof water loss could be made. During the desiccation process,both the evaporation rate and the dielectric properties becomezero at a critical hydration Hc, indicating that for H <Hc,the residual water must be in a tightly bound state. The absolutevalues of Hc, depend on the nature of the sample, but they arein the range 0·1–0·3, well above the hydrationlimit for active ion uptake (H=0·02). The kinetic properties of phosphoenolpyruvate (PEP) carboxylasehave been studied among several Flaveria species: the C3 speciesF. cronquistii, the C3–C4 species F. pubescens and F.linearis, and the C4 species F. trinervia. At either pH 7 or8, the maximum activities (in µmol.mg Chl–1.h–1)for F. pubescens and linearis (187–513) were intermediateto those of the C3 species (12–19) and the C4 species(2,182–2,627). The response curves of velocity versusPEP concentration were hyperbolic for the C3 and C3–C4species at either pH 7 or 8 while they were sigmoidal for theC4 species at pH 7 and hyperbolic at pH 8. The Km values forPEP determined from reciprocal plots were lowest in the C3 species,and of intermediate value in the C3–C4 species comparedto the K' values of the C4 species determined from Hill plotsat either pH 7 or 8. Glucose-6-phosphate (G6P) decreased theKm values for PEP at both pH 7 and 8 in the C3 and C3–C4species. In the C4 species, G6P decreased the K' values at pH8 but increased the K' values at pH 7. In all cases, G6P hadits effect by influencing the activity at limiting PEP concentrationswith little or no effect on the maximum activity. At pH 8 andlimiting concentrations of PEP the degree of stimulation ofthe activity by G6P was greatest in the C4 species, intermediatein F. linearis, a C3–C4 species, and lowest in the C3species. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. The metabolism of [14C]GA20 during seed maturation of Phaseolusvulgaris was studied using cell-free systems from embryos rangingin age from 10 DAF (day after flowering) to 24 DAF. Enzyme preparationsfrom very immature seeds actively converted GA20 to GA1, GA5and GA6. The ratio of incubation products suggested the biosyntheticpathway of GA20—GA5—GA6. Fluctuation in the levelsof endogenous C19-GAs, namely GA1, GA4, GA5, GA6, GA8, GA9 andGA20 was analyzed by GC-SIM using internal standards to compareenzyme activity with the levels of endogenous GAs. The introduction and repression of CAM in Sedurn telephiunmL, a temperate succulent, was investigated in watered, progressivelydrouglited and rewatered plants in growth chambers. Measurementswere made of water vapour and CO2 exchange, titratable acidity(TA) and xylem sap tension. Effects of photoperiod were alsostudied. CAM was induced by drought under long or short days,although when watered no CAM activity was expressed. C3-CAM intermediate plants were used for the investigation ofwater supply. Those which had received water and those drought-stressedboth displayed a similar nocturnal increase in TA, with a day-nightmaximum (H+) of 69 µmol g–1 fr. wt. The wateredplants took up CO2 at a maximum rate of 2?2 µmol m–2s–1 only in the light period, while the droughted plantsshowed a maximum nocturnal CO2 uptake rate of 0?69 µmolm–2 s–1. Subsequently, as CAM was repressed, thewatered S. telephiwn displayed little variation in TA, withconstant levels at 42 µmol g–1 fr. wt. (day 10).After 10 d of drought stress, the CAM characteristics of S.telephiurn were aLso affected, with reduced net CO2 uptake andH+. The transition between C3 and CAM in S. telephium can be describedas a progression in terms of the proportion of respiratory CO2which is recycled and refixed at night as malic acid, in comparisonwith net CO2 uptake. Recycling increased from 20% (day 1) to44% (day 10) as a result of the drought stress and was highin both the CAM-C3 stage (no net CO2 uptake at night) and alsoin the drought-stressed CAM stage (reduced net CO2 uptake atnight). The complete C3-CAM transition occurred in less than8 d, and the stages could be characterized by xylem sap tensionmeasurements: CAM = 0?50 MPa C3-CAM = 0?36 MPa C3 = 0?29 MPa. Positive linear relationships were shown between constant temperaturesand the rates of progress of germination to different percentiles,G, for single populations of each of five genotypes of chickpea(Cicer anetinum L.). The base temperature, Tb, at which therate of germination is zero, was 0·0°C for all germinationpercentiles of all genotypes. The optimum temperature, To(G),at which rate of germination is most rapid, varied between thefive genotypes and also between percentiles within at leastone population. Over the sub-optimal temperature range, i.e.from Tb to To(G), the distribution of thermal times within eachpopulation was normal. Consequently a single equation was appliedto describe the influence of sub-optimal temperatures on rateof germination of all seeds within each population of each genotype.The precision with which optimum temperature, Tb(G), could bedefined varied between populations. In each of three genotypesthere was a negative linear relationship between temperatureabove Tb(G) and rate of germination. For all seeds within anyof these three populations thermal time at supra-optimal temperatureswas constant. Variation in the time taken to germinate at supra-optimaltemperatures was a consequence of normal variation in the ceilingtemperature, To(G)—the temperature at or above which rateof progress to germination percentile G is zero. A new approachto defining the response of seed germination rate to temperatureis proposed for use in germplasm screening programmes. In two populations final percentage germination was influencedby temperature. The optimum constant temperature for maximumfinal germination was between 10°C and 15°C in thesepopulations; approximately 15°C cooler than the optimumtemperature for rate of germination. The water content and osmotic potential (2) of the differentparts of the pea fruit have been measured during developmentof the seed in two lines near-isogenic except for the r locus.During the early development of both genotypes, the testa possesseda more negative 2 than either embryo, endosperm or pod while,at stages when liquid endosperm was present, the embryo alwaysmaintained 2, more negative than the endosperm. A clear effectof the r locus on water content and 2 was only observed in embryotissue — wrinkled (rr) embryos possessing more water andmaintaining a more negative 2 than round (RR) for most of thedevelopmental period studied. The more negative 2 of wrinkledembryos correlated with their greater uptake of water in vivo. When cultured in vitro, the embryos of both genotypes showedmaximum growth (fresh or dry weight) if 10 per cent sucrosewas added to the medium (equivalent to about — 1.2 MPa).Round embryos, however, increased in weight more than wrinkledat all sucrose concentrations examined. An intercomparative study was carried out to investigate possibleeffects on primary productivity measurements when using NaH14C03solutions prepared by different methods. Five different ampoulebatches coded A, B, C, D and E were tested. Three of the batches(A, B and D) had been produced by direct dilution of industriallyproduced NaH14CO3, of high specific activity. A and D were dilutedwith distilled water added carrier, whereas no information onhow batch B was diluted could be obtained. Batch E was preparedby trapping 14CO2, gas — released by strong HCl from Ba14CO3— in sodium hydroxide. In the case of batch C, the processof manufacture was not known. The tests were carried out ondifferent phytoplankton material with low algal density. Twobatches (B and C) showed significant inhibitory effects on Pcalc(–5–44%), on the slope of the ascending part ofthe light adaptation curve (), and on Pmax. Batch A showed minor,but still significant effects. The four batches A, B, C andD carried rather high amounts of non-volatile rest activity(between 13 and 194 d.p.m./µCi), which made measurementsof the release of extracellular dissolved organic carbon (EOC)almost impossible. This phenomenon,per se, would if uncor-rectedproduce considerably higher per cent EOC release in low-productivewaters, as has been reported by many authors. As to the standardizationof the working solutions, two batches (C and D) showed a pooraccuracy (16 and 18% deviation, respectively) when tested atthe C14C, and two batches (A and B) showed unacceptably highvariability between ampoules of the same batch. The study indicatesthat it is not recom-mendable to use working solutions preparedby direct dilution of industrially produced NaH14CO3 of highspecific activity without prior testing of possible effectson algal photosynthesis. It is recommended that the specificactivity of the working solution be measured if it is not exactlyspecified by the manufacturer by a ‘Certificate of Quality’.  相似文献

Measurements of the rate of light-saturated photosynthesis (Pmax)were made on terminal leaflets of potato plants growing in cropssupplied with 0, 3, 6, 12, 24 and 36 g N m–2. Measurementswere made between 100 and 154 d after planting. Two types ofleaf were selected—the fourth leaf on the second-levelbranch (L4, B1) and the youngest terminal leaflet that was measurable(LYM). Later, the total nitrogen concentration of each leaflet(NL) was measured. A linear regression between Pmax and NL,common to both leaf positions, explained 68.5% of the totalvariation. With L4, B1 leaves there was a significant improvementin the proportion of variation explained when regressions withseparate intercepts and a common slope were fitted to individualfertilizer treatments. These results suggest that an increasingproportion of leaf nitrogen was not associated with the performanceof the photosynthetic system with increasing nitrogen supply.This separation between nitrogen treatments was not as clearfor LYM leaves. Stomatal conductance to transfer of water vapourwas neither influenced by leaf position nor directly by nitrogensupply. Arginine decarboxylase (EC 4.1.1.19 [EC] ) was purified from soybean,Glycine max, hypocotyls by a procedure which includes ammoniumsulfate fractionation, acetone precipitation, gel filtrationchromatography, and affinity chromatography. Using this procedure,ADC was purified to one band in non-denaturing PAGE. The purifiedADC has an Mr of 240 kDa based on gel filtration chromatographyand is a trimer of identical subunits which has an estimatedMr of 74 kDa based on SDS-PAGE. ADC is active between 30 and50°C and has a Km value of 46.1 µM. ADC is very sensitiveto agmatine or putrescine but not to spermidine or spermine.In the presence of 0.5 mM agmatine (or putrescine), the enzymeactivity was inhibited by 70%. Partially purified homoserine dehydrogenase was prepared frompea seedlings. The optimum pH for this enzyme is approximately 5.4. The Kmvaluesfor ASA and TPNH are 4.6xl0–4Af and 7.7xl0–5M, respectively.This enzyme can also utilize DPNH but less effectively thanTPNH. In contrast with yeast homoserine dehydrogenase whichis insensitive to — SH reagents, the pea enzyme is inhibitedalmost completely by 10–4MPCMB and 10–5MHgCl2, theinhibition being removed by 10–2M thioglycolate. Homoserinedehydrogenase was found not only in decotylized seedlings, butalso in cotyledons. The significance of this enzyme in homoserine biosynthesis ingerminating pea seeds has been discussed. Previously, it was established that the peptide YYWIGIRK-NH2inhibits both myeloid cell adhesion to selectins in vitro andneutrophil influx into inflammatory sites in vivo (Briggs etal., 1995). Initial structure/activity studies revealed thatat least one Y residue at the N-terminus of the peptide wasessential for these bioactivities but that the C-terminal Kresidue was unnecessary for inhibitory activity. We have nowsynthesized a new series of peptides which contain single residuesubstitutions at each position of the reference peptide, YYWIGIR-NH2and have tested these peptides for inhibitory activity in aselectin cell binding assay. In addition, peptides containingsingle D-amino acids at selected positions, or an all D-configuredreference peptide sequence, or the retro-inverso version (rigiwyy-NH2of the reference peptide sequence have also been analyzed forinhibitory activity in the same assays. Finally, the abilityof the reference peptide and a specifically designed controlsequence (YY(AIB)IGIR-NH2 to discriminate between potentialsynthetic saccharide ligands, including sialyl-Lewis x, Lewisx, and sialyl-N-acetyl-lactosamine, was investigated using isothermaltitration calorimetry. The results of these studies demonstratethat whereas many single amino acid substitutions are toleratedin the peptide without complete loss of inhibitory activity,substitution at some positions (e.g., the W residue) resultsin relatively inactive compounds, clearly pointing to the importanceof these residues in making critical contacts with the appropriatesaccharide ligand. Titration calorimetry revealed that the referencepeptide does not discriminate between Lewis x or sialyl-Lewisx in vitro, but binds these saccharides with nearly 40-foldhigher affinity (KD 25 µM) than the nonfucosylated trisaccharide,sialyl-N-acetyl-lactosamine. We can infer from these studiesthat the presence of a sialyl group, per Se, is not a requisitefor complex formation between the reference peptide and itssaccharide ligand. Substitution of single D-amino acid residuesat various po sitions in the reference peptide sequence reducesor elimi nates all inhibitory properties. However, the all Dconfigured peptide or the retro-inverso peptide sequence havegreater activity than the all L-configured reference peptidein the in vitro biological assays, and each was an effectiveinhibitor of neutrophil infiltration in a thioglycol late-inducedmouse peritonitis model. These results, combined with the resultsof titration, allow us to conclude that binding between thereference peptide and its saccharide ligand, which affords itsinhibitory properties, is mediated by the presence of a contiguous,nonpolar surface, or face, presented at the N-terminus of thereference peptide, likely encompassing the sequence YYWI. Rumex obtusifolius plants were grown for several months in daylitenvironment chambers (Solardomes) force-ventilated with aircontaining 350 or 600 µ;mol mol–1 C02. ElevatedCO2 was found to accelerate the natural ontogenic decline inphotosynthesis, but did not reduce leaf duration. In both CO2treatments photosynthetic rates declined progressively withincreasing leaf age, the decline being greater for plants grownin elevated C02 such that rates became lower than in ambientCO2. The degree of CO2-induced photosynthetic down-regulationas determined by A/C1 analysis was found to be dependent onleaf age. The major contribution to the decline in photosynthesiswas likely to be a reduction in Rubisco activity as changesin stomataland mesophyll limitations were small. Instantaneouswater use efficiency (WUE1) was greater for plants in elevatedCO2, but these values declined rapidly with leaf age, whereasin ambient CO2 values were always lower, but were maintainedfor longer. Effects of temperature on partition of assimilate between leaves,stems and panicles of pearl millet are analysed in terms ofa duration (tw) over which a structure increased in weight,and a partition factor (p)—the fraction of new dry matterallocated to the structure during tw. The value of tw was, forall structures, inversely proportional to temperature abovea base of 10 °C and below an optimum of 28 to 30 °C.For stems and panicles, the value of p was, with one exception,little affected by temperature. The dry weight of these structureswas, therefore, proportional to tw, and decreased with risein temperature. (The exception was panicles at the lowest temperature,19 °C, for which p was reduced by 40% because few grainswere set.) For leaves, however, p increased with rise in temperature,counteracting the effect on tw, such that dry weight changedlittle with temperature. The optimum temperature for reproductiveyield was 22 °C, but the proportion of the total dry matterallocated to reproductive structures changed little between22 °C and 31 °C.
