Summary The activities of seven enzymes were studied during the first 24 h of spherule formation (differentiation) in cultures of Physarum polycephalum. These enzymes were: isocitrate dehydrogenase, glucose-6-phosphate dehydrogenase, glutamate dehydrogenase, acid phosphatase, phosphodiesterase, -glucosidase, and histidase. Isocitrate dehydrogenase and glucose-6-phosphate dehydrogenase showed a decrease, glutamate dehydrogenase and phosphodiesterase increased about eight-fold, acid phosphatase about two-fold, -glucosidase showed an activity peak at about 15 h after starvation which decreased, after 24 h to its original value, and histidase showed no significant activity change during the period. The inhibition of protein synthesis by cycloheximide resulted in a decrease of enzymic activity at all times during the process, whereas the inhibition of RNA synthesis by actinomycin D had no effect during the early stages of spherulation but resulted in a rather rapid decrease of enzymic activity when added 21 h after the initiation of spherule formation. Attempts to induce higher enzymic activities during spherulation above the levels already observed were unsuccessful.
Zusammenfassung Der Aktivitätsverlauf, von sieben Enzymen wurde während der ersten 24 Std der Mikrosklerotienbildung verfolgt. Es handelte sich um die Enzyme: Isocitratdehydrogenase, Glucose-6-Phosphat Dehydrogenase, Glutamat-dehydrogenase, saure Phosphatase, Phosphodiesterase, -Glucosidase und Histidase. Isocitratdehydrogenase und Glucose-6-Phosphat Dehydrogenase nahmen während dieser Zeit ab, Glutamatdehydrogenase und Phosphodiesterase stiegen auf das 8fache des Ausgangswertes, saure Phosphatase auf das 2fache. -Glucosidase zeigte ein Aktivitätsmaximum nach ungefähr 15 Std, das aber nach etwa 24 Std wieder bis zu dem Ausgangswert abnahm, während Histidase sich nicht signifikant veränderte. Die Hemmung der Proteinsynthese durch Cycloheximid resultierte zu jedem Zeitpunkt in einer sofortigen Abnahme der Enzymaktivität, während die Inhibition der RNS-Synthese in der ersten Zeit ohne Effekt war, jedoch bei einer Zugabe nach 21 Std nach der Induktion der Spherulation eine ziemlich schnelle Abnahme der Enzymaktivität bewirkte. Versuche, die Enzymaktivitäten zusätzlich zu den schon beobachteten zu induzieren, schlugen fehl.

Activity of malate dehydrogenase (oxalacetate reduction) was 10.9 mumol/min/mg protein; the apparent Km was 64 microM. 3. The activity of NADP-isocitric dehydrogenase (IDH) was 110 nmol/min/mg with apparent Km of 35 microM. 4. NAD-IDH showed allosteric properties with AMP as a positive modulator. The apparent Km for the unmodulated activity, 2 mM, was decreased to 0.95 mM by 0.13 mM AMP. 5. Succinic dehydrogenase activity was estimated as three times higher than that of alpha-glycerophosphate dehydrogenase. 6. Mitochondria contained significant amounts of phenolic compounds. The cultivation of Physarum polycephalum amoebae in two media with different protein contents revealed a regulation of aminopeptidases and proteases depending on the albumin content of the medium: in growing amoebae and plasmodia the aminopeptidases have similar isoenzyme patterns and relative activities against nitroanilides. One alanine and four leucine aminopeptidase isoenzymes were found within the slightly acid pH range. During growth amoebae secrete—different from plasmodia—leucine aminopeptidase into the medium with low protein content. In an albumin-rich medium additional alanine aminopeptidase activity was found. Out of nine plasmodial proteases four were found in amoebae too. Only one band (pI 3.6) was present in the protein-poor medium. The cell-cycle-related activities of the cAMP- and cGMP-dependent phosphodiesterases of Physarum polycephalum were assayed. the activities of plasmodial homogenate and of selected subcellular fractions were measured. the results suggested the presence of both cAMP- and cGMP-dependent phosphodiesterase in the isolated nuclei of P. polycephalum. In addition, they reveal that the cAMP- and cGMP-dependent phosphodiesterase activities of the subcellular fractions fluctuate throughout the cell cycle. the whole-cell homogenates exhibit no cell-cycle-related changes in the presence of 5 × 10-4 m cGMP. Kinetic data suggest the presence of multiple phosphodiesterase activities in the homogenate and its particulate fractions for the cGMP-dependent enzyme. Multiple cAMP activities are also suggested for the particulate fractions. the Km values indicate that the substrate affinities of the phosphodiesterases from P. polycephalum are similar to those found previously in mammalian systems.  相似文献

The levels of glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, lactate dehydrogenase, and cyclic phosphodiesterase activities were examined in growing and starving plasmodia of Physarum polycephalum. The activities of lactate dehydrogenase, 6-phosphogluconate dehydrogenase, and glucose-6-phosphate dehydrogenase decreased whereas that of cyclic phosphodiesterase increased. The change in activity of lactate dehydrogenase was the result of the variation of the activity of a single enzyme quite similar to the lactate dehydrogenases of higher animals.  相似文献

1. Sinapis alba L. seedlings contain glycosyltransferase catalyzing the synthesis of sterol glucosides in the presence of UDPglucose as sugar donor. The major activity occurs in the membranous fraction sedimenting at 300--9000 x g. Successive treatment of the particulate enzyme fraction with acetone and Triton X-100 affords a soluble glucosyltransferase preparation which can be partly purified by gel filtration on Sephadex G-150. Molecular weight of the glucosyltransferase is 1.4 . 10(5). Apparent Km values for UDPglucose and sitosterol are 8.0 . 10(-5) M and 5.0 . 10(-6) M, respectively. 2. Comparison was made of the S. alba glucosyltransferase with a similar sterol-glucosylating enzyme isolated from non-photosynthesizing organism Physarum polycephalum (Myxomycetes). UDPglucose was the most efficient glucose donor in both cases but the enzyme from Ph. polycephalum can also utilize CDPglucose and TDPglucose. Glucose acceptors are, in case of both enzymes, sterols containing a beta-OH group at C-3 and a planar ring system (5 alpha-H or double bond at C-5). The number and position of double bonds in the ring system and in the side chain, as well as the presence of additional alkyl groups in the side chain at C-24 are of secondary importance. 3. The present results indicate that both enzymes can be regarded as specific UDPglucose:sterol glucosyltransferases. Certain differences in their specificity towards donors and acceptors of the glucosyl moiety suggest, however, a different structure of the active sites in both enzymes.  相似文献

利用BrUTP免疫标记技术,研究了多头绒泡菌(Physarum polycephalum Sclhw.)间期细胞核中RNA的转录状况。结果表明:在整修间期核仁中的rRNA都在活跃转录;核质中hnRNA的转录呈逐渐上升趋势,早S期转录水平很低,晚S期转录活性升高1倍,G2期转录达到最高水平;整修间期核质中RNA的转录水平增加了5-6倍。  相似文献

The expression and cytological distribution of acetylated alpha-tubulin was investigated in Physarum polycephalum. A monoclonal antibody specific for acetylated alpha-tubulin, 6-11B-1 (Piperno, G., and M. T. Fuller, 1985, J. Cell Biol., 101:2085-2094), was used to screen for this protein during three different stages of the Physarum life cycle--the amoeba, the flagellate, and the plasmodium. Western blots of two-dimensional gels of amoebal and flagellate proteins reveal that this antibody recognizes the alpha 3 tubulin isotype, which was previously shown to be formed by posttranslational modification (Green, L. L., and W. F. Dove, 1984, Mol. Cell. Biol., 4:1706-1711). Double-label immunofluorescence demonstrates that, in the flagellate, acetylated alpha-tubulin is localized in the flagella and flagellar cone. Similar experiments with amoebae interestingly reveal that only within the microtubule organizing center (MTOC) are there detectable amounts of acetylated alpha-tubulin. In contrast, the plasmodial stage gives no evidence for acetylated alpha-tubulin by Western blotting or by immunofluorescence.  相似文献

The total concentration of calcium in the endoplasm of plasmodia of was measured using a calcein fluorescence-quenching technique. The calcium concentration of the endoplasm increases with the time of cultivation on different substrates under culture conditions frequently used for routine experiments. Calcium accumulation within endo- and ectoplasm as well as in microplasmodia is most pronounced when plasmodia are starved and illuminated. Starvation and illumination lead to calcium concentrations frequently exceeding 200 mM in the case of macroplasmodia and 20 mM in the case of microplasmodia.  相似文献
