Zygogonium ericetorum, the type species of the genus, was studied from a natural population collected in Mt. Schönwieskopf, Tyrol, Austria. Generic concepts of Zygogonium and Zygnema were tested with atpB, psbC, and rbcL gene sequence analysis, which showed a sister relationship between Z. ericetorum and Mesotaenium, in an early branching clade sister to a grouping of Zygnema and several other filamentous and unicellular zygnematalean taxa. A variety of light, confocal, transmission electron microscopy, and cytochemical techniques provided new data on the variable chloroplast shape of Z. ericetorum, and its aplanospore structure and development, which has been previously considered taxonomically important but has been ambiguously interpreted. Zygogonium can be distinguished from other zygnematophytes (particularly Zygnema), based on the combination of two characters: (i) irregular, compressed plate‐like chloroplasts and (ii) residual cytoplasmic content left in sporangia outside of the fully developed aplanospores or zygospores. The presence of a sporangial wall that separates the spores from the parent cell should be excluded from the definition of Zygogonium, because it is also observed in Zygnema. Similarly, the ecological characterization of Zygogonium as acidophilic is not unique to the genus. The names of 18 species currently belonging to Zygogonium are here changed to Zygnema, because of incompatibility with this new proposed Zygogonium concept. In the species transferred to Zygnema, chloroplasts are typically stellate in three‐dimensions, and the entire content of fertile cells is transformed into the spore, so there is no cytoplasmic residue.  相似文献

The filamentous green alga Zygogonium ericetorum (Zygnematophyceae, Streptophyta) was collected in a high‐alpine rivulet in Tyrol, Austria. Two different morphotypes of this alga were found: a purple morph with a visible purple vacuolar content and a green morph lacking this coloration. These morphotypes were compared with respect to their secondary metabolites, ultrastructure, and ecophysiological properties. Colorimetric tests with aqueous extracts of the purple morph indicated the presence of soluble compounds such as phenolics and hydrolyzable tannins. High‐performance liquid chromatography‐screening showed that Z. ericetorum contained several large phenolic peaks with absorption maxima at ~280 nm and sometimes with minor maxima at ~380 nm. Such compounds are uncommon for freshwater green microalgae, and could contribute to protect the organism against increased UV and visible (VIS) irradiation. The purple Z. ericetorum contained larger amounts (per dry weight) of the putative phenolic substances than the green morph; exposure to irradiation may be a key factor for accumulation of these phenolic compounds. Transmission electron microscopy of the purple morph showed massive vacuolization with homogenous medium electron‐dense content in the cell periphery, which possibly contains the secondary compounds. In contrast, the green morph had smaller, electron‐translucent vacuoles. The ecophysiological data on photosynthesis and desiccation tolerance indicated that increasing photon fluence densities led to much higher relative electron transport rates (rETR) in the purple than in the green morph. These data suggest that the secondary metabolites in the purple morph are important for light acclimation in high‐alpine habitats. However, the green morph recovered better after 4 d of rehydration following desiccation stress.  相似文献

Athetis lepigone has been recorded in many countries in Europe and Asia, but it had never been documented as an agricultural pest until 2005. For the purpose of using the sex pheromone to control this pest, we conducted a study to identify the sex pheromone of A. lepigone by gas chromatography with an electroantennographic detector (GC‐EAD) and GC coupled with mass spectrometry (GC/MS) analyses. Three pheromone candidates were detected by GC‐EAD analysis in the extracts of the female sex pheromone gland, and two candidates were identified as (Z)‐7‐dodecenyl acetate (Z7‐12:OAc) and (Z)‐9‐tetradecenyl acetate (Z9‐14:OAc) in a ratio of 1:5 by mass spectral analysis of natural pheromone components and dimethyl disulphide adducts. In the field male trapping test, the traps baited with the binary blend captured high number of males, while traps with single component hardly caught males, indicating that the two components are essential for the male attractiveness. In addition, the optimum ratios of Z7‐12:OAc and Z9‐14:OAc were determined as 3:7–7:3, and the best doses for the binary blend (at ratio of 3:7 between Z7‐12:OAc and Z9‐14:OAc) were 0.25–0.5 mg/trap, based on the number of male catches. The identification of a highly attractive sex pheromone will help in developing efficient strategies for monitoring and control of A. lepigone.  相似文献

Geographic range size and genetic diversity are key correlates of extinction risk and evolutionary potential of a species, with species occupying smaller geographic ranges and showing limited genetic diversity assumed to be more threatened by environmental changes. The Italian Aesculapian snake Zamenis lineatus is a narrow‐range endemic of southern Italy and Sicily, once considered as part of the widespread species Z. longissimus. To date, we still lack comprehensive data on geographic range and intraspecific diversity of Z. lineatus. In this study, we analysed 106 Aesculapian snakes across the Italian Peninsula and Sicily in order to define the genetic diversity and distribution range of Z. lineatus, its possible range overlap with Z. longissimus and to assess whether hybridization occurs at the species’ range boundaries. We combined genetic data from mitochondrial and nuclear DNA markers with phenotypic data suitable for taxonomic identification. The observed phylogeographic pattern of Z. lineatus suggests: (i) a reduced peninsular range size, about a half of what is currently considered; (ii) limited genetic diversity and weak population structure; (iii) the occurrence of pervasive introgressive hybridization with Z. longissimus in the eastern contact zone. Together, results from this study indicate a higher extinction risk for Z. lineatus than previously appreciated and provide directions for future studies on the hybridization at the contact zone(s) between Z. lineatus and Z. longissimus.  相似文献

The cocoa pod borer (CPB), Conopomorpha cramerella (Snellen), sex pheromone was previously identified as a blend of (E,Z,Z)‐ and (E,E,Z)‐4,6,10‐hexadecatrienyl acetates and corresponding alcohols. These pheromone components were synthesized by modification of an existing method and the relative attractiveness of synthetic blends that included different levels of non‐target pheromone components and chemical purities was tested in a cocoa field using Delta traps. Male captures were not significantly different among traps baited with pheromone blends containing 5% to 47% (based on four identified pheromone components) of other geometric acetates [(E,Z,E)‐, (Z,Z,Z)‐, (Z,E,Z)‐ and (Z,E,E)‐4,6,10‐hexadecatrienyl acetates], indicating that C. cramerella males did not discriminate among the pheromone components and other geometric isomers in the blends. Therefore, neither antagonistic nor synergistic effects from other pheromone geometric isomers were observed. The modified synthetic pathway offers the prospect of more economical production of CPB sex pheromone. During 17 weeks when C. cramerella monitoring coincided with the main cocoa pod harvest period in 2013–2014, CPB trap catch data from some blends showed a good correlation with the number of pods with C. cramerella infestation symptoms.  相似文献

The phylogenetic relationships between western Palaearctic Zamenis and Rhinechis ratsnakes have been troubled, with recent estimates based on the supermatrix approach questioning their monophyly and providing contradictory results. In this study, we generated a comprehensive molecular data set for Zamenis and closely related ratsnakes to assess their phylogenetic and systematic relationships and infer their spatial and temporal modes of diversification. We obtained a fully resolved and well‐supported phylogeny, which is consistent across markers, taxon‐sets and phylogenetic methods. The close phylogenetic relationship between Rhinechis and Zamenis is well‐established. However, the early branching pattern within this clade, and the position of R. scalaris and Z. hohenackeri, remains poorly supported. The Persian ratsnake Z. persicus is sister to the Mediterranean species Z. situla, Z. longissimus and Z. lineatus, of which Z. situla is sister to a clade containing the latter two species. These results are consistent with a recent phylogenomic study on ratsnakes based on hundreds of loci. Whereas, topological tests based on our data and evidence from such phylogenomic study strongly rejected previous phylogenetic estimates based on the supermatrix approach and demonstrate that these “mega‐phylogenies”, with hundreds of taxa and high levels of missing data, have recovered inconsistent relationships with spurious nodal support. Biogeographical and molecular dating analyses suggest an origin of the ancestor of Rhinechis and Zamenis in the Aegean region with early cladogenesis during the Late Miocene associated with the Aegean arch formation and support a scenario of east‐to‐west diversification. Finally, while we have little morphological and phylogenetic evidence for the distinctiveness between Rhinechis and Zamenis, a classification of them in a single genus, and the designation of Zamenis scalaris (Schinz, 1822), reflects better their evolutionary relationships.  相似文献

Cotton‐ and maize‐producing insecticidal crystal (Cry) proteins from the bacterium, Bacillus thuringiensis (Bt), have been commercialized since 1996. Bt plants are subjected to environmental risk assessments for non‐target organisms, including natural enemies that suppress pest populations. Here, we used Cry1F‐resistant Spodoptera frugiperda (J.E. Smith) and Cry1Ac and Cry2Ab‐resistant Trichoplusia ni (Hübner) as prey for the assassin bug, Zelus renardii (Kolenati), a common predator in maize and cotton fields. In tritrophic studies, we assessed several fitness parameters of Z. renardii when it fed on resistant S. frugiperda that had fed on Bt maize expressing Cry1F or on resistant T. ni that had fed on Bt cotton expressing Cry1Ac and Cry2Ab. Survival, nymphal duration, adult weight, adult longevity and female fecundity of Z. renardii were not different when they were fed resistant‐prey larvae (S. frugiperda or T. ni) reared on either a Bt crop or respective non‐Bt crops. ELISA tests demonstrated that the Cry proteins were present in the plant at the highest levels, at lower levels in the prey and at the lowest levels in the predator. While Z. renardii was exposed to Cry1F and Cry1Ac and Cry2Ab when it fed on hosts that consumed Bt‐transgenic plants, the proteins did not affect important fitness parameters in this common and important predator.  相似文献

In moth species, females emit a species‐specific sex pheromone that is perceived over long distance by conspecific males. The species‐specificity in the chemical communication channel is achieved by a combination of unique components in specific ratios and sometimes also by interspecific behavioural antagonists to deter sympatrically occurring heterospecific males. In this study, we determined possible antagonistic effects in Helicoverpa gelotopoeon Dyar (Lepidoptera: Noctuidae) males to the major sex pheromone component of sympatrically occurring heliothine moths, Z11‐16:Ald, as well as to the sex pheromone of the sympatrically occurring Heliothis virescens (Fabricius) (Lepidoptera: Noctuidae) (Z11‐16:Ald and Z9‐14:Ald). We also explored whether other co‐occurring species are attracted to these pheromone blends. Our field experiments showed that the addition of Z11‐16:Ald alone or in combination with Z9‐14:Ald inhibited trap catches of H. gelotopoeon males and that this inhibition depended on the concentration of these compounds. In addition, other moth species were attracted to the blends. Together, our results confirm the antagonistic effect of heterospecific sex pheromone compounds of H. virescens to H. gelotopoeon.  相似文献

Aim To test alternative biogeographic hypotheses related to the diversification of a montane mammal (Zapus hudsonius luteus) endemic to the American Southwest. Location South‐western United States. Methods We used statistical phylogeographic analyses of mitochondrial DNA (1512 bp; two genes) from 93 individuals from six geographic regions to test diversification hypotheses. Species distribution models of climate and fossil records were integrated to assess contemporary and historical distributions and barriers to gene flow. We calculated dates of divergence and examined historical demography using coalescent simulations. Results We documented monophyly of Z. h. luteus represented by 19 segregated haplotypes. Predicted current distribution generally coincided with known localities, while predicted paleodistributions suggested that this lineage was widespread throughout lower elevations of the American Southwest and on the Edwards Plateau (as documented by the fossil record). Population size did not change substantially during a westward shift in range that occurred in the last 100 k generations. Results supported fragmentation of a common ancestor during the Holocene as the most plausible explanation for genetic structure. Main conclusions Monophyletic Z. h. luteus reflects fragmentation of a common ancestor with recent (Holocene) upslope colonization of disjunct montane areas. We refute the hypotheses of in situ divergence or origins from a Colorado Piedmont ancestor. Instead, westward colonization from the Edwards Plateau during the Wisconsin followed by Holocene fragmentation, which serves as a generalized biogeographic hypothesis for species associated with mesic graminoid habitats in the American Southwest. Further exploration of these signatures using independent nuclear DNA is warranted. Key conservation implications are (1) Z. h. luteus is a monophyletic lineage on an independent evolutionary trajectory; (2) Z. h. luteus shared a recent common ancestor with Z. h. pallidus (not Z. h. preblei); (3) mtDNA does not reflect recent population declines; and (4) coalescent simulations and species distribution models reflect Holocene fragmentation.  相似文献

The increasing demand for food production and predicted climate change scenarios highlight the need for improvements in crop sustainability. The efficient use of water will become increasingly important for rain‐fed agricultural crops even in fertile regions that have historically received ample precipitation. Improvements in water‐use efficiency in Zea mays have been limited, and warrant a renewed effort aided by molecular breeding approaches. Progress has been constrained by the difficulty of measuring water‐use in a field environment. The stable carbon isotope composition (δ13C) of the leaf has been proposed as an integrated signature of carbon fixation with a link to stomatal conductance. However, additional factors affecting leaf δ13C exist, and a limited number of studies have explored this trait in Z. mays. Here we present an extensive characterization of leaf δ13C in Z. mays. Significant variation in leaf δ13C exists across diverse lines of Z. mays, which we show to be heritable across several environments. Furthermore, we examine temporal and spatial variation in leaf δ13C to determine the optimum sampling time to maximize the use of leaf δ13C as a trait. Finally, our results demonstrate the relationship between transpiration and leaf δ13C in the field and the greenhouse. Decreasing transpiration and soil moisture are associated with decreasing leaf δ13C. Taken together these results outline a strategy for using leaf δ13C and reveal its usefulness as a measure of transpiration efficiency under well‐watered conditions rather than a predictor of performance under drought.  相似文献

Polyketide synthases (PKSs) occur in many bacteria, fungi and plants. They are highly versatile enzymes involved in the biosynthesis of a large variety of compounds including antimicrobial agents, polymers associated with bacterial cell walls and plant pigments. While harmful algae are known to produce polyketide toxins, sequences of the genomes of non‐toxic algae, including those of many green algal species, have surprisingly revealed the presence of genes encoding type I PKSs. The genome of the model alga Chlamydomonas reinhardtii (Chlorophyta) contains a single type I PKS gene, designated PKS1 (Cre10.g449750), which encodes a giant PKS with a predicted mass of 2.3 MDa. Here, we show that PKS1 is induced in 2‐day‐old zygotes and is required for their development into zygospores, the dormant stage of the zygote. Wild‐type zygospores contain knob‐like structures (~50 nm diameter) that form at the cell surface and develop a central cell wall layer; both of these structures are absent from homozygous pks1 mutants. Additionally, in contrast to wild‐type zygotes, chlorophyll degradation is delayed in homozygous pks1 mutant zygotes, indicating a disruption in zygospore development. In agreement with the role of the PKS in the formation of the highly resistant zygospore wall, mutant zygotes have lost the formidable desiccation tolerance of wild‐type zygotes. Together, our results represent functional analyses of a PKS mutant in a photosynthetic eukaryotic microorganism, revealing a central function for polyketides in the sexual cycle and survival under stressful environmental conditions.  相似文献

A new peritrichous ciliate, Zoothamnium palmphlatum nov. spec., was collected from an estuary in Yantai, China. It was investigated, using both live observation and silver staining. The new species can be identified by a palm‐shaped colony consisting of highly developed and alternately arranged secondary branches, a double‐layered peristomial lip, and an infundibular polykinety 3 composed of two parallel kinetosomal rows. Phylogenetic analyses of the small subunit (SSU) rRNA gene sequence show that Z. palmphlatum clusters with other members of the family Zoothamniidae. Furthermore, the comparison of primary and secondary SSU rDNA structures indicates that Z. palmphlatum is distinctly different from its morphologically similar species (93.2–97.0% in sequence similarity) in combination of H10 and H31 regions. Although Zpalmphlatum shares highest sequence similarity with Zoothamnium mucedo (98.9%), the new species has distinctly different structures in the H11, H12, H18, and H31 regions compared to Z. mucedo, which indicates that sequence similarity may not determine the similarity of the secondary structure.  相似文献

We report in this study large induction of volatile organic compounds (VOCs) from a single inflorescence of mossy sorrel (Rumex confertus Willd., Polygonaceae), by herbivory of the weevil (Hypera rumicis L., Coleoptera: Curculionidae). VOCs blend induced by the weevil herbivory included 1 green leaf volatiles (GLVs) ((Z)‐3‐hexen‐1‐yl acetate), five terpenes ((Z)‐β‐ocimene, linalool, geranyl acetate, β‐caryophyllene and (E)‐β‐farnesene), three esters (benzyl acetate, methyl salicylate and methyl anthranilate) and one aromatic heterocyclic organic compound (indole). Uninjured plants produced only detectable amounts of VOCs. A Y‐tube experiment revealed that both females and males of H. rumicis were not attracted to any of tested concentrations (1, 5, 25, 125 ng/min). Also both females and males were significantly repelled by the highest concentrations (25 and 125 ng/min). Additionally, concentration of 5 ng/min proved to be repellent for females of H. rumicis.  相似文献

Selection for plant traits important for agriculture can come at a high cost to plant defenses. While selecting for increased growth rate and yield, domestication and subsequent breeding may lead to weakened defenses and greater susceptibility of plants to herbivores. We tested whether expression of defense genes differed among maize, Zea mays ssp. mays L. (Poaceae), and its wild relatives Zea mays ssp. parviglumis Iltis & Doebley and Zea diploperennis Iltis et al. We used two populations of Z. mays ssp. parviglumis: one expected to express high levels of an herbivore resistance gene, wound‐inducible protein (wip1), and another expected to have low expression of wip1. To test whether maize and wild Zea differed in induction of defenses against Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae), we quantified expression of several genes involved in plant defense: wip1, maize protease inhibitor (mpi), pathogenesis‐related protein (PR‐1), and chitinase. Moreover, we compared growth, development, and survival of caterpillars on maize and wild Zea plants. We found that maize expressed low levels of all but one of the genes when attacked by caterpillars, whereas the wild relatives of maize expressed induced defense genes at high levels. Expression of wip1, in particular, was much greater in the Z. mays ssp. parviglumis population that we expected to naturally express high levels of wip1, with expression levels 29‐fold higher than in herbivore‐free plants. Elevated expression of defenses in wild plants was correlated with higher resistance to caterpillars. Larvae were 15–20% smaller on wild Zea compared with maize, developed 20% slower, and only 22% of them survived to pupation on Z. mays ssp. parviglumis with high levels of wip1. Our results suggest that domestication has inadvertently reduced the resistance of maize, and it is likely that expression of wip1 and other genes associated with defenses play an important role in this reduction in resistance.  相似文献

Recently, Schroeder et al. (2010, Ibis 152: 368–377) suggested that intronic variation in the CHD1‐Z gene of Black‐tailed Godwits breeding in southwest Friesland, The Netherlands, correlated with fitness components. Here we re‐examine this surprising result using an expanded dataset (2088 birds sampled from 2004 to 2010 vs. 284 birds from 2004 to 2007). We find that the presence of the Z* allele (9% of the birds) is not associated with breeding habitat type, egg size, adult survival, adult body mass or adult body condition. The results presented here, when used in synergy with the previously reported results by Schroeder et al., suggest that there might be a tendency towards female adults with the Z* allele laying earlier clutches than adult females without the Z* allele. The occurrence of the Z* allele was also associated with a higher chick body mass and return rate. Chicks with the Z* allele that had hatched early in the breeding season were heavier at birth than chicks without the Z* allele and chicks with the Z* allele that had hatched late. Collectively, the results suggest that variation in the CHD1‐Z gene may indeed have arisen as a byproduct of selection acting on females during the egg fase and on chicks during the rearing stages of the reproductive cycle.  相似文献

The crude extract from Cnaphalocrocis medinalis (Lepidoptera, Pyralidae) has been identified to be (Z)‐11‐octadecenal (Z11‐18:Ald), (Z)‐13‐octadecenal (Z13‐18:Ald), (Z)‐11‐octadecen‐1‐ol (Z11‐18:OH) and (Z)‐13‐octadecen‐1‐ol (Z13‐18:OH) in the GC‐MS, confirming the results reported previously in Japan. Field evaluation in China showed that the optimal blend is Z11‐18:Ald, Z13‐18:Ald, Z11‐18:OH and Z13‐18:OH at a ratio of 3 : 25 : 3 : 3. The optimal dosage is 500 μg Z13‐18:Ald per PVC tubing lure. In August, trap catches were highest between 3:00 and 4:00 a.m., and no moths were caught before 12:00 a.m. or after 6:00 a.m. The impact of trap design and trap height placement was also evaluated in the paddy field. The sticky wing trap was better than the water pan trap. The number of moths captured by the electric grid trap was 1.7 times greater than caught by the sticky wing trap. The optimal trap height was 20 cm below the top of the rice plants (80 cm high). Pheromone lure formulated from 60 μg Z11‐18:Ald, 500 μg Z13‐18:Ald, 60 μg Z11‐18:OH and 60 μg Z13‐18:OH was effective for population monitoring in trial sites in Zhejiang, Shanghai, Jiangxi, Hunan, Hubei, Guangxi, Guizhou, Chongqing and Hainan Provinces, encompassing both subtropical and temperate regions. Use of this synthetic pheromone blend provides a promising alternative to the currently used disturb and count method (DCM) for monitoring the population of C. medinalis in China.  相似文献

The genus Pseudo‐nitzschia contains potentially toxic species of problematic taxonomy, making it one of the most intensively studied diatom genera. The study of 35 clonal strains isolated from the Bilbao estuary, an area that experiences recurrent blooms of Pseudo‐nitzschia, revealed the presence of two new species, P. abrensis and P. plurisecta, differing from their congeners in both morphology and gene sequence. The morphological features were analyzed by LM and EM, whereas molecular analyses were based on the internal transcribed spacer (ITS) and large subunit (LSU) regions of the rDNA. P. plurisecta appears closely related to P. cuspidata/P. pseudodelicatissima in the phylogenetic tree, whereas P. abrensis forms a moderately supported clade with P. heimii/P. subpacifica and P. caciantha/P. circumpora. Comparison of the secondary structure of ITS2 regions reveals marked differences in the most highly conserved regions among related taxa. Morphologically, the new species differ from their closest congeners in the arrangement of the poroid sectors and the density of valve striae and fibulae. The two species share similar pigment composition, and belong to the group of Pseudo‐nitzschia species containing only chlorophyll c2 and c3.  相似文献

Phylogenetic relationships of Oceanian staple yams (species of Dioscorea section Enantiophyllum) were investigated using plastid trnL‐F and rpl32‐trnL(UAG) sequences and nine nuclear co‐dominant microsatellites. Analysis of herbarium specimens, used as taxonomic references, allowed the comparison with samples collected in the field. It appears that D. alata, D. transversa and D. hastifolia are closely related species. This study does not support a direct ancestry from D. nummularia to D. alata as previously hypothesized. The dichotomy in D. nummularia previously described by farmers in semi‐perennial and annual types was reflected by molecular markers, but the genetic structure of D. nummularia appears more complex. Dioscorea nummularia displayed two haplotypes, each corresponding to a different genetic group. One, including a D. nummularia voucher from New Guinea, is closer to D. tranversa, D. alata and D. hastifolia and encompasses only semi‐perennial types. The second group is composed of semi‐perennial and annual yams. However, some of these annual yams also displayed D. alata haplotypes. Nuclear markers revealed that some annual yams shared alleles with D. alata and semi‐perennial D. nummularia, suggesting a hybrid origin, which may explain their intermediate morphotypes and the difficulty met in classifying them.  相似文献
