Eleven polymorphic microsatellite loci were isolated and characterized from an AC-enriched genomic library of Akebia trifoliate ssp. australis. The number of allele per locus ranged from 3 to 14. The observed heterozygosity and expected heterozygosity at population
level were 0.196–1.000 and 0.522–0.902, respectively. In addition, this set of microsatellites produced robust cross-species
amplification in other two related taxa, suggesting these microsatellite markers should provide a useful tool for genetic

Robust, polymorphic microsatellite DNA markers (simple sequence repeats—SSRs) are valuable tools for a range of tree conservation
and breeding applications. SSRs are routinely used in the study of population genetic structure and diversity, pedigree reconstruction
and genetic linkage mapping. Their abundance in the genome, co-dominant inheritance and potential for cross-species amplification
make microsatellites highly prized markers. This paper characterises 22 novel genomic polymorphic microsatellite loci for
Sitka spruce (Picea sitchensis (Bong.) Carr.). Amplification of DNA from Sitka spruce material was carried out both with a set of unrelated trees to obtain
diversity statistics for each locus, and with the progeny of a full-sib family to test simple Mendelian inheritance. Observed
heterozygosity ranged from 0.38 to 0.91 and allele number per locus ranged from 6 to 21, with a mean of 12.2. In addition,
the primer pairs were tested with DNA from Norway spruce (P. abies) and white spruce (P. glauca) to investigate their potential for cross-species amplification and ten loci amplified in all three species. The results
from these genomic microsatellites are compared to data generated from microsatellites derived from Picea EST libraries. In summary, this novel, highly polymorphic markers represent a significant addition to the rapidly expanding
Picea genomics tool-box.

Achnatherum sibiricum is a threaten and toxic perennial bunchgrass mainly in the north of China. We developed 11 polymorphic microsatellite loci
from A. sibiricum by combining biotin capture method. After validating and scoring, these loci were polymorphic in a test population with the
range of alleles from four to 13 per locus. The observed and expected heterozygosities ranged from 0.1649 to 0.5624 and from
0.3071 to 0.8826, respectively. All 11 microsatellite markers were in Hardy–Weinberg equilibrium. These polymorphic microsatellites
will be useful for genetic diversity analysis and population structure construction for A. sibiricum. 相似文献

We report the development of 15 new microsatellite markers for Emys orbicularis and Emys trinacris. A survey of 20 individuals showed that all loci are highly polymorphic with 3–14 alleles per locus. Additionally, 22 Glyptemys muhlenbergii primers were checked for cross-species amplification, with 14 being amplified successfully and polymorphic (2–14 alleles).
A set of eight markers was selected and combined into two multiplex PCRs. Levels of genetic diversity were assessed in 648
individuals covering the complete distribution area. The number of alleles ranged from 13 to 24 and observed heterozygosity

Isolation and Characterization of Twenty Microsatellite Loci in Japanese Sea Cucumber (Stichopus japonicus) 总被引:5,自引:0,他引:5
Twenty microsatellite markers were first developed from the Japanese sea cucumber Stichopus japonicus using an enrichment protocol. Of the 20 microsatellite loci, 19 loci were polymorphic in the population examined. At these polymorphic loci, the number of alleles per locus varied from 2 to 15, and the observed heterozygosities ranged from 0.03 to 0.97, which is considerably higher than those previously found for allozymes. The high variability of the microsatellite markers identified in this study will make them excellent tools for genetic analyses of S. japonicus. 相似文献

Five new polymorphic microsatellite loci were developed for the coral reef damselfish Pomacentrus amboinensis. Twenty-four individuals from two Great Barrier Reef populations were genotyped at the five loci, with numbers of alleles
per locus ranging from 6–23 and observed heterozygosity between 0.42–0.92. In addition, the cross-species testing of six primers
developed for Stegastes partitus revealed one primer (SpGATA40) that was also polymorphic for P. amboinensis. Due to high levels of polymorphism (≥14 alleles) in at least four of the six loci and a high proportion of tetranucleotide
repeats, these microsatellite markers should be useful for parentage assignment as well as other investigations of individual

Chinese alligator (Alligator sinensis) is a critically endangered species endemic to China. In this study, the extent of genetic variation in the captive alligators
of the Changxing Reserve Center was investigated using microsatellite markers derived from American alligators. Out of 22
loci employed, 21 were successfully amplified in the Chinese alligator. Sequence analysis showed loci in American alligators
had a bigger average size than that of the Chinese alligators and the longest allele of an individual locus almost always
existed in the species with longer stretch of repeat units. Eight of the 22 loci were found to be polymorphic with a total
of 26 alleles present among 32 animals scored, yielding an average of 3.25 alleles per polymorphic locus. The expected heterozygosity
(H
E) ranged at a moderate level from 0.4385 to 0.7163 in this population. Compared to that in the American alligators, a lower
level of microsatellite diversity existed in the Changxing population as revealed by about 46% fewer alleles per locus and
smaller H
E at the homologous loci. The average exclusion power and the ability to detect shared genotypes and multiple paternity were
evaluated for those markers. Results suggested that when the polymorphic loci were combined, they could be sensitive markers
in genetic diversity study and relatedness inference within the Chinese alligator populations. The level of genetic diversity
present in the current Changxing population indicated an important resource to complement reintroductions based on the individuals
from the other population. In addition, the microsatellite markers and their associated diversity characterized in this population
could be utilized to further investigate the genetic status of this species. 相似文献

Cupressus chenggiana S. Y. Hu (Cupressaceae) is an endemic and endangered conifer species in southwest China. In order to study the population
genetics and design the effective conservation methods, we aimed to develop microsatellite primers for this species in the
present study. We developed eight new microsatellite loci for this species through biotin capture method. Polymorphism of
each locus was further assessed in 18 individuals from three geographically distant populations. The number of alleles per
locus ranged from 6 to 11 with an average of 8.13. The observed and expected heterozygosities ranged from 0.219 to 0.296 and
from 0.374 to 0.470, with averages of 0.254 and 0.417, respectively. We further found that three of nine microsatellite loci
developed previously for another congeneric species showed polymorphic banding patters. We performed primer-crossing tests
of these loci in the other two congeneric species which are closely related to C. chenggiana (C. gigantea and C. duclouxiana). These microsatellite markers would be effective for analyzing genetic diversity and population genetic structure of this
species and its morphological differentiation with the close relatives. 相似文献

Twenty-eight polymorphic microsatellite loci were developed and characterized for the endangered tree Berchemiella wilsonii var. pubipetiolata. The observed number of allele ranged from two to seven. The ranges of observed and expected heterozygosity were 0.039–1.000 and 0.038–0.816, respectively. In addition, this set of microsatellites produced robust cross-species amplification in other two endangered taxa: Berchemiella berchemiaefolia and Berchemiella wilsonii, suggesting these microsatellite markers should provide a useful tool for genetic and conservation studies of the globally endangered genus Berchemiella. 相似文献

A total of 12 polymorphic microsatellite loci were developed and characterized for a Chinese medicinal plant, Epimedium brevicornum (Berberidaceae). A genomic DNA enrichment protocol was used to isolate microsatellite loci and polymorphism was explored
using 38 individuals from one natural population. The observed number of alleles ranged from 2–14. The ranges of observed
and expected heterozygosity were 0.00–0.83 and 0.15–0.88, respectively. In addition, successful cross-species amplification

We developed six new microsatellite markers containing tetranucleotide repeat motifs (GATA/CTAT) for Lanyu scops owl (Otus elegans botelensis) from an enriched partial library. All these loci are polymorphic and conform to Hardy–Weinberg equilibrium. We cross-species
tested these and 12 other microsatellite primer pairs previously developed from O. elegans on four other species of owls (O. lettia, O. spilocephalus, O. scops, and Ninox scutulata). Results showed that the degree of polymorphism decreased with increasing phylogenetic distance to O. elegans. Most loci (66.7, 83.3, and 100%) were polymorphic in the three Otus owls but only five (27.8%) were polymorphic in N. scutulata. These microsatellites should be very useful genetic markers in studying the mating system, population genetics, and conservation

We report on the isolation and characterization of nine microsatellite markers in the takin (Budorcas taxicolor) from genomic DNA-enriched libraries. Twenty-eight microsatellites were screened from the libraries, and nine of the screened
microsatellites were polymorphic. The number of observed alleles for each locus in 28 individuals ranged from two to seven,
and the expected and observed heterozygosity was 0.105–0.758 and 0.071–0.821, respectively. Four loci (TK01, TK02, TK04 and
TK08) of nine deviated from Hardy-Weinberg expectation and no significant linkage association was found among all these loci.
These microsatellite markers provide useful tool for population genetic studies of the takin. 相似文献

Barfin flounder (Verasper moseri) and spotted halibut (Verasper variegatus) are two commercially important flatfish species in the Northeast Asia. In the present study, we reported polymorphic microsatellite
markers in V. moseri and V. variegatus by the cross-species amplification of microsatellite primers developed previously in two other related marine fish species.
A total of 244 polymorphic microsatellite markers were selected for cross-species amplification in V. moseri and V. variegatus, of which 182 markers deriving from Atlantic halibut (Hippoglossus hippoglossus) and 62 markers deriving from Japanese flounder (Paralichthys olivaceus). A sample of 10 individuals were detected. As a result, a total of 67 loci showed polymorphisms in V. moseri, and 62 loci showed polymorphisms in V. variegatus, with the observed number of alleles per locus ranging from two to five in V. moseri, and from two to seven in V. variegatus, respectively. This paper provided more candidate microsatellite markers which could be useful for construction of genetic
linkage maps, evaluation of population genetic structure and stock management of V. moseri and V. variegatus. 相似文献

We isolated 23 polymorphic microsatellite loci of tetranucleotide, dinucleotide or compound repeat motif in the black-faced
spoonbill (Platalea minor). In a panel of up to 20 individuals, number of alleles per locus ranged from two to 19 (mean = 8) and heterozygosity from
0.278 to 0.950 (mean = 0.606). Allele frequencies of five loci deviated from that expected under Hardy–Weinberg Equilibrium,
and one of them was likely Z-linked. Excluding these loci, the remaining 18 loci should provide a sufficient set of molecular
markers for use in ecological and conservation genetic investigation of this globally endangered wader species, and cross-species

New microsatellite loci for Tawny Pipit Anthus campestris were isolated from a genomic library. We were able to unambiguously score six loci: two were dinucleotide, one trinucleotide,
two tetranucleotide and one pentanucleotide that turned out to be sex-linked. Four out of six loci were polymorphic with 7–23
alleles in our population and an observed heterozygosity ranging between 0.286 and 0.936. Cross-utility of these markers was
tested in other 17 steppe-bird species of six families. In addition, 16 microsatellite loci developed for other species were
tested for cross-species amplification in A. campestris. Eight microsatellite markers were successfully amplified; seven of them were polymorphic with 2–43 alleles and an observed
heterozygosity of 0.040–0.863. Overall, 14 functional locus markers have been characterized for A. campestris that could be useful for future studies of paternity, genetic variability and population structure. 相似文献

In this study, twelve polymorphic microsatellite markers were developed for Coilia ectenes (synonym C. nasus). In a sample of 30 C. ectenes individuals in a Wuhan population from Yangtze River, a total of 78 alleles were detected, and the number of alleles per
locus ranged from 3 to 12. The observed and expected heterozygosities ranged from 0.100 to 1.000 and from 0.098 to 0.899,
respectively. Six loci significantly deviated from the Hardy-Weinberg equilibrium after Bonferroni correction (P < 0.004), and no significant linkage disequilibrium between pairs of loci was found. The applicability of these markers in
a closely related species, C. mystus, was evaluated by cross-species amplification. These microsatellite markers will be useful in studies on population genetics,

Random amplified polymorphic DNA (RAPD) and microsatellite markers were applied to evaluate the genetic variation in endemic
and endangered yellow catfish, Horabagrus
brachysoma sampled from three geographic locations of Western Ghat, South India river systems. In RAPD, of 32 10-mer RAPD primers screened
initially, 10 were chosen and used in a comparative analysis of H. brachysoma collected from Meenachil, Chalakkudy and Nethravathi River systems. Of the 124 total RAPD fragments amplified, 49 (39.51%)
were found to be shared by individuals of all 3 populations. The remaining 75 fragments were found to be polymorphic (60.48%).
In microsatellites, six polymorphic microsatellite loci were identified by using primers developed for Pangasius
hypophthalmus, Clarias
macrocephalus and Clarias
gariepinus. The identified loci were confirmed as microsatellite by sequencing after making a clone. The nucleotide sequences of 6 loci
were published in NCBI genbank. The number of alleles across the six loci ranged from 4 to 7 and heterozygosities ranged from
0.07 to 0.93. The mean number of alleles and effective number of alleles per locus were 5.00 and 3.314, respectively. The
average heterozygosity across all investigated samples was 0.72, indicating a significant deficiency of heterozygotes in this
species. RAPD and microsatellite methods reported a high degree of gene diversity and genetic distances depicted by UPGMA
