益生元对抗生素引起的肠道菌群失调的作用   总被引:1,自引:0,他引:1  
目的 评价益生元菊糖和大蒜多糖在治疗肠道菌群失调中的作用.方法 72只Balb/c小鼠随机分为6组,随机取出12只作为正常对照组,其余灌胃盐酸林可霉素和头孢曲松钠混合抗生素溶液3d,建立小鼠肠道失调模型后,以生理盐水灌胃作为自然恢复组,其余4组分别灌胃菊糖、低剂量大蒜多糖、高剂量大蒜多糖以及丽珠肠乐5d.受试小鼠无菌取粪,观察各组肠道菌群的数量变化、测量血清IgG,粪便内毒素含量、有机酸含量变化.结果 菊糖和大蒜多糖高剂量灌胃能加速恢复抗生素导致的小鼠肠道菌群失调,这两种益生元对内毒素的清理、免疫力的恢复也有积极的效果,效果均优于丽珠肠乐和自动恢复.结论 益生元有加快肠道菌群恢复和降低抗生素的破坏效果,有益于小鼠肠道健康.  相似文献

目的 探讨香菇多糖对微生态失调小鼠肠道菌群及免疫功能的调节作用.方法 经盐酸林可霉素灌胃建立肠道微生态失调小鼠模型,香菇多糖灌胃治疗,同时设正常对照组、自然恢复组和丽珠肠乐组.7d后处死各组小鼠,进行肠道菌群定量、免疫器官体重及其淋巴细胞转化率检测.结果 用香菇多糖对肠道微生态失调小鼠进行治疗后,小鼠肠道双歧杆菌、乳酸杆菌数量显著增加,而肠杆菌和肠球菌的数量显著降低;脾脏指数明显增加,对胸腺指数无影响;显著增强了淋巴细胞转化率.结论 盐酸林克霉素灌胃能诱导微生态失调小鼠模型的有效建立.香菇多糖能调整小鼠肠道菌群及免疫功能.  相似文献

目的应用复方树舌液作为微生态调节剂对肠道微生态失调小鼠进行调节。方法用盐酸林可霉素造成菌群失调模型,用复方树舌液进行治疗,在治疗期间分别检测各组小鼠的肠道优势菌群、乙酸、内毒素含量及肝脏肠杆菌易位。结果小鼠肠道菌群失调得到恢复、小鼠肠道内乙酸含量增加、内毒素含量下降、易位至肝脏的肠杆菌数量减少,与自然恢复组相比,差异有统计学意义(P〈0.05),树舌组优于丽珠肠乐组。结论复方树舌液作为益生元对小鼠肠道菌群失调具有调节作用。  相似文献

目的探讨中药马齿苋多糖对衰老小鼠微生态调节作用。方法皮下注射D-半乳糖建立衰老小鼠模型,用马齿苋多糖进行治疗,同时设正常对照组、阳性对照组和阴性对照组,用药30d后处死小鼠,进行肠道微生态指标检测。结果用D-半乳糖造模以后,小鼠肠道双歧杆菌及乳酸杆菌数量降低(P〈0.05),而肠杆菌及肠球菌数量增加说明模型建立,血内毒素含量也升高(P〈0.05)。用马齿苋治疗后,益生菌数量升高而肠杆菌和肠球菌数量减少,血内毒素含量也降低(P〈0.05)。结论马齿苋多糖对衰老小鼠的肠道菌群和血内毒素具有调节作用。  相似文献

目的研究金银花水提取物作为微生态调节剂对肠道微生态失调大鼠肠道菌群的调整作用。方法结扎大鼠胆总管造成肠道菌群失调模型后分别以金银花、丽珠肠乐、金银花与丽珠肠乐合剂、生理盐水灌胃,于灌胃4d后测定各组大鼠肠道菌群组分、乙酸含量及肝脏中肠杆菌易位情况。结果大鼠肠道菌群失调得到恢复,肠道内乙酸含量增加,易位至肝脏的肠杆菌数量减少,与自然恢复组相比,差异有统计学意义(P〈0.05),金银花与丽珠肠乐合剂组效果最佳。结论金银花水提物作为益生元对大鼠肠道菌群失调具有调整作用。  相似文献

目的探索蒲公英多糖对小鼠肠道微生态的调节作用。方法测定蒲公英多糖总糖含量及单糖组成。将小鼠分为正常组、模型组、阳性组和给药组(A1~A7),使用林可霉素灌胃制备肠道菌群失调模型,观察蒲公英多糖对小鼠一般状况、体重、肠道菌群、血清内毒素、小肠黏液s Ig A和血清IL-2的影响。结果蒲公英7组多糖中,相对分子质量100 000和6 000~10 000部分占总糖比例最高(86.40%)。经蒲公英多糖实验性治疗后,小鼠一般状况改善,体重有所增加,小鼠肠道内双歧杆菌和乳酸杆菌数量增加,肠杆菌和肠球菌数量减少。与模型组比较,给药组外周血内毒素含量减少(P0.01)、小肠黏液s Ig A和血浆IL-2含量均增加(P0.05),其中尤以A1、A2、A6组增加明显(P0.01)。结论蒲公英多糖能够改善林可霉素致小鼠肠道菌群失调,具有微生态调节作用。  相似文献

目的探讨蒙古黄芪2种不同成分提取物对肠道微生态失调小鼠的调节作用。方法从60只昆明种清洁级小鼠中随机取出10只作为正常对照组,其余50只每天灌胃0.3 g/ml的盐酸林可霉素,每日2次,每次0.3 ml,连续3 d,诱导小鼠肠道微生态失调,正常对照组以等量生理盐水处理。第4天除正常对照组外,所有小鼠均灌胃造成肠道微生态失调模型,将模型小鼠随机分为4组:黄芪多糖组、黄芪皂苷组、丽珠肠乐组和自然恢复组,并灌胃相应浓度药物进行调整治疗,自然恢复组以等量生理盐水处理,连续7 d。结果与自然恢复组相比,黄芪多糖、黄芪皂苷2种药物均能扶植模型小鼠肠道正常菌群生长,提高乙酸含量,降低内毒素水平,有效控制肠杆菌向肝脏的易位(P0.05);黄芪多糖的各项检测指标与其他组相比,差异有显著性,有统计学意义(P0.05)。结论蒙古黄芪不同成分提取物对肠道微生态失调小鼠均具有一定的调节作用,黄芪多糖的疗效可能优于黄芪皂苷。  相似文献

目的研究应用马齿苋多糖对肠道微生态失调小鼠进行调整治疗,达到从微生态学角度防治感染的目的。方法应用林可霉素灌胃建立肠道微生态失调小鼠模型,然后用马齿苋多糖进行治疗,同时设正常对照组、阳性对照组和阴性对照组,于给药7 d后处死小鼠,进行肠道菌群定量、肠内容物挥发性脂肪酸检测及肠黏膜电镜观察。结果林可霉素灌胃3 d后,小鼠肠道菌群失调,肠内容物挥发性脂肪酸含量明显下降,肠黏膜损伤严重。持续7 d治疗后,治疗组小鼠肠道双歧杆菌和乳酸杆菌数量明显上升,肠内容物挥发性脂肪酸含量明显上升,损伤的肠黏膜基本修复。结论应用林可霉素可以成功建立肠道微生态失调动物模型;马齿苋多糖具有扶植肠道正常菌群生长,调整菌群失调,防治感染的作用,是理想的中药微生态调节剂。  相似文献

为了探讨铁皮石斛多糖对脾虚便秘小鼠免疫、肠道微生物及酶活性的影响,明确多糖对脾虚便秘的治疗作用,通过灌胃番泻叶水煎液7 d后控制饮食,饥饱失常8 d,建立小鼠脾虚便秘模型。造模成功后,正常组和模型组灌胃等量无菌水,治疗组灌胃铁皮石斛多糖溶液,分析小鼠体重、脾脏系数、胸腺系数、肠道菌群和酶活性的变化情况。结果表明,模型组的小鼠体重、体重变化率、脾脏指数、胸腺指数、五类可培养微生物(细菌总数、大肠杆菌、乳酸菌、双歧杆菌和真菌)数量与正常组比较差异显著(P0.01或P0.05)。治疗完成后,小鼠体重和体重变化率显著增高,高于正常组(P0.05);小鼠脾脏指数和胸腺指数均有不同程度恢复,接近正常组(P0.05);大肠杆菌大量得到恢复,但远没有达到正常组水平(P0.01);乳酸菌和双歧杆菌明显地减少,但总数显著超过正常组(P0.01);木聚糖酶和蛋白酶接近正常组,淀粉酶显著增加,纤维素酶显著减少(P0.01)。说明铁皮石斛多糖对增强机体免疫力、调控肠道微生态平衡和肠道酶活性方面有一定的促进作用,能优化肠道环境,改善脾虚便秘症状。  相似文献

目的了解玉米紫色植株花色苷色素对小鼠肠道菌群的影响。方法按中华人民共和国卫生部《保健食品检验与评价技术规范》中的动物实验调节肠道菌群功能的检验方法,观察玉米紫色植株色素对小鼠肠道中肠杆菌、乳杆菌、双歧杆菌数量的影响。结果灌服后小鼠肠道乳杆菌、双歧杆菌数量变化差异均无显著性,低剂量组肠扦菌数量明显低于灌服前(P〈0.05),其他组肠杆菌数量变化差异无显著性。结论玉米紫色植株色素对小鼠肠道菌群可能有影响。  相似文献

In the present study the effect of garlic, in a form more similar to how most people eat garlic, on lipid and antioxidant metabolism in rats was investigated. The antioxidant activity was determined by the efficacy to scavenge 2, 2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) derived radicals in garlic samples. The highest results were estimated in aqueous fraction in comparison with other extracts divided on the basis of polarity. Wistar male rats were randomly divided into 10 diet groups, each with seven animals. The groups were named: Control, RG (raw garlic), BG (boiled garlic for 20 min), AERG (aqueous extract of raw garlic), AEBG (aqueous extract of boiled garlic), Ch (Cholesterol), Ch/RG, Ch/BG, Ch/AERG and Ch/AEBG. All experimental diets were supplemented with 25 mg of lyophilized garlic/kg body weight obtained from raw, boiled and their aqueous extracts over a period of 30 days.Serum lipid (total cholesterol, LDL-cholesterol and triglycerides) concentrations were higher in all groups fed cholesterol (Ch); however, the increase was significant only in Ch group, without garlic supplementation. In groups of rats fed diets with cholesterol, garlic samples significantly hindered the rise of TC and LDL-C (P < 0.05). A significant increase (P < 0.05) in the plasma antioxidant activity was registered in experimental groups of rats fed cholesterol-free diets supplemented with garlic; oppositely, a significant decrease was only in group of rats given food containing cholesterol without garlic.The protein spectra has shown that during short boiling some proteins change their functional properties such as solubility and mobility, resulting in a number of protein bands in SDS-electrophoresis.

Conclusions

Raw and boiled garlic improved plasma lipid metabolism and plasma antioxidant activity in an experiment on rats. Thus, dietary hypolipidemic garlic was effective in reducing the oxidant stress, which was indicated by an increase of antioxidant activity and a decrease of lipids in the rats' blood. It was found that garlic boiled for 20 min has the same bioactivity as raw garlic in its antioxidant and protein spectra. Therefore it should be added at this time to foods. The selenium and copper content of raw garlic is not altered by boiling. The protein electrophoretic pattern of raw garlic is altered by boiling.  相似文献

The purpose of this study was to determine the effects of dietary garlic powder on diethylnitrosamine (DEN)- induced hepatocarcinogenesis and cytochrome P450 (CYP) enzymes in weaning male Sprague-Dawley rats by using the medium-term bioassay system of Ito et al. The rats were fed diets that contained 0, 0.5, 2.0 or 5.0% garlic powder for 8 weeks, beginning the diets with the intraperitoneal (i.p.) injection of DEN. The areas of placental glutathione S-transferase (GST-P) positive foci, an effective marker for DEN-initiated lesions, were significantly decreased in the rats that were fed garlic powder diets; the numbers were significantly decreased only in the 2.0 and 5.0% garlic-powder diets. The p-Nitrophenol hydroxylase (PNPH) activities and protein levels of CYP 2E1 in the hepatic microsomes of the rats that were fed the 2.0 and 5.0% garlic powder diet were much lower than those of the basal-diet groups. Pentoxyresorufin O-dealkylase (PROD) activity and CYP 2B1 protein level were not influenced by the garlic-powder diets and carcinogen treatment. Therefore, the suppression of CYP 2E1 by garlic in the diet might influence the formation of preneoplastic foci during hepatocarcinogenesis in rats that are initiated with DEN.  相似文献

Inulin, a fructooligossacharide, is a prebiotic that plays an important role in the immune function in mammals, but it has never been assayed in other vertebrate groups. Thus, we have studied the inulin effects on the gilthead seabream (Sparus aurata L.) innate immune response both in vitro and in vivo. For the in vitro study, head-kidney leucocytes were incubated with inulin (ranging from 0 to 1000 microg ml(-1)) for 30, 90, 180 and 300 min and 24h and any effect was observed on leucocyte viability or the main innate cellular immune responses (leucocyte peroxidase, phagocytic, respiratory burst and natural cytotoxic activities). For the in vivo study, seabream specimens were fed for 1 or 2 weeks with a commercial diet supplemented with inulin: 0 (control), 5 or 10 g inulin kg(-1) diet (0.5 and 1%, respectively). Inulin produced a significant inhibition in phagocytosis and respiratory burst in leucocytes from specimens fed diets containing 0.5% or 1% of inulin for 1 week. Based on the present results, inulin does not seem to be a good immunostimulant for seabream, though its effects in other species and combined with other immunostimulans (i.e. probiotics) might be of great interest.  相似文献

The influence of the cloned-cattle meat diets upon reproduction in mammals was rarely studied. This study was performed to analyze the effects of the diets containing cloned-cattle (Korean native beef, Hanwoo) meat on the reproductive physiology in rats. The male and female rats were fed with the diets containing 5% or 10% of normal- (N-5 or N-10) or cloned- (C-5 or C-10) cattle meat during test periods. The rats fed with commercial pellets were used as control. Lower food consumption in normal- and cloned-cattle meat diet groups is detected in both male and female rats compared with that of control (P < 0.05, 0.01 and 0.001). No signs of cloned-cattle meat diets on male reproductive parameters are found in all groups, except for lower sperm deformity in C-5 group (P < 0.05) and higher testosterone concentration in C-10 group (P < 0.05), respectively. There are no significant test substance-related differences of Caesarean section and delivery in dams and external examination and physiological development test in neonate compared with control and normal meat groups. Based on these results, it can be postulated that there are no obvious negative effects on the reproductive physiology in rats fed with cloned-cattle meat diets compared to their comparators.  相似文献

We explored whether bifidobacteria and lactobacilli numbers and other selected bacteria in the upper intestine and the caecum of growing pigs were affected by diet and intake of inulin. Starting at two weeks after weaning (28 d) 72 pigs were fed two types of diets (wheat/barley (WB) or maize/gluten (MG)), without or with 3% inulin (WB + I, MG + I) for three and six weeks. Intestinal bacteria were quantified by fluorescence-in-situ-hybridization (n = 8/group). Duration of feeding had no effect on the variables tested, so data for both periods were pooled. Gastric total bacteria amounted to log(10) 7.4/g digesta. Bifidobacteria were detected in stomach and duodenum two weeks after weaning and disappeared thereafter. In jejunum and caecum bifidobacteria were present at a level of log(10) 7.0/g digesta. Inulin did not alter numbers of lactobacilli, bifidobacteria, enterococci, enterobacteria and bacteria of the Clostridium coccoides/Eubacterium rectale-group. Inulin disappearance in stomach plus jejunum was higher with the MG diet (73.7 vs. 60.7%, p = 0.013). Caecal acetate was lower in inulin-supplemented diets (p < 0.05) whereas propionate and butyrate were higher in pigs fed the WB diets (p < 0.05). With the WB diet total caecal short chain fatty acids concentration was higher which resulted in a lower pH value (p < 0.05).  相似文献

The edible brown seaweed sugar Kombu (Saccharina latissima) is a good source of dietary fiber (DF) and associated compounds. Besides it presents antioxidant capacity in vitro due to their sulfated polysaccharides and polyphenols. The effect of a DF-rich sugar Kombu diet on biochemical parameters and antioxidant and prebiotic effects in healthy rats was evaluated. Thus, rats were fed either a basal diet or a supplemented one with 10 % sugar Kombu for 4 weeks. Several health-promoting effects were found such as a decrease in triglycerides (TGL) and uric acid (UrA), and an increase in antioxidant status both in serum and cecum. Regarding prebiotic effect, higher cecum weight and total short chain fatty acid (SCFA) content were evidenced in the seaweed-fed group, without significant differences on total bacterial count of feces. Sugar Kombu and sulfated polysaccharides from its DF could be used as functional ingredients for further nutraceutical applications.  相似文献

This study describes the effects of mixtures of xanthan gum and galactomannan, guar gum, or locust bean gum, on the lipids in plasma and liver in non-diabetic and diabetic rats. Non-diabetic rats were fed cholesterol-free diets with 3% guar gum, locust bean gum, or xanthan gum (3G, 3L, and 3X), or a mixture of xanthan gum and guar gum or locust bean gum (1:2, w/w) (2G1X, 2L1X) for 2 weeks. Rats fed diets not containing these polysaccharides were used as controls. The total cholesterol in plasma and the triacylglycerol in liver were significantly lowered in rats fed the 2G1X diet. The 3G, 3X, 3L, and 2L1X diets showed no significant effect on the total cholesterol and triacylglycerol in plasma and liver. In the streptozotocin-induced (STZ) diabetic rats, the total cholesterol in plasma was lowered in rats fed the 3G, 3X or 2G1X diet for 4 weeks, and the 2G1X diet was more effective than the 3G and 3X diets. The triacylglycerol in plasma in STZ diabetic rats was also significantly lowered by the 2G1X diet. These results showed that a mixture of xanthan gum and guar gum has an improved hypolipidemic effect on non-diabetic and STZ diabetic rats. The effects of the 2G1X diet on the diabetic symptoms in STZ diabetic rats, suppression of food and water intakes, decrease in glucose in urine, and lowering of plasma glucose, were also observed.  相似文献

Massive, toxic doses of vitamin D have been shown to cause nephrocalcinosis in rats, but the effect of this vitamin within its range of fluctuation in commercial rat diets was unknown. Therefore, in two experiments with young female rats, the effect on nephrocalcinosis of a moderately increased level of vitamin D in the diet was studied, that is 5000 IU/kg versus the recommended concentration of 1000 IU/kg. This was done using purified diets with 0.5% (w/w) calcium and 0.04% magnesium containing either 0.2 or 0.6% phosphorus (P). Rats fed the diets containing 0.6% P showed severe kidney calcification compared to those fed the 0.2%-P diets. The level of vitamin D in the 0.2 and 0.6%-P diets did not affect kidney calcification. Bone density was increased after feeding diets containing 5000 instead of 1000 IU of vitamin D/kg. This study suggests that, within 28 days, a moderate increase of the amount of vitamin D in the diet has no influence on the development of kidney calcification. This in turn suggests that the variation in nephrocalcinosis severity and incidence seen in practice in rats fed different commercial diets is unlikely to be related to the different vitamin D concentrations in these diets. However, in rats fed such diets bone metabolism may be influenced differently.  相似文献
