In continuation of our work on the conformational analysis of succinic acid (SA) and maleic acid (MA) in different solvents, we present here the experimental dielectric and IR and also the ab initio Hartree–Fock calculations of the two dicarboxylic acids in tetrahydrofuran (THF). The dielectric measurements are carried out at microwave X-band frequency of 9.7 GHz and the calculations are performed at STO-3G and 6-31G(d) basis sets. The dielectric data and the dipole moment determined experimentally are compared with the dipole moment determined from the conformal analysis. It is seen that the dielectric properties of SA/MA in THF are much different from that of SA/MA in 1-4, dioxane (1-4D) that we had reported previously. The IR spectra of SA–THF system is also reported here. The present study indicates the possible formation of nano-clusters of SA/MA in THF due to incomplete solvation by THF.  相似文献

Fatty acid amide hydrolase (EC 3.5.1.4.) is the enzyme responsible for the rapid degradation of lipid-derived chemical messengers such as anandamide, oleamide, and 2-arachidonoylglycerol. The pharmacological characterization of this enzyme in vivo has been hampered by the lack of selective and bioavailable inhibitors. We have developed a simple, radioactive, high-throughput-compatible assay for this enzyme based on the differential absorption of the substrate and its products to activated charcoal. The assay was validated using known inhibitors. It may be applied for the identification of new inhibitors from a compound library.  相似文献

The three-dimensional (3D) model of the human fatty acid amide hydrolase (hFAAH) was constructed based on the crystal structure of the rat FAAH (PDB code 1MT5) in complex with a substrate using Modeller9v2 program. With the aid of molecular mechanics and molecular dynamics method, the last model was obtained and further assessed by Profile-3D, Prosa2003 and Procheck, which confirms that the refined model is reliable. Furthermore, the docking results of propofol and its structural analogue into the active site of hFAAH indicate that 2,6-di-sec-butyl phenol is a more preferred ligand than others, which is in good agreement with the experimental results. From the docking studies, we also suggest that Phe192, Ile238, Thr377, Leu380, Phe381, Phe388 and Leu404 in the hFAAH are seven important determinant residues in binding as they have strong van der Waal interactions with the ligand.  相似文献

Polycarboxylic acids have been used as nonformaldehyde crosslinking agents for cotton with sodium hypophosphite (NaH2PO2) as the catalyst to replace the formaldehyde-based dimethyloldihydroxyethleneurea (DMDHEU). Maleic acid (MA), an α, β-unsaturated bifunctional carboxylic acid, can esterify cotton but is not able to form crosslinking between two cellulose molecules by itself. In this research, we discovered that the wrinkle resistance of the cotton fabric treated with MA and NaH2PO2 was significantly increased and phosphorus was bound to cotton when the treated fabric was exposed to temperatures higher than that required for esterification of cotton by MA. Elevation of the fabric wrinkle resistance and increase in quantity of the phosphorus bound to cotton had similar dependency on curing temperature, on MA concentration, and on NaH2PO2 concentration. All the data support the hypothesis that H-P-(residual of NaH2PO2) added to >CC< of the MA already bound to cotton by esterification, thus forming a new crosslink between two cotton cellulose molecules. The cotton fabrics treated by MA/NaH2PO2 showed fabric wrinkle resistance similar to that treated with DMDHEU, but the breaking strength and tearing strength of the MA-treated cotton fabrics were significantly improved.  相似文献

A new nano-sized lead(II) coordination polymer of maleic acid (H2Mal), [Pb(μ7-Mal)]n (1), has been synthesized by sonochemical method and characterized by scanning electron microscopy, X-ray powder diffraction, elemental analyses and IR spectroscopy. The compound 1 was structurally characterized by single-crystal X-ray diffraction. Thermal stability of nano and bulk samples of compound 1 were studied and compared with each other. After calcination of nano-sized compound 1 at 600 °C, pure phase micro-sized lead(II) oxide has been produced.  相似文献

A new coordination polymer, [Zn2(mal)(1,10-phen)Cl]n (1), (mal = malate, 1,10-phenanthroline), has been synthesized with malic acid and fumaric acid which are generated from maleic acid under hydrothermal reactions. At about the same condition, we get [Cd(fma)(2,2′-bpy)(H2O)]n (2) (fma = fumarate, 2,2′-bpy=2, 2′-bipyridine). The diverse products illustrate that the carbon-carbon doublebond of the maleic acid has two kinds of reaction trends under different conditions. Complex 1, which displays a two-dimensional (4, 8) lattice-type network, is formed from Zn and maleic through the addition reaction with water molecule. If the Zn is changed by Cd, at the same reaction condition with 1, a two-dimensional supramolecular network complex 2 is formed through the conformation transform reaction. To our knowledge, a lot of coordination polymers have been constructed from malic acid and fumaric acid directly; however, these kinds of complexes have seldom been synthesized from maleic acid under hydrothermal reaction. As is known, the rigid carbon-carbon double bond makes maleic acid lead to some unique structural features which the saturated aliphatic acid does not possess. To illustrate this clearly, a simple one dimensional complex 3, [Cd(glut)(1,10-phen)(H2O)]n (glut = glutarate), is synthesized. Furthermore, complex 1 and complex 3 exhibit intense photoluminescent property at room temperature.  相似文献

pH-sensitive liposomes are designed to undergo acid-triggered destabilization. First generation pH-sensitive liposomes, based on the cone-shaped lipid dioleoylphosphatidylethanolamine (DOPE), have been shown to lose fusogenicity in the presence of serum. Here, we report the design and evaluation of novel serum-resistant pH-sensitive liposome formulations that are based on the composition of egg phosphatidylcholine (PC), cholesteryl hemisuccinate (CHEMS), oleyl alcohol (OAlc), and Tween-80 (T-80). When loaded with the fluorescent probe calcein, these liposomes exhibited excellent stability at pH 7.4 and underwent rapid destabilization upon acidification as shown by calcein dequenching and particle size increase. Adjusting the mole percentages of T-80 and OAlc in the formulation could regulate the stability and pH-sensitive properties of these liposomes. Liposomes with a higher T-80 content exhibited greater stability but were less sensitive to acid-induced destabilization. Meanwhile, formulations with a higher OAlc content exhibited greater content release in response to low pH. The pH-triggered liposomal destabilization did not produce membrane fusion according to an octadecylrhodamine B chloride (R18) lipid-mixing assay. Compared to DOPE-based pH-sensitive liposomes, the above formulations showed much better retention of their pH-sensitive properties in the presence of 10% serum. These liposomes were then evaluated for intracellular delivery of entrapped cytosine-β-d-arabinofuranoside (araC) in KB human oral cancer cells, which have elevated folate receptor (FR) expression. The FR, which is amplified in many types of human tumors, has been shown to mediate the internalization of folate-derivatized liposomes into an acidic intracellular compartment. FR-targeted OAlc-based pH-sensitive liposomes, entrapping 200 mM araC, showed ∼17-times greater FR-dependent cytotoxicity in KB cells compared to araC delivered via FR-targeted non-pH-sensitive liposomes. These data indicated that pH-sensitive liposomes based on OAlc, combined with FR-mediated targeting, are promising delivery vehicles for membrane impermeable therapeutic agents.  相似文献

Starch/poly(butylene adipate-co-terephthalate) films were obtained by one-step reactive extrusion using maleic anhydride (MA) and citric acid (CA) as compatibilizers. The mechanical, structural, optical and barrier properties of the films were analyzed when glycerol (GLY), CA and MA were added to the starch/PBAT (55:45, w/w) according to mixture design. FTIR analysis showed that CA and MA were able to promote esterification/transesterification reactions and that CA induced them more efficiently. When a greater proportion of compatibilizer (1.5 wt%) was used, the resulting films were more opaque and had a greater tensile strength. A greater proportion of GLY (10.0%, w/w) improved the elongation at the break of the films. The barrier properties to water vapor of the films were improved by high levels of CA (1.5 wt%) and intermediate levels of GLY (9.25 wt%). The inclusion of compatibilizers resulted in blends with improved properties, representing a potential replacement for non-biodegradable films.  相似文献

Anandamide and other fatty acid amides are metabolised by the enzyme fatty acid amide hydrolase (FAAH), which thereby regulates their endogenous levels. Here we demonstrate that cultured rat cortical microglia express FAAH at low levels. The potent FAAH inhibitor URB597 reduced the LPS stimulated microglial expression of cyclo-oxygenase 2 and inducible nitric oxide, with concomitant attenuation of the release of PGE2 and NO. Additional of supplemental exogenous anandamide did not increase the magnitude of attenuation of mediator release. The effect of URB597 on LPS stimulated PGE2 release was not blocked by selective CB1 or CB2 receptor antagonists.  相似文献

A novel fluorescent assay to continuously monitor fatty acid amide hydrolase (FAAH) activity that is simple, sensitive, and amenable to high-throughput screening (HTS) of compound libraries is described in this article. Stable Chinese hamster ovary (CHO) cell lines expressing either human FAAH or an inactive mutant, FAAH-S241A, were established. Arachidonyl 7-amino, 4-methyl coumarin amide (AAMCA), a novel fluorogenic substrate for FAAH, was designed and synthesized. FAAH catalyzes the hydrolysis of AAMCA to generate arachidonic acid and a highly fluorescent 7-amino, 4-methyl coumarin (AMC). The assay was done at 25 degrees C by incubating whole cell or microsomal preparations from FAAH-expressing cells with AAMCA. Release of AMC was monitored continuously using a fluorometer. Microsomal FAAH catalyzed the hydrolysis of AAMCA with an apparent K(m) of 0.48muM and V(max) of 58pmolmin(-1)mgprotein(-1). The assay is specific for FAAH given that microsomes prepared from cells expressing FAAH-S241A or vector alone had no significant activity against AAMCA. Furthermore, the activity was inhibited by URB-597, an FAAH-specific inhibitor, in a concentration-dependent manner with an IC(50) of 33.5nM. The assay was optimized for HTS and had a Z' value ranging from 0.7 to 0.9. The assay is also compatible with ex vivo analysis of FAAH activity.  相似文献

The aim of this study was to examine the role of fatty acid amide hydrolase (FAAH) on ethanol sensitivity, preference, and dependence. The deletion of FAAH gene or the inhibition of FAAH by carbamoyl-biphenyl-3-yl-cyclohexylcarbamate (URB597) (0.1 mg/kg) markedly increased the preference for ethanol. The study further reveals that URB597 specifically acts through FAAH and that cannabinoid-1 (CB1) receptor is critical for N -arachidonoyl ethanolamide (AEA) mediated ethanol-reinforced behavior as revealed by lack of URB597 effect in both FAAH and CB1−/− mice compared with vehicle-treated −/− mice. The FAAH −/− mice displayed a lower sensitivity to hypothermic and sedative effects to acute ethanol challenge. The FAAH −/− mice also exhibited a reduction in the severity of handling-induced convulsions following withdrawal from chronic ethanol exposure. The CB1 receptor and proenkephalin gene expressions, and CB1 receptor and μ-opioid (MO) receptor-mediated G-protein activation were found to be significantly lower in the caudate-putamen, nucleus accumbens core and shell of FAAH −/− than +/+ mice. Interestingly, the MO receptor-stimulated G-protein signaling was greater in the striatum of FAAH −/− than +/+ mice following voluntary ethanol consumption. These findings suggest that an elevation in the AEA content and its action on the limbic CB1 receptor and MO receptor might contribute to ethanol-reinforced behavior. Treatment with drugs that decrease AEA tone might prove useful in reducing excessive ethanol consumption.  相似文献

Long-chain fatty acid amides are cell-signaling lipids identified in mammals and, recently, in invertebrates, as well. Many details regarding fatty acid amide metabolism remain unclear. Herein, we demonstrate that Drosophila melanogaster is an excellent model system for the study long-chain fatty acid amide metabolism as we have quantified the endogenous levels of N-acylglycines, N-acyldopamines, N-acylethanolamines, and primary fatty acid amides by LC/QTOF-MS. Growth of D. melanogaster on media supplemented with [1-13C]-palmitate lead to a family of 13C-palmitate-labeled fatty acid amides in the fly heads. The [1-13C]-palmitate feeding studies provide insight into the biosynthesis of the fatty acid amides.  相似文献

An inherited deficiency of arylsulfatase A (ASA) causes the lysosomal storage disease metachromatic leukodystrophy (MLD) characterized by massive intralysosomal storage of the acidic glycosphingolipid sulfatide and progressive demyelination. Lyso-sulfatide, which differs from sulfatide by the lack of the N-linked fatty acid, also accumulates in MLD and is considered a key driver of pathology although its concentrations are far below sulfatide levels. However, the metabolic origin of lyso-sulfatide is unknown. We show here that ASA-deficient murine macrophages and microglial cells express an endo-N-deacylase that cleaves the N-linked fatty acid from sulfatide. An ASA-deficient astrocytoma cell line devoid of this activity was used to identify the enzyme by overexpressing 13 deacylases with potentially matching substrate specificities. Hydrolysis of sulfatide was detected only in cells overexpressing the enzyme fatty acid amide hydrolase (FAAH). A cell-free assay with recombinant FAAH confirmed the novel role of this enzyme in sulfatide hydrolysis. Consistent with the in vitro data, deletion of FAAH lowered lyso-sulfatide levels in a mouse model of MLD. Regardless of the established cytotoxicity of lyso-sulfatide and the anti-inflammatory effects of FAAH inhibition seen in mouse models of several neurological diseases, genetic inactivation of FAAH did not mitigate, but rather exacerbated the disease phenotype of MLD mice. This unexpected finding was reflected by worsening of rotarod performance, increase of anxiety-related exploratory activity, aggravation of peripheral neuropathy, and reduced life expectancy. Thus, we conclude that FAAH has a protective function in MLD and may represent a novel therapeutic target for treatment of this fatal condition.  相似文献

A binding assay for human fatty acid amide hydrolase (FAAH) using the scintillation proximity assay (SPA) technology is described. This SPA uses the specific interactions of [3H]R(+)-methanandamide (MAEA) and FAAH expressing microsomes to evaluate the displacement activity of FAAH inhibitors. We observed that a competitive nonhydrolyzed FAAH inhibitor, [3H]MAEA, bound specifically to the FAAH microsomes. Coincubation with an FAAH inhibitor, URB-597, competitively displaced the [3H]MAEA on the FAAH microsomes. The released radiolabel was then detected through an interaction with the SPA beads. The assay is specific for FAAH given that microsomes prepared from cells expressing the inactive FAAH-S241A mutant or vector alone had no significant ability to bind [3H]MAEA. Furthermore, the binding of [3H]MAEA to FAAH microsomes was abolished by selective FAAH inhibitors in a dose-dependent manner, with IC50 values comparable to those seen in a functional assay. This novel SPA has been validated and demonstrated to be simple, sensitive, and amenable to high-throughput screening.  相似文献

The effect of lipopolysaccharide inhalation upon lung anandamide levels, anandamide synthetic enzymes and fatty acid amide hydrolase has been investigated. Lipopolysaccharide exposure produced a dramatic extravasation of neutrophils and release of tumour necrosis factor alpha into the bronchoalveolar lavage (BAL) fluid, which was not accompanied by epithelial cell injury. The treatment, however, did not change significantly the levels of anandamide and the related compound palmitoylethanolamide in the cell-free fraction of the BAL fluid. The activities of the anandamide synthetic enzymes N-acyltransferase and N-acylphosphatidylethanolamine phospholipase D and the activity of fatty acid amide hydrolase in lung membrane fractions did not change significantly following the exposure to lipopolysaccharide. The non-selective fatty acid amide hydrolase inhibitor phenylmethylsulfonyl fluoride was a less potent inhibitor of lung fatty acid amide hydrolase than expected from the literature, and a dose of 30 mg/kg i.p. of this compound, which produced a complete inhibition of brain anandamide metabolism, only partially inhibited the lung metabolic activity.  相似文献
