Phytochemical investigation of the 70% aqueous EtOH extract of Lycopodium complanatum led to six new polyhydroxy serratene triterpenoids (serrat A-F, 16), along with a known analogue (7). Their structures and configurations were elucidated by data analysis of HRESIMS, 1D and 2D NMR, in combination with comparisons of reported experimental spectroscopic data. All the isolates were evaluated cytotoxic activities against HepG2 cells, MCF-7 cells and series human lung cancer cell lines A549, Calu-6, NCI-H441, NCI-H226 and NCI-H1975. The results indicated that certain compounds inhibited proliferation of human cancer cells. Moreover, all compounds possessed selective cytotoxic activities on MCF-7 cells. Further, possible biosynthesis pathways of these compounds were proposed.  相似文献

A new tricoumarin glycoside, triumbelletin-7-O-β-d-glucoside (1) and a new biflavonoid, wikstroflavone A (2), together with two known compounds, wikstaiwanone A (3) and wikstaiwanone B (4), were isolated from the rhizome of Wikstroemia indica. The structures of new compounds were elucidated by extensive spectroscopic techniques (UV, IR, HRESIMS, 1D, 2D NMR and CD), in combination with quantum chemical calculations of 13C NMR and ECD spectra. All isolates were tested for their antineoplastic activities against cancer-derived cell lines HCT116, SW480, U87 and T98G. Compounds 24 exhibited moderate cytotoxic activities to the four cell lines. The flow cytometry assay and western blot analysis revealed that the cytotoxic effects were possibly attributed to the induced apoptotic cell death.  相似文献

Two new prenylated flavonol glycosides, epimedigrandiosides A and B (1 and 2), and 28 previously known compounds including prenylated flavonol derivatives, flavonol glycoside, megastigmanes, phenyl alkanoids, sesquiterpenoid glycoside, lignan, and hexene glucoside were isolated from the methanol extract of Epimedium grandiflorum. Structure elucidation was achieved by means of spectroscopic and spectrometric techniques including 1D and 2D NMR and HRESIMS. The absolute configuration of sugars was determined by chemical methods Structure elucidation of 3‴-carbonyl-2″-β-l-quinovosyl icariin (19) was not previously described, so its 1H and 13C NMR data were reported for the first time. The methanol extract and the isolated compounds were evaluated for their activity towards several targets related to inflammation and metabolic disorder including NF-κB, iNOS, PPARα and PPARγ. Moreover, their cytotoxic activity against four cancer cell lines (SK-MEL, KB, BT-549, SK-OV-3) and two noncancerous kidney cell lines (LLC-PK1 and Vero) were also evaluated.  相似文献

A new sulfated holostane glycoside, leucospilotaside B ( 1 ), together with the two related structurally known compounds holothurin B2 ( 2 ) and holothurin B ( 3 ), was isolated from sea cucumber Holothuria leucospilota collected from the South China Sea. The structure of 1 was elucidated by spectral analysis (1H‐, 13C‐, and 2D‐NMR, ESI‐MS, and HR‐ESI‐MS) and chemical methods. The compounds 1 – 3 possess the same disaccharide moiety, but were different in the side chains of the triterpene aglycone. Compound 1 showed significant cytotoxicities against four human tumor cell lines, HL‐60, MOLT‐4, A‐549, and BEL‐7402.  相似文献

Two new hemiterpene glycosides, named as securiterpenoside B (1), and securiterpenoside C (2), and one new phenolic glycoside, named as securiphenoside A (3) were isolated from the roots of Securidaca inappendiculata Hassk. Their structures were elucidated on the basis of 1D and 2D NMR data and HRESIMS and comparisons with published data. Moreover, compounds 13 were evaluated for cytotoxicities against A549 (Lewis lung cancer), Hela (human cervical cancer) and MCF-7 (human breast cancer) cell lines.  相似文献

Phytochemical investigation of the dichloromethane extract of the dried aerial parts of Corydalis rupestris (Papaveraceae) resulted in the identification of four new isoquinoline alkaloids rupestrines A-D and one known isoquinoline alkaloid, namely, stylopine. The structures of these compounds were characterized by extensive spectroscopic methods including 1D- (1H and 13C) and 2D NMR experiments (COSY, HSQC, HMBC, and NOESY) as well as HRESIMS analyses. In addition, the absolute configurations of rupestrines A-D were determined using modified Mosher’s method. Cytotoxic effects of alkaloids and their interaction with albumin were also investigated in this study.  相似文献

As part of our search of new bioactive saponins from Cameroonian medicinal plants, phytochemical investigation of the roots of Albizia boromoensis led to the isolation of four new oleanane-type saponins, named boromoenosides A–D (1–4). Their structures were established by direct interpretation of their spectral data, mainly HRESIMS, 1D NMR (1H, 13C NMR, and DEPT) and 2D NMR (COSY, NOESY, HSQC and HMBC), and by comparison with the literature data. All isolated saponins were assayed for their cytotoxicity against U-87 MG human glioblastoma cell lines and TG1 glioblastoma stem-like cells with no positive activity detected.  相似文献

As a part of our continuing interest in identifying anticancer drug leads from natural sources, we have investigated the in vitro growth inhibitory effects of the hexane fraction of the root bark of Calotropis procera (Ait) R. Br. (Asclepiadaceae). This study reports the isolation and structure elucidation of four new ursane-type triterpenes named calotroprocerol A (1), calotroproceryl acetate A (2), calotroprocerone A (3) and calotroproceryl acetate B (4) in addition to five known compounds including pseudo-taraxasterol acetate (5), taraxasterol (6), calotropursenyl acetate B (7), stigmasterol (8) and (E)-octadec-7-enoic acid (9). Their structures were established on the basis of 1D and 2D NMR studies (1H–1H COSY, HSQC, and HMBC) and HRMS spectral data. The in vitro growth inhibitory activity of the isolated compounds was evaluated against three human cancer cell lines including the A549 non-small cell lung cancer (NSCLC), the U373 glioblastoma (GBM) and the PC-3 prostate cancer cell lines.  相似文献

Two new rotenoids, named millettiaosas A–B (12), together with four known compounds were isolated from the roots of Millettia speciosa. Their structures were elucidated on the basis of spectroscopic analysis including 1D and 2D NMR techniques and HRESIMS. Evaluation of the two new compounds for cytotoxicity against four human cancer cell lines (MCF-7, HCT-116, A549 and HepG-2) showed moderate activities (10 μM < IC50 < 26 μM).  相似文献

Two new cembranoids, together with fifteen known ones, were isolated from the flowers of Nicotiana tabacum L. The structures of the new compounds were established as (1βH,2E,4αOH,6αOH,7E,10αH,11αH,12βOH)-10,11-epoxy-2,7-cembradiene-4,6,12-triol (1) and (1βH,2E,4αOH,6αOH,7E,10βH,11βH,12αOH)-10,11-epoxy-2,7-cembradiene-4,6, 12-triol (2) by using spectroscopic analysis, including HRESIMS, IR, one- and two-dimensional NMR. A plausible biogenetic relationship of the isolated cembranoids was proposed. The antitumor activities of selected compounds against a panel of three human cancer HepG2, A549 and HCT-116 cell lines were evaluated by the MTT assay. Compound 5 exhibited moderate activity against Hep-G2 cell lines with an IC50 value of 14.38 μM.  相似文献

Phytochemical study on the methanolic extract of Sansevieria cylindrica aerial parts lead to the isolation, characterization and structure elucidation of a new steroidal saponin, 1β-hydroxy-kryptogenin-1-O-α-l-rhamnopyranosyl-(1  2)-α-l-arabinopyranoside (1), a new homoisoflavanone, (3S)-3,7-dihydroxy-8-methoxy-3-(3′,4′-methylenedioxybenzyl) chroman-4-one (2) and the known saponin alliospiroside A (3). To the best of our knowledge, the genin 1β-hydroxy-kryptogenin is reported here for the first time. The structures of the new compounds were determined by UV, IR, EIMS, HRESIMS together with 1D (1H and 13C) and 2D (HSQC and HMBC) NMR spectral analysis. The isolated compounds 1–3 were tested for their radical scavenging activity (DPPH). Compound 2 exhibited activity compared to that of ascorbic acid as a standard. The cytotoxicity of the isolated compounds and the standard doxorubicin was tested against the three human tumor cell lines HT116, MCF-7 and PC-3. The results showed that the isolated compounds were inactive.  相似文献

Nine new 8-O-4′ neolignans, named pinnatifidanin B I–IX (19), together with 9 known analogs (1018) were isolated from the seeds of Crataegus pinnatifida. The structures of 118 were determined by spectroscopic methods, including 1D, 2D NMR, CD and HRESIMS analysis. Compounds 811, 17 and 18 displayed potent cytotoxic activities against human cancer cell lines, and most interestingly, none of the 6 compounds displayed inhibitory activity against human lung cell line (Mrc5). The 6 cytotoxic compounds are considered to be potential as antitumor agents, which could significantly inhibit the cancer cell growth in a dose-dependent manner and are probably safer than positive control drug.  相似文献

Three new cassane furanoditerpenes, phanginins N–P (13), together with four known ones were isolated from the seeds of Caesalpinia sappan. Their structures were elucidated on the basis of spectroscopic analysis including HRESIMS, 1D and 2D NMR techniques. Two new ceramides, irisamides A (1) and B (2), together with a new isoflavone, iridin S (3) have been isolated from the MeOH extract of the rhizomes of Iris germanica L. (Iridaceae). Chemical investigation of the marine sponge Dactylospongia elegans, collected from the South China Sea, afforded three new dimeric sesquiterpene quinones, popolohuanones G – I ( 1 – 3 ), together with two known analogs, popolohuanones B ( 4 ) and C ( 5 ). The new structures were determined by HR‐ESI‐MS and 1D‐ and 2D‐NMR analyses. All the five compounds showed no cytotoxic activity against five human cancer cell lines, while popolohuanone H ( 2 ) showed potent inhibitory activity against IL‐6, an inflammatory cytokine, at the concentration of 10 μm .  相似文献

Euphoroids A–C (13), three new ent-abietane-type diterpenoids, together with ten known analogues (413) were obtained from the roots of Euphorbia ebracteolata. The structures of these compounds were determined by extensive spectroscopic data analysis, including UV, HRESIMS, 1D-, and 2D-NMR data. The inhibitory effects of compounds 113 on human cancer cells were determined using the MTT assay. The results revealed that new compounds 2 and 3 showed moderate cytotoxic activities against human cancer cell lines. Three new thiohydantoins with a 2-thioxoimidazolidin-4-one nucleus, named macathiohydantoins P–R (13), along with one known analog (4), were isolated from the roots of Lepidium meyenii. Their structures were elucidated by means of analysis of extensive spectroscopic data, including 1D NMR, 2D NMR and HRESIMS. The thiohydantoins (14) were isolated as racemic mixtures, which were further separated by chiral semi-preparative HPLC to afford the corresponding enantiomers. Their absolute configuration of C-5 was assigned by comparison of optical rotation values with those reported in the literature. Chemical investigation of the mangrove fungal endophytes, Phomopsis spp. xy21 and xy22, afforded four new cytochalasins, named phomopsichalasins D-G, along with six known analogues. The structures of these cytochalasins were elucidated on the basis of HRESIMS and extensive NMR spectroscopic data. Phomopsichalasins D (1) and E (2) represent the first two 10-phenyl[11]-cytochalasans containing a 12-carboxyl function. All of the isolates were evaluated for their cytotoxicities against eight human cancer cell lines by the MTT method. Phomopsichalasin G (4) exhibited inhibitory activities against HCT-8, HCT-8/T, A549, MDA-MB-231, and A2780 cancer cell lines with IC50 values of 7.5, 8.6, 6.4, 3.4, and 7.1 μM, respectively.  相似文献

Three new flavonoids, 6,7-dimethoxy-4′-hydroxy-8-formylflavon (1), 8-formyl-4′,6,7-trimethoxyflavon (2), 4′,7-dihydroxy-8-formyl-6-methoxyflavon (3), together with fifteen known flavonoids (418) were isolated from the leaves of oriental tobacco (a variety of Nicotiana tabacum L). Their structures were determined by means of HRESIMS, extensive 1D and 2D NMR spectroscopic studies and chemical evidences. The cytotoxicity against five human tumor (NB4, A549, SHSY5Y, PC3, and MCF7) cell lines of compounds 13 were also evaluated. The results showed that compounds 1 and 3 showed high cytotoxicity against PC3 and A549 cell lines with IC50 values of 2.6 and 1.6 μM, respectively.  相似文献

Flower of Chrysanthemum morifolium is widely used in China and Japan as a folk medicine in treatment of many diseases. However, its active compounds remain largely unknown. In the present work, we have isolated, purified and characterized six new compounds (16), including two new arylnaphthalene lignans and four new phenolic glycosides, together with eight known compounds (714), from the flower of C. morifolium. Their structures and absolute configurations were elucidated in detail using 1D and 2D NMR, UV, IR, ORD, HRESIMS and ECD spectrometric data. In addition, compounds 13 possessed the significant neuroprotective activity against hydrogen peroxide-induced neurotoxicity in human neuroblastoma SH-SY5Y cells.  相似文献
