Coronary artery disease (CAD) is a major cause of death in Canada and the United States. Single photon emission computed tomography (SPECT) myocardial perfusion imaging (MPI) is a useful diagnostic test in the management of patients with CAD. The widely used SPECT MPI agents, 99mTc sestamibi and 99mTc tetrofosmin, exhibit less than ideal pharmacokinetic properties with decreasing uptake with higher flows. 123I has a similar energy as 99mTc, an ideal half life, and is readily available from cyclotrons. The objective of this study was to develop an 123I labeled MPI agent based on rotenone, a mitochondrial complex I inhibitor, as an alternative to currently available SPECT MPI agents. Methods: 123I-CMICE-013 was synthesized by radiolabeling rotenone with 123I in trifluoroacetic acid (TFA) with iodogen as the oxidizing agent at 60 °C for 45 min, followed by RP-HPLC purification. The product was formulated in 5% EtOH in 10 mM NaOAc pH 6.5. The inactive analog 127I-CMICE-013 was isolated and characterized by NMR and mass spectrometry, and the structure determined. Micro-SPECT imaging studies were carried out in normal and infarcted rats. Biodistribution studies were performed in normal rats at 2 h (n = 6) and 24 h (n = 8) post injection (p.i.). Results: 123I-CMICE-013 was isolated with >95% radiochemical purity and high specific activity (14.8–111 GBq/μmol; 400–3000 mCi/μmol). Structural analysis showed that rotenone was iodinated at 7′-position, with removal of the 6′,7′-double bond, and addition of a hydroxy group at 6′-position. MicroSPECT images in normal rats demonstrated homogeneous and sustained myocardial uptake with minimal interference from lung and liver. Absent myocardial perfusion was clearly identified in rats with permanent left coronary artery ligation and ischemia-reperfusion injury. In vivo biodistribution studies in normal rats at 2 h p.i. showed significant myocardial uptake (2.01 ± 0.48%ID/g) and high heart to liver (2.98 ± 0.93), heart to lung (4.11 ± 1.04) and heart to blood (8.37 ± 3.97) ratios. At 24 h p.i., the majority of 123I-CMICE-013 was cleared from tissues, and a significant amount of tracer was found in the thyroid, indicating in vivo deiodination of the tracer. Conclusion: 123I-CMICE-013 is a promising new radiotracer for SPECT MPI with high myocardial uptake, very good target to background ratios and favorable biodistribution characteristics.  相似文献

One to four routes of synthesis are described for 8′-hydroxyrotenone, 5′-hydroxyrotenone, two epimers of 6′,7′-dihydro-6′,7′-dihydroxyrotenone, two epimers of 6′,7′-epoxyrotenone and the four rotenolones derived from each of these compounds. The stereochemical relationships are determined, in each case, by chemical interconversion, ORD and monochromatic rotation to assess the absolute configuration of the B/C ring juncture and by IR, MS and NMR for the cis- or trans-nature of this juncture. The new compounds described are useful standards for studies on the metabolites and photodecomposition products of rotenone insecticide chemical.  相似文献

A population of Xiphinema hunaniense Wang and Wu, 1992 with all four juvenile stages was found in the rhizosphere of Pinus sp. in Hangzhou, Zhejiang, China. Morphometrics of 18 females and 35 juveniles of this population are given herein. Detailed morphology and morphometrics of the four juvenile stages are provided. Further comparisons based on morphometrics of the population with previous studies of the females and the first-stage juveniles of X. hunaniense with X. radicicola are given, and morphological variation in X. hunaniense populations are discussed. A revised polytomous key code of Loof and Luc (1990) for X. hunaniense identification is provided, i.e., A1- B4- C4- D4/5- E1- F2(3)- G2- H2-I3- J4- K2- L1. In addition, the sequence of the D2 and D3 expansion region of the 28S rRNA gene was analyzed and compared with sequences of closely related species downloaded from the NCBI database. Cluster analysis of sequences confirmed and supported the species identifications.  相似文献

Chemical investigation of the aerial parts of Oncocalyx glabratus resulted in the isolation of three new flavan derivatives, 5,3′,4′-trihydroxyflavan 7-O-gallate (1), 5,4′-dihydroxyflavan 7-3′-O-digallate (2) and 5,3′-dihydroxyflavan 7-4′-O-digallate (3), named oncoglabrinol A, B and C, respectively, together with four known flavonols, (+)-catechin (4), (+)-catechin-7-O-gallate (5), catechin-7-4′-O-digallate (6A) and catechin-7-3′-O-digallate (6B). The structures of the compounds were established by 1D, 2D NMR and ESI-HRMS spectral analyses. The biological activity of the compounds was tested through a series of in vitro assays designed for determining cytotoxicity, antiviral activity against hepatitis B virus, and antidiabetic activity. All compounds were found non-toxic and showed moderate anti-HBV activity. Compounds 3 and 6 showed dual PPAR agonistic activity while others were not effective.  相似文献

For the investigation of rotenone biosynthesis, acetate-2-14C, mevalonic acid-2-14C lactone and methionine-methyl-14C were administered to Derris elliptica plants, respectively, and the distribution of carbon-14 in the labeled rotenone was determined by degradation. When mevalonic acid-2-14C lactone was incorporated into rotenone, the radioactivity was found equally in the carbons at both C-7′ and C-8′, indicating that these carbons are derived from the carbon-2 of mevalonic lactone. In the case of methionine-methyl-14C about 80% of the total radioactivity was found to enter two methoxyl groups. This result demonstrates that methionine is an efficient precursor of the methoxyl group. Furthermore, it is also suggested that methionine may be a precursor of the carbon at C-6.  相似文献

DEAE-cellulose chromatography, in the presence and absence of Ca2+, of the 16,000g supernatant from bovine carotid artery smooth muscle has been used to separate four different types of cyclic nucleotide phosphodiesterase (3′:5′-cyclic-nucleotide 5′-nucleotidohydrolase, EC 3.1.4.17) activity, designated types A, B, C, and D. Type A is a high affinity, cyclic AMP-specific form of phosphodiesterase (Km = 1.6 μM) and elutes at relatively high ionic strength. Type B is a high affinity (Km = 2 μM), cyclic GMP-specific form which elutes at low ionic strength. Type C is a mixed substrate form, displaying anomalous kinetics for the hydrolysis of both cyclic AMP and cyclic GMP. It elutes from DEAE-cellulose at an ionic strength intermediate to that of types A and B. Type D is also a mixed substrate form of phosphodiesterase. However, its elution pattern from DEAE-cellulose differs, depending on whether Ca2+ is present or not, suggesting a Ca2+-dependent interaction between this enzyme form and the acidic Ca2+-dependent regulator protein (CDR). The hydrolytic activity of type D is stimulated by CDR, and activation requires the simultaneous presence of Ca2+ and CDR. Kinetic analysis of cyclic AMP hydrolysis by type D gives a linear double reciprocal plot; activation has no effect on the Km but increases the velocity approximately sixfold. Activation of cyclic GMP hydrolysis apparently affects both the Km and V. At all concentrations tested, the degree of activation is higher with cyclic AMP than with cyclic GMP. It is suggested that while the activable form of phosphodiesterase may play a relatively minor role in the overall hydrolysis of cyclic nucleotides, Ca2+-dependent activation may have a more important role in regulating the level of cyclic AMP than that of cyclic GMP in vascular smooth muscle.  相似文献

Obesity, insulin resistance, and hyperandrogenism are considered crucial parameters of polycystic ovary syndrome (PCOS) which might be related to vitamin D metabolism. The aim of this study was to investigate the associations between polymorphisms (TaqI and ApaI) in the vitamin D receptor gene (VDR) and PCOS among Egyptian women. We aimed also to elucidate the impact of these polymorphisms on vitamin D level, hormonal and metabolic parameters of PCOS. One hundred and fifty Egyptian women with PCOS and 150 unrelated controls were enrolled in this study. Polymorphisms of VDR Taq-I T/C (rs731236) and Apa-I A/C (rs7975232) gene were genotyped using polymerase chain reaction restriction fragment length polymorphism (PCR–RFLP). Serum 25 hydroxy vitamin D [25(OH) D] levels were measured by high-performance liquid chromatography. PCOS women had significantly lower levels of 25(OH) D compared to healthy women. Our results revealed that Taq-I CC genotype and C allele were associated with increased risk of PCOS, while the Apa-I polymorphism was not. Haplotype Taq-I C/ Apa-I C was associated with a higher PCOS risk more than controls. Moreover, there was a significant decrease of 25(OH) D levels in carriers of haplotype Taq-I C/ Apa-I C (with variant alleles) compared to the non-carriers. Results showed also that there was an obesity- VDR Taq-I genotypes interactions. These results suggested that, VDR Taq-I gene polymorphism is associated with increased risk of PCOS in Egyptian women.  相似文献

The sharptooth catfish Clarias gariepinus is an emerging global invader for which control strategies might include the use of piscicides such as rotenone. Experimental exposure demonstrated that C. gariepinus was less susceptible to rotenone than most other fish species, with unexpected survival observed at rotenone concentrations of 87.5 and 100 µg L?1. C. gariepinus were also observed exhibiting avoidance behaviour to rotenone treated water and were found to be capable of recovering from rotenone exposure. As such, effective eradication might not be attainable even at a dose exceeding 100 µg L?1 with exposures of longer than 24 h. This exposure scenario may pose an unacceptable risk to non-target fauna and highlights the difficulty associated with managing current and future invasions.  相似文献

23 S ribosomal ribonucleic acid gene from the extreme thermophile eubacterium Thermus thermophilus HB8 has been cloned in pBR322, and the nucleotide sequence of region D has been determined, which encompasses 873 nucleotides at the 3′-end of the RNA. We compare the primary and secondary structure of this region with the respective part of the 23 S rRNA from Escherichia coli and Bacillus stearothermophilus. A high level of structural conservation can be observed, throughout the RNA domain, albeit the usage of G/C basepairs is substantial even in comparison with another thermophilic eubacterium B. stearothermophilus. It is surprising that, in contrast to the usage of 3′U-G5′, the occurrence of 3′G-U5′ is comparable in E. coli as well as in B. stearothermophilus and T. thermophilus. Furthermore, it is most remarkable that the use of 3′A-U5′ and 3′U-A5′ is, compared to E. coli, only slightly reduced in B. stearothermophilus, but drastically decreased in T. thermophilus.  相似文献

Phytoplankton monitoring in Wickford Cove, Rhode Island, US (41°34′10.13″N, 71°26′45.76″W), located in Narragansett Bay, detected an unusual species of Alexandrium in the spring of 2009. Thecal plate analysis using brightfield and SEM microscopy revealed a plate morphology consistent with that of Alexandrium peruvianum (Balech and de Mendiola) Balech and Tangen. Molecular analyses indicated that the sequences of the SSU, ITS1, 5.8S, ITS2 and LSU through the D region of the 18S gene were similar to those of A. peruvianum from North Carolina. Toxin analyses of cells brought into culture revealed saxitoxins, gymnodimine and fast-acting spiroimines were present in the cultured clone. Saxitoxins detected included GTX 2, GTX3, B1, STX, C1 and C2. Also present in the Wickford cove isolates of A. peruvianum were 12-methyl gymnodimine and 13-desmethyl spirolide C. A. peruvianum was detected at four sites in lower Narragansett Bay: at two sites in Wickford and two sites in Jamestown, RI. A. peruvianum was observed in the spring of 2009, 2010, 2011 and 2012 at maximum abundance levels ranging from tens of cells per liter to 14,000 cells L−1. The discovery of A. peruvianum in Rhode Island coastal waters, with its potential threat to public health, is notable as it appears to be an emergent bloom species globally. The presence of A. peruvianum in Narragansett Bay is the third confirmed observation of this species on the Atlantic coast of North America. Monitoring efforts in the southern New England region should incorporate measures to detect the presence of A. peruvianum toxins.  相似文献

Polymerase chain reaction analysis revealed four alternatively spliced variants of each of the γ and δ isoforms of calmodulin-dependent protein kinase II (CaM-kinase II) in rabbit liver. Among the four variants of the γ isoform, two were novel ones, designated as CaM-kinase II γ-H and γ-I. The γ-I variant possessed both of the two deletable exons, D2a and D2b, which had never been found together in any variant. Sequence analysis of the γ-I indicated that the D2a was upstream of the D2b and that they were contiguous with each other in the γ-I. Among the four variants of the δ isoform, two were also novel ones, designated as CaM-kinase II δ-11 and δ-12, and the other two were the already-reported ones, δ-2 and δ-6. The δ-11 and δ-12 were identical to the δ-2 and δ-6, respectively, except that three bases (CAG) located at a splicing junction was deleted in the δ-11 and δ-12, suggesting two splicing sites of a single intron. Thus, the diverse splicing patterns may produce many more variants than those so far considered.  相似文献

Karyotypes of 24 diploid (2n=12)Tragopogon species are similar with one long pair of chromosomes (A), two medium-length pairs (B and C), and three short pairs (D, E, and F). These species may be divided into three karyotypic groups: 1) seven species with a satellite on A and on D; 2) 14 species with a satellite on A only; and 3) three species with a satellite on D only. Most species within karyotype groups may be separated from each other either by distinctive features of certain chromosomes or by statistical differences in length of chromosome arms or long arm: short arm ratios of chromosome A. Three tetraploid (2n=24) species had two long pairs (A,A′), four medium-length pairs (B,B′;C,C′), and six short pairs (D,D′;E,E′;F,F′). Three flavone glycosides, sudachiins B, C and D, were isolated from the green peel of Citrus sudachi. On the basis of UV, 1H NMR and 13C NMR spectral data sudachiins B and C were identified as sudachiin A 6″- (3-hydroxy-3-methyl)glutarateand 7-O-β-D-glucosyl sudachitin 6″-(3-hydroxy-3-methyl)glutarate, respectively. Sudachiin D was found to be a unique glycoside in which sudachiin A and 7-O-β-D-glucosylsudachitin were esterified at their 6″-positions with 3-hydroxy-3-methylglutaric acid.  相似文献

The novel biotransformation process of 4′-demethylepipodophyllotoxin to produce 4′-demethylepipodophyllic acid was developed for the first time. Based on the capability of modifying 4′-demethylepipodophyllotoxin structure, Bacillus fusiformis CICC 20463, Bacillus subtilis CCTCC AB93174, Pseudomonas aeruginosa CCTCC AB93066 and Pseudomonas oleovorans CGMCC 1.1641 were screened out from the eight tested strains. Among them, B. fusiformis was selected for the following study because of its high substrate conversion. The biotransformation product was separated from the biotransformation broth by macroporous resin D312, and identified as 4′-demethylepipodophyllic acid by the comparison analysis of 1H NMR, 13C NMR, and ESI-MS spectrums with those of 4′-demethylepipodophyllotoxin. This suggests that B. fusiformis, P. aeruginosa, B. subtilis and P. oleovorans had the ability to transform 4′-demethylepipodophyllotoxin into 4′-demethylepipodophyllic acid. The established biotransformation process will give reference to the biotransformation study of other bioactive natural leading compounds.  相似文献

Four homoisoflavonoids named portulacanones A−D, identified as 2′-hydroxy- 5,7-dimethoxy-3-benzyl-chroman-4-one, 2′-hydroxy-5,6,7-trimethoxy-3-benzyl-chroman-4-one, 5,2′-dihydroxy-6,7-dimethoxy-3-benzyl-chroman-4-one, and 5,2′-dihydroxy-7-methoxy-3-benzylidene-chroman-4-one, were isolated from aerial parts of the plant Portulaca oleracea along with nine other known metabolites. Their structures were established on the basis of extensive spectroscopic analyses. Portulacanones A−D is the first group of homoisoflavonoids so far reported from the family Portulacaceae. They represent a rare subclass of homoisoflavonoids in nature with a structural feature of a single hydroxyl group substituted at C-2′ rather than at C-4′ in ring B of the skeleton. Three homoisoflavonoids and the known compound 2,2′-dihydroxy-4′,6′-dimethoxychalcone selectively showed in vitro cytotoxic activities towards four human cancer cell lines. Especially 2,2′-dihydroxy-4′,6′-dimethoxychalcone showed cytotoxic activity against cell line SGC-7901 with an IC50 value of 1.6 μg/ml, which was more potent than the reference compound mitomycin C (IC50 13.0 μg/ml).  相似文献

Toxicity tests of three strains of Bacillus sphaericus against late instars of 12 culicine mosquito species indicated a wide range of susceptibility. Culex pipiens and C. salinarius were highly susceptible (LC50s < 104 spores/ml) to strain 1593, and C. pipiens and C. restuans were highly susceptible to strain 2013-4. The potency of strain SSII-1 was approximately one-tenth that of strains 1593 and 2013-4 against C. pipiens. Susceptibility of Aedes species to strain 1593 was highly variable. At temperatures ≥ 20°C, A. fitchii, A. intrudens, A. stimulans, and A. vexans were moderately to highly susceptible (LC50s 6 × 103−4 × 104 spores/ml), A. triseriatus was only slightly susceptible (LC50 > 106 spores/ml), and A. aegypti was refractory. Susceptibility of Aedes mosquitoes to strain SSII-1 was less variable, with LC50s against A. aegypti, A. canadensis, A. stimulans, and A. triseriatus all being between 104 and 106 vegetative cells + spores/ml. All species of mosquitoes tested were, in general, highly susceptible to B. thuringiensis var. israelensis (LC50s 2.3 × 103−2.5 × 104 spores/ml). In B. sphaericus toxicity tests, decreased temperatures resulted in up to a 16-fold increase in LC50 and a substantial reduction in probit line slope. First-instar A. aegypti larvae were more susceptible to B. sphaericus strain SSII-1 than the three later instars, which were approximately equally susceptible; however, no significant difference was observed in the susceptibility of the four instars of A. triseriatus.  相似文献

A 125I-labelled photolabile derivative of cholic acid has been used to investigate the organic anion-binding Y′ fraction from rat liver, prepared by the method of Sugiyama, Y., Yamada, T. and Kaplowitz, N. (1982) Biochimica Biophysica Acta, 709, 342–352. The use of this photoaffinity probe led to the discovery of previously undescribed bile acid-binding proteins. A comprehensive purification scheme for the Y′ proteins which allows the isolation of these novel binding species is described. Electrophoretic analysis shows that the Y′ binders can be divided into two groups. The proteins in group 1 are dimeric and the 5B, 6E and 7F binding species consist of subunits with approximate molecular masses of 19.6, 15.6 and 14.9 kDa, respectively. The group 2 binding proteins, 5C, 5D and 8C, are monomeric and have molecular masses of approximately 36.2, 36.2 and 33 kDa, respectively. Calculation of the incorporation of 125I by these proteins showed that the group 1 proteins displayed a significantly greater specific incorporation of radioactivity than group 2. The specificity of 125I-labelled 3β-azidocholylhistamine is further demonstrated by analysis of tryptic digests of photoaffinity labelled Y′ binders and glutathione S-transferases AA, A, D and F by reverse-phase high-performance liquid chromatography (HPLC) and thin-layer chromatography (TLC). The majority of the radioactivity was shown to be incorporated into a single component, which was not coincident with the free photoaffinity label.  相似文献

Dendrocalamus sinicus, which is the largest bamboo species in the world, has broad prospects in the fields of bioenergy and biorefinery application. In this study, dewaxed D. sinicus samples were sequentially treated with 80 % ethanol containing 0.025 M HCl, 80 % ethanol containing 0.5 % NaOH, and aqueous alkaline solutions (containing 2.0, 5.0, and 8.0 % NaOH, respectively) at 75 °C for 4 h, in which 9.63, 8.71, 21.83, 21.09, and 13.09 % of the original lignin were isolated, respectively. The lignin fractions obtained were comparatively characterized by chemical composition, molecular weights, and structural features by wet chemical and instrumental analysis methods. It was found that the bamboo lignin fractions isolated by ethanol had lower weight-average molecular weights (1,360–1,380 g?mol?1) and contained much higher amounts of associated hemicelluloses, while the lignin fractions isolated by aqueous alkaline solutions had higher weight-average molecular weights (5,300–6,040 g?mol?1) and contained lower amounts of associated hemicelluloses. Spectroscopy analyses indicated that the bamboo lignin was a typical grass lignin, consisting of p-hydroxyphenyl (H), guaiacyl (G), and syringyl (S) units. A small percentage of the lignin side-chain was found to be acetylated at the γ-carbon, predominantly at syringyl units. The major interunit linkages present in the bamboo lignin obtained were β-O-4′ aryl ether linkages, together with lower amounts of β-β′, β-5′, and β-1′ linkages.  相似文献

A bacterium (strain A1) isolated from a ditch synthesized three types of intracellular alginate lyases: A1-I (molecular weight [M.W.] 60,000), A1-II-2 (M.W. 25,000) and A1-III (M.W. 38,000). The nucleotide sequence of the gene for A1-I lyase, which has been cloned in Escherichia coli DH1 was determined. The open reading frame of the gene encoded 622 amino acids with a calculated M.W. of 69,153. The N-terminal amino acid sequence of A1-I lyase purified from strain A1 or E. coli DH1 cells transformed with the A1-I lyase gene was consistent with the deduced sequence from 55His to 74Ala, indicating that the A1-I lyase was synthesized as a precursor with a M.W. of 69,153 and then processed to a mature form with a M.W. of 63,681. The N-terminal sequence of the first twenty amino acids of A1-III lyase was found to match that of A1-I lyase. The N-terminal sequence of the first twenty amino acids of A1-II-2 lyase was consistent with the deduced amino acid sequence from 414Ala to 433Val in the nucleotide sequence of the A1-I lyase gene. These results indicated that the A1-I lyase was further processed to generate A1-II-2 and A1-III lyase species.  相似文献
