In the leaf of rice (Oryza sativa L.) cultivar Yunnan purple rice,the anthocyanins with an obvious absorption peak at 530nm were distributed in the cells of upper and lower epidermis,bulliform tissue and bristle. The maximal photosynthetic oxygen evolution rate and chlorophyll content in flag leaves were 28% and 23%,respectively,more than the common green leaf rice cultivar Chijiaoru-anzhan. Higher chlorophyll content is probably one of the physiological adaptations for enhancing light harvesting capacity of the antenna in photosystems in this cyanic leaves species. Upon the photooxidation of leaf segments mediated by methyl viologen in weak light for 3 days,the distinct bleaching of anthocyanins in purple rice was associated with the reduction of scavenging ability to DPPH· free radical ability and the increase in membrane leakage rate. But almost no changes in contents of flavonoids and total phenolics were observed. Chlorophyll fluorescence parameters Fv/Fo,qP and φPSⅡ decreased with the increase in NPQ and DES of xanthophylls cycle after photooxidation treatment. Green rice leaves showed more decrease in DPPH· scavenging rate and more increase in cell membrane leakage rate but showed a trace of anthocyanins during photooxidation. It is sug-gested that anthocyanin may be a beneficial and primary antioxidant in sun cyanic rice leaves against oxidative stress induced by environmental adversity. And photooxidation could induce different changing patterns of anthocyanins between the tested purple and green rice leaves.  相似文献

Morphological responses of American cranberry (Vacciniummacrocarpon Ait, Ericaceae) to different light conditions (red,far-red, white light and sunlight) were examined. Root growth and development,stem elongation, leaf enlargement, de-etiolation of stem and leaf, flower budformation, and flowering of American cranberry were measured under each lightcondition and in the dark. It was found that red light promotes development ofroots and leaves, flowering, and de-etiolation of stem and leaf of Americancranberry. Stem elongation and etiolation of stem and leaf were shown infar-redlight and dark. Anthocyanin biosynthesis as phytochemical response in cranberryplants was most sensitive to red light. Estimation of anthocyanin levels indifferent parts of cranberry plant suggested that anthocyanins were presentonlyin red fruit skins, and not in peeled fruits, green fruits, green leaves, greenstems, roots and seeds.  相似文献

High-light leaves of the evergreen herb Galax urceolata exhibit a striking color change from green to red during winter months due to anthocyanin synthesis in outermost mesophyll cells. Here we investigate three possible functions of this color change. To test the hypothesis that anthocyanins function as light attenuators, maximum photosystem II efficiency (F(v)/F(m)) of red and green leaves was measured during and after exposure to wavelengths either strongly or poorly absorbed by anthocyanin. To determine whether anthocyanins elevate radical-scavenging capacity, antioxidant activity of red and green leaves was assessed using the alpha,alpha-diphenyl-beta-picrylhydrazyl assay. Nonstructural carbohydrate levels were analyzed to test the hypothesis that anthocyanins function as a carbon sink. Declines in F(v)/F(m) under white and green light were significantly greater for green than red leaves, but were comparable under red light. Anthocyanin content positively correlated with antioxidant activity. Although levels of anthocyanins did not appear to be related to nonstructural carbohydrate concentration, high levels of sugars may be necessary for their photo-induction. Results suggest that anthocyanins function as light attenuators and may also contribute to the antioxidant pool in winter leaves.  相似文献

Common hazel (Corylus avellana L., Fusca rubra Dipp.) juvenile leaves from the periphery of the canopy and thus subjected to high fluxes of solar radiation are characterized by red coloration due to anthocyanin accumulation disappearing in mature leaves. To elucidate the physiological role of anthocyanin accumulation, the interrelations between anthocyanin content, a degree of attenuation by the pigments of the light reaching the photosynthetic apparatus (PSA), and PSA tolerance to photoinhibition in C. avellana juvenile leaves were studied. Absorption spectra were calculated taking into account the light losses due to reflection by the leaf. The analysis of the spectra showed that, in red common hazel leaves accumulating high amounts of anthocyanins in the vacuoles of the upper and lower epidermal cells, up to 95% of visible radiation entering the leaf blade was absorbed by these pigments. The rate of the linear electron transport (ETR) in the chloroplast electron transport chain (ETC) was closely correlated with the anthocyanin content (r 2 = 0.87). In red leaves, the saturation of ETR dependence on irradiance was observed at the higher values of PAR than in green leaves. In red juvenile leaves, this value was close to that in mature green leaves tolerant to high light. There were no differences between red and green leaves in the level of non-photochemical quenching, the content of violaxanthin cycle pigments, a degree of their de-epoxidation under natural illumination and at irradiation with high PAR fluxes. Basing on the data obtained, one may conclude that anthocyanins in C. avellana juvenile leaves serve PSA photoprotection, preventing injury of immature PSA with excessive fluxes of PAR.  相似文献

本文研究南天竹不同叶色的叶片叶绿素、花青素、可溶性糖含量和气孔密度、POD、CAT活性的变化。结果表明,南天竹叶色差异与花青素、叶绿素含量有关,绿叶花青素含量低、叶绿素含量高;红叶则相反。此外,红叶下表皮气孔数目少,气孔小;绿叶气孔数目多,气孔大。红叶的可溶性糖含量明显高于绿叶,而POD和CAT活性低于绿叶。  相似文献

Red autumn colouration of trees is the result of newly synthesized anthocyanin pigments in senescing autumn leaves. As anthocyanin accumulation is costly and the trait is not present in all species, anthocyanins must have an adaptive significance in autumn leaves. According to the coevolution hypothesis of autumn colours, red autumn leaves warn herbivorous insects – especially aphids that migrate to reproduce in trees in the autumn – that the tree will not be a suitable host for their offspring in spring due to a high level of chemical defence or lack of nutrients. The signalling allows trees to avoid herbivores and herbivores to choose better host trees. In this study the coevolution hypothesis was tested with four deciduous tree species that have red autumn leaf colouration – European aspen (Populus tremula L.) (Salicaceae), rowan (Sorbus aucuparia L.) (Rosaceae), mountain birch [Betula pubescens ssp. czerepanovii (NI Orlova) Hämet‐Ahti], and dwarf birch (Betula nana L.) (Betulaceae), and with two generalist herbivores, the autumnal moth [Epirrita autumnata (Borkhausen)] and the winter moth [Operophtera brumata (L.)] (both Lepidoptera: Geometridae). Anthocyanin concentrations of autumn leaves were determined from leaf samples and the growth performance parameters of the moth larvae on the study trees were measured in the spring. Trees with higher anthocyanin concentration in the autumn were predicted to be low‐quality food for the herbivores. Our results clearly showed that anthocyanin concentration was not correlated with the growth performance of the moths in any of the studied tree species. Thus, our study does not support the coevolution hypothesis of autumn colours.  相似文献

Two different α‐glucosidase‐producing thermophilic E134 strains were isolated from a hot spring in Kozakli, Turkey. Based on the phenotypic, phylogenetic and chemotaxonomic evidence, the strain was proposed to be a species of G. toebii. Its thermostable exo‐α‐1,4‐glucosidases also were characterized and compared, which were purified from the intracellular and extracellular fractions with estimated molecular weights of 65 and 45 kDa. The intracellular and extracellular α‐glucosidases showed optimal activity at 65 °C, pH 7·0, and at 70 °C, pH 6·8, with 3·65 and 0·83 Km values for the pNPG substrate, respectively. Both enzymes remained active over temperature and pH ranges of 35–70 °C and 4·5–11·0. They retained 82 and 84% of their activities when incubated at 60 °C for 5 h. Their relative activities were 45–75% and 45–60% at pH 4·5 and 11·0 values for 15 h at 35 °C. They could hydrolyse the α‐1,3 and α‐1,4 bonds on substrates in addition to a high transglycosylation activity, although the intracellular enzyme had more affinity to the substrates both in hydrolysis and transglycosylation reactions. Furthermore, although sodium dodecyl sulfate behaved as an activator for both of them at 60 °C, urea and ethanol only increased the activity of the extracellular α‐glucosidase. By this study, G. toebii E134 strain was introduced, which might have a potential in biotechnological processes when the conformational stability of its enzymes to heat, pH and denaturants were considered. 该研究以陕西汉中、河南林州、河北涉县和北京中国科学院植物研究所4个种源黄连木(Pistacia chinensis Bunge)的苗木为对象,用分光光度计和色差仪对其叶绿素、类胡萝卜素、花色素苷含量及叶色参数(L*、a*、b*)进行了测定分析,探讨不同种源苗木秋季叶色变化规律及差异,揭示黄连木叶色呈现与叶片色素含量之间的内在关联,为筛选适合城市绿化的优良黄连木种源提供依据。结果表明:(1)在秋季叶片转色期,随着时间的推移,4个种源黄连木叶片的叶绿素、类胡萝卜素和花色素苷含量的比例呈现不同的变化趋势,其中:河北种源的花色素苷含量较高,叶片呈现红色;陕西种源叶绿素含量较高,叶片呈现绿色的时间较长;河南、北京种源处于两者之间。(2)各个种源黄连木的叶色参数a*值(红/绿)均与花色素苷含量呈正相关关系,与叶绿素含量呈负相关关系,且相关系数均达到显著水平(P0.05),各个种源叶色参数L*值(光泽明亮度)也与叶绿素含量间表现出显著或极显著的正相关性。研究发现,河北种源黄连木秋季的叶色最红,陕西种源黄连木叶片呈现绿色的时间最长;色差仪的应用实现了叶色和各色素含量间量化的关系。  相似文献

This work describes the study of the chemical composition and bioactivity of the essential oils (EOs) of the different organs (leaves, flowers, stems and roots) from Eruca vesicaria. According to the GC and GC/MS analysis, all the EOs were dominated by erucin (4‐methylthiobutyl isothiocyanate) with a percentage ranging from 17.9 % (leaves) to 98.5 % (roots). The isolated EOs were evaluated for their antioxidant (DPPH, ABTS and β‐carotene/linoleic acid), antibacterial and inhibitory property against α‐amylase and α‐glucosidase. Most EOs exhibited an interesting α‐glucosidase and α‐amylase inhibitory potential. The roots essential oil was found to be the most active with IC50 values of 0.80±0.06 and 0.11±0.01 μg mL?1, respectively. The essential oil of roots exhibited the highest antioxidant activity (DPPH, PI=92.76±0.01 %; ABTS, PI=78.87±0.19; and β‐carotene, PI=56.1±0.01 %). The isolated oils were also tested for their antibacterial activity against two Gram‐positive and three Gram‐negative bacteria. Moderate results have been noted by comparison with Gentamicin used as positive control.  相似文献

Purified anthocyanin extracts show strong antioxidant properties in vitro, but it is not known whether they can scavenge reactive oxygen in living cells. The oxidative responses in red and green portions of Pseudowintera colorata leaf laminae were compared by the real‐time imaging of H2O2 in cells after mechanical injury. An oxidative burst was elicited almost immediately from chloroplasts in the palisade mesophyll, as evidenced using the fluorochromes dichlorofluorescein and scopoletin. H2O2 accumulated in green lamina regions for 10 min, and then decreased slowly. By contrast, red regions recovered rapidly, and maintained consistently low levels of H2O2. Infusion of cells with N‐acetyl‐l ‐cysteine accelerated the depletion of H2O2 from green regions. Wounded leaves ultimately developed a localized necrotic lesion and an intense anthocyanic band. The red regions were enriched in anthocyanins, flavonols, dihydroflavonols, and hydroxycinnamic acids. Only the anthocyanins were suitably located to account for the enhanced rates of H2O2 scavenging. The data support the hypothesis that red cells have elevated antioxidant capabilities in vivo.  相似文献

At suboptimal temperatures, anthocyanins accumulate in the illuminated leaf surface of some maize genotypes and, if the anthocyanins shade chloroplasts, they can effectively reduce the risk of photo‐inhibition but also photo‐synthesis. To investigate this phenomenon, gas exchange, fluorescence, superoxide dismutase activity and xantho‐phyll composition of anthocyanin‐containing HOPI and anthocyanin‐deficient W22 maize genotypes were measured in either white or red light, where the latter is not absorbed by anthocyanins. Despite differences in light absorption in chloroplasts, photosynthesis did not differ between HOPI and W22 under either light source, suggesting that neither CO2 supply nor photochemistry were more limiting in red leaves than in green leaves. In fact, no major differences in transpiration were detected. The ΔF/Fm (photosystem II quantum yield) of HOPI in white light was higher than in red light and higher than ΔF/Fm of W22 with either light source. This probably compensated for the lower white light absorption of HOPI chloroplasts compared with W22 because of the presence of anthocyanins and led to similar rates of calculated electron transport for both genotypes. After exposure to high white light at 5 °C, xanthophyll de‐epoxidation and superoxide dismutase activity were lower in HOPI than in W22. Further, HOPI could be exposed to a much higher irradiance than W22 before Fv/Fm was reduced to that of W22.  相似文献

为探究秋季枫叶呈色的关键生理因素,该文以转色期叶色为绿色、黄色和红色的枫香单株为试材,研究了L*、a*、b*值变化与叶片色素、可溶性糖及可溶性蛋白质含量变化的相关性。结果表明:(1)在变色期,3种色彩枫香叶片叶绿素a、叶绿素b、总叶绿素和类胡萝卜素均大量降解,花色素苷不同程度积累。(2)绿色叶单株叶绿素和类胡萝卜素始终保持较高含量,花色素苷含量上升4.2倍,叶片内色素含量比值始终保持稳定; 黄色叶单株叶绿素和类胡萝卜素含量最低,花色素苷含量上升4.4倍,b*值与叶绿素含量极显著负相关,与类胡萝卜素含量显著负相关,与花色素苷/类胡萝卜素含量比值极显著正相关; 红色叶单株叶绿素和类胡萝卜素含量略高于黄色叶单株,花色素苷含量上升27.2倍,a*值与叶绿素含量、类胡萝卜素含量极显著负相关,与花色素苷含量显著正相关,与色素含量比值无显著相关性。(3)红色叶单株具有较高的可溶性糖含量和可溶性蛋白质含量。因此,在枫香叶片变色期,保持较高的叶绿素和类胡萝卜素含量,维持色素含量比值稳定使叶片呈现绿色; 叶绿素和类胡萝卜素的大量降解,以及花色素苷/类胡萝卜素含量比值的升高使叶片呈现黄色; 叶绿素的降解和花色素苷的大量合成使叶片呈现红色。  相似文献

We evaluated the association of red coloration with senescence in sugar maple (Acer saccharum Marsh.) leaves by assessing differences in leaf retention strength and the progression of the abscission layer through the vascular bundle of green, yellow, and red leaves of 14 mature open-grown trees in October 2002. Computer image analysis confirmed visual categorization of leaves as predominantly green, yellow or red, and chemical quantification of leaf pigment concentrations verified that leaf color reflected underlying differences in leaf biochemistry. Significantly lower chlorophyll concentrations within red and yellow leaves indicated that senescence was more advanced in leaves from these color categories relative to green leaves. Among leaf types, only red leaves contained high concentrations of anthocyanins. There were significant differences in leaf retention capacity among color categories, with the petioles of green leaves being the most firmly attached to twigs, followed by red and then yellow leaves. Microscopic analysis indicated that yellow leaves had the most advanced extension of the abscission layer through the vasculature, with green and red leaves having significantly less abscission layer progression than yellow. A more limited progression of the abscission layer through vascular bundles may be evidence of delayed leaf senescence that could extend resorption of mobile leaf constituents. Together, results from this study suggest an association between leaf anthocyanin content and functional delays in senescence.  相似文献

Leaf developmental patterns were characterized for three tropical tree species with delayed greening. Changes in the pigment contents, photosynthetic capacity, stomata development, photosystem 2 efficiency, rate of energy dissipation, and the activity of partial protective enzymes were followed in developing leaves in an attempt to elucidate the relative importance of various photoprotective mechanisms during leaf ontogeny. Big leaves of Anthocephalus chinensis, a fast-growing light demanding species, expanded following an exponential pattern, while relatively small leaves of two shade-tolerant species Litsea pierrei and Litsea dilleniifolia followed a sigmoidal pattern. The juvenile leaves of A. chinensis and L. pierrei contained anthocyanin located below the upper epidermis, while L. dilleniifolia did not contain anthocyanin. Leaves of A. chinensis required about 12 d for full leaf expansion (FLE) and photosynthetic development was delayed 4 d, while L. pierrei and L. dilleniifolia required 18 or 25 d for FLE and photosynthetic development was delayed 10 or 15 d, respectively. During the leaf development the increase in maximum net photosynthetic rate was significantly related to changes in stomatal conductance and the leaf maturation period was positively related to the steady-state leaf dry mass per area for the three studied species. Dark respiration rate of leaves at developing stages was greater, and pre-dawn initial photochemical efficiency was lower than that of mature leaves. Young leaves displayed greater energy dissipation than mature leaves, but nevertheless, the diurnal photoinhibition of young L. dilleniifolia leaves was higher than that of mature leaves. The young red leaves of A. chinensis and L. pierrei with high anthocyanin contents and similar diurnal photoinhibition contained more protective enzymes (superoxide dismutase, ascorbate peroxidase) than mature leaves. Under water‐limiting conditions excitation energy harnessed by chlorophyll can lead to the formation of reactive oxygen species (ROS). Resurrection plants minimize their formation by preventing the opportunity for light–chlorophyll interaction but also quench them via antioxidants. Poikilochlorohyllous species such as Xerophyta humilis break down chlorophyll to avoid ROS formation. Homoiochlorophyllous types retain chlorophyll. We proposed that leaf folding during drying of Craterostigma wilmsii and Myrothamnus flabellifolius shades chlorophyll to avoid ROS (Farrant, Plant Ecology 151, 29–39, 2000). This was tested by preventing leaf folding during drying in light. As controls, plants were dried without light, and X. humilis was included. Craterostigma wilmsii did not survive drying in light if the leaves were prevented from folding, despite protection from increased anthocyanin and sucrose and elevated antioxidant enzyme activity. Membranes were damaged, electrolyte leakage was elevated and plastoglobuli (evidence of light stress) accumulated in chloroplasts. Restrained leaves of M. flabellifolius survived drying in light. Leaf folding allows less shading, but the extent of chemical protection (anthocyanin content and antioxidant activity) is considerably higher in this species compared with C. wilmsii. Chemical protection appears to be light regulated in M. flabellifolius but not in C. wilmsii. Drying in the dark resulted in loss of viability in the homoiochlorophyllous but not the poikilochlorophyllous species. It is hypothesized that some of the genes required for protection are light regulated in the former.  相似文献

结合民族植物学和药理学的研究方法,对西双版纳地区傣族、哈尼族和基诺族等3个少数民族民间利用番石榴(Psidium guajava)、余甘子(Phyllanthus emblica)和水柳(Homonoia riparia)的传统知识进行调查研究及体外抗菌活性实验。结果表明:番石榴和余甘子在村寨中较为常见,当地少数民族将其种植于庭院中,常作为果蔬食用,食用番石榴嫩叶可缓解拉肚子的症状,治疗腹痛、腹泻。水柳生长在水边,傣族会将其叶作为腌酸鱼的配料之一。根据文献记载,番石榴、余甘子和水柳的叶部位作为药使用时,常煎水外洗,治疗皮肤瘙痒。对这3种药用植物叶部位采用80%乙醇浸泡制备的提取物进行体外抗菌实验,结果显示番石榴、余甘子和水柳3种药用植物对金黄色葡萄球菌和大肠埃希菌均有较好的抑菌和杀菌活性,其最小抑菌浓度MIC在98~390 μg·mL 1之间,最小杀菌浓度MBC在98~781 μg·mL 1之间。番石榴和水柳叶对铜绿假单胞菌有一定抑菌和杀菌活性,其MIC和MBC范围均为6 250~12 500 μg·mL 1。由此可见,这3种药用植物的民间利用具有一定的合理性和药用开发价值。  相似文献
