Cultures of the coenobial four-celled chlorococcal algaScenedesmus quadricauda were synchronized. Ontogeny of the species, proceeding analogously to that of the unicellular forms, was studied; however, the cells in the coenobium ripen successively. The paramoters of the synchronized cultures were followed and according to the obtained results an objective method of evaluation of the synchronization-degree of the algal cultures, on the basis of information theory, was suggested.  相似文献

Summary The kinetics of Cu uptake in nutritionally starved cells of the diazotrophic cyanobacteriumNostoc calcicola Bréb. have been compared with those in cells recovering from starvation. Unstarved cyanobacterial cells assimilated 97.0 nmol Cu mg–1 protein within 1 h when incubated in medium containing 40 M Cu. Uptake was markedly inhibited in carbon-starved cells and, to a lesser extent, in cells starved of nitrogen or sulphur. The intracellular concentrations of protein and photopigments were markedly lower in cells starved of carbon, nitrogen, sulphur or phosphorus, whilst that of carbohydrate was lower in cells starved of carbon, sulphur or phosphorus, but almost doubled in cells starved of nitrogen. The ability to assimilate Cu was partially restored in cells after 72 h of recovery from phosphorus or sulphur deprivation, but showed little improvement during recovery from carbon or nitrogen starvation. A possible role of phosphorus in regulating Cu transport and accumulation is discussed.  相似文献

Summary Absorption spectra of a young and an old culture of the diatom Pheodactylum tricornutum were measured in thin layers between two opal glass sheets. The spectra at 24° and at -196°C were replotted to give equal areas from 730–625 m to allow direct comparison. At 24°C the spectrum for the difference between the two cultures had a negative component of 18 m half width centered at 675 m and a positive region of W0.5=26 m near 700 m.The spectra at -196°C may be somewhat distorted by clumping of the cells during freezing but nevertheless the 16 day culture clearly showed a smaller proportion of Ca 670 to Ca 680. This older culture has a shoulder due to a 707 m component. The difference curve at -196°C shows the decrease of an unsymmetrical band peaking at 669 m and an increase at 695 m in addition to the 707 m component. Due to the possibility of distortion, the presence of an actual component at 695 is doubtful in these particular cultures.The room temperature spectrum in the chloropyhll a region for the 5 day culture can be closely fitted by a single probability curve at 675 m having a half-width of 31 m. The sum of two components, with widths more reasonable for chlorophylls, also matched the data well enough. These two probability curves, of 22 m half width, centered on 669 and 683.2 m and had a height ratio, h669/h683 of 1.18. In the 16 day culture the ratio for these bands changed to 1.11 and there was extra absorption around 700 m.Dedicated to Professor C. B. van Niel on the occasion of his 70th birthday  相似文献

By means of circular dichroism (CD) spectroscopy, we have characterized the organization of the photosynthetic complexes of the diatom Phaeodactylum tricornutum at different levels of structural complexity: in intact cells, isolated thylakoid membranes and purified fucoxanthin chlorophyll protein (FCP) complexes. We found that the CD spectrum of whole cells was dominated by a large band at (+)698 nm, accompanied by a long tail from differential scattering, features typical for psi-type (polymerization or salt-induced) CD. The CD spectrum additionally contained intense (−)679 nm, (+)445 nm and (−)470 nm bands, which were also present in isolated thylakoid membranes and FCPs. While the latter two bands were evidently produced by excitonic interactions, the nature of the (−)679 nm band remained unclear. Electrochromic absorbance changes also revealed the existence of a CD-silent long-wavelength (∼545 nm) absorbing fucoxanthin molecule with very high sensitivity to the transmembrane electrical field. In intact cells the main CD band at (+)698 nm appeared to be associated with the multilamellar organization of the thylakoid membranes. It was sensitive to the osmotic pressure and was selectively diminished at elevated temperatures and was capable of undergoing light-induced reversible changes. In isolated thylakoid membranes, the psi-type CD band, which was lost during the isolation procedure, could be partially restored by addition of Mg-ions, along with the maximum quantum yield and the non-photochemical quenching of singlet excited chlorophyll a, measured by fluorescence transients.  相似文献

Summary Differentiating cells ofMicrasterias denticulata have been treated with aqueous solutions of the antibiotic gougerotin. Strong and characteristic cytomorphogenetic aberrations, resembling those of the anuclear type of development could be observed. It has been speculated that the aberrant growth of the growing half cell is the result of inhibition of protein synthesis by gougerotin.In addition to the morphogenetic influence, nuclear migration has been strongly inhibited by the drug. Therefore, it might be suggested that gougerotin is an active anti-microtubule agent.  相似文献

The uptake pattern of Ca2+ by the cyanobacterium Nostoc calcicola Bréb in its freely suspended and immobilized form is comprised of two distinct phages; (a) rapid uptake for 1st 10 min followed by (b) slower transport at least up to 60 min. Entrapment of cyanobacterial cells in polyvinyl foam always maintained a higher Ca2+ profile over freely suspended cells. Also, the intracellular Ca2+ concentration was three times more in the former under similar experimental conditions. Whereas, illumination supported maximum Ca2+ transport in all the sets, darkness resulted in drastic reduction (90%) of Ca2+ uptake in freely suspended cells and least (15%) in polyvinyl entrapped cyanobacterial cells. Exogenously added ATP (10 μM) on the other hand, enhanced Ca2+ uptake in dark incubated freely suspended cells; ATP at the same concentration failed to bring out any significant enhancement in cation uptake in immobilized cells facing dark exposure. It was observed that these cells were still able to sustain sufficient ATP preserves to drive active transport of Ca2+ even in the dark. Furthermore, the immobilized cells exhibited remarkable Ca2+ transport rate even at the age of 20 and 50 days at which its free living counterpart took up insignificant Ca2+. These findings suggest the improved metabolic efficiency of polyvinyl foam entrapped cells over freely suspended cells in terms of Ca2+ accumulation and its possible use as a bioreactor for metal accumulation/removal in repetitive cycles without any measurable loss in cell biomass. This study reports further information on mareninne, awater-soluble blue-green pigment synthesized by the diatom Hasleaostrearia, which is essential to the greening of maturing oysters inFrench production areas. The extraction process is reported, as well aspreliminary characterization of a partially purified marennine extract,including quantitative spectrophotometric analysis of intracellular pigment. Amean specific extinction coefficient, E 1% 1cm = 17.2 at 669 nm, is proposed. Results forquantitative determination of marennine accumulated in cells during batchcultures are presented, and their importance for pigment production isconsidered in relation to potential industrial applications.  相似文献

The diatom Haslea ostrearia that lives in oyster ponds has the distinctive feature of synthesizing “marennine”, a blue-green pigment of which the chemical nature still remains unknown. This pigment is responsible for the greening of oyster gills. Here, we report a new method for extraction and purification of intracellular (accumulated in the apex of the cell) and extracellular (released into the external medium) forms of the pigment. Intracellular marennine is obtained by extraction from blue algal pellets with a carbonate buffer. The extract is then centrifuged and filtered. Extracellular marennine is obtained by clarification of blue-coloured culture medium. Both extracts are then purified by a semi-preparative process, using ultrafiltration through membranes and anion-exchange chromatography. This procedure allows us to produce native pigment displaying the degree of purity required to enter upon the molecular characterisation of marennine. By this process, about 35% of the initial amount of pigment can be recovered. If necessary, this method could be easily scaled up to a larger production system to accommodate potential industrial applications.  相似文献

Spacing between individuals in populations of Harmothoë imbricata has been investigated both on the shore and in the laboratory. Males tend to occur closer together than females, and the mean male‐male individual distance measured was less than the mean distance between females; male—female distances for immature worms were intermediate. When worms mature they pair: the male mounts the female and lies across her dorsal surface. There is evidence that after spawning the members of a pair separate. Contact responses between worms have been investigated in the laboratory. Most encounters between immature worms lead to separation, as a result of one or both worms moving rapidly away from the other or fighting; females show more marked avoidance behaviour than males. The majority of male‐male and female‐female encounters between mature worms also lead to separation but in male‐female encounters the male usually mounts the female. A male which has mounted a female becomes highly aggressive and will attack intruding males but not females.  相似文献

Zusammenfassung Mit Hilfe einer modifizierten Färbemethode nach Crafts (1931) konnte festgestellt werden, daß die bei Teilungsstadien von Micrasterias denticulata nach Turgorverminderung auftretenden Zellwandverliekungen von plasmatischen Fäden durchzogen sind. Das periphere Plasma löst sich daher trotz der Sekretion von Wandmaterial nicht vollständig von der Primärwand.Es wird die mögliche Funktion von plasmatischen Fäden unterschiedlicher Dehnbarkeit für die Form der Wandverdickung diskutiert.
Protoplasmic structures in artificially produced cell-wall thickenings
Summary With the aid of a modified technique by Crafts (1931) it could be shown that cell-wall thickenings in Micrasterias denticulata, artificially produced by turgor-reduction during cell-growth, are penetrated by protoplasmic filaments. The outer protoplasmic layer is therefore not removed completely from the primary wall during the secretion of cell-wall material. The possible function of protoplasmic filaments differing in their extensibility is discussed in relation to the characteristic form and pattern of the wall-thickenings.

Haslea ostrearia is a common marine tychopelagic diatom which has the particularity of synthesizing a blue-green hydrosoluble pigment called “marennine”. This pigment, when released into the external medium, is known to be responsible for the colour of oyster gills. Here we present results for main biophysical and biochemical characteristics of pure intra- and extracellular marennine. Tests for chemical determination show that the nature of the two forms of marennine cannot be distinguished and could be related to a polyphenolic compound. Nevertheless, based on spectral properties and the molecular weight, which is about 10751 ± 1 and 9893 ± 1 Da, for the intracellular and extracellular forms respectively, we assess that the pigment accumulated in the apex of the cell and the one released in the external medium have probably distinct molecular structures.  相似文献

Summary Exposure of growingMicrasterias cells to high (32°–36°C) and low (3°–10°C) temperatures produces changes in morphology that are accompanied by several ultrastructural alterations. Whereas low temperatures essentially cause simplification of cell ornamentation, a variety of cell malformations result from high temperature treatment. These are the loss of cell symmetry leading to markedly aberrant cell shapes and an increase of main lobes with reduced degree of differentiation. Preliminary studies indicate that a shift in the distribution of membrane-associated Ca2+ by elevated temperatures probably underlies these abnormal cytomorphogenetic events. Both, low and high temperature cause a reduction in size of the young half cell and affect cytoplasmic streaming. Moreover, nuclear migration is retarded and chloroplast arrangement is influenced by temperature treatment at both ranges. Growth velocity of primary wall responsible for cell shaping is increased at high and slowed down at low temperatures compared to cells grown at 20°C.The main ultrastructural alterations induced by high temperatures are an increase in amount and length of ER cisternae, the appearance of heat shock granule aggregations localized in the cytoplasm, a reduced number of ribosomes and polysomes, the presence of oil bodies in growing cells and a varying thickness of the primary wall. Influences of low temperatures on ultrastructure are less pronounced. They are manifested in the formation of large aggregations of ER cisternae slightly differing from those found in untreated cells, a disturbed arrangement of the microtubule system surrounding the nucleus and a decrease of the number of cell wall forming cytoplasmic vesicles.It is thought that most of the temperature effects are due to an influence on membranes probably an alteration of ionic currents and, in addition, a modulation of normal protein synthesis.Dedicated to my teacher Professor Oswald Kiermayer in deep gratitude  相似文献

A clone ofScenedesmus quadricauda, isolated from Tjeukemeer, exhibits a high degree of morphological variation in synchronized cultures. Cells are synchronized by light-dark cycles. During the photoperiod they build up the capacity to divide. First division into 2- and 4-celled coenobia is induced, then during the second half of the photoperiod the induction of division into 8 unicells takes place. Division itself and the subsequent liberation of daughter cells occur in the dark period.By giving a definite photoperiod the formation of either coenobia or unicellular stages is determined. The formation of both coenobia and unicells is followed using a light microscope. In both cases only the pattern of cytokinesis is similar. After cytokinesis the unicells become ovoid in shape and form two spines at each pole. They are released from the parental wall as separate cells and show remarkable similarity to theChodatella-like cells described by SWALE (1967) and FOTT (1968). The coenobial cells elongate, adhere to one another and each of the two outmost cells forms two spines (SMITH, 1914).  相似文献

The work is concerned with the question of the effect of humic acids on the biological action of agropyrene, the effective substance from the rootstock of Agropyron repens (L.) P. BEAUV. The test organism was the alga Scenedesmus obliquus (TURP.) KRÜGER, cultivated in mineral solution. The basic biological test was the determination of the number and size of the cells of this alga. It was found that the way in which agropyrene acts on combined application with humus acids depends on which humus fraction is used. It can display a synergic action if humic acid is used, since this substance probably facilitates the penetration of agropyrene into the cells. On the other hand, fulvic acid counteracts the activity of agropyrene and this most probably by mutual binding and blocking of active groups and links.  相似文献

Blood flow to the testis, measured by the133-Xenon isotope clearance technique, initially increased after rams were exposed to elevated (32°C) temperature. However, after 5 and 7 days continuous exposure, blood flow decreased significantly. Similar changes in blood flow to the testis were found during whole body exposure to elevated temperature, or when the temperature of the testis was increased by scrotal heating. After one week of heating the wall of the spermatic artery in the middle region of the pampiniform plexus had thickened, and the arterial lumen had decreased significantly. The PGF 2 content in the testis of rams exposed to elevated temperature increased significantly. It is tentatively postulated that the higher levels of PGF 2 in the testis of rams exposed to elevated temperature may be responsible for impaired spermatogenic function by constricting the spermatic artery in the pampiniform plexus region and thereby reducing blood flow to the testis.Published with approval of the Director of Kentucky Agricultural Experiment Station as Journal Article 75-5-150.Presented at the Seventh International Biometeorological Congres, 17–23 August 1975. The light regime effect on the growth rate variability of clones of one chlorococcal alga strain growing on the surface of a solid medium was studied from the point of view of the clone selections manifesting the desired grade of this complex character. Several combinations of light regimes used for the cultivation of coenobia treated by UV-light and untreated control on two types of solid media proved the significance of manifestation degree changes of this character due to the given light conditions. The sizes of the alga colonies after a certain growth time on the surface of solid medium were evaluated statistically and the values obtained indicated the degree of growth rate manifested by the clones of a given population. The \(\bar x\) and sx values of their growth rate variability were considered as the indicators facilitating the choice of selec-tion type and screening level. This paper is a partial contribution to the solution of the problems concerning the primary selection of alga clones on the surface of solid media.  相似文献

A reproducible and efficient protoplast to plant-system has been developed for strawberry. By using young shoots growing on medium supplemented with cytokinin we were able to exclude the detrimental enzyme Pectolyase from our enzyme solution. The more gentle isolation procedure reported in this paper, together with the agarose-embedding system had a pronounced effect on protoplast viability and growth. Plant regeneration was enhanced by using thidiazuron instead of benzyladenine. Flow cytometric measurement of the DNA content in protoplast derived plants revealed that 27 out of 51 plants contained the amount of DNA corresponding to octoploid level. Fifteen plants exhibited chromosome doubling(16×), one was diplodecaploid(12×) and eight plants mixoploid.  相似文献
