Summary Allophycocyanin is one of the most important marine active peptides. Previous studies suggested that recombinant allophycocyanin (rAPC) could remarkably inhibit the S-180 carcinoma in mice, indicating its potential pharmaceutical uses. Based on intergeneric conjugal transfer, heterologous expression of rAPC was first achieved in marine Streptomyces sp. isolate M097 through inserting the apc gene into the thiostrepton-induced vector pIJ8600. The transformation frequency for this system was approximately 10−4 exconjugants/recipient. In the transformed Streptomyces sp. isolate M097, the yield of purified rAPC could amount to about 38 mg/l using a simple purification protocol, and HPLC analysis showed that the purity of the protein reached about 91.5%. In vitro activity tests also revealed that the purified rAPC had effective scavenging abilities on superoxide and hydroxyl radicals. This would widen the usefulness of the marine Streptomyces as a host to express the rAPC and to offer industrial strain for the production of rAPC.  相似文献

采用涂布法,从胶州湾海泥样品中分离到224株放线菌菌株,并且从海水样品中分离到32株放线菌菌株。根据形态学特性,菌株被分成了7个组。同时利用杯碟法测定了它们的抗菌活性,其中约7%对金黄色葡萄球菌有活性,11%对八叠球菌有效,10%对大肠杆菌有效,11%对绿脓杆菌有效,2%对白色念珠菌有效,5%对隐球菌有效,6%对绿色产色链霉菌有效和6%对米赫毛霉有效。分离到的22%放线菌对所测定的病原微生物有抗性作用。我们的结果表明胶州湾放线菌能够产生对病原微生物有抗性作用的不同代谢物,这些代谢物可以作为筛选新颖天然产物的独特和丰富的资源。  相似文献

本实验从辽宁大连海域的海水、海泥和海参养殖圈的样品中分离得到38株海洋放线菌,以金黄色葡萄球菌、溶壁微球菌、枯草芽孢杆菌、副溶血性弧菌和铜绿假单胞杆菌为指示菌,筛选出两株抑菌活性高的菌株,分别命名为HS-B31和HS-B34。16S r DNA测序鉴定及构建系统发育树的结果显示,它们都属于放线菌目、链霉菌属的不同种。对两株链霉菌的发酵上清液进行萃取、粗提和浓缩得到粗提物,即B31和B34;抗菌作用的结果显示,这两种粗提物的抗菌效果均为显著。经薄层层析分析,并利用制备型层析板对粗提物B31和B34进行活性物质的分离制备,共得到八个组分,即B31-1、B31-2、B31-3、B31-4、B31-5和B34-1、B34-2、B34-3。用滤纸片法对这些组分进行抗菌检测,结果显示组分B31-3和B34-3不仅对革兰氏阳性菌金黄色葡萄球菌有较好的抑菌作用,而且对海洋致病菌革兰氏阴性菌副溶血性弧菌也显示出较强的抑菌效果。研究为新型抗生素的研制和应用提供了微生物新资源。  相似文献

Totally 25 marine soil samples were collected from the region of Palk Strait of Bay of Bengal, Tamil Nadu, and were subjected to the isolation of actinomycetes. Sixty-eight morphologically distinct isolates were obtained and 37% (25) of them had antimicrobial activity. The potential producer was named as Streptomyces sp. VPTS3-1 and the phylogenetic evaluation on the basis of 16S rDNA sequence further categorized the organism as Streptomyces afghaniensis VPTS3-1. Further, the antimicrobial compound was extracted from the isolate using various solvents and the antimicrobial efficacies were tested against bacterial and fungal pathogens. In addition, in vitro optimization of parameters for the antimicrobial compound production revealed that the suitable pH as 7–8, the period of incubation as 9 days, temperature (30°C), salinity (2%), and starch and KNO3 as the suitable carbon and nitrogen sources respectively in starch–casein medium.  相似文献

Four marine actinobacteria tolerant to 200 g NaCl l−1 were screened for antibacterial activity against eight patient-derived multiple drug resistant (MDR) bacteria. The active compound (MW 300.2, predicted molecular formula C20H28O2) from an actinobacterium, was inhibitory to three Gram-positive and three Gram-negative MDR bacteria, seven non-clinical Gram-positive, four Gram-negative bacteria and five fungi (MIC: 3.5–4.0 µg ml−1). Also, 54% of human leukemia (HL-60) cells were killed by the compound at 0.05 µg ml−1. Bioreactor production demonstrated unusual primary metabolite kinetics. Molecular phylogenetic analysis showed this typical intertidal inhabitant to be a member of the Streptomyces genus and distinct from other salt-tolerant actinobacteria. As no compound was found to match the properties in several electronic databases, our screening strategy should increase the possibility of discovering bioactive molecules from rare actinobacteria.  相似文献

Plastic wastes have a negative impact on marine environments; however, they can be used as carbon sources and habitats by certain microbes. Microbes in the marine plastisphere can migrate worldwide through the ocean and cause serious environmental problems when they encounter suitable environments. Therefore, efforts to investigate the microbes inhabiting the marine plastisphere are increasing. In the present study, fungal strains were isolated from plastic wastes buried in Korean sea sands and mudflats and identified using molecular and morphological analyses. Five species were identified that were previously unrecorded from South Korea: Cladosporium funiculosum, Neosetophoma poaceicola, Neosetophoma rosigena, Parasarocladium gamsii, and Trichoderma fomiticola. Their molecular phylogenies and morphological characteristics are described in this study.  相似文献

A novel reactor system, the rotating disk bioreactor (RDBR), was used to mimic the niche environmental conditions of three salt-tolerant estuarine actinobacteria isolated from the Sundarbans region off the Bay of Bengal, designated MS310 (99% similar in its 16S rRNA gene sequence to Streptomyces parvallus), MS3/20 and MS1/7. The RDBR, operated at a rotational speed of one revolution per day, 50% submergence of discs, aeration rate of 1.0 vvm, and with a sucrose-containing medium, faithfully mimicked the intertidal estuarine habitat of these marine isolates, and supported biofilm formation and production of antimicrobial metabolites-in particular, actinomycin D by MS310. Onset of antibiotic production by MS310 occurs at 20 h in the RDBR compared to 55 h in a conventional stirred-tank bioreactor (STBR). Furthermore, peak antimicrobial activity is attained much earlier in the RDBR with MS310 (at 45 h) than that reported with a terrestrial strain of S. parvallus grown in a STBR (at 144 h). Peak antimicrobial activity of metabolites produced by MS1/7 and MS3/20 were also attained earlier in the RDBR (at 25 and 12 h, respectively) than in a STBR (at 80 and 28 h, respectively). Antibiotic synthesis in the three isolates, in general, appears to be associated with their growth. Overall, the RDBR may be considered the preferred alternative to the STBR for production of antimicrobials by biofilm-forming estuarine bacteria for its much higher surface/volume ratio, lower costs, and easy operability.  相似文献

We aimed at determining the antagonistic behavior of bacteria derived from marine biofilms against terrestrial phytopathogenic fungi. Some bacteria closely related to Bacillus mojavensis (three isolates) and Bacillus firmus (one isolate) displayed antagonistic activity against Colletotrichum gloeosporioides ATCC 42374, selected as first screen organism. The four isolates were further quantitatively tested against C. gloeosporioides, Colletotrichum fragariae, and Fusarium oxysporum on two culture media, potato dextrose agar (PDA) and a marine medium-based agar [yeast extract agar (YEA)] at different times of growth of the antagonists (early, co-inoculation with the pathogen and late). Overall antagonistic assays showed differential susceptibility among the pathogens as a function of the type of culture media and time of colonization (P < 0.05). In general, higher suppressive activities were recorded for assays performed on YEA than on PDA; and also when the antagonists were allowed to grow 24 h earlier than the pathogen. F. oxysporum was the most resistant fungus while the most sensitive was C. gloeosporioides ATCC 42374. Significant differences in antagonistic activity (P < 0.05) were found between the different isolates. In general, Bacillus sp. MC3B-22 displayed a greater antagonistic effect than the commercial biocontrol strain Bacillus subtilis G03 (Kodiak). Further incubation studies and scanning electronic microscopy revealed that Bacillus sp. MC3B-22 was able to colonize, multiply, and inhibit C. gloeosporioides ATCC 42374 when tested in a mango leaf assay, showing its potential for fungal biocontrol. Additional studies are required to definitively identify the active isolates and to determine their mode of antifungal action, safety, and biocompatibility.  相似文献

An oxidative and SDS-stable alkaline protease secreted by a marine haloalkalophilic Bacillus clausii isolated from the tidal mud flats of the Korean Yellow Sea near Inchon City was investigated in batch fermentation in shake flasks and in a bioreactor under a range of conditions. The isolate produced maximum protease yields (15,000 U ml−1) under submerged fermentation conditions at 42 °C for 40 h with an aeration of 1.5 v/v/min and agitation of 400 rev/min in a formulated soybean—casein medium (pH 9.6) containing (w/v): soybean meal (2%), casein (1%), corn starch (0.5%), NH4Cl (0.05%), NaCl (0.05%), KH2PO4(0.04%), K2HPO4(0.03%), MgSO4(0.02%), yeast extract (0.01%) and Na2CO3(0.6%). The optimal pH and temperature of activity of the partially purified enzyme were 11.5 and 80 °C, respectively. The alkaline protease showed extreme stability towards SDS and oxidizing agents, retaining its activity above 96 and 75% on treatment for 72 h with 5% SDS and 5% H2O2, respectively. The inhibition profile exhibited by phenylmethanesulphonyl fluoride suggested that the protease from B. clausii belongs to the family of serine proteases.  相似文献

Two new pyripyropenes, 13‐dehydroxy‐1,11‐deacetylpyripyropene A ( 1 ) and 1‐deacetylpyripyropene A ( 2 ), together with six known compounds, were isolated from a marine fungus Fusarium lateritium 2016F18‐1 which was associated with the sponge Phyllospongia foliascens. Their structures were established mainly based on NMR and MS data. Their cytotoxic activities against human cancer cells CNE1, CNE2, HONE1, SUNE1, GLC82, and HL7702 were evaluated.  相似文献

The red alga Laurencia nipponica comprises various chemical races distributed relative to the ocean current in Japanese coastal areas. We investigated the chemical compositions and chemical races of L. nipponica distributed from the Kunashiri and Etorofu Islands, the confluence of the Soya warm current and Oya-shio cold current. Two new halogenated secondary metabolites, deacetylneonipponallene ( 1 ) and neopacifenol ( 2 ), along with four known compounds, deoxyprepacifenol ( 3 ), pacifenol ( 4 ), halo-chamigrene diether ( 5 ), and isolaurallene ( 6 ) were isolated from L. nipponica collected at Chikappunai, Kunashiri Island, while Zaimokuiwa (Kunashiri Island) and Sana (Etorofu Island) populations contained 3 , 7-hydroxylaurene ( 7 ), 2,10-dibromo-3-chloro-9-hydroxy-α-chamigrene ( 8 ), and (3Z)-laurefucin ( 9 ). The structures of 1 and 2 were established using spectroscopic methods. The chemical races of L. nipponica distributed in this area were divided into 6 - and 9 -producing races. Interestingly, both races contained 4 as an additional race-index, as well as its derivatives, 2 and 5 . To the best of our knowledge, this is the first example of a race comprising a mixture of two race-index compounds, suggesting that the convergence of two currents causes the production of new and diverse chemical races in this species.  相似文献

 This study furthers our understanding of the role of socioeconomics in coral reef management by demonstrating the importance, as well as means, of incorporating socioeconomic information into coral reef management. A case study analysis was made of the socioeconomic context of the three primary user groups in Montego Bay Marine Park, Jamaica: fishers, hoteliers and watersports operators. The primary means of data collection were document analysis, interviews with individuals representative of user groups, focus groups and participant observation. The results regarding user’s awareness, user groups’ relations with the managing agency, relations among and within user groups, resource use patterns, and sociocultural values demonstrate the need to: increase public awareness of the benefits of the Park, increase user awareness of Park management activities, increase user involvement in Park management, and develop intersectoral coordination. These findings provide useful guidance for future Park management and, in a more general context, illustrate the importance of socioeconomic assessments for reef management. Recent studies suggest that lysozyme, rich in hen egg, has an antitumor function. In the present study, we investigated the antitumor and antiangiogenesis effects of a newly isolated marine lysozyme both in vitro and in vivo. First, we showed that this marine-derived lysozyme specifically inhibits the proliferation of endothelial cells (ECV304) in a dose-dependent manner with no cytotoxicity (IC50 = 3.64 μM). Second, we showed that this marine lysozyme directly suppresses neovascularization in chicken embryos using chorioallantoic membrane assay. Third, we demonstrated that this marine lysozyme markedly inhibits tumor growth in mice bearing either sarcoma 180 or hepatoma 22. Unexpectedly, hen egg lysozyme has no effects on the proliferation of endothelial cells in vitro or neovascularization in chicken embryos or tumor growth in nude mice at the same dosage range. Taken together, our studies clearly show that the newly identified marine lysozyme is a potent antitumor molecule, which may inhibit tumor growth and inhibit angiogenesis. We believe that this marine lysozyme may have a therapeutic value in antitumor drug development.  相似文献

Two metabolites enalin A (1) and B (2), together with hydroxymethyl furfural (3) and three cyclodipeptides (4, 5 and 6), were isolated from the mangrove fungus Verruculina enaria from a salt lake in the Bahamas. 为获得能明显抑制水华蓝藻的化感物质,本研究从28株大型海藻内共生真菌中筛选出了4株菌株,其粗提物能显著抑制水华鱼腥藻(Anabaenaflos-aquae)和铜绿微囊藻(Microcystis aeruginosa)的生长。对这4株真菌扩大培养后,检测了从其次生代谢产物中分离出的32种单体化合物的抑藻活性。 其中, 化合物sterigmatocystin、(2’E,4’E,6’E)-6-(1’-carboxyocta-2’,4’,6’-triene)-11,12-epoxy-9,11-dihydroxydrim-7-ene、 6,8-di-O-methylnidurufin和aversin对水华鱼腥藻的抑制率显著大于50%(P 〈 0.05), 24 h半数有效浓度(EC50,24h)分别为6.865、4.302、1.824、2.319 μg/mL;sterigmatocystin、indol-3-carboxaldehyde和(2’E,4’E,6’E)-6-(1’-carboxyocta-2’,4’,6’-triene)-11,12-epoxy-9,11-dihydroxydrim-7-ene对铜绿微囊藻的抑制率显著大于50%(P 〈 0.05), EC50,24h 分别为4.167、5.374、8.386 μg/mL。  相似文献

From the dried leaves of Ohwia caudata, two new compounds, namely (4E)-(4-hydroxyphenyl)-3-butenoic acid butyl ester ( 1 ), and 4-benzyl-1,3-phenylenedicarbamic acid methyl ester ( 2 ), together with five known compounds, were isolated and identified. The structures of compounds 1 and 2 were established using 1D-NMR, 2D-NMR and HR-ESI-MS spectral analysis. Previous studies on O. caudata had been reported to protect against Alzheimer's disease, two new compounds were evaluated for their neuroprotective effect against lipopolysaccharide-induced BV2 microglia cells. The result indicated two compounds showed well anti-neuroinflammatory activity at 12.5 μM.  相似文献

Cell walls of brown algae are complex supramolecular assemblies containing various original, sulfated, and carboxylated polysaccharides. Among these, the major marine polysaccharide component, alginate, represents an important biomass that is successfully turned over by the heterotrophic marine bacteria. In the marine flavobacterium Zobellia galactanivorans, the catabolism and uptake of alginate are encoded by operon structures that resemble the typical Bacteroidetes polysaccharide utilization locus. The genome of Z. galactanivorans contains seven putative alginate lyase genes, five of which are localized within two clusters comprising additional carbohydrate-related genes. This study reports on the detailed biochemical and structural characterization of two of these. We demonstrate here that AlyA1PL7 is an endolytic guluronate lyase, and AlyA5 cleaves unsaturated units, α-l-guluronate or β-d-manuronate residues, at the nonreducing end of oligo-alginates in an exolytic fashion. Despite a common jelly roll-fold, these striking differences of the mode of action are explained by a distinct active site topology, an open cleft in AlyA1PL7, whereas AlyA5 displays a pocket topology due to the presence of additional loops partially obstructing the catalytic groove. Finally, in contrast to PL7 alginate lyases from terrestrial bacteria, both enzymes proceed according to a calcium-dependent mechanism suggesting an exquisite adaptation to their natural substrate in the context of brown algal cell walls.  相似文献

A new polychlorinated triphenyl diether named microsphaerol ( 1 ), has been isolated from the endophtic fungus Microsphaeropsis sp. An intensive phytochemical investigation of the endophytic fungus Seimatosporium sp., led to the isolation of a new naphthalene derivative named seimatorone ( 2 ) and eight known compounds, i.e., 1‐(2,6‐dihydroxyphenyl)‐3‐hydroxybutan‐1‐one ( 3 ), 1‐(2,6‐dihydroxyphenyl)butan‐1‐one ( 4 ), 1‐(2‐hydroxy‐6‐methoxyphenyl)butan‐1‐one ( 5 ), 5‐hydroxy‐2‐methyl‐4H‐chromen‐4‐one ( 6 ), 2,3‐dihydro‐5‐hydroxy‐2‐methyl‐4H‐chromen‐4‐one ( 7 ), 8‐methoxynaphthalen‐1‐ol ( 8 ), nodulisporins A and B ( 9 and 10 , resp.), and daldinol ( 11 ). The structures of 1 and 2 were elucidated by detailed spectroscopic analysis including 1H‐ and 13C‐NMR, COSY, HMQC, HMBC, and HR‐EI‐MS, while the structures of the known compounds were deduced from comparison of their spectral data with those in the literature. Preliminary studies revealed that microsphaerol ( 1 ) showed good antibacterial activities against B. Megaterium and E. coli, and good antilagal and antifungal activities against C. fusca, M. violaceum, respectively. On the other hand, seimatorone ( 2 ) exhibited moderate antibacterial, antialgal, and antifungal activities.  相似文献
