Seven out of nine Penicillium isolates from mouldy maize in Yugoslavia have been differentiated into the adjacent species P. aurantiogriseum and P. commune. Nephrotoxicity of cultured mycelia in the rat has been demonstrated for all isolates of both species and was correlated usefully, though indirectly, with the production of benzodiazepine secondary metabolites, notably auranthine. Shredded wheat (22 g) moulded by an example of each species and fed to a rat over 4 days elicited renal pathology in the P3 segment of proximal tubules, involving frequent pyknosis and extensive mitosis typical of this as yet uncharacterised toxin. The effect was attributed in P. aurantiogriseum at least partly to the spores. Prominent pathology was elicited by only lg of spores given over 4 days.  相似文献

Extracts of 14 Penicillium species, P. aurantiogriseum, P. brevicompactum, P. citrinum, P. chrysogenum, P. expansum, P. glabrum, P. hirsutum, P. italicum, P. janthinellum, P. melini, P. oxalicum, P. purpurescens, P. simplicissimum, and P. viridicatum were investigated by total protein, specific enzyme determinations, isoelectric focusing (IEF), sodium dodecylsulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting using pooled human atopic IgE. Considerable variation was observed between the Penicillium species with respect to protein yield and the number of distinct protein bands resolved in IEF. Using the Api-Zym system, the most common activities observed among the extracts included acid and alkaline phosphatase, phosphodiamidase and β-glucosaminidase. The number of discrete atopic IgE-reactive bands in immunoblots of Penicillium extracts ranged from 1 (P. chrysogenum) to 9 (P. viridicatum). Certain allergens showed potential for cross-reactivity between species, including 52 and 54 kDa proteins in P. citrinum, P. purpurescens, P. viridicatum and 40 kDa proteins in several species. The extracts were also nonmutagenic when tested with the Ames assay using Salmonella strains TA98 and TA100. The results tend to indicate that P. viridicatum, P. janthinellum, P. oxalicum, P. brevicompactum and P. italicum, which are highly immunogenic as well as allergenic, could possibly be good candidates for allergen cloning studies through the construction of cDNA libraries. Penicillium glabrum, P. purpurescens, P. decumbens, P. expansum, P. chrysogenum, P. crustosum andP. aurantiogriseum are responsible for some of the alterations noticed on Moroccan table grapes cold stored. Contamination of these table grapes withPenicillium species occurs in the vineyard and inside the cooling station where other fruits which are often fungus-contaminated are also kept. All of thesePenicillium species were able to grow at 0°C apart fromP. glabrum andP. purpurescens. Consequently, refrigeration of grapes during long-term storage is not sufficient in itself in preserving their initial qualities.  相似文献

Penicillium andAspergillus have been recognized as important aeroallergens for more than 30 years, and are especially significant in indoor environments. There are over 400 species ofPenicillium andAspergillus combined, but there is little information on which species occur most frequently in the environment, or if each exhibits unique allergenic properties. A preliminary study showed no overlap between those species isolated from an outdoor site in Tulsa, Oklahoma and the species used in immunotherapy at allergy clinics in the Tulsa area. Pursuing this line of research, air samples were collected as three seasonal samples (over a 6 month period) in the homes or offices of ten allergy patients known to be allergic toPenicillium and/orAspergillus. Twenty three species ofPenicillium and 12 species ofAspergillus were identified from these samples through isolation, macroscopic, and microscopic examination.Penicillium corylophilum, P. glabrum, Aspergillus niger, andA. flavipes were the most abundant species isolated, supporting the data obtained in a preliminary study. At least in the Tulsa area, it appears that atopic patients are being tested and treated with extracts ofPenicillium andAspergillus species that are either not present or not abundant in the local indoor or outdoor environments. Additional research is necessary to determine if the environmental isolates share allergens with those species used in immunotherapy.  相似文献

Production of fungitoxic extrolites was evaluated in culture filtrates of several isolates belonging to Penicillium canescens and P. janczewskii that showed some extent of inhibitory activity against the plant pathogenic fungus Rhizoctonia solani. In addition to griseofulvin and dechlorogriseofulvin that are already known in these species, curvulinic acid, previously unreported in Penicillium, was produced by all isolates assayed. Another extrolite recently characterized from a P. verrucosum strain by the name of Sch 642305 was detected in 5 isolates of P. canescens only. The purified compound completely inhibited mycelial growth of isolates of Rhizoctonia solani and other plant pathogenic fungi in␣vitro. The role of this extrolite as a possible biochemical determinant of antagonism toward plant pathogenic fungi, and implications concerning chemotaxonomy are discussed.  相似文献

Several strains of the genusPenicillium isolated from swabs from uranium miners working environment were screened for production of known mycotoxins; and embryotoxicity of chloroform extracts from the isolates was investigated in chick embryos.Penicillium aurantiogriseum was found to produce chaetoglobosin A. ED50 of this mycotoxin for 2-, 3- and 4-day-old chick embryos was found 0.040 (0.031–0.052), 0.074 (0.051–0.107), and 0.180 (0.113–0.229) µg/embryo, respectively. The effect was purely embryolethal with no signs of teratogenicity recorded. This is the first report on the isolation of chaetoglobosin A from the genusPenicillium.  相似文献

Aims: To assess the ability of fungi isolated from grapes to produce patulin and citrinin. Methods and Results: A total of 446 Aspergillus isolates belonging to 20 species and 101 Penicillium isolates were inoculated in Czapek yeast extract agar and yeast extract sucrose agar and incubated for 7 days at 25°C. Extracts were analysed for patulin and citrinin by thin‐layer chromatography. None of the isolates of Aspergillus spp. produced either patulin or citrinin. Patulin was produced by three isolates of Penicillium expansum and two of Penicillium griseofulvum. Citrinin was produced by five isolates of P. expansum, two of Penicillium citrinum and one of Penicillium verrucosum. Conclusions: Our results show that the Aspergillus and Penicillium species commonly isolated from grapes are not a source of the mycotoxins, patulin and citrinin. Significance and Impact of the Study: The possibility of co‐occurrence of patulin and citrinin with ochratoxin A in grapes and grape products remain low, owing to the low frequency of isolation of potentially producing species.  相似文献

Fungi have been only rarely isolated from glacial ice in extremely cold polar regions and were in these cases considered as random, long-term preserved Aeolian deposits. Fungal presence has so far not been investigated in polar subglacial ice, a recently discovered extreme habitat reported to be inhabited exclusively by heterotrophic bacteria. In this study we report on the very high occurrence (up to 9000 CFU L−1) and diversity of filamentous Penicillium spp. in the sediment-rich subglacial ice of three different polythermal Arctic glaciers (Svalbard, Norway). The dominant species was P. crustosum, representing on the average half of all isolated strains from all three glaciers. The other most frequently isolated species were P. bialowiezense, P. chrysogenum, P. thomii, P. solitum, P. palitans, P. echinulatum, P. polonicum, P. commune, P. discolor, P. expansum, and new Penicillium species (sp. 1). Twelve more Penicillium species were occasionally isolated. The fungi isolated produced consistent profiles of secondary metabolites, not different from the same Penicillium species from other habitats. This is the first report on the presence of large populations of Penicillium spp. in subglacial sediment-rich ice.  相似文献

The virulence of two isolates of the hyphomycete fungi, Beauveria bassianaand B. brongniartii, and additional fungal species isolated from diseased Bactrocera oleae pupae and Sesamia nonagrioideslarvae were assessed against adults of the olive fruit fly B. oleae and the Mediterranean fruit fly Ceratitis capitata (Diptera: Tephritidae). Contact and oral bioassays revealed that moderate to high mortality rates for the olive fruit fly occurred when the adults were exposed to conidia of Mucor hiemalis, Penicillium aurantiogriseum, P. chrysogenum and B. bassianaisolates. A strain of M. hiemalis isolated from S. nonagrioides larvae was the most toxic resulting in 85.2% mortality to the olive fruit fly adults. B. brongniartiiand B. bassiana were the most pathogenic to the C. capitataadults causing 97.4 and 85.6% mortality. Metabolites collected from the M. hiemalis and P. chrysogenum isolates were toxic to adults of both species.  相似文献

Decayed wood from fallen white spruce (Picea glauca) and trembling aspen (Populus tremuloides) collected in northeastern Alberta, Canada, was the source of new isolates of species in the ascomycete generaGymnostellatospora andPseudogymnoascus. In addition to new reports ofG. japonica, G. frigida andP. roseus, two new species are described.Gymnostellatospora canadensis sp. nov. resemblesG. japonica but differs in producing brown ascomata and in the formation of an arthroconidial anamorph.Gymnostellatospora subnuda sp. nov. is distinct in lacking differentiated peridial hyphae.Gymnostellatospora alpina was not found in decayed wood but is reviewed based on extralimital material. 本文报导青霉属的三个新种,它们不同于此属已发表的诸种.异型青霉(Penicillium heteromorphum Kong et Qi),根据帚状枝的结构和分生孢子的某些特征,近于两型孢青霉(Penicillium dimorphosporum Swart),但此种在查氏琼脂上生长很稀疏,并有两种大小的分生孢子,大分生孢子直径可达10 pm,壁光滑,在观察菌落期间,有时大分生孢子落在基质上,很快萌发生长成简单的分生孢子结构,这些特征易于与后者区分.结节青霉(Penicillium nodulum Kong et Qi)在某些方面类似纠缠青霉(Penicilliumimplicatum Biourge),此新种的菌落特征,柄顶端膨大似顶囊,呈结节状膨大的营养菌丝等特征而不同于纠缠青霉.神农架青霉(Penicillium shennong jianum Kong et Qi)和毡毛青霉(Penicillium velutinum van Heyma)相近,而新种在多种培养基上生长都局限,菌落面呈带褐的橄榄色,分生孢子壁光滑等特征可与毡毛青霉区分开来.  相似文献

Eight of eleven ochratoxigenic isolates of Penicillium nordicum and Penicillium verrucosum produced guttation droplets when grown on Czapek yeast extract (CYA) agar for 10–14 days at 25°C. Parallel cultivation of one strain each of P. nordicum and P. verrucosum on malt extract agar demonstrated that higher volumes of exudate are produced on this agar. However, HPLC analyses revealed higher concentrations of ochratoxin A (OTA) and B (OTB) in droplets originating from cultures on CYA. For quantitative determination of the mycotoxin contents, triplicates of three isolates each of P. nordicum and P. verrucosum were grown as single spot cultures on CYA for up to 14 days at 25°C. Guttation droplets were carefully collected between day 11 and 14 with a microliter syringe from each culture. Extracts from exudates and corresponding mycelia as well as fungal free agar were analyzed by HPLC for the occurrence of ochratoxin A (OTA) and B (OTB). Mean concentrations ranging between 92.7–8667.0 ng OTA and 159.7–2943.3 ng OTB per ml were detected in the guttation fluids. Considerably lower toxin levels were found in corresponding samples of the underlying mycelia (9.0–819.3 ng OTA and 4.5–409.7 ng OTB/g) and fungal free agar (15.3–417.0 ng OTA and 12.7–151.3 ng OTB/g). This is the first report which shows that high amounts of mycotoxins could be excreted from toxigenic Penicillium isolates into guttation droplets.  相似文献

Penicillium spp. are well known to produce a variety of beneficial metabolites for plant growth and survival, as well as they defend their hosts from attack of certain pathogens. In this study, effects of culture filtrate of different Penicillium spp. were tested on tomato seeds. On the whole, presoaking of seeds in the filtrates of the nine Penicillium isolates tested significantly increased seed germination when compared with the control seeds. Cultural extracts of P. expansum and P. bilaii were highly effective in growth promotion up to 90%. It was also observed that P. implicatum and P. oxalicum significantly enhanced the root growth in tomato seedling as compared to other species. In case of shoot length, P. verrucosum (3.38 cm), P. granulatum (2.81 cm) and P. implicatum (2.62 cm) were effective. P. implicatum was quite promising in increasing shoot and root length in tomato seedlings, while P. simplicissimum and P. citrinum were less effective on seedling growth. The plant growth-promoting ability of Penicillium strains may help in growth promotion in other plants and crops. Penicillium spp. are already known for producing various mycotoxins and enzymes. Plant growth-promoting ability of Penicillium spp. will open new aspects of research and investigations. The role of Penicillium spp. in tomato plant growth requires further exploration.  相似文献

A number of Penicillium isolates were recovered in association to Rhizoctonia solani strains pathogenic on tobacco and from soil on plates pre-colonized by the pathogen itself. Their antagonism toward R. solaniAG-2-1 was evaluated in dual cultures in vitro. Inhibition of growth was evident to some extent in most pairings, while hyphal interactions referable to mycoparasitic relationships were not observed. However, the occurrence of plasmolysis and/or vacuolisation and the induction of monilioid cells were indicative of the release of bioactive compounds. Therefore, production of fungitoxic metabolites was tested by adding concentrated culture filtrates of each Penicillium isolate to the growth medium of R. solani. Complete and lasting inhibition was incited by culture filtrates of some isolates belonging to P. brevicompactum, P. expansum, and P. pinophilum. Three purified compounds, respectively mycophenolic acid, patulin and 3-O-methylfunicone, which were extracted from culture filtrates, were able to inhibit R. solani in vitro. Their production was also detected in dual cultures of the same Penicilliumstrains with R. solani prepared in sterilized soil and when the Penicilliumstrains were cultured directly on R. solani mycelium harvested from liquid cultures. The possible role of such metabolites in antagonism of the above-mentioned Penicilliumspecies against R. solani is discussed.  相似文献

Corn (Zea mays) is the main cereal produced in and exported from Argentina. The risk of contamination by mycotoxins is related to the mycoflora associated with the corn kernels. This paper reports on the identification of internal and external mycoflora of corn kernels harvested in the main production area in Argentina in 1990. A mycological survey was carried out on 178 corn samples, from five locations in that area and the isolation frequency and relative density of the prevalent fungal genera compared. GenusFusarium was the most prevalent component of the internal seedborne mycoflora in the five locations.Penicillium was prevalent in all locations, taking into account the frequency. However, this genus was predominant only in two locations, when the relative density was considered. The predominantFusarium wasF. moniliforme and the most frequently isolated species ofAlternaria, Aspergillus andPenicillium wereA. alternata, A. flavus andP. decumbens, respectively.Diplodia species were not isolated from any of the samples.  相似文献

Forty-four fungal species belonging to 20 genera were isolated from 30 samples of qat leaves. The most frequent genera wereAspergillus, Alternaria, Penicillium, andCladosporium followed byFusarium, Drechslera, Chœtomium, andMucor. The most prevalent species in above genera wereAspergillus niger, A. flavus, A fumigatus, Alternaria alternata, Penicillium chrysogenum, P. citrinum, Cladosporium cladosporioides, andFusarium verticillioides. From these fungi, 17 species (39%) related to 7 genera (35%) proved to be true endophytes. Eleven out of 75 isolates were mycotoxigenic.A. alternata produced alternariol and alternariol monomethyl ether whereasA. flavus produced aflatoxins B1 and B2. Ochratoxin A, sterigmatocystin, citrinin and T-2 toxin were produced byA. ochraceus, A. versicolor, P. citrinum andF. oxysporum, respectively. The presence of such toxigenic fungi associated with qat leaves is considered to be a threat to public health.  相似文献

Imazalil tolerant isolates ofPenicillium spp. were recovered from sampling natural spore populations in storage rooms and apples collected from packinghouses in Lleida (Spain). ThePenicillium resistant strains belong to the speciesP. cyaneofulvum, P. variabile, P. rugulosum, P. minioluteum andP. pinophilum. 85% of the tested strains were resistant to imazalil in final concentrations of imazalil ranging from 4,500 µg/ml to 11,000 µg/ml. The resistance of these moulds to this fungicide was constant during successive subcultures. 89% ofPenicillium studied strains produced decay in the determination of parasitic fitness at 10 days.  相似文献

A system is described for identifying grain-inhabiting nephrotoxicPenicillium spp. based on their colony characters on Czapek yeast extract agar, yeast extract sucrose agar, and malt extract agar media, and their secondary metabolite profiles on thin layer chromatography plates. Using this system, the identity of 11Penicillium species, or their chemotypes, producing nephrotoxic metabolites could be confirmed. The species areP. verrucosum chemotype I, P.verrucosum chemotype II,P. expansum, P. citrinum, P. aurantiogriseum, P. freii, P. tricolor, P. polonicum, P. viridicatum, P. cyclopium, and P. melanoconldlum. Other non-nephrotoxicPenicillium species present on stored grains were separated from nephrotoxic species by their colony characters and metabolite profiles.  相似文献

Patterns of nodulation, growth, andFrankia — host specificity have not been well characterized for the actinorhizal genera in the family Rosaceae because of the scarcity ofFrankia isolates from these taxa. Furthermore, the few isolates available from actinorhizal Rosaceae have consistently failed to nodulate plants from the host genus. In a series of experiments, species of rosaceousDryas, Cowania, Cercocarpus, Fallugia, andPurshia were inoculated withFrankia isolates, crushedDryas actinorhizae, and neoglacial soils to ascertain whether any of these inocula would effectively induce nodulation. Neoglacial soils from Alaska and Canada nodulated not only the localDryas drummondii, but alsoCercocarpus betuloides, Cowania mexicana andPurshia tridentata from distant and ecologically diverse locales as well as nonrosaceous, actinorhizal species ofAlnus, Elaeagnus, Myrica, andShepherdia. But of eightFrankia isolates, including two fromPurshia tridentata and one fromCowania mexicana, none were able to induce nodulation onPurshia orCowania species. Globular, actinorhizae-like nodules incapable of acetylene reduction were produced onC. betuloides inoculated withFrankia isolates. Crushed nodule suspensions fromDryas drummondii nodulated rosaceousCowania, Dryas andPurshia, as well as non-rosaceousElaeagnus, Myrica, andShepherdia species. Nodules produced by inoculation ofCowania mexicana andPurshia tridentata with crushed, dried nodule suspensions fromDryas drummondii reduced acetylene to ethylene, indicating nitrogenase activity for these nodulated plants. These data suggest that a similar microsymbiont infects the actinorhizal genera in the family Rosaceae.  相似文献
