Fat mobilyzing activity of synthetic tetradecapeptide, which corresponds in 31--44 amino acid sequence of human growth hormone, is studied in vitro in human fat tissue. The peptide at concentrations of 3--33 microng/ml considerably stimulated lipolysis in subdermal fat tissue, omentum and shoulder ateroma. Minimal efficient peptide concentration was 3 microng/ml in most experiments, sometimes it was 0.3 microng/ml. Direct dependency between dose logarithm and lipolysis rate was observed at dose interval of 0.3--18 microng/ml. Native growth hormone produced no activity in human fat tissue even in concentrations of 50--100 microng/ml.  相似文献

The influence of ACTH fragments, possessing structural elements, common for certain groups of peptide hormones and kinins--"common" fragments and cluster of basic amino-acids--(Lys 17,18-ACTH 11-18-NH2--I; ACTH 11-13-NH2--II; NH2CO-ACTH18-20-NH2--III) on lipolytic effect of ACTH in rat isolated fat cells and on the steroidogenic effect of ACTH in isolated rat adrenal cells was studied. Fragment I exerts a steroidogenic effect (alpha=0,84) at concentrations of 1--100 microng/ml. At low concentrations (10(-8)--10(-3) microng/ml) fragment I potentiates ACTH-induced steroidogenesis. Fragment I has no effect on the lipolysis;however, it potentiates ACTH-induced lipolysis at concentrations of 10--100 microng/ml. The results obtained support our previous supposition that "common" fragments are essential secondary non-specific active sites of hormones.  相似文献

The authors studied the effect of adrenotropic substances on lipolysis in rat epididymal adipose tissue in albumin medium in vitro. On using albumins of different origin (human, bovine), the pD2 values for catecholamines differed by more than one order, in correlation to the type of albumin used. The isopropylnorsynephrine pD2 values did not differ. The addition of ascorbic acid (100 microng/ml) raised the catecholamine pD2 values and completely equalized the pD2 values found in both media. The pD2 values for the synephrine derivative did not alter. The propranolol pA2 values were not negatively affected by the addition of ascorbic acid. Ascorbic acid also produced a mild increase in the maximum lipid-mobilizing values obtained with any of the given substances in either medium. It was concluded in the discussion that catecholamines are oxidized at different rates in different albumin media and that this oxidation can be inhibited by adding ascorbic acid. Ascorbic acid likewise mildly stimulates the maximum lipid-mobilizing effect. The authors recommend the addition of ascorbic acid to albumin medium as a regular component for the study of adrenergic lipid mobilization.  相似文献

The influence of proteinase inhibitors on the lipolytic effect of the pituitary polypeptide hormones and epinephrine in an isolated adipose tissue of rabbits and rats has been studied. Neither of proteinase inhibitors changed the basal rate of lipolysis. Trasylol, a serine proteinase inhibitor, suppressed completely growth hormone (GH) effect and partially reduced the effect of adrenocorticotropin (ACTH) and beta-lipotropin (beta-LPH) but did not change the effect of epinephrine. Bacitracin proved ineffective with regard to the effect of polypeptide hormones. Pepstatin, an acid proteinase inhibitor, partially blocked the stimulation of lipolysis by ACTH without affecting the effect of GH and beta-LPH. The influence of proteinase inhibitors on the ACTH effect in rat adipose tissue was similar to that found in rabbit tissue. The Trasylol-induced inhibition of the hormone-stimulated lipolysis decreased to a considerable extent after GH or ACTH incubation with rabbit plasma or partial GH digestion with pepsin. This decrease was not observed when plasma serine proteinases were blocked during GH incubation with plasma. The results demonstrate an involvement of some proteolytic enzymes in the realization of the polypeptide hormone lipolytic effect and permit to suppose the requirement of preliminary activation of the hormones by means of proteolytic modification.  相似文献

Three different insulin-like effects of growth hormone were studied in segments of adipose tissue obtained from hypophysectomized rats. The onset of each response was preceded by a characteristic lag period: antilipolysis was seen only after a 10--15 minute exposure to growth hormone; stimulation of glucose oxidation was significant 20 minutes after exposure to growth hormone and increased leucine oxidation was seen only after 30 minutes. Each of the responses was measurable without a detectable delay when the tissues were exposed to hormone during a prior incubation period. Accelerated leucine oxidation was detected when 0.01 microgram/ml growth hormone was present in the incubation medium; the other responses required a minimum of 0.1 microgram/ml. Inhibitors of protein synthesis of concentrations which decreased the incorporation of 14C leucine into protein by 99% had no effect on either the antilipolytic action of growth hormone or the stimulatory action on glucose oxidation, but abolished the acceleration of leucine oxidation. In contrast to findings in diaphragm muscle, theophylline was without effect on any of the insulin-like actions of growth hormone in adipose tissue, even though it decreased the basal rate of glucose and leucine oxidation.  相似文献

Bovine diabetogenic protein has been further purified by gel filtration yielding a fraction (Mr 25 000–28 000) having increased diabetogenic and in vitro lipolytic activity. Using rat epididymal fat pads, this fraction was shown to be lipolytic at concentrations as low as 1–10 μg/ml. The in vitro lipolytic effect was unaffected by the nutritional state of the animals, was not potentiated by dexamethasone, could be demonstrated in the presence and absence of glucose and was not mediated by α- and β-adrenergic receptors. A lag phase of > 1 h was observed before diabetogenic protein induced lipolysis occurred, suggesting that protein synthesis might be involved. Cycloheximide (10 μg/ml), added initially, prevented the diabetogenic protein-induced lipolysis. This direct effect of the purified protein on adipose tissue helps explain the elevation of free fatty acids seen when bovine diabetogenic hormone is administered in vivo and suggests that this anterior pituitary protein may be a new lipid-mobilizing hormone.  相似文献

The time course for the decrease in norepinephrine concentration of rat pineal explants in culture indicated a significant fall starting at the 4th hour and completed after 16-24 h of incubation. Significant decreases of serotonin and 5-hydroxyindoleacetic acid (HIAA) levels in tissue, an increase of HIAA/serotonin ratio, and an increase of melatonin production rate in vitro were also observed as a function of the incubation time. Estradiol (10(-7)-10(-5) M) increased rat pineal melatonin content, testosterone (10(-5) M) decreased it and progesterone was devoid of activity when incubated with explants for up to 6 h. The in vitro stimulatory effect of estradiol on rat pineal methoxyindole synthesis was blocked by propranolol but not by phentolamine; propranolol also blocked the increase of nuclear estradiol-receptor complex produced by estrogen exposure of pineal explants. TSH (1-100 ng/ml), growth hormone (10-100 ng/ml) and LH (10 ng/ml) augmented rat pineal melatonin content while 100 ng/ml of FSH decreased it significantly. Prolactin exerted a biphasic effect on rat pineal explants, the lowest concentration augmenting melatonin content while the high concentration depressed it. Deep, intermediate and superficial segments of guinea-pig pineal glands showed an increase in melatonin concentration after a 6-h incubation in the presence of 10(-7)-10(-5) M estradiol.  相似文献

The effect of bovine growth hormone on adenylate cyclase activity was studied in bovine and rat renal medulla. Highly purified growth hormone (lot B1003A) increased adenylate cyclase activity in plasma membranes from bovine renal medulla from 132+/-6 pmol cyclic AMP formed/mg protein per 10 min to 364+/-10 pmol cyclic AMP formed/mg protein per 10 min. Similar results were seen with homogenates of rat renal medulla. The minimum effective concentration of bovine growth hormone required to activate adenylate cyclase was 0.5 mug/ml and maximum activation was detected at 500 mug/ml. The amount of vasopressin determined by radioimmunoassay to contaminate the growth hormone caused an increase in adenylate cyclase activity comparable to that of the corresponding concentration of growth hormone that was tested. Dialysis of growth hormone and vasopressin resulted in parallel reductions in the effect of each hormone on adenylate cyclase activity. Similarly, both growth hormone and vasopressin produced increases in short circuit current in isolated toad bladders but these effects were not detectable after dialysis of the hormones. In contrast, the effect of growth hormone on the uptake of 35SO2-4 by cartilage from hypophysectomized rats was not decreased after dialysis. These results indicate that available preparations of growth hormone are contaminated by small but physiologically significant amounts of vasopressin and that the activation of adenylate cyclase activity in renal medulla in response to growth hormone can be explained by this contamination rather than by an effect of growth hormone per se.  相似文献

The in vitro effect of growth hormone on adipose tissue lipoprotein lipase was studied in rats. Epididymal adipose tissue was incubated with human growth hormone in the presence of heparin. Growth hormone at a concentration of 0.1 μg per ml decreased by approximately 20% (p<0.005) the heparin-releasable lipoprotein lipase in rat adipose tissue. Discussion was focussed on the reciprocal changes caused by growth hormone of the activities of lipoprotein lipase and hormone-sensitive lipase in the rat adipose tissue.  相似文献

The influence of fragment ACTH 11-14 analogues with amino acid sequences H-Lys-Pro-Val-Gly-OH (fragment I) and H-Lys-Pro-Val-Gly-NH2 (fragment II), possessing structural elements, similar for certain groups of peptide hormones and kinins, on lipolytic effect of ACTH in adipose tissue and isolated epydidymal fat cells of rat, was studied. Both fragments have no effect on the lipolysis; they potentiate the ACTH-induced lipolysis 1,5--2,0 fold, but do not alter the maximal effect at concentrations 0,1--1,0 mkg/ml in tissue and fragment I--at concentrations from 0,01 to 0,1 mkg/ml in isolated fat cell system. The role of "common" fragments in hormone-receptor interactions as well as mechanism of their potentiating effect is discussed. It is assumed that the "common" fragment of ACTH--ACTH11-14--is a second, non-specific active site of hormone directly involved in secondary signal formation.  相似文献

Growth stimulating activity of the molecular weight 6,500-peptide purified from rat pancreatic juice was measured on 3T3 fibroblasts. This peptide was reported to be a cholecystokinin-releasing peptide and to stimulate pancreatic enzyme secretion in the rat small intestine in response to food intake. Incorporation of [3H]thymidine and [35S]methionine into 3T3 was significantly stimulated and the cell number was also increased after 24-48 hr incubation with 10-100 ng/ml of the peptide. The increase in [3H]thymidine incorporation was dose-related and started 12 hr after the incubation, a peak being reached 24 hr after the incubation. These results show that this peptide exhibits growth stimulating activity to the mammalian cells, and suggest that the peptide might have a physiological effect in vivo.  相似文献

Both adipokinetic hormone and octopamine have a stimulating effect on lipid release from locust fat body in vitro, when incubated in diluted haemolymph. The presence of adipokinetic hormone results in the formation of the flight-specific haemolymph lipoprotein A+ accepting the increased amount of lipids released into the incubation medium. In contrast, interconversions of lipoproteins do not occur when octopamine is added to the incubation medium, which is in line with the expectations: the lipid-mobilizing effect of octopamine is a limited and short-term effect. When fat body tissue is incubated with isolated haemolymph protein fractions, the lipid-mobilizing effect of adipokinetic hormone only occurs when the incubation medium contains both lipoprotein, Ay and protein fraction C, resulting in the formation of lipoprotein A+. In similar control incubations with the hormone omitted, some lipoprotein A+ is also formed (concomitant with a slight amount of lipid released), though significantly less than in incubations with hormone. Besides a stimulating function on lipolytic processes in the fat body, adipokinetic hormone is suggested to influence haemolymph lipoprotein rearrangement. A possible counteracting function of another factor in the haemolymph is discussed.  相似文献

Ghrelin and synthetic growth hormone secretagogues have diverse effects on the hypothalamus including effects on appetite and the growth hormone axis as well as on the hypothalamus-pituitary-adrenal (HPA) axis. We previously studied the effect of synthetic growth hormone secretagogues on CRH and AVP release from rat hypothalami in vitro, and now report on the effects of ghrelin on CRH and AVP release. The ghrelin protein content and ghrelin output from rat hypothalamic explants was measured using a specific novel ghrelin enzyme immunoassay. The effect of 10(-8) M to 10(-6) M ghrelin on CRH and AVP release was studied in the rat hypothalamic explants, where stimulation with des-octanoyl ghrelin was used as control. The presence of both ghrelin mRNA and protein could be shown in the rat hypothalamus. Ghrelin output was detected in the incubation fluid of rat hypothalamic explants and could be stimulated with high potassium concentrations. Our data also demonstrated a dose-dependent effect of ghrelin on both CRH and AVP release, while des-octanoylated ghrelin showed no effect on either peptide. In summary, the current data suggest that ghrelin is expressed in the hypothalamus both at RNA and the protein levels. Ghrelin stimulates the HPA axis in the rat via stimulation of both CRH, and particularly, AVP release from the hypothalamus. The local autocrine/paracrine and endocrine effects of ghrelin in the hypothalamus could influence all the hormonal systems involved in ghrelin effects, including growth hormone release, the HPA axis and appetite.  相似文献

Antiserum YJC 13-31 against the rat ghrelin conjugated to bovine serum albumin (BSA) was produced in the rabbit and a double antibody radioimmunoassay (RIA) for ghrelin has been developed. Characterization results of this antiserum revealed no cross-reaction with human growth hormone and somatostatin. Weak cross-reactions with insulin (0.1%), rat growth hormone (0.1%) and glucagon (0.3%) were observed, which scarcely interfered the assay system. The sensitivity of this RIA was 5 pg per assay tube. With the rat serum samples, the within-assay precision was 7.1% and the between-assay precision was 12.3%. The RIA was also available to detect the ghrelin in rat tissue extracts with good parallelism to the rat ghrelin standard. In application, the serum ghrelin and corticosterone levels in weaned rats were measured by RIA. Gavage of saline was sufficient to raise serum ghrelin from 2.6 +/- 0.18 to 6.7 +/- 0.7 ng/ml (P < 0.01). Gavage with nonylphenol (NP) suppressed the elevation of serum ghrelin levels in a dose-dependent manner. Besides, gavages of saline elevated the serum levels of corticosterone from 108.8 +/- 13.5 to 188.7 +/- 23.5 ng/ml (P < 0.01) but the elevation effects of corticosterone from gavages were overcome by NP in the low dose of 50 mg/kg. It can be speculated that ingestion of NP is harmful to young animals during growth and environmental adaptation.  相似文献

The effects of colchicine and lumicolchicine on the ultrastructure of non-myelinated axons in cat autonomic nerves were studied using in vitro preparations of inferior mesenteric ganglion/hypogastric nerves. After 24 hrs of in vitro incubation with colchicine added to the medium (10 microng/ml) a significant decrease in number of neurotubules per 1 axon was observed. In the presence of a solution of alpha-beta and gamma-lumicolchicine (10 microng/ml) severe degenerative changes occured in axons and Schwann cells. At a lower dose of lumicolchicine (3 microng/ml) these changes were less frequent and the number of neurotubules per 1 axon did not differ from that in control nerves.  相似文献

The authors studied changes in adrenergic lipolysis in the epididymal adipose tissue of rats to which diethylstilboestrol and oestradiol combined with the anti-oestrogen clomiphene were administered. The maximum lipid-mobilizing effect of isoprenaline was increased not only by subcutaneously administered diethylstilboestrol, but also by the highest dose of the antioestrogen clomiphene used (p.o., 200 micrograms.kg-1 b.w.). Under the given experimental conditions, with 4 1/2 h incubation of adipose tissue, clomiphene was also effective when added in vitro. Its own oestrogenic effect probably stimulated the lipid-mobilizing action of isoprenaline. On combining the administration of increasing doses of clomiphene (p.o., 1-5 days) with a constant dose of oestradiol (200 micrograms.kg-1, s.c. on the 8th day, i.e. 24 h before the actual experiment), changes in isoprenaline lipolysis depended on the dose of clomiphene. In low doses clomiphene inhibited the stimulating effect of subsequently administered oestradiol on isoprenaline-induced lipolysis, but in large doses (100 and 200 micrograms.kg-1 daily) it potentiated, together with oestradiol, the lipid-mobilizing effect of isoprenaline. The results show that the non-steroid oestrogen diethylstilboestrol and the antioestrogen clomiphene may be included among the hormones capable of altering the response of adipose tissue to sympathomimetics (isoprenaline). We attribute the fact that clomiphene acted either as an antagonist or as an agonist of oestradiol to its combined oestrogenic and anti-oestrogenic effects.  相似文献

The rabbit fetal placenta plays an important physiological role in luteal maintenance in pregnancy, probably via the secretion of an unidentified placental "luteotropin." The objective of these studies was to examine conditioned medium from fetal placental-tissue incubations (FPI) for the presence of placental luteotropic hormones/factors, using the stimulation of progesterone accumulation by rabbit granulosa-lutein cells in culture, as an in vitro luteotropic bioassay. Progesterone accumulation by rabbit granulosa-lutein cells (during 5 days of culture) was increased (compared with controls), 1.5-fold by 10(-8) M estradiol-17 beta (E2) and 11.5-fold by 100 ng/ml luteinizing hormone (oLH). FPI stimulated progesterone accumulation (approximately 3-fold) and this was further increased in the presence of E2 (FPI + E2; approximately 6-fold). Luteotropic bioactivity in FPI (+ E2) was retained after dialysis (6000-8000 MW cutoff; 7.8-fold) and heating (90-95 degrees C for 1 h; 7.5-fold), but was destroyed after incubation with trypsin (1 mg/ml, 1 h at 37 degrees C; 0.9-fold). Media conditioned with skeletal muscle (1.2-fold), heart (1.6-fold), liver (1.5-fold), and uterus (0.5-fold) and 5-10% serum (less than 1-fold), from pseudopregnant rabbits, had little or no luteotropic bioactivity. These data indicate that FPI contains a luteotropic hormone/factor that is probably a heat-stable, trypsin-sensitive, protein/peptide of greater than 6000-8000 MW that acts in synergy with E2 to promote granulosa-lutein cell steroidogenesis. This placental hormone/factor is a good candidate for the elusive rabbit placental luteotropin.  相似文献

The effect of insulin on the activity of Na, K-ATPase was studied in rat brain microsomes. Addition of insulin to the incubation medium in a dose of 0.18 U/ml coupled with strophanthine did not change the enzyme activity. The raising of the hormone dose to 0.36 U/ml was accompanied by inhibition of the enzyme activity. The incubation duration (10 and 30 min) did not influence the Na-pump. Preincubation of brain microsomes with insulin for 5 min significantly activated Na, K-ATPase. It has been thus demonstrated that insulin is capable of influencing the activity of Na, K-ATPase of rat brain microsomes in vitro. The effect obtained depends both on the dose of the hormone introduced into the incubation medium and the experimental conditions.  相似文献

The ability of growth hormone (GH) to stimulate lipolysis was examined using chicken abdominal adipose tissue explants incubated in vitro and purified pituitary and bacterially derived chicken and bovine GH. Consistently in the fourth hour of incubation, lipolysis (as determined by glycerol release) was increased by the presence of GH (1 micrograms/ml), irrespective of pituitary or bacterial derivation or of chicken or bovine origins. This effect of GH was observed with adipose tissue originating from young (6-8 weeks old) intact and hypophysectomized chicks and adult (6-9 months old) male chickens. Glycerol release was also enhanced by lower doses of GH (10 ng/ml with tissue from young and 100 ng/ml with tissue from adult chickens).  相似文献

The influence of diabetes, hypophysectomy and adrenalectomy on glucose oxidation in rat aorta was studied. Diabetes was induced in normal, adrenalectomized and hypophysectomized-cortisone substituted rats by streptozotocin (65 mg/kg body weight). The oxidation of glucose to CO2 was determined during incubation of rat aorta in vitro for 2-3 hours. The aortic glucose oxidation was reduced after hypophysectomy but was unaffected by adrenalectomy. After streptozotocin treatment the rise in blood glucose concentration was similar in normal, adrenalectomized and hypophysectomized-cortisone substituted rats. In shamoperated diabetic rats the aortic glucose oxidation was reduced after a diabetes duration of 4 days. In adrenalectomized diabetic rats the aortic glucose oxidation was not significantly affected after 4 days but was reduced after a diabetes duration of 14 days. When adrenalectomized diabetic rats were treated with hydrocortisone the aortic glucose oxidation was reduced after diabetes for 4 days. After incubation of normal rat aorta in vitro for 6 hours with cortisol (1 microgram/ml) in the incubation medium a decrease in the aortic glucose oxidation was found. Incubation of aorta with only growth hormone had no effect. These results suggest that cortisol is of importance for the lowered glucose oxidation in diabetic rat aorta.  相似文献
