The objectives of this study were to evaluate the sensitivity of two bacterial tests commonly used in metal toxicity screening — the Vibrio fischeri bioluminescence inhibition test and the Pseudomonas putida growth inhibition test — in comparison to the standard acute Daphnia magna test, and to estimate applicability of the selected methods to the toxicity testing of environmental samples. The D. magna acute test proved to be more sensitive to cadmium (Cd), zinc (Zn) and manganese (Mn) than the two bacterial assays, whereas P. putida seems to be the most sensitive species to lead (Pb). Manganese appears to be slightly toxic to D. magna and non-toxic to the two selected bacteria. This leads to the conclusion that even in regions with high background concentrations, manganese would not act as a confounding factor. Low sensitivity of V. fischeri to heavy metals questions its applicability as the first screening method in assessing various environmental samples. Therefore, it is not advisable to replace D. magna with bacterial species for metal screening tests. P. putida, V. fischeri and/or other bacterial tests should rather be applied in a complex battery of ecotoxicological tests, as their tolerance to heavy metals can unravel other potentially present toxic substances and mixtures, undetectable by metal-sensitive species.  相似文献

Reactivators of phosphorylated acetylcholinesterase (oximes) are substances used as a human antidotal therapy for organophosphate poisoning. The objective of our study was to examine if juveniles of freshwater microcrustacean Daphnia magna could be employed as test animals in early screen toxicity tests of those substances as a first step for further experiments with daphnids intoxicated by organophosphates. For this purpose, seven different oximes were investigated. It was found that toxicity of all tested oximes increased with time. Mono-quaternary oximes were approximately ten fold (EC50, 14.9 mg.l? 1) more toxic in 24 hour tests and five fold (EC50 was 79.46 mg.l? 1) more toxic in 48 hour tests than bis-quaternary oximes. Tests with daphnids were shown to be easy to carry out at low cost and provided valuable results which could be used as a starting point for further research.  相似文献

This study investigates the effects of lifelong exposure to reversed geomagnetic and zero geomagnetic fields (the latter means absence of geomagnetic field) on the life history of Daphnia carinata King from Australia and Daphnia magna Straus from Europe. Considerable deviation in the geomagnetic field from the usual strength, leads to a decrease in daphnia size and life span. Reduced brood sizes and increased body length of neonates are observed in D. magna exposed to unusual magnetic background. The most apparent effects are induced by zero geomagnetic field in both species of Daphnia. A delay in the first reproduction in zero geomagnetic field is observed only in D. magna. No adaptive maternal effects to reversed geomagnetic field are found in a line of D. magna maintained in these magnetic conditions for eight generations. Integrally, the responses of D. magna to unusual geomagnetic conditions are more extensive than that in D. carinata. We suggest that the mechanism of the effects of geomagnetic field reversal on Daphnia may be related to differences in the pattern of distribution of the particles that have a magnetic moment, or to moving charged organic molecules owing to a change in combined outcome and orientation of the geomagnetic field and Earth's gravitational field. The possibility of modulation of self-oscillating processes with changes in geomagnetic field is also discussed.  相似文献

This paper deals with some methods for breeding Daphnia magna. Critical variables are the filtration of the medium on charcoal, the composition of the medium, the amount of food, the addition of vitamins with yeast extract and the extent of refreshing the medium. Daphnia magna is a commonly used test animal in aquatic toxicology.Test procedures for acute and chronic test are described, together with the influence of several variables on their results.The practicability of the methods was checked with four model compounds, viz. 1,1,2-trichloroethane, dieldrin, pentachlorophenol and 3,4-dichloroaniline. The toxicity of deltamethrin, a synthetic pyrethroid insecticide, was determined under standardized conditions (ISO, 1982) in neonates and juveniles of Daphnia magna. Neonates (6 to 24 h old) were more sensitive than juveniles (48 to 72 h old). The 24- and 48-h EC50s (immobilization) in neonates were 0.113 and 0.031 µg l–1, respectively. The toxicity of deltamethrin was highly toxic. The 96-h EC50 was in the ppt (µg l–1) range. Toxicity tests with Daphnia may be used to detect toxic residues in water and sediment in areas treated with deltamethrin and other highly toxic pyrethroid pesticides.  相似文献

Triclosan has been widely used as an antimicrobial agent. However, triclosan was found to cause toxicity, including muscle contraction disturbances, carcinogenesis, and endocrine disorders. In addition, it was found to affect central nervous system function adversely and even have ototoxic effects. Conventional methods for detecting such triclosan can be performed easily. However, the conventional detection methods are inadequate in precisely reflecting the impact of toxic substances on stressed organisms. Therefore, a test model for the toxic environment at the molecular level through the organism is needed. From that point of view, Daphnia magna is being used as a ubiquitous model. D. magna has the advantages of easy cultivation, a short lifespan and high reproductive capacity, and high sensitivity to chemicals. Therefore, the protein expression pattern of D. magna that appear in response to chemicals can be utilized as biomarkers for detecting specific chemicals. In this study, we characterized the proteomic response of D. magna following triclosan exposure via two-dimensional (2D) gel electrophoresis. As a result, we confirmed that triclosan exposure completely suppressed D. magna 2-domain hemoglobin protein and evaluated this protein as a biomarker for triclosan detection. We constructed the HeLa cells in which the GFP gene was controlled by D. magna 2-domain hemoglobin promoter, which under normal conditions, expressed GFP, but upon triclosan exposure, suppressed GFP expression. Consequently, we consider that the HeLa cells containing the pBABE-HBF3-GFP plasmid developed in this study can be used as novel biomarkers for triclosan detection.  相似文献

Cyanobacteria have become an important environmental concern due to their ability to produce a wide range of natural toxins. At present, very few studies describe concentration response curves for cyanobacteria other than Microcystis. However, field evidence highlights that both cyanobacterial concentration as well as cyanobacterial species composition vary considerably with season and year. Therefore, the aim of this study was to investigate the effects of different cyanobacteria at various concentrations of these cyanobacteria in the diet on the reproduction of Daphnia pulex and Daphnia magna. Those two species were chosen to assess whether the cyanobacteria-daphnid dynamics could be generalized for the Daphnia genus. Results demonstrated that both slope and EC50 of the concentration response curves depend upon the Daphnia species, the cyanobacteria species and the potential interaction between the two. This has two major consequences. First, the differences in sensitivity to cyanobacteria between D. magna and D. pulex depend upon concentration of the specific cyanobacteria. Second, we noted different mechanisms of toxicity for the two zooplankton species, a more general mechanism of toxicity for D. pulex and a more specific one for D. magna. Our data therefore suggest that results of studies investigating effects of cyanobacteria at different concentrations cannot be generalized across species. Furthermore, mechanisms of toxicity are not only cyanobacteria specific, but also dependent on the exposed species, even for rather closely related species such as in the Daphnia genus. Whenever possible, we therefore propose to combine a multi-species approach together with a full concentration response analysis to reach more general conclusions concerning the effects of cyanobacteria on zooplankton.  相似文献

Two chronic toxicity experiments were conducted with Daphnia magna. In a semi-static experiment with cohorts, the no effect level for bromide in respect of the intrinsic rate of natural increase (derived from age-specific survival and fecundity) was 10 mg l–1. A second test was started with small populations in an intermittent-flow system. These populations had a stable age distribution, were composed of cohorts of different ages, and showed an almost perfect logistic growth. Model calculations showed that bromide reduced the upper numerical limit (carrying capacity). It also increased the time-lag required to attain the maximum reproduction rate. For the first parameter, a no effect level of 14 mg l–1 was calculated. For the latter a threshold could not be detected. The EC50 and EC10 were 27 and 18 mg l–1, respectively. Additional experiments showed that individual growth of D. magna in time could also be described by a logistic equation. The age structure of the populations changed when food became limiting. This was parallelled by a reduction of the mean brood size. In conclusion it is stated that the results of the toxicity studies with populations support Halbach's view (1984), that population dynamics can be used like a magnifying glass to detect small sublethal ecotoxicological effects of environmental pollutants.  相似文献

A new rearing technique ideally suitable for promoting either the parthenogenetic or the gametogenetic (ephippia) type of reproduction ofDaphnia magna in the laboratory is described. Biomanipulation, through the reduction of fish abundance resulting in an increase of large filter feeders and a stronger top-down control on algae, is commonly used as a lake restoration tool in eutrophic lakes. However, cyanobacteria, often found in eutrophic ponds, can influence the grazing capacity of filter feeding zooplankton. We performed grazing experiments in hypertrophic Lake Blankaart during two consecutive summers (1998, with and 1999, without cyanobacteria) to elucidate the influence of cyanobacteria on the grazing pressure of zooplankton communities. We compared the grazing pressure of the natural macrozooplankton community (mainly small to medium-sized cladocerans and copepods) with that of large Daphnia magna on the natural bacterioplankton and phytoplankton prey communities. Our results showed that in the absence of cyanobacteria, Daphnia magna grazing pressure on bacteria was higher compared to the grazing pressure of the natural zooplankton community. However, Daphnia grazing rates on phytoplankton were not significantly different compared to the grazing rates of the natural zooplankton community. When cyanobacteria were abundant, grazing pressure of Daphnia magnaseemed to be inhibited, and the grazing pressure on bacteria and phytoplankton was similar to that of the natural macrozooplankton community. Our results suggest that biomanipulation may not always result in a more effective top-down control of the algal biomass.  相似文献

The role that temperature plays in excluding large daphnid species from subtropical and tropical ponds, and competitive relationships between Daphnia laevis and Daphnia magna, are explored. D. magna, a large temperate species is probably not excluded from subtropical or tropical systems by elevated temperatures. However, D. magna was a poorer competitor, under a restricted set of laboratory conditions, than D. laevis, the only pond dwelling daphnid in subtropical Florida. Competition is proposed as one mechanism that may limit the number of daphnid species in subtropical and tropical ponds and lakes. Reduced environmental fluctuations in subtropical and tropical systems (compared with temperate systems) may allow zooplankton populations to reach an environmental carrying capacity where competition limits the number of similar, coexisting species in a habitat.  相似文献

The relationship between respiration rate and dry weight forDaphnia magna was investigated in the absence of food. In addition the respiration and feeding rates of matureDaphnia magna females were measured simultaneously at differentChlorella concentrations. It was found that, besides the expected effect of food concentration on the feeding rate, there also was an effect of food concentration on the respiration rate. For food concentrations above a certain critical level a negative correlation between respiration rate and food concentration was found. Preferential feeding behaviour by Daphnia magna was shown when Daphnia were fed on a mixture of 14C-labelled algae (Chlorella vulgaris or Scenedesmus quadricauda) and 3H-labelled bacteria (Escherichia coli). Daphnia consumption of bacteria was equal or higher in the presence of algae. On the other hand, in the presence of bacteria, algal consumption decreased by 40–70% compared to algal consumption in the absence of bacteria. 14C radioactive uptake was in good agreement with the chlorophyll content and demonstrates the preferential feeding behaviour of Daphnia.  相似文献

The effects of sodium salt of 2.4-Dichlorophenoxi-acetic acid (2.4-D-Na) on the mortality and reproduction ofDaphnia magna have been tested.The mortality tests were performed in experiments with duration time of 24, 48 and 96 h at the temperatures of 15 and 20 °C. The toxic effect increased by 11–16 regarding 47% with increasing of temperature and exposure time.This herbicide (Dikonirt) affected adversely the reproduction of daphnids at concentration lower than tenth of LC50 value.  相似文献

Daphnia magna is used widely as a standard ecotoxicological indicator organism, and protocols exist for its use in assessing the toxicity of substances under acute and chronic experimental conditions. Problems exist in repeatability of such bioassays between laboratories. Sources of variation are identified using a simple quantitative genetics model. Presenting specific examples, we conclude that these problems are tractable, but only if the genotype and culture conditions prior to and during tests are strictly controlled.  相似文献

1. Acute and chronic toxicity tests with endosulfan were conducted on Daphnia magna. The 24-hr static lc50 was 0.62 mg/l with a coefficient of variation of 14.2%.2. The sublethal effects of 0.12, 0.15, 0.20, 0.25 and 0.31 mg/l endosulfan on the survival, reproduction and growth of D. magna were monitored for 21 days.3. The parameters used to determined the effect of the pesticide on reproduction were: mean total young per female, maximum number of broods, mean brood size, mean number of broods, mean time to first reproduction and intrinsic rate of natural increase (r).4. Growth, as measured by body length, was depressed significantly at all endosulfan concentrations tested. The highest concentrations used (0.20, 0.25 and 0.31 mg/l) caused a decrease in survival and mean total young per female, and an increase in the time to first reproduction. The intrinsic rate of natural increase (r) and the mean number of broods were decreased significantly at all the sublethal concentrations of endosulfan tested.  相似文献

Summary The usefulness of autoregression equations as empirical models of population change in an experimental population of Daphnia magna was studied. A two-term and a ten-term autoregression equation were fit to the data. Tests of the significance of the terms of the ten-term equation were carried out, and the fit of the second-order autoregression tested. The problem of autocorrelated error terms is discussed. The ability of the autoregressions to generate the observed changes in population density of the Daphnia magna population was studied by running Monte Carlo simulations using both the two-term and the ten-term models. Both models were fairly accurate up to a period of about 60 days. The second order autoregression model appeared to be more appropriate at high densities than the ten-term model, but the ten-term model was more effective at population densities less than 80 individuals.  相似文献

Food concentration supplied during subchronic and chronic toxicity tests, is one of the most important factors that might influence the response of test organisms to toxicants. The green microalga Scenedesmus incrassatulus was used as food for the cladoceran Daphnia magna, in a chronic toxicity test with the toxicant sodium dodecyl sulphate (SDS). Test concentrations were 0.625, 1.25, and 2.5 mg l-1 of SDS, equivalent to 1/40, 1/20, and 1/10 of the average 48-h LC50 previously determined. Food concentrations were 9.5, 19, and 38 mg l-1 (dry wt.). Survival and reproduction were recorded, and the data were analyzed using a life-table approach. After 55 days, the main findings were as follows:
1.  Average clutch size decreased as food concentration increased and there was a negative interaction between the toxicant and the algal concentration.
2.  The Net Reproductive Rate (R 0) of the controls fed the lowest food concentration was approximately 10 times higher than the controls fed the highest food concentration.
3.  R 0's for the treatments with SDS fed the lowest food concentration, were 2.5 to 5 times higher than those fed the highest food concentration.
4.  Reproduction in controls and treatments with the same SDS concentrations, became similar as food level increased.
According to these results, the food concentration to be supplied in chronic toxicity tests should be accurate, as an incorrect food concentration might negatively affect the survival and reproduction of test organisms, thus masking or confounding the effects of the toxicant being examined. A concentration around 10 mg l-1 (dry wt.) of S. incrassatulus seems to be appropriate for D. magna economic toxicity tests.  相似文献
