Populations of the Pacific blue shrimp, Penaeus stylirostris, reared at the University of Arizona's experimental shrimp culture facility on Oahu in Hawaii from late 1980 through 1981, were severely affected by a highly acute and lethal disease of viral etiology. Also found to be susceptible to the disease were P. vannamei and P. monodon. The disease was named infectious hypodermal and hematopoietic necrosis (IHHN) disease to describe the principal lesions observed. The histopathology of acute and subacute IHHN disease in these species was dominated by the presence of conspicuous eosinophilic intranuclear-inclusion bodies of the Cowdry type A variety in ectodermally (especially the cuticular hypodermis) and mesodermally (especially the hematopoietic tissues) derived tissues that were undergoing necrosis. Electron microscopy of affected tissues demonstrated the presence of two or three types of virus-like particles with cubic morphology and diameters of 17 to 27 nm that suggest IHHN virus to be either a parvo- or picornavirus. 相似文献

Adult penaeid shrimp were collected from mariculture raceways, holding tanks, and near-shore waters of Galveston Island, Texas. Those with darkly pigmented lesions were chosen for study. When observed by scanning electron microscopy, diseased cuticle shows dense populations of bacteria within a lesion, whereas normal cuticle shows a relatively low number of bacteria. Some lesions exhibited a morphologically homogeneous population of bacteria while others revealed a heterogeneous or mixed infection. Bacteriological analysis of a homogenate of each lesion revealed the presence of a wide variety of organisms which produced extracellular lipases, proteases, and chitinases. Lipolytic and proteolytic organisms were consistently isolated from all 20 lesions examined, while chitinoclastic bacteria were found in all but two lesions. According to the presumptive isolation scheme used, 10 distinct phenotypes of bacteria were differentiated among the isolates from 20 lesions. These phenotypes were classified into four genera: Vibrio, Alteromonas, Spirillum, and Flavobacterium. Groups of 10 shrimp were abraded and inoculated with representative isolates from each of the 10 phenotypes. Four isolates were able to initiate lesion formation. These were two Vibrio species, one Alteromonas species, and one Spirillum species. All of these four pathogens were lipolytic, while only two were chitinoclastic. This evidence suggests that there is not one specific chitinoclastic bacterium which causes brown spot symptoms, but that a variety of bacteria acting alone or in groups may be the causal agents. 相似文献

The acute and subacute toxicity of aflatoxin B1 to the marine shrimp Penaeus stylirostris and P. vannamei (Order: Decapoda, Class: Crustacea) was investigated. Experimental shrimp were exposed to a range of concentrations of the toxin directly by intramascular injection (from 2 to 160 μg aflatoxin B1/g body weight), or by multiple per os dosing with the feed (from 53 to 300 μg aflatoxin B1/g feed) for up to 25 days. The histopathogenesis of aflatoxicosis in the aflatoxin-exposed animals was followed and found to be time and dose dependent in the hepatopancreas, mandibular organ, and in the hematopoietic organs. Less significant and/or inconsistent lesions were also observed in other organs and tissues, but a time-dose dependency was not noted. The principal lesions of aflatoxicosis in penaeid shrimp occur in the hepatopancreas and the mandibular organ. In the former organ, subacute and acute aflatoxicosis is expressed as necrosis of the hepatopancreatic tubule epithelium that proceeds from the proximal (older) portion of the tubules, in the center of the organ, to the peripheral (younger) tubule tips. A marked intertubular hemocytic inflammation followed by encapsulation and fibrosis of affected tubules follows in subacute aflatoxicosis, but is not as developed as in acute aflatoxicosis. The mandibular organ in aflatoxicosis displays a necrosis of the peripheral epithelial cells of the cords within the gland that progresses proximally to the central vein. Only a slight hemocytic inflammation accompanies the degenerative changes in this latter organ. 相似文献

设计8对引物分片段扩增桃拉综合征病毒中国分离株ZHZC3全基因组,病毒两末端序列采用末端快速扩增方法(RACE)获取。扩增产物克隆到pMD18-T载体并测序,用DNAstar软件拼接全序列及同源性比较。结果显示ZHZC3全序列除去3′poly(A)尾,由10202个碱基组成,有两个开放阅读框,分别编码2107和1011个氨基酸的聚蛋白。与美国参考株HI94相比,在编码区没有核苷酸的缺失和插入,但在5′UTR缺失3个A,两者整体核酸同源性达97.9%。ORF1中ZHZC3与HI94及巴西株(BLZ01)的核酸同源性分别为97.6%、97.7%,在ORF2中ZHZC3与HI94、BLZ01的核酸同源性则分别为98.3、97%。与国外株ORF2的部分序列比较发现:ZHZC3和中国台湾株均与美国株HI94同源性最高。克隆分析6株TSV中国大陆株主要结构蛋白CP2基因,发现其编码的氨基酸存在三个高变区,中国大陆株更有其独特的氨基酸变异模式,312(S),449(A),451(Q)和468(H)。表明该病毒的整体变异性不高,但中国的流行株已形成其自己的遗传演变特征。在此基础上,利用生物学软件对CP2蛋白功能域和三维结构进行了预测,为进一步分析CP2蛋白结构与功能关系奠定了基础。ZHZC3株是第一个测定全序列的TSV中国株。 相似文献

Penaeid shrimp aquaculture is an important industry in the Americas, and the industry is based almost entirely on the culture of the Pacific White Shrimp, Litopenaeus vannamei. Western Hemisphere shrimp farmers in 14 countries in 2004 produced more than 200,000 metric tons of shrimp, generated more than $2 billion in revenue, and employed more than 500,000 people. Disease has had a major impact on shrimp aquaculture in the Americas since it became a significant commercial entity in the 1970s. Diseases due to viruses, rickettsial-like bacteria, true bacteria, protozoa, and fungi have emerged as major diseases of farmed shrimp in the region. Many of the bacterial, fungal and protozoan caused diseases are managed using improved culture practices, routine sanitation, and the use of chemotherapeutics. However, the virus diseases have been far more problematic to manage and they have been responsible for the most costly epizootics. Examples include the Taura syndrome pandemic that began in 1991-1992 when the disease emerged in Ecuador, and the subsequent White Spot Disease pandemic that followed its introduction to Central America from Asia in 1999. Because of their socioeconomic significance to shrimp farming, seven of the nine crustacean diseases listed by the World Animal Organization (OIE) are virus diseases of shrimp. Of the seven virus diseases of penaeid shrimp, five are native to the Americas or have become enzootic following their introduction. The shrimp virus diseases in the Americas are increasingly being managed by exclusion using a combination of biosecurity and the practice of culturing domesticated specific pathogen-free (SPF) stocks or specific pathogen-resistant (SPR) stocks. Despite the significant challenges posed by disease, the shrimp farming industry of the Americas has responded to the challenges posed by disease and it has developed methods to manage its diseases and mature into a sustainable industry. 相似文献

The temporal distribution patterns of the predominantly occurring postlarvae and juvenile shrimps in the mangrove and associated habitats of Muthupet, India were investigated for two years from February 1984 to January 1986. Among the eight commercially important species recorded, Penaeus indicus H. Milne Edwards, P. merguiensis De Man, P. monodon Fabricus and Metapenaeus dobsoni (Miers) were predominant. The postlarval recruitment size variedwith species: P. indicus and P. merguiensis recruited at the size of 9–11 mm total length (TL), P. monodon at 12–14 mm TL and M. dobsoni at 4–6 mm TL. The species P. indicus, P. merguiensis and M. dobsoni were observed continuously throughout the study period with maximum abundance occurring from July to September in 1984–85 and from August through October in 1985–86. P. monodon occurred seasonally from November to January in both years. Postlarvae and juvenile catches were low during low salinity and high salinity periods and a higher density was observed in the months of moderate water salinity. Large numbers of P. indicus, P. merguiensis and M. dobsoni clearly showed the preference to the detritus rich muddy substrate, whereas P. monodon did not show any preference and was equally abundant over different substrate types. 相似文献

The Infectious hypodermal and hematopoietic necrosis virus (IHHNV) and Taura syndrome virus (TSV) are two important shrimp viruses in cultured shrimp in America. These two viruses were transmitted to China at the beginning of the 21st century. In this study, 214 shrimp samples of Penaeus vannamei were collected from seven different areas of China and tested by PCR for IHHNV and TSV infection. The results showed that there were a high prevalence of IHHNV (65.42%) and low prevalence of TSV (3.27%) in the tested samples. Several samples were found to be co-infected with these two viruses. A 3 kb fragment of 7 positive IHHNV samples and a structure protein region (ORF2) of three TSV positive samples were amplified and sequenced. The sequence comparison indicated that both IHHNV and TSV sequenced in China have a low genetic variations compared with the prototype IHHNV and TSV from Hawaii. Phylogenetic analysis showed that TSV isolates were clustered into two groups, Asia and America group, which was genetically correlated to geographic distribution. Taura syndrome virus (TSV) is a highly virulent pathogen of Litopenaeus vannamei, has affected shrimp aquaculture throughout the world, and threatens wild populations. Despite its importance, little work has been done on the pathogen's formal epidemiology. Therefore we developed a compartment model for epidemics of TSV in closed populations of L. vannamei. The model includes five compartments, uninfected susceptible, prepatently infected, acutely infected, chronically infected, and dead infected shrimp. The transmission coefficients, patency coefficient, virulence coefficients, and removal coefficient (disappearance of dead infected shrimp) control the dynamics of the model. We estimated the coefficients in laboratory studies and inserted the estimates in the model to characterize TSV epidemics and to estimate the basic reproduction ratio R(0) and threshold density for TSV epidemics in L. vannamei. Further we examined through computer simulation the effect of varying the coefficients on R(0). Decreases in transmission decrease R(0), decreases in virulence increase R(0), increases in patency do not affect R(0), and increases in recovery most likely increase R(0) but under some conditions might decrease it. 相似文献

The greatest threat to the future of world shrimp aquaculture is disease, in particular the virulent untreatable viruses, infectious hypodermal and haematopoietic necrosis virus (IHHNV), taura syndrome virus (TSV), yellow head virus (YHV), and white spot syndrome virus (WSSV). To overcome these hazards, the industry of the future must be based on: (i) specific pathogen-free and genetically improved shrimp stocks; (ii) biosecure systems including enclosed, reduced water-exchange/increased water-reuse culture systems; (iii) biosecure management practices; and (iv) co-operative industry-wide disease control strategies. Specific pathogen-free shrimp are those that are known to be free of specified pathogens and such stocks will ensure that seed shrimp are not the conduit for introduction of pathogens and that if pathogens are encountered the stocks will not be severely affected. Commercially acceptable biosecure culture systems that are under cover and use recirculated sea water will need to be developed for shrimp production. Adherence to operating protocols that incorporate strict biosecurity practices, including restricted access and disinfection strategies, will need to become standard. Co-operative efforts will include: early warning surveillance; co-ordination of harvest and water exchange schedules of contaminated ponds; processor co-operation to ensure that processing wastes are not threats; quick response to outbreaks. 相似文献

Early in Australia's prawn aquacultural history it was very clear that all viruses in aquaculture had been preceded by their discovery in the wild prawns from the fishery. During the farming intensification process, it was also very clear that the housing of spawners from the fishery was providing evidence of potential problems. There is no evidence that there has been any introduction of viruses novel to Australia, but rather the viruses have been always present in the wild prawns awaiting a chance to be amplified by the increasing population density of aquaculture. The information on prawn viruses adds weight to the view that Australia's marine disease faunas are part of the greater Asian area. Oropouche virus is the aetiological agent of Oropouche fever, a zoonotic disease mainly transmitted by midges of the species Culicoides paraensis. Although the virus was discovered in 1955, more attention has been given recently to both the virus and the disease due to outbreaks of Oropouche fever in different areas of Brazil and Peru. Serological studies in human and wild mammals have also found Oropouche virus in Argentina, Bolivia, Colombia, and Ecuador. Several mammals act as reservoirs of the disease, although the sylvatic cycle of Oropouche virus remains to be assessed properly. Oropouche fever lacks key symptoms to be differentiated from other arboviral febrile illnesses from the Americas. Sporadic cases of aseptic meningitis have also been described with good prognosis. The cultivation of exotic Penaeus vannamei in Thailand began on a very limited scale in the late 1990s, but a Thai government ban on the cultivation of P. monodon in freshwater areas in 2000 led many Thai shrimp farmers to shift to cultivation of P. vannamei. Alarmed by the possibility of Taura syndrome virus (TSV) introduction, the Thai Department of Fisheries required that imported stocks of P. vannamei be certified free of TSV by RT-PCR (Reverse Trasciption Polymerase Chain Reaction) testing. During the interval of allowed importation, over 150,000 broodstock shrimp were imported, 67% of these from China and Taiwan. Despite the safeguards, TSV outbreaks occurred and we confirmed the first outbreak by RT-PCR in early 2003. This resulted in a governmental ban on all shrimp broodstock imports from February 2003, but TSV outbreaks have continued, possibly due to original introductions or to the continued illegal importation of stocks. To determine the origin of the TSV in Thailand, the viral coat protein gene VP1 was amplified by RT-PCR from several shrimp specimens found positive for TSV by RT-PCR from January to November 2003. These included 7 samples from P. vannamei disease outbreaks in Thailand, 3 other non-diseased shrimp samples from Thailand and Burma and 6 samples including P. vannamei and P. japonicus from China. Comparison revealed that the Thai, Burmese and Chinese TSV types formed a clade distinct from a clade of TSV types from the Americas. 相似文献

Aim: To study the accumulation and retention of recombinant proteins in Artemia gut for optimizing paratransgenic disease control in shrimp aquaculture. Methods and Results: Transgenic Escherichia coli expressing fluorescent marker proteins and the transgenic cyanobacterium Synechococcus bacillarus expressing a functional murine single chain antibody, DB3, were fed to Artemia franciscana. Stable expression and retention of several marker molecules (e.g. GFP, DS Red and DB3) up to 10 h after of feeding with E. coli were evident within the gut of Artemia. Engineered strains of S. bacillarus expressing DB3 accumulated within the gut of Artemia with detectable antibody activity for 8–10 h of feeding via ELISA, coincident with the time period of the highest density of transgenic S. bacillarus in the Artemia gut. Conclusions: Artemia fed transgenic bacteria or algae accumulated recombinant proteins for up to 10 h that retained biological activity. Co‐delivery of multiple recombinant proteins simultaneously in the gut of Artemia was also demonstrated. Significance and Impact of the Study: Expression of molecules that target infectious agents of mariculture in shrimp via commonly deployed feed organisms such as Artemia could potentially offer powerful new tools in the ongoing global effort to increase food supply. 相似文献
