Ultracentrifugation studies of diphtheria antitoxin showed that: 1. Purified antitoxin of high activity obtained from horse plasma without enzymatic treatment has exactly the same sedimentation constant as the globulin fraction obtained in a similar way from normal horse plasma s 20 water = 6.9 x 10–13. 2. Purified antitoxin obtained with trypsin digestion of the toxin-antitoxin complex has a sedimentation constant of s 20 water = 5.5 ± 0.1 x 10–13, a diffusion constant of D 20 water = 5.76 x 10–7, and a molecular weight of about 90,000. Electrophoresis experiments demonstrated that: 1. The trypsin-purified antitoxin has an isoelectric point not far from pH 7.0. 2. The reversible spreading noticed at about pH 7.3 cannot be attributed to heterogeneous preparation. 3. The large increase in the γ-globulin fraction occurring during immunization consists either of antitoxin of various degrees of activity or of some inert protein in addition to the antitoxin.  相似文献

A Gram-negative, aerobic, rod shaped, non-spore-forming bacterial strain, designated Dae08T, was isolated from sediment of the stream near Daechung dam in South Korea, and was characterized in order to determine its taxonomic position, using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain Dae08T belongs to the family Xanthomonadaceae of the Gammaproteobacteria, and is related to Lysobacter brunescens ATCC 29482T (97.3%). The phylogenetic distances from any other species with validly published names within the genus Lysobacter were greater than 3.7%. The G+C contents of the genomic DNA of strain Dae08T was 69.3 mol%. The detection of a quinone system with Q-8 as the predominant compound and a fatty acid profile with iso-C15:0, iso-C17:1, ω9c, iso-C17:0, iso-C16:0, and iso-C11:0 3-OH as the major acids supported the affiliation of strain Dae08T to the genus Lysobacter. DNA-DNA relatedness between strain Dae08T and its phylogenetically closest neighbour was 28%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Dae08T (= KCTC 12600T) should be classified in the genus Lysobacter as the novel species, for which the name Lysobacter daecheongensis sp. nov. is proposed.  相似文献

In consideration of their origin the adaptive strategies of the evergreen species of the Mediterranean maquis were analysed. Rosmarinus officinalis L., Erica arborea L., and Erica multiflora L. had the lowest net photosynthetic rate (PN) in the favourable period [7.8±0.6 mol(CO2) m–2s–1, mean value], the highest PN decrease (on an average 86 % of the maximum) but the highest recovery capacity (>70 % of the maximum) at the first rainfall in September. Cistus incanus L. and Arbutus unedo L. had the highest PN during the favourable period [15.5±5.2 mol(CO2) m–2s–1, mean value], 79 % decrease during drought, and a lower recovery capacity (on an average 54 %). Quercus ilex L., Phillyrea latifolia L., and Pistacia lentiscus L. had an intermediate PN in the favourable period [9.2±1.3 mol(CO2) m–2s–1, mean value], a lower reduction during drought (on an average 63 %), and a range from 62 % (Q. ilex and P. latifolia) to 39 % (P. lentiscus) of recovery capacity. The Mediterranean species had higher decrease in PN and stomatal conductance during drought and a higher recovery capacity than the pre-Mediterranean species. Among the pre-Mediterranean species, P. latifoliahad the best adaptation to long drought periods also by its higher leaf mass per area (LMA) which lowered leaf temperature thus decreasing transpiration rate during drought. Moreover, its leaf longevity determined a more stable leaf biomass during the year. Among the Mediteranean species, R. officinalis was the best adapted species to short drought periods by its ability to rapidly recover. Nevertheless, R. officinalis had the lowest tolerance to high temperatures by its PN dropping below half its maximum value when leaf temperature was over 33.6°C. Summary One well-defined competitive interaction amongst rhizobia is that between compatible and non-compatible strains of Rhizobium leguminosarum with respect to the nodulation of some primitive pea genotypes. The Middle Eastern pea cv Afghanistan is nodulated effectively can R. leguminosarum TOM, but its capacity to nodulate can be blocked if a mixed inoculation is made with R. leguminosarum PF2. This PF2 phenotype (Cnb) is encoded by its symbiotic plasmid and cosmid clones thereof. We found that Cnb is also encoded by the well-characterized Sym plasmid pRL1JI of R. leguminosarum strain 248. We have isolated and characterized a 6.9 kb HindIII fragment of pSymPF2 which confers the Cnb+ phentoype on other (Cnb) rhizobia. A Tn5 site-directed Cnb mutant was constructed by homogenotization and was also found to be Nod on the European pea cv Rondo. DNA hybridization and complementation analysis indicated that the 6.9 kb Cnb+ fragment contained the nodD, nodABC and nodFE operons. Analysis of the Cnb phenotype of nod::Tn5 alleles of pRL1JI showed that mutations of nodC, nodD or nodE all abolished Cnb activity whereas mutants in nodI and nodJ reduced activity to 50% of the wild-type level.  相似文献

Summary A method for determining the mass flow rate of xylem water in thin stems under natural field conditions is presented. Diurnal courses of xylem water flow and stomatal conductance of the vines Entadopsis polystachya, Cyclanthera multifoliolata, and Serjania brachycarpa were examined in a tropical deciduous forest on the west coast of Mexico. E. polystachya (leaf area 23.6 m2) had a maximum water flow rate of 6.50 kg h-1 or 1.44 kg cm-2 stem basal area h-1; daily water use was 2.00 kg m-2 leaf area day-1. S. brachycarpa (leaf area 4.5 m2) and C. multifoliolata (leaf area 3.6 m2) had a maximum water flow rate of 0.72 and 0.19 kg h-1 or 0.63 and 0.92 kg cm-2 stem basal area h-1. Daily water use was 1.26 and 0.39 kg m-2 leaf area day-1, respectively. The daily courses of xylem water flow were strongly influenced by the orientation of the leaf area to irradiance and its intensity. While leaves of E. polystachya had a constant high stomatal conductance during the day, S. brachycarpa had a maximum stomatal opening in the morning followed by continuous closure during the rest of the day. In contrast to the woody species, the herbaceous C. multifoliolata exhibited a strong midday depression of stomatal conductance and wilting of its leaves. The leaf biomass accounted for 8% (Entadopsis), 16% (Serjania), and 23% (Cyclanthera) of above-ground biomass. The relation of sapwood area to leaf area supplied (Huber value) was 0.19 (Entadopsis), 0.18 (Serjania), and 0.06 (Cyclanthera) cm2 m-2  相似文献

We have applied enzyme kinetic analysis to electrophysiological steady-state data of Zhou et al. (Zhou, J.J., Trueman, L.J., Boorer, K.J., Theodoulou, F.L., Forde, B.G., Miller, A.J. 2000. A high-affinity fungal nitrate carrier with two transport mechanisms. J. Biol. Chem. 275:39894–9) and to new current-voltage-time records from Xenopus oocytes with functionally expressed NrtA (crnA) 2H+-NO 3 symporter from Emericella (Aspergillus) nidulans. Zhou et al. stressed two Michaelis-Menten (MM) mechanisms to mediate the observed nitrate-induced currents, I NO 3 . We show that a single straightforward reaction cycle describes the data well, pointing out that during exposure to external substrate, S = (2H++NO 3 )o, the product concentration inside, [P] = [H+] i 2 · [NO 3 i, may rise substantially near the plasma membrane, violating the condition [P] [S] for MM kinetics. Here, [P] and its changes during experimentation are treated explicitly. K 1/2 20 µM for I NO 3 at pHo from Zhou et al. is confirmed. According to our analysis, NrtA operates between about 0.2 and 0.6 of the electrical distance in the membrane (outside 0, inside 1). In absence of thermodynamic gradients, the predominant orientation of the binding site(s) is probably inwards. The activity of the enzyme is sensitive to the transmembrane voltage, V, with an apparent gating charge of +1.0 ± 0.5 for inactivation, and transition probabilities of 0.3–1.3 s–1 at V = 0. This gating mode impedes loss of cellular NO 3 during depolarization.  相似文献

Type II toxin‐antitoxin (TA) modules, which are important cellular regulators in prokaryotes, usually encode two proteins, a toxin that inhibits cell growth and a nontoxic and labile inhibitor (antitoxin) that binds to and neutralizes the toxin. Here, we demonstrate that the res‐xre locus from Photorhabdus luminescens and other bacterial species function as bona fide TA modules in Escherichia coli. The 2.2 Å crystal structure of the intact Pseudomonas putida RES‐Xre TA complex reveals an unusual 2:4 stoichiometry in which a central RES toxin dimer binds two Xre antitoxin dimers. The antitoxin dimers each expose two helix‐turn‐helix DNA‐binding domains of the Cro repressor type, suggesting the TA complex is capable of binding the upstream promoter sequence on DNA. The toxin core domain shows structural similarity to ADP‐ribosylating enzymes such as diphtheria toxin but has an atypical NAD+‐binding pocket suggesting an alternative function. We show that activation of the toxin in vivo causes a depletion of intracellular NAD+ levels eventually leading to inhibition of cell growth in E. coli and inhibition of global macromolecular biosynthesis. Both structure and activity are unprecedented among bacterial TA systems, suggesting the functional scope of bacterial TA toxins is much wider than previously appreciated.  相似文献

Toxin‐antitoxin systems are mediators of diverse activities in bacterial physiology. For the ParE‐type toxins, their reported role of gyrase inhibition utilized during plasmid‐segregation killing indicates they are toxic. However, their location throughout chromosomes leads to questions about function, including potential non‐toxic outcomes. The current study has characterized a ParDE system from the opportunistic human pathogen Pseudomonas aeruginosa (Pa). We identified a protective function for this ParE toxin, PaParE, against effects of quinolone and other antibiotics. However, higher concentrations of PaParE are themselves toxic to cells, indicating the phenotypic outcome can vary based on its concentration. Our assays confirmed PaParE inhibition of gyrase‐mediated supercoiling of DNA with an IC50 value in the low micromolar range, a species‐specificity that resulted in more efficacious inhibition of Escherichia coli derived gyrase versus Pa gyrase, and overexpression in the absence of antitoxin yielded an expected filamentous morphology with multi‐foci nucleic acid material. Additional data revealed that the PaParE toxin is monomeric and interacts with dimeric PaParD antitoxin with a KD in the lower picomolar range, yielding a heterotetramer. This work provides novel insights into chromosome‐encoded ParE function, whereby its expression can impart partial protection to cultures from selected antibiotics.  相似文献

[目的]来自Paenibacillus polymyxa WLY78的固氮基因簇(nifBHDKEfNXhesAnifV)可以转化入Escherichia coli中表达并使重组大肠杆菌合成有固氮活性的固氮酶。本文拟通过对重组大肠杆菌E.coli 78-7的转录组分析以提高其固氮能力。[方法]对固氮条件(无氧无NH4+)和非固氮条件(空气和100 mmol/L NH4+)培养的重组大肠杆菌E.coli 78-7进行转录组分析。[结果]nif基因在两种培养条件下显著表达,说明在重组大肠杆菌中可规避原菌中氧气和NH4+nif基因的负调控。对于固氮过程必需的非nif基因,如参与钼、硫、铁元素转运的modcysfeoAB,这些基因在两种培养条件下表达水平有差异。而参与铁硫簇合成的sufisc基因簇在两条件下表达水平差异巨大。此外,参与氮代谢的基因在固氮条件下显著上调。[结论]重组大肠杆菌中与固氮相关的非nif基因在该菌的固氮过程中具有较大影响,本文对在异源宿主中调高固氮酶活性研究具有重要意义。  相似文献

Summary The presence of a Ca2+ channel in the plasmalemma of tonoplast-freeNitellopsis obtusa cells was demonstrated and its characteristics were studied using current- and voltage-clamp techniques. A long-lasting inward membrane current (I m ), recorded using a step voltage clamp, consisted of a single component without time-dependent inactivation. Increasing either [Ca2+] o or [Cl] o 1) enhanced the maximum amplitude of inwardI m ((I m ) p ) and 2) shifted the peak voltage ((V m ) p ) at(I m ) p to more positive values under ramp-shaped voltage clamping and 3) depolarized the peak value of action potentials. This behavior is consistent with predictions based on the Nernst equation for Ca2+ but not for Cl. DIDS (4,4-diisothiocyano-2,2-disulfonic acid stilbene) did not suppress(I m ) p in tonoplast-free cells, in contrast with its effect on normal cells. La3+ and nifedipine blocked(I m ) p irreversibly. On the other hand, Ca2+ channel agonist, BAY K 8644 irreversibly enhanced(I m ) p . Both Sr2+ influx and K+ efflux increased upon excitation. The charge carried by Sr2+ influx was compensated for by K+ efflux. It is concluded that only the Ca2+ channel is activated during plasmalemma excitation in tonoplast-free cells. In terms of the magnitude of(I m ) p , Sr2+ could replace Ca2+, but Mn2+, Mg2+ and Ba2+ could not. External pH affected(I m ) p and the membrane conductance (g m ) at(I m ) p ((g m ) p ). Increasing the external ionic strength caused increases in both(I m ) p and(g m ) p , and shifted(V m ) p to positive values. At the same time, Sr2+ influx increased. Thus Ca2+ channel activation seems to be enhanced by increasing external ionic strength. The possible involvement of surface potential is discussed.  相似文献

叶面积指数(LAI)是评价森林的生长、结构及众多服务功能的重要参数,受坡面上环境条件变化影响而有坡面变化与尺度效应,对此需要深入理解和精细刻画。在六盘山半湿润区香水河小流域选择了33年生华北落叶松人工林的一个斜坡长480.6m、水平长398.2m的东南坡向的典型坡面,在整个坡面上建立了宽30 m的调查样带,均匀分为空间连续的16个样地,在2014年生长季中期测定了林冠层LAI,并分析其坡面变化规律。结果表明:林冠层LAI存在明显的坡面差异,其坡面平均值为3.11,变化在2.66—3.49,变幅为0.83,变异系数为0.09;LAI总体上随着从坡顶向下的坡长增加呈波动性增大趋势,在坡面中部(水平坡长188.45 m时)达到最大,之后稍微减小。森林冠层LAI存在着坡面空间尺度效应,即冠层LAI的顺坡滑动平均值(Y_1)随水平坡长(X,m)增加而逐渐增大,平均每100 m升高0.12,其回归关系式为:Y_1=-2×10~(-8)X~3+8×10~(-6)X~2+5×10~(-4)X+2.6523,(R~2=0.99);各样地LAI与整个坡面平均值的比值(Y_2,小数)随水平坡长(X_1,m)增加呈现为波动性的非线性变化,其回归关系式为:Y_2=-9×10~(-9)X_1~3+2×10~(-6)X_1~2+1×10~(-3)X1+0.829,(R~2=0.78),可基于此式将特定坡位样地的实测LAI推算整个坡面的估计值。造成研究坡面上LAI坡位变化的主要原因是不同坡位(海拔)样地的气温与土壤含水量的差异。  相似文献

The orientation ofRhodobacter sphaeroides reaction center complexes (RC complexes) in proteoliposomal membranes was investigated by a direct electrometric method. Conditions were found that allow monitoring of only that RC complex fraction that is oriented with its donor side to the inner part of the proteoliposome. It is shown thato-phenanthroline, an inhibitor of electron transfer between primary (QA) and secondary (QB) quinone acceptors, can also inhibit the photoinduced QA reduction. The efficiency of this inhibition depends on the concentration of added ubiquinone. It is assumed that the laser flash-inducedo-phenanthroline inhibition of primary dipole (P-870+ · Q A ) formation is of a competitive nature.  相似文献

The genetic model for hybrid sterility that an allelic interaction at the S 5 locus induces the abortion of megaspores has been proposed as partial clarification of the wide compatibility in Asian rice cultivars; this model predicts the presence of an egg-killer. The present study was carried out in order to confirm that the proposed S 5 i allele acts as an egg-killer against its counterpart, the S 5 j allele, in the Indica-Japonica hybrid. A conspicuous feature of an egg-killer is the high rate of its transmission into the progeny through the egg. Backcrossing experiments were conducted using the Indica-Japonica hybrid in which the S 5 i and S 5 j alleles were assumed to be involved. Although an egg-killer was easily identified by these backcross experiments, it was not detected in the Indica-Japonica hybrid, which suggests that the proposed genetic mechanism for hybrid sterility in Asian rice should be viewed with caution.  相似文献

A chlorophyll b-less mutant of Chlamydomonas reinhardtii (Pg 27) was isolated after UV irradiation of the wild type cells. This photosynthetically competent mutant totally lacks chlorophyll b and the CP2 chlorophyll-protein complex. However, SDS-PAGE, proteolytic digestions and immunodetections demonstrated that the 24–25 Kd apoproteins of the lacking CP2 complex are still present in thylakoids of the Pg27 mutant. It is concluded that this CP2-less mutant is affected in the biosynthesis pathway of chlorophyll b.This CP2-less mutant was crossed with a CP1-less mutant (Fl5) Fluorescence emission spectra and fluorescence inductions in the presence of DCMU were analysed in the resulting (cp 2 , cp 1 + ), (cp 2 + , cp 1 ), (cp 2 + , cp 1 + ), cp 2 , cp 1 )tetratype. Differences in PS 2 optical cross section and in the relative amplitude or localisation of fluorescence emission peaks fit well with a quadripartite model where PS1 and PS2 would each correspond to a reaction centre core complex (CP1 and CP2 respectively) associated to a light harvesting antenna (LHC1 and LHC2 respectively). 为探讨氮沉降对九寨沟藓类植物的影响,该研究以当地优势藓类植物锦丝藓(Actinothuidium hookeri)和塔藓(Hylocomium splendens)为对象,以NH4NO3为氮源,设置对照(0 kg N·hm-2·a-1)、低浓度(20 kg N·hm-2·a-1)、高浓度(50 kg N·hm-2·a-1)3种处理,开展为期6个月的氮沉降模拟实验。结果表明:(1)氮沉降处理导致两种藓类植物的活性氧、丙二醛、叶绿素、脯氨酸和可溶性蛋白含量显著增加,同时锦丝藓过氧化氢酶、过氧化物酶、超氧化物歧化酶、抗坏血酸过氧化物酶活性增加。(2)对于生长旺期和生长末期的塔藓,氮沉降导致其过氧化物酶、过氧化氢酶、抗坏血酸过氧化物酶活性降低。(3)锦丝藓的综合隶属函数值随氮沉降浓度增大而增加,在生长旺期和生长末期,塔藓综合隶属函数值对氮沉降的响应存在差异。综上认为,两种藓类植物对氮沉降处理的生理响应存在差异,高浓度氮沉...  相似文献

Using X-ray mutagenesis and a chlorate-resistance selection method, a nitrate reductase (NR)-deficient mutant of Chlorella ellipsoidea nrm-4 was isolated, which is incapable of using NO3 as a nitrogen source. NR activity was not detected in nrm-4, suggesting this is a null mutation. Molecular analysis revealed that nrm-4 has a two base deletion at position 2348–49 of its DNA sequence, which produced a frame-shift mutation. The nrm-4 mutation is stable and nrm-4 algae could only use NH4+ and NO2 as nitrogen sources. Expression of a wild-type NR gene could complement this NR-deficient mutant. Furthermore, this transgenic strain was able to grow in NO3 medium, when the growth rate of the nrm-4 strain was equal to that of the wide type alga. The nrm-4 strain could potentially be used as a bioreactor that uses nitrate as a selectable marker instead of an antibiotic or herbicide.  相似文献

This paper reports the observation on the intersubgenomic heterosis for seed yield among hybrids between natural Brassica napus (AnAnCnCn) and a new type of B. napus with introgressions of genomic components of Brassica rapa (ArAr). This B. napus was selected from the progeny of B. napus × B. rapa and (B. napus × B. rapa) × B. rapa based on extensive phenotypic and cytological observation. Among the 129 studied partial intersubgenomic hybrids, which were obtained by randomly crossing 13 lines of the new type of B. napus in F3 or BC1F3 to 27 cultivars of B. napus from different regions as tester lines, about 90% of combinations exceeded the yield of their respective tester lines, whereas about 75% and 25% of combinations surpassed two elite Chinese cultivars, respectively. This strong heterosis was further confirmed by reevaluating 2 out of the 129 combinations in a successive year and by surveying hybrids between 20 lines of the new type of B. napus in BC1F5 and its parental B. napus in two locations. Some DNA segments from B. rapa were identified with significant effects on seed yield and yield components of the new type of B. napus in BC1F5 and intersubgenomic hybrids in positive or negative direction. It seems that the genomic components introgressed from B. rapa contributed to improvement of seed yield of rapeseed.  相似文献

Isolated hepatocytes from starved rats were loaded with N-[14C]acetylglutamate by preincubating them with [14C]bicarbonate, oleate, NH3, ornithine and lactate. Turnover of N-acetylglutamate in these cells was subsequently measured in an unlabelled medium under conditions of minimal flux (oleate alone present) and maximal flux (oleate, NH3, ornithine and lactate present) through the urea cycle. 1. Direct measurement of the distribution of N-[14C]acetylglutamate across the mitochondrial membrane in the hepatocytes showed that, under the conditions studied, the rate of degradation of total intracellular N-[14C]acetylglutamate was about equal to the rate of efflux of N-acetylglutamate from the mitochondria. 2. In the presence of oleate alone, intramitochondrial N-acetylglutamate decreased because mitochondrial N-acetylglutamate efflux predominated over the synthesis of N-acetylglutamate in the mitochondria. 3. In the presence of oleate, NH3, ornithine and lactate both the rate of synthesis of N-acetylglutamate and the rate of its transport out of the mitochondria were increased when compared with the condition with oleate alone. However, the intramitochondrial concentration of N-acetylglutamate increased because initially the rate of its synthesis exceeded that of its efflux from the mitochondria. Finally, a steady state was reached in which both rates were equal. 4. The data indicate that in hepatocytes from starved rats N-acetylglutamate transport out of the mitochondria takes place at a rate proportional to its intramitochondrial concentration. It is concluded that transport of N-acetylglutamate either occurs by diffusion or is mediated by a transport system with a high Km for intramitochondrial N-acetylglutamate.  相似文献
