1. A pigment can be extracted from Blepharisma undulans by heat treatment of a concentrated suspension of the deeply pigmented animals. 2. In the presence of this pigment, various colorless protozoans are sensitized to light and killed if exposed long enough. The protozoans show a differential sensitivity, some being much more sensitive than others. 3. Bleached colorless blepharismas are not sensitized to their own pigment even after prolonged exposure in the most concentrated solutions available. 4. Blepharisma is also less sensitive than any of the protozoans tested to such photodynamic dyes as rose bengal. 5. The pigment is not extracted from wet blepharismas by non-polar solvents, but is readily extracted into such polar organic solvents as the alcohols. 6. When the alcohol extract is dried, the amorphous residue is readily soluble in a variety of organic solvents, but not in the most non-polar. 7. The pigment is highly stable in alcohol extract and has been kept in the dark for years. 8. The pigment is bleached by light in a photooxidation. 9. Absorption maxima are found at wave lengths 5800, 5400, 4800, and 3300 A, the latter being the largest. Similar peaks are found in alcohol and water solutions, although the heights are not exactly the same. In both alcoholic and aqueous solutions pH had an effect on the absorption spectrum. Heat has little effect but illumination with intense visible light or exposure to ultraviolet light bleaches the pigment with decreases in the characteristic peaks. 10. Preliminary absorption column experiments indicate a single pigment in the alcohol extract. 11. Experiments on the migration of zoopurpurin in agar and gelatin gels indicate that it diffuses at about the same rate as eosin and is therefore probably not a large molecule.  相似文献

In the vertebrate retina the presence of synaptic ribbons (SRs) is well documented in two sites only, viz., in photoreceptor axon terminals in the outer plexiform layer and in bipolar cell axons in the inner plexiform layer. The present paper reports the presence of non-photoreceptor SRs in the outer plexiform layer of cattle and mouse, where they were seen in small numbers in thin cell processes near cone pedicles of light-adapted animals. They were never seen near rod spherules. Quantitative data obtained in mice killed at different time-points revealed that the SRs under consideration increased in number during day time and were absent during the dark phase. Moreover, under high light intensity of 10000 lux they were more frequent in number compared to 100-lux-exposed animals. It is concluded that the cell processes revealing the temporary presence of SRs are processes of flat bipolar cells which may provide a feedback to cones during the light phase.  相似文献

Measurements were made of the growth and pigment content of developing flower buds of Salpiglossis sinuata. From the time the buds were approximately 10 mm long they grew in length exponentially until they reached their final length. The logarithm of bud length increased linearly with time and served as a convenient morphological index on which to relate the progress of anthocyanin synthesis. Buds shorter than about 42 mm had no anthocyanin, but when buds reached this length, anthocyanin production was initiated and proceeded rapidly. The maximum relative pigment concentration (pigment/mg fresh weight) was attained by the buds about 17 hr after the initiation of pigment synthesis. In the mahogany-colored variety used in these studies, two anthocyanidins were found and identified as cyanidin and delphinidin. Buds excised from the plants could be cultured in vitro. Buds started in culture at a length of 30–35 mm when they contained no anthocyanins developed pigment during their growth. The amount of pigment formed increased with increasing light intensity, while only small amounts of pigment could be formed in buds cultured in darkness. The anthocyanidins of these cultured buds were the same as those of the intact flowers, but the ratio of delphinidin to cyanidin decreased with decreasing light intensity. Brief daily irradiation of dark-grown buds with red, far-red or blue light did not increase pigment synthesis nor change the anthocyanidin ratio. If buds were placed in culture at 20–25 mm and grown in darkness, they developed a third anthocyanidin, identified as malvidin, which was not present in intact flowers, light-grown buds or 30–35-mm buds cultured in darkness.  相似文献

Summary Under anaerobic conditions Fusarium aquaeductuum is able to synthesize carotenogenic enzymes but does not produce pigments. If illumination of the mycelia in the presence of oxygen is followed by an incubation in the dark under N2 atmosphere, the strictly concurrent formation of the different carotenoids sets off as soon as aerobic conditions are restored. The paraboloidal increase of pigment production possibly indicates that synthesis of carotenogenic enzymes is also resumed. Blocking this enzyme synthesis by addition of cycloheximide leads to a simultaneous and linear increase of each carotenoid portion as soon as oxygen is replenished. This is interpreted to mean that light induces carotenogenic enzymes in a coupled group. On the other hand, our present and earlier results do not support any hypothesis on the existence of a carotenogenic multienzyme complex. The composition of the pigment after carotenoid production has ceased provides evidence for a selective inhibition of the synthesis of individual carotenogenic enzymes. Changes in pigment composition caused by an incubation of the mycelia for 12 h under anaerobic conditions are also reported.  相似文献

The non-heterocystous filamentous cyanobacterium Plectonema boryanum UTEX 594 grew rapidly microaerobically under nitrogen-starvation conditions in continuous high light intensity by conducting oxygenic photosynthesis and oxygen sensitive nitrogen-fixation in alternating cycles. During diazotrophic phase, the light harvesting pigment phycocyanin declined with a concomitant depression in light dependent oxygen evolution by the cyanobacterium. A substantial component of light dependent carbon dioxide fixation during diazotrophic phase was not inhibited by DCMU in spite of complete cessation of photosynthetic oxygen evolution. Endogenous-reductant dependent electron transfer to photosystem I during diazotrophic phase is postulated even during photoautotrophic growth.  相似文献

Locomotor activity in the freshwater crab, Potamon fluviatile, was studied both in the field, by direct counts of animals along a stream section, and by means of actographs. General metabolism was evaluated through the analysis of oxygen consumption. Sex, size, and environmental factors proved to influence both the intensity and the timing of locomotion. In the laboratory, the two sexes behaved in the same way and consumed oxygen to the same extent, while the higher females' activity recorded in the field was due to their reproductive state. Younger age classes were more cryptic in the field and also less active in the laboratory, even though they had a higher weight-relative oxygen consumption. A rise in temperature, at least under the range examined in the present study, increased the crabs' activity rate, and also caused a change in their rhythmicity pattern, with a passage from diurnalism to nocturnalism. A lowering in relative humidity increased locomotion intensity, as a hygrokinetic response.  相似文献

When the larvae of a saturniid silkmoth, Antheraea yamamai, are maintained under high intensity light (5000 lux), they produce green cocoons whereas the cocoons produced under light of low intensity (e.g., 50 lux) or in darkness are yellow. The green colour of the cocoon is due to the presence of a blue bilin pigment in combination with yellow pigment, and light stimulates the accumulation of blue bilin. In the present study, we show that two blue bilins, with similar characteristics to the sarpedobilin in the green cocoon, can be induced in larval haemolymph both in vivo and in vitro. In both conditions, the amount of these bilins increased with increasing intensity or duration of light exposure. Induction also occurred at 0 degrees C. In contrast, the chromophore of the constitutive biliprotein of the haemolymph did not change depending on light conditions. Size fractionation of the haemolymph indicates that the precursor of the blue bilins induced by light is bound to a protein with a molecular mass of 5000 Da or more. Thus, in these insects, the blue bilin responsible for green colouration is facultative under photochemical stimulation.  相似文献

Irradiation with visible light of human serum albumin in aqueous solution at pH 8, in the presence of catalytic amounts of rose bengal or methylene blue, resulted in random oxidation of the histidine residues in the protein under consumption of one mole O2, and release of somewhat less than one proton, per histidine residue degraded. An increase of light absorption at 250 nm was proportional to the amount of oxygen consumed. Bilirubin bound to the oxidized protein showed an increased light absorption at its maximum, 460 nm, and a decreased binding affinity, indicating a conformational change of the protein on oxidation of histidine residues. This change also resulted in a slight perturbation of tyrosine light absorption, corresponding to a shift of the chromophore to more polar surroundings. Further, a sensitized oligomerization of albumin was observed, independent of oxidation of the histidine residues, and not consuming oxygen. Irradiation of a complex of human serum albumin with one molecule of bound bilirubin, in the absence of a sensitizing dye, resulted in a fast, non-oxygen consuming process whereby the light absorption maximum of the pigment was shifted 4 nm towards longer wavelength and part of the bilirubin was converted to a more polar pigment, bound less firmly to the protein. This was followed by a relatively slow oxidation of the pigment under uptake of one mole O2. Parallel photooxidation of the protein carrier could not be detected. It is considered possible that the fast, anaerobic process is operative in phototherapy of hyperbilirubinemia in the newborn. Serum albumin is probably not oxidized during this treatment.  相似文献

In the transient receptor potential (trp) mutant of Drosophila, the receptor potential appears almost normal in response to a flash but quickly decays to baseline during prolonged illumination. Photometric and early receptor potential measurements of the pigment suggest that the pigment is normal and that the decay of the trp response during illumination does not arise from a reduction in the available photopigment molecules. However, there is reduction in pigment concentration with age. Light adaptation cannot account for the decay of the trp response during illumination: in normal Drosophila a dim background light shortens the latency and rise time of the response and also shifts the intensity response function (V-log I curve) to higher levels of light intensity with relatively little reduction in the maximal amplitude (Vmax) of response. In the trp mutant, a dim background light or short, strong adapting light paradoxically lengthens the latency and rise time of the response and substantially reduces Vmax without a pronounced shift of the V-log I curve along the I axis. The effect of adapting light on the latency and V-log I curve seen in trp are associated with a reduction in effective stimulus intensity (reduction in excitation efficiency) rather than with light adaptation. Removing extracellular Ca+2 reduces light adaptation in normal Drosophila, as evidenced by the appearance of "square" responses to strong illumination. In the trp mutant, removing extracellular Ca+2 does not prevent the decay of the response during illumination.  相似文献

A mathematical formulation of the relationship between optical absorption coefficient of photosynthetic pigment molecules and light intensity was developed. It showed that physical parameters of photosynthetic pigment molecule (i.e., light absorption cross-section of photosynthetic pigment molecule, its average lifetime in the excited state, total photosynthetic pigment molecules, the statistical weight, or degeneracy of energy level of photosynthetic pigment molecules in the ground state and in the excited state) influenced on both the light absorption coefficient and effective light absorption cross-section of photosynthetic pigment molecules. Moreover, it also showed that both the light absorption coefficient and effective light absorption cross-section of photosynthetic pigment molecules were not constant, they decreased nonlinearly with light intensity increasing. The occupation numbers of photosynthetic pigment molecules in the excited states increased nonlinearly with light intensity increasing.  相似文献

Levels of chloroplast antioxidants and enzymes that scavenge oxygen racidals were followed in the leaves of pea plants (Pisum sativum L. cv. Meteor) grown under glasshouse conditions between April 1984 and May 1985. While little variation in pigment levels or superoxide dismutase activity was detected during this period, plants grown in early summer (May–June) contained appreciably higher levels of ascorbate, ascorbate peroxidase and glutathione reductase than plants grown in winter (Dec–Jan.). The role of light intensity in regulating levels of chloroplast antioxidants was examined further using pea plants grown in a constant environment chamber under 100 or 400 μmol m−2 s −1 photon flux density. Chloroplasts isolated from plants grown at the higher light intensity contained significantly higher levels of ascorbate, ascorbate peroxidase, glutathione reductase and dehydroascorbate reductase. These data suggest that light intensity may have an important influence on the level and activity of chloroplast antioxidants and oxygen radical scavenger enzymes.  相似文献

The mechanism of action of the p-nitrodiphenyl ether herbicides has remained ambiguous because of conflicting reports in the literature. The diphenyl ether herbicide oxyfluorfen causes a light induced consumption of oxygen which resembles the electron acceptor reaction of paraquat. However, this reaction is not linked to the transport of electrons through photosystem I. This conclusion is based on the observation that the rate of oxygen consumption, in the presence of oxyfluorfen, does not demonstrate a first order rate dependence on light intensity. Using the bleaching of N,N-dimethyl p-nitrosoaniline as a specific detector of singlet oxygen, we demonstrate that oxyfluorfen is a potent generator of this toxic radical. The production of singlet oxygen occurs in the presence of inhibitors of photosynthetic electron transport (oxyfluorfen at 10−4 molar and paraquat) and also under temperature conditions (3°C) which prevent electron transport. This light induced reaction results in oxygen consumption and is the primary cause of lethality for oxyfluorfen. The production of singlet oxygen occurs rapidly and at low herbicide concentrations (10−9 molar). The reaction occurs without photosynthetic electron transport but does require an intact thylakoid membrane.  相似文献

Leaf beetles of the genus Macroplea live permanently under water. Species‐specific preferences for either freshwater or brackish water are available in the literature. To detect potential physiological differences, the oxygen consumption of Macroplea mutica and Macroplea appendiculata from habitats with differing salinities is measured at two different salinities (0 and 10). The specific oxygen consumption does not depend on oxygen saturation of the water (values in the approximate range of 25–100% occur during the experiments). There is no difference between species or sampling locations. Similarly, the salinity during the measurements does not affect the oxygen consumption of the beetles, either when compared as salinity per se (0 versus 10), or when classified as home salinity or atypical salinity. Comparisons with other chrysomelid beetles and aquatic insects (using available published data) reveal that the two Macroplea species have relatively low metabolic rates. This finding is discussed in the light of activity patterns and morphology, especially the reduction of flight muscles, which comprise a highly metabolically active tissue.  相似文献

以适宜不同栽培条件的3个甜瓜品种为材料,研究了遮荫对其光合色素含量、净光合速率及比叶重等叶片特性的影响。结果表明:与正常光照相比,遮光处理能诱使甜瓜叶片叶绿素和类胡萝卜素含量显著提高,叶绿素a/b显著降低。全光照条件下,3个甜瓜品种的净光合速率日变化曲线均呈双峰型,有明显的"光合午休"现象,而在遮荫条件下则呈单峰曲线变化,且光合峰值出现的时间比全光照下推迟;遮光条件下甜瓜叶片气孔导度日变化曲线与其净光合速率日变化类似。3个品种间比叶重在全光照条件下差异不显著,但遮荫显著降低了壮龄叶片的比叶重,且遮荫强度越重,比叶重越小;品种‘黄河蜜3号’壮龄叶的比叶重降幅(31.83%)显著大于‘银帝’(27.22%)和‘玉金香’(26.01%)。可见,遮荫降低了甜瓜叶片的净光合速率和功能叶片的比叶重,植株通过增加自身叶片光合色素含量以增强对环境的适应性,缓解遮荫对其的影响,品种‘银帝’表现出较强的耐弱光性。  相似文献

N-Acetyltransferase (NAT) activity was determined in the pineal gland of frogs (Rana tigrina) of different ages using 2-aminofluorene and p-aminobenzoic acid as substrates, and assayed by high-pressure liquid chromatography. Frogs of different ages were either killed during the light phase or exposed to darkness or light for 1 min during the dark phase of the lighting cycle, then returned to their cages in darkness for 30 min before being killed. The pineal gland NAT activity of 1-month-old frogs was inhibited when the animal was nocturnally exposed to 1 min of light. Nocturnal light exposure did not inhibit NAT activity in 1-month-old frogs, even though these animal displayed clear light-dark differences in pineal gland NAT activity. Nocturnal light exposure did not inhibit night-time levels of NAT activity in 1-month-old animals which had been bilaterally enucleated, thus suggesting that this effect is retinally mediated. Pretreatment of 1-month-old and 6-month-old animals with isoproterenol (a beta-adrenoceptor agonist drug) prevented the nocturnal light-induced inhibition of NAT activity. From the different sensitivity of 1-month-old and 6-month-old animals to different intensities or durations of nocturnal light exposure it was found that the duration or intensity of light exposure was not able to inhibit nocturnal NAT activity. The NAT activity was at least 4–5-fold greater in 1-month-old frogs than in 6-month-old frogs. This is the first demonstration of the retino-pineal gland pathway that appears to produce light-induced changes in pineal glands of frogs 1-month-old or older, but this pathway only functions in 1-month-old frogs, and does not appear to function in 6-month-old frogs.  相似文献

Seven healthy male subjects underwent a treadmill incremental work test in control conditions and during an intravenous epinephrine infusion (10 micrograms/min). At all exercise intensities, epinephrine increased heart rate, ventilation, respiratory quotient and plasma lactate levels without significant changes in oxygen consumption. Under epinephrine infusion, the "anaerobic threshold", considered as the critical intensity at which ventilation began to increase non linearly with oxygen consumption, appeared at a lower intensity and for a higher plasma lactate level than in control conditions. We conclude that the hyperventilation threshold does not necessarily reflect a muscular hypoxia. It could be due to an effect of catecholamines on peripheral chemoreceptors, maybe by alpha-adrenergic vasoconstriction in the carotid bodies.  相似文献

采用野外研究与室内研究相结合的方法研究了月光周期和光照对夜行性动物艾虎活动的影响。野外无线电遥测结果表明,艾虎的日活动时间不受月光周期的影响,雌雄艾虎在不同的季节及繁殖期和非繁殖期都有相对稳定的活动时间,其开始活动和结束活动的时间与月光周期和日出、日落都没有明显的相关性。室内模拟研究结果也表明,由于环境条件的变化,艾虎增加了白天的活动时间,但仍然以夜间活动为主,光照强弱对艾虎的活动格局和活动时间没有明显的影响。以上结果表明昼夜变换使艾虎形成了相对稳定的生物种节律,艾虎在取食过程中可能利用洞道等隐蔽场所降低明亮月光期的高捕食风险。  相似文献

Rat sperm are mechanically immobilized by the mucin immobilin during storage in the cauda epididymidis. We thought this mechanical immobilization might lower the respiration rate of caudal sperm since it is well established that both the respiration rate and adenosine triphosphate (ATP) dephosphorylation rate of certain invertebrate sperm are markedly reduced when they are mechanically immobilized. To measure the oxygen consumption rates of rat sperm, we used a conventional polarographic oxygen sensor. However, we used an unstirred sample chamber since we found that sperm are unusually fragile and are quickly killed by even moderate stirring. When caudal sperm were diluted in physiological salt solutions, they became vigorously motile and consumed oxygen at a rate of approximately 20 microliter O2/10(8) sperm/hour at 34.5 degrees C for dilutions between 20- and 200-fold. In confirmation of the work of Brokaw et al., we found that when the motility of sea urchin sperm was reduced progressively with increasing amounts of methylcellulose, the sperm consumed oxygen at a markedly reduced rate. In contrast, we found that when caudal rat sperm were mechanically immobilized, either by methylcellulose or by immobilin, their oxygen consumption rate did not differ detectably from that of vigorously motile sperm. Thus, the coupling of metabolism to motility differs significantly between these two types of sperm. We conclude that immobilin does not change the respiration rate of caudal rat sperm.  相似文献
