Prolactin receptors were monitored by measuring 125I-labeled prolactin binding to collagenase-dissociated mammary epithelial cells of lactating BALB/c mice. Specific receptors for iodine-labeled prolactin with an apparent dissociation constant (Kd) of 0.99 · 10?9 M were present on the dissociated mammary cells. The binding was inhibited by ovine prolactin, human growth hormone and human placental lactogen but not by follicle stimulating hormone, luteinizing hormone, thyroid stimulating hormone, bovine growth hormone or insulin. Adrenal ablation of nursing mothers caused a reduction of the number of prolactin receptors and this effect was preventable by hydrocortisone therapy. Hydocortisone injections to mothers 3 days after adrenalectomy also induced a replenishment of the prolactin receptors on the mammary cells. Injections of progesterone failed to sustain the high level of mammary cell prolactin receptors in adrenalectomized animals. Stimultaneous injections of hydrocortisone and progesterone to animals 3 days after adrenalectomy caused a partial suppression of the stimulatory action of hydrocortisone alone. The results suggest that hydrocortisone can exert a modulatory influence on mammary cell prolactin receptors in non-hypophysectomized post-partum mice without altering the dissociation constant (Kd) of the receptors.  相似文献

Statistically significant (P < 0.05) thyroid hyperactivity (131I% uptakes) occurs on certain days in Plasmodium berghei infected C3H mice. Male mice thyroid glands are made more hyperactive by the malaria than the female glands. Hyperactive thyroid glands may be at least one cause of hypocholesterolemia in plasmodium-infected rodents. Thyroid hyperplasia was found only in experimental mice (620 males; 820 females). A hyperactive thyroid gland appears to be an unreported aspect of experimental acute P. berghei infections in mice. The hyperthyroidism may be due to possible toxic substances acting either directly on the thyroid gland, or indirectly on the hypothalamus affecting TSH production.  相似文献

The properties and activity of brown adipose tissue have been investigated in suckling, pre-obese, obob mice in order to determine whether decreased thermogenesis in the tissue precedes the development of obesity in this mutant. At 14 days of age there was no difference between the obob and normal animals in the total amount of interscapular brown adipose tissue, and the DNA content, protein content, and cytochrome oxidase activity of the tissue were similar in the two groups of mice. Respiration rates of brown adipose tissue mitochondria in the presence of albumin were, however, greater in the normal than the obob animals, although after the addition of GDP to recouple the mitochondria there was no difference between the two groups. The mitochondrial membrane potential, measured with [3H]methyltriphenylphosphonium, was less affected by exogenous GDP in obob mice than in normal animals. GDP binding to brown adipose tissue mitochondria, an index of the proton conductance pathway, was much greater in normal than in obob mice at both 10 and 14 days of age; the decreased GDP binding in the mutant animals was found to result from a reduction in the number of binding sites. It is concluded that brown adipose tissue mitochondria of pre-obese obob mice are more tightly coupled than those of normal siblings, and that the activity of the ‘thermogenic’ proton conductance pathway is lower in the mutant animals. A decrease in thermogenesis in brown adipose tissue is therefore an early event in the development of the obob mouse and precedes the appearance of obesity.  相似文献

Relatively large amounts of immunoreactive prolactin were measured in homogenates of human decidual tissue obtained immediately after delivery of normal term pregnancies. In order to study the release and possible synthesis of prolactin by this tissue, explants of decidua were incubated for 24 hours at 37°C in oxygenated Gey's buffer containing 20% fetal calf serum. When cycloheximide was added to the medium in concentrations sufficient to prevent invitro protein synthesis, 85–90% of the prolactin present in the tissue was released into the medium during the first 3 hours of incubation. No additional prolactin accumulated in either the medium or the tissue during the remainder of the incubation period. In the absence of cycloheximide, the prolactin concentration in the medium increased progressively during incubation, so that after 24 hours the total amount of hormone present in the tissue and medium was significantly greater than that in the tissue and medium prior to incubation (37.6 ± 9.6 ng/ml at 0 time vs 82.2 ± 7.7 ng/ml at 24 hours). When 3H-1-leucine (100 u Ci) was supplied during incubation, radioactive proteins were detected in the medium at 24 hr, 14–20% of which were specifically precipitated by antiserum to human pituitary prolactin. When aliquots of this medium were chromatographed on Sephadex G-100, 80–95% of the 3H-proteins precipitated by antiserum to pituitary prolactin eluted in the same position as did purified, iodinated pituitary prolactin. These data indicate that a species of prolactin which is identical to pituitary prolactin by the criteria of immunoprecipitation and gel chromatography is synthesized by human decidual tissue invitro.  相似文献

Lactogenic hormones from the placenta and pituitary are primarily responsible for the growth and function of the mammary gland during pregnancy and lactation. In the present study we describe the optimal conditions for the measurement of 125I-labeled ovine prolactin binding to mammary gland slices of pregnant and lactating rats. Prolactin binding is saturable (Kd approx. 2.36 · 10?9 M), hormone specific and destroyed by proteases. The hormonal environments of pregnancy and lacation dramatically influence the availability and measurement of prolactin binding sites. Whereas binding consistently appears to be low in mammary glands removed from rats during pregnancy, binding levels rise 7–8-fold shortly after birth and remain high during the 22 days of lactation. However, the removal of the ovaries and gravid uteri at specific times during pregnancy results in prompt 3–6-fold increase in prolactin binding. Elevated levels in potential prolactin binding capacity appear in mammary tissue coincident with the reported rise in serum rat placental lactogen between the eight and eleventh days. We suggest that high levels of this lactogenic hormone promote the appearance of prolactin binding sites during pregnancy and mask the sites such that they are not available for measurement in vitro.  相似文献

Epithelial cells in explants from the mammary glands of euthyroid mature virgin mice are proliferatively dormant. They must undergo DNA synthesis and traverse the cell cycle in vitro before they are able to differentiate fully in response to insulin, hydrocortisone, and prolactin, and synthesize enzymatically active alpha-lactalbumin (measured as lactose synthetase activity). In contrast, glands from hyperthyroid mature virgin mice do not require DNA synthesis in vitro to differentiate. Explants from the euthyroid virgin tissue overcome their dependence on DNA synthesis when 10(-9) M 3,5,3'-triiodo-L-thyronine is added directly to the cultures in addition to the other three hormones. Explants from involuted mammary glands from euthyroid primiparous mice do not require DNA synthesis in vitro to make the milk protein even though they, like explants from mature euthyroid virgin tissue, are proliferatively dormant and do not contain detectable lactose synthetase activity in vivo. Glands from primiparous animals made mildly hypothyroid by ingestion of 0.1% thiouracil in drinking water during 7 wk of involution remain morphologically indistinguishable from glands of their euthyroid counterparts. However, explants from the glands of these hypothyroid animals revert to a state of dependence on DNA synthesis to differentiate functionally. These observations suggest that the dependence on DNA synthesis and cell cycle traversal for hormonal induction of lactose synthetase activity in the mouse mammary gland is controlled by thyroid hormones.  相似文献

PGA1 was incubated with rabbit renal cortical homogenates containing HSA (0–4.5%). The ability of this tissue to readily metabolize PGA1 progressively decreased with increasing HSA levels in the incubates The rate of disappearance of 3H-PGA1 was twice as rapid in rats treated with salicylic acid (S. A.) in comparison to control animals; since only 30% of the injected radioactivity was bound to the plasma of the S.A. treated rats, as compared to 90% bound to control plasma, an association may exist between the degree of binding of 3H-PGA1 and its rate of clearance. The studies indicate that PGA1 interaction with HSA decreases its metabolism in vitro, and slows down its clearance in vivo, implicating HSA as a possible factor in prostaglandins metabolism in vivo.  相似文献

The ability of morphine, leu-enkephalin and β-endorphin to antagonize the binding of 3H-spiroperidol to bovine anterior pituitary membranes was studied. All three drugs were virtually inactive despite their ability to stimulate prolactin secretion invivo and the reported ability of morphine to antagonize the inhibitory effect of dopamine on prolactin release from rat hemi-pituitaries. These results suggest that opiates do not produce their direct effect on prolactin secreation at the pituitary level through an effect on the 3H-spiroperidol binding site. The opiates may antagonize the effect of dopamine at a component of the dopamine receptor which is independent of the 3H-spiroperidol binding site, or the opiates may stimulate prolactin secretion by an effect on the lactotrophes which is independent of dopamine.  相似文献

The potency of seven substituted benzamine drugs (AHR-5531B, AHR-5645B, AHR-6092, AHR-8764, bromopride, sultopride and tiapride) to stimulate rat prolactin (PRL) secretion in vivo was found to be three orders of magnitude greater than that of non-benzamide neuroleptic drugs relative to their respective abilities to inhibit 3H-spiperone binding to bovine anterior pituitary membranes. Nevertheless, the IC50 values for the inhibition of 3H-spiperone binding by the seven substituted benzamide drugs was significantly correlated with their high potency to stimulate rat PRL secretion in vivo. Further, the slope of the regression line for these substituted benzamides paralleled that of a series of butyrophenone, phenothiazine, morphanthridine and dibenzodiazepine neuroleptic drugs. Two benzamide (sulpiride and metoclopramide) and three non-benzamide neuroleptic drugs gave intermediate results. This data suggests that blockade of different subgroups of dopamine receptors in the anterior pituitary gland labeled by 3H-spiperone may be responsible for the in vivo stimulation of PRL secretion by the benzamide and non-benzamide neuroleptioc drugs.  相似文献

Male, weanling rats divided into three groups were maintained for 15 days on a semipurified diet containing either 5% casein fed ad libitum (group 1), 20% casein pair-fed to group 1 (group 2), or 20% casein fed ad libitum (group 3). Animals on day 16 were injected i.p. with 3H-AFB1 (1.90 mg/kg) and were sacrificed six hours later. In both the control and protein deficient animals, binding of AFB1 to DNA was greater than that for chromatin protein. In the protein deficient animals, there was a consistent decrease (70%) in binding to chromatin, DNA and chromatin protein. The decrease in binding to nuclear macromolecules in protein deficient animals is correlated with carcinogenicity and mixed function oxidase (MFO) enzyme activity, and the relationships between carcinogenicity, MFO activity, and binding are discussed.  相似文献

Met5-enkephalin amide, D-Ala2-Met5-enkephalin amide, D-Ala2-Leu5-enkephalin amide, morphine sulfate and naloxone hydrochloride were examined for their effects on growth hormone and prolactin release invivo and invitro. Subcutaneous injection of D-Ala2-Met5 enkephalin amidea, D-Ala2-Leu5 enkephalin amideb and morphine sulfate, but not Met5-enkephalin and amidec, resulted in significant elevations in the serum growth hormone and prolactin of immature female rats. Naloxone blocked the hormone-stimulatory effect of the opioid receptor agonists and when administered alone significantly reduced serum growth hormone and prolactin concentrations. None of the drugs demonstrated a direct action on anterior pituitary tissue growth hormone or prolactin release invitro.  相似文献

Some opiates with morphinan- and benzomorphan-structures possess affinities for neuroleptic receptors as revealed by their abilities to compete with 3H-spiroperidol for common binding sites in rat striatum in vitro (IC50 in the range between 10?6 and 10?5M). The binding of these opiates to neuroleptic receptors appears to be of pharmacological significance, since in vivo studies in mice revealed a small but significant displacement of spiroperidol by high doses of the opiate antagonist levallorphan from specific binding sites in the striatum. In addition, there exists some correlation between the ability of opiates to bind to neuroleptic receptor sites in vitro and their potency to evoke “bizarre behavior” in rats in vivo. In contrast, a wide variety of other opiates having morphine-, morphinone- or oripavine-structure showed no affinity for neuroleptic binding sites in vitro (IC50 greater than 10?4 M). Of the opioid peptides (methionine-enkephalin, leucine-enkephalin and β-endorphin) none has an affinity for neuroleptic binding sites. A variety of other peptides were also investigated but did not interfere with spiroperidol binding. Only ACTH showed a moderate affinity for neuroleptic binding sites.  相似文献

Propoxyphene (Darvon) was compared to SKF 525-A, a prototypical inhibitor of hepatic microsomal mixed function oxidases, to assess propoxyphene's potential to inhibit drug metabolism in morphine tolerant/dependent mice. In vitro, both propoxyphene (Ki = 3.5 × 10?5M) and SKF 525-A (Ki = 4.3 × 10?6M) inhibited the activity of aminopyrine N-demethylase competitively in hepatic microsomes from tolerant/dependent animals. Propoxyphene and SKF 525-A were weaker, noncompetitive inhibitors of aniline hydroxylase activity. In vivo, equimolar doses (0.24 mmoles/kg, i.p.) of each compound inhibited both of the above monooxygenases in the 10,000g supernatant fractions of livers from the tolerant/ dependent animals. Propoxyphene was 40–50% as potent an inhibitor of these activities as SKF 525-A. A dose (300 mg/kg) of propoxyphene napsylate, shown to prevent narcotic abstinence signs with no observable toxicity in withdrawing mice, significantly prolonged the blood levels of injected pentobarbital and tripled pentobarbital sleeping time in these animals. When administered at 300 mg/kg chronically, propoxyphene napsylate acted as an inducer of its own metabolism. Propoxyphene napsylate, then, given acutely to narcotic tolerant/dependent mice, is a potent inhibitor of microsomal drug metabolizing capacity. In skeletal muscle of animals with the phosphorylase b kinase deficiency gene there is < 1% of the normal activity to convert phosphorylase b to a in the presence of Ca++, Mg++, and ATP (1). Correspondingly, there is < 1% of the normal activity to phosphorylate phosphorylase b. Nevertheless, under the same conditions, these extracts catalyze the phosphorylation of troponin at a rate 57% of normal. Phosphorylase b converting activity can be sedimented from skeletal muscle of control mice by centrifugation. This fraction isolated from I strain skeletal muscle extracts phosphorylates troponin at a rate 29–39% of the control. EGTA1 (15 mM) inhibits troponin phosphorylation by 50–60% in this fraction from both strains. The EGTA inhibition is reversed by 15 mM Ca++. Thus the phosphorylase b kinase in skeletal muscle of animals with the phosphorylase b kinase deficiency gene can phosphorylate troponin B, although it shows little or no activity with phosphorylase as a substrate. This observation is consistent with the normal muscle contractility of I strain animals.  相似文献

Rice callus tissue in suspension culture was labeled invivo with 32PO4?3. Approximately 4% of the total RNA extracted from the tissue could be bound to nitrocellulose filters in the presence of 0.5 M KCl. The base composition of this material was: A, 30.6; U, 32.7; G, 18.0; and C, 18.6%. If the RNA was digested with both T1 and pancreatic ribonucleases prior to binding, the base composition of the bound material was: A, 97.8; U, 0.0; G, 1.6; and C, 0.5%. About 14% of the RNA isolated from purified polyribosomes could be bound to filters in 0.5 M KCl. It is concluded that higher plant RNA contains polyadenylate sequences.  相似文献

Dystrophic mice of the C57B1 dy2Jdy2J strain and of the ReJ 129 dydy strain and littermate controls were used to prepare met-tRNAfMet binding factors. The tissues were homogenized and fractions were obtained which contained ribosomes. The binding factors were assayed by the binding of [35S]methionyl-tRNA to control liver ribosomes. The binding, i.e. eukaryotic initiation factor 2 (elF 2) activity, was measured in brain, liver and muscle and in all of these tissues there was a significant decrease in the dystrophic mice. This decrease in initiation factor activity of hindleg muscle resembled, in the direction of the effect, the decrease in elongation factor activity of hindleg muscle resembled, in the of dydy mice previously reported by our laboratory. Thus these two defects, taken together may help to explain the marked wasting of the muscles. The decrease in brain in both strains provides evidence for nervous tissue involvement in genetic dystrophy.  相似文献

The site at which opiate agonists and antagonists act to alter secretion of prolactin, growth hormone and luteinizing hormone as well as the pharmacological specificity of the opiate receptors mediating these effects were examined in rats. Injection of β-endorphin but not a 10 fold higher dose of the non opiate peptide β-endorphin, increased release of prolactin and growth hormone in male rats while inhibiting luteinizing hormone release in ovariectomized, estrogen primed female rats. Prior treatment with naltrexone i.p. blocked these responses. Injection of naltrexone into the hypothalamus lowered prolactin release. In rats with a surgically formed hypothalamic island systemic administration of morphine or naltrexone altered prolactin release in the same manner as was observed in intact animals. In contrast no effects of β-endorphin or naltrexone were observed on the spontaneous secretion of prolactin invitro. In addition β-endorphin did not alter the inhibition of prolactin release produced by apomorphine invitro. The ED50 for stimulation of prolactin release following intraventricular administration of β-endorphin or the synthetic enkephalin analog FK 33-824 was the same, approximately 0.1 ng/rat. However FK 33-824 at 0.2 ng/rat was able to produce much greater analgesia and catatonia than β-endorphin. The metabolism and distribution of β-endorphin was examined but did not account for these differential effects. These results indicate that opiate agonists and antagonists can act at the hypothalamic but not the anterior pituitary level to alter release of prolactin, growth hormone and luteinizing hormone. In addition the data suggest that the opiate receptors mediating release of prolactin may have a different pharmacological specificity from those involved with analgesia and catatonia.  相似文献

The binding in vitro of tritiated aldosterone to domestic duck (Anas platyrhynchos) kidney tissue has been investigated. Using tissue from animals on a normal diet, tritiated aldosterone was specifically bound to kidney cytosol with an apparent equilibrium dissociation constant of about 9 nM and number of binding sites in the 20 fmol/mg protein range. These values did not show statistically significant changes when the cytosol originated from animals with salt activated nasal glands. Kidney cytosols labeled with tritiated aldosterone sedimented with a single peak at 8S in a linear sucrose gradient (10–30%) and this peak was quenched by excess, radioinert aldosterone. Following incubation of labeled cytosols with crude nuclei, the cytosols became depleted of the label and aldosterone was translocated to the Tris-soluble and Tris-insoluble, 0,4 M KCl soluble nuclear fractions. Kidney cytosols metabolized aldosterone extensively to a compound presumed to be 3α,5β-tetrahydroaldosterone. However, only unchanged aldosterone became receptor-bound. It was concluded that the duck kidney possesses aldosterone receptors, though competition studies indicated that the specificity of these receptors might be different from those described in the mammalian kidney.  相似文献

Various dopamine antagonists, including two novel non-neuroleptic drugs domperidone and halopemide, stimulated apomorphine-suppressed prolactin secretion from cultured rat pituitary cells. The potency of these drugs closely paralleled their rank-order in displacing in vitro H3-haloperidol binding in rat striatum reported by others (10). Concentration-effect curves were parallel except those of pimozide and clopimozide which were biphasic : prolactin secretion was stimulated at low concentrations but depressed at concentrations above 25nM. When added alone, pimozide and clopimozide, but none of the other drugs tested, also depressed prolactin secretion. The present findings indicate that prolactin secretion from cultured pituitary cells may provide an in vitro test system suitable to differentiate antagonists of dopamine receptors and possibly to distinguish pure from partial antagonists.  相似文献

This research describes the effects of short-term elemental iodine (I2) and iodide (I) replacement on thyroid glands and mammary glands of iodine-deficient (ID) Sprague-Dawley female rats. Iodine deficiency causes atypical tissue and physiologic changes in both glands. Tissue histopathology and the endocrine metabolic parameters, such as serum TT4, tissue and body weights, and vaginal smears, are compared. A moderate reduction in thyroid size from the ID control (IDC) was noted with both I and I2, whereas serum total thyroxine approached the normal control with both I and I2, but was lower in IDC. Thyroid gland IDC hyperplasia was reduced modestly with I2, but eliminated with I. Lobular hyperplasia of the mammary glands decreased with I2 and increased with I when compared with the IDC; extraductal secretions remained the same as IDC with I2, but increased with I; and periductal fibrosis was markedly reduced with I2, but remained severe with I. Thus, orally administered I2 or I in trace doses with similar iodine availability caused different histopathological and endocrine patterns in thyroid and mammary glands of ID rats. The significance of this is that replacement therapy with various forms of iodine are tissue-specific.  相似文献
