The effects of temperature and salinity on in vitro development of Perkinsus sp. prezoosporangia isolated from cultured Manila clams in Korea were investigated, and the difference in resistance to low temperature between prezoosporangia collected in winter and those collected in summer was compared. Temperature and salinity had significant effects on the development of prezoosporangia, and the developmental rates increased with increasing temperature and salinity. Prezoosporangia isolated in winter sporulated and released motile zoospores at 10 degrees C, although the rates were significantly lower than those at 20 and 30 degrees C. However, no prezoosporangia collected in summer sporulated at 10 degrees C. Low salinities (< or = 10/1000) had a significant negative effect on the development of prezoosporangia. A small number of prezoosporangia sampled in summer did sporulate at 5/1000, but further developments including formation and release of zoospores were not observed. However, prezoosporangia sampled in winter and incubated at 5/1000 released motile zoospores, although the rates were significantly lower than those at higher salinities.  相似文献

The effects of temperature, pH, and NaCl concentrations on the infectivity of zoospores of Leptolegnia chapmanii (Argentine isolate) were determined for Aedes aegypti and Culex pipiens under laboratory conditions. Zoospores of L. chapmanii were infectious at temperatures between 10 and 35 degrees C but not at 5 or 40 degrees C. At the permissive temperatures, mortality rates in young instars were much higher than in older instars and larvae of Ae. aegypti were more susceptible to L. chapmanii than larvae of Cx. pipiens. At 25 degrees C, Ae. aegypti larvae challenged with L. chapmanii zoospores resulted in 100% infection at pH levels ranging from 4 to 10. Larvae of Cx. pipiens exposed to similar pH and zoospore concentrations resulted in increasing mortality rates from 62% to 99% at pH 4 to 7, respectively, and then decreased to 71% at pH 10. Aedes aegypti larvae exposed to L. chapmanii zoospores in NaCl concentrations ranging from 0 to 7 parts per thousand (ppt) at 25 degrees C resulted in 100% mortality while mortality rates for Cx. pipiens decreases from 96% in distilled water to 31.5% in water with 6 ppt NaCl. Control Cx. pipiens larvae died when exposed at a NaCl concentration of 7 ppt. Vegetative growth of L. chapmanii was negatively affected by NaCl concentrations. These results have demonstrated that the Argentinean isolate of L. chapmanii tolerated a wide range of temperatures, pH, and salinity, suggesting that it has the potential to adapt to a wide variety of mosquito habitats.  相似文献

Swimming endurance of whiteleg shrimp, Litopenaeus vannamei exposed to various temperatures (15, 20, and 25 degrees C) and salinities (15, 32, and 40 per thousand) was determined in a swimming channel against one of five flow velocities (5.41, 6.78, 8.21, 10.11, and 11.47 cm s(-1)) for up to 9000 s. No shrimp swam the full 9000 s throughout the experiment. The swimming endurance decreased as swimming speed was increased at any of the temperatures and salinities tested and was significantly affected by temperature and salinity (P<0.05). The power model (nu x t(b) = a) showed a better fit to the relationship between swimming endurance (t, in s) and swimming speed (nu, in cm s(-1)) at any of the temperatures and salinities tested. The swimming ability index (SAI), defined as SAI = integral(0)(9000) vdt x 10(-4) (cm), was found to be temperature- and salinity-dependent in L. vannamei. The optimum temperature and salinity and corresponding maximum SAI were Topt = 21.3 degrees C and SAI(max21.3) = 7.37 cm; Sopt = 27.6 per thousand and SAI(max27.6) = 7.47 cm, respectively. The range of temperatures and salinities within which SAI is >90% of the maximum was estimated between 17.6 and 24.9 degrees C and between 18.5 and 36.7 per thousand, respectively. The results suggest that the power model fits well to the observed endurance estimates and the SAI is a good index to quantitatively describe the overall swimming ability of L. vannamei. Furthermore, temperature and salinity can limit the swimming performance of L. vannamei.  相似文献

Oomycete infections caused by Aphanomyces invadans occur in freshwater and estuarine fishes around the world. Along the east coast of the USA, skin ulcers caused by A. invadans are prevalent in Atlantic menhaden, Brevoortia tyrannus. From laboratory observations low salinities appear crucial to transmission of the pathogen. To better understand aspects of transmission, we characterized sporulation and cyst formation of secondary zoospores of two isolates of A. invadans at different salinities and temperatures. Sporulation occurred only at low salinities. At room temperature (ca. 20-22 C), using "pond water" augmented with artificial sea salts, the endemic strain WIC and the Thailand strain PA7 of A. invadans produced free-swimming secondary zoospores at salinities of 0, 1 and 2 psu (practical salinity unit = per thousand), but not at 4 psu or higher. Secondary zoospores of another species, ATCC-62427 (Aphanomyces sp.), were observed at 1, 2, 4 and 8 psu but not at 0 and 12 psu. Secondary zoospores of all three isolates, especially WIC, were abundant and motile 1-2 d postsporulation. Sporulation was temperature dependent and occurred over a relatively narrow range. No sporulation occurred at 4, 30 or 35 C for either WIC or PA7. For both strains zoospore production within 1-3 d after the initiation of sporulation was more prolific at 25 C than at 20 and 15 C. At 15 C production of zoospores was sustained over 11 d for WIC and 5 d for PA7. At room temperature single WIC secondary zoospores remained motile 12-18 h. Salinities exceeding 4 psu or vigorous shaking caused immediate cyst formation of WIC secondary zoospores. Exposure to menhaden tissue, but not tissues of other fishes to secondary zoospores (WIC), caused rapid (2 h) cyst formation. Cysts were capable of excysting when transferred to 1 psu water within 2-3 h of cyst formation. Cysts that had remained encysted in 6.5 psu for 24 h did not excyst when transferred to 1 psu water. Salinity and temperature requirements for sporulation indicate that juvenile menhaden must acquire infections during rain or in low salinity oligohaline waters.  相似文献

Seaweed cultivation is imperative to augment increasing industrial demand. Ulva fasciata Delile is a potential seaweed for cultivation with applications in food industries. There is a renewed interest in large-scale aquaculture of this species in India due to its envisaged demand in snack food products. In the present study, we have successfully demonstrated the possibility of inducing zoospores in vegetative tissue, effective regeneration and improved growth in this seaweed by manipulating salinity (from 15 to 30 psu) and temperature (from 15 to 35°C). The optimum salinity and temperature requirement for zoospores induction were found to be 15 psu and 25°C, respectively. The quadriflagellate zoospores showed negative phototaxis and the settlement and germination pattern similar to several other green seaweeds. The optimum regeneration (78.53?±?10.05%) was recorded at 25°C and 30 psu salinity. The maximum daily growth rate (16.1?±?0.28%) was at 25°C and 30 psu salinity which corresponded to the field conditions. This method could be further refined at nursery culture to achieve artificial seeding essential for the success of commercial cultivation of this seaweed.  相似文献

In the Marseilles region (French Mediterranean coast), A. clausi is one of the most abundant copepod species of the Gulf of Fos while A. tonsa constitutes the almost exclusive copepod species of the Berre lagoon, a neighbouring semi-closed brackish area communicating with the gulf. As different ecophysiological capabilities to stand the various temperature, salinity and food conditions could explain why these two species do not coexist in the same environment, comparative experiments were performed on metabolism and feeding. The respiration and ammonia excretion of the two species were measured in different combinations of temperature (10, 15 and 20 degrees C) and salinity (15, 25 and 35 per thousand). For each temperature, at the salinity of 35 per thousand, respiration rates were less in A. clausi than in A. tonsa, the contrary being observed at the lowest salinity. At any temperature ammonia excretion was greater at the intermediate salinity in A. tonsa and least in A. clausi. In Acartia tonsa, Q(10) of respiration and excretion were minimum at the lowest salinity, while in A. clausi they were unaffected by salinity variation. The O:N atomic ratio (from respiration and ammonia excretion rates) was significantly more elevated in A. clausi (mean 21.2; range 13.6-28.7) than in A. tonsa (mean 11.3; range 4.2-25) suggesting a more proteinic oriented metabolism in the later. Feeding experiments where Dunaliella tertiolecta30 per thousand) or lagoon (<16 per thousand) salinity. The relationships between ingestion and food concentration in the two species were not significantly different. These different results are compared to other ecophysiological information concerning these Acartia species (survival tolerances, osmotic regulation, feeding behaviour) and are discussed in relation with the characteristics of their niches in the studied region.  相似文献

Physiological responses (oxygen consumption) and behavioral responses (feeding and activity) of the mud snails Hydrobia ulvae and Hydrobia glyca at different salinities (20 per thousand-80 per thousand) and temperatures (20 degrees and 30 degrees C) were studied. After 24 h under experimental conditions, both Hydrobia species already showed maximal activities (>90%) for a wide salinity range (30 per thousand-70 per thousand), with significant differences in activity between species only outside the usual salinity range of the studied lagoon. In contrast, egestion rates of H. glyca were significantly higher at the lowest salinities tested (30 per thousand and 40 per thousand) irrespective of water temperature, whereas egestion rates of H. ulvae were always significantly higher (57% on average) at 20 degrees C than at 30 degrees C and at the usual salinities found in the field (40 per thousand and 50 per thousand). Both species showed an oxyregulatory response to dissolved oxygen concentrations ranging from saturation to 1.5 mg O(2) L(-1), although specific oxygen consumption rates were significantly higher at 30 degrees C than at 20 degrees C (Q(10)=1.47+/-0.08 for H. ulvae and Q(10)=12.1+/-0.06 for H. glyca) and at the lowest salinities (30 per thousand-50 per thousand for H. ulvae and 30 per thousand-40 per thousand for H. glyca). On average, specific rates were higher for the smaller-sized H. glyca (1.64+/-0.03 microg O(2) mg(-1) ash-free dry weight [AFDW]) than for H. ulvae (1.35+/-0.03 microg O(2) mg(-1) AFDW). Despite the overlapping of their tolerances to high temperatures and salinities, the observed interspecies differences could play a certain role in the distribution of H. ulvae and H. glyca in the studied habitat. In particular, the decreasing feeding activity but increasing respiration of H. ulvae at 30 degrees C for salinities that usually occur in the studied lagoon could represent disadvantages to H. glyca during the warm period.  相似文献

Osmoregulation in Litopenaeus vannamei was studied in a factorial experiment at four temperatures (20, 24, 28 and 32 degrees C) and six salinities (10, 16, 22, 28, 34 and 40 per thousand). The isosmotic related points for 20, 24, 28, and 32 degrees C were 754, 711, 822, and 763 mmol/kg, respectively. This species hyperregulates between at salinities of 10 and 20 per thousand and hyporegulates between 20 and 40 per thousand. The isosmotic point in L. vannamei exposed to constant salinities changed in relation to temperature from 717 to 823 mmol/kg. For these experimental conditions, the T-S combination of 32 degrees C and 28 per thousand produced the best growth.  相似文献

Trophozoite, prezoosporangium and zoospore are the 3 main developmental stages that form the life cycle of protozoa of the genus Perkinsus. Several studies have shown that the differentiation of Perkinsus species from the trophozoite to the prezoosporangium stage involves a substantial modification of the antigenic characteristics of these molluscan parasites. With the aim of determining the presence and distribution of antigenic determinants conserved during trophozoite to prezoosporangium differentiation, a polyclonal serum was raised against trophozoites of P. atlanticus purified from parasitized gills of the clam Tapes semidecussatus. Immunocytochemical analyses showed that the serum generated against P. atlanticus trophozoites strongly cross-reacted with the prezoosporangium stage. Immunogold electron microscopy studies revealed that the granular component of the nucleolus, chromatin, cell wall, plasmalemma, lomasomes and vacuolar membrane are the main subcellular structures where the immunodominant epitopes consistently expressed by trophozoites and prezoosporangia are located. Furthermore, analysis of the immunogold staining pattern revealed that the labelling density obtained for prezoosporangia in the nucleolus, cell wall, plasmalemma and lomasomes was significantly higher than that obtained for trophozoites. The most immunoreactive structure in trophozoites was the granular component of the nucleolus, whereas in prezoosporangia it was the lomasome. Interestingly, the main antigenic compartment of P. atlanticus, considering both developmental stages, was the lomasome of the prezoosporangium. These findings show that P. atlanticus trophozoite to prezoosporangium differentiation is accompanied by significant qualitative and quantitative changes in the ultrastructural distribution of the immunodominant antigens shared by these 2 developmental stages.  相似文献

The Mozambique tilapia, Oreochromis mossambicus, is capable of surviving a wide range of salinities and temperatures. The present study was undertaken to investigate the influence of environmental salinity and temperature on osmoregulatory ability, organic osmolytes and plasma hormone profiles in the tilapia. Fish were acclimated to fresh water (FW), seawater (SW) or double-strength seawater (200% SW) at 20, 28 or 35 degrees C for 7 days. Plasma osmolality increased significantly as environmental salinity and temperature increased. Marked increases in gill Na(+), K(+)-ATPase activity were observed at all temperatures in the fish acclimated to 200% SW. By contrast, Na(+), K(+)-ATPase activity was not affected by temperature at any salinity. Plasma glucose levels increased significantly with the increase in salinity and temperature. Significant correlations were observed between plasma glucose and osmolality. In brain and kidney, content of myo-inositol increased in parallel with plasma osmolality. In muscle and liver, there were similar increases in glycine and taurine, respectively. Glucose content in liver decreased significantly in the fish in 200% SW. Plasma prolactin levels decreased significantly after acclimation to SW or 200% SW. Plasma levels of cortisol and growth hormone were highly variable, and no consistent effect of salinity or temperature was observed. Although there was no significant difference among fish acclimated to different salinity at 20 degrees C, plasma IGF-I levels at 28 degrees C increased significantly with the increase in salinity. Highest levels of IGF-I were observed in SW fish at 35 degrees C. These results indicate that alterations in gill Na(+), K(+)-ATPase activity and glucose metabolism, the accumulation of organic osmolytes in some organs as well as plasma profiles of osmoregulatory hormones are sensitive to salinity and temperature acclimation in tilapia.  相似文献

We established monoclonal in vitro cultures of a Perkinsus sp. isolated from the baltic clam Macoma balthica and compared morphological features of various life stages by light and transmission electron microscopy to those of the currently accepted Perkinsus species: Perkinsus marinus, Perkinsus olseni, Perkinsus atlanticus, and Perkinsus qugwadi. Except that trophozoites were slightly larger than those of P. marinus, and that they underwent zoosporulation in culture, observation of our isolate under light microscopy did not reveal striking differences from any Perkinsus species. Perkinsus sp. from M. balthica shared fine structural characteristics with other Perkinsus species that clearly place it within this genus. Although zoospores of Perkinsus sp. from M. balthica were slightly smaller than those from other species, the ultrastructural arrangement and appearance of the apical complex and flagella seem to be identical to those of P. marinus and P. atlanticus. Our isolate also appeared, in some sections, to have cortical alveolar expansions of the plasmalemma at regions other than the anterior end and lobulated mitochondria that were reported as unique for P. qugwadi. Little consensus exists among authors in the assignment of taxonomic weight to any particular morphological feature to designate Perkinsus species. The present study of gross morphology and ultrastructure was complemented with molecular studies reported elsewhere, which propose that Perkinsus sp. from Macoma balthica is a distinct species.  相似文献

The undulated surf clam Paphia undulata supports Thailand's largest shellfishery in the Gulf of Thailand, with landings in 1999 recorded at 70000 t (metric tonnes) yr(-1). We report, for the first time, the prevalence of Perkinsus sp. in clams in the Gulf. A monthly survey from January to December 2001 utilizing the fluid thioglycollate medium (FTM) method showed that average monthly prevalence was 84.7% (n = 360). The monthly percentage of infected clams was generally 100%, with low prevalence in May (66.7%) and no infection in September. The monthly mean infection intensity in terms of Perkinsus sp. cells g(-1) tissue varied from 0 in September to 187 759 +/- 18970 (x +/- SE) in October. No obvious annual variation in intensity and prevalence was observed. Prezoosporangia that developed in FTM were 25 to 75 pm in diameter. A few days after incubation in aerated seawater, the prezoosporangia underwent successive binary cell division and formed motile zoospores (2 to 5 microm long). The zoospores were released into the seawater through a discharge tube formed during the 2- and 4-cell stages. Serial semi-thin sections (1 to 4 pm thickness) of clam tissue (n = 120 clams) showed developing trophozoites 3 to 6 pm in diameter within gills, connective tissue, gonads and, especially, the digestive glands. Microscopic features of different life stages indicated that Perkinsus sp. in Thailand closely resembled P. olseni (= P. atlanticus) reported in Australia, New Zealand, Korea, Japan, Spain and Portugal.

Continuous in vitro cultures of the clam Tapes decussatus parasite Perkinsus atlanticus were established from infected gill fragments, infected haemolymph and parasite hypnospores isolated from infected gill fragments following incubation in Ray's fluid thioglycollate medium (RFTM). No continuous cultures could be initiated from P. atlanticus zoospores. Cultures initiated from hypnospores yielded the highest percentage of continuous cultures (100%, 6/6), followed by cultures initiated from gill fragments (93%, 43/46) and from haemolymph (30%, 3/10). Failures to establish continuous cultures were due to microbial contamination. The source of parasite influenced the success rate, the time taken to establish cultures and the size of cultured cells. In vitro proliferation of parasite cells was mainly by vegetative multiplication. Zoosporulation, yielding motile biflagellated zoospores, was observed at a low frequency (< 1% of dividing cells) in every culture. Morphology of cultured cells examined with light and transmission electron microscopy corresponded to that of P. atlanticus found in clam tissues. Cultured cells enlarged in RFTM and stained blue-black with Lugol's solution, which are characteristics of the Perkinsus species cells. DNA sequences of the internal transcribed spacer (ITS) region of the ribosomal RNA gene complex matched those of P. atlanticus. All cultures were established in a medium designated JL-ODRP-2A that was similar in composition to the culture medium JL-ODRP-1 originally used to propagate Perkinsus marinus in vitro. Proliferation of P. atlanticus in vitro could be supported by the commercial culture medium (1:2 v/v) DME:Ham's F-12 with fetuin.  相似文献

Excretion rates of ammonia-N, nitrite-N, nitrate-N, and dissolved organic nitrogen (DON) for juvenile Litopenaeus vannamei (3.85+/-0.83 g) were quantified in response to nine different combinations of temperature (24, 28, and 32 degrees C) and salinity (10, 25, and 40 ppt) under laboratory conditions. Results indicated that L. vannamei is ammonotelic, with ammonia-N accounting for 61.9-84.3% of total nitrogen (TN) excretion. There were significant effects of temperature and salinity, but no significant interaction between them, on ammonia-N excretion rate (R(AN)). R(AN) increased with increasing temperature, over the interval 24-32 degrees C. R(AN) was lower at 25 ppt than at 10 and 40 ppt, at all temperatures. DON excretion rate (R(DON)) was not significantly influenced by either temperature or salinity; the overall mean R(DON) was about 5.24 μg-N g -1 h -1. However, the percentages of DON in TN (P(DON)) varied from 15.4 to 36.4% under the various temperature-salinity combinations. P(DON) at 28 and 32 degrees C was significantly lower than at 24 degrees C, and P(DON) at 10 ppt was significantly lower than at 25 and 40 ppt. Only very small amounts of nitrogen were excreted by L. vannamei as nitrite-N and nitrate-N.  相似文献

Cyanobacterial mats of the Arabian Gulf coast of Saudi Arabia experience extreme conditions of temperature and salinity. Because they are exposed to continuous oil pollution, they form ideal models for biodegradation under extreme conditions. We investigated the bacterial diversity of these mats using denaturing gradient gel electrophoresis and 16S rRNA cloning, and tested their potential to degrade petroleum compounds at various salinities (fresh water to 16%) and temperatures (5 to 50 degrees C). Cloning revealed that c. 15% of the obtained sequences were related to unknown, possibly novel bacteria. Bacteria belonging to Beta-, Gamma- and Deltaproteobacteria, Cytophaga-Flavobacterium-Bacteroides group and Spirochetes, were detected. The biodegradation of petroleum compounds at different salinities by mat microorganisms showed that pristine and n-octadecane were optimally degraded at salinities between 5 and 12% (weight per volume NaCl) whereas the optimum degradation of phenanthrene and dibenzothiophene was at 3.5% salinity. The latter compounds were also degradable at 8% salinity. The same compounds were degraded at temperatures between 15 and 40 degrees C but not at 5 and 50 degrees C. The optimum temperature of degradation was 28-40 degrees C for both aliphatics and aromatics. We conclude that the studied microbial mats from Saudi Arabia are rich in novel halotolerant and thermotolerant microorganisms with the potential to degrade petroleum compounds at elevated salinities and temperatures.  相似文献

Summary Sporangial formation was found restricted within the range of temperature from 20–31° C. Atmospheric humidity of 100 % did not favour very much in developing sporangia. Free water appeared to be essential in sporangial formation. The range of temperature within which the liberation of zoospores took place was even narrower, 19 to 25° C. Temperature from 21 to 23° C appeared to be best condition for maximum liberation of zoospores. Similarly as with sporangial development atmospheric humidity as high as 100 % had very little effect on the liberation of zoospores unless there was condensation of water. The liberated zoospores in free water were found to germinate only within 20–24° C. At about 22° C the germination rate was found optimum.  相似文献

Physiological adjustment to water of reduced salinity requires energy expenditure. In this study we sought to determine the fitness costs associated with such adjustment in the euryhaline polychaete Capitella sp. I, and the extent to which such costs could be explained by increased rates of energy expenditure. In a series of experiments conducted at 20 degrees C, salinity was reduced from 30 per thousand to either 25, 20, 15, 12, or 10 per thousand within 72 h after the larvae had been induced to metamorphose. Juveniles were reared on fine, organic-rich sediment. Over the next 15-30 days, we determined survival, growth, fecundity, and rates of respiration and feeding (via fecal pellet production). Larval salinity tolerance was also determined. Juvenile survival at salinities as low as 12-15 per thousand was comparable to that at 30 per thousand. The lower limit of salinity tolerance was 10-12 per thousand at 20 degrees C for both larvae and juveniles. Juveniles grew significantly more slowly at 12-15 per thousand in six of the seven experiments. Fecundity, however, was generally highest at intermediate salinities of 20-25 per thousand, and comparable at 30 and 15 per thousand. No individuals released embryos at 12 per thousand over the approximately 30-day observation periods in any of the three experiments in which the worms were reared at this low salinity. Reduced growth rates were not explained by differences in rates of respiration at different salinities: at reduced salinity, respiration rates were either statistically equivalent to (P>0.10) or significantly below (P<0.05) those recorded for animals maintained at 30 per thousand. Lower growth rates at lower salinities were best explained by reduced feeding rates. Further studies are required to determine whether digestive efficiency, growth hormone concentrations, or reproductive hormone concentrations are also altered by low salinity in this species.  相似文献
