Membrane cross bonding--an adhesion between opposing areas of the cytoplasmic face of the red cell membrane--was achieved by treating red cells with heat, diamide, N-ethymaleimide, urea, or by ATP depletion in conjunction with cell shrinking. Membrane cross bonding could be recognized by the shape of the cells upon swelling. Quantitated by the percentage of cross-bonded red cells the effectivity of the treatments decreased in the order given above. Cross bonding was hardly reversible by reducing the diamide-induced S-S bonds with dithioerythritol. The effect of heat and urea treatment as well as ATP depletion was partly reversible. Transmission electron micrographs of the cross-bonded region showed basically parallel membranes. The distance between the respective phospholipid bilayers varied between 40 and 120 nm from cell to cell. Hb-free ghosts prepared from diamide-treated red cells could also be cross bonded. The following conclusions are drawn: spectrin provides the molecular cross link in membrane cross bonding. Aggregation and enrichment of spectrin in the cross-bonded region are probably involved in membrane cross bonding. 相似文献

Interaction of calmodulin with the red cell and its membrane skeleton and with spectrin 总被引:1,自引:0,他引:1
The binding of calmodulin to red cell membrane cytoskeletons and to purified spectrin from red cells and bovine brain spectrin (fodrin) has been examined. Under physiological solvent conditions binding can be measured by ultracentrifugal pelleting assays. The membrane cytoskeletons contained a single class of binding sites, with a concentration similar to that of spectrin dimers and an association constant of 1.5 X 10(5) M-1. Binding is calcium dependent and is suppressed by the calmodulin inhibitor trifluoperazine. The binding showed a marked dependence on ionic strength, with a maximum at 0.05 M, and a steep dependence on pH, with a maximum at pH 6.5. It was unaffected by 5 mM magnesium. An azidocalmodulin derivative, under the conditions of our experiments, did not label the spectrin-containing complex, although it could be used to demonstrate binding to fodrin. Binding of calmodulin to spectrin tetramers and fodrin in solution could be demonstrated by a pelleting assay after addition of F-actin. Calculations (which are necessarily rough) suggest that at the free calcium concentration prevailing in a normal red cell about 1 in 20 of the calmodulin binding sites in spectrin will be occupied; this proportion will rise rapidly with increasing intracellular calcium. To determine whether inhibition of calmodulin binding to red cell proteins disturbs the control of cell shape, as has been suggested, calcium ions were removed from the cell by addition of an ionophore and of ethylene glycol bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid to the external medium. The elastic properties of the human red blood cell membrane have been measured as functions of temperature. The area compressibility modulus and the elastic shear modulus, which together characterize the surface elastic behavior of the membrane, have been measured over the temperature range of 2-50 degrees C with micropipette aspiration of flaccid and osmotically swollen red cells. In addition, the fractional increase in membrane surface area from 2-50 degrees C has been measured to give a value for the thermal area expansivity. The value of the elastic shear modulus at 25 degrees C was measured to be 6.6 X 10(-3) dyne/cm. The change in the elastic shear modulus with temperature was -6 X 10(-5) dyne/cm degrees C. Fractional forces were shown to be only on the order of 10-15%. The area compressibility modulus at 25 degrees C was measured to be 450 dyne/cm. The change in the area compressibility modulus with temperature was -6 dyne/cm degrees C. The thermal area expansivity for red cell membrane was measured to be 1.2 X 10(-3)/degrees C. With this data and thermoelastic relations the heat of expansion is determined to be 110-200 ergs/cm2; the heat of extension is 2 X 10(-2) ergs/cm2 for unit extension of the red cell membrane. The heat of expansion is of the order anticipated for a lipid bilayer idealized as twice the behavior of a monolayer at an oil-water interface. The observation that the heat of extension is positive demonstrates that the entropy of the material increases with extension, and that the dominant mechanism of elastic energy storage is energetic. Assuming that the red cell membrane shear rigidity is associated with "spectrin," unit extension of the membrane increases the configurational entropy of spectrin by 500 cal/mol. 相似文献

The electroporation technique, with field strengths slightly below the critical value Ec for electroporation of red blood cells (RBC), enables the insertion of xeno-proteins into the RBC membrane without damaging the cells. The electro-insertion has been used to insert biotinylated human glycophorin into human RBC membrane and human glycophorin into murine RBC membrane. Binding anti-human glycophorin antibody (10F7) to the murine RBC bearing human glycophorin indicates extracellular orientation of inserted glycophorin. Insertion of about 10(5) glycophorin molecule per cell has been estimated by whole cell ELISA. 相似文献

Properties of red cell membrane proteins: mechanism of spectrin and band 4.1 interaction 总被引:3,自引:0,他引:3
Interactions between human red cell's band 4.1 and spectrin were studied by fluorescence resonance energy transfer and batch microcalorimetry techniques. The association constant (Ka = 8.6 X 10(7) M-1), the stoichiometry (one molecule of band 4.1 to one molecule of spectrin), the reversibility, and the enthalpy (delta H = -6 kcal/mol) were determined. A proton uptake was observed to take place as a result of the spectrin-band 4.1 complex formation. In addition to the protonation of the reaction products, the entropic contribution (-T delta S) has been observed to be responsible for approximately 50% of the binding free energy. We concluded that the environment plays a significant role in the stabilization of the complex. Since band 4.1 has been required for the maintenance of the cytoskeletal stability, small alterations of the binding energies or the degree of interaction could have a pronounced effect on the structure of the erythrocyte membrane. 相似文献

The self-association equilibrium of spectrin has been studied by separating the molecular species present in the cooled reaction mixture by gel electrophoresis. The association constant for formation of the hexamer from dimer and tetramer is lower by an order of magnitude than that for the association of two dimers. The association constant for the formation of the octamer from the hexamer is appreciably larger, and the value appears to reach a constant level for higher oligomers. These observations are explained in terms of conformational strain due to formation of cyclic structures, the distortion being greatest on passing from the tetramer to the hexamer. The association for a single-site interaction between the dimer and a univalent fragment has also been analyzed. The results show that the free energy generated by a single-point interaction is much greater than that obtained by averaging over all pairwise interactions within the oligomers, correcting for the effect of cratic entropy. The results are related to the association state of the spectrin prevailing in the cell. Phosphorylation at the physiological sites in the dimer does not appreciably change the thermodynamics of self-association, at least up to the hexamer. 相似文献

When red blood cells are osmotically shrunk, opposing regions of the inner membrane surface touch each other in the dimple area. In normal red cells such a mechanical contact is undone by reswelling the cells. When the cells are treated with the SH reagents diamide or N-ethylmaleimide, or simply heated to temperatures between 42 and 48 degrees C such a mechanical contact can be made permanent by a process termed 'membrane cross bonding'. Cross bonding also occurred when the cells were treated before mechanical contact was established. The bridge between the two cross-bonded membrane regions may be assumed to be formed by membrane skeletal material. Membrane bridges become visible microscopically when the cells are swollen. These bridges are strong enough to resist the membrane tensions occurring at osmotic lysis. Bridged red cells can be a useful tool in rheology, since they are deformable but cannot adapt to shear flows by membrane tank treading. 相似文献

Phloretin binding to red blood cell components has been characterized at pH6, where binding and inhibitory potency are maximal. Binding to intact red cells and to purified hemoglobin are nonsaturated processes approximately equal in magnitude, which strongly suggests that most of the red cell binding may be ascribed to hemoglobin. This conclusion is supported by the fact that homoglobin-free red cell ghosts can bind only 10% as much phloretin as an equivalent number of red cells. The permeability of the red cell membrane to phloretin has been determined by a direct measurement at the time-course of the phloretin uptake. At a 2% hematocrit, the half time for phloretin uptake is 8.7s, corresponding to a permeability coefficient of 2 x 10(-4) cm/s. The concentration dependence of the binding to ghosts reveals two saturable components. Phloretin binds with high affinity (K diss = 1.5 muM) to about 2.5 x 10(6) sites per cell; it also binds with lower affinity (Kdiss = 54 muM) to a second (5.5 x 10(7) per cell) set of sites. In sonicated total lipid extracts of red cell ghosts, phloretin binding consists of a single, saturable component. Its affinity and total number of sites are not significantly different from those of the low affinity binding process in ghosts. No high affinity binding of phloretin is exhibited by the red cell lipid extracts. Therefore, the high affinity phloretin binding sites are related to membrane proteins, and the low affinity sites result from phloretin binding to lipid. The identification of these two types of binding sites allows phloretin effects on protein-mediated transport processes to be distinguished from effects on the lipid region of the membrane. 相似文献

It is not dubious that the regulation of erythrocyte metabolism occurs across the membrane and that accordingly interactions between cytosolic enzymes and membrane components necessarily exist. Several aspects of such relationships were reviewed. The results of some experiments carried out in non-physiological conditions should be carefully interpreted. However it can be accepted that some enzymes undergo reciprocal translocations between cytosol and membrane and that very probably these transfers play a role in the control of metabolic regulation in the red cell. 相似文献

The two main proposals found in the literature for the stress-free shape of the red cell membrane are (a) the bioconcave shape and (b) the sphere of the same surface area. These possibilities are evaluated in this paper using theoretical modeling of equilibrium membrane shapes according to Zarda et al. (1977. J. Biomech. 10:211-221) and by comparison to experiments on red cells whose membrane shear modulus has been increased by treatment with diamide. Neither proposal is found to be compatible with all the experimental behaviour of native red cells. Neither proposal is found to be compatible with all the experimental behaviour of native red cells. To account for this discrepancy we propose that either the shear modulus of the native membrane is dependent on the membrane strain or that the bending stiffness is higher than estimated by Evans (1980. Biophys. J. 30:265-286). These studies suggest that the bioconcave disk is the more likely possibility for the stress-free shape. 相似文献

Using the patch-clamp technique, a non-selective voltage-activated Na+ and K+ channel in the human red blood cell membrane was found. The channel operates only at positive membrane potentials from about +30 mV (inside positive) onwards. For sodium and potassium ions, similar conductances of about 21 pS were determined. Together with the recently described K+(Na+)/H+ exchanger, this channel is responsible for the increase of residual K+ and Na+ fluxes across the human red blood cell membrane when the cells are suspended in low ionic strength medium. 相似文献

Analysis of the ternary interaction of the red cell membrane skeletal proteins spectrin, actin, and 4.1 总被引:10,自引:0,他引:10
Spectrin dimers interact weakly with F-actin under physiological solvent conditions (with an association constant of about 5 X 10(3) M-1 at 20 degrees C). In the presence of the membrane skeletal constituent, protein 4.1, strong binding is observed; an analysis of the profiles for formation of a ternary complex leads to an association constant of about 1 X 10(12) M-2. This association becomes weaker at low ionic strength, whereas the opposite applies to the spectrin-actin interaction. The stability of the ternary complex is maximal at physiological ionic strength and somewhat above. The effect of temperature in the range 0-20 degrees C on the formation of the ternary complex is small, whereas the spectrin-actin interaction almost vanishes at low temperature. There is no detectable calcium sensitivity in either the binary or the ternary system within the limits of precision of our assay. The ternary complex resembles the natural system in the membrane in that the actin is resistant to dissociation and unavailable in the deoxyribonuclease assay; after selective proteolytic destruction of spectrin and 4.1, all the actin becomes available. In the absence of 4.1, spectrin dimers do not measurably protect the actin against dissociation. 相似文献

Filipin, a polyene antibiotic, interacts with beta-hydroxy sterols such as cholesterol in most cell membranes, forming bumps and pits that are visible by electron microscopy of freeze-fracture replicas. The markedly reduced perturbability of the red blood cell (RBC) membrane, compared to other cells, has been attributed to the constraining influence of the red cell membrane skeleton, the undercoat composed of spectrin, actin, and protein 4.1. To test the influence of the membrane skeleton on filipin-induced perturbation of the RBC membrane, we studied the interaction of filipin with red cells that were inherently devoid of spectrin and RBC in which spectrin had been crosslinked or denatured. These spectrin-deficient, crosslinked, and denatured cells have a fivefold increase in the number of filipin-induced perturbations as compared to control cells, despite equivalent membrane cholesterol content. These findings confirm that the spectrin-based membrane skeleton strongly influences the organization of the membrane so as to limit perturbation by filipin:cholesterol interaction and that for membranes in which the cholesterol content is known, filipin is a useful probe for testing the avidity of spectrin-based cytoskeletal attachment. 相似文献

A possible physical interpretation of the partial detachment of the membrane skeleton in the budding region of the cell membrane and consequent depletion of the membrane skeleton in red blood cell vesicles is given. The red blood cell membrane is considered to consist of the bilayer part and the membrane skeleton. The skeleton is, under normal conditions, bound to the bilayer over its whole area. It is shown that, when in such conditions it is in the expanded state, some cell shape changes can induce its partial detachment. The partial detachment of the skeleton from the bilayer is energetically favorable if the consequent decrease of the skeleton expansion energy is larger than the corresponding increase of the bilayer-skeleton binding energy. The effect of shape on the skeleton detachment is analyzed theoretically for a series of the pear class shapes, having decreasing neck diameter and ending with a parent-daughter pair of spheres. The partial detachment of the skeleton is promoted by narrowing of the cell neck, by increasing the lateral tension in the skeleton and its area expansivity modulus, and by diminishing the attraction forces between the skeleton and the bilayer. If the radius of the daughter vesicle is sufficiently small relative to the radius of the parent cell, the daughter vesicle can exist either completely underlaid with the skeleton or completely depleted of the skeleton. 相似文献

The elastic properties of the cell membrane play a crucial role in determining the equilibrium shape of the cell, as well as its response to the external forces it experiences in its physiological environment. Red blood cells are a favored system for studying membrane properties because of their simple structure: a lipid bilayer coupled to a membrane cytoskeleton and no cytoplasmic cytoskeleton. An optical trap is used to stretch a red blood cell, fixed to a glass surface, along its symmetry axis by pulling on a micron-sized latex bead that is bound at the center of the exposed cell dimple. The system, at equilibrium, shows Hookean behavior with a spring constant of 1.5×10(-6)?N/m over a 1-2 μm range of extension. This choice of simple experimental geometry preserves the axial symmetry of the native cell throughout the stretch, probes membrane deformations in the small-extension regime, and facilitates theoretical analysis. The axisymmetry makes the experiment amenable to simulation using a simple model that makes no a priori assumption on the relative importance of shear and bending in membrane deformations. We use an iterative relaxation algorithm to solve for the geometrical configuration of the membrane at mechanical equilibrium for a range of applied forces. We obtain estimates for the out-of-plane membrane bending modulus B≈1×10(-19)?Nm and an upper limit to the in-plane shear modulus H<2×10(-6)?N/m. The partial agreement of these results with other published values may serve to highlight the dependence of the cell's resistance to deformation on the scale and geometry of the deformation. 相似文献

The quaternary behaviour of DT diaphorase in solution has been investigated by hydrodynamics under a range of conditions.
At neutral pH DT diaphorase is shown to exist as a tightly-associated homodimer in a dimer-tetramer equilibrium. Concentrations
of the chaotropic agent potassium thiocyanate (KSCN) of greater than 200 mm result in irreversible loss of the FAD cofactor and denaturation of the homodimer though this agent appears to be ineffective
in disrupting intermolecular association. These data conform to a model in which under extreme dissociation conditions, the
folded dimer is in equilibrium with the unfolded monomer and are consistent with evidence from the X-ray structure and proposed
catalytic mechanism where both monomers are catalytically interdependent.
