Leukotriene B4 acts through its receptors, BLT(1) and BLT(2), however, their expression in rheumatoid arthritis is unknown. In this experiment, BLT(1) and BLT(2) mRNA expressions in the synovium of rats with collagen-induced arthritis (CIA) at days 1, 3, 7 and 14 after CIA onset were analyzed by RT-PCR. The expression of two immunological and inflammatory factors, S100A8 and S100A9, in the synovium of the arthritic rats was also determined at the indicated time. At d14, the differential expressions of BLT(1) and BLT(2) in the synovium, spleen, peripheral blood mononuclear cells (PBMC) and thymus of CIA rats were analyzed. The results showed that, in the synovium of the arthritic rats, the BLT(1) mRNA expression increased after CIA onset, reached the highest value between d1 and d3, and declined afterwards while the BLT(2) expression increased with time and reached its peak at d14. Both S100A8 and S100A9 expression reached the peak levels between d1 and d3, and decreased to lower levels between d7 and d14. For the analyzed tissues from CIA rats at d14, BLT(1) mRNA was expressed in the thymus with the highest level, followed by the spleen, PBMC and synovium. BLT(2) mRNA was expressed in the thymus the highest as well, but followed by the synovium, spleen and PBMC. Since BLT(1) and BLT(2) play distinct roles during CIA, this study may provide basis for new therapies targeting BLT(1) and BLT(2), respectively, for the treatment of arthritic inflammation at different stages.  相似文献

This study evaluated the hypothesis that neutral (APN) and dipeptidyl-IV (DPPIV) aminopeptidase activity levels would be critical for the susceptibility to arthritis in collagen-induced model (CIA). The macroscopic signs of arthritis in CIA rats were checked and peripheral blood, synovial fluid and synovial tissue from knee joint were withdrawn. Soluble (SF) and solubilized membrane-bound (MF) fractions from the synovial tissue and peripheral blood mononuclear cells (PBMCs) were obtained. APN and DPPIV activities were fluorometrically quantified. Severe swelling in both the entire hind paws was the minimum criterion to select CIA rats with arthritis. These arthritic rats had high APN in plasma, synovial fluid and SF of the synovial tissue, together with low APN and DPPIV in MF of PBMCs and hallmark histological changes in tibio-tarsal joint. CIA rats with no macroscopic signs of arthritis were diagnosed as resistant and they had low APN in MF of the synovial tissue, low DPPIV in SF of PBMCs and high DPPIV in plasma together with histological aspects of tibio-tarsal joint similar to healthy control rats. Data suggested that APN and DPPIV activity levels are related to the development of arthritis, being protective or inducer of the susceptibility. Understanding what is controlling the compartment-specific changes of these peptidases and looking at ways in which to manipulate their activities may lead to a better knowledge of the arthritic processes and novel treatments.  相似文献

Chronic autoimmune inflammation, which is commonly observed in rheumatoid arthritis (RA), disrupts the delicate balance between bone resorption and formation causing thedestruction of the bone and joints. We undertook this study to verify the effects of natural grape-seed proanthocyanidin extract (GSPE), an antioxidant, on chronic inflammation and bone destruction. GSPE administration ameliorated the arthritic symptoms of collagen-induced arthritis (CIA), which are representative of cartilage and bone destruction. GSPE treatment reduced the formation of tartrate-resistant acid phosphatase (TRAP)-positive multinucleated cells and osteoclast activity and increased differentiation of mature osteoblasts. Receptor activator of NFκB ligand expression in fibroblasts from RA patients was abrogated with GSPE treatment. GSPE blocked human peripheral blood mononuclear cell-derived osteoclastogenesis and acted as an antioxidant. GSPE improved the arthritic manifestations of CIA mice by simultaneously suppressing osteoclast differentiation and promoting osteoblast differentiation. Our results suggest that GSPE may be beneficial for the treatment of inflammation-associated bone destruction.  相似文献

Previous studies indicated that disruption of glial function in the spinal cord enhanced electroacupuncture (EA) analgesia in arthritic rats, suggesting glia is involved in processing EA analgesia. To probe into the potential value for clinical practice, the present study was to investigate the effect of propentofylline, a glia inhibitor, on EA analgesia in rats. Mechanical allodynia induced by tetanic stimulation of sciatic nerve (TSS) was used as a pain model. On day 7 after TSS, EA treatment induced a significant increase in paw withdrawal threshold to mechanical stimulation. Intrathecal or intraperitoneal injection of propentofylline relieved TSS-induced mechanical allodynia. The combination of low dosage of propentofylline and EA produced more potent anti-allodynia than propentofylline or EA alone. Immunohistochemistry exhibited that TSS-induced activation of microglia and astrocytes was inhibited significantly by propentofylline. These results indicate that propentofylline and EA induce synergetic analgesia by interrupting spinal glial function.  相似文献

Mangueira Lagoon, located in the extreme south of Brazil, has water with physicochemical characteristics such as alkaline pH and carbonate levels propitious for the growth of the cyanobacterium Spirulina platensis. Previously published studies have shown that Mangueira Lagoon water supplemented with small quantities of carbon and nitrogen is suitable for S. platensis cultivation and can significantly reduce production costs. We studied mixed cultures of Spirulina platensis and the toxic cyanobacterium Microcystis aeruginosa using a 2(3) factorial design in which the three factors were the initial biomass concentration of S. platensis and M. aeruginosa and the type of culture medium (100% Zarrouk's medium or 80% Mangueira Lagoon water plus 20% Zarrouk's medium). The highest S. platensis maximum specific growth rate (mu(max)) occurred in the culture with the highest M. aeruginosa biomass concentration and when undiluted culture medium was used (micro(max) = 0.283 d(-1)). The highest M. aeruginosa specific death rate (k) was obtained in the presence of S. platensis (k = 0.555 d(-1)) and was independent of the initial M. aeruginosa biomass concentration and culture medium, demonstrating that S. platensis cultures are not susceptible to contamination by M. aeruginosa. The culture medium had no significant influence (p > 0.05) on S. platensis micro(max) values, indicating that production costs could be reduced by using a medium consisting of 80% Mangueira Lagoon water plus 20% Zarrouk's medium.  相似文献

Cyanobacterial strains of the genus Spirulina have recently been identified as an excellent source of sulfolipids, some of which possess anti-HIV properties. Thus, to investigate the distribution of sufolipid biosynthesis pathways in Spirulina, a genetic screening/phylogentic study was performed. Five different strains of Spirulina [Spirulina (Jiangmen), Spirulina sp., S. platensis, S. maxima, and Spirulina seawater] sourced from different locations were initially classified via 16S rDNA sequencing, and then screened for the presence of the sulfolipid biosynthesis genes sqdB and sqdX via a PCR. To assess the suitability of these strains for human consumption and safe therapeutic use, the strains were also screened for the presence of genes encoding nonribosomal peptide synthases (NRPSs) and polyketide synthases (PKSs), which are often associated with toxin pathways in cyanobacteria. The results of the 16S rDNA analysis and phylogenetic study indicated that Spirulina sp. is closely related to Halospirulina, whereas the other four Spirulina strains are closely related to Arthrospira. Homologs of sqdB and sqdX were identified in Spirulina (Jiangmen), Spirulina sp., S. platensis, and the Spirulina seawater. None of the Spirulina strains screened in this study tested positive for NRPS or PKS genes, suggesting that these strains do not produce NRP or PK toxins.  相似文献

在钝顶螺旋藻(Spirulina platensis)和极大螺旋藻(Spirulina maximum)接种的第5天至第10天添加不同浓度的亚碲酸钠,研究Te(Ⅳ)胁迫对两种螺旋藻的生长、抗氧化活性系统和脂质过氧化作用的影响。结果表明,通过调节添加碲的时间,可以有效地调节碲胁迫强度,随着加碲时间依次后移,碲胁迫强度逐渐减小,螺旋藻的最终生物量递增。碲对螺旋藻的生物效应与碲胁迫强度有关,也与添加碲时藻所处的生长期有关。实验组Ⅲ(分别在第7、8、第9天三天添加碲)的碲胁迫强度并不是最低的,两种螺旋藻的最终生物量也不是最大,但MDA的含量却是最低的,表明该实验组的生长状态是最佳的。各种抗氧化酶(SOD、GPX、CAT、APX和POD)的活性变化情况较复杂。其中,在碲胁迫下,GPX的活性显著提高。    相似文献

It is now well established that rheumatoid arthritis patients have reduced levels of galactose on their immunoglobulin G (IgG) molecules compared with normal individuals. We have investigated whether, in an experimentally induced model of arthritis, similar glycosylation changes on IgG are to be found. Serum IgG was isolated from collagen-induced arthritic DBA/1 mice and a control group, and the glycosylation of the IgG in these preparations was compared using lectin blotting. The glycosylation of IgG in immune complexes was also analysed. Arthritic mice exhibited similar glycosylation changes on their IgG as observed for rheumatoid arthritis patients. On average, there was less galactose on the IgG from arthritic mice than from the control group, but this difference was of borderline significance. However, theN-acetylglucosamine content of IgG was significatly elevated in arthritic mice. There was no difference in the sialic acid content of IgG in the two groups. The results for immune complexes were similar to those obtained for serum IgG, but the data were limited by insufficient numbers. The similarity in glycosylation changes in collagen-induced arthritis and in patients with rheumatoid arthritis suggests that common pathogenic mechanisms may be involved.  相似文献

对钝顶螺旋藻生物富集Cr(Ⅲ)的影响因素进行了研究。发现螺旋藻对Cr(Ⅲ)的生物富集主要经历了快速的吸附和缓慢的吸收两个步骤;化学键较弱的Cr(Ⅲ)化合物具有较高的富集效率;藻细胞浓度一定时,随着Cr(Ⅲ)浓度的增加,单位重量螺旋藻对Cr(Ⅲ)的富集量不断增加,最后趋于饱和;当Cr(Ⅲ)浓度一定时,随着藻细胞浓度的增加,螺旋藻对Cr(Ⅲ)的总富集量逐渐增加而单位重量藻体的富集量减少。研究还证实,螺旋藻干粉比新鲜藻能富集更多的Cr(Ⅲ);pH值是影响Cr(Ⅲ)生物富集的一个重要影响因素,最佳pH在7左右;温度升高和加强光强均可加强Cr(Ⅲ)的富集;阳离子对Cr(Ⅲ)的富集存在一定的促进或抑制作用。  相似文献

Rheumatoid arthritis (RA) has a negative impact on muscle mass, and reduces patient's mobility and autonomy. Furthermore, RA is associated with metabolic comorbidities, notably in lipid homeostasis by unknown mechanisms. To understand the links between the loss in muscle mass and the metabolic abnormalities, arthritis was induced in male Sprague Dawley rats (n = 11) using the collagen-induced arthritis model. Rats immunized with bovine type II collagen were compared to a control group of animals (n = 11) injected with acetic acid and complete Freund's adjuvant. The clinical severity of the ensuing arthritis was evaluated weekly by a semi-quantitative score. Skeletal muscles from the hind limb were used for the histological analysis and exploration of mitochondrial activity, lipid accumulation, metabolism and regenerative capacities. A significant atrophy in tibialis anterior muscle fibers was observed in the arthritic rats despite a non-significant decrease in the weight of the muscles. Despite moderate inflammation, accumulation of triglycerides (P < 0.05), reduced mitochondrial DNA copy number (P < 0.05) and non-significant dysfunction in mitochondrial cytochrome c oxidase activity were found in the gastrocnemius muscle. Concomitantly, our results suggested an activation of the muscle specific E3 ubiquitin ligases MuRF-1 and MAFbx. Finally, the adipose tissue from the arthritic rats exhibited decreased PPARγ mRNA suggesting reduced adipogenic capacities. In conclusion, the reduced adipose tissue adipogenic capacity and skeletal muscle mitochondrial capacity are probably involved in the activation of protein catabolism, inhibition of myogenesis, accumulation of lipids and fiber atrophy in the skeletal muscle during RA.  相似文献

AIM: To investigate an effective harvesting method for Spirulina platensis. METHODS AND RESULTS: Eighty per cent of S. platensis cells in the logarithmic growth phase were harvested by flotation when the cells were set in a static condition for 2 h. The optimum harvesting time was about day 6 of cultivation. The flotation activity of S. platensis cells was enhanced by the addition of NaCl. CONCLUSIONS: The harvesting of S. platensis by flotation is a cost-effective and straightforward method that can retain the algal quality. The optimum harvesting time of S. platensis can be predicted by the cellular protein to carbon ratio. SIGNIFICANCE AND IMPACT OF THE STUDY: Flotation harvesting is also applicable to other cyanobacteria with gas vesicles.  相似文献

研究了钝顶螺旋藻和极大螺旋藻在含CdCl2水体中的生长状况与摄Cd能力.结果表明:两种螺旋藻皆对CdCl2有较强的耐受能力,但是有不同的摄Cd行为.当CdCl2浓度为6~24mg.L-1,培养96h时,两种螺旋藻对Cd的摄取作用主要表现为藻细胞外的表面吸附;培养10d时,钝顶螺旋藻的胞内Cd含量依然甚微,而极大螺旋藻对Cd的细胞内吸附量却明显增加,24mg.L-1CdCl2处理的极大螺旋藻胞内的Cd吸附量为12mg.L-1CdCl2处理的11.6倍,且略超过细胞表面吸附量.表明在高浓度Cd的长时间胁迫下,两种螺旋藻的摄Cd行为和对Cd的耐受机制具有明显差异,其中钝顶螺旋藻为胞外机制,而极大螺旋藻却为胞内、胞外混合机制,且以胞内机制为主.  相似文献

研究了盐泽螺旋藻的形态、生理生化特性和在不同条件下的生长状况,并与其他螺旋藻进行了比较。盐泽螺旋藻、极大螺旋藻和钝顶螺旋藻在蛋白质的含量、氨基酸组分以及可见光吸收光谱等方面差别不大。盐泽螺旋藻的生长速度最快(世代时间为8.4h)。极大螺旋藻、钝顶螺旋藻1926和钝顶螺旋藻2340的世代时间分别为11、11.8和14.8h。盐泽螺旋藻的光合作用和呼吸作用强度亦大于极大螺旋藻和钝顶螺旋藻。此外,这种藻对盐分和温度还具有较宽的适应范围,在形态上也和其他3种螺旋藻有较大之差异。    相似文献

According to previous reports, adjuvant-induced arthritic rats present reduced activities of the hepatic glucose 6-phosphatase. A kinetic study was done in order to characterize this phenomenon. Microsomes were isolated from livers of arthritic and control rats (Holtzman strain) and the glucose 6-phosphatase was measured at various temperatures (13-37 degrees C) and glucose 6-phosphate concentrations. Irrespective of the temperature, the enzyme from arthritic rats presented a reduction of both V(max) and K(M). Detergent treatment of liver microsomes from control rats increased the activity, but no increase was found when microsomes from arthritic rats were treated in the same way. The mannose 6-phosphatase activity of detergent-treated microsomes from arthritic rats was only 25% of the activity found with detergent-treated microsomes from control rats. Without detergent treatment, the mannose 6-phosphatase activities of both control and arthritic rats were minimal. The activation energy, derived from V(max), was not changed by arthritis. In vivo arthritic rats presented higher hepatic glucose 6-phosphate concentrations, a phenomenon that is consistent with a reduced activity of glucose 6-phosphatase. It was concluded that in arthritic rats, the hydrolase is probably reduced, without a similar change in the translocase activity.  相似文献

The gene encoding ribosomal proteins S12 and probably S7 as well as protein synthesis elongation factors Tu (EF-Tu) and G (EF-G) of Spirulina platensis have been identified and cloned. Gene expression was determined for ribosomal protein S12 by genetic complementation of the appropriate Escherichia coli mutant, whereas for the EF-Tu gene it was determined by production of the protein in E. coli minicells. On the basis of these experiments we suggest the following gene order in the S. platensis chromosome: S12, S7, EF-G, EF-Tu.  相似文献

CD81 belongs to a family of cell-surface protein (tetraspanin) known as one of the up-regulated elements in rheumatoid arthritis synoviocytes. In this study, the therapeutic effect of small interfering RNA targeting CD81 (siCD81) was examined by in vivo electroporation method. Treatment with siCD81 significantly ameliorated paw swelling of collagen-induced arthritic (CIA) rats. In histological examination, hypertrophy of synovium, bone erosion, and degeneration of articular cartilage were milder in rats treated with siCD81 than in the control group and the non-specific siRNA group. Expression of synoviolin, a rheumatoid regulator, was suppressed by siCD81. Thus, therapeutic intervention by targeting CD81 may be used in the treatment of rheumatoid arthritis.  相似文献

A lymphotoxin-beta (LTbeta) receptor-Ig fusion protein (LTbetaR-Ig) was used to evaluate the importance of the lymphotoxin/LIGHT axis in the development and perpetuation of arthritis. Prophylactic treatment with the inhibitor protein LTbetaR-Ig blocked the induction of collagen-induced arthritis in mice and adjuvant arthritis in Lewis rats. Treatment of mice with established collagen-induced arthritis reduced the severity of arthritic symptoms and joint tissue damage. However, in a passive model of anti-collagen Ab-triggered arthritis, joint inflammation was not affected by LTbetaR-Ig treatment precluding LT/LIGHT involvement in the very terminal immune complex/complement/FcR-mediated effector phase. Collagen-II and Mycobacterium-specific T cell responses were not impaired, yet there was evidence that the overall response to the mycobacterium was blunted. Serum titers of anti-collagen-II Abs were reduced especially during the late phase of disease. Treatment with LTbetaR-Ig ablated follicular dendritic cell networks in the draining lymph nodes, suggesting that impaired class switching and affinity maturation may have led to a decreased level of pathological autoantibodies. These data are consistent with a model in which the LT/LIGHT axis controls microenvironments in the draining lymph nodes. These environments are critical in shaping the adjuvant-driven initiating events that impact the subsequent quality of the anti-collagen response in the later phases. Consequently, blockade of the LT/LIGHT axis may represent a novel approach to the treatment of autoimmune diseases such as rheumatoid arthritis that involve both T cell and Ab components.  相似文献

以鄂尔多斯高原碱湖特有的钝顶螺旋藻S1、引进的钝顶螺旋藻S2和极大螺旋藻S3为材料,采用Folin法和茚三酮法分别测定低温下细胞外渗液中可溶性蛋白和游离氮基酸含量。结果表明:处理温度越低,时间越长,螺旋藻细胞外渗液中可溶性蛋白和游离氨基酸的含量越高。S1膜损伤程度最轻,比引进种S2、S3有较强的抗寒能力。判断低温对螺旋藻膜损伤的程度,用可溶性蛋白较游离氨基酸更接近于实际。  相似文献

Increasing interest is being shown in the cyanobacterium Spirulina platensis because of its nutritional properties when used as food supplement and possible therapeutic effects. One of the most important areas being studied is the development of alternative nutrient sources which can be used to decrease the production costs of commercially produced S. platensis and obtain high productivity. Water from Mangueira Lagoon (Rio Grande do Sul State, Brazil) has high levels of carbonates and a high pH and has the potential to be used as a culture medium for S. platensis, although some nutrient supplementation may be required. We tested the effect of unsupplemented Mangueira Lagoon water (MLW) or MLW supplemented with 1.125 or 2.250 mg/l of urea and/or 21 or 42 mg/l of sodium bicarbonate on the growth of S. platensis in fed-batch culture using a 3(2) factorial design and found that there the addition of 1.125 mg/l of urea resulted in a 2.67 fold increase times in the final biomass concentration of S. platensis.  相似文献
