The phytoremediation potential and responses of Sagittaria trifolia to diesel were investigated. In order to elucidate the biochemical and physiological responses of S. trifolia to diesel, the chlorophyll content, root vitality, soluble protein content and antioxidant enzymes activity (peroxidase (POD), catalase (CAT) and antioxidant enzymes superoxide dismutase (SOD)) were determined in the plant tissues after 50 d of diesel treatment. The results showed the presence of S. trifolia significantly improved the removal ratios of diesel, from 21~36% in the control soils to 54~85% in the planted soils. The chlorophyll content, root vitality and soluble protein content all increased at low diesel concentration, then decreased at high diesel concentration. The activities of CAT and POD exhibited peak values at 5 g·kg?1 diesel treatment and declined at higher diesel concentrations. However, the activity of SOD kept stable at lower diesel concentration (1 and 5 g·kg?1), and also declined at higher diesel concentration. Collectively, S. trifolia had the ability to tolerate certain amount of diesel, but when the concentration was up to 10 g·kg?1, the growth of S. trifolia would be restrained. The results also showed that variation of antioxidant enzyme activity was an important response in plants to diesel pollution.  相似文献

Salicylic acid (SA) can alleviate damage to rice plants induced by heat stress, but its role in preventing spikelet degeneration under high-temperature stress has not been documented. Rice plants pretreated with SA (0–50 mmol L?1) were subjected to heat stress at 40?°C at the pollen mother cell meiosis stage for 10 days. The results indicated that there were no significant differences in grain yields and yield components among rice plants that were exogenously sprayed with SA (0–50 mmol L?1) under natural conditions. Under heat stress, the grain yield, spikelet number per panicle and setting rate in response to SA treatments were higher than under the control (0 mmol L?1 SA or NON-SA) treatment, especially with 1 and 10 mmol L?1 SA. A higher grain yield, spikelet number per panicle and setting rate were recorded in these two SA treatments compared with the NON-SA treatment. During this process, soluble sugars, proline, phytohormones including ABA, GA3, BRs, IAA, ZR and JA, and antioxidant enzymes, such as superoxide dismutase, peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), were induced by SA. Moreover, soluble sugars, IAA, POD and APX in the spikelets with SA treatments were not only higher than with the NON-SA treatment but the changing patterns were also similar to that of the spikelet number per panicle under natural conditions and heat stress. Therefore, our results suggest that sugars, antioxidant enzymes and IAA might mediate SA to prevent spikelet degeneration caused by heat stress.  相似文献

We investigated genotoxicity and oxidative stress in the gills of Labeo rohita exposed to 33.6, 67.1, and 100.6 mg L–1of cadmium chloride at 96 h. Genotoxicity was assessed using single cell gel electrophoresis whereas oxidative stress was monitored through lipid peroxidation induction and antioxidant response parameters, namely reduced glutathione (GSH), glutathione peroxidase, glutathione-S-transferase, superoxide dismutase, and catalase (CAT) activities. Significant (p < .05) effect of both concentration and time of exposure was observed on the extent of DNA damage in treated fish. Similarly, malondialdehyde content, level of GSH, and activities of antioxidant enzymes were significantly elevated in treated groups, except CAT. The increased DNA damage and lipid peroxidation (LPO) content along with fluctuation in antioxidant defense system in fish indicated the interaction of cadmium (Cd) with DNA repair processes and production of reactive oxygen species. Thus, Cd is liable for induction of LPO, alteration of antioxidant defenses, and DNA damage in gills of L. rohita.  相似文献

Duckweed Lemna minor L. was grown on Wang culture medium supplemented with lead ions for 24 hours. Metal was tested at 1.5, 3 and 6 mg·dm−3 concentrations. The response of antioxidant enzymes, such as superoxide dismutase, catalase and peroxidase in lead-treated roots of duckweed was investigated. Lead ions had no effect on the spectrum of catalase and peroxidase isoenzymes while a new isoform of superoxide dismutase appeared on the Pb treated roots. A lead-depended increase in activities of superoxide dismutase and peroxidase was observed, whereas catalase activity was maintained at relatively constant values at lower lead concentrations and then decreased markedly below control level.  相似文献

Potassium phosphite (KPhi) is widely used as a resistance inducer to protect plants against fungal pathogens. In the present study, the effect of KPhi on the activation of defence-related enzymes and biochemicals in Pseudoperonospora cubensis-challenged cucumber plants was investigated. Cucumber plants were treated with KPhi before or after inoculation with P. cubensis and leaf samples were collected at different time courses for physiological and biochemical assessments. Results revealed that the activity of reactive oxygen species (ROS)-scavenging enzymes like catalase, guaiacol peroxidase, superoxide dismutase and ascorbate peroxidase as well as proline and total carbohydrates contents were significantly increased by KPhi application, while hydrogen peroxide (H2O2) concentration, as a disease damage indicator was reduced. The maximum activity of ROS-scavenging system was achieved 3–4 days after KPhi application. These findings suggest that KPhi application prior to pathogen infection efficiently triggers plant defence responses which may reduce the disease severity.  相似文献

This study describes various biochemical processes involved in the mitigation of cadmium toxicity in green alga Ulva lactuca. The plants when exposed to 0.4 mM CdCl2 for 4 days showed twofold increase in lipoperoxides and H2O2 content that collectively decreased the growth and photosynthetic pigments by almost 30% over the control. The activities of antioxidant enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR) and glutathione peroxidase (GPX) enhanced by twofold to threefold and that of catalase (CAT) diminished. Further, the isoforms of these enzymes, namely, Mn-SOD (~85 kDa), GR (~180 kDa) and GPX (~50 kDa) responded specifically to Cd2+ exposure. Moreover, the contents of reduced glutathione (3.01 fold) and ascorbate (1.85 fold) also increased substantially. Lipoxygenase (LOX) activity increased by two fold coupled with the induction of two new isoforms upon Cd2+ exposure. Among the polyunsaturated fatty acids, although n ? 3 PUFAs and n ? 6 PUFAs (18:3n ? 6 and C18:2n ? 6) showed relatively higher contents than control, the latter ones showed threefold increase indicating their prominence in controlling the cadmium stress. Both free and bound soluble putrescine increased noticeably without any change in spermidine. In contrast, spermine content reduced to half over control. Among the macronutrients analysed in exposed thalli, the decreased K content was accompanied by higher Na and Mn with no appreciable change in Ca, Mg, Fe and Zn. Induction of antioxidant enzymes and LOX isoforms together with storage of putrescine and n ? 6 PUFAs in cadmium exposed thallus in the present study reveal their potential role in Cd2+ induced oxidative stress in U. lactuca.  相似文献

Tea (Camellia sinensis (L.) O. Kuntze) hyper-accumulates fluoride (F), mainly in the leaves. To understand how tea copes with the stress caused by F, we tracked photosynthesis, antioxidant defense, and cell ultrastructure under different F concentrations (0–50 mg L?1). High F (≥5 mg L?1) caused decreases in photosynthetic and chlorophyll fluorescence parameters. Activated oxygen metabolism was altered by F, as manifested in increasing lipid peroxidation, electrolyte leakage (EL), and accumulation of H2O2. The activities of ascorbate peroxidase (APX, EC 1.11.1.1) and catalase (CAT, EC 1.11.1.6) increased at 0–5 mg L?1 F, but sharply decreased less than 10–50 mg L?1 F. The activity of manganese superoxide dismutase (Mn-SOD, EC 1.15.1.1) decreased with increasing F concentration. Expression of genes encoding antioxidant enzymes were in accordance with their measured activities. The results suggest that the antioxidant enzymes in the tea plant can eliminate reactive oxygen species (ROS) at <5 mg L?1 F, but not at 20–50 mg L?1 F. High F increased the number of epidermal hairs on tea leaves and decreased the stomatal aperture, reducing water loss. The leaf cellular structure appeared normal under 1–50 mg L?1 F, although starch grains in chloroplast increased with increasing F. Proline and betaine play important roles in osmotic regulation in tea plant tolerating F stress. ROS scavenging and greater number of epidermal hairs are likely parts of the tea plant F-tolerance mechanism.  相似文献

Salsola ikonnikovii (Chenopodiaceae), a drought-tolerant plant species that is distributed in sand or light-saline soil in Xinjiang, China, produces seeds (fruits) with attached winged perianths. To study the role of the wing in seed germination under salt stress and to further investigate the growth and physiological responses of the plants to salt stress, the germination behaviour of S. ikonnikovii was determined after winged and non-winged seeds were treated with 0–1000 mmol · L?1 NaCl. Several parameters of two-month old plants that had been treated with NaCl for three weeks were measured. The results revealed that the winged perianths limited germination but protected the seeds from salt damage. The growth of the plants was stimulated by lower concentrations of salt (≤100 mmol · L?1 NaCl), while increasing salt concentrations inhibited growth. The level of reactive oxygen species and malondialdehyde increased significantly at high concentrations of salt. Correspondingly, concentrations of the osmolytes proline, betaine, and soluble sugars, and the activities of antioxidative enzymes (catalase, peroxidase and superoxide dismutase) increased, but the levels of non-enzymatic antioxidants (carotenoids, glutathione) were significantly reduced at high salt concentrations. These results imply that osmotic adjustment and the antioxidative system may work synergistically to ensure that a plant grows normally under high salt concentrations.  相似文献

Heavy metal contaminated agricultural soils are one of the most important constraints for successful cultivation of crops. The current research was conducted to evaluate the role of potassium (K) on plant growth and amelioration of cadmium (Cd) stress in Gladiolus grandiflora under greenhouse conditions. G. grandiflora corms were sown in media contaminated with 0 (C), 50 (Cd50) and 100 (Cd100) mg Cd kg?1 soil. The plants growing in Cd-contaminated media exhibited reduced gas exchange attributes, chlorophyll (Chl) contents, vegetative and reproductive growth as compared to control. The plants raised in Cd contaminated media showed reduced nutrition yet higher Cd contents. However, supplementation of 60 mg Kg?1 K in treated plants (C+K, Cd50+K and Cd100+K) improved quantity of total soluble protein and proline (Pro) along with activity of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD) and ascorbate peroxidase (APX) under Cd stress. Similarly, K supplementation reduced the level of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in treated plants. Potassium supplemented plants exhibited better vegetative and reproductive growth. The improved stress tolerance in K supplemented plants was attributed to the reduced quantity of MDA and H2O2, enhanced synthesis of protein, proline, phenols, flavonides and improved activity of antioxidant enzymes. The present research supports the application of K for alleviation of Cd stress in G. grandiflora.  相似文献

Pot experiments were conducted to investigate the role of selenium in alleviating cadmium stress in Solanum lycopersicum seedlings. Cadmium (150 mg L?1) treatment caused a significant reduction in growth in terms of height and biomass accumulation and affected chlorophyll pigments, gas exchange parameters, and chlorophyll fluorescence. Selenium (10 μM) application mitigated the adverse effects of cadmium on growth, chlorophyll and carotenoid contents, leaf relative water content, and other physiological attributes. Lipid peroxidation and electrolyte leakage increased because of cadmium treatment and selenium-treated plants exhibited considerable reduction because of the decreased production of hydrogen peroxide in them. Cadmium-treated plants exhibited enhanced activity of antioxidant enzymes that protected cellular structures by neutralizing reactive free radicals. Supplementation of selenium to cadmium-treated plants (Cd + Se) further enhanced the activity of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) by 19.69, 31.68, 33.14, and 54.47%, respectively. Osmolytes, including proline and glycine betaine, increased with selenium application, illustrating their role in improving the osmotic stability of S. lycopersicum under cadmium stress. More importantly, selenium application significantly reduced cadmium uptake. From these results, it is clear that application of selenium alleviates the negative effects of cadmium stress in S. lycopersicum through the modifications of osmolytes and antioxidant enzymes.  相似文献

To understand protective roles of nitric oxide against salt stress, the effects of exogenous sodium nitroprusside on activities of lipoxygenase, peroxidase, phenylalanine ammonialyase, catalase, superoxide dismutase enzymes, proline accumulation, and distribution of sodium in soybean plants under salt were determined. Application of sodium nitroprusside + bacterium enhanced plant growth-promotion characteristics, activities of different enzymes, and proline accumulation in the presence of sodium nitroprusside under salt stress. Treatment with NaCl at 200 mM and sodium nitroprusside (0.1 mM) reduced Na+ levels but increased K+ levels in leaves in comparison with the NaCl-treated plants. Correspondingly, the plants treated with exogenous sodium nitroprusside and NaCl maintained a lower ratio of [Na+]/[K+] in NaCl-stressed plants.  相似文献

The current research was performed to evaluate the effect of Bacillus megaterium MCR-8 on mitigation of nickel (Ni) stress in Vinca rosea grown on Ni-contaminated soil (50, 100, and 200 mg Ni kg?1 soil). The treated plants exhibited reduced growth, biomass, gas exchange capacity, and chlorophyll (Chl) content under Ni stress. The inoculated plants growing in Ni-contaminated media exhibited relatively higher growth, total soluble protein, and proline contents. Similarly, bacterial inoculation improved the activity of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) under Ni stress. The Ni stress alleviation in inoculated plants was attributed to the reduced level of malondialdehyde (MDA) and hydrogen peroxide (H2O2), enhanced synthesis of protein, proline, phenols, and flavonides in conjunction with improved activity of antioxidant enzymes. The growth-promoting characteristics of microbe such as 1-aminocyclopropane-1-carboxylate deaminase (ACCD) and phosphate solubilization activity, siderophore, and auxin production capability also improved the growth and stress mitigation in inoculated plants. Furthermore, the inoculated plants exhibited higher value for bioconcentration factor (BCF), translocation factor (TF), and resulted in higher loss of Ni content from soil. The current results exhibited the beneficial role of B. megaterium MCR-8 regarding stress alleviation and Ni phytoextraction by V. rosea.  相似文献

The potential of an aquatic fern, Azolla filiculoides, in phytoremediation of a mono azo dye solution, C.I. Acid Blue 92 (AB92), was studied. The effects of operational parameters such as reaction time, initial dye concentration, fern fresh weight, pH, temperature and reusability of the fern on biodegradation efficiency were investigated. The intermediate compounds produced by biodegradation process were analyzed using GC–MS analysis. An artificial neural network (ANN) model was developed to predict the biodegradation efficiency. The findings indicated that ANN provides reasonable predictive performance (R2 = 0.961). The effects of AB92 solutions (10 and 20 mg L?1) on growth, chlorophylls and carotenoids content, activity of antioxidant enzymes such as superoxide dismutase, peroxidase and catalase and formation of malondialdehyde were analyzed. AB92 generally showed inhibitory effects on the growth. Moreover, photosynthetic pigments in the fronds significantly decreased in the treatments. An increase was detected for lipid peroxidation and antioxidant enzymes activity, suggesting that AB92 caused reactive oxygen species production in Azolla fronds, which were scavenged by induced activities of antioxidant enzymes.  相似文献

Activated oxygen species such as superoxide radicals, singlet oxygen, hydrogen peroxide and hydroxyl radicals can be produced in plants exposed to low, non-freezing, non-injurious temperatures. To prevent or alleviate oxidative injury, plants have evolved several mechanisms which include scavenging by natural antioxidants and enzymatic antioxidant systems such as superoxide dismutases, catalase and peroxidases. Although overproduction of hydrogen peroxide and increased tolerance to oxidative stress can be induced in wheat by low-temperature treatments, data concerning changes in the enzymatic antioxidant systems are almost absent. With the aim to provide this information, antioxidant enzyme (superoxide dismutases, catalase and peroxidases) activities were analysed in leaves and roots of Triticum aestivum cvs Brasilia (frost resistant in field) and Eridano (less frost resistant in field) seedlings grown at day/night temperatures of 24/22°C (control treatment) and 12/5°C (low-temperature treatment). Our data showed that superoxide dismutase activities were unaffected by low-temperature treatment both in leaves and roots. Catalase activity in leaves and roots was decreased in 12/5°C-grown seedlings, but Brasilia maintained higher catalase activity than Eridano. Differences were also observed in guaiacol peroxidase activities between control and acclimated seedlings: Higher guaiacol peroxidase activities were found in the leaves of 12/5°C-grown seedlings while in roots these activities were lower. Moreover, Brasilia guaiacol peroxidase activities were higher than Eridano. Superoxide dismutase and peroxidase zymogram analyses showed that synthesis of new isoforms was not induced by low-temperature treatment. Changes in the activities of antioxidant enzymes induced by cold acclimation support the hypothesis that a frost-resistant wheat cultivar, in comparison with a less frost-resistant one, maintains a better defence against activated oxygen species during low-temperature treatment.  相似文献

Iron toxicity reduces growth of rice plants in acidic lowlands. Silicon nutrition may alleviate many stresses including heavy metal toxicity in plants. In the present study, the ameliorating effects of silicon nutrition on rice (Oryza sativa L.) plants under toxic Fe levels were investigated. Plants were cultivated in greenhouse in hydroponics under different Fe treatments including 10, 50, 100, and 250 mg L?1 as Fe-EDTA and silicon nutrition including 0 and 1.5 mM sodium silicate. Iron toxicity imposed significant reduction in plant fresh weight, tiller, and leaf number. The activity of catalase, cell wall, and soluble peroxidases, and polyphenol oxidase in shoots decreased due to moderate Fe toxicity (50 and 100 mg L?1), but increased at greater Fe concentration. Ascorbate peroxidase activity increased in both roots and shoots of Fe-stressed plants. Iron toxicity led to increased tissue hydrogen peroxide and lipid peroxidation. Silicon nutrition improved plant growth under all Fe treatments and alleviated Fe toxicity symptoms, probably due to lower Fe concentration of Si-treated plants. Silicon application could improve the activity of antioxidant enzymes such as catalase, ascorbate peroxidase, and soluble peroxidase under moderate Fe toxicity, which resulted in greater hydrogen peroxide detoxification and declined lipid peroxidation. Thus, silicon nutrition could ameliorate harmful effects of Fe toxicity possibly through reduction of plant Fe concentration and improvement of antioxidant enzyme activity.  相似文献

The current investigation was carried out to examine the role of arbuscular mycorrhizal fungi (AMF) in alleviating adverse effects of salt stress in Ephedra aphylla. The plants were exposed to 75 and 150 mM sodium chloride (NaCl) stress with and without application of AMF. Salt stress caused significant decrease in chlorophyll and carotenoid contents; however, the application of AMF restored the pigments content in salt-affected plants. Proline, phenols, and lipid peroxidation were increased with increasing concentration of NaCl, but lower accumulation has been reported in plants treated with AMF. NaCl stress also showed increase in different antioxidant enzymes activities (catalase, ascorbate peroxidase, peroxidase, glutathione reductase, and superoxide dismutase), and further increase was observed in plants treated with AMF. The nutrient uptake, Na+ and Na/K ratio increased and potassium and phosphorus were decreased with increasing concentration of NaCl in the present study. However, the colonization with AMF significantly increased K+ and P and reduced Na+ uptake. It is concluded that presown soil treatment with AMF reduced severity of salt stress in E. aphylla through alterations in physiological parameters, antioxidants and uptake of nutrients.  相似文献

Summary The antioxidant enzymes superoxide dismutase, glutathione peroxidase, and catalase were measured in the rete mirabile and gas gland epithelium area of the swim bladder of the toadfish Opsanus tau. When the concentration of enzyme in the swim bladder was compared with the concentration in other organs (kidney, heart, gills) of the same fish, the swim bladder was found to have the highest concentration of superoxide dismutase but relatively low levels of glutathione peroxidase and catalase.Cytochemical assay for the peroxidatic activity of catalase confirmed that virtually no catalase is present in epithelial cells of the gas gland. A similar assay for peroxidase revealed a cyanide-sensitive peroxidase in the multilamellar bodies of these cells. Most of the catalase and peroxidase in the rete mirabile appears to be confined to the granules of neutrophils and the cytoplasm of erythrocytes. Enzyme activity in the neutrophils is not inhibited by 10-1 M KCN. The activity of enzymes participating in the systems of antioxidant protection was assayed in the second leaf and roots of 21-day-old wheat seedlings (Triticum aestivum L.) grown in a medium with nitrate (NO 3 treatment), ammonium (NH+ 4 treatment), or without nitrogen added (N-deficiency treatment). The activities of superoxide dismutase (SOD), peroxidase, ascorbate peroxidase, glutathione reductase, and catalase in the leaves and roots of the NH+ 4 plants was significantly higher than in the plants grown in the nitrate medium. The activity of SOD decreased and ascorbate peroxidase markedly increased in leaves, whereas the activity of ascorbate peroxidase increased in the roots of N-deficient plants, as compared to the plants grown in nitrate and ammonium. Low-temperature incubation (5°, 12 h) differentially affected the antioxidant activity of the studied plants. Whereas leaf enzyme activities did not change in the NH+ 4 plants, the activities of SOD, peroxidase, ascorbate peroxidase, and catalase markedly increased in the NO 3 plants. In leaves of the N-deficient plant, the activity of SOD decreased; however, the activity of other enzymes increased. In response to temperature decrease, catalase activity increased in the roots of NO 3 and NH+ 4-plants, whereas in the N-deficient plants, the activity of peroxidase increased. Thus, in wheat, both nitrogen form and nitrogen deficiency changed the time-course of antioxidant enzyme activities in response to low temperature.  相似文献

The protection of human diploid fibroblasts against high oxygen tension was investigated using various combinations of the three major antioxidant enzymes: superoxide dismutase, catalase and gluthathione peroxidase. α-Tocopherol, a well-known hydrophobic antioxidant, was also tested in combination with the different enzymes. Microinjection of solutions containing different combinations of the three enzymes was compared with the injection of each single enzyme. We observed that the protections given by catalase or superoxide dismutase on the one hand, and by glutathione peroxidase on the other hand, were additive. Surprisingly, the combinations of catalase and superoxide dismutase were less effective than catalase alone and was even toxic at low SOD concentrations. Addition of α-tocopherol following the injection of any of the three enzymes was highly beneficial, but the strongest synergistic effect was obtained with glutathione peroxidase. These results stress the importance of membrane protection by α-tocopherol and indirectly by glutathione peroxidase. They also showed that any injection leading to the decrease in the O2?. or H2 O 2 concentration combined with one of these two protectors is very beneficial for the cells probably by decreasing the OH concentration. This is also proven by the very good protective effect obtained with desferrioxamine.  相似文献

The influence of arsenic (As) on the growth and the antioxidant system of Lemna gibba L. exposed to five concentrations of As (0.0, 0.25, 0.5, 1.0, and 1.5 mg L?1) was studied. Although As exposure reduced relative growth rate, L. gibba continued to grow even after the high accumulation of this element after five days of exposure. The concentration of the superoxide anion was unaffected by As, whereas an increase of hydrogen peroxide concentrations was observed with the increasing of As concentration. Increasing concentrations of As also increased the enzyme activity of superoxide dismutase, peroxidase, and glutathione reductase and the total and nonprotein thiols, up to 0.5 mg L?1, whereas the anthocyanin content increased constantly with As concentration. Catalase and ascorbate peroxidase activities as well as the content of chloroplastic pigments were reduced in plants exposed to all As concentrations. These results support a major role of anthocyanins, nonprotein thiols, and antioxidant scavenging enzymes in L. gibba tolerance to toxic As concentrations.  相似文献
