The aim was to develop an assay for the flowering stimulus ofa photoperiodically-sensitive plant. Phloem exudate solutionswere obtained from photoperiodically induced and non-inducedleaves of Perilla crispa (Thunb.) Tanaka, following treatmentof excised leaves with solutions of EDTA or phytic acid. Theamounts of exudate obtained were estimated polarimetrically,and the conditions for obtaining maximum exudate yields weredetermined. Shoot explants from non-induced P. crispa plantswere grown on a nutrient medium. Under short days the explantsreached anthesis after c. 35 d. In continuous light a smallproportion of the explants showed signs of flowering after 100d. The effects of test substances and phloem exudate on theflowering of explants grown in continuous light was investigated.(?)-ABA (4.0 µM), sucrose (14.6 mM) and phloem exudatefrom both induced and non-induced leaves caused some promotionof flowering. In three experiments, phloem exudate from inducedleaves enhanced flowering to a greater extent than exudate fromnon-induced leaves; in other experiments the effects of thetwo types of exudate were similar. There was no evidence thatABA or sucrose in the phloem exudate caused flowering. Concentrationsof phloem exudate above 2.0 g I–1 were phytotoxic to theexplants. The phloem exudate prepared from the cotyledons of Pharbitisseedlings that had been exposed to a single dark period (oflonger than 10 h) induced flowering in cultured apices excisedfrom non-induced seedlings. The flower-inducing activity ofthe exudate increased as the seedlings were exposed to longerperiods of darkness. The highest activity was associated withthe exudate taken from cotyledons exposed to a single 16-h darkperiod. The activity of the exudate taken from cotyledons exposedto an inductive dark period was clearly reduced by interruptionof the dark period. The addition of exudate taken from threecotyledons to 10 ml of medium resulted in the highest flower-inducingactivity. About 50% of cultured apex explants formed floralbuds, even when the concentration of the exudate was reducedto 0.1 cotyledon equivalents per 10 ml of medium. The cytokinin activity of the root exudate, the leaves, and the apices of vegetative and flowering white lupin plants (Lupinus albus L.) was investigated. The level of cytokinin activity in the root exudate decreased over the 11-week experimental period. Four peaks of cytokinin activity were recorded in the root exudate of 8-week-old plants after fractionation on Sephadex LH-20. Two of these peaks co-eluted with zeatin and zeatin riboside. It is suggested that the remaining peaks represent nucleotide and glucoside cytokinins. The cytokinin levels in extracts of the mature leaves fluctuated slightly over the experimental period. Three peaks of activity co-eluting with zeatin, zeatin riboside and the glucoside cytokinins were recorded in extracts of mature leaves of 8-week-old plants. In the apices cytokinin activity could only be detected in the inflorescences of flowering plants. It would appear that cytokinins produced by the roots accumulate in the fully expanded mature leaves, but are utilized in the rapidly growing apical region and in young expanding leaves.  相似文献

The growth and development of the main stem and the two uppermostaxillary apices of maize were studied during the period fromemergence until flowering. Plants were grown in the field undervarying levels of applied nitrogen fertilizer at two times ofsowing. The effects of daylength were isolated from those oftemperature by making comparisons of growth and developmenton a thermal time basis. The growth and development of the terminal (male) apex and thetwo uppermost axillary (female) apices followed the same patterns,with apex volumes increasing curvilinearly with increase innumber of leaf or husk primordia. The RGR(relative growth rateof volume) of the terminal apex was, however, only approximatelyone-tenth of the axillary apices. There was no difference ingrowth and development between the first and second axillaryapices before flowering: other factors, such as accumulationof dry weight, rather than primordia production, must be responsiblefor an axillary apex's potential to bear grain. Applied N, andto a lesser extent short days, increased the rates of growthand development of all the apices. For example, applied N increasedthe RGR (volume) of the apical domes, and the rate of productionof spikelet primordia, by about 25%. All axillary apices and treatments showed a single relationshipbetween number of spikelets and surface area of the ear: a favourableenvironment (e.g. high N) simply accelerated the progressionof spikelet production and area expansion along this singlepath. We conclude that this path is probably determined geneticallyand that N and time of sowing influence potential yield of maizethrough effects on the surface area of the ear but not on thedensity of spikelets formed. Response of vernalization to low temperature (2C) was studiedin a winter-annual form of Lolium temulentum L., using imbibedseeds, excised mature and immature embryos, shoot apical meristemsand callus tissue. Excised embryos, as early as 5 days afteranthesis, and excised shoot apices could be vernalized as effectivelyas imbibed seeds. Cold treatment of developing embryos withinthe ear, however, appeared to have no vernalizing effect. Plantsderived from callus by somatic embryogenesis showed varyingdegrees of vernalization response. The vernalization response in L. temulentum, as in winter annualcereals, appears to be located in the shoot apical meristemand the vernalized condition can be transmitted to new axillarymeristems formed from it. Flowering of seedlings of Pharbitis nil, strains Violet andTendan, cultured in modified White's medium, was promoted bymedium dilution, the critical dark period being shortened byabout 15 min. Dilution of the N source alone was enough to causethe medium-dilution effect. Dilution of the culture medium duringthe day before and on the day of exposure to the dark-period(a total of two days) caused the maximum dilution effect. TheC and N contents of the cotyledons and of the shoot apices changedrapidly in response to medium dilution. In 1/2-strength White'smedium with 1/1,000 strength NO3 which was most effectivefor flower promotion, the C-N ratio was highest. In 1/2-strengthmodified White's medium, in which flowering was lowest withthe longest critical dark period, the C-N ratio was lowest.Thus, there is a close relation between flowering response andthe C-N ratio in cotyledons or shoot apices of Pharbitis nil. Procedures are described for producing axillary shoots fromseedling apices and adventitious shoots from petioles and leaf-derivedcallus of sugar beet cultivars. The rate of adventitious shootregeneration from petioles was influenced by temperature, BAPconcentration of the medium, and the time in culture of theseedling apices from which the petioles were excised. Petiolesectioning confirmed that adventitious shoots originated inthe sub-epidermal parenchyma. Two distinct types of callus wereproduced from leaf explants, but only white friable callus wascapable of shoot development. This callus developed from browntissue and was composed of thin-walled cells with dense cytoplasmand prominent nuclei. Green compact callus with thick-walledlignified cells developed from green tissue, but did not produceshoots. Successful seed sterilization and shoot regenerationfrom petiole explants and callus was cultivar-dependent. Adventitiousshoots were rooted and successfully transplanted to pottingcompost under glasshouse conditions. After removal of the embryo from developing seeds of Vicia fabaL. and Pisum sativum L., the ‘empty’ ovules werefilled with a standard solution (pH 5.5). Seed coat exudatesof both species were collected during relatively long experiments(up to about 12 h) and the concentration of sugar (mainly sucrose),amino acids and phosphate in the exudate measured. A discussionis presented on the amino acid/sugar ratio and the phosphate/sugarratio in the seed coat exudate. A pretreatment (15 min) withp-chloromercuribenzenesulphonic acid (PCMBS) reduced the releaseof sugar, amino acids and phosphate from broad bean seed coats.After excision of ‘empty’ ovules of Vicia faba andPisum sativum from the maternal plant, 2–4 h after thistreatment a strong difference became visible between sucroserelease from excised seed coats and sucrose release from attachedseed coats. Similarly, when the rate of phloem transport ofsucrose into an ‘empty’ ovule of Vicia faba or Pisumsativum was reduced by a sub-optimal mannitol concentrationin the solution, a reduced rate of sugar release from the seedcoat could be observed. Excision and treatment with a sub-optimalmannitol concentration reduced the release of amino acids toa lesser extent than for sucrose. The crude water extracts of leaves of many plant species belongingto Spermatophyta and some belonging to Bryophyta induced floweringof Lemna paucicostata 151 (PI51) under continuous light, atthe concentrations equivalent to 0.1 to 10 mg fr wt leaf per10 ml culture medium (mg fr wt/10 ml). The extract of Salvinia(Pterydophyta) added together with the extract of Lemna at aconcentration lower than that necessary to cause flowering alsoinduced flowering. The activity of the water extracts of someplants varied considerably from experiment to experiment dueto unknown factors, but the extracts of Pharbitis nil strainViolet, a sensitive short-day plant, always showed a high activity,as did the extracts of Lemna paucicostata reported previously. The extract of Pharbitis cotyledons induced flowering of P151even at 0.3 mg fr wt/10 ml, and significantly promoted floweringof L. paucicostata 441 and 6746 at 1–3 mg fr wt/10 ml.Ex-udate from the cuttings of the seedlings was also active.However, neither the activity of the water extract nor thatof the exudate could be correlated with photoperiodic floralinduction. On the other hand, the extract of leaves or cotyledonshad higher activity (on a fr wt basis) than that of other organs,and the activity of the extract of cotyledons changed with theirage roughly in parallel with their photoperiodic sensitivity. The roots of light-grown pea seedlings (Pisum sativum L. cv.Alaska) were excised at 250, 500 or 1500 µm from the body/capjuncture. Tips were sampled for 7 d after excision to monitorthe polarity and structure of the regeneration response. Inroots excised at 250 µm a new single apex regeneratedwithout swelling or any sign of repair. The new root cap appearedidentical to the control. After the 500 µm excision tworesponses occurred. In 45% of the roots examined, a single newapex regenerated, in 55% two or three new apices regeneratedto form a dichotomized or trichotomized root in the same longitudinalaxis as the primary root. In roots excised at 1500 µm,one, two or three lateral roots formed adjacent to the xylemat right angles to the primary root axis. In most instancesthe new roots developed triarch xylem. The discussion concentrateson the relationship of the differentiation state of the vascularcylinder and the pericycle, at the excision site, to the regenerationresponse. Tussocks of three clones of the perennial grass Deschampsiacaespitosa were exposed to natural temperatures, for successiveperiods with increments of one week, throughout each seasonin four consecutive autumn/winter seasons. After exposure theywere returned to a warm, illuminated glasshouse and the incidence,intensity and timing of flowering were recorded. The year-to-year variations in exposure time requirement forfull flowering were very highly correlated (r2 > 0.98) withmean air temperature in the period between mid-September andearly December. The relationship with the natural logarithmof accumulated air temperature was similarly good. Daily meanair temperature was also demonstrated to be a good indicatorof daily mean tussock temperature (r2 = 0.91), in a series ofmeasurements in the autumn of 1976. More prolonged exposure increased the proportion of apices ina tussock which flowered, but the maximum value at the end ofa whole winter was only about 15 per cent. The cumulative vernalizationeffects of naturally fluctuating temperature regimes are analysedand the significance of these findings for the maintenance ofperenniality in clonal grasses is considered. The relative abundance of ureides [(ureide-N/ureide-N + NO3–N+ amino-N)x 100] in vacuum-extracted xylem exudate of soyabean(Glycine max [L.] Merr.) can be used to quantify N2 fixationactivity. We report experiments designed to (1) develop appropriateprocedures for sampling and storage of xylem exudate extractedfrom plants in the field and (2) develop a single plant, non-destructivesampling procedure, useful for evaluation of segregating breedingmaterial. Soyabean cultivars Bragg, Essex, Davis, Hardee andDodds were grown at three sites at Brecza, New South Wales,on a deep, alkaline vertisol. All plots were inoculated witheffective Bradyrhizobium japonicum CB 1809. Vacuum strengthsof between 50•5 and 80•8 kN m–2 did not affectthe relative abundance of ureides of xylem exudate. Delays betweenplant sampling and extraction of exudate, however, resultedin time-dependent increases in the relative ureide contents.Extraction should immediately follow shoot detachment to allowvalid comparison of treatments and accurate interpretation ofdata. The relative ureide contents of exudate showed only minordiurnal fluctuations. Exudate, once collected, could be stabilizedby dilution (1:1) in absolute ethanoL Both diluted and undilutedsamples were stable for at least 7 d at 4°C. Differencesduring early flowering, but not subsequently, in the compositionsof N solutes of exudate extracted from either the whole shoot,the top half or the lower half indicated the possibility ofnon-destructive sampling where seed, as well as xylem exudate,could be harvested. Best results in terms of volume of exudateextracted, treatment separation and seed production were obtainedwhen single plants were sampled during late flowering by detachmentof shoots at internode 5. Plants of Lolium temulentum L. cv. Ceres grown under short days (SDs) can be induced to initiate inflorescences either by exposure to one long day (LD) or by single applications of some gibberellins (GAs), which also enhance the flowering response to one LD. Single doses of up to 25 μg per plant of C-16, 17-dihydro-GA5 were about as effective as GA5 for promoting flowering after one LD but inhibited stem elongation by up to 40% over three weeks. The promotion of flowering but not the inhibition of elongation by 16, 17-dihydro-GA5 was reduced in SDs or in LDs low in far-red (FR) radiation. With shoot apices cultured in vitro, 16, 17-dihydro-GA5 was more florigenic than GA3 for apices excised after one LD of 14 h or more, but less florigenic for apices excised from plants in shorter days. 16, 17-Dihydro-GA5 was ineffective compared with GA1, GA3 and GA5 for α-amylase production by half-seeds of Lolium, a response concordant with its effect on stem elongation. As with GA5, 16, 17-dihydro derivatives of GA1, GA3, GA20 and several other GAs were more effective for flowering and less effective for stem elongation than the GAs from which they were derived. Hydroxylation at C-17 and/or C-16 generally reduced the effectiveness of 16, 17-dihydro-GA5 for flowering. These results extend the known features of GA structure which favour flowering relative to stem elongation in L. temulentum. Moreover, C-16, 17-dihydro-GA5 mimics, in its daylength- and wavelength-dependence and lack of stem elongation, characteristics of the LD stimulus in L. temulentum.  相似文献

The flowering mutant dn in sweet pea was used as a tool to study14C-assimilate and dry matter partitioning with respect to nutrientdiversion theories on the control of flower initiation. Wildtype plants (Dnh) are photoperiodic and exhibit late floweringand profuse basal branching in short days while mutant plants(dn) are day neutral, early flowering and devoid of basal laterals.In short days, dn plants exported a significantly greater proportionof assimilate acropetally than (Dnh) plants and the upper portionof dn plants had a greater dry weight. These differences werereduced dramatically when basal laterals were excised regularlyfrom the (Dnh) plants although the difference in flowering remained.However, the effect of dn on resource allocation within theapical region may be more important in regard to flowering thanthe effect on acropetal versus basipetal movement. In shortdays, the dn plants partitioned significantly more resourcesinto their internodes and petioles, and less into their leaflets,than Dnh plants as shown by dry weight and 14C-assimilate measurements.These differences were apparent from as early as node 7 up tothe node of flower initiation in dn plants (node 30) and theywere not eliminated by removal of basal laterals from Dnh plants.Differences between dn and Dnh plants in partitioning and floweringwere largely eliminated under long days. The fact that in thisspecies a single gene influences both resource allocation andflower initiation lends further support to nutrient diversionhypotheses on the control of flowering. The complete removal of the reproductive structures once andshoot apices of soybeans (Glycine max L. Merrill) during earlyanthesis but before the rapid seed development stage significantlyincreased flowering and pod set in greenhouse and field grownplants. The treated plants had darker green leaves, shorterstems and petioles and retained their chlorophyll content longerthan control plants. Pod maturation was also delayed. Althoughdecapitation and the removal of reproductive structures increasedthe number of 3- or more-seeded pods in all varieties tested,seed weight per plant was not consistently increased. The role of cotyledons in hypocotyl elongation caused by gibberellicacid was studied using young seedlings of lettuce, Lactuca saliva,var. ‘Grand Rapids’. Removal of cotyledons fromintact seedlings resulted in a depression of hypocotyl elongationcaused by gibberellic acid. Gibberellic acid-induced hypocotylelongation in decotylized seedlings, was however, substantiallyenhanced by incubating the seedlings together with excised cotyledons.The exudate from excised cotyledons also enhanced the effectof gibberellic acid on hypocotyl elongation in decotylized seedlings.This active principle (named the cotyledon factor) in the cotyledonexudate was stable against heating at 100?C for 15 min, permeatedthe dialysis membrane, and was extractable with ethyl acetate.Biological activity of the cotyledon factor was not replacedby indole-3-acetic acid, kinetin, cyclic AMP, vitamins, sucroseor inorganic nutrients. The development of primordia as leaves, petals, or as organsintermediate between leaves and petals can be regulated by photoperiodin Impatiens. In intermediate organs only some parts of theorgan differentiated as petal, and then only in some cell layers.Allometric measurements of primordium shape suggested that intermediateorgans may begin development as petals, and that their intermediatecharacter at maturity resulted from a switch of some parts ofthe organs from petal to leaf development when the primordiawere between 0.5 and 1 mm long. In reverted apices made to re-flower,primordia were not completely determined as leaves until theywere about 750 µm long. Determination typically occurredfirst at the tips and last at the bases of these primordia.The determination of primordia as leaves or petals in Impatiensis discussed in relation to primordium determination in otherspecies. It is suggested that the lack of commitment to flowermay result in relatively late primordium determination in Impatiens. In Chenopodium rubrum there exists a correlation between theage of the seedlings and the effectiveness of photoperiodicinduction. The younger the plants the more effective was photoperiodictreatment. In three-day-old seedlings one short day was sufficientto promote incomplete flowering, while two short days broughtabout 100 per cent flowering. With six-, eight-, and ten-day-oldplants exposed to two or three short days quantitative differenceswere observed in the earliness of flowering and the percentageof flowering plants. The effects of continuous light and ofshort days with a light break preceding the inductive treatmentwere compared. The results obtained indicate that the inhibitoryeffect of plant age cannot be attributed solely to the appearanceof inhibitors under continuous light but changes of growth patternin plants of different age should also be taken into consideration. The inhibition of RNA synthesis in shoot apices brought aboutby 6-azauridine resulted also in a flowering stimulation, providedthat the inhibitor was applied one or two days prior to inductionand the inductive process itself remained undisturbed. Thisstimulation was accompanied by inhibition of vegetative growthand by a decrease of RNA concentration in the cytoplasm as estimatedcytophotometrically. The competition between growth of vegetative organs and floraldifferentiation affects the response to inductive treatment.The suppression of growth can result in enhancement of flowering.  相似文献
