While the mutualistic interaction between plants and AM fungi is of obvious importance to ecosystem processes, the factors influencing the ecological and evolutionary dynamics within this interaction are poorly understood. The mutual interdependence of plant and AM fungal relative growth rates could generate complex dynamics in which the composition of the AM fungal community changes due to association with host and this change in fungal composition then differentially feeds back on plant growth. I first review evidence for feedback dynamics and then present an approach to evaluating such complex dynamics. I specifically present evidence of host-specific differences in the population growth rates of AM fungi. Pure cultures of AM fungi were mixed to produce the initial fungal community. This community was then distributed into replicate pots and grown with one of four co-occurring plant species. Distinct compositions of AM fungal spores were produced on different host species. The AM fungal communities were then inoculated back onto their own host species and grown for a second growing season. The differentiation observed in the first generation was enhanced during this second generation, verifying that the measure of spore composition reflects host-specific differences in AM fungal population growth rates. In further work on this system, I have found evidence of negative feedback through two pairs of plant species. The dynamic within the AM fungal community can thereby contribute to the coexistence of plant species.  相似文献

应用样方法,以三叶草(Trifolium repens)作盆栽扩繁,利用湿筛倾析法和蔗糖离心法分离土壤孢子,研究了广东省常绿阔叶林、人工林(台湾相思林、湿地松林、刚果按林)和木薯地等3种植物群落11个土样的丛枝菌根菌(Arbuscular mycorrhizal fungi,AMF)孢子组成及分布状况.根据孢子形状、颜色、大小、孢壁饰纹及连孢菌丝等形态,将所分离的孢子划分为20个形态型(morphotypes),分属于3科5属.其中球囊霉属(Glomus)有10种形态型,无梗囊霉属(Acaulospora)5种,巨孢囊霉属(Gigaspora)和盾巨孢囊霉属(Scutellospora)各2种,内养囊霉属(Entrophospora)1种.在20个形态型中,以球囊霉属最丰富,分布最广.其中4种类型Glomus dolichosporum(TypeI)、Glomus eburneum(TypeJ)、Glomus versiforme(TypeM)和Acaulospora scrobiculata(TypeD)在常绿阔叶林、人工林和木薯地中均有出现,其余类型仅见个别土样.  相似文献

The effects of spatial heterogeneity in negative biological interactions on individual performance and species diversity have been studied extensively. However, little is known about the respective effects involving positive biological interactions, including the symbiosis between plants and ectomycorrhizal (EM) fungi. Using a greenhouse bioassay, we explored how spatial heterogeneity of natural soil inoculum influences the performance of pine seedlings and composition of their root‐associated EM fungi. When the inoculum was homogenously distributed, a single EM fungal taxon dominated the roots of most pine seedlings, reducing the diversity of EM fungi at the treatment level, while substantially improving pine seedling performance. In contrast, clumped inoculum allowed the proliferation of several different EM fungi, increasing the overall EM fungal diversity. The most dominant EM fungal taxon detected in the homogeneous treatment was also a highly beneficial mutualist, implying that the trade‐off between competitive ability and mutualistic capacity does not always exist.  相似文献

1.  Many grass species are associated with maternally transmitted fungal endophytes. Increasing evidence shows that endophytes enhance host plant success under varied conditions, yet studies have rarely considered alternative mechanisms whereby these mutualistic symbionts may affect regeneration from seed.
2.  We performed a microcosm experiment to evaluate whether infection with Neotyphodium occultans affects recruitment in the annual grass Lolium multiflorum either directly, by infecting the seeds, or indirectly, by altering the suitability of recruitment microsites through the litter shed by host plants. Endophyte effects on establishment were tested for different litter depths and watering regimes under natural herbivory by leaf-cutting ants.
3.  Seed infection increased seedling emergence through the litter as well as final recruitment, irrespective of microsite conditions. However, litter produced by infected plants delayed emergence and decreased density of both infected and non-infected grass populations.
4.  Individual plant biomass did not change with seed infection but was increased under deep litter from endophyte-infected plants. Although seed infection did not protect establishing plants from leaf-cutting ants, herbivory was reduced in the presence of deep litter shed by infected plants.
5.  We conclude that fungal endophytes may affect host plant recruitment across subsequent generations not only by infecting the seeds but also through the host's dead remains. While the former effect entailed an advantage to infected plants, litter-mediated effects did not discriminate by infection status, and generally promoted the establishment of fewer and larger plants. Thus hidden foliar symbionts may play an underappreciated role in maintaining host species dominance through the litter produced by prior patch occupants.  相似文献

Summary Increasing atmospheric carbon dioxide (CO2) concentration is expected to increase plant productivity and alter plant/plant interactions, but little is known about its effects on symbiotic interactions with microorganisms. Interactions between perennial ryegrass, Lolium perenne (a C3 plant), and purpletop grass, Tridens flavus (a C4 plant), and their clavicipitaceous fungal endophytes (Acremonium lolii and Balansia epichloe, respectively) were investigated by growing the grasses under 350 and 650 l l 1 CO2 at two nutrient levels. Infected and uninfected perennial ryegrass responded with increased growth to both CO2 enrichment and nutrient addition. Biomass and leaf area of infected and uninfected plants responded similarly to CO2 enrichment. When growth analysis parameters were calculated, there were significant increases in relative growth rate and net assimilation rate of infected plants compared to uninfected plants, although the differences remained constant across CO2 and nutrient treatments. Growth of purpletop grass did not increase with CO2 enrichment or nutrient addition and there were no significant differences between infected and uninfected plants. CO2 enrichment did not alter the interactions between these two host grasses and their endophytic-fungal symbionts.  相似文献

When molasses was added during ensilage of three tropical grasses [hamil grass (Panicum maximum cv. Hamil), pangola grass (Digitaria decumbens) and setaria (Setaria sphacelata cv. Kazungula)] the final pH, concentration of fermentation acids (except lactic acid) and NH3–N content were all similar after 100 days of incubation. Pangola grass silage had significantly higher lactic acid content (66 g/kg dry matter) than the other two. Adding either 4 or 8% (w/w) molasses reduced NH3–N, volatile fatty acid content and pH but increased lactic acid content in the final silages. Numbers of lactic acid bacteria remained approximately constant during the course of the fermentation, although large differences were noted in the species composition of the populations. At the time of ensiling, only Pediococcus spp. and Leuconostoc spp. were detected. By 5 days, the homo-fermentative population, notably Lactobacillus plantarum, dominated (43%) and remained dominant. Hetero-fermentative rods were only detected in the 100-day silage, where they represented 29% of the strains isolated. Homo-fermenters were more abundant in pangola (60%) and setaria (47%) silages than hamil (27%) silages. Homo-fermenter populations were lowest in the 12-week forage. Molasses additions increased homo-fermenter populations. Pangola grass gave the best quality silage but, since the water-soluble carbohydrate content in the grasses was insufficient to promote a strong lactic fermentation, the addition of 20 to 30 kg molasses/tonne should achieve satisfactory preservation.M. To test the hypothesis that hypoxia centrally affects performance independently of afferent feedback and peripheral fatigue, we conducted two experiments under complete vascular occlusion of the exercising muscle under different systemic O(2) environmental conditions. In experiment 1, 12 subjects performed repeated submaximal isometric contractions of the elbow flexor to exhaustion (RCTE) with inspired O(2) fraction fixed at 9% (severe hypoxia, SevHyp), 14% (moderate hypoxia, ModHyp), 21% (normoxia, Norm), or 30% (hyperoxia, Hyper). The number of contractions (performance), muscle (biceps brachii), and prefrontal near-infrared spectroscopy (NIRS) parameters and high-frequency paired-pulse (PS100) evoked responses to electrical muscle stimulation were monitored. In experiment 2, 10 subjects performed another RCTE in SevHyp and Norm conditions in which the number of contractions, biceps brachii electromyography responses to electrical nerve stimulation (M wave), and transcranial magnetic stimulation responses (motor-evoked potentials, MEP, and cortical silent period, CSP) were recorded. Performance during RCTE was significantly reduced by 10-15% in SevHyp (arterial O(2) saturation, SpO(2) = ~75%) compared with ModHyp (SpO(2) = ~90%) or Norm/Hyper (SpO(2) > 97%). Performance reduction in SevHyp occurred despite similar 1) metabolic (muscle NIRS parameters) and functional (changes in PS100 and M wave) muscle states and 2) MEP and CSP responses, suggesting comparable corticospinal excitability and spinal and cortical inhibition between SevHyp and Norm. It is concluded that, in SevHyp, performance and central drive can be altered independently of afferent feedback and peripheral fatigue. It is concluded that submaximal performance in SevHyp is partly reduced by a mechanism related directly to brain oxygenation.  相似文献

Introduced, non-native organisms are of global concern, because biological invasions can negatively affect local communities. Arbuscular mycorrhizal (AM) fungal communities have not been well studied in this context. AM fungi are abundant in most soils, forming symbiotic root-associations with many plant species. Commercial AM fungal inocula are increasingly spread worldwide, because of potentially beneficial effects on plant growth. In contrast, some invasive plant species, such as the non-mycorrhizal Alliaria petiolata, can negatively influence AM fungi. In a greenhouse study we examined changes in the structure of a local Canadian AM fungal community in response to inoculation by foreign AM fungi and the manipulated presence/absence of A. petiolata. We expected A. petiolata to have a stronger effect on the local AM fungal community than the addition of foreign AM fungal isolates. Molecular analyses indicated that inoculated foreign AM fungi successfully established and decreased molecular diversity of the local AM fungal community in host roots. A. petiolata did not affect molecular diversity, but reduced AM fungal growth in the greenhouse study and in a in vitro assay. Our findings suggest that both introduced plants and exotic AM fungi can have negative impacts on local AM fungi.  相似文献

The well documented biochemical profile of Brassicaceae, oligophagy of the herbivore Plutella xylostella (L.) (Lepidoptera: Plutellidae), and host specialization of the parasitoid Diadegma insulare (Cresson) (Hymenoptera: Ichneumonidae) provide an ideal system for investigating tritrophic interactions mediated by nutritional quality of plants. We evaluated the bottom-up effects of five soil fertility regimes on nutritional quality of canola (Brassica napus L.) and then on several fitness correlates of female and male D. insulare as mediated through P. xylostella. Variation in soil fertility influenced the nutritional quality of host plants and this in turn affected the performance of D. insulare. In general, D. insulare performed best on plants grown with 3.0 g fertilizer pot−1; these plants had 2.06-, 3.77-, and 1.02-fold more nitrogen, phosphorous and potassium, respectively than ones grown without any added fertilizer. P. xylostella escape from D. insulare was highest (32%) on plants grown at 1.0 g fertilizer, and this could be attributed to both physical and physiological defense mechanisms mediated by host plant nutritional quality. Plant stress and plant vigor are competing paradigms pertaining to the performance of herbivorous insects on their host plants. These hypotheses were originally proposed to predict responses of herbivores, but may also explain the effects of plant quality on koinobiont parasitoids, such as D. insulare.  相似文献

The growth response of the hyphae of mycorrhizal fungi has been determined, both when plant and fungus together and when only the fungus was exposed to a temperature change. Two host plant species, Plantago lanceolata and Holcus lanatus, were grown separately in pots inoculated with the mycorrhizal fungus Glomus mosseae at 20/18 degrees C (day/night); half of the pots were then transferred to 12/10 degrees C. Plant and fungal growth were determined at six sequential destructive harvests. A second experiment investigated the direct effect of temperature on the length of the extra-radical mycelium (ERM) of three mycorrhizal fungal species. Growth boxes were divided in two equal compartments by a 20 micro m mesh, allowing only the ERM and not roots to grow into a fungal compartment, which was either heated (+8 degrees C) or kept at ambient temperature. ERM length (LERM) was determined on five sampling dates. Growth of H. lanatus was little affected by temperature, whereas growth of P. lanceolata increased with temperature, and both specific leaf area (SLA) and specific root length (SRL) increased independently of plant size. Percentage of colonized root (LRC) and LERM were positively correlated with temperature when in symbiosis with P. lanceolata, but differences in LRC were a function of plant biomass. Colonization was very low in H. lanatus roots and there was no significant temperature effect. In the fungal compartment LERM increased over time and was greatest for Glomus mosseae. Heating the fungal compartment significantly increased LERM in two of the three species but did not affect LRC. However, it significantly increased SRL of roots in the plant compartment, suggesting that the fungus plays a regulatory role in the growth dynamics of the symbiosis. These temperature responses have implications for modelling carbon dynamics under global climate change.  相似文献

In tropical ecosystems with highly weathered soils, transformation of organic phosphorus (P) to bioavailable inorganic P plays a crucial role for the nutrition of organisms. In these ecosystems, P is suspected to be growth-limiting and might therefore be affected by atmospheric nutrient deposition occurring even in remote areas such as montane rainforests. We assessed effects of P and nitrogen (N) addition on net and gross P mineralization rates, and microbial P immobilization in the organic layer along an altitudinal gradient of a tropical montane rainforest in Ecuador. Net P mineralization rates amounted to 1.8 ± 0.9 (at 1000 m a.s.l.), 3.7 ± 0.6 (at 2000 m) and 2.0 ± 0.4 (at 3000 m) mg P kg?1 day?1. Altitudinal differences led to an increased microbial P immobilization at 1000 m compensating for a higher gross P mineralization. P addition increased net P mineralization rates at 2000 and 3000 m, suggesting a higher P demand at 1000 m. At higher altitudes, P likely was released as a by-product during organic matter decomposition. Gross P mineralization, determined by means of the isotopic dilution approach, could not be calculated for control and N treatments due to rapid microbial P immobilization. For P (+N) treatments, gross P mineralization rates were lowest at the 1000 m site towards the end of the long-term incubation period. Atmospheric P deposition in the tropics might lead to P fertilization effects through direct input as well as through acceleration of P release from organic matter, thereby increasing P availability for organisms.  相似文献

Abstract

Nutrient addition experiments initiated in 1980 on the Keen of Hamar, Shetland Isles, have produced a unique dataset of long-term vegetation response to amendments of major plant growth nutrients (N, P, NP, NPK and NPKCa). Previous studies have reported the notable impact of phosphorus on the ‘serpentine debris’ community, and the negligible effect of nitrogen. However, a survey in 2010 provided our first indication that the experimentally-induced phosphorus effect was weakening and this was consolidated by further surveys of vegetation cover and community composition. This community shift might have been different had the local climate acted synergistically with phosphorus additions: in the last few years of the study the Shetland Isles experienced particularly low spring rainfall – the dry spell may have been a well-timed environmental filter driving community recovery rather than a permanent change of state to heathland on an organic soil. The longevity of our investigation is a unique opportunity to explore vegetation response to the key drivers of global environmental change, namely climate change, eutrophication as a result of agricultural intensification, and the potential for invasion of species as new resource-rich niches become available.  相似文献

Genetic modification of plants has become common practice. However, root-specific genetic modifications have only recently been advocated. Here, a review is presented regarding how root-specific modifications can have both plant internal and rhizosphere-mediated effects on aboveground plant properties and plant performance. Plant internal effects refer to pleiotropic processes such as transportation of the modified gene product. Rhizosphere-mediated effects refer to altered plant-rhizosphere interactions, which subsequently feed back to the plant. Such plant-soil feedback mechanisms have been demonstrated both in natural systems and in crops. Here how plant internal and rhizosphere-mediated effects could enhance or counteract improvements in plant properties for which the genetic modification was intended is discussed. A literature survey revealed that rice is the most commonly studied crop species in the context of root-specific transgenesis, predominantly in relation to stress tolerance. Phytoremediation, a process in which plants are used to clean up pollutants, is also often an objective when transforming roots. These two examples are used to review potential effects of root genetic modifications on shoots. There are several examples in which root-specific genetic modifications only lead to better plant performance if the genes are specifically expressed in roots. Constitutive expression can even result in modified plants that perform worse than non-modified plants. Rhizosphere effects have rarely been examined, but clearly genetic modification of roots can influence rhizosphere interactions, which in turn can affect shoot properties. Indeed, field studies with root-transformed plants frequently show negative effects on shoots that are not seen in laboratory studies. This might be due to the simplified environments that are used in laboratories which lack the full range of plant-rhizosphere interactions that are present in the field.  相似文献

The community of ectomycorrhizal (ECM) and co-associated fungi from a serpentine site forested with Pinus sylvestris and Quercus petraea was explored, to improve the understanding of ECM diversity in naturally metalliferous soils. ECM fungi were identified by a combination of morphotyping and direct sequencing of the nuclear ribosomal internal transcribed spacer region 2 and of a part of the large-subunit region. Co-associated fungi from selected ECM were identified by restriction fragment length polymorphism and sequencing of representative clones from libraries. Polymerase chain reaction with species-specific primers was applied to assess patterns of association of ECM and co-associated fungi. Twenty ECM species were differentiated. Aphyllophoralean fungi representing several basidiomycete orders and Russulaceae were dominant. Phialocephala fortinii was the most frequently encountered taxon from the diverse assemblage of ECM co-associated fungi. A ribotype representing a deeply branching ascomycete lineage known from ribosomal deoxyribonucleic acid sequences only was detected in some ECM samples. A broad taxonomic range of fungi have the potential to successfully colonise tree roots under the extreme edaphic conditions of serpentine soils. Distribution patterns of ECM-co-associated fungi hint at the importance of specific inter-fungal interactions, which are hypothesised to be a relevant factor for the maintenance of ECM diversity.  相似文献

Background

Current knowledge of plant-soil feedback is based largely on single end point studies with soils conditioned by monocultures, but accounting for variability in the ecological impacts of feedback effects may require understanding how feedback develops over time and in multi-species plant communities.

Methods

To examine temporal development and additivity of feedback, two pairs of native and non-native congeneric grasses were grown alone or in mixtures to create six soil conditioning treatments. We measured plant growth and feedback on the soils over 19 months and addressed whether plant biomass was additive or non-additive between soils treated by mixtures and their constituent monocultures.

Results

For native grasses, plant-soil feedback either became progressively more negative through time or switched from neutral to negative. Feedback to non-native grasses was variably neutral to positive. Final biomass of the grasses growing on soils conditioned by mixtures was generally an additive function of growth on soils conditioned by the component monocultures, except native grasses growing in soils conditioned by their own congener mixtures, which were non-additive.

Conclusions

Temporal variation and non-additivity in feedback suggest that extrapolation to communities may be complex. More work is needed to assess the generality of temporal and scaling effects.  相似文献

Fungal endophytes of the genus Epichloë live intercellulary in above ground plant parts of many pooid ‘grasses of the temperate regions. The associations are characterized by single genotype entities since a given host individual normally contains a single endophyte genotype. They can persist over the life span of the hosts. This study examines whether two fungal genotypes can co-exist within a host plant, and how fungal genotypes are distributed within a host in the case of double infections. We selected four Epichloë bromicola strains that we identified as unique genotypes through RAPD‘ analysis. Young Bromus erectus plants, derived from callus cultures, were artificially inoculated with all possible double-strain mixtures of these fungal genotypes. For identification of fungal genotypes in planta, we designed genotype-specific primer pairs that flanked size-variable loci in the fungal genomes. Diagnostic PCR revealed that only one fungal genotype was present in most inoculated plants, but double infections were also observed with a frequency of 8% of all infected plants. Subsequent analyses of individual tillers of doubly infected plants revealed that, in a given tiller, both the leaf-blade and the leaf-sheath were colonized with only one endophyte genotype. Tillers without any detectable fungal DNA were also observed. Thus, co-existence of multiple endophyte genotypes within a single host plant is governed by mutual exclusion at the tiller level.  相似文献

Arbuscular mycorrhizal symbiosis is induced upon a series of recognition events involving the reorganization of both plant and fungal cellular programs culminating in the formation of appressoria on the epidermal root cells. In this work we monitored for the first time the genetic changes occurring in the fungal partner during early appressorium development. We established an in vitro system of Glomus mosseae and Petroselinum crispum for studying appressorium formation and found that after 120 h first appressoria developed in the root epidermis. We have constructed a fungal subtractive suppressive library enriched in genes up-regulated at this stage. Our aim was to identify early signaling events during plant recognition leading to appressoria formation. The library contains 375 clones with an average size of 500 bp. From these, 200 clones were sequenced and most of them represent gene fragments with no known homologues (63%) and therefore putative new genes specific to the mycorrhiza symbiosis. Reverse-Northern blot and RT-PCR analyses confirmed that ca. 30% of the genes present in the library were up-regulated upon plant induction after 120 h. Among the genes with homologues in other organisms we found several genes common to other plant-microbe interactions including some genes related to Ca2+-dependent signaling. The up-regulation of these genes opens the possibility that Ca2+ plays a role in the early stages of mycorrhiza formation as it has been found in other plant-microbe interactions such as the Rhizobium symbiosis or the Magnaporthe grisea/rice pathogenic interaction.  相似文献
